Micropropagated Choisya, Daphne, Delphinium, Hemerocallis, Hosta, Iris and Photinia were found to adjust the pH of Murashige and Skoog's plant tissue culture medium (initial pH 5.6 or 3.5) to different values depending on the species. The effect of changes in pH of the medium from 4 to 10 on the action potential and its first derivative was studied at the original resting potential and during hyperpolarization of the membrane in experiments on single nodes of Ranvier. Raising the pH of the medium from 7 to 9 led to a decrease in amplitude of the action potential and of its derivative (Vmax). During hyperpolarization of the membrane these parameters were fully restored. Lowering the pH of the solution led to an increase in the action potential and a decrease in Vmax. During hyperpolarization of the membrane the action potential and its derivative were not completely restored. Under the influence of solutions with low and high pH values the duration of the action potential was increased. Changes in the action potential and in Vmax with an increase in pH can be attributed to increased inactivation of the sodium permeability of the membrane, and in solutions with low pH to a decrease in the maximal sodium permeability and to weakening of its inactivation.A. The responses of a chemostat Saccharomyces cerevisiae culture (D = 0.1 h-1) to a stepwise increase or decrease in the pH of the medium were shown to be asymmetric. When the pH was lowered from 6.5 to a value above 0.3, the rates of oxygen uptake and carbon dioxide evolution rose for a sort period of time whereas the optical density of the culture fell down. The detected changes in the properties of the culture were identical with those which had been observed in the course of spontaneous undamped oscillations in the physiological parameters of the continuous C. cerevisiae culture. Apparently, in both cases, the energy status of cells changed when the oxidative type of metabolism was substituted by fermentation. When the pH of the medium was elevated within the same range (4.7-6.5), the response of the culture was three times as low and its properties changed in the opposite direction. When the pH of the medium was changed in a cyclic sinusoidal manner, oscillations in the physiological characteristics of the culture, identical with spontaneous oscillations were induced at certain values of the amplitude and the frequency of pH changes.  相似文献

The effects of various organic solvents on penicillin acylase-catalyzed synthesis of β-lactam antibiotics (pivampicillin and ampicillin) have been investigated in water-solvent mixtures. The rates of penicillin acylase-catalyzed reactions were found to be significantly reduced by the presence of a small amount of organic solvent. In particular, the rate of enzyme catalysis was extremely low in the presence of ring-structured solvents and acids while enzyme activities were fully restored after removing the solvents. This indicates that interactions between the solvents and the enzyme are specific and reversible. To correlate the inhibitory effects of organic solvents with solvent properties the influence of solvent hydrophobicities and solvent activity on the rate of pivampicillin synthesis was examined. The reaction rate was found to decrease with increasing solvent hydrophobicities, and a better correlation was observed between the reaction rate and solvent activity. The effects of ionic strength on the synthesis of pivampicillin and ampicillin were also examined. The ionic strength dependence indicates that electrostatic interactions are involved in the binding of ionic compounds to the enzyme. On the basis of the active site structure of penicillin acylase, a possible mechanism for molecular interactions between the enzyme and organic solvents is suggested.  相似文献

在黑暗条件下,利用不同形态的氮源(硝酸盐氮,氨氮,有机氮和硝酸盐氮,有机氮)培养蓝藻水华优势种铜绿微囊藻(Microcystis aeruginosa),分析其氮代谢和对水体pH的影响.研究结果表明,在不同氮源的培养液中铜绿微囊藻密度在最初的24 h内出现波动,之后下降.培养液中pH值在试验最初的24 h显著下降,之后趋于稳定,在硝态氮培养液中pH值下降最大,从8.18下降到7.19,其反硝化作用产生的NO-2浓度也最大.不同氮源培养液中总氮含量都有所下降,以混合氮源培养液中总氮减少量最大,说明化合态氮经过反硝化作用生成了氮气并溢出培养液,因此,在黑夜条件下藻华水体中存在反硝化作用.  相似文献

Changes in the pH level of the fermentation medium used for preliminary cultivation of C. tropicalis were studied with respect to its initial aciditv or alkalinitv. When C. tropicalis was grown on the medium used for levorin fermentation with ph 5.1--10.3, the yeast changed it in 24 hours to the level of 6.2--7.9. As dependent on the initial pH values for cultivation of C. trophicalis, production of levorin on subsequent inoculation of Act. levoris changed. The antibiotic activity increased and ranged within 120--178% of the control. Synthesis of levoristatin, a non-polyenic antibiotic equally increased under such conditions and ranged within 153--163% of the control. The pH values of 9.4--10.3 of the initial fermentation medium were optimal for mixed cultivation of Act levoris and C. tropicalis and maxium production of levorin and levoristatin.  相似文献

The hydrodynamical and optical properties of DNA were investigated in the wide-range of pH by the methods of streaming birefringence, viscometry and spectrophotometry for the different ionic strengths of environment. The measurements of the intrinsic viscosity as a function of pH allow us to determine the compactization of protonated DNA without the destruction of double-helical conformation. This transition is accompanied by a decrease in the optical anisotropy of DNA and the coefficient of molar extinction E260 (P). The increase of volume and persistence length of DNA was observed in the alkaline range of pH. Analyses of experimental data lead to an assumption that the predominant cause of these effects is the change of flexibility of DNA as a result of ionization of its bases. The data obtained were compared with those for polycationic molecules.  相似文献

Lyophilized chloroplasts of Pisum sativum (pea) have been extracted with petroleum ether of different polarity (obtained by adding varying amounts of ethanol to the petroleum ether). Extracted thylakoids have then been solubilized by sodium dodecyl sulphate (SDS) and chlorophyll-protein complexes have been isolated by polyacrylamide gel electrophoresis (PAGE). Absorption- and low temperature fluorescence emission spectro-scopy have been used to characterize thylakoids and purified chlorophyll-protein complexes. Weakly polar solvents extracted mainly chlorophyll a. SDS-PAGE scan profiles of similarly extracted thylakoids contained no photosystem II chlorophyll a reaction center antennae (CP-an) and the amount of photosystem I chlorophyll a reaction center antennae (CP-a1) was reduced as compared with an unextracted control. This was due partly to the extraction of chlorophyll a prior to SDS-PAGE, and partly to the increased solubilization of chlorophyll a by SDS as a result of β-carotene extraction. By increasing the polarity of the solvent CP-a1 also disappeared in the scan profile, leaving only the light-harvesting chlorophyll a/b-protein complex (CP-a/b) and SDS complexed chlorophyll. From these results we conclude that the chlorophyll molecules in the reaction center antennae are relatively more hydrophobically associated than the molecules in the light-harvesting CP-a/b complex. The chlorophyll a of CP-au and the far red absorbing chlorophyll a fraction of CP-a1 appear to be the most hydrophobically associated chlorophyll molecules.  相似文献

The features of the interaction in a system biocomposite (peat)-solvent (H2O, 0.1 M NaOH, C2H5OH) have been considered. Protein glycation is believed to play an important role in the development of long-term disorders associated with diabetes. Previous studies have shown that copper could activate this process; however, these experiments were performed under non-physiological conditions. In this study, in vitro experiments were carried out at near-physiological conditions to examine the catalytic activity of copper on the interaction of albumin with glucose. Changes in pH and phosphate buffering capacity were shown to affect albumin glycation. Under stable pH conditions, copper activates albumin glycation only at low protein concentrations (<30 gl–1). Copper had no effect on albumin glycation at higher protein concentrations probably because the metal is chelated by the protein.  相似文献
