A System for Measuring Effects of Sulphur Dioxide on Gas Exchange of Plants   总被引:3,自引:0,他引:3  
Apparatus is described for exposing plants to low concentrationsof SO2 (50–500µg m–3 in air) and for measuringeffects on photosynthesis, dark respiration, and transpiration.Temperature, humidity, and irradiance in the chambers were controlledindependently, and fans ensured that leaf boundary layer resistanceswere low. Experiments with plants of Vicia faba in clean andpolluted air showed that: (i) a depression of net photosynthesisby 50 µg m–3 SO2 depended on boundary layer resistanceand on irradiance; (ii) stomatal resistance was increased ordecreased by 50 µg m–3 SO2 when relative humidityin the chambers was low (35% r.h., 22 °C) or high (50% r.h.,22 °C) respectively.  相似文献

Net photosynthesis rates (PN) of Viciafaba plants were measuredin chambers containing either clean air or air containing between50 and 300 parts 10–9 ozone (O3) with or without 40 parts10– 9 SO2. Exposure to O3 concentrations greater than 50 parts 10–9 for periods of 4 h resulted in reductions in PN with greaterreductions at higher concentrations. After exposure ceased,recovery of pre-exposure PN depended on O3 concentration. Exposureto less than 90. 100 parts 10– 9 was followed by completerecovery after 20 h whereas 200-300 parts 10– 9 resultedin visible injury and irreversible depression of PN. The additionof 40 parts 10– 9 SO2 to O3 significantly decreased Psbut the effect of SO2 declined with increasing O3. The additionof SO2 had no significant affect on recovery patterns. It is postulated that Vicia faba plants are able to toleratethe absorption of O, up to a threshold above which progressivedecreases in PN occur due to effects on photosynthetic processes.The slow and sometimes incompletely reversible effects of O3on PN indicate cellular differences in the injury mechanismand repair capacity compared with the action of SO2 alone. Lowconcentrations of O3 with SO2 result in additive injury to PNsuggesting independent action of the pollutants, but the reductionin SO2 action with increasing O3 indicates a limit for potentialphysiological injury More the onset of visible injury.  相似文献

The effects of exposure to SO2 (50 and 400 µg m–3SO2) on the growth, photosynthesis, and respiration of perennialryegrass (Lolium perenne L.) cv. S23 were examined in two successivegrowth periods of 29 and 22 d. At the higher concentration ofSO2, there was some visible injury of the leaves and specificleaf area was reduced, but yield, net photosynthesis, and darkrespiration of the plants were not significantly affected byexposure. The treatment was also without effect on the transpirationcoefficient of the plants and their number of tillers. The plantsexposed to the lower concentration of SO2 showed no signs ofinjury and did not differ in any of the measured characteristicsfrom plants grown in SO2-free air. Content of S in the shootsincreased linearly with the concentration of SO2, the additionalS being found in the sulphate fraction whilst organic S wasunchanged. The results are discussed in relation to earlierfindings that yield of ryegrass exposed to SO2 may be reducedwithout visible signs of injury.  相似文献

Agrostis capillaris L.5, Festuca vivipara L. and Poaalpina L.were grown in outdoor open-top chambers at either ambient (340 3µmol mol–1) or elevated (6804µmol mol–1)concentrations of atmospheric carbon dioxide (CO2) for periodsfrom 79–189 d. Photosynthetic capacity of source leaves of plants grown atboth ambient and elevated CO2 concentrations was measured atsaturating light and 5% CO2. Dark respiration of leaves wasmeasured using a liquid phase oxygen electrode with the buffersolution in equilibrium with air (21% O2, 0.034% CO2). Photo-syntheticcapacity of P. alpina was reduced by growth at 680 µmolmol–1 CO2 by 105 d, and that of F. vivipara was reducedat 65 d and 189 d after CO2 enrichment began, suggesting down-regulationor acclimation. Dark respiration of successive leaf blades ofall three species was unaltered by growth at 680 relative to340 µmol mol–1 CO2. In F. vivipara, leaf respirationrate was markedly lower at 189 d than at either 0 d or 65 d,irrespective of growth CO2 concentration. There was a significantlylower total non-structural carbohydrate (TNC) concentrationin the leaf blades and leaf sheaths of A. capillaris grown at680µmol mol–1 CO2. TNC of roots of A. capillariswas unaltered by CO2 treatment. TNC concentration was increasedin both leaves and sheaths of P. alpina and F. vivipara after105 d and 65 d growth, respectively. A 4-fold increase in thewater-soluble fraction (fructan) in P. alpina and in all carbohydratefractions in F. vivipara accounted for the increased TNC content. In F. vivipara the relationship between leaf photosyn-theticcapacity and leaf carbohydrate concentration was such that therewas a strong positive correlation between photosynthetic capacityand total leaf N concentration (expressed on a per unit structuraldry weight basis), and total nitrogen concentration of successivemature leaves reduced with time. Multiple regression of leafphotosynthetic capacity upon leaf nitrogen and carbohydrateconcentrations further confirmed that leaf photosynthetic capacitywas mainly determined by leaf N concentration. In P. alpina,leaf photosynthetic capacity was mainly determined by leaf CHOconcentration. Thus there is evidence for down-regulation ofphotosynthetic capacity in P. alpina resulting from increasedcarbohydrate accumulation in source leaves. Leaf dark respiration and total N concentration were positivelycorrelated in P. alpina and F. vivipara. Leaf dark respirationand soluble carbohydrate concentration of source leaves werepositively correlated in A. capillaris. Changes in source leafphotosynthetic capacity and carbohydrate concentration of plantsgrown at ambient or elevated CO2 are discussed in relation toplant growth, nutrient relations and availability of sinks forcarbon. Measuring the Canopy Net Photosynthesis of Glasshouse Crops   总被引:3,自引:0,他引:3  
A null balance method is described for measuring net photosynthesisof mature canopies of cucumber and other protected crops overperiods of 10 min in a single-span glasshouse (c. 9m x 18m inarea). Accuracy of control of the CO2 concentration in the greenhouseatmosphere is within ±10 vpm of the normal ambient level(c. 350 vpm). The amounts of CO2 used in canopy net photosynthesisare measured with linear mass flowmeters accurate to within±0.80g. The total errors incurred in measuring canopynet photosynthesis at an ambient CO2 level are estimated tobe of the order of ± 1·2% in bright light (350W m–2, PAR)and ±3·6% in dull light (100W m–2, PAR). Measurements of the rates of net photosynthesis of a maturecanopy of a cucumber crop were made at near-ambient CO2 concentrationsover a range (0–350 W m–2) of natural light fluxdensities. A model of light absorption and photosynthesis applicableto row crops was used to obtain a net photosynthesis versuslight response curve for the cucumber crop. At a light fluxdensity of 350 W m–2 the fitted value of canopy net photosynthesiswas 2.65 mg CO2 m–2s–1 (equivalent to over 95 kgCO2 ha–1h–1). The results are discussed in relationto the need for CO2 supplements to avoid depletion in both ventilatedand unventilated glasshouses during late spring and summer. The effects of exposure of up to 2 h with sulphur dioxide ona range of plant species was observed by measuring changes inthe rate of net photosynthesis under closely controlled environmentalconditions. Ryegrass, Lolium perenne ‘S23’ was thespecies most sensitive to SO2; significant inhibition was detectedat 200 nl l–1. Fumigations at 300 nl l–1 also inhibitedphotosynthesis in field bean (Vicia faba cv. ‘Three FoldWhite’ and ‘Blaze’) and in barley (Hordeumvulgare cv. ‘Sonja’). No effect was detected inwheat (Triticum aestivum cv. ‘Virtue’) at concentrationsup to 600 nl l–1 SO2, or in oil-seed rape (Brassica napuscv. ‘Rafal’) except at 800 nl l–1 SO2). Recoverycommenced immediately after the fumigation was terminated andwas complete within 2 h when inhibition had not exceeded 20%during the SO2 treatment. The mechanism of SO2-induced changes in stomatal conductance(g) of alder was examined to determine if SO2 affects guardcell function directly or indirectly through the SO2-inducedchanges in photosynthesis. During experimental fumigations at SO2 concentrations of 3–3µmol m–3 (0.08 µl l–1), stomatal closurepreceded declines in net photosynthetic rate (A), indicatingthat SO2 can directly affect guard cells. From these and otherstudies it appears that the sequence of A and g responses maybe influenced by SO2 concentration as well as by species. Fumigation with SO2 did not cause increases in g, even whenthe intercellular substomatal CO2 concentration (ci) was reducedby 50 µmol mol–1. In two experiments, the functioning and metabolism of nodulesof white clover, following a defoliation which removed abouthalf the shoot tissue, were compared with those of undefoliatedplants. In one experiment, the specific respiration rates of nodulesfrom undefoliated plants varied between 1160 and 1830 µmolCO2 g–1h–1, of which nodule ‘growth and maintenance’accounted for 22 ± 2 per cent, or 27 ± 3.6 percent, according to method of calculation. Defoliation reducedspecific nodule respiration and nodule ‘growth and maintenance’respiration by 60–70 per cent, and rate of N2 fixationby a similar proportion. The original rate of nodule metabolismwas re-established after about 5 d of regrowth; during regrowthnodule respiration was quantitatively related to rate of N2,fixation: 9.1 µmol CO2 µmol–1N2. With the possible exception of nodules examined 24 h after defoliation,the efficiency of energy utilization in nitrogenase functioningin both experiments was the same in defoliated and undefoliatedplants: 2.0±0.1 µmol CO2 µmol–1 C2H4;similarly, there was no change in the efficiency of nitrogenasefunctioning as rate of N2 fixation increased with plant growthfrom 1 to 22 µmol N2 per plant h–1. Exposure of nodulated white clover root systems to a 10 percent acetylene gas mixture resulted in a sharp peak in rateof ethylene production after 1.5–2.5 min; subsequently,rate of ethylene production declined rapidly before stabilisingafter 0.5–1 h at a rate about 50 per cent of that initiallyobserved. Regression of ‘peak’ rate of ethyleneproduction on rate of N2 fixation indicated a value of 2.9 µmolC2H4 µmol–1 N2, for rates of N2 fixation between1 and 22 µmol N2 per plant h–1. The relationshipsbetween nitrogenase respiration, acetylene reduction rates andN2 fixation rates are discussed. Permeability coefficients (PS values) for CO2 of the plasmamembrane (PM) of the unicellular green algae Eremosphaera viridis,Dunaliella parva, and Dunaliella acidophila, and of mesophyllprotoplasts isolated from Valerianella locusta were determinedfrom 14CO2 uptake experiments using the rapid separation ofcells by the silicone oil layer centrifugation technique. Theexperimental PS values were compared with calculated numbersobtained by interpolation of Collander plots, which are basedon lipid solubility and molecular size, for D. parva cells,mesophyll protoplasts isolated from Spinacia oleracea, mesophyllcells and guard cells of Valerianella, and guard cell protoplastsisolated from Vicia faba. The conductivity of algal plasma membranes for CO2 varies between0.1 and 9 ? 10–6 m s–1, whereas for the plasmalemmaof cells and protoplasts isolated from leaves of higher plantsvalues between 0.3 and 11 ? 10–6 m s–1 were measured.By assuming that these measurements are representative for plantsand algae in general, it is concluded that the CO2 conductivityof algal PM is of the same order of magnitude as that of thehigher plant cell PM. Ps values of plasma membranes for CO2are lower than those for SO2, but are in the same order of magnitudeas those measured for H2O. On the basis of these results itis concluded that theoretical values of about 3000 ? 10–6m s–1 believed to be representative for higher plant cells(Nobel, 1983) and which are frequently used for computer-basedmodels of photosynthesis, lack experimental confirmation andrepresent considerable overestimations. However, with severalsystems, including higher plant cells, the conductance of thePM for CO2 was significantly higher in light than in darkness.This suggests that in light, additional mechanisms for CO2 uptakesuch as facilitated diffusion or active uptake may operate inparallel with diffusional uptake. The CO2 dynamics of the lacunal air and the relative contributionof external and internal CO2 sources to dark CO2 assimilationwas examined in the submerged aquatic CAM species Littorellauniflora (L.) Aschers. Refixation of internal CO2, released by dark respiration, constitutedabout 30–35% of the total dark CO2 assimilation. At aCO2 concentration of 0·2 mol m–3 around the leavesthe external CO2 uptake through the roots increased from 45%of the total CO2 uptake at 0·7 mol m–3 CO2 to 100%at 1·6 mol m–3 and 3·1 mol m–3 CO2around the roots. The negligible importance of leaf CO2 uptakeat high CO2 concentrations around the roots was the result ofa causative high CO2 concentration in the leaf lacunae. The CO2 permeability of Littorella leaves was high relativeto root permeability. This has at least two ecological implications:(1) it enhances the potential diffusive release of CO2 fromthe sediment C02-pool via the lacunal system of the plants.This loss of CO2, however, was found to be greatly reduced byCAM activity of the plants. (2) The high permeability of theleaf surface to CO2 exchange allows the plants to assimilateCO2 from the water surrounding the leaves when the concentrationis high, i.e. during extensive epiphyte dark respiration. Thus,CAM tends to facilitate retension of a high CO2 pool in thesediment-plant system and at the same time allows the plantsto exploit the water column CO2 source when it is abundant.This result is in accordance with the general idea that CAMin aquatics constitute a carbon conserving mechanism. Stomatal Responses to Sulphur Dioxide and Vapour Pressure Deficit   总被引:5,自引:0,他引:5  
Stomatal conductances (gs) of plants of Vicia faba, Raphanussativus, Phaseolus vulgaris, Heilanthus annuus, and Nicotianatabacum were measured in chambers containing either clean airor air containing between 18 and 1000 parts 10–9 SO2 atwater vapour pressure deficits (vpd) ranging from 1·0to 1·8 kPa. When vpd was low (<1·3 kPa at 22 °C) and stomatawere open, exposure to SO2 induced rapid and irreversible increasesin gs in V. faba. This response persisted throughout the exposure(3 d). The increase in gs, 20–30% compared with cleanair, was independent of SO2 concentration up to 350 parts 10–9Stomatal conductances of polluted plants at night were greaterthan controls. When stomata were closed before exposure to SO2,there was no effect on gs. When vpd was varied, gs of unpolluted plants of P. vulgarisshowed no response, but that of R. sativus increased slightlywith increasing vpd. In both species exposure to SO2 causedan increase in gs at all vpd values. gs of unpolluted plantsof V. faba, H. annuus, and N. tabacum decreased with increasingvpd. At low vpd values exposure to SO2 in these species causedan increase in gs, but, above a certain value of vpd, dependingon species, gs decreased with exposure to SO2. It is postulated that SO2, once in the substomatal cavity, entersthe stomatal complex via adjacent epidermal cells and at lowconcentrations leads to a reduction in turgor in these cellsand consequently to stomatal opening. In vpd-sensitive species,increased transpiration from guard cells or epidermal cellsadjacent to the stomata induced by SO2 may lead to stomatalclosure at large vpd levels. Stomatal sensitivity to vpd insuch cases may be enhanced because adjacent epidermal cell turgoris lowered by SO2. At high SO2 concentrations direct disruptionof guard cell structure may lead to a loss of turgor and stomatalclosure.  相似文献

Non-selected and Na2SO-, K2SO4- or KCl-selected callus culturesof Vaccinium corymbosum L. cv. Blue Crop were grown on mediasupplemented with 0, 25 and 50 mM Na2SO4 (non-selected and Na2SO(-selectedonly), 0, 25 and 50mMK2SO4 (non-selected and K2SO4-selectedonly) or 0, 50 and 100 mM KCl (non-selected and KCl-selectedonly). On all media, growth of selected callus (on a fresh-weightor dry-weight basis) was greater than that of non-selected callus,and selected callus grew optimally on the level and type ofsalt on which it was selected. Selected callus was friable andmaintained a higher f. wt:d. wt ratio. Tissue water potentialin selected callus was more negative than in non-selected callus. Flame photometry and chloridometry showed Na+, K+ and Claccumulated in callus to concentrations equal to or greaterthan the initial concentration in the medium. Turbidometry showedthat tissue SO42- concentration was lower than the concentrationin the medium. In most cases selected callus accumulated moreNa+, Ksup, SO42– or Cl than non-selected callus.Vacuolar ion concentration was measured by electronprobe X-raymicroanalysis, and on most media selected callus had highervacuolar ion concentrations than non-selected callus. SO42–and Cl were accumulated in the vacuoles at concentrationshigher than the external medium, but vacuolar Na+ concentrationdid not reach external concentration on Na2SO4 and on potassiumsalts was maintained between 12 and 17 mM. Vacuolar K+ concentration(approx. 142–191 mM on no salt) decreased on Na2SO4 andincreased on K2SO4 and KCl. There was no precise correlation between total or specific ionaccumulation (Na+, K+, SO42– and Cl and fresh-weightyield. Results suggest that selection results in adaptationin response to decreased water potential of the medium. Young plants of uniculm barley were grown singly in pots ina growth room at 23/21 °C, and an irradiance of 655 µEm–2s–1 during each 12 h photoperiod. At the fifth leaf stage,they were subjected to 80 h of continuous darkness during whichthe rates of CO2 efflux of vegetative shoot meristems, and maturefully expanded leaves, were separately monitored. Respiratoryefflux from the meristematic tissue was initially high, 12–15mg CO2 g–1 h–1, equivalent to a daily loss in weightof 20–25 per cent. It remained high, or even rose slightly,during what would have been the normal dark period, but thenfell sharply. Even so, it was still three times that of themature tissue at the end of the experimental period. The rateof CO2 efflux of the mature tissue began low, and fell evenfurther during the first 12 h of darkness. It then levelledoff at a rate of 2·0–2·5 mg CO2 g–1h–1, equivalent to a daily loss in weight of about 3 percent. It is suggested that the rate of ‘mature tissue’respiration, established after 12–24 h of darkness, mightbe a useful selection criterion to employ in attempts to increasethe total dry matter yield of the grass crop by breeding. The rate of net photosynthesis (P) of whole plant stands oftomato (Lycopersicon esculentum Mill.), cucumber (Cucumis sativusL.) and sweet pepper (Capsicum annuum L.) was measured in sixlong-term experiments in large greenhouses under normal operatingconditions and CO2-concentrations between 200 and 1200 µmolmol-1. The objective was to quantify the responses to lightand carbon dioxide and to obtain data sets for testing simulationmodels. The method of measuring canopy photosynthesis involvedan accurate estimation of the greenhouse CO2 balance, usingnitrous oxide (N2O) as tracer gas to determine, on-line, theexchange rate between greenhouse and outside air. The estimatedrelative error in the observed P was about ± 10%, exceptthat higher relative errors could occur under particular conditions. A regression equation relating P to the photosynthetically activeradiation, the CO2 concentration and the leaf area index explained83-91% of the variance. The main canopy photosynthesis characteristicscalculated with the fitted regression equations were: canopyPmax 5-9 g m-2 h-1 CO2 uptake; ratio Pmax/LAI 1·5-3 gm-2 h-1; light compensation point 32-86 µmol s-1 m-2;light use efficiency (quantum yield) at low light 0·06-0·10µmol µmol-1 and CO2 compensation point 18-54 µmolmol-1. The respiratory effluxes of nodules and of roots of FiskebyV soyabean (Glycine max (L.) Merr.), grown in a controlled environment,were measured at intervals in air and 3% O2 from shortly afterthe onset of N2 fixation until plant senescence. The respiratoryburdens linked with nitrogenase plus ammonia metabolism, andnodule growth and maintenance, were calculated from gas exchangedata and related to the concurrent rates of N2 fixation. The specific respiration rates of nodules increased to a maximumof 21 mg CO2 g–1 h–1 at the time pods began development:the equivalent maximum for roots was c. 4.5 mg CO2 g–1h–1. Maximum nodule and root respiration rates per plantwere attained about 25 d later at the time N2 fixation peakedat 15 mg N d–1 plant–1. The relationship between nodule respiration and N2 fixationindicated an average respiratory cost of 13.2 mg CO2 mg–1N until the last few days of plant development Separation ofnodule respiration into the two components: nitrogenase (+ NH3metabolism) respiration and nodule growth and maintenance respiration,indicated that the latter efflux accounted for c. 20% of nodulerespiration while N2 fixation was increasing and new noduletissue was being formed. When nodule growth ceased and N2 fixationdeclined, this component of respiration also declined. The respiratorycost of nitrogenase activity plus the associated metabolismof NH3 varied between 11 mg CO2 mg–1 N during vegetativeand early reproductive growth, to 12.5 mg CO2 mg–1 N duringthe later stages of pod development. In whole filaments of Anabaena cylindrica dark nitrogen-fixingactivity (measured as acetylene reduction) and respiration increasedwith the light intensity of a fixed period of preillumination,saturating at ca. 10,000 lux. With saturating light during preillumination,the amount and duration of dark nitrogen-fixing activity increasedwith length of preillumination, but respiration declined rapidlyin the dark. At dark respiration rates below 250 nmol O2 uptake mg protein–1?h–1(State 1) no significant nitrogen-fixing activity is observed.From 250 to 550 nmol O2 uptake?mg protein–1?h–1(State 2), nitrogen-fixing activity depends on O2 uptake whileabove 550 nmol O2 uptake?mg protein–1?h–1 (State3), nitrogen-fixing activity no longer increases with furtherincrease in O2 uptake rate. Absorption rates of SO2 by pecan (Carya illinoensis (Wang) K.Koch) leaflets exposed to 2.6, 5.2, and 7.8 mg SO2 m–3were measured over a 2 h period. SO2 was rapidly absorbed bythe leaflets in all treatments during the initial 30–50min; the rate of uptake decreased to a rather constant levelthereafter. Total SO2 absorbed during the 2 h period was 15.6,25.6, and 38.9 nmol cm–2 for the low, medium, and highSO2 concentrations, respectively. Reductions in net photosyntheticrates were proportional to ambient SO2 concentrations and totalSO2 absorbed. Partial photosynthetic recovery occurred in alltreatments during a 2 h post-treatment period and full recoveryoccurred during a 12 h dark period. Exposure to SO2 resultedin slight increases in stomatal and boundary layer resistancesto CO2 and substantial increases in residual resistances. Absorptionrates of SO2 by alfalfa (Medicago saliva L.) exposed to 5.2mg SO2m–3 for 1 h were approximately double those of pecanexposed to the same ambient SO2 concentration. Alfalfa net photosyntheticrates were reduced 74% after 1 h exposure to 5.2 mg SO2 m–3while a depression of 42% occurred in pecan.  相似文献

Treatment of S-sufficient or S-deficient Chlorella pyrenoidosacells with NaHSO3, during an 8-h period in the light, significantlydecreased their chlorophyll and dry matter contents when thecells were incubated in the presence or absence of SO42+. Incontrols lacking HSO3, when the starting pH was 7.5,dry matter and chlorophyll contents increased slightly, whereasno significant changes in either occurred at a starting pH of3.0 when the cells and medium contained SO4. In the dark,at both pH 3.0 and 7.5, dry matter and chlorophyll contentsdecreased slightly. Bisulphite treatment in the dark causedlittle decrease of either dry matter or chlorophyll when cellsand medium contained SO42–. However, in its absence, drymatter decreased markedly, but there was little change of chlorophyllcontent in the dark. The interactions between HSO3 asa source of S and as an inhibitor of growth and chlorophyllformation are discussed in the context of the changes inducedby light and alternative sources of S. Overall, the harmfuleffects of HSO3 outweigh any role it has as a sourceof S, since its effects are ameliorated by SO42–.  相似文献

Antarctic plankton community respiration rates were determinedfrom in vitro changes in dissolved oxygen. Oxygen consumptionrates, measured at in situ temperatures between 0 and 6°C,were found to lie in the range 0.3–3.7 µmol O2 l–1per 24 h. Water samples were collected between East FalklandIsland and South Georgia, South Atlantic Ocean, and incubatedshipboard in the dark at up to 36 temperatures between –2and 14–C. A respiration rate at each temperature was thendetermined and used to calculate the temperature coefficient(Q10) of Antarctic planktonic community respiration from theArrhenius equation. Fourteen Q0 values lay in the range 1–3,with four further values >5. This range of temperature coefficientvalues for community respiration is comparable to the publishedrange of values for plankton photosynthesis. Frequency distributionsof temperature coefficients for the two processes show similarmodal Q105 of 2–3. Thus, this study does not lend supportto the hypothesis of a differential response of photosynthesisand community respiration to low temperature.  相似文献
