为探讨新鲜无花果枝提取物(FBE)对体外培养的人胃癌细胞株BGC-823增殖和凋亡的影响,用不同浓度FBE处理胃癌BGC-823细胞,采用细胞形态学观察,细胞活力测定(MTY法)及免疫组织化学法检测增殖细胞核抗原(PCNA)表达情况来评价FBE对BGC-823细胞体外增殖的影响,应用流式细胞术的膜联蛋白V/碘化丙啶技术(Annexin V/PI法)检测FBE诱导BGC-823细胞体外凋亡的情况.形态学观察发现中高剂量(>0.5mg/mL)组处理4 h细胞出现明显凋亡,24 h检测到中高剂量(>0.5 mg/mE)组细胞增殖活力和增殖细胞核抗原表达显著降低(P<0.05),流式细胞仪检测到FBE处理4 h所有剂量组细胞平均凋亡率(9.76%,10.87%,14.29%,49.67%,71.37%)均高于对照组(6.1%)(P<0.05或P<0.01),以上作用效果呈现剂量依赖性.以上结果说明无花果枝提取物能够通过诱导胃癌BGC-823细胞凋亡从而抑制其体外的生长与增殖.  相似文献

为探讨B-RAF基因特异的siRNA干扰对胃癌BGC823细胞的增殖和凋亡的影响, 设计并合成B-RAF小分子干扰RNA(B-RAF-siRNA)和阴性对照siRNA, 用TransMessenger介导转染胃癌BGC823细胞, RT-PCR分析检测胃癌BGC823细胞中B-RAF基因以及Bcl-2基因的表达; MTT检测胃癌BGC823细胞增殖情况; 流式细胞仪检测细胞凋亡情况, 并与对照组进行比较。TransMessenger能够有效介导B-RAF-siRNA和阴性对照siRNA转染胃癌BGC823细胞, TransMessenger介导的B-RAF-siRNA有效地抑制胃癌BGC823细胞B-RAF以及Bcl-2基因的表达, 与对照组相比, 抑制率达90.0%以上, 最高达100%; 同时明显抑制胃癌BGC823细胞增殖; 促进胃癌BGC823细胞的凋亡(P < 0.01)。B-RAF基因特异的siRNA干扰能有效地抑制胃癌BGC823细胞中B-RAF基因以及Bcl-2基因的表达, 同时促进胃癌细胞凋亡和抑制胃癌细胞增殖。  相似文献

目的:通过康莱特联合顺铂对宫颈癌SiHa细胞增殖和凋亡的影响,探讨其作用机制。方法:体外培养宫颈癌Siha细胞,分别将康莱特(浓度为1,2,4,6,8 mg/mL),顺铂(浓度梯度为1.5,3,6,9,12μg/mL),单独作用于宫颈癌SiHa细胞,加药24h、48h用噻唑蓝(MTT法)检测细胞增殖情况。用流式细胞术检测康莱特组和顺铂组细胞24h凋亡率,选取合适的药物浓度(康莱特6 mg/mL,顺铂3μg/mL),进行联合用药,加药24h、48h用MTT法检测细胞增殖情况,用流式细胞术检测24h细胞凋亡率。结果:①MTT法显示加药后两组的24h、48h,宫颈癌SiHa细胞的抑制率均高于对照组(P0.05),并且在一定程度上呈浓度和时间依赖性。②联合用药时,细胞的抑制率和凋亡率要显著高于单独用药(P0.01)。结论:康莱特、顺铂单独或联合作用均能抑制SiHa细胞的增殖,促进其凋亡,且康莱特联合顺铂的作用要显著高于单独用药,康莱特与化疗药物联合使用可提高肿瘤细胞对化疗药物的敏感性。  相似文献

目的:研究青藤碱(sinomenine,SIN)对宫颈癌Hela细胞增殖的影响及其机制,为SIN在宫颈癌的预防和治疗上提供实验依据。方法:不同浓度SIN分别处理体外培养的人宫颈癌细胞系Hela细胞后,采用噻唑蓝(Mar)法检测处理24h、48h、72h后Hela细胞的增殖活性,流式细胞仪测定细胞周期和细胞凋亡。结果:1.0.1、0.2、0.4、0.625、1.25、2.5mmml/L SIN处理Hela细胞24h、48h、72h后,细胞增殖明显受到抑制,呈时间和剂量依赖性特点;2.流式细胞仪细胞周期分析表明,SIN处理组G1期细胞比例明显增加,S期细胞比例明显减少,两组比较有统计学意义;3.细胞凋亡分析表明,SIN处理组细胞凋亡率较对照组升高,呈时间和剂量依赖性特点;结论:SIN在体外能有效抑制宫颈癌细胞生长,其机制可能与其阻滞细胞周期、诱导细胞凋亡有关,SIN有望应用于宫颈癌的辅助治疗。  相似文献

为了寻找能抑制肿瘤细胞增殖的药物,本研究探讨了不同浓度白花蛇舌草(Hedyotis diffusa)对人胃癌MKN-45细胞的影响,使用不同浓度的白花蛇舌草处理人胃癌MKN-45细胞24 h、48 h、72 h,共聚焦显微镜下观察细胞形态,流式细胞术检测细胞周期,qRT-PCR和Western blotting检测CCNB1和CCND1的表达水平。结果表明,在一定浓度范围内,白花蛇舌草可抑制人胃癌MKN-45细胞增殖且呈剂量时间依赖性;白花蛇舌草处理人胃癌MKN-45细胞24 h、48 h、72 h后的IC50值分别为111.759μg/mL、26.878μg/mL、16.179μg/mL;随着白花蛇舌草的浓度增加,显微镜视野下细胞数量减少;G1期细胞减少,S期细胞增多;CCNB1和CCND1基因表达水平显著降低。白花蛇舌草能能抑制人胃癌MKN-45细胞增殖,阻滞细胞于S期,降低CCNB1和CCND1基因的表达。  相似文献

目的通过观察大豆低聚糖对胃癌癌细胞株BGC-823细胞的细胞周期和细胞凋亡的影响,探索乳酸杆菌发酵滤液对胃癌细胞作用的可能机制。方法用光镜和流式细胞仪分析不同浓度大豆低聚糖对BGC-823细胞的凋亡诱导效果;用流式细胞仪分析不同浓度大豆低聚糖对BGC-823细胞细胞周期的影响。结果大豆低聚糖可以诱导BGC-823细胞的凋亡。形态学观察处理后的BGC-823细胞,可见细胞变形,细胞皱缩,体积变小,细胞间隙增大,细胞核固缩。流式细胞仪分析50 mg/ml和100 mg/ml大豆低聚糖作用48 h和72 h BGC-823细胞的凋亡比例,分别为6.76%和7.93%。50 mg/ml大豆低聚糖作用48 h,引起BGC-823细胞G1期阻滞,100 mg/ml大豆低聚糖作用48 h,引起BGC-823细胞出现S期阻滞。结论大豆低聚糖可诱导部分BGC-823细胞凋亡。大豆低聚糖对BGC-823细胞的生长抑制作用在低浓度时可能通过G1期阻滞实现,在高浓度时可能通过S期阻滞实现。  相似文献

目的:探讨选择性环氧合酶(COX-2)抑制剂塞来昔布对胃癌细胞株BGC823多药耐药(mdr)1表达的影响.方法:胃癌细胞株BGC823经浓度分别为0、10、100μ mol/L的塞来昔布处理后,酶联免疫吸附试验检测塞来昔布对胃癌细胞前列腺素E2(PGE2)分泌的影响,24、48 h后用RT-PCR检测多药耐药(mdr)1 mRNA表达,48 h后用免疫细胞化学染色法检测P-gp表达.结果:塞来昔布可显著抑制胃癌细胞株BGC823 PGE2分泌.并呈浓度依赖性(P<0.05).不同浓度塞来昔布作用于细胞后,胃癌细胞株BGC823的mdrl/P-gp表达受不同程度抑制,100μ mol/L的塞来昔布对mdrl mRNA表达抑制作用强于10μ mol/L(P<0.01).不同浓度药物与测量时间为交互作用,作用48h与24h相比,塞来昔布对mdrl mRNA表达的抑制作用更强(P<0.01).结论:塞来昔布可抑制BGC823的mdrl/P-gp表达,且呈剂量效应关系.塞来昔布可能通过抑制COX-2活性,抑制COX-2下游产物PGE2表达,从而抑制P-gp表达.选择性COX-2抑制剂可能有助于减轻肿瘤细胞对化疗药物的耐药性.  相似文献

为研究层粘连蛋白(laminin,LN)促进肿瘤细胞生长作用,采用脉冲标记计数有丝分裂百分率(percentage labeling mitosis,PLM)法测得体外培养人胃癌 (BGC 823) 细胞周期时间为41 h,其中G1期时间为24.5 h. 分裂细胞脱离法获取分裂期细胞,继续培养23 h,在细胞运行进入G1晚期时,将其置于LN 0、0.11、0.55、1.10 μmol/L基质上孵育4 h; 细胞荧光光度计检测晚G1期细胞内Ca2+浓度、钙调蛋白、DNA含量. 结果显示,LN与其膜上受体结合后引起细胞内Ca2+浓度、钙调蛋白、DNA含量增加,尤以在0.55 μmol/L LN作用显著(P<0. 001).蛋白质免疫印迹分析证明,cPKC α呈现表达,提示 LN与其受体结合可增强其细胞cPKC-α的活性;分析G1期细胞周期蛋白E(cyclinE)、细胞周期蛋白依赖性激酶CDK2表达水平,呈现逐渐增强的趋势; LN可诱导c-Myc蛋白呈现高表达,提示 LN与其受体结合增强与细胞增殖密切相关的基因表达;在LN作用前后的BGC 823细胞均未检测到Bax蛋白表达.结果提示,在人胃癌 (BGC 823)细胞G1/S期交界处,层粘连蛋白与其膜上受体结合引起细胞内Ca2+浓度升高,诱导钙调蛋白的释放,其含量增加,增强蛋白激酶C的活化,导致细胞内DNA含量增加、G1/S期细胞周期蛋白与CDK表达增强、诱导原癌基因c-Myc呈持续表达状态,而凋亡基因Bax不表达.  相似文献

目的:初步探索EphA3与胃癌细胞BGC823增殖的关系。方法:用T_4DNA连接酶将EphA3的全长cDNA片段连接到酶切后的慢病毒载体pCDH-EF1-MCS-T2A-Puro中制备慢病毒,感染胃癌BGC823细胞建立过表达EphA3细胞系,Western印迹检测目的蛋白的表达,CCK8细胞生长实验和裸鼠成瘤实验检测EphA3对BGC823细胞增殖的影响。结果:双酶切鉴定与基因测序表明过表达EphA3慢病毒载体构建成功;Western印迹表明建立了过表达EphA3的BGC823细胞系,高表达EphA3对BGC823细胞的增殖和成瘤后生长有明显的促进作用。结论:EphA3促进BGC823胃癌细胞增殖,为胃癌的靶向性治疗及个性化治疗提供了线索。  相似文献

目的:探究长非编码RNA SNHG18对胃癌细胞增殖和凋亡的影响。方法:采用实时定量PCR(qRT-PCR)技术检测人胃癌组织及癌旁组织和胃癌细胞系中lncRNA SNHG18的表达;采用MTT和克隆形成试验观察转染SNHG18过表达质粒后胃癌细胞BGC823增殖活力的变化;通过流式细胞术检测lncRNA SNHG18对胃癌细胞BGC823凋亡的影响。结果:相较于癌旁组织和胃正常粘膜上皮细胞系GSE-1,胃癌组织及胃癌细胞系中SNHG18的表达水平显著降低(P0.05);胃癌细胞过表达SNHG18增殖活力以及克隆形成的能力均显著降低(P0.05),而细胞凋亡率明显升高(P0.05)。结论:胃癌组织中长非编码RNA SNHG18呈低表达,可促进胃癌细胞增殖并抑制其凋亡,可能在胃癌发生发展过程中发挥重要作用。  相似文献

FSK88, a forskolin derivative, was extracted and purified from cultured tropical plant roots, Coleus forskohlii. Our previous studies have demonstrated that FSK88 can inhibit HL-60 cell proliferation and induce the differentiation of HL-60 cells to monocyte macrophages. In this study, we showed that FSK88 can induce apoptotic death of human gastric cancer BGC823 cells in a dose- and time-dependent manner. Results showed that FSK88-induced apoptosis was accompanied by the mitochondrial release of cytochrome c and activation of caspase-3 in BGC823 cells. Furthermore, treatment with caspase-3 inhibitor (z-DEVD-fmk) was capable of preventing the FSK88-induced caspase-3 activity and apoptosis. FSK88-induced apoptosis in human gastric cancer BGC823 cells was also accompanied by the up-regulation of Bax, Bad and down-regulation of Bcl-2. Theses results clearly demonstrated that the induction of apoptosis by FSK88 involved multiple cellular and molecular pathways and strongly suggest that pro- and anti-apoptotic Bcl-2 family genes, mitochondrial membrane potential (Deltapsi(m)), cytochrome c, and caspase-3, participate in the FSK88-induced apoptotic process in human gastric cancer BGC823 cells.  相似文献

目的探讨沉默生存素(survivin)基因表达的干扰RNA对人胃癌BGC-823细胞增殖和成瘤能力的影响。方法应用已经在细胞上验证能够有效沉默survivin的小分子干扰RNA(shRNA-survivin-1),并在体外实验的基础上,建立稳定表达干扰RNA细胞系,进一步探讨干扰RNA稳定表达对胃癌BGC-823细胞生长和裸鼠移植成瘤的影响。结果 shRNA-survivin-1有效沉默人胃癌BGC-823细胞survivin mRNA的表达,成功筛选shRNA-sur-vivin-1稳定表达细胞株BGC/siRNA-1细胞,实验表明,BGC/siRNA-1细胞的生长曲线缓慢上升,细胞增殖能力下降;BGC/siRNA-1细胞裸鼠移植成瘤体积与对照组相比,明显减小(P〈0.05)。结论 shRNA-survivin-1可以沉默survivin基因的表达,可以显著抑制胃癌BGC-823细胞的增殖,并降低胃癌BGC-823细胞的成瘤能力,本研究为靶向survivin的RNA干扰在胃癌的基因治疗提供了有力的理论依据和技术储备。 更多还原  相似文献

Cancer is the second leading cause of death worldwide. Edible medicinal mushrooms have been used in traditional medicine as regimes for cancer patients. Recently anti-cancer bioactive components from some mushrooms have been isolated and their anti-cancer effects have been tested. Pleurotus ferulae, a typical edible medicinal mushroom in Xinjiang China, has also been used to treat cancer patients in folk medicine. However, little studies have been reported on the anti-cancer components of Pleurotus ferulae. This study aims to extract bioactive components from Pleurotus ferulae and to investigate the anti-cancer effects of the extracts. We used ethanol to extract anti-cancer bioactive components enriched with terpenoids from Pleurotus ferulae. We tested the anti-tumour effects of ethanol extracts on the melanoma cell line B16F10, the human gastric cancer cell line BGC 823 and the immortalized human gastric epithelial mucosa cell line GES-1 in vitro and a murine melanoma model in vivo. Cell toxicity and cell proliferation were measured by MTT assays. Cell cycle progression, apoptosis, caspase 3 activity, mitochondrial membrane potential (MMP), migration and gene expression were studied in vitro. PFEC suppressed tumor cell growth, inhibited cell proliferation, arrested cells at G0/G1 phases and was not toxic to non-cancer cells. PFEC also induced cell apoptosis and necrosis, increased caspase 3 activity, reduced the MMP, prevented cell invasion and changed the expression of genes associated with apoptosis and the cell cycle. PFEC delayed tumor formation and reduced tumor growth in vivo. In conclusion, ethanol extracted components from Pleurotus ferulae exert anti-cancer effects through direct suppression of tumor cell growth and invasion, demonstrating its therapeutic potential in cancer treatment.  相似文献

Understanding the molecular mechanism of gastric cancer cell apoptosis is pivotal for the development of precise therapies targeting this disease. In the present study, we examined the effects of annexin A7 inhibition on the apoptosis of gastric cancer cells and the growth of tumour xenografts in vivo. Expression of annexin A7 in BGC823 cells was suppressed by small interference RNA, and cells apoptosis was assessed by flow cytometry. The mechanism by which annexin A7 mediates apoptosis in BGC823 cells was explored by determining the expression of key apoptosis regulators. In addition, by suppressing annexin A7 in BGC823 cells with small hairpin RNA, we studied the effects of annexin A7 inhibition on in vivo tumour growth. Our results showed that inhibiting annexin A7 expression induced more than fivefold increase in BGC823 cell apoptosis in vitro. This was in concord with a significant decrease of Bcl-2 expression and increases of Bax, Caspase-3, and Caspase-9. The activities of caspase-3 and caspase-9 were increased by 2.95?±?0.18 and 3.70?±?0.33 times, respectively, upon the annexin A7 downregulation in BGC823 cells. Importantly, suppressing annexin A7 showed the same apoptotic mechanism in vivo and significantly inhibited the growth of BGC823 xenografts in mice. These data suggest that annexin A7 likely protects gastric cells from apoptosis and targeting it may represent a valuable strategy in future therapeutic development.  相似文献

摘要目的:观察生半夏、南星中药水提物对缺氧环境中人胃癌细胞株BGC823细胞HIF-lα蛋白表达和侵袭力的影响。方法:运用CoCl2(氯化钴)诱导BCG823细胞缺氧,使得细胞中HIF-1α蛋白表达升高.实验组加入生半夏、南星水提物对细胞进行预处理,然后在进行缺氧诱导。通过甲基噻唑基四唑法(MTT)检测细胞活性,使用Transwell检测细胞侵袭能力变化,RT-PCR、Weste-rn blotting分别检测HIF-lαmRNA及蛋白含量及变化。结果:生半夏、南星水提物能抑制人胃癌BGC823细胞的增殖;生半夏、南星水提物均能抑制缺氧诱导胃癌细胞的侵袭力,并且能降低HIF-1αmRNA及蛋白表达。结论:生半夏、南星水提物可抑制人胃癌BGC823细胞的增殖,抑制人胃癌BGC823细胞侵袭力,可能通过降低HIF-1α蛋白表达有关。  相似文献

The aim of this study was to investigate the biological characteristics of the RASAL1 gene in a well-differentiated gastric cancer cell line MKN-28 and a poorly differentiated gastric cancer cell line BGC-823 cells, using RNA interference and gene transfection technology, respectively. MKN-28 cells were transfected with the shRNA of RASAL1 and BGC-823 cells were transfected with the pcDNA 3.1 plasmid vector containing RASAL1. RT-PCR and western blotting were then used to detect the expression of RASAL1 mRNA and protein. The activities of RAS and extracellular signal-regulated kinase 1/2 were analyzed by the pull-down method and western blotting. The proliferate capacity, apoptosis rate, invasive and migratory potentials of MKN-28 or BGC-823 cells were also measured by Cell Counting Kit-8 cell proliferation assay, propidium iodide/Annexin V staining coupled with flow cytometry, and transwell chamber assays, respectively. Measurement of RASAL1 mRNA and protein expression in two cells revealed successful transfection of the shRNA of RASAL1 and RASAL1-pcDNA3.1 plasmid into these two cells. Moreover, decreased expression of RASAL1 in MKN-28 cells resulted in increased expression of RAS-GTP and p-ERK1/2. Interestingly, decreased expression of RASAL1 inhibited apoptosis and facilitated cell proliferation, invasion and migration. The increased expression of RASAL1 in BGC-823 cells caused declined expression of RAS-GTP and p-ERK1/2, as well as promoted apoptosis and restrained cell proliferation, invasion and migration. The down-regulation of RASAL1 promoted the proliferation, invasion and migration of gastric cancer MKN-28 cells, and up-regulation of RASAL1 inhibited the proliferation, invasion and migration of BGC-823 gastric cancer cells by regulating the RAS/ERK signaling pathway. Thus, our results suggest that RASAL1 may play an important role as a tumor suppressor gene in gastric cancer.  相似文献

P130Cas和paxillin分子是整合素家族下游重要的衔接分子.为了探索这两个分子在肿瘤细胞抗失巢凋亡中的作用,应用RNAi技术分别抑制抗失巢凋亡的胃癌细胞BGC82 3中paxillin和p130cas基因的表达,观察它们对细胞失巢性生长的影响.依据siRNA设计原则,分别设计针对p130cas和paxillin的两条序列;成功的构建了特异性封闭上述两分子的载体pWH1 p130cas和pWH1 paxillin .构建的载体瞬时转染贴壁培养和失巢培养的BGC82 3后,RT PCR和Western印迹检测发现paxillin和p130Cas分子在mRNA及蛋白水平的表达量均明显降低;倒置显微镜下观察发现,贴壁培养的胃癌细胞发生皱缩、脱落;失巢培养的细胞聚集成团的现象受到明显抑制,细胞团比对照组小,且较松散;MTT实验结果表明,失巢培养的BGC82 3pWH1 paxillin 组细胞存活率(32 19%±6 11% )和BGC82 3pWH1 p13 0cas组细胞存活率(2 8 5 2 %±5 0 2 % )与对照组相比显著下降(P <0 0 1vscontrol) ;FCM实验结果发现与失巢培养的对照组相比,BGC82 3pWH1 paxillin和BGC82 3pWH1 p13 0cas组细胞G1期抬高,并出现了凋亡峰.运用RNAi技术分别抑制了BGC82 3细胞中paxillin和p130Cas分子的表达,初步证明paxillin和p130Cas是细胞存活的重要信号分子,在肿瘤细胞抗失巢凋亡过程中具有重要的作用.  相似文献

【目的】研究从双歧杆菌属两歧双歧杆菌(Bifidobacterium bifidum)提取的细胞表面成分胞外多糖(Exopolysaccharide, B.EPS)对人胃癌细胞BGC-823的生长抑制作用及对端粒酶限速因子hTERT活性的影响。【方法】将三种不同浓度B.EPS体外作用于胃癌细胞BGC823,MTT法检测细胞生长抑制率并辅以形态学观察;异硫氰酸盐(FITC)联合PI染色,通过流式细胞术检测肿瘤细胞初期调亡情况;肿瘤细胞端粒酶限速因子hTERT mRNA经RT-PCR检测B.EPS对端粒酶活性抑制作用;通过荧光分光光度计显示B.EPS 对胃癌细胞作用后胞内Ca2+含量变化。【结果】经过检测发现,B.EPS对人胃癌细胞BGC823的生长显著抑制(P<0.05)呈剂量时间反应关系;细胞中hTERT mRNA在B.EPS的作用下表达降低(P<0.05),有一定剂量效应关系;随着B.EPS对肿瘤细胞的抑制,细胞内Ca2+含量显著增加(P<0.05)。【结论】B.EPS诱导人胃癌细胞BGC823调亡的机制可能与改变肿瘤细胞端粒酶限速因子hTERT mRNA表达量和细胞内钙离子浓度有关。  相似文献
