No data are available on whether rising carbon dioxide concentration [CO2] or increased air temperature can alter the establishment and persistence of common ragweed ( Ambrosia artemisiifolia L.) within a plant community following soil disturbance. To determine ragweed longevity, we exposed disturbed soil with a common seed bank population to an in situ temperature and [CO2] gradient along an urban–rural transect beginning in early 2002. No other consistent differences in meteorological variables (e.g. wind speed, humidity, PAR, tropospheric ozone) as a function of urbanization were documented over the course of the study (2002–2005). Above-ground measurements of biomass over this period demonstrated that ragweed along the transect responded to urban induced increases in [CO2]/temperature with peak biomass being observed at this location by the end of 2003. However, by the Fall of 2004, and continuing through 2005, urban ragweed populations had dwindled to a few plants. The temporal decline in ragweed populations was not associated with increased disease, herbivory or auto-allelopathy, but was part of a demographic reduction in the total number of annual plant species observed for the urban location. In a separate experiment, we showed that such a demographic shift is consistent with CO2/temperature induced increases in biomass and litter accumulation, with a subsequent reduction in germination/survival of annual plant species. Overall, these data indicate that [CO2]/temperature differences associated with urbanization may increase initial ragweed productivity and pollen production, but suggest that long-term, multi-year persistence of ragweed in the urban macro-environment may be dependent on other factors.  相似文献

Testing whether and how subordinate individuals differ from dominants in the utilization of enriched CO2 atmospheres is important for understanding future stand and community structure. We hypothesized that subordinate and dominant Ambrosia artemisiifolia L. (Asteraceae) (common ragweed) plants growing in dense stands would not equally acquire or utilize carbon gains from CO2-enrichment, and that the resulting disproportionate growth gains to subordinates would reduce size inequalities in competing stands. We grew experimental stands of A. artemisiifolia in either ambient (360 μL L−1) or twice ambient (720 μL L−1) levels of atmospheric CO2. We compared the relative growth, photosynthetic capacity, and architecture of subordinate and dominant plants in each treatment, and assessed size inequalities using the stand-level coefficient of variation (CV). In elevated CO2, plants grew larger, but subordinate plants shifted more mass to upper stem allocation than dominants. Dominant plants demonstrated reduced leaf-level photosynthetic gains in elevated CO2 compared with subordinate plants. Reduced CVs in plant size reflected smaller proportional growth gains by dominants over subordinates in elevated vs. ambient stands. We conclude that differences in the architectural and physiological responses of subordinate and dominant ragweed plants reduce competition and allow subordinate plants to catch up to dominants in elevated CO2 conditions.  相似文献

Although the evolutionary and ecological processes that contribute to plant invasion have been the focus of much research, investigation into the molecular basis of invasion is just beginning. Common ragweed (Ambrosia artemisiifolia) is an annual weed native to North America and has been introduced to Europe where it has become invasive. Using a custom‐designed NimbleGen oligoarray, we examined differences in gene expression between five native and six introduced populations of common ragweed in three different environments (control, light stress and nutrient stress), as well as two different time points. We identified candidate genes that may contribute to invasiveness in common ragweed based on differences in expression between native and introduced populations from Europe. Specifically, we found 180 genes where range explained a significant proportion of the variation in gene expression and a further 103 genes with a significant range by treatment interaction. Several of these genes are potentially involved in the metabolism of secondary compounds, stress response and the detoxification of xenobiotics. Previously, we found more rapid growth and greater reproductive success in introduced populations, particularly in benign and competitive (light stress) environments, and many of these candidate genes potentially underlie these growth differences. We also found expression differences among populations within each range, reflecting either local adaptation or neutral processes, although no associations with climate or latitude were identified. These data provide a first step in identifying genes that are involved with introduction success in an aggressive annual weed.  相似文献

The processes of pollen grain development and germination depend on the uptake and metabolism of pollen sugars. In pepper (Capsicum annuum L.), initial sugar metabolism includes sucrose hydrolysis by invertase and subsequent phosphorylation of glucose and fructose by hexose kinases. The main objective of this study was to investigate changes in fructokinase (EC 2.7.1.4) and hexokinase (EC.2.7.1.1) activities in pepper flowers during their development, and to study the possible roles of these enzymes in determining pollen germination capacity under high temperature and under CO(2) enrichment, previously shown to modify sugar concentrations in pepper pollen (Aloni et al., 2001 Physiologia Plantarum 112: 505-512). Fructokinase (FK) activity was predominant in pepper pollen, and increased during pollen maturation. Pollen hexokinase (HK) activity was low and did not change throughout pollen development. High-temperature treatment (day/night, 32/26 degrees C) of pepper plants reduced the percentage of pollen that germinated compared with that under normal temperatures (26/22 degrees C), and concomitantly reduced the activity of FK in mature pollen. High temperature also reduced FK and HK activity in the anther. Under high ambient CO(2) (800 micro l l(-1)) pollen FK activity was enhanced. The results suggest that pollen and anther FK may play a role in the regulation of pollen germination, possibly by providing fructose-6-phosphate for glycolysis, or through conversion to UDP-glucose (UDPG) to support the biosynthesis of cell wall material for pollen tube growth. High temperature stress and CO(2) enrichment may influence pollen germination capacity by affecting these pathways.  相似文献

Abstract

This work investigates the relationship between plant growth, grain yield, nutrient acquisition and partitioning in rice (Oryza sativa L.) under elevated CO2. Plants were grown hydroponically in growth chambers with a 12-h photoperiod at either 370 or 700 µmol CO2 mol?1 concentration. Plant dry mass (DM), grain yield and macro- and micronutrient concentrations of vegetative organs and grains were determined. Elevated CO2 increased biomass at tillering, and this was largely due to an increase in root mass by 160%. Elevated CO2 had no effect on total nutrient uptake (N, P, K, Mg and Ca). However, nutrient partitioning among organs was significantly altered. N partitioning to leaf blades was significantly decreased, whereas the N partitioning into the leaf sheaths and roots was increased. Nutrient use efficiency of N, P, K, and Mg in all organs was significantly increased at elevated CO2. At harvest maturity, grain yield was increased by 27% at elevated CO2 while grain (protein) concentration was decreased by a similar magnitude (28%), suggesting that critical nutrient requirements for rice might need to be reassessed with global climate change.  相似文献

Background

Peroxiredoxins have diverse functions in cellular defense-signaling pathways. 2-Cys-peroxiredoxins (2-Cys-Prx) reduce H2O2 and alkyl-hydroperoxide. This study describes the purification and characterization of a genuine 2-Cys-Prx from Vigna unguiculata (Vu-2-Cys-Prx).

Methods

Vu-2-Cys-Prx was purified from leaves by ammonium sulfate fractionation, chitin affinity and ion exchange chromatography.

Results

Vu-2-Cys-Prx reduces H2O2 using NADPH and DTT. Vu-2-Cys-Prx is a 44 kDa (SDS-PAGE)/46 kDa (exclusion chromatography) protein that appears as a 22 kDa molecule under reducing conditions, indicating that it is a homodimer linked intermolecularly by disulfide bonds and has a pI range of 4.56–4.72; its NH2-terminal sequence was similar to 2-Cys-Prx from Phaseolus vulgaris (96%) and Populus tricocarpa (96%). Analysis by ESI-Q-TOF MS/MS showed a molecular mass/pI of 28.622 kDa/5.18. Vu-2-Cys-Prx has 8% α-helix, 39% β-sheet, 22% of turns and 31% of unordered forms. Vu-2-Cys-Prx was heat stable, has optimal activity at pH 7.0, and prevented plasmid DNA degradation. Atomic force microscopy shows that Vu-2-Cys-Prx oligomerized in decamers which might be associated with its molecular chaperone activity that prevented denaturation of insulin and citrate synthase. Its cDNA analysis showed that the redox-active Cys52 residue and the amino acids Pro45, Thr49 and Arg128 are conserved as in other 2-Cys-Prx.

General significance

The biochemical and molecular features of Vu-2-Cys-Prx are similar to other members of 2-Cys-Prx family. To date, only one publication reported on the purification of native 2-Cys-Prx from leaves and the subsequent analysis by N-terminal Edman sequencing, which is crucial for construction of stromal recombinant 2-Cys-Prx proteins.  相似文献

Elevated atmospheric CO2 may alter decomposition rates through changes in plant material quality and through its impact on soil microbial activity. This study examines whether plant material produced under elevated CO2 decomposes differently from plant material produced under ambient CO2. Moreover, a long‐term experiment offered a unique opportunity to evaluate assumptions about C cycling under elevated CO2 made in coupled climate–soil organic matter (SOM) models. Trifolium repens and Lolium perenne plant materials, produced under elevated (60 Pa) and ambient CO2 at two levels of N fertilizer (140 vs. 560 kg ha?1 yr?1), were incubated in soil for 90 days. Soils and plant materials used for the incubation had been exposed to ambient and elevated CO2 under free air carbon dioxide enrichment conditions and had received the N fertilizer for 9 years. The rate of decomposition of L. perenne and T. repens plant materials was unaffected by elevated atmospheric CO2 and rate of N fertilization. Increases in L. perenne plant material C : N ratio under elevated CO2 did not affect decomposition rates of the plant material. If under prolonged elevated CO2 changes in soil microbial dynamics had occurred, they were not reflected in the rate of decomposition of the plant material. Only soil respiration under L. perenne, with or without incorporation of plant material, from the low‐N fertilization treatment was enhanced after exposure to elevated CO2. This increase in soil respiration was not reflected in an increase in the microbial biomass of the L. perenne soil. The contribution of old and newly sequestered C to soil respiration, as revealed by the 13C‐CO2 signature, reflected the turnover times of SOM–C pools as described by multipool SOM models. The results do not confirm the assumption of a negative feedback induced in the C cycle following an increase in CO2, as used in coupled climate–SOM models. Moreover, this study showed no evidence for a positive feedback in the C cycle following additional N fertilization.  相似文献

Despite the recognition that the capacity to acquire N is critical in plant response to CO2 enrichment, there is little information on how elevated CO2 affects root N uptake kinetics. The few available data indicate a highly variable pattern of response to elevated CO2, but it is presently unclear if the observed inconsistencies are caused by differences in experimental protocols or by true species differences. Furthermore, if there are interspecific variations in N uptake responses to elevated CO2, it is not clear whether these are associated with different functional groups. Accordingly, we examined intact root‐system NH4+ and NO3 uptake kinetic responses to elevated CO2 in seedlings of six temperate forest tree species, representing (i) fast‐ vs. slow‐growers and (ii) broad‐leaves vs. conifers, that were cultured and assayed in otherwise similar conditions. In general, the species tested had a higher uptake capacity (Vmax) for NH4+ than for NO3. Species substantially differed in their NO3 and NH4+ uptake capacities, but the interspecific differences were markedly greater for NO3 than NH4+ uptake. Elevated CO2 had a species‐dependent effect on root uptake capacity for NH4+ ranging from an increase of 215% in Acer negundo L. to a decrease of about 40% in Quercus macrocarpa Michx. In contrast, NO3 uptake capacity responded little to CO2 in all the species except A. negundo in which it was significantly down‐regulated at elevated CO2. Across species, the capacity for NH4+ uptake was positively correlated with the relative growth rate (RGR) of species; however, the CO2 effect on NH4+ uptake capacity could not be explained by changes in RGR. The observed variation in NH4+ uptake response to elevated CO2 was also inconsistent with life‐form differences. Other possible mechanisms that may explain why elevated CO2 elicits a species‐specific response in root N uptake kinetics are discussed. Despite the fact that the exact mechanism(s) for such interspecific variation remains unresolved, these differences may have a significant implication for competitive interactions and community responses to elevated CO2 environment. We suggest that differential species responses in nutrient uptake capacity could be one potential mechanism for the CO2‐induced shifts in net primary productivity and species composition that have been observed in experimental communities exposed to elevated levels of CO2.  相似文献

Bread wheat (hexaploid AABBDD genome; 16 billion basepairs) is a genetically complex, self-pollinating plant with bisexual flowers that produce short-lived pollen. Very little is known about the molecular biology of its gametophyte development despite a longstanding interest in hybrid seeds. We present here a comprehensive characterization of three apparently homeologous genes (TAA1a, TAA1b and TAA1c) and demonstrate their anther-specific biochemical function. These eight-exon genes, found at only one copy per haploid complement in this large genome, express specifically within the sporophytic tapetum cells. The presence of TAA1 mRNA and protein was evident only at specific stages of pollen development as the microspore wall thickened during the progression of free microspores into vacuolated-microspores. This temporal regulation matched the assembly of wall-impregnated sporopollenin, a phenylpropanoid-lipid polymer containing very long chain fatty alcohols (VLCFAlc), described in the literature. Our results establish that sporophytic genes contribute to the production of fatty alcohols: Transgenic expression of TAA1 afforded production of long/VLCFAlc in tobacco seeds (18 : 1; 20 : 1; 22 : 1; 24 : 0; 26 : 0) and in Escherichia coli (14 : 0; 16 : 0; 18 : 1), suggesting biochemical versatility of TAA1 with respect to cellular milieu and substrate spectrum. Pollen walls additionally contain fatty alcohols in the form of wax esters and other lipids, and some of these lipids are known to play a role in the highly specific sexual interactions at the pollen-pistil interface. This study provides a handle to study these and to manipulate pollen traits, and, furthermore, to understand the molecular biology of fatty alcohol metabolism in general.  相似文献

Effects on sugar beet ( Beta vulgaris L.) of current and elevated CO2 and temperature alone and in combination and their interactions with abundant and deficient nitrogen supply (HN and LN, respectively) have been studied in three experiments in 1993, 1994 and 1995. Averaged over all experiments, elevated CO2 (600 μ mol mol–1 in 1993 and 700 μ mol mol–1 in 1994 and 1995) increased total dry mass at final harvest by 21% (95% confidence interval (CI) = 21, 22) and 11% (CI = 6, 15) and root dry mass by 26% (CI = 19, 32) and 12% (CI = 6, 18) for HN and LN plants, respectively. Warmer temperature decreased total dry mass by 11% (CI = – 15, – 7) and 9% (CI = – 15, – 5) and root dry mass by 7% (CI = – 12, – 2) and 7% (CI = – 10, 0) for HN and LN plants, respectively. There was no significant interaction between temperature and CO2 on total or root dry mass. Neither elevated CO2 nor temperature significantly affected sucrose concentration per unit root dry mass. Concentrations of glycinebetaine and of amino acids, measured as α -amino-N, decreased in elevated CO2 in both N applications; glycinebetaine by 13% (CI = – 21, – 5) and 16% (CI = – 24, – 8) and α -amino-N by 24% (CI = – 36, – 11) and 16% (CI = – 26, – 5) for HN and LN, respectively. Warmer temperature increased α -amino-N, by 76% (CI = 50, 107) for HN and 21% (CI = 7, 36) for LN plants, but not glycinebetaine.  相似文献

The aim of this work was to examine the response of wheat plants to a doubling of the atmospheric CO2 concentration on: (1) carbon and nitrogen partitioning in the plant; (2) carbon release by the roots; and (3) the subsequent N uptake by the plants. The experiment was performed in controlled laboratory conditions by exposing fast-growing spring wheat plants, during 28 days, to a 14CO2 concentration of 350 or 700 L L–1 at two levels of soil nitrogen fertilization. Doubling CO2 availability increased total plant production by 34% for both N treatment. In the N-fertilized soil, the CO2 enrichment resulted in an increase in dry mass production of 41% in the shoots and 23% in the roots; without N fertilization this figure was 33% and 37%, respectively. In the N-fertilized soil, the CO2 increase enhanced the total N uptake by 14% and lowered the N concentration in the shoots by 23%. The N concentration in the roots was unchanged. In the N-fertilized soil, doubling CO2 availability increased N uptake by 32% but did not change the N concentrations, in either shoots or roots. The CO2 enrichment increased total root-derived carbon by 12% with N fertilization, and by 24% without N fertilization. Between 85 and 90% of the total root derived-14C came from respiration, leaving only 10 to 15% in the soil as organic 14C. However, when total root-derived 14C was expressed as a function of root dry weight, these differences were only slightly significant. Thus, it appears that the enhanced carbon release from the living roots in response to increased atmospheric CO2, is not due to a modification of the activity of the roots, but is a result of the increased size of the root system. The increase of root dry mass also resulted in a stimulation of the soil N mineralization related to the doubling atmospheric CO2 concentration. The present study investigates cadmium (Cd) ability to enhance superoxides (O2) and nitric oxide (NO) production (as nitrites) in haemocytes of mussel Mytilus galloprovincialis as well as the possible involvement of Na+/H+ exchanger (NHE) in the induction of NADPH oxidase and NO synthase activity. PMA, a well-known PKC-mediated NADPH oxidase as well as NO synthase stimulator was also used, in order to verify Cd effects on both O2 and NO generation. According to the results of the present study, micromolar concentrations of Cd (0.05, 5, 10 and 50 μM) seemed to enhance O2 and NO generation in haemocytes of mussels. Moreover, O2 and NO generation in haemocytes exposed to Cd could be enhanced by its ability to induce reactive oxygen species (ROS) but respiratory burst activation as well. Inhibition of NO synthase with 10 μM l-NAME, significantly attenuated Cd ability to enhance O2 production and diminished NO generation, thus leading to the suggestion that Cd toxic effects, started at concentration of 50 μM, could enhance NADPH oxidase and NO synthase stimulation in haemocytes of mussels. NHE seems to play a regulatory role in the induction of either O2 or NO generation in haemocytes exposed to the metal, since its inhibition with the use of 10 μM EIPA significantly decrease both O2 and NO production. The involvement of NHE in the induction of O2 and NO generation, probably via PKC-mediated NADPH oxidase and NO synthase activation, is likely to be crucial to haemocytes exposed to heavy metals, such as Cd.  相似文献

The chiral recognition mechanism of a cinchona alkaloid-based chiral stationary phase (CSP) showing high enantiomer discrimination potential for 2-methoxy-2-(1-naphthyl)propionic acid (MalphaNP acid) was investigated. Conformational and structural analyses of the 1:1 complexes of 9-O-(tert-butylcarbamoyl) quinine selector (SO) and MalphaNP acid (selectand, SA) were carried out employing NMR spectroscopy in solution, Fourier-transform infrared (FT-IR) spectroscopy, and solid-state X-ray diffraction analysis. Intramolecular NOEs of a soluble analogue of the CSP afforded the conformational states of the free and complexed form of the selector. The (1)H-NMR spectra revealed that the free form of the SO constitutes anti-open as well as anti-closed and/or syn-closed conformers. Upon complexation with the (S)-MalphaNP acid enantiomer to form the more stable diastereomeric associate, a conformational transition of the selector takes place, resulting in the synthesis of the anti-open conformer nearly exclusively. FT-IR spectra reveal that, besides the primary ion-pairing interaction, stereoselective hydrogen bonding stabilizes the more stable complex via the amide hydrogen of the SO. X-ray diffraction analysis of 9-O-(tert-butylcarbamoyl)quinine and (S)-MalphaNP acid complex further revealed the occurrence of a bidentate H-bond-mediated ionic interaction between SO and SA as well as the lack of pi-pi interaction in the 1:1 complex, and corroborated the conclusions derived from spectroscopic and chromatographic studies.  相似文献

Naturally grown trees of Mediterranean evergreen oak (Quercus ilex L.), representing the climax species of the region, were enclosed in six large open-top chambers and exposed to ambient and elevated CO2 concentrations during a 3 year period. Maximum daily net photosynthetic rates measured at the two different CO2 concentrations were from 30 to 100% higher in elevated than in ambient [CO2] throughout the experimental period. The increase in maximum daily photosynthesis was also accompanied by a 93% rise in the apparent quantum yield of CO2 assimilation, measured during periods of optimum soil moisture conditions. Hence, no clear evidence of down-regulation of net photosynthetic activity was found. Interactions between atmospheric CO2 concentration and plant water stress were studied by following the natural evolution of drought in different seasons and years. At each level of water stress, the maximum rate of carbon assimilation was higher in elevated than in ambient [CO2] by up to 100%. Analysis of in vivo chlorophyll fluorescence parameters in normal (21%) and low (2%) oxygen concentrations provided useful insights into the functioning and stability of the photosynthetic processes. The photochemical efficiency of PSII (Fv/Fm) progressively decreased as drought conditions became more evident; this trend was accentuated under elevated [CO2]. Thermal de-excitation processes were possibly more significant under elevated than under ambient [CO2], in a combination of environmental stresses. This research suggests two possible conclusions: (i) a ‘positive’ interaction between elevated [CO2] and carbon metabolism can be obtained through relief of water stress limitation in the summer months, and (ii) elevated [CO2], under drought conditions, may also enhance the significance of slow-relaxing quenching.  相似文献

Summary Non-nodulatedalnus glutinosa plants were grown for 6 weeks in nutrient solutions using 3 combined-N treatments (NO3; NO3/NH4; and NH4) at a total N level of 4 meq.l–1, and growth was ccmpared with nodulated plants at zero N (N2 fixation). Of the combined-N sources, 100 per cent NH4 resulted in the highest dry matter yields when the solution pH was adjusted daily atc. 6. The dry matter yield was lowest with NO3.During the first 3 weeks, the yield of the N2-fixing plants was as high as that of the NH4 plants, but fell relatively behind during the second 3-week period. These effects could be attributed to higher initial N contents and higher shoot:root ratios, respectively, in the N2-fixing plants. Specific rates of N acquisition in the root were of a comparable order of magnitude for the combined-N and zero-N treatments.When NO3 was taken up, it was almost completely reduced in the roots. Regardless of N source there was a large excess of cations (C) relative to inorganic anions (A) in the plants, which was presumed to be balanced by an equivalent amount of organic anions (C-A). The relatively small differences in generation of organic anions for the various modes of N supply indicated the relative importance of the proton pump when NH4 or N2 was the N source. Proton or hydroxyl-ion effluxes, calculated on the basis of plant analyses, were generally in good agreement with measured excretion values. The acidity generation with N2 fixation amounted toc. 0.5 meq H+.mmol–1 Norg, which was distinctly higher than the range of 0.1–0.2 mentioned by Raven and Smith43 for dinitrogen-fixing plants.Without pH adjustment, specific rates of cation uptake and carboxylate generation were strongly depressed as the acidity increased, when NO3/NH4, NH4 and N2 were the N sources. Growth ofAlnus glutinosa appeared to be still normal at a pH ofc. 2.8. During the final 3 weeks, only the NH4 plants ceased growing at a pH of 2.6.  相似文献
