Sporotrichosis is a fungal infection of subcutaneous or chronic evolution, inflammatory lesions characterized by their pyogranulomatous aspect, caused by the dimorphic fungus Sporothrix schenckii. Adenosine deaminase (ADA) is a "key" enzyme in the purine metabolism, promoting the deamination of adenosine, an important anti-inflammatory molecule. The increase in ADA activity has been demonstrated in several inflammatory conditions; however, there are no data in the literature associated with this fungal infection. The objective of this study was to evaluate the activity of serum ADA (S-ADA) and lymphocytes (L-ADA) of rats infected with S. schenckii. We used seventy-eight rats divided into two groups. In the first experiment, rats were infected subcutaneously and in the second experiment, infected intraperitoneally. Blood samples for hematologic evaluation and activities of S-ADA and L-ADA were performed at days 15, 30, and 40 post-infection (PI) to assess disease progression. Sporotrichosis, a disease caused by the saprophytic, dimorphic fungus Sporothrix schenckii, is currently diagnosed worldwide, especially in some tropical and subtropical areas. The infection usually occurs after traumatic inoculation of soil, plants, and organic matter containing the fungus. Certain activities, such as floriculture, agriculture, mining, and wood exploitation, and zoonotic transmission are associated with the mycosis. In humans, the disease is limited to skin, subcutaneous tissue, and the proximal lymphatic. It occurs commonly as lymphocutaneous or fixed lesions predominantly affect the upper limbs and face, the latter location is frequent in children. However, sporotrichosis in children is uncommonly seen. Data about the disease on this specific group of patients is scanty. The gold standard for diagnosis is culture. Nevertheless, there are other recently added methods (serological, histopathological, and molecular) useful for an accurate diagnosis. Itraconazole is the first choice of treatment for sporotrichosis; however, potassium iodide is also an effective option, mainly in children.  相似文献

Sporotrichosis is an emergent subcutaneous mycosis of humans and some animals caused by dimorphic fungi of the genus Sporothrix. The disease occurs worldwide but is endemic or hyperendemic in tropical and subtropical areas. The epidemiology of the disease is changing dramatically, and it is now considered an important zoonosis with high morbidity rates, principally in Brazil, and an opportunistic infection in immunocompromised patients. Due to the limited options currently available to treat invasive fungal infections, including sporotrichosis, and the emergence of drug resistance and toxicity, the development of anti-Sporothrix vaccines has become an area of great interest. This work provides a brief analysis of the feasibility of the development of prophylactic and therapeutic vaccines against sporotrichosis, the main advances achieved to date, future challenges and prospects.  相似文献

I trapped, marked, sexed and examined moult and softparts on 101 free-living Greater Kestrels Falco rupicoloides near Pretoria, South Africa, during 1975–1988.1 collected and plucked one kestrel to determine the number and mass of its feather tracts, as an index of materials invested in moult. I also examined moult on 72 museum specimens from southern Africa and observed plumage and softpart changes in four captive birds. During 1975–1979,1 made focal observations on free-living adult kestrels (males 181 h, females 262 h) and recorded monthly samples of their visual communication behaviours to the nearest minute.

Greater Kestrels can be aged for three successive plumage and softpart stages (juvenile, post-juvenile, adult). Plumage changes with potential signal effects involved only small investments at the post-juvenile and first adult moults (3.7% and 11% of total plumage mass respectively). Changes in colouration and communication behaviour correlated with relevant stages of the annual cycle, possibly to miminze conflict with territorial adults and age of first breeding. Remex and rectrix moult starts on average much later in relation to egglaying for Greater and some tropical kestrels than for some temperate species. This and other individual variation in timing may result for variations in nutritional status.  相似文献

Body measurements, plumage and moult of Sacred Ibis Threskiomas aethiopicus were studied at Pretoria from July 1973 to June 1974. Adult and immature Sacred Ibises are sexually dimorphic in size. Bill length alone can be used to sex most birds. Body mass, wing, tarsus and tail lengths overlap greatly between the sexes but males are generally larger than females. The sexes show similar patterns of variation in body mass and gonad size throughout the year. Juveniles follow a different pattern of variation in these parameters. The plumages of adults, immatures and juveniles are described and compared. There is no sexual dimorphism in plumage pattersn. Moult in adults occurs mainly in the post-breeding period from January to August, and in juveniles and immatures throughout the year. Adult Sacred Ibises have an-extensive, irregular and asymmetrical moult. Factors affecting sexual size dimorphism in African and Australian populations are discussed.  相似文献

The Pretoria Bone Collection began with the inception of the Department of Anatomy and the Medical School at the University of Pretoria in August 1942. Since then the collection has grown from a student aid to a resource for research. In the year 2000, the Pretoria Bone Collection was reorganised. The research material was divided into skulls, complete postcranial and incomplete postcranial remains. The collection presently contains 290 complete skeletons, 704 complete skulls and 541 complete postcranial remains. This paper presents information on the composition of this collection and hopes to heighten both national and international awareness of the collection and research opportunities in South Africa.  相似文献

The endemic mycoses traditionally include coccidioidomycosis, histoplasmosis, blastomycosis and paracoccidioidomycosis. Although sporotrichosis and chromomycosis are technically not included among the endemic mycoses, they are frequently diagnosed in Mexico. Most systemic endemic mycoses are a consequence of inhaling the fungi, while subcutaneous mycoses are acquired through the inoculation of vegetable matter or soil containing the organism. Coccidioidomycosis is caused by Coccidioides spp., a dimorphic pathogenic fungus. Approximately 60% of exposures result in asymptomatic infection; in the rest there are protean manifestations that range from a benign syndrome also known as "Valley Fever" to progressive pulmonary or extrapulmonary disease. Histoplasmosis, caused by the dimorphic fungus Histoplasma capsulatum, is endemic to the Americas. Pulmonary histoplasmosis manifestations are protean, ranging from a brief period of malaise to a severe, prolonged illness. The spectrum of illness in disseminated histoplasmosis ranges from a chronic, intermittent course to an acute and rapidly fatal infection. Paracoccidioidomycosis is a chronic, granulomatous systemic disease caused by Paracoccidioides brasiliensis that characteristically produces a primary pulmonary infection, often asymptomatic, and then disseminates to form ulcerative granulomata of the oral, nasal and occasionally the gastrointestinal mucosa. Sporotrichosis, caused by Sporothrix schenckii, has diverse clinical manifestations; the most frequent is the lymphocutaneous form. Generally, infection results from inoculation of the fungus through thorns, splinters, scratches and small traumas. Chromomycosis (Chromoblastomycosis) is a slowly progressive cutaneous and subcutaneous mycosis attributed to various saprophyte Hypomycetes fungi. The primary lesion is also thought to develop as a result of percutaneous traumatic inoculation.  相似文献

Approximately 5% of the Republic of South Africa is set aside for national parks, nature reserves, private game reserves or wilderness areas. Within these areas a large variety of animals and plant species is protected. Many of these species are indigenous to the Southern African subregion. Most of the zoological gardens in South Africa are owned privately or by a municipality. The zoo in Pretoria is the only one which has national status. In addition to facilities in Pretoria, the National Zoological Gardens owns two game breeding centres covering an area of more than 7000 ha. The game breeding centres act as important gene pool reservoirs and are used for the breeding of threatened species. They thus fulfill an important role in conserving threatened species and, in some cases, supplementing dwindling populations in protected areas.  相似文献

BackgroundCutaneous mucormycosis (zygomycosis), with subcutaneous spreading and dissemination, in immunocompetent patients is an uncommon disease caused by species belonging to the fungal genera Apophysomyces, Rhizopus and Saksenaea, among others.Case reportA case of necrotising fasciitis by Saksenaea vasiformis in an immunocompetent woman is described. The infection was acquired through a car accident resulting in multiple injuries affecting mainly her right arm. After the surgical reduction of fractures, skin lesions worsened and led to necrosis. The patient quickly developed a severe necrotising fasciitis with negative cultures at first. Despite the extensive surgical debridement and the aggressive antifungal treatment, the patient died. The histopathological study showed a fungal infection due to a fungus belonging to the Mucorales order, which was confirmed by culturing the clinical sample on Sabouraud agar, and identifying the species by cultures on Czapek-Dox agar, and sequencing of the ITS region of the ribosomal DNA.ConclusionsThis case confirm the presence of this fungus in Spain, the value of histopathology for the mucormycosis diagnosis, as well as the need to perform special cultures to facilitate their isolation and identification to the species level by the combined use of Czapek-Dox agar and sequencing of the ITS region.  相似文献

Secretarybirds in three adjacent territories were monitored from 1977 to 1988 on grass- and croplands near Pretoria, South Africa. Most observations of breeding biology and behaviour confirmed or extended previous studies. There was no correlation between pairs in occupancy of territory, productivity or development periods of young: this confirms the flexible breeding abilities which are unusual for such a large bird. Some aspects of breeding biology (egg shape and texture, watering of chicks) and behaviour (Wings open and Up-down greeting displays) may be homologous with storks and important in understanding the phylogeny and evolution of the Sagitariidae and other diurnal raptors.  相似文献

Sporotrichosis, caused by several species from the genus Sporothrix, mainly affects men working in agricultural labor. It is reported mostly from endemic regions located in tropical and subtropical areas. Sporotrichosis usually affects subcutaneous tissue with no symptoms or mild symptoms, but it causes disseminated and even fatal disease if the patient is immunocompromised. Significant progress in the knowledge of etiologic agents of sporotrichosis has been achieved recently, but evaluations of treatment using well-designed clinical trials have been neglected. This article reviews the drugs currently used to treat subcutaneous disease, describing the differences in their efficacy, and reviews recent findings about the proposed new Sporothrix species of clinical interest, as well as the role of melanin as a virulence factor in Sporothrix schenckii.  相似文献

Sporotrichosis is a subcutaneous mycosis, which affects mainly small animals, and is considered an important public health disease. This paper describes the epidemiological and laboratory characteristics of 103 clinical cases of sporotrichosis diagnosed over a 10-year period in southern Brazil. The 92 cats and 11 dogs from eight municipalities in Rio Grande do Sul State developed especially the disseminated cutaneous and fixed cutaneous forms of the disease. Respiratory signs such as sneezing, serous nasal discharge and dyspnea were found in about 57% of the animals. The detection of Sporothrix schenckii in different clinical samples showed highest isolation in testicles (46.6%), oral cavity (45.2%) and conjunctival mucosa (38.1%). A differentiated histological pattern was found between the fixed cutaneous and disseminated cutaneous (DC) manifestations of the disease; well-organized granulomas of nodular distribution and various fungal structures prevailed in the DC form in cats. Melanin detection in S. schenckii cells by the Fontana–Masson technique was positive in 45.4% of the samples. The study revealed that the State of Rio Grande do Sul is an endemic sporotrichosis area and demonstrated the possibility of involvement of other pathways in the infection and spread of the disease. In addition, it emphasized the importance of laboratory tests for mycosis confirmation, especially in dogs that develop clinical manifestations without the presence of cutaneous lesions.  相似文献

Sporotrichosis is a chronic granulomatous mycosis caused by the dimorphic fungus Sporothrix schenckii, which is widely distributed in nature, and presents a saprophytic mycelial form on plant debris and soil. The immunological mechanisms involved in the prevention and control of sporotrichosis are not yet fully understood. In this study, mice were studied after infection with Sporothrix schenckii. In the first week after infection, fungal loading increased and thence decreased drastically 14 days after infection. Analysis by immunoblotting showed that the sera of all mice tested had antibodies reacting only with a 70 kDa antigen, with predominance of IgG1 and IgG3. Taken together, our results show that antigens from S. schenckii induced a specific humoral response in infected mice.  相似文献

SUMMARY. Fungal infection of calanoid copepod ( Diaptomus novamexicanus ) eggs was observed in each of three years in an alpine lake (Castle Lake, California, U.S.A.). Stages in the infection process were examined by light and scanning electron microscopy and evidence was obtained that the Lagenidium -like fungus concerned was a virulent parasite. Fungal destruction of eggs varied in timing and severity from year to year. The maximum impact of the disease was an estimated 48.4% decrease in potential copepod recruitment in 1976 due to the onset of a severe epidemic early in the summer growing season. The minimum impact, a 5.6% decrease in potential recruitment, was recorded in 1975. In this year the proportion of infected eggs was reduced and large numbers of juveniles had been released before the fungal disease began. The 1974 epidemic was intermediate in severity. The effect of these epidemics on Castle Lake calanoid populations is discussed in relation to temperature, food availability and predation.  相似文献

It is well established that resistance or susceptibility to Paracoccidioidis brasiliensis infection in mice is under strict host's genetic control. Mice from A/Sn strain inoculated by the ip route are resistant to fungal infection while infection induced in mice from B10.A strain results in a fatal disease. The early cellular events of infection in both strains are characterized by a marked neutrophilic infiltration that is more prominent in B10.A mice. A peculiar characteristic of the Paracoccidioides brasiliensis-mouse model is that the subcutaneous (sc) inoculations of the fungus either in resistant (A/Sn) or susceptible (B10.A) mice is self-curing and turns mice from the B10.A strain able to express typical DTH reaction to fungal antigens, as observed in A/Sn mice. Here we report the investigation on the early events of the inflammatory response induced by the inoculation of live fungus into the hind footpad of A/Sn (resistant) and B10.A (susceptible) mice. The influence of neutrophils on the inflammatory response and antibody titers or DTH response to gp43, the major fungal antigen, was also evaluated. Results showed a different course of the inflammatory response induced by fungal inoculation in A/Sn and B10.A mice. Neutrophil depletion before infection differently influenced the kinetics of the inflammatory process in both mice strains but did not modifythe fungal load in the lesions. In neutrophil depleted mice from both strains, a decrease in DTH response and an increase in total antibody titers to gp43 were observed. The significant increase in the fungal load in lesions seen in nude mice indicates that the self-limited infection evoked by fungal inoculation into the subcutaneous tissue is a T-cell dependent phenomenon. The implications of these observations in the pathogenesis of paracoccidioidomycosis are discussed.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

Laying dates for a population of individually marked Greater Kestrels Falco rupicoloides, on grass- and croplands near Pretoria, South Africa, were determined for 89 nesting attempts during 1975–1988. Laying occurred in the austral spring between 23 July and 11 November. Timing of laying was negatively correlated with rainfall of the previous summer and positively correlated with rainfall of the winter and spring preceding laying. Success in fledging young was significantly higher for clutches laid before the median laying date of 1 September and for clutches laid after seasons of above average rainfall.  相似文献

Two new species of Tachygonetria, T. africana n. sp. and T. pretoriensis n. sp. are described. Both are parasites of Geochelone pardalis (Bell) from South Africa. In addition, two subspecies, T. longicollis fitzsimmonsi Petter, 1966 and T. macrolaimus macrolaimus (Linstow, 1899), also parasites of Geochelone pardalis (from Pretoria Zoo, South Africa), are redescribed and new information on their morphology is presented. Thielaviopsis basicola is a soil-borne fungal pathogen with a wide host range and a cosmopolitan distribution. It causes disease on many agricultural crops, and in South Africa is the causal agent of black pod rot of groundnuts and black root rot on chicory. Knowledge of the population diversity of T. basicola could provide valuable information regarding management strategies, the possible movement, origin, and reproductive strategies of the fungus. The objective of this study was to determine the population diversity of T. basicola isolates from groundnuts and chicory in South Africa using co-dominant polymorphic markers. These markers were also used to compare isolates from South Africa with those from other hosts and geographic regions. Seven loci revealed nine alleles and two genotypes, one on groundnut and one on chicory, differing at only two loci. T. basicola isolates from eight different countries and ten different hosts revealed 17 genotypes across the seven loci with 39 different alleles. The lack of diversity for the two South African host-related populations of isolates suggests that T. basicola was introduced into South Africa. Some evidence is provided for a European origin of the pathogen, possibly linked to trade in root crops.  相似文献
