A new xylose fermenting yeast was isolated from over-ripe banana by enrichment in xylose-containing medium. The phylogenetic analysis of ITS1-5.8S-ITS2 region sequences of ribosomal RNA of isolate BY2 revealed that it shows affiliation to genus Pichia and clades with Pichia caribbica. In batch fermentation, Pichia strain BY2 fermented xylose, producing 15 g l?1 ethanol from 30 g l?1 xylose under shaking conditions at 28°C, with ethanol yield of 0.5 g g?1 and volumetric productivity of 0.31 g l?1 h?1. The optimum pH range for ethanol production from xylose by Pichia strain BY2 was 5–7. Pichia strain BY2 also produced 6.08 g l?1 ethanol from 30 g l?1 arabinose. Pichia strain BY2 can utilize sugarcane bagasse hemicellulose acid hydrolysate for alcohol production, efficiency of fermentation was improved by neutralization, and sequential use of activated charcoal adsorption method. Percent total sugar utilized and ethanol yield for the untreated hydrolysate was 17.14% w/v and 0.33 g g?1, respectively, compared with 66.79% w/v and 0.45 g g?1, respectively, for treated hemicellulose acid hydrolysate. This new yeast isolate showed ethanol yield of 0.45 g g?1 and volumetric productivity of 0.33 g l?1 h?1 from sugarcane bagasse hemicellulose hydrolysate detoxified by neutralization and activated charcoal treatment, and has potential application in practical process of ethanol production from lignocellulosic hydrolysate.  相似文献

The possibilities of parallel lactic acid and biomass production in batch and fed-batch fermentation on distillery stillage from bioethanol production were studied. The highest lactic acid yield and productivity of 92.3 % and 1.49 g L?1 h?1 were achieved in batch fermentation with initial sugar concentration of 55 g L?1. A significant improvement of the process was achieved in fed-batch fermentation where the concentration of lactic acid was increased to 47.6 % and volumetric productivity for 21 % over the batch process. A high number of Lactobacillus rhamnosus ATCC 7469 viable cells of 109 CFU ml?1 was attained at the end of fed-batch fermentation. The survival of 92.9 % of L. rhamnosus cells after 3 h of incubation at pH 2.5 validated that the fermentation media remained after lactic acid removal could be used as a biomass-enriched animal feed thus making an additional value to the process.  相似文献

Mutants of Candida magnoliae NCIM 3470 were generated by treatment of ultra-violet radiations, ethyl methyl sulphonate and N-methyl-N′-nitro-N-nitrosoguanidine. Mutants with higher reductase activity were screened by means of 2,3,5-triphenyl tetrazolium chloride agar plate assay. Among the screened mutants, the mutant R9 produced maximum mannitol (i.e. 46 g l?1) in liquid fermentation medium containing 250 g l?1 glucose and hence was selected for further experiments. Preliminary optimization studies were carried out on shake-flask level which increased the mannitol production to 60 g l?1 in liquid fermentation medium containing 300 g l?1 glucose. A two-stage fermentation process comprising of growth phase and production phase was employed. During the growth phase, glucose was supplemented and aerobic conditions were maintained. Thereafter, the production phase was initiated by supplementing fructose and switching to anaerobic conditions by discontinuing aeration and decreasing the speed of agitation. The strategy of two-stage fermentation significantly enhanced the production of mannitol up to 240 g l?1, which is the highest among all fermentative production processes and corresponds to 81 % yield and 4 g l?1 h?1 productivity without formation of any by-product.  相似文献

The evolutionary adaptation was carried out on the thermotolerant yeast Kluyveromyces marxianus NIRE-K1 at 45 °C up to 60 batches to enhance its xylose utilization capability. The adapted strain showed higher specific growth rate and 3-fold xylose uptake rate and short lag phase as compared to the native strain. During aerobic growth adapted yeast showed 2.81-fold higher xylose utilization than that of native. In anaerobic batch fermentation, adapted yeast utilized about 91 % of xylose in 72 h and produced 2.88 and 18.75 g l?1 of ethanol and xylitol, respectively, which were 5.11 and 5.71-fold higher than that of native. Ethanol yield, xylitol yield and specific sugar consumption rate obtained by the adapted cells were found to be 1.57, 1.65 and 4.84-fold higher than that of native yeast, respectively. Aforesaid results suggested that the evolutionary adaptation will be a very effective strategy in the near future for economic lignocellulosic ethanol production.  相似文献

Carbon distribution and kinetics of Candida shehatae were studied in fed-batch fermentation with xylose or glucose (separately) as the carbon source in mineral medium. The fermentations were carried out in two phases, an aerobic phase dedicated to growth followed by an oxygen limitation phase dedicated to ethanol production. Oxygen limitation was quantified with an average specific oxygen uptake rate (OUR) varying between 0.30 and 2.48 mmolO2 g dry cell weight (DCW)?1 h?1, the maximum value before the aerobic shift. The relations among respiration, growth, ethanol production and polyol production were investigated. It appeared that ethanol was produced to provide energy, and polyols (arabitol, ribitol, glycerol and xylitol) were produced to reoxidize NADH from assimilatory reactions and from the co-factor imbalance of the two-first enzymatic steps of xylose uptake. Hence, to manage carbon flux to ethanol production, oxygen limitation was a major controlled parameter; an oxygen limitation corresponding to an average specific OUR of 1.19 mmolO2 g DCW?1 h?1 allowed maximization of the ethanol yield over xylose (0.327 g g?1), the average productivity (2.2 g l?1 h?1) and the ethanol final titer (48.81 g l?1). For glucose fermentation, the ethanol yield over glucose was the highest (0.411 g g?1) when the specific OUR was low, corresponding to an average specific OUR of 0.30 mmolO2 g DCW?1 h?1, whereas the average ethanol productivity and ethanol final titer reached the maximum values of 1.81 g l?1 h?1 and 54.19 g l?1 when the specific OUR was the highest.  相似文献

Erythritol is an important natural sweetener, industrially produced only by fermentation on glucose media. Glycerol is an important renewable feedstock as it is the major by-product of the biodiesel production process; here we present an alternative way to convert this low-cost substrate into value-added products, such as erythritol. Repeated batch cultures (RBC) were performed to improve the productivity of erythritol from pure and crude glycerol. An acetate negative mutant of Yarrowia lipolytica Wratislavia K1 was found to be applicable for the production of high amounts of erythritol in RBC. When 20 % of fresh replaced medium was added, the strain Wratislavia K1 was able to produce 220 g l ?1 erythritol, which corresponded to a 0.43 g g?1 yield and a productivity of 0.54 g l?1 h?1. Additionally, the activity of the culture remained stable for more than 1,000 h, i.e., 11 cycles of the repeated batch bioreactors.  相似文献

Clostridium beijerinckii optinoii is a Clostridium species that produces butanol, isopropanol and small amounts of ethanol. This study compared the performances of batch and continuous immobilized cell fermentations, investigating how media flow rates and nutritional modification affected solvent yields and productivity. In 96-h batch cultures, with 80 % of the 30 g L?1 glucose consumed in synthetic media, solvent concentration was 9.45 g L?1 with 66.0 % as butanol. In a continuous fermentation using immobilized C. beijerinckii optinoii cells, also with 80 % of 30 g L?1 glucose utilization, solvent productivity increased to 1.03 g L?1 h?1. Solvent concentration reached 12.14 g L?1 with 63.0 % as butanol. Adjusting the dilution rate from 0.085 to 0.050 h?1 to allow extended residence time in column was required when glucose concentration in fresh media was increased from 30 to 50 g L?1. When acetate was used to improve the buffer capacity in media, the solvent concentration reached 12.70 on 50 g L?1 glucose. This continuous fermentation using immobilized cells showed technical feasibility for solvent production.  相似文献

This study, comprising three independent experiments, was conducted to optimize the zinc (Zn) application through seed coating for improving the productivity and grain biofortification of wheat. Experiment 1 was conducted in petri plates, while experiment 2 was conducted in sand-filled pots to optimize the Zn seed coating using two sources (ZnSO4, ZnCl2) of Zn. In the first two experiments, seeds of two wheat cultivars Lasani-2008 and Faisalabad-2008 were coated with 0.25, 0.50, 0.75, 1.00, 1.25, 1.50, 1.75 and 2.00 g Zn kg?1 seed using ZnSO4 and ZnCl2 as Zn sources. The results of experiment I revealed that seed coating with 1.25 and 1.50 g Zn kg?1 seed using both sources of Zn improved the seedling emergence. However, seed coated with 1.25 and 1.50 g Zn kg?1 seed using ZnSO4 was better regarding improvement in seedling growth and seedling dry weight. The results of the second experiment indicated that seed coated with 1.25 and 1.50 g Zn kg?1 seed using ZnSO4 improved the seedling emergence and seedling growth of tested wheat cultivars. However, seed coating beyond 1.5 g Zn kg?1 seed using either Zn source suppressed the seedling emergence. Third experiment was carried out in glass house in soil-filled earthen pots. Seeds of both wheat cultivars were coated with pre-optimized treatments (1.25, 1.50 g Zn kg?1 seed) using both Zn sources. Seed coating with all treatments of ZnSO4 and seed coating with 1.25 g Zn kg?1 seed using ZnCl2 improved the seedling emergence and yield-related traits of wheat cultivars. Seed coating with 1.25 g Zn kg?1 seed also improved the chlorophyll a and b contents. Maximum straw Zn contents, before and after anthesis, were recorded from seed coated with 1.5 g Zn kg?1 seed using either Zn source. Increase in grain yield from seed coating followed the sequence 1.25 g Zn kg?1 seed (ZnSO4) >1.25 g Zn kg?1 seed (ZnCl2) >1.5 g Zn kg?1 seed (ZnSO4). However, increase in grain Zn contents from seed coated was 1.5 g Zn kg?1 seed (ZnCl2) >1.25 and 1.5 g Zn kg?1 seed (ZnCl2, ZnSO4) >1.25 g Zn kg?1 seed (ZnSO4). Seed coating with Zn increased the grain Zn contents from 21 to 35 %, while 33–55 % improvement in grain yield was recorded. In conclusion, wheat seeds may be coated with 1.25 g Zn kg?1 seed using either source of Zn for improving the grain yield and grain Zn biofortification.  相似文献

The persistent edaphic stress on microbial succession due to dynamic changes during composting was explored for selection of multi-stress tolerant microbe(s) desirable for ethanol production. A total of 23 strains were isolated from mango compost using four successive enrichments in YP broth (g l?1): glucose, 100; 150; 250 with ethanol (40) and cycloheximide (0.4) at 40 °C, pH 6.0. Based on multi-gene ribotyping, 14 yeasts (61 %) of Saccharomycetaceae, 2 filamentous fungi (8.6 %) and 7 bacteria (30.4 %) were obtained. Phenetic and phylogenetic analysis of the 14 yeasts revealed 64.3 % tolerant to 500 g l?1 glucose, growth at 45 °C and resemblance to Candida sp. (14.3 %), Kluyveromyces marxianus (35.7 %), Pichia kudriavzevii (21.4 %) and Saccharomyces cerevisiae (28.6 %). Assessment of the 14 yeasts in glucose fermentation medium (pH 4.5 at 40 °C) showed ethanol productivity of ≥92 % by 12 yeasts with theoretical yields of 90–97 %. Fermentation of molasses (150 g l?1 glucose equivalent) by P. kudriavzevii D1C at 40 °C resulted in 73.70 ± 0.02 g l?1 ethanol and productivity of 4.91 ± 0.01 g l?1 h?1. Assessment of P. kudriavzevii D1C revealed multi-stress tolerance towards 5-hydroxymethyl furfural, ethanol (20 %, v/v), high gravity and H2O2 (0.3 M) indicating suitability for ethanol production using high gravity molasses and pre-treated lignocellulosic biomass fermentation.  相似文献

In this study, after the expression of a pyruvate carboxylase gene (PYC) cloned from Meyerozyma guilliermondii in a marine-derived yeast Yarrowia lipolytica SWJ-1b, a transformant PG86 obtained had much higher PYC activity than Y. lipolytica SWJ-1b. At the same time, the PYC gene expression and citric acid (CA) production by the transformant PG86 were also greatly enhanced. When glucose concentration in the medium was 60.0 g L?1, CA concentration formed by the transformant PG86 was 34.02 g L?1, leading to a CA yield of 0.57 g g?1 of glucose. During a 10-L fed-batch fermentation, the final concentration of CA was 101.0 ± 1.3 g L?1, the yield was 0.89 g g?1 of glucose, the productivity was 0.42 g L?1 h?1 and only 5.93 g L?1 reducing sugar was left in the fermented medium within 240 h of the fed-batch fermentation. A novel butanol fermentation process was developed in which sweet sorghum bagasse (SSB) was pretreated using liquid hot water (LHW) pretreatment technique followed by enzymatic hydrolysis and butanol (acetone butanol ethanol (ABE)) fermentation. A pretreatment temperature of 200 °C resulted in the generation of a hydrolyzate that inhibited butanol fermentation. When SSB pretreatment temperature was decreased to 190 °C (0-min holding time), the hydrolyzate was successfully fermented without inhibition and an ABE productivity of 0.51 g L?1 h?1 was achieved which is comparable to the 0.49 g L?1 h?1 observed in the control fermentation where glucose was used as a feedstock. These results are based on the use of 86 g L?1 SSB solid loadings in the pretreatment reactors. We were also able to increase SSB solid loadings from 120 to 200 g L?1 in the pretreatment step (190 °C) followed by hydrolysis and butanol fermentation. As pretreatment solid loadings increased, ABE yield remained in the range of 0.38–0.46. In these studies, a maximum ABE concentration of 16.88 g L?1 was achieved. Using the LHW pretreatment technique, 88.40–96.00 % of polymeric sugars (cellulose + hemicellulose) were released in the SSB hydrolyzate. The LHW pretreatment technique does not require chemical additions and is environmentally friendly, and the hydrolyzate can be used successfully for butanol fermentation.  相似文献

Arabic date is overproduced in Arabic countries such as Saudi Arabia and Iraq and is mostly composed of sugars (70–80 wt%). Here we developed a fed-batch fermentation process by using a kinetic model for the efficient production of lactic acid to a high concentration from Arabic date juice. First, a kinetic model of Lactobacillus rhamnosus grown on date juice in batch fermentation was constructed in EXCEL so that the estimation of parameters and simulation of the model can be easily performed. Then, several fed-batch fermentations were conducted by employing different feeding strategies including pulsed feeding, exponential feeding, and modified exponential feeding. Based on the results of fed-batch fermentations, the kinetic model for fed-batch fermentation was also developed. This new model was used to perform feed-forward controlled fed-batch fermentation, which resulted in the production of 171.79 g l?1 of lactic acid with the productivity and yield of 1.58 and 0.87 g l?1 h?1, respectively.  相似文献

A newly-isolated strain of Serratia marcescens, G12, was characterized for 2,3-butanediol (2,3-BD) production. In shake-flask and batch fermentations, 2,3-BD reached 48.5 and 51 g l?1, respectively. Low amounts of (~8 g l?1) of acetoin were also formed. In fed-batch fermentations, strain G12 produced 72.8 g 2,3-BD l?1 with glucose initially at 130 g l?1. When aeration rate was increased to 2.5 vvm for the fermentation process, 2,3-BD reached 87.8 g l?1 and the highest productivity was 1.6 g l?1 h?1. Acetoin was at 6.2 g l?1. G12 therefore may be a suitable candidate strain for large-scale production of 2,3-BD.  相似文献

Background

Corn stover, as one important lignocellulosic material, has characteristics of low price, abundant output and easy availability. Using corn stover as carbon source in the fermentation of valuable organic chemicals contributes to reducing the negative environmental problems and the cost of production. In ethanol fermentation based on the hydrolysate of corn stover, the conversion rate of fermentable sugars is at a low level because the native S. cerevisiae does not utilize xylose. In order to increase the conversion rate of fermentable sugars deriving from corn stover, an effective and energy saving biochemical process was developed in this study and the residual xylose after ethanol fermentation was further converted to l-lactic acid.

Results

In the hybrid process based on the hydrolysate of corn stover, the ethanol concentration and productivity reached 50.50 g L?1 and 1.84 g L?1 h?1, respectively, and the yield of ethanol was 0.46 g g?1. The following fermentation of l-lactic acid provided a product titer of 21.50 g L?1 with a productivity of 2.08 g L?1 h?1, and the yield of l-lactic acid was 0.76 g g?1. By adopting a blank aeration before the inoculation of B. coagulans LA1507 and reducing the final cell density, the l-lactic acid titer and yield reached 24.25 g L?1 and 0.86 g g?1, respectively, with a productivity of 1.96 g L?1 h?1.

Conclusions

In this work, the air pumped into the fermentor was used as both the carrier gas for single-pass gas stripping of ethanol and the oxygen provider for the aerobic growth of B. coagulans LA1507. Ethanol was effectively separated from the fermentation broth, while the residual medium containing xylose was reused for l-lactic acid production. As an energy-saving and environmental-friendly process, it introduced a potential way to produce bioproducts under the concept of biorefinery, while making full use of the hydrolysate of corn stover.

The effects of redox potential used as a control parameter on the process of succinic acid production in batch cultures of Actinobacillus succinogenes NJ113 have been investigated. In batch fermentation, cell growth and metabolite distribution were changed with redox potential levels in the range of ?100 to ?450 mV. From the results, the ORP level of ?350 mV was preferable, which resulted in high succinic acid yield (1.28 mol mol?1), high succinic acid productivity (1.18 g L?1 h?1) and high mole ratio of succinic acid to acetic acid (2.02). The mechanism of redox potential regulation was discussed by metabolic flux analysis and the ratio of NADH/NAD+. We expected that redox potential can be used as a valuable parameter to monitor and control much more anaerobic fermentation production.  相似文献

Cow manure with bedding is renewable organic biomass available around the year on dairy farms. Developing efficient and cost-effective psychrophilic dry anaerobic digestion (PDAD) processes could contribute to solving farm-related environmental, energy, and manure management problems in cold-climate regions. This study was to increase the organic loading rate (OLR), fed to a novel psychrophilic (20 °C) dry anaerobic digestion of 27 % total solid dairy manure (cow feces and wheat straw) in sequence batch reactor (PDAD-SBR), by 133 to 160 %. The PDAD-SBR process operated at treatment cycle length of 21 days and OLR of 7.0 and 8.0 g total chemical oxygen demand (TCOD)?kg?1 inoculum day?1 (5.2?±?0.1 and 5.8?±?0.0 g volatile solids (VS)?kg?1 inoculum day?1) for four successive cycles (84 days) produced average specific methane yields (SMYs) of 147.1?±?17.2 and 143.2?±?11.7 normalized liters (NL)?CH4?kg?1 VS fed, respectively. PDAD of cow feces and wheat straw is possible with VS-based inoculum-to-substrate ratio of 1.45 at OLR of 8.0 g TCOD kg?1 inoculum day?1. Hydrolysis was the limiting step reaction. The VS removal averaged around 57.4?±?0.5 and 60.5?±?5.7 % at OLR 7.0 and 8.0 g TCOD kg?1 inoculum day?1, respectively.  相似文献

Biomass productivity was quantified for the marine macroalga Derbesia tenuissima cultivated outdoors at seven stocking densities from 0.25 to 8 g L?1 for 5 weeks. Total lipids and fatty acid quantity and quality was measured from samples that were freeze-dried, dried by oven (75 °C), food dehydrator (60 °C), or outdoor in the sun (40 °C) or shade (38 °C). Stocking densities of 0.25 to 2 g L?1 yielded the highest biomass productivities (>20 g dry weight m?2 day?1) with no effect on total lipid quantity (11 %), or fatty acid quantity (5.3 %) or quality at any density tested. However, there was an interactive effect of stocking density and drying technique, with a decrease of up to 40 % in polyunsaturated fatty acids in sun-dried compared to freeze-dried biomass. Notably, while fatty acid and biomass productivity may be inseparable in macroalgae, cultivation conditions have a significant carryover effect in the post-harvest delivery of high-quality bio-oils.  相似文献

The suitability of poplar (Populus × canadensis Moench.—genotype ‘Neva’), black locust (Robinia pseudoacacia L.), and eucalyptus (Eucalyptus bridgesiana R. Baker) growing in short rotation coppice (SRC) system in a Mediterranean area (southern Italy), and under two management regimes, was evaluated in terms of survival, biomass yield, biomass quality, and soil fertility. The high management regime (H treatment) consisted of high plant density (6667 trees ha?1) and a 2-year harvesting cycle; the low management regime (L treatment) consisted of low plant density (1667 trees ha?1) and a 4-year harvesting cycle. The dry biomass production was 36, 13, and 9 t dry matter (dm)?ha?1 in the H treatment and 25, 14, and 7 t dm ha?1 in the L treatment for eucalyptus, black locust, and poplar, respectively. The analysis of the biomass showed a superior quality for the black locust feedstock because of its low moisture and ash percentages, high heating value (HHV), and low alkali metal concentrations, although, from an environmental point of view, the high N (12.3 g kg?1) and S (0.7 g kg?1) biomass concentrations would increase the pollutant emissions generated by combustion. Eucalyptus showed a high HHV, especially for the H treatment (18.70 MJ kg?1). Its high concentrations of K (4 g kg?1) and Mg (0.8 g kg?1) could provoke slagging and fouling in combustion equipment, and the high concentrations of S and N, if leaves are considered in the harvested biomass, indicate the low quality of its feedstock. No specific poplar feedstock stood out, although it had a good HHV (19.02 MJ kg?1). The soil fertility was not affected negatively after the 4-year SRC cycle, while S content in soil showed a tendency to increase in the case of black locust cultivation.  相似文献

Among several fatty acids tested, oleic acid was selected as the most efficient inducer for the production of 4-hydroxydodecanoic acid, a metabolite of β-oxidation, by Waltomyces lipofer. Cells were induced by incubation for 12 h in a medium containing 10 g l?1 yeast extract, 10 g l?1 peptone, 5 g l?1 oleic acid, 1 g l?1 glucose, and 0.05 % (w/v) Tween 80. The optimal reaction conditions for the production of γ-lactones by induced cells were pH 6.5, 35 °C, 200 rpm, 0.71 M Tris, 60 g l?1 hydroxy fatty acid, and 20 g l?1 cells. Non-induced cells produced 38 g l?1 γ-dodecalactone from 60 g l?1 10-hydroxystearic acid after 30 h, with a conversion yield of 63 % (w/w) and a productivity of 1.3 g l?1 h?1 under the optimized conditions, whereas induced cells produced 51 g l?1 γ-dodecalactone from 60 g l?1 10-hydroxystearic acid after 30 h, with a conversion yield of 85 % (w/w) and a productivity of 1.7 g l?1 h?1. The conversion yield and productivity of induced cells were 22 % and 1.3-fold higher, respectively, than those of non-induced cells. Induced cells also produced 28 g l?1 γ-decalactone and 12 g l?1 γ-butyrolactone from 60 g l?1 12-hydroxystearic acid and 60 g l?1 10-hydroxydecanoic acid, respectively, after 30 h. The concentration, conversion yield, and productivity of γ-dodecalactone and γ-decalactone are the highest reported thus far. This is the first study on the biotechnological production of γ-butyrolactone.  相似文献

This study explores the possibility of producing ethanol using the acid hydrolysate of three abundant agar-containing red seaweeds (agarophytes): Gelidium amansii, Gracilaria tenuistipitata, and Gracilariopsis chorda. The main component in the seaweed samples was agar, which ranged from 20 to 51 % (g g?1 dry weight). After optimizing acid hydrolysis, 100 g of seaweed was hydrolyzed at 130 °C for 15 min with 0.2 M H2SO4. Then, 120 mL of a 1:2 mixture of the hydrolysate broth and basal medium was fermented in a 200-mL bottle at 30 °C for 96 h. Of the three seaweeds, G. amansii had the best ethanol yield, producing 0.23 g g?1 of galactose or 45 % of the theoretical yield. This yield increased to 60 % after detoxification of the hydrolysate with activated carbon.  相似文献
