Summary InCyclamen persicum Anneke, explants from the various vegetative organs of aseptic seedling formed embryoids. The optimal responses were recorded in Murashige and Skoog (MS) medium enriched with 5.0µM 2,4-dichlorophenoxyacetic acid (2,4-D), 0.5µM kinetin and 3–6% sucrose. Embryogenesis was enhanced at higher temperature of 25–30°C. On the other hand, light inhibited embryogenesis. Background

Rootstocks play an essential role to determining orchard performance of fruit trees. Pyrus communis and Cydonia oblonga are widely used rootstocks for European pear cultivars. The lack of rootstocks adapted to different soil conditions and different grafted cultivars is widely acknowledged in pear culture. Cydonia rootstocks (clonal) and Pyrus rootstocks (seedling or clonal) have their advantages and disadvantages. In each case, site-specific environmental characteristics, specific cultivar response and production objectives must be considered before choosing the best rootstock. In this study, the influence of three Quince (BA 29, Quince A = MA, Quince C = MC) and a local European pear seedling rootstocks on the scion yield, some fruit quality characteristics and leaf macro (N, P, K, Ca and Mg) and micro element (Fe, Zn, Cu, Mn and B) content of ‘Santa Maria’ pear (Pyrus communis L.) were investigated.

Results

Trees on seedling rootstock had the highest annual yield, highest cumulative yield (kg tree−1), largest trunk cross-sectional area (TCSA), lowest yield efficiency and lowest cumulative yield (ton ha−1) in the 10th year after planting. The rootstocks had no significant effect on average fruit weight and fruit volume. Significantly higher fruit firmness was obtained on BA 29 and Quince A. The effect of rootstocks on the mineral element accumulation (N, K, Ca, Mg, Fe, Zn, Cu, Mn and B) was significant. Leaf analysis showed that rootstocks used had different mineral uptake efficiencies throughout the early season.

Conclusion

The results showed that the rootstocks strongly affected fruit yield, fruit quality and leaf mineral element uptake of ‘Santa Maria’ pear cultivar. Pear seedling and BA 29 rootstock found to be more prominent in terms of several characteristics for ‘Santa Maria’ pear cultivar that is grown in highly calcareous soil in semi-arid climate conditions. We determined the highest N, P (although insignificant), K, Ca, Mg, Fe and Cu mineral element concentrations on the pear seedling and BA 29 rootstocks. According to the results, we recommend the seedling rootstock for normal density plantings (400 trees ha−1) and BA 29 rootstock for high-density plantings (800 trees ha−1) for ‘Santa Maria’ pear cultivar in semi-arid conditions.  相似文献

The application of 10?3 g 1?1 of 2,4 dichlorophenoxyacetic acid, 2-methyl-4-chlorophenoxyacetic acid, 3-amino-1,2,4-triazole and 6-chloro-2-ethylamino-4-isopropyl-amino-1,3,5-triazine toNicotiana tabacum cv. ‘Samsun’ plants inoculated with tobacco mosaic virus results in an increase in the content of this virus in the tissues. When whole plants are used, TMV content increases by 20% after herbicide application; when leaf dises are used, the amount of the virus can be doubled by the herbicide in comparison with control untreated inoculated discs. The results clearly show that the used non-selective herbicides act as stimulators of virus biosynthesis, probably via enhanced pentose phosphate cycle activity which enables an enhanced formation of viral RNA.  相似文献

Achillea filipendulina (family Asteraceae) is widespread throughout temperate North America. In order to clean stock plants from endemic fungal and bacterial contaminations a method for large-scale propagation of A. filipendulina through meristem culture was sought and found and is described in this paper. The best conditions for propagating A. filipendulina was found to be MS (Murashige and Skoog) salt medium supplemented with 3% sucrose and 1 mg l–1 IAA (indole-3-acetic acid) plus 2 mg l–1 BA (6-benzyladenine) under 16 h of cool fluorescent light. Rooted plants were successfully acclimatized within a short time after propagating on this medium. The propagation via tissue culture did not affect plant's presentation. The use of clean stock plants made it possible to increased Israeli production of Achillea from about 150,000 stems a year to about 1,300,000 stems a year.  相似文献

Radiation frost events, which have become more common in the Mediterranean Basin in recent years, inflict extensive damage to tropical/subtropical fruit crops. During radiation frost, sub-zero temperatures are encountered in the dark, followed by high light during the subsequent clear-sky day. One of the key processes affected by these conditions is photosynthesis, which, when significantly inhibited, leads to the enhanced accumulation of reactive oxygen species (ROS) and damage. The use of ‘chemical priming’ treatments that induce plants' endogenous stress responses is a possible strategy to improve their coping with stress conditions. Herein, we studied the effects of priming with sodium hydrosulfide (NaHS), a donor of hydrogen sulfide (H2S), on the response of photosynthesis to overnight frost and day high-light conditions in ‘Hass’ avocado (Persea americana Mill). We found that priming with a single foliar application of NaHS had positive effects on the response of grafted ‘Hass’ plants. Primed plants exhibited significantly reduced inhibition of CO2 assimilation, a lower accumulation of hydrogen peroxide as well as lower photoinhibition, as compared to untreated plants. The ability to maintain a high CO2 assimilation capacity after the frost was attained on the background of considerable inhibition in stomatal conductance. Thus, it was likely related to the lower accumulation of ROS and photodamage observed in primed ‘Hass’ plants. This work contributes toward the understanding of the response of photosynthesis in a subtropical crop species to frost conditions and provides a prospect for chemical priming as a potential practice in orchards during cold winters.  相似文献

In the rosaceous subtribe Pyrinae (formerly subfamily Maloideae), pathogen attack leads to formation of biphenyls and dibenzofurans. Accumulation of these phytoalexins was studied in greenhouse-grown grafted shoots of Malus domestica cv. ‘Holsteiner Cox’ and Pyrus communis cv. ‘Conference’ after inoculation with the fire blight bacterium, Erwinia amylovora. No phytoalexins were found in leaves. However, both classes of defence compounds were detected in the transition zone of stems. The flanking stem segments above and below this zone, which were necrotic and healthy, respectively, were devoid of detectable phytoalexins. The transition zone of apple stems contained the biphenyls 3-hydroxy-5-methoxyaucuparin, aucuparin, noraucuparin and 2′-hydroxyaucuparin and the dibenzofurans eriobofuran and noreriobofuran. In pear, aucuparin, 2′-hydroxyaucuparin, noreriobofuran and in addition 3,4,5-trimethoxybiphenyl were detected. The total phytoalexin content in the transition zone of pear was 25 times lower than that in apple. Leaves and stems of mock-inoculated apple and pear shoots lacked phytoalexins. A number of biphenyls and dibenzofurans were tested for their in vitro antibacterial activity against some Erwinia amylovora strains. The most efficient compound was 3,5-dihydroxybiphenyl (MIC = 115 μg/ml), the immediate product of biphenyl synthase which initiates phytoalexin biosynthesis.  相似文献

Axillary shoot producing cultures were obtained from microcuttings and shoot tips of Calluna vulgaris cv. H.E. Beale. For cultures derived from microcuttings the highest multiplication rate of 38 shoots (5 mm or longer) was obtained on a reduced salt medium with the addition of 0.5 mgl-1 2-isopentenyladenine (2iP) during an 8 week subculture. For shoot tip derived cultures 0.2 mgl-1 6-benzyladenine (BA) was the best cytokinin and led to a multiplication rate of 26 for a 6 week subculture. The addition of 1 g/l casein hydrolysate to a multiplication medium enhanced shoot proliferation in presence of 0.5 mgl-1 BA.Despite various auxin treatments shoots formed no roots in vitro but rooted readily if transferred to a peat substrate ex vitro. A high rooting percentage (80%) was also obtained with shoots taken from the end of a multiplication phase and rooted directly. An additional subculture on low auxin containing media before transfer to peat substrate is recommended because the shoot condition can be improved in this way. A high number of rooted plantlets was produced, so the methods described will allow mass propagation.  相似文献

In vitro flowering is an alternative breeding tool for generating hybrid Cucumis spp. as it is able to overcome limitations caused by interspecific incompatibility. The present study describes an efficient method for induction of multiple shoots and in vitro flowering from shoot tip explants of cucumber (Cucumis sativus L.). Shoot tip explants were excised from 7-day-old seedlings and cultured on Murashige and Skoog (MS) medium fortified with different concentrations of 6-benzylaminopurine (BAP; 0.5–2.5 mg/L) alone or in combination with 0.5 mg/L kinetin (KIN). The highest frequency (93.1%) of multiple shoot formation with maximum number of shoots (15.2 shoots/explant) was achieved on MS medium supplemented with 1.0 mg/L BAP. For in vitro flowering, shoots were cultured on MS medium supplemented with 0.5 mg/L BAP and different concentrations of sucrose. Flowering occurred on about 95% of in vitro shoots cultured on MS medium fortified with 6% (w/v) sucrose and 0.5 mg/L BAP after 15 d. For rooting, shoots (>2 cm) were cultured on MS medium augmented with various concentrations of indole-3-butyric acid (IBA; 0.5–2.5 mg/L) alone or in combination with 0.5 mg/L KIN. Among the combinations tested, supplementation with IBA (1.5 mg/L) and KIN (0.5 mg/L) induced maximum rooting rates (95.4%) with 7.8 roots/shoot. Rooted plantlets were successfully transferred into plastic cups containing a mixture of soil and sand (1:1), established in the greenhouse, and subsequently acclimatized in the field. The in vitro flowering reported in this study may facilitate rapid hybridization in Cucumis species and offers a model system for studying the physiological mechanisms involved in flowering.  相似文献

Two rare anthocyanins, cyanidin 3-(6-malonylglucoside)-7,3′-di(6-sinapylglucoside) and the demalonyl derivative, were characterised as the purple floral pigments of Dendrobium cv. ‘Pompadour’. Nine known flavonol glycosides were also identified, including the 3-rutinoside-7-glucosides of kaempferol and quercetin. One new glycoside was detected: the ferulyl ester of quercetin 7-rutinoside-7-glucoside. These flavonoid patterns are typical for plants in the family Orchidaceae.  相似文献

The effects of humic substances on in vitro culture of Golden Delicious apple are reported. Potassium humate (KH) when used in proliferation showed a negative interaction with BA while it enhanced rooting when IBA was not present in the culture medium. In the presence of IBA, KH increased root number and reduced root growth. The highest concentration tested, 500 mg l-1, caused a drastic reduction in root system development. 50 mg l-1 KH hastened rooting and plants grew more rapidly when transferred to soil.  相似文献

Stem sections from shoot cultures maintained in vitro were used to produce transgenic plants of the potato, Solanum tuberosum L. cv. Russet Burbank. Stem internode pieces inoculated with Agrobacterium tumefaciens containing coat protein genes from potato virus X and potato virus Y, produced shoots with a frequency of 60% in the absence of selection and 10% on medium containing 100 mg/l kanamycin monosulfate. Regenerated shoots were assayed for kanamycin resistance by placing stem segments on callus induction medium containing an increased level of kanamycin. Of a total 255 regenerated shoots, 47 (18%) were kanamycin resistant. Of the kanamycin resistant shoots, 25 (53%) expressed the PVX or PVY coat protein genes as assayed by enzyme-linked immunosorbent assay or Western immunoblot analysis.  相似文献

Mass propagation of date palm through indirect somatic embryogenesis or organogenesis has attracted the interest of commercial producers. But, this technique still faces some problems that hindered the production of date palm plantlets in vitro. Tissue browning is one of the serious problems that reduce callus growth and shoot regeneration. So the objective of the present study is to investigate the effect of cold pretreatment on callus growth, shoot regeneration, and polyphenol oxidase (PPO) activity during the callus culture. Results showed that a high survival rate of callus cultures (100%) were obtained when cultures were incubated in low temperature (cold treatment) for 45 and 75?days. On the other hand, total amount on phenolic compounds was also reduced to 0.47 and 0.53?mg GAE/g after same period of incubation (45 and 75?days respectively) at low temperature. In additional, our results showed that the highest frequency of shoot formation (66.67 and 73.34, %) and the highest shoot numbers (7.8 and 8.6 shoots/100?mg) were obtained from callus treated with low temperature for 45 and 75?days, respectively.  相似文献

There are more than nine thousand cultivars of Hibiscus rosa-sinensis L., with a series of flowers with shapes, colors and new cultivars continues as generated through both traditional and modern breeding techniques. In this study, advanced biotech methods of in vitro culture have been used to identify a technique for the efficient mass multiplication of H. rosa-sinensis ‘White Butterfly’, using phenyl urea, N-(2-Chloro-4-pyridyl)-N′-phenylurea (4-CPPU). For the first time, the effects of 4-CPPU for stimulating axillary shoot proliferation and multiple shoot regenerations from nodal explants were evaluated, and the optimal nutrient media deduced. From the diverse concentrations as 0.1, 0.5, 2.5, 5.0 & 10.0 µM of 4-CPPU, the highest frequency of shoots was recorded at 2.5 µM supplied in Murashige and Skoog (MS, pH-5.8) medium. After eight-weeks of culture, on an average of 6.7 shoot were obtained on this media with shoot heights of 4.2 cm from each explant. With the involvement of 0.5 µM-IBA (indole-3-butyric acid) in MS medium the regenerated shoots were rooted and followed by successful acclimation to ex vitro conditions. The ploidy consistency among the micro-plants was analyzed using flow cytometry and compared with ex vitro grown plants. No differences in the ploidy levels were observed among the 4-CPPU induced plants, when compared with the donor plants.  相似文献

The effect of irrigation intervals was studied on physiological, morphological, and antioxidant traits of two marigold (Calendula officinalis L.) cultivars in Karaj, Iran, in a split-plot experiment based on a randomized complete block design with three replications. The experimental treatments included irrigation at three levels of I1 (irrigation interval of 3 days), I2 (irrigation interval of 5 days), and I3 (irrigation interval of 7 days) as the main plot and cultivar at two levels of V1 (cv. ‘sparse petal’) and V2 (cv. ‘compact petal’) as the sub-plot. The results based on the comparison of the means showed that the increase in irrigation interval from 3 to 7 days decreased the leaf area index, crop growth rate, relative growth rate, and net assimilation rate by 73.53, 85.76, 93.47, and 94.81%, respectively. It also decreased the flower yield, plant height, flower number, and leaf number by 71.92, 41.84, 99.31, and 58.67%, respectively. The interaction between irrigation and cultivar revealed that I3V1 had the highest total phenol content (3013.59 g gallic acid 100 g?1 tissue) and antioxidant capacity (60.8%). It can be inferred that the treatment of I1 and cv. ‘compact petal’ give the best results for flower yield and physiological and morphological traits, and the treatment of I3 and both cultivars provide the best results for antioxidant traits in the climatic conditions of the Karaj region.

Changes in net photosynthetic rate (PN), stomatal conductance (gs), intercellular CO2 concentrations (Ci), transpiration rate (E) and water use efficiency (WUE) were measured in Plantago major L. plants grown under sufficient soil water supply or under soil water stress conditions. The plants had high PN in a wide range of soil water potential and temperature regimes. Soil water had little effect on PN under ambient CO2 concentrations, which was explained by a high carboxylation rate, but increased the dark respiration rate. Carboxylation activity at low Ci depended on RuBP regeneration, whereas at high Ci it depended on the phosphate regeneration rate. The gs and E values were low in plants under stress as compared to the controls that resulted in an increase of WUE. The results obtained show that Plantago major plants have different ways of adaptation to soil water deficit conditions.  相似文献

The chlorosis susceptible Vitis vinifera L. cv. Pinot blanc was grafted on two hybrid rootstocks with different iron efficiency, as follows: V. Berlandieri × V. rupestris 140 Ru (iron-efficient) and V. riparia × V. rupestris 101-14 (iron-inefficient). The grafted vines were grown in pots of a calcareous and a non-calcareous soil. The shoot growth was periodically checked and leaves, selected at two different times (at the middle of the annual growing period), were assayed for total chlorophyll, ferrous iron, ash alkalinity, percentage of dry matter and chlorosis score. At the end of the growing cycle the roots were oven-dried and weighed. The most significant findings of the trial were: (a) the soil strongly affected the shoot growth, with canes about twice as long in the non-calcareous soil; (b) the iron-efficient rootstock (140 Ru) did not induce chlorosis when growing on the calcareous soil, while the opposite occurred with the iron-inefficient rootstock (101=14); and (c) a high ash alkalinity occurred in light chlorotic leaves compared to green ones, under the same iron concentration.  相似文献

In two hydroponic experiments it was found that phosphorus induced necrotic leaf symptoms and reduced the growth of Grevillea cv. Poorinda Firebird, a member of the Proteaceae family in which a number of species are known to be affected by phosphorus toxicity. The addition of high levels of calcium was found to aggravate these effects while high nitrogen or potassium levels alleviated them. Tissue analysis of plants receiving high levels of phosphorus showed a tenfold increase in leaf phosphorus compared with plants receiving no phosphorus.
