Background

Piglets are born with eight sharp teeth that during nursing can cause facial lesions on littermates and teat lesions on the sow. Teeth grinding in piglets is therefore often practiced to reduce these lesions. The aim of this study was to assess the consequences of grinding piglet teeth in regard to the occurrence of lesions. In this study the piglets' teeth were grinded in 28 litters, and in 36 litters the piglets' teeth were kept intact. Twice, one time during the first week and one time during the second week after birth facial lesions of the piglets were scored and the teats of the sows were examined for lesions. The facial lesion score accounted for the amount and severity of lesions. The individual observations on piglets in the litter were synthesized in a litter facial lesion score.

Findings

69.8% and 43.5% of the piglets had facial lesions in week 1 and week 2 respectively. The effect of treatment was not significant on litter facial lesion score. The litter facial lesion score was higher in week 1 than in week 2 (p < 0.001) and higher in large litters (p = 0.003) than in small litters. Mortality between week 1 and week 2 was higher in litters with intact teeth (p = 0.02). Sow teat lesions only occurred if litters had intact teeth.

Conclusions

According to our results teeth grinding is only justifiable in large litters.  相似文献

Background

Heart failure (HF)-induced skeletal muscle atrophy is often associated to exercise intolerance and poor prognosis. Better understanding of the molecular mechanisms underlying HF-induced muscle atrophy may contribute to the development of pharmacological strategies to prevent or treat such condition. It has been shown that autophagy-lysosome system is an important mechanism for maintenance of muscle mass. However, its role in HF-induced myopathy has not been addressed yet. Therefore, the aim of the present study was to evaluate autophagy signaling in myocardial infarction (MI)-induced muscle atrophy in rats.

Methods/Principal Findings

Wistar rats underwent MI or Sham surgeries, and after 12 weeks were submitted to echocardiography, exercise tolerance and histology evaluations. Cathepsin L activity and expression of autophagy-related genes and proteins were assessed in soleus and plantaris muscles by fluorimetric assay, qRT-PCR and immunoblotting, respectively. MI rats displayed exercise intolerance, left ventricular dysfunction and dilation, thereby suggesting the presence of HF. The key findings of the present study were: a) upregulation of autophagy-related genes (GABARAPL1, ATG7, BNIP3, CTSL1 and LAMP2) was observed only in plantaris while muscle atrophy was observed in both soleus and plantaris muscles, and b) Cathepsin L activity, Bnip3 and Fis1 protein levels, and levels of lipid hydroperoxides were increased specifically in plantaris muscle of MI rats.

Conclusions

Altogether our results provide evidence for autophagy signaling regulation in HF-induced plantaris atrophy but not soleus atrophy. Therefore, autophagy-lysosome system is differentially regulated in atrophic muscles comprising different fiber-types and metabolic characteristics.  相似文献

Seventeen new-born piglets of hybrid stock were tested for defects of neuromuscular transmission by stimulation electromyography (EMG). Nine of these displayed extreme symptoms of muscle weakness (splayleg), while the others were their clinically normal littermates. Muscles from four different functional groups were investigated, including the gastrocnemius, tibialis anterior, knee flexors and thigh adductors. Repetitive stimulation of muscle nerves at 3 Hz gave comparable peak-to-peak amplitudes of the EMG response in splayleg and control piglets (mean values from 5 to 10 mV). The lowest mean EMG response values at this stimulation frequency were found in splayleg adductor muscles of the thigh which were not, however significantly different from the controls. Higher frequencies of stimulation (30, 50 and 100 Hz), in general, led to a less pronounced decrease of EMG amplitude in splayleg piglet muscles than in the controls, with the exception of knee flexors. Neither splayleg nor control muscles exhibited post-activation exhaustion or post-tetanic potentiation. It is being concluded from these results that congenital myofibrillar hypoplasia is not primarily a myasthenia-like syndrome, but that either excitation-contraction coupling or the contractile mechanism itself are primarily affected.  相似文献

Introduction

Muscle symptoms in systemic sclerosis (SSc) may originate from altered skeletal muscle microcirculation, which can be investigated by means of blood oxygenation level dependent (BOLD) magnetic resonance imaging (MRI).

Methods

After ethics committee approval and written consent, 11 consecutive SSc patients (5 men, mean age 52.6 years, mean SSc disease duration 5.4 years) and 12 healthy volunteers (4 men, mean age 45.1 years) were included. Subjects with peripheral arterial occlusive disease were excluded. BOLD MRI was performed on calf muscles during cuff-induced ischemia and reactive hyperemia, using a 3-T whole-body scanner (Verio, Siemens, Erlangen, Germany) and fat-suppressed single-short multi-echo echo planar imaging (EPI) with four different effective echo times. Muscle BOLD signal time courses were obtained for gastrocnemius and soleus muscles: minimal hemoglobin oxygen saturation (T2*min) and maximal T2* values (T2*max), time to T2* peak (TTP), and slopes of oxygen normalization after T2* peaking.

Results

The vast majority of SSc patients lacked skeletal muscle atrophy, weakness or serum creatine kinase elevation. Nevertheless, more intense oxygen desaturation during ischemia was observed in calf muscles of SSc patients (mean T2*min -15.0%), compared with controls (-9.1%, P = 0.02). SSc patients also had impaired oxygenation during hyperemia (median T2*max 9.2% vs. 20.1%, respectively, P = 0.007). The slope of muscle oxygen normalization was significantly less steep and prolonged (TTP) in SSc patients (P<0.001 for both). Similar differences were found at a separate analysis of gastrocnemius and soleus muscles, with most pronounced impairment in the gastrocnemius.

Conclusions

BOLD MRI demonstrates a significant impairment of skeletal muscle microcirculation in SSc.  相似文献

Summary The histochemical pattern of muscle fiber types of the longissimus dorsi and biceps femoris muscles was investigated in normal and splaylegged piglets at birth and seven days later. Only slight differences between the muscle fibers at birth were found using histochemical reactions for alkaline adenosine triphosphatase (ATPase), succinate dehydrogenase (SDH), phosphorylase (PH) activities, and for the periodic acid-Schiff (PAS) reaction. With the method for acid-preincubated ATPase activity, high activity was observed in Type I muscle fibers and low activity in Type II muscle fibers in animals of both groups investigated. However, a higher number of Type I fibers was found in muscles of normal piglets, suggesting a faster and more advanced process of transformation of Type II into Type I muscle fibers in unaffected animals. Thus the histochemical conversion appears to be retarded in muscles of splaylegged animals, which have a histochemical pattern similar to that of normal prenatal animals. Cholinesterase activity in motor endplates was well developed; its staining revealed smaller sized and irregularly arranged endplates in muscles of affected piglets. Fiber type differentiation in muscles of animals which recovered from splayleg becomes fully developed and comparable to normal piglets seven days after birth. The number of fibers which became converted from Type II to Type I was increased; the fiber types were differentiated with regard to the PAS reaction and to their ATPase, SDH and PH activities. Morphological features of motor endplates in muscles of normal and surviving splaylegged piglets are similar.Histochemical investigation of the fiber type differentiation thus suggests that full recovery occurs within the first week of postnatal life in muscles affected by pathological changes accompanying splayleg.  相似文献

Background and aims

Mixing effects during litter decomposition could occur between two or more different litter species because of the potential nutrient transfer. However, evidence of mixing effects is variable and the underlying mechanisms remain unclear. Using a three-year decomposition experiment, we aim to examine for the effects of litter mixing and position on decomposition rates and nitrogen (N) and phosphorus (P) dynamics.

Methods

We studied litter decomposition of Stipa krylovii (Sk) and Astragalus galactites (Ag), two dominant species with contrasting litter quality, in a typical steppe of northern China in both single decomposition and three mixing treatments. The three mixing treatments included thorough mixing (Sk-Ag), Ag over Sk (Ag/Sk), and Sk over Ag (Sk/Ag).

Results

Both the Sk-Ag and the Sk/Ag mixture had negative mixing effects on the mass loss of the litter mixture, while the Ag/Sk mixture had a neutral mixing effect. The percent mass loss was higher when the litter species was placed at the top (25.0 and 51.9 % of mass remaining for Ag and Sk, respectively) than at the bottom (38.3 and 61.8 % of mass remaining for Ag and Sk, respectively). The Sk/Ag mixture had negative effects on the release of N while all three mixing treatments had positive effects on the release of P.

Conclusions

Our results indicate that: (1) mixing treatments can induce different mixing effects; (2) environmental factors likely play an important role in controlling the mixing effect; and (3) litter-mixtures have different non-additive effects on N and P, which may further increase the heterogeneity of N and P availability as the two litter species may fall differentially in terms of space and time.  相似文献

Background

Microarray profiling has the potential to illuminate the molecular processes that govern the phenotypic characteristics of porcine skeletal muscles, such as hypertrophy or atrophy, and the expression of specific fibre types. This information is not only important for understanding basic muscle biology but also provides underpinning knowledge for enhancing the efficiency of livestock production.  相似文献

Background

In order to increase the weak database concerning the organogenesis of Acoela – a clade regarded by many as the earliest extant offshoot of Bilateria and thus of particular interest for studies concerning the evolution of animal bodyplans – we analyzed the development of the musculature of Symsagittifera roscoffensis using F-actin labelling, confocal laserscanning microscopy, and 3D reconstruction software.

Results

At 40% of development between egg deposition and hatching short subepidermal fibres form. Muscle fibre development in the anterior body half precedes myogenesis in the posterior half. At 42% of development a grid of outer circular and inner longitudinal muscles is present in the bodywall. New circular muscles either branch off from present fibres or form adjacent to existing ones. The number of circular muscles is higher than that of the longitudinal muscles throughout all life cycle stages. Diagonal, circular and longitudinal muscles are initially rare but their number increases with time. The ventral side bears U-shaped muscles around the mouth, which in addition is surrounded by a sphincter muscle. With the exception of the region of the statocyst, dorsoventral muscles are present along the entire body of juveniles and adults, while adults additionally exhibit radially oriented internal muscles in the anterior tip. Outer diagonal muscles are present at the dorsal anterior tip of the adult. In adult animals, the male gonopore with its associated sexual organs expresses distinct muscles. No specific statocyst muscles were found. The muscle mantles of the needle-shaped sagittocysts are situated along the lateral edges of the animal and in the posterior end close to the male gonopore. In both juveniles and adults, non-muscular filaments, which stain positively for F-actin, are associated with certain sensory cells outside the bodywall musculature.

Conclusion

Compared to the myoanatomy of other acoel taxa, Symsagittifera roscoffensis shows a very complex musculature. Although data on presumably basal acoel clades are still scarce, the information currently available suggests an elaborated musculature with longitudinal, circular and U-shaped muscles as being part of the ancestral acoel bodyplan, thus increasing the possibility that Urbilateria likewise had a relatively complicated muscular ground pattern.  相似文献

Background

The aim of this study was to investigate the association between walking ability and muscle atrophy in the trunk and lower limbs.

Methods

Subjects in this longitudinal study were 21 elderly women who resided in nursing homes. The thicknesses of the following trunk and lower-limb muscles were measured using B-mode ultrasound: rectus abdominis, external oblique, internal oblique, transversus abdominis, erector spinae, lumbar multifidus, psoas major, gluteus maximus, gluteus medius, gluteus minimus, rectus femoris, vastus lateralis, vastus intermedius, biceps femoris, gastrocnemius, soleus, and tibialis anterior. Maximum walking speed was used to represent walking ability. Maximum walking speed and muscle thickness were assessed before and after a 12-month period.

Results

Of the 17 measured muscles of the trunk and lower limbs, age-related muscle atrophy in elderly women was greatest in the erector spinae, rectus femoris, vastus lateralis, vastus intermedius, and tibialis anterior muscles. Correlation coefficient analyses showed that only the rate of thinning of the vastus lateralis was significantly associated with the rate of decline in maximum walking speed (r = 0.518, p < 0.05).

Conclusions

This longitudinal study suggests that reduced walking ability may be associated with muscle atrophy in the trunk and lower limbs, especially in the vastus lateralis muscle, among frail elderly women.  相似文献

Background and aims

Litter decomposition is a key process controlling flows of energy and nutrients in ecosystems. Altered biodiversity and nutrient availability may affect litter decomposition. However, little is known about the response of litter decomposition to co-occurring changes in species evenness and soil nutrient availability.

Methods

We used a microcosm experiment to evaluate the simultaneous effects of species evenness (two levels), identity of the dominant species (three species) and soil N availability (control and N addition) on litter decomposition in a Mongolian pine (Pinus sylvestris var. mongolica) plantation in Northeast China. Mongolian pine needles and senesced aboveground materials of two dominant understory species (Setaria viridis and Artemisia scoparia) were used for incubation.

Results

Litter evenness, dominant species identity and N addition significantly affected species interaction and litter decomposition. Higher level of species evenness increased the decomposition rate of litter mixtures and decreased the incidence of antagonistic effects. A. scoparia-dominated litter mixtures decomposed faster than P. sylvestris var. mongolica- and S. viridis-dominated litter mixtures. Notably, N addition increased decomposition rate of both single-species litters and litter mixtures, and meanwhile altered the incidence and direction of non-additive effects during decomposition of litter mixtures. The presence of understory species litters stimulated the decomposition rate of pine litters irrespective of N addition, whereas the presence of pine litters suppressed the mass loss of A. scoparia litters. Moreover, N addition weakened the promoting effects of understory species litters on decomposition of pine litters.

Conclusions

Pine litter retarded the decomposition of understory species litters whereas its own decomposition was accelerated in mixtures. Nitrogen addition and understory species evenness altered species interaction through species-specific responses in litter mixtures and thus affected litter decomposition in Mongolian pine forests, which could produce a potential influence on ecosystem C budget and nutrient cycling.  相似文献

Background

The standard textbook information that annelid musculature consists of oligochaete-like outer circular and inner longitudinal muscle-layers has recently been called into question by observations of a variety of complex muscle systems in numerous polychaete taxa. To clarify the ancestral muscle arrangement in this taxon, we compared myogenetic patterns during embryogenesis of Ophryotrocha diadema with available data on oligochaete and polychaete myogenesis. This work addresses the conflicting views on the ground pattern of annelids, and adds to our knowledge of the evolution of lophotrochozoan taxa.

Results

Somatic musculature in Ophryotrocha diadema can be classified into the trunk, prostomial/peristomial, and parapodial muscle complexes. The trunk muscles comprise strong bilateral pairs of distinct dorsal and ventral longitudinal strands. The latter are the first to differentiate during myogenesis. They originate within the peristomium and grow posteriorly through the continuous addition of myocytes. Later, the longitudinal muscles also expand anteriorly and form a complex arrangement of prostomial muscles. Four embryonic parapodia differentiate in an anterior-to-posterior progression, significantly contributing to the somatic musculature. Several diagonal and transverse muscles are present dorsally. Some of the latter are situated external to the longitudinal muscles, which implies they are homologous to the circular muscles of oligochaetes. These circular fibers are only weakly developed, and do not appear to form complete muscle circles.

Conclusion

Comparison of embryonic muscle patterns showed distinct similarities between myogenetic processes in Ophryotrocha diadema and those of oligochaete species, which allows us to relate the diverse adult muscle arrangements of these annelid taxa to each other. These findings provide significant clues for the interpretation of evolutionary changes in annelid musculature.  相似文献

Background

Intermediate forms in the evolution of new adaptations such as transitions from water to land and the evolution of flight are often poorly understood. Similarly, the evolution of superfast sonic muscles in fishes, often considered the fastest muscles in vertebrates, has been a mystery because slow bladder movement does not generate sound. Slow muscles that stretch the swimbladder and then produce sound during recoil have recently been discovered in ophidiiform fishes. Here we describe the disturbance call (produced when fish are held) and sonic mechanism in an unrelated perciform pearl perch (Glaucosomatidae) that represents an intermediate condition in the evolution of super-fast sonic muscles.

Results

The pearl perch disturbance call is a two-part sound produced by a fast sonic muscle that rapidly stretches the bladder and an antagonistic tendon-smooth muscle combination (part 1) causing the tendon and bladder to snap back (part 2) generating a higher-frequency and greater-amplitude pulse. The smooth muscle is confirmed by electron microscopy and protein analysis. To our knowledge smooth muscle attachment to a tendon is unknown in animals.

Conclusion

The pearl perch, an advanced perciform teleost unrelated to ophidiiform fishes, uses a slow type mechanism to produce the major portion of the sound pulse during recoil, but the swimbladder is stretched by a fast muscle. Similarities between the two unrelated lineages, suggest independent and convergent evolution of sonic muscles and indicate intermediate forms in the evolution of superfast muscles.  相似文献

Aims

Plant litter decomposition plays an important role in the storage of soil organic matter in terrestrial ecosystems. Conversion of native vegetation to agricultural lands and subsequent land abandonment can lead to shifts in canopy structure, and consequently influence decomposition dynamics by alterations in soil temperature and moisture conditions, solar radiation exposure, and soil erosion patterns. This study was conducted to assess which parameters were more closely related to short-term decomposition dynamics of two predominant Mediterranean leaf litter types.

Methods

Using the litterbag technique, we incubated leaf litter of Pinus halepensis and Rosmarinus officinalis in two Mediterranean land-uses with different degree of vegetation cover (open forest, abandoned agricultural field).

Results

Fresh local litter lost between 20 and 55% of its initial mass throughout the 20-month incubation period. Rosemary litter decomposed faster than pine litter, showing net N immobilization in the early stages of decomposition, in contrast to the net N release exhibited by pine litter. Parameters related to litter quality (N content or C:N) or land-use/site conditions (ash content, an index of soil deposition on litter) were found to explain the cross-site variability in mass loss rates for rosemary and Aleppo pine litter, respectively.

Conclusions

The results from this study suggest that decomposition drivers may differ depending on litter type in this Mediterranean ecosystem. While rosemary litter was degraded mainly by microbial activity, decomposition of pine litter was likely driven primarily by abiotic processes like soil erosion.  相似文献

Background and Aims

Increased N availability induced by agricultural fertilization applications and atmospheric N deposition may affect plant nutrient resorption in temperate wetlands. However, the relationship between nutrient resorption and N availability is still unclear, and most studies have focused on leaf nutrient resorption only. The aim of our study was to examine the response of leaf and non-leaf organ nutrient resorption to N enrichment in a temperate freshwater wetland.

Methods

We conducted a 7-year N addition experiment to investigate the effects of increased N loading on leaf, sheath and stem nutrient (N and P) resorption of two dominant species (Deyeuxia angustifolia and Glyceria spiculosa) in a freshwater marsh in the Sanjiang Plain, Northeast China.

Results

Our results showed that, for both leaf and non-leaf organs (sheath and stem), N addition decreased N resorption proficiency and hence increased litter N concentration. Moreover, the magnitude of N addition effect on N resorption proficiency varied with fertilization rates for D. angustifolia sheaths and stems, and G. spiculosa leaves. However, increased N loading produced inconsistent impacts on N and P resorption efficiencies and P resorption proficiency, and the effects only varied with species and plant organs. In addition, N enrichment increased litter mass and altered litter allocation among leaf, sheath and stem.

Conclusions

Our results highlight that leaf and non-leaf organs respond differentially to N addition regarding N and P resorption efficiencies and P resorption proficiency, and also suggest that N enrichment in temperate freshwater wetlands would alter plant internal nutrient cycles and increase litter quality and quantity, and thus substantially influence ecosystem carbon and nutrient cycles.  相似文献

Background and aims

Freeze events can strongly influence many ecosystem processes. However, the effects of freeze events on litter production, litter quality, and decomposition are rarely documented.

Methods

In this study, litter fall was measured monthly for 2 years. Two litter decomposition experiments were also performed using freeze-damaged litter and non-damaged litter in a loblolly pine forest.

Results

The freeze event in November 2009 caused a pronounced pulse of needle litter fall. The freeze-damaged needle litter was shown to have higher N concentration and lower C/N ratio compared with the normal falling needle litter. This finding indicates that freeze damage significantly increased needle quality because of incomplete nutrient resorption. The decomposition of freeze-damaged needle litter was faster than that of normal falling yellow needle litter and slower than that of hand-picked green needle litter. The decomposition rate constant (k) was negatively correlated with the C/N ratio in the needle litter. Our results also showed that the different climatic conditions influence patterns of litter decomposition.

Conclusions

This study suggests that freeze events significantly alter litter quantity and quality, thus affecting litter decomposition rates in a loblolly pine forest in central China.  相似文献

Aims

We assessed the effects of native and exotic tree leaf litter on soil properties in two contrasting scenarios. The native Quercus robur and Pinus pinaster tree species coexist with the aliens Eucalyptus globulus and Acacia dealbata in acid soils of NW Spain. The native trees Fraxinus angustifolia and Ulmus minor coexist with the aliens Ailanthus altissima, Robinia pseudoacacia and Ulmus pumila in eutrophic basic riparian soils in Central Spain.

Methods

Four plastic trays per species were filled with homogenized top-soil of the site and covered with leaf litter. Before and after 9?months of incubation, litter mass, soil pH, organic matter, mineral and total N were measured. Available mineral N (NO 3 ? -N and NH 4 + -N) was assessed every 2?months.

Results

Soil biological activity was higher in the basic than in the acid soil. Litter of the exotic trees tended to decompose less than litter of native species, probably due to the presence of secondary metabolites in the former. Soil pH, mineral and total N responded differently to different litter types, irrespective of their exotic or native origin (acid soil), or was similar across litter treatments (basic riparian soil). The similar response of the basic soil to the addition of different litter types may be due to the low contrast of litter quality between the species. E. globulus litter inhibitied soil microbial activity much more than the rest of the studied litter types, leading to a drastic impoverishment of N in soils.

Conclusion

Litter of exotic N-fixing trees (A. dealbata and R. pseudoacacia) did not increase soil N pools because of the inhibition of microbial activity by secondary compounds. Therefore, secondary metabolites of the litter played a major role explaining exotic litter impact on soil properties.  相似文献
