Summary Plants with chromosome numbers (2n=18–54) lower than that of the normal 2n=56 were found in the C1 progeny of colchicine induced octoploid form ofHordeum murinum. Pollen mother cells in three octoploid plants had reduced chromosome complements at meiosis. Some of these plants had the original chromosome number of 2n=28. A number of these cells had regular bivalent pairing at first metaphase. Evidence indicated independent as well as nonindependent assortment of chromosomes at somatic reduction, the latter type resulting in ancestral type pollen mother cells (14II) and gametes respectively. Since normal tetraploid plants were recovered in the progeny (C2) of chromosome mosaics, ancestral type fully balanced egg cells must have also been formed. It is believed that the recovered tetraploids originated presumably from the fusion of double reduced gametes. 【目的】揭示蝉棒束孢居群形态性状的变异式样。【方法】通过对15个居群,75个蝉棒束孢菌株17个显微形态性状的观察和测量,采用SAS 8.1软件对蝉棒束孢显微形态性状数据进行描述性、巢式方差和Q聚类分析,获得蝉棒束孢显微形态变异数据。【结果】蝉棒束孢具有大小两型分生孢子,产孢细胞的形态可分为葫芦状和瓶状2种类型;蝉棒束孢在PDA培养基中易形成数量众多的厚垣孢子,菌丝间多能形成融合菌丝,并可观察到少量的融合菌丝在厚垣孢子与菌丝间、产孢细胞间形成。描述性分析表明,蝉棒束孢居群17个形态性状的变异系数(CV)在13.07%–104.09%之间,在物种水平上表现出丰富的变异;巢式方差分析表明,蝉棒束孢显微形态性状在居群间平均方差分量占总变异的11.29%,居群内个体间平均方差分量占总变异的15.27%,菌株内观察值间平均方差分量占总变异的73.44%。【结论】蝉棒束孢菌株形态性状变异丰富,是形态性状变异的主要来源,蝉棒束孢居群显微形态性状不存在明显的地理相关性。  相似文献

运用单因素方差分析、主成分分析、判别分析和聚类分析,对高原鼢鼠(Eospalax baileyi)4个地区(临潭、天祝、玛曲和碌曲)8个地理群体的头骨形态特征进行综合分析,比较13个特征变量的地理分异,挖掘形态特征的主要变量,并探讨其形态变异与环境的关系。结果发现,群体间13个特征变量均存在显著差异,主成分分析表明13个头骨特征变量可用3个主成分来表述,其累计贡献率达96.981%。颅全长、上齿列长及齿隙长是高原鼢鼠头骨发生分化的主要变量。天祝种群与碌曲种群,及玛曲种群与临潭种群都有重叠,其中碌曲的两个种群相似性最高,玛曲种群和临潭种群与其他种群几乎无重叠,显示完全分化,聚类分析结果与判别分析结果一致。头骨形态与地理因子的相关性分析发现头骨形态大小与海拔成显著的正相关关系。综合分析认为地理隔离、栖息地海拔因子影响下的生态条件等是高原鼢鼠头骨分化的主要因素。  相似文献

该文使用简单重复序列间( ISSR)分子标记,对送春与多花兰种间杂交后代进行了研究。结果表明:从80个ISSR引物中筛选出14个扩增效果稳定的ISSR引物,对两亲本和59个F1代个体进行了ISSR扩增,得到107个扩增位点,扩增的片段大小位于90~2100 bp之间,平均每个引物扩增7.64条条带,得到11种类型的带。 ISSR标记在送春×多花兰的F1代中表现出一定的多态性,分离频率为44.86%,分离位点有83.33%符合孟德尔1︰1或3︰1的分离规律,产生偏孟德尔分离的位点占12.50%,余下的4.17%属于特殊分离带型。可能导致后代变异的位点为偏孟德尔分离的6条带、缺失的8条带或新生成的2条带。聚类图中父本和母本与F1代个体间的遗传距离较远,59个杂交后代先聚集成一组,再同母本相聚为一组,最后才同父本聚在一起,59个杂种均偏母本型。送春与多花兰的杂交后代在植株形态、染色体、遗传物质方面都具备双亲特点,61个个体间的ISSR分子量标记结果和植株形态学特征都说明,59个F1代杂种包含送春和多花兰的遗传特性是真杂种;F1代杂种既有双亲的互补特征带,又有双亲的重组片断即产生新的特异带,这说明送春与多花兰的杂交后代具有遗传变异的特点。该研究结果可以有效地对杂交后代进行定向选择,为兰花的杂交育种提供了分子依据。  相似文献

在光学显微镜下观察了不同产地何首乌叶表皮结构特征,应用多元回归方法对不同产地何首乌的叶表皮特征与气候因子的关系进行了分析。观察的叶表皮特征指标有气孔密度、气孔指数、气孔器长、气孔器宽、气孔极区角质加厚、气孔器类型、表皮细胞垂周壁性状及叶表面角质条纹。观察结果:上表皮有少量气孔器分布;在下表皮,气孔器类型为非典型不等细胞型和不规则型,有少量腺鳞分布,气孔器密度每1mm2为241.7(64~573)个,气孔指数为17.1(7.5~26.5)%,气孔器长31.1(20~44)μm,气孔器宽23.1(16~38)μm。随着产地的不同,何首乌叶下表皮结构有明显差异。分析结果显示气孔器、气孔器宽度以及气孔密度均与纬度关系密切,随纬度的升高,气孔器长、气孔器宽呈减小的趋势,气孔密度呈增加的趋势,R2值分别为0.619、0.729、0.772。  相似文献

Dps(DNAprotection during starvation)蛋白是原核生物中特有的一类具有铁离子结合和抗氧化损伤功能的重要蛋白。利用体外PCR扩增技术和体内同源重组方法,获得了耐辐射奇球菌(Deinococcus radiodurans)dps全基因(DRB0092)缺失突变株。对突变株和野生型分别进行不同浓度过氧化氢(H2O2)处理,结果表明:与野生型菌株R1相比,dps突变株在低浓度H2O2(≤10mmol/L)条件下存活率急剧下降,而高浓度(≥30mmol/L)下则完全致死。Native-PAGE活性染色结果显示,稳定生长期dps突变株体内两种过氧化氢酶(KatA和KatB)的活性较野生型R1分别上调2.3倍和2.6倍。通过质粒构建和大肠杆菌诱导表达,获得可溶性Dps蛋白。体外结合和DNA保护实验结果显示:Dps具有明显的DNA结合功能,并能保护质粒DNA免受羟自由基攻击。本研究证明,Dps蛋白在耐辐射奇球菌抗氧化体系中发挥重要作用,可能对该菌极端抗性机制有重要贡献。  相似文献

Two species associated with the Gibberella fujikuroi species complex, G. circinata (the cause of pitch canker in pines) and G. subglutinans (avirulent on pine), were found to have limited interfertility in hybrid crosses. MAT idiomorphs, polymorphisms in the histone H3 gene, vegetative compatibility, and virulence phenotypes were used to verify recombination. The MAT idiomorphs appeared to be assorting independently, but the histone H3 haplotype was disproportionately represented by that of the G. subglutinans parent. Ninety-eight percent (45/46) of the progeny tested were vegetatively incompatible with both parents. All F1 progeny were avirulent to pine, but a wide range of virulence was restored through a backcross to the virulent parent (G. circinata). Attempts at hybrid crosses using other isolate combinations were rarely successful (1/26). This limited interfertility supports retention of G. circinata and G. subglutinans as separate species, but offers opportunities to characterize the inheritance of virulence to pine.  相似文献

Individual plants from the BC1F6 and BC1F8 backcross progenies of barley-wheat [H. marinum subsp. gussoneanum Hudson (=H. geniculatum All.) (2n = 28) × T. aestivum L. (2n = 42)] and the BC1F6 progeny of their amphiploids were used to obtain alloplasmic euploid (2n = 42) lines L-28, L-29, and L-49 and alloplasmic telocentric addition (2n = 42 + 2t) lines L-37, L-38, and L-50. The lines were examined by genomic in situ hybridization (GISH), microsatellite analysis, chromosome C-banding, and PCR analysis of the mitochondrial 18S/5S repeat. Lines L-29 and L-49 were characterized by substitution of wild barley chromosome 7H1 for common wheat chromosome 7D. In line L-49, common wheat chromosomes 1B, 5D, and 7D were substituted with homeologous barley chromosomes. Lines L-37, L-38, and L-50 each contained a pair of telocentric chromosomes, which corresponded to barley chromosome arm 7H1L. A survey on the distribution of surface flavonoid aglycones in 52 samples of 29 species of Veronica resulted in detection of eight flavone aglycones. Apigenin and luteolin were the most common constituents of the studied exudates, followed by the methylated derivatives apigenin 4′-methyl ether, apigenin 7,4′-dimethyl ether and luteolin 3′-methyl ether. Some intraspecific variations and the main ecological trends of flavonoid aglycone accumulation in Veronica were discussed. The observed exudate profiles appeared to be characteristic for some related groups within the genus, in agreement with the morphological, karyological, molecular and other chemical data.  相似文献

Cytological studies on Rubenstrensa exasperatum were carried out. The number of chromosomes in diploid cells (2n=16), and the morphological characteristics of the karyotype were determined. All the chromosomes (from Nos 2 to 8) had a well-defined short arm and, in accordance with the Ic values, were determined to be meta-submetacentric. Polymorphism was established in the length of the 2 bomologues of the 1st chromosome. This pair is probably linked with sex determination.  相似文献

Plant populations at high elevation face extreme climatic conditions and resource limitations. The existence of distylous species at different elevations can help us investigate their adaptation to high altitudes, the evolution of their morphological characteristics, as well as their responses to limited resources. Here, 17 populations of Primula nivalis at different elevations were evaluated regarding variations in plant morphological characteristics, biomass allocation, and morphological plasticity in a heterogeneous environment. Our results demonstrate that heterogeneous environments can affect plant morphological characteristics and resource allocation in each sexual morph of these plants. Moreover, environmental variations reduced morphological plasticity in the two plant morphs, and the plasticity of long style (LS) plants was greater than that of short style (SS) plants. There were significant negative correlations between morphological characteristics and elevation, rainfall, temperature, and sunshine, and these are the main variables that affect morphological characteristics and resource allocation of both morphs of P. nivalis plants in heterogeneous environments. The morphological characteristics of P. nivalis plants transplanted from high to lower elevations were not significantly different in either population. LS plants had greater morphological plasticity and adaptability in heterogeneous environments than SS plants. Elevational gradients and heterogeneous environments differentiated both morphs of P. nivalis plants with regards to morphology as well as adaptations. LS plants showed a higher level of adaptability than SS plants.  相似文献

Biological and morphological characters of the thelytokous and arrhenotokous forms ofA. mytilaspidis and their hybrids were investigated. Some biological differences between the thelytokous and arrhenotokous forms were found. The former is adapted to warmer condition and will parasitizeHemiberlesia lataniae as well asDiaspis echinocacti, while the latter will not parasitizeH. lataniae. The overall fecundity of the thelytokous form is lower than that of the arrhenotokous form, whereas the “effective fecundity”, female progeny per female parent, is higher in the thelytokous form. 为了解近年来入侵我国的菊方翅网蝽的生物学特性,并为该昆虫种群在我国未来发展的趋势及防治提供科学依据,我们在温度(25±2) ℃、湿度(80±5)%、光周期L∶D=14∶10的实验室条件下,饲养、观察并记录了其各龄幼虫的形态特征,测定了该虫生长发育历期、存活率和产卵量等,构建了实验种群繁殖特征生命表,并计算了种群动态的相关参数.结果表明: 菊方翅网蝽的卵期为(14.58±1.17) d,若虫期为(14.88±1.45) d,成虫寿命为(59.88±5.85) d,单雌产卵量为(87.2±17.8)粒.该实验种群的内禀增长率(rm)为0.05,周限增长率(λ)为1.06,世代平均历期(T)为46.11 d,净繁殖率(R0)为11.88,种群加倍时间(t)为12.91 d;此外,在种群的稳定年龄组配中,若虫期占59.3%,成虫期占40.7%.预计菊方翅网蝽有可能会在中国进一步扩散并造成潜在的危害.  相似文献

油松天然群体的种实性状表型多样性研究   总被引:1,自引:0,他引:1  
为了揭示油松天然种群在不同地理环境条件下表型变异的程度和规律,在油松整个天然分布范围内选择了12个具有代表性的居群作为研究对象,对其球果、种子、种翅等12个种实性状的变异程度及其与环境因子间关系进行了比较分析。结果显示:(1)各个性状在居群内和居群间均存在较大的变异(CV>12%)。其中千山(QS),曾家镇(ZJ)和互助(HZ)3个居群表现出了较高的变异(CV>20%),而球果干重(CDW)和种子长(CL)是所有表型性状中变异幅度最大的(CV分别为31%和21%),但种翅性状与其他性状相比具有较高的稳定性。(2)巢式设计方差分析表明,在居群内表型分化系数(Vst)变化在3.18%~89.86%之间,而群体间的Vst为38.97%;与其他针叶树种相比,油松拥有较高的表型分化系数,且居群内的变异程度远高于居群间的变异,尤其是千山(QS)、曾家镇(ZJ)和互助(HZ)3个居群,这说明油松具有较高的环境异质性适应能力或恶劣环境耐受能力。(3)相关性分析表明,该研究的各形态特征与潜在蒸发量均为负相关,且大部分形态指标间及它们与潜在蒸发量间存在显著相关性,表明潜在蒸发量是油松形态特征变化的最重要环境影响因子,预示油松最适宜生长于温暖潮湿的环境中;并表明因各形态特征间相互紧密关联,所以它们受环境条件影响而共变。  相似文献

Long-term morphological changes were investigated in the conodont genus Palmatolepis, using a Fourier analysis of the outline of platform elements. Three time-slices and four Late Frasnian - Early Famennian sections on North Gondwana were studied. No difference existed between locations, but shape variations were recorded associated with the successive Kellwasser events. The Middle Famennian period was analyzed in a section located at the Baltica margin across the annulata event. Some morphological changes may be related to the deposition of the corresponding black shales, but the main feature is a heterogeneity of the populations, which can be split into two morphological types. Latest Famennian conodonts from another North Gondwanan section show only minor shape changes during this time interval, which is environmentally stable prior to the global Hangenberg event. Based on synthetic shape variations across the whole period an morphological trend towards narrower elements is observed. This morphological change happens step-wise in response to global events. The amplitude of the shape shift is proportionally related to the extend of the successive Late Devonian global events.  相似文献

Leaf and ray flower flavonoids were investigated for the seven species of Helianthus series Angustifolii. Flavone aglycones occur in small glandular trichomes located on leaf undersurfaces of H. angustifolius, H. floridanus and H. simulans. Other species lacked both glandular trichomes and flavone aglycones. Flavonol glycosides occur in low concentrations in leaves of all species but were not characterized. Anthochlors (chalkones) occur in leaves of H. heterophyllus and H. longifolius. Ray flower flavonoids include anthochlor and flavonol glycosides and occur in the basal region of the ligule producing a band of UV A around the head. Anthochlors are the predominant ray flower flavonoids in H. angustifolius, H. heterophyllus and H. longifolius, whereas they are absent and quercetin 7-glucoside is present in H. carnosus and H. floridanus. Cladistic analysis of flavonoid and morphological characters indicates that evolution in the series has been a radiation from ancestral types rather than a linear sequence of progressively more derived species.  相似文献

Summary Two monosomics of Phaseolus vulgaris (2n = 22) were found among selfed progeny of plants treated with colchicine. The monosomic chromosomes involved were identified as chromosomes H and J according to the previously suggested Giemsa karyotype. Both monosomic plants had slower growth rate and smaller size as compared with their respective euploid sibs. However, no apparent morphological characteristics distinguished the two monosomics. The frequencies of transmission through selfing of monosomics H and J were 9% and 10 % respectively.  相似文献

为拓宽白菜育种的基因资源,改良白菜品质,以白菜(Brassica campestris,2n=20,AA)和甘蓝(B.oleracea L.var.capitata,2n=18,CC)的子叶和下胚轴为材料分离、制备原生质体。采用40%聚乙二醇(Polyethylene glycol,PEG)进行原生质体融合。融合细胞在以0.3 mol/L蔗糖、0.3 mol/L葡萄糖为渗透稳定剂,附加1.0 mg/L 2,4-D+0.5 mg/L 6-苄氨基嘌呤(6-BA)+0.1 mg/L 1-萘乙酸(NAA)+1.0 mg/L激动素(Kinetin,Kin)的改良K8p培养基中培养并诱导细胞分裂。小愈伤组织经增殖培养后在MS+0.2 mg/L玉米素(Zeatin,ZEA)+1 mg/L 6-BA+0.5 mg/L Kin+0.4 mg/L NAA的固体分化培养基上诱导出不定芽。30 d后再转入MS基本培养基,获得完整的再生植株。将生根的植株转移到花盆,并对其杂种性质进行形态学、细胞学和分子生物学鉴定。结果表明,经细胞融合分裂出的320个愈伤组织中,获得了35棵再生植株,其再生率达10.94%。形态学观察显示,绝大多数再生植株的叶面积较大,株型和叶型为两种杂交亲本的中间型,部分植株的叶片浓绿、肥厚。染色体计数结果显示,36.4%的再生植株染色体数为2n=38;36.4%的再生植株的染色体数为2n=58 60;27.2%的再生植株的染色体数为2n=70 76,超过两个融合亲本的染色体数的总和。流式细胞仪测定DNA含量显示,再生植株DNA含量变化比较大,其结果与染色体鉴定结果相吻合。随机扩增多态性DNA(Randomamplified polymorphic DNA,RAPD)和基因组原位杂交(Genomic in situ hybridization,GISH)分析结果证明再生植株具有双亲基因组。体细胞杂种花粉育性比较低,杂交、回交后其育性逐渐获得恢复,与白菜回交后代逐渐恢复了育性。通过体细胞杂交和回交、杂交获得了形态变化广泛的个体,为白菜的品种育种提供多样的种质资源。  相似文献
