几丁质酶具有降解几丁质的作用,其降解产物氨基寡糖和几丁低聚糖在农业、食品、医药等领域具有重要的应用价值和广泛的应用前景。自然界中多种微生物可以产生几丁质酶,而其在细菌体内的合成受精密调控。粘质沙雷氏菌作为一种高产几丁质酶的菌种,在农业生防领域具有很大的应用潜力。本综述从粘质沙雷氏菌几丁质酶基因的分类和结构特征、几丁质酶基因克隆、表达和调控以及几丁质酶的应用等方面论述了粘质沙雷氏菌几丁质酶基因的研究进展,为粘质沙雷氏菌几丁质酶基因和功能蛋白的利用提供理论基础。  相似文献

【目的】从松材线虫的媒介天牛蛹室及其气管中获得的黏质沙雷氏菌Serratia marcescens AHPC29对松材线虫具有致病能力,本研究旨在探究黏质沙雷氏菌AHPC29对松材线虫引起的松树萎蔫病的防治效果以及该菌株在实验室的增菌条件。【方法】通过对温室内人工感染松材线虫的松树灌溉菌剂,分析黏质沙雷氏菌AHPC29对松树萎蔫病的作用效果;通过单组分筛选和正交实验确定培养基组分和培养条件对其生长的影响,探究黏质沙雷氏菌AHPC29的最佳增菌条件。【结果】对于感染松材线虫的松树,灌溉黏质沙雷氏菌AHPC29菌液的处理组生长状态优于对照组,并且树内松材线虫含量显著降低;黏质沙雷氏菌AHPC29增菌的最佳培养基配比为0.1%乳糖、0.5%复合氨基酸、0.5% KNO3、1.5% MgSO4,其中影响最大的组分为氮源;培养时其菌液最佳接种量为7%,最佳装液量为40%,最佳转速为180 r·min-1,最佳温度为30 ℃,最适培养时长为36 h。【结论】本研究获得了黏质沙雷氏菌AHPC29的最佳增菌条件,并证实该菌具有良好的松树萎蔫病防治应用潜力,为其作为生防菌的应用提供了理论基础。  相似文献

根据生理生化特征和16S rDNA序列,对一株从新疆苦豆子Sophora alopecuroides中分离的、对耐甲氧西林金黄色葡萄球菌(Methicillin-resistant Staphylococcus aureus,MRSA)具有较强拮抗作用的内生细菌菌株KDNB6进行鉴定;并通过硅胶柱、凝胶柱层析从该菌发酵产物中分离纯化了抗MRSA的活性物质。结果表明菌株KDNB6各项特征与粘质沙雷氏菌Serratia marcescens最为接近,鉴定为S.marcescens。从菌株KDNB6的代谢产物中分离到一个抗MRSA的活性成分。  相似文献

病原褪色沙雷氏菌的有效分离与鉴定对控制由褪色沙雷氏菌引起的养殖东亚飞蝗感染病均具有一定的参考与应用价值,同时将为对蝗灾的生物防治研究提供借鉴。对从发病死亡的养殖东亚飞蝗中分离的10株细菌,进行了表观分类学指征、16S r RNA基因序列与系统发育、人工感染的致病作用等方面的检验与分析。结果表明为沙雷氏菌属的褪色沙雷氏菌,对供试养殖东亚飞蝗显示强致病性。褪色沙雷氏菌对东亚飞蝗有强致病性,是蝗虫养殖的重要致病菌。  相似文献

黑果枸杞不同组织内生细菌群落多样性   总被引:3,自引:1,他引:2  
【目的】黑果枸杞是我国荒漠区特有的药用盐生植物,本研究分析了黑果枸杞不同组织中内生细菌群落多样性特征及分布规律。【方法】应用Illumina MiSeq高通量测序技术对黑果枸杞内生细菌的16S rRNAV5-V7区域序列进行测定,并分析群落组成、多样性及功能等生物学信息。【结果】黑果枸杞不同组织内生细菌群落多样性及功能均有较大的差异。花、叶、果、茎和根产生的OTUs分别是182、173、119、187和254,群落多样性表现为根花果、茎叶。从门水平上看,变形菌门是优势菌门,在不同组织中均有分布,花、叶、果、茎和根中的相对丰度分别为87.66%、41.51%、81.76%、97.67%和61.85%。在属水平上显示内生细菌的分布表现出器官差异性。花部能够准确分类的优势菌属为沙雷氏菌属和不动杆菌属,相对丰度分别为11.57%和8.55%。叶部为红球菌属和慢生根瘤菌属,相对丰度分别为29.68%和5.53%。果实中为泛菌属、红球菌属和沙雷氏菌属,相对丰度分别为23.12%、5.52%和4.29%。茎部为沙雷氏菌属和假单胞菌属,相对丰度分别为12.03%和17.71%。根部为盐单胞菌属、Fodinicurvata和Lipingzhangella,相对丰度分别为24.18%、5.16%和4.86%。在不同组织中分布较广的盐单胞菌、沙雷氏菌、不动杆菌、红球菌、泛菌等菌属均具有较高耐盐性和促生、生防、降解有机污染物及抗氧化等功能。PICRUSt功能预测分析显示,黑果枸杞组织中内生细菌功能中涉及丰富的多糖、萜类和酮类、酶及维他命等次生代谢产物的生物合成。【结论】黑果枸杞内生细菌具有丰富的群落和功能多样性,拥有多种益生功能性状,也含有多个与人和植物体代谢相关的功能信息。不同组织优势菌属和功能信息各有不同,其中根部的内生细菌物种最丰富,花部和茎部参与各种代谢调控的细菌丰度最高。此外,不同组织中还含有大量未知种属的微生物类群,这些都为内生细菌功能利用和挖掘新的有益微生物资源提供广阔的发展空间。  相似文献

植物病原真菌是农业生产的主要威胁之一,使用生物制剂防治病原真菌被认为是更安全和可持续的方式。芽胞杆菌能产生多种抗真菌活性物质(脂肽、细菌素和酶等),是目前应用最广泛的生防菌。基于芽胞杆菌及其代谢产物的生物防治剂,可有效防治植物病原真菌,在农业生产中发挥着重要的作用。本文聚焦于芽胞杆菌代谢产物的生物防治潜力及其对抗3种常见植物病原真菌(稻瘟菌、尖孢镰刀菌、灰葡萄孢菌)的拮抗属性和机理研究等,通过调研近年来已发表的芽胞杆菌代谢产物抗菌的相关文献,对几种重要的芽胞杆菌代谢产物进行介绍,并总结了芽胞杆菌代谢产物对重要植物病原真菌的抗菌效果及其机制,同时对芽胞杆菌代谢产物对病原真菌的壁膜损伤、抑制真菌孢子萌发和菌丝生长以及竞争性结合真菌DNA等机制的研究手段和效果进行了总结,期望为今后芽胞杆菌类生防制剂的制备和应用提供指导策略。  相似文献

198 9年 ,筛选了 1株高产 phospholipaseC(PLC)的CW W 90 3菌株[1,2 ] ,据其形态特征、生理生化反应 ,初步将其归于弧菌科气单胞菌属[3 ] ,由于该菌株的许多生理生化特性与粘质沙雷氏菌相同。但其极生单鞭毛和无色素及少许生理生化特性与粘质沙雷氏菌相异。后经AutomatedBacteriaIdentificationSystem BiologMicroStationSystem检测 96种C源和N源的利用及其个体群体发育 ,说明其与粘质沙雷氏菌 (Serratiamarcescens)相符 ;并在基因组水平上研究该菌株的系统发育 ,从分子水平上对该菌株进行 16SrRNA序列分析煌同源性比较。根据CW W 90 3菌株 16SrRNA与GeneBank数据库中Serratiamarcescens的 16SrRNA的序列具有 99%的同源性 ,终将CW W 90 3菌株鉴定为粘质沙雷氏菌武汉株 (SerratiamarcescensWuhanstrain)。  相似文献

链霉菌是一类具有复杂的形态分化周期和强大的次级代谢能力的高GC含量的放线菌,能够利用其初级代谢产生的前体化合物和能量,合成多种结构复杂、功能多样的具有生物活性的次级代谢产物,在农业、食品、畜牧业、工业以及医药研究等领域都具有重要的价值。在链霉菌的形态分化后期常常伴随着次级代谢产物的生物合成,并且两者都受到复杂的网络调控;同时链霉菌的形态对次级代谢产物的产量和种类造成很大影响。对链霉菌生长周期的全面理解将加深对链霉菌形态分化与次级代谢产物合成关系的认识。本文将对链霉菌的形态分化过程、形态分化和抗生素合成两者共同的调控因子以及链霉菌形态与抗生素产量之间的关系进行综述,这将有助于理解抗生素的合成过程,也将会在缩短发酵周期、构建高产工程菌株、新型杀菌剂的研发以及新型抗生素的合成等方面给予我们启发。  相似文献

Nine fungal strains of Dichotomomyces cejpii were studied for their ability to produce antibiotic metabolites. It was found that they produced secondary exo- and endo metabolites with antimicrobial activity against Gram-negative and Gram-positive bacteria, yeasts and moulds and with a toxic effect against animal organs in vitro. A model oligotrophic aquatic system involving localization of fatty acids on a solid surface was used to quantitate scavenging by three bacteria; Leptospira biflexa patoc 1 which adheres reversibly, pigmented Serratia marcescens EF190 which adheres irreversibly, and a non-pigmented hydrophilic mutant of EF190. The Leptospira and pigmented Serratia displayed two distinct scavenging strategies which are related to their different methods of adhesion. The Leptospira efficiently scavenged [1-14C] stearic acid from the surface in 24 h, whereas the pigmented hydrophobic Serratia initially showed a faster rate of removal but the overall rate was considerably slower than that of the Leptospira. The hydrophilic, non-pigmented Serratia required 50h incubation to remove significant amounts of the labelled fatty acid. The greater scavenging ability of the hydrophobic pigmented Serratia strain compared to the hydrophilic non-pigmented mutant could not be attributed to differences in viability of fatty acid metabolism. The hydrophobicity of the pigmented Serratia allows for firmer adhesion and greater interaction with the surface localized nutrients.  相似文献

A soil bacterium capable of utilizing methyl parathion as sole carbon and energy source was isolated by selective enrichment on minimal medium containing methyl parathion. The strain was identified as belonging to the genus Serratia based on a phylogram constructed using the complete sequence of the 16S rRNA. Serratia sp. strain DS001 utilized methyl parathion, p-nitrophenol, 4-nitrocatechol, and 1,2,4-benzenetriol as sole carbon and energy sources but could not grow using hydroquinone as a source of carbon. p-Nitrophenol and dimethylthiophosphoric acid were found to be the major degradation products of methyl parathion. Growth on p-nitrophenol led to release of stoichiometric amounts of nitrite and to the formation of 4-nitrocatechol and benzenetriol. When these catabolic intermediates of p-nitrophenol were added to resting cells of Serratia sp. strain DS001 oxygen consumption was detected whereas no oxygen consumption was apparent when hydroquinone was added to the resting cells suggesting that it is not part of the p-nitrophenol degradation pathway. Key enzymes involved in degradation of methyl parathion and in conversion of p-nitrophenol to 4-nitrocatechol, namely parathion hydrolase and p-nitrophenol hydroxylase component “A” were detected in the proteomes of the methyl parathion and p-nitrophenol grown cultures, respectively. These studies report for the first time the existence of a p-nitrophenol hydroxylase component “A”, typically found in Gram-positive bacteria, in a Gram-negative strain of the genus Serratia. Biological control is being considered as an alternative or a supplemental way of reducing the use of chemicals in agriculture. An endophytic strain G3 with potential as a biocontrol agent was isolated from the stems of Triticum aestivum L. It was classified by 16S rDNA sequencing as a member of Serratia. Strain G3 displayed a broad spectrum of antifungal activity in vitro against a number of phytopathogens such as Botrytis cinerea, Cryphonectria parasitica, Rhizoctonia cerealis and Valsa sordida. Molecular mechanisms involved in biocontrol by Serratia sp. G3 was investigated for its potential application to plant health management. The results showed that G3 produces an array of antimicrobial exoproducts, including chitinase, protease, antibiotic pyrrolnitrin, and siderophores for iron competition. Moreover, it also produced the plant growth hormone indole-3-acetic acid, suggesting that multiple mechanisms and their synergistic effects may be involved in biocontrol of plant diseases. Additionally, strain G3 can produce at least ten N-acyl homoserine lactones (AHLs) signal molecules for cell to cell communication, including unsubstituted, 3-oxo, and 3-hydroxy at the C3 position through liquid chromatography-tandem mass spectrometry (LC-MS/MS), which is different from the previously reported Serraia species. For the first time, N-3-oxo-heptanoyl-homoserine lactone, one of the main molecules was reported in the genus Serratia. The role of AHL-dependent quorum sensing system in the interactions between the endophytic strain G3 and host plants and its potential application in improving biocontrol efficacy will be further explored.  相似文献

【背景】一直以来,链霉菌都是活性物质的主要生产者,近年来随着抗生素滥用引起的环境和微生物抗药性问题越发严重,挖掘高效生物防治因子和新型抗生素成为了解决以上问题的重要手段。【目的】通过获得植物内生链霉菌SAT1全基因组序列和次级代谢基因簇信息,利用比较基因组学和泛基因组学分析SAT1菌株的特殊性以及与其他链霉菌的共性,为阐明SAT1抑菌和内生机制提供理论基础,为揭示链霉菌的生态功能提供可靠数据。【方法】通过三代测序平台PacBio Sequel完成SAT1基因组测序,利用生物信息学技术进行注释和功能基因分类;分别利用RAxML和PGAP软件进行系统发育树的构建和泛基因组分析;次级代谢基因簇的预测和分析通过antiSMASH网站完成。【结果】获得SAT1菌株的全基因组完成图,该菌线性染色体长度约7.47 Mb,包含有4个质粒,GC含量近73%,共预测到7 550个蛋白编码基因,含有37个次级代谢基因簇,分属29个类型,其中默诺霉素基因簇与加纳链霉菌具有较高相似性。42株代表链霉菌中,单个菌株次级代谢基因簇数量为20-55个,主要类型为PKS类、Terpene类和Nrps类,而且含有大量杂合基因簇,各个菌株中特有基因数目较为庞大。【结论】链霉菌SAT1菌株在基因组特点以及次级代谢基因簇的数量和类型上与其余41株链霉菌具有一定的共性,其中潮霉素B基因簇和默诺霉素基因簇合成的相关物质可能与SAT1抑菌活性密切相关。42株链霉菌中次级代谢基因簇数量的多少与基因组大小成正相关,同时大量杂合基因簇以及庞大的特有基因数目的存在说明链霉菌在长期进化过程中存在了很高程度的水平基因转移现象,可能具有重要的生态功能。  相似文献

Summary Several endophytic diazotrophs were isolated from cultivars of rice and screened for their diazotrophy by nitrogenase assay and amplification of partial nifH gene. Ability of one of the diazotrophic endophytes, Serratia sp. (isolate EDA2 from cultivar ADT36) to colonize the rice seedlings grown in the presence of flavonoids and growth hormones, under gnotobiotic condition was assessed in cultivar ADT36 using a strain marked with transposon-based egfp and Km r . The endophytic colonization was monitored through re-isolation from different parts of rice seedlings in LB+Km plates. Addition of the flavonoids quercetin and diadzein to the growth medium increased the extent of endophytic colonization of the conjugant in rice seedlings by colonizing throughout the plant. Population and in planta nitrogenase activity of Serratia in rice seedlings were significantly increased by addition of flavonoids, quercetin and diadzein, whereas growth hormones, IAA and NAA reduced the efficiency of Serratia. The inoculation of Serratia sp. with flavonoids increased the plant biomass and biochemical constituents of rice seedlings under controlled condition.  相似文献

Transformed roots of Lupinus mutabilis cv. Potosi induced by Agrobacterium rhizogenes strain R1601 were cultured on Murashige and Skoog-based medium lacking kanamycin sulphate, or with this antibiotic at 40 mg l−1. The neomycin phosphotransferase gene in the genome of transformed roots was confirmed by non-radioactive Southern hybridisation. Neomycin phosphotransferase protein was detected by ELISA. Transformed roots synthesised isoflavones, but not quinolizidine alkaloids; the latter are typical secondary metabolites of lupin normally produced in aerial parts of the plant. Genistein and 2′-hydroxygenistein, were the main secondary metabolites in cultured, transformed roots, whereas the glycoside genistin was more abundant in roots of non-transformed plants. Wighteone concentrations in transgenic roots were higher than those of non-transformed roots. Transformed roots produced twice the concentration of isoflavones compared with roots from non-transformed plants, indicating that Ri plasmid T-DNA genes modified isoflavone concentration and pattern of biosynthesis.  相似文献

Forty-one isolates ofFusarium sambucinum sensu lato were screened for production of secondary metabolites in agar cultures. Of 16 strains ofF. sambucinum sensu stricto all but two strains produced diacetoxyscirpenol and two unidentified metabolites, TB1 and TB2 respectively. The two remainingF. sambucinum strains produced T-2 toxin, TB1 and TB2.Fusarium venenotum (6 strains) produced diacetoxyscirpenol and an unidentified metabolite BB.Fusarium torulosum (8 strains) produced wortmannin and antibiotic Y. The three species could be differentiated by their pattern of identified and unidentified metabolites detected by agar plug TLC combined with chemical data from HPLC-diode array detection of fungal extracts, and data on growth rates on potato sucrose agar and tannin sucrose agar.  相似文献
