本文报道了分布在云南的4种树蛙(峨眉泛树蛙,黑点泛树蛙,无声囊泛树蛙和棕褶树蛙)垢核型,C-带及Ag-NORs它们具有相同的二倍体数(2n=26),除无声囊泛树蛙是6+7,其余均为5+8核型模式,种间SM的数目和顺序,次缢痕和Ag-NORs相互间有所差异。黑点泛树蛙Nos.3,12为SM,Ag-NORs位于10p^finter,峨眉泛树蛙Nfos2,3,4,6.9为SM,随体和Ag-NORs在12  相似文献

用酶组织化学和免疫组织化学双标技术,观察了正常SD大鼠基底前脑内侧隔核(MS)、斜角带垂直支(VDB)和水平支(HDB)中NOS阳性神经元的形态和分布及NOS与胆碱能神经元标志物ChAT、NGF受体(NGF-R)和AChE之间的共存关系。结果发现,MS、VDB和HDB的头端NOS阳性神经元较多、胞体较大、突起多,尾端NOS阳性神经元数目较少、胞体较小、突起少而短。NOS+ChAT双标神经元占NOS阳性神经元总数的90%,占ChAT阳性神经元总数的39%;NOS+NGF-R双标神经元占NOS阳性神经元总数的83%,占NGF-R阳性神经元总数的40%;NOS+AChE双标神经元占NOS阳性神经元总数的96%,占AChE阳性神经元总数的39%。这些结果为研究Alzheimer'sdisease病理过程中基底前脑隔区胆碱能神经元退变与NO的关系提供了形态学依据。  相似文献

本文对我国云南南部的白须长臂猿(H.leucogenys)染色体的G带、C带、晚复制带及Ag-NORs进行了较为详细的研究。它的2n=52,核型公式为44(M或SM)+6(A),XY(M,A)。C带表明一些染色体着丝点C带弱化;有的染色体出现插入的和端位的C带;X染色体两臂有端位C带,Y染色体是C带阳性和晚复制的。Ag-NORs的数目,雌体有4个,雄体有5个,Y染色体上具NOR。本文对白颊长臂猿与其它长臂猿间的亲缘关系、核型进化的可能途径进行了讨论。  相似文献

采用带血管灌流的离体大鼠胃模型,就铃蟾肽类肽能神经介质C(NMC)对促胃液素分泌的影响及其与胆碱能神经的关系,以及铃蟾肽受体拮抗剂(BN-Ant)D-Phe~6-BN(6-13)OMe对NMC诱导的促胃液素分泌的影响等问题进行探讨。实验结果表明,NMC可明显刺激大鼠胃分泌促胃液素(从基础的550±126pg/10min增加到刺激时的1060±180pg/10min),与对照组或自身基础状态比较,差异非常显著(P<0.01);此效应(净增量)被D-Phe~6-BN(6-13)OMe完全消除或被阿托品抵消约60%(P<0.01)。本实验结果证实,铃蟾肽类肽能神经递质NMC是促胃液素分泌的强刺激剂;新近问世的BN-AntD-Phe~6-BN(6-13)OMe具有高效拮抗NMC刺激促胃液素分泌的作用;同时揭示,NMC的作用机制不仅直接对胃G细胞、而且还可能通过影响胆碱能神经而发挥效应。  相似文献

以鹿角菜胶(CAR)注射到大鼠一侧后爪的足底皮下作为伤害性刺激模型,分别于CAR刺激后6、12h和1、3d处死动物,对照组动物仅将盐水注入一侧后爪足底皮下,用原位杂交法和免疫组织化学法观察前原脑啡肽(PPE)mRNA阳性神经元、亮氨酸脑啡肽(L-ENK)和μ阿片受体(MOR)样阳性结构在大鼠脊髓背角(SDH)的分布和变化。对照组大鼠SDH内可见到大量PPEmRNA阳性神经元,这些阳性神经元主要分布于Ⅰ、Ⅱ层和Ⅴ、Ⅵ层,CAR刺激后6h,刺激侧SDH中PPEmRNA阳性神经元的数量明显增多,12h和1d达到最高水平,3d时略有下降,但仍高于正常水平。L-ENK样阳性纤维和终末主要分布于正常大鼠SDH的Ⅰ、Ⅱ层,CAR刺激后1d,L-ENK样阳性结构在刺激侧SDH中的密度略有升高,3d后下降直至低于正常水平。MOR阳性胞体和纤维主要分布于SDH的Ⅱ层,CAR刺激后1d,刺激侧Ⅱ层中MOR阳性结构明显增加,并持续到刺激后3d。上述结果提示阿片类物质在伤害性信息调控中具有重要作用。  相似文献

目的探讨肥大细胞与肠嗜铬细胞在人胃溃疡发病中的作用。方法收集胃溃疡标本90例,正常胃组织30例。采用甲苯胺兰检测肥大细胞的数量、分布,免疫组化染色检测类胰蛋白酶、5-羟色胺表达,另取5例新鲜胃溃疡组织进行电镜检测。结果溃疡组与对照组比较肥大细胞及肠嗜铬细胞数量均明显增加(P〈0.05),肥大细胞在溃疡组脱颗粒现象明显,线性相关分析显示肥大细胞与肠嗜铬细胞数量呈正相关,相关系数为0.741。结论肥大细胞与肠嗜铬细胞在人胃溃疡发病中均发挥了作用。  相似文献

Several studies have examined the role of mast cells in the myocardial response to injury such as that caused by hypertension and ischemia-reperfusion. However, little is known about the influence of mast cells on normal myocardial structure and function. The present experiments examined cardiac function in Langendorff-perfused hearts isolated from 6- and 9-mo-old male mast cell-deficient (Ws/Ws) and mast cell-competent rats. A fluid-filled balloon catheter was used to measure left ventricular diastolic and systolic function at increasing preload volumes. At 6 mo of age, mast cell-deficient rats showed a slight cardiac hypertrophy (as monitored by heart weight and heart weight-to-body weight ratio) but no significant change in maximum observed systolic or diastolic function. In contrast, at 9 mo of age, the mast cell-deficient group showed no signs of hypertrophy but displayed a diastolic dysfunction characterized by decreased compliance without a significant decline in maximum observed basal -dP/dtmax. There were no significant differences in maximum observed values for measures of systolic function (developed pressure and +dP/dtmax). In summary, the results of this study in adult rats suggest that mast cells influence cardiac function in the absence of injury and that observed differences between mast cell-competent and -deficient animals vary with age. Thus it is important to consider these "physiological" actions and resulting changes in function when studying effects of insult in mast cell-deficient models.  相似文献

Specimens of O. americanus collected in eight localities from Rio Grande do Sul and four from Paraná (Brasil) were studied. Patterns of chromosomal localization and activity of nucleolar organizer regions (NORs) were established using the silver banding technique. It was shown that diploid and tetraploid specimens from all localities have homologues of group 4 bearing active NORs. In addition, some populations showed NOR activity on only one homologue number 5 (Uruguaiana, Prudentópolis), 8 (Itaqui) or 2 (Pato Branco). Intrapopulation polymorphism was found, some specimens showing the extra band and others not. Most cells analysed from the intestinal epithelium of an individual showed the same particular staining pattern of NORs, which were unequally stained, suggesting differences in their activity. The tetraploid genomes showed a maximum of five and a minimum of two active NORs. Diploid genomes showed two active NORs and four in the case of one specimen from Pato Branco. Four diploid populations and sympatry of 2n and 4n animals in Santa Bárbara do Sul (RS) were detected for the first time in South Brasil. Karyological evolution, as well as the meaning of number and unequal activity of NORs on tetraploid genomes are discussed.  相似文献

Changes of nucleolar organizer region in HL-60 cells after treated with retinoic acid (RA) were studied with techniques of silver-staining nucleolar organizer region (Ag-NOR) in metaphase karyotypes, Brachet's reaction and with our improved TEM techniques for studying silver-stained active nucleolar organizer region (Ag-aNOR) in interphase nucleoli. Number of Ag-NOR in HL-60 cells is 4.5/cell on average. The Ag-NOR number of cells treated with RA showed no remarkable difference from that of control group. Ag-aNOR number treated with RA was reduced obviously as compared with that of control group. Meanwhile, the changes of nucleolus number showed by Brachet's reaction were in accordance with those of Ag-aNOR. Therefore, it may be concluded: (1). Though the number of active rRNA genes did not changed after the differentiation of HL-60 cells induced by RA, their expression was clearly inhibited: (2). The relationship between the changes of Brachet-No and Ag-aNOR is in positive correlation (r = 0.98, p less than 0.01). EM examination of Ag-aNOR of HL-60 cells reveals that Ag-protein (RNA polymerase I) only presented in fibrillar centers (FC) and the dense fibrillar components (DFC) of nucleolus. In addition, in control group, large amount of Ag-protein, FC, DFC and granular components (GC) were observed, and there were many large nucleoli in a nucleus, meanwhile, the cells of the treated group tended to be mature, with a decrease in the amount of Ag-protein, FC, DFC and GC accordingly, and the nucleoli reduced both in size and number significantly.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

对35例食管癌与30例胃癌间质中的肥大细胞与淋巴细胞进行光镜与电镜观察。结果发现,两种癌间质中的肥大细胞数量在恶性程度低组均比恶性程度高组与对照组显著增加。在各组食管癌或胃癌中,有淋巴细胞高度浸润并形成淋巴小结者,肥大细胞也增多。用Alcian蓝—藏红染色,对照组Alcian蓝阳性和藏红阳性的肥大细胞都可以见到;而在食管癌胃癌间质中,大部分肥大细胞均呈Alcian蓝阳性。电镜观察,对照组肥大细胞颗粒多具有涡卷状结构,而癌组者则显示多样化形态。一些肥大细胞和淋巴细胞与癌细胞接触,肥大细胞可见活跃脱颗粒现象。结果提示,在抗肿瘤生长的防御机制中,肥大细胞与淋巴细胞有协同作用。  相似文献

目的 探讨肥大细胞在输卵管妊娠中的数量变化及其与血清性激素的关系。方法 :取输卵管妊娠时的输卵管及月经周期的增生期、分泌期和正常宫内早孕时的输卵管 ,常规石蜡切片 ,用甲苯胺蓝染色法显示肥大细胞 ;用酶免疫分析法检测输卵管妊娠患者、正常育龄未孕妇女 (增生期和分泌期 )及正常宫内早孕妇女血清雌二醇和孕酮水平。结果 :输卵管妊娠患者血清雌二醇和孕酮水平均高于正常育龄未孕妇女 (增生期和分泌期 ) ,低于正常宫内早孕妇女 ,四组间两两比较差异均有显著性 (P <0 0 5 ) ;肥大细胞主要分布于输卵管肌层 ,其数量变化为 :输卵管妊娠组较增生期和分泌期这两组均少 ,差异均有显著性 (P <0 0 5 ) ,而增生期和分泌期这两组肥大细胞数量变化不明显 ,差异无显著性 (P >0 0 5 ) ;两例正常宫内早孕时的输卵管壁肥大细胞数量明显比输卵管妊娠组多 ,与增生期和分泌期这两组比较 ,肥大细胞数量变化不明显。结论 :1 人输卵管壁内肥大细胞的数量不受血清性激素水平的影响。 2 输卵管妊娠时肥大细胞数量减少  相似文献

The tongue, pinna and dorsal skin of adult male C-1 mice were removed at 03.00, 06.00, 09.00, 12.00, 15.00, 18.00, 21.00 and 24.00 h, fixed in basic lead acetate and stained with Alcian blue-safranin or 0.5% toluidine blue. The mast cell numbers of these regions were counted and analyzed statistically by analysis of variance. It was found that there were circadian variations in the mast cell number in the tongue, pinna and dorsal skin. The difference between the minimum and maximum of circadian variation in mast cell number in all three regions was highly significant (p less than 0.01). Furthermore, the time points of the maximum and minimum of mast cell number varied between the different regions. The time point of the minimum in the tongue and pinna was at 06.00 h, whereas it was at 09.00 h in the dorsal skin. The time point of the maximum in the tongue and dorsal skin was at 21.00 h, but in the pinna it was at 18.00 h.  相似文献

研究肥大细胞在人胎儿甲状腺发育中数量、分布及组化性质的改变,以探讨胎儿器官发育中肥大细胞的差异。取45例不同胎龄的人胎甲状腺石蜡切片做甲苯胺蓝染色和阿尔辛蓝--藏红染色,并测定肥大细胞的临界电解质浓度值及进行硫酸小蘖硷荧光染色。结果显示:3月龄胎儿甲状腺内开始出现肥大细胞,数量极少,主要分布在被膜及小叶间结缔组织内,甲苯胺蓝染色肥大细胞颗粒呈淡紫蓝色,阿尔辛蓝--藏红染色呈蓝色,临界电解质浓度值较低,硫酸小蘖硷染色未见显黄色荧乐的肥大细胞,从3月龄到足月随着胎龄增长,肥大细胞数量缓慢增多,8月龄时肥大细胞经甲苯胺蓝染色,其颗粒呈紫红色,阿尔辛蓝--藏红染色出现少量含红色和红蓝混合染色颗粒的肥大细胞,临界电解质浓度值偏高,可见少量显黄色荧光的肥大细胞,结果表明:在人胎儿3月龄时甲状腺发育中开始出现肥大细胞,但随胎儿发育肥大细胞的组化性质改变不明显。  相似文献

本研究用雌性中国1号小鼠12只,分实验组和对照组,实验组分别腹腔注射天花粉后处死,取子宫组织作超薄切片,电镜观察。在天花粉作用下,常见子宫肥大细胞脱颗粒,呈功能活化状态,并与子宫平滑肌细胞紧密相邻。提示肥大细胞可能通过脱颗粒释放组胺促进子宫平滑肌的收缩。同时,也见肥大细胞与子宫结缔组织中的成纤维细胞,淋巴细胞等密切接触,提示成纤维细胞和淋巴细胞与天花粉作用下子宫肥大细胞的数量增多有关。  相似文献

We have investigated differences between the actions of encephalomyocarditis virus (EMCV) on cytometric indices in cultured NIH 3T3 and HEp-2 cells, which are characterized by different levels of transformation. HEp-2 cells surviving 48 h after EMCV infection showed lower nuclear ploidy, reduced nuclear area, fewer nucleoli and a higher percentage of euploid cells. There was a significant increase of nucleolar/nuclear DNA 6-24 h after EMCV infection. However, EMCV had markedly different effects on NIH 3T3 cells: there was a consistent increase in population ploidy, but the average number of nucleoli and the number of euploid cells in the population remained constant. The nucleolar/nuclear DNA ratio was almost unchanged. These different viral effects might be explained by the contrasting levels of differentiation of the cultured cell lines. The number of nucleoli does not depend on the amount of nuclear DNA in either viral-infected or intact cells but on the euploidy-to-aneuploidy ratio. The ratio of the sums of the nucleolar perimeters to the nuclear perimeter increases linearly with the number of nucleoli per nucleus in both intact and virus-infected cells. In both cell lines, the amount of DNA per nucleolus decreases as the number of nucleoli increases.  相似文献
