Polychlorinated biphenyls (PCBs) are ubiquitous in the environment. Midway Atoll, located in the North Pacific Ocean, was occupied by the military during and after World War II. However, Midway Atoll has become a national wildlife refuge and home to many different seabirds today, including the black-footed albatross (Diomedea nigripes) (BFAL). The profiles and toxic equivalents (TEQ) of PCB congeners in the plasma and preen oil of BFAL chicks and adults were determined in this study. The concentrations of the total PCBs in the plasma samples of chicks and adults collected in Midway Atoll ranged from 2.3 to 223.8 (mean 80.1) and 22.8 to 504.5 (mean 158.6) ng g-1 (wet weight, ww), respectively. The TEQs ranged from 0.2 to 0.6 (mean 0.4) and 0.4 to 1.6 (mean 0.9) pg g-1 ww, respectively, in the plasma samples of chicks and adults from Midway Atoll. The major congeners in the plasma samples of chicks and adults included PCBs 31, 87, 97, 99, 118, 138, 153, and 180, accounting for 70% of the total PCBs. The concentrations of the total PCBs in the adult preen oil samples ranged from 1693 to 39404 (mean 10122) ng g-1 (ww), of which 97% were PCBs 105, 118, 128, 138, 153, 161, 172, and 183.  相似文献

Intermedin/adrenomedullin-2 (IMD) is a member of the adrenomedullin/CGRP peptide family. Less is known about the distribution of IMD than for other family members within the mammalian cardiovascular system, particularly in humans. The aim was to evaluate plasma IMD levels in healthy subjects and patients with chronic heart failure. IMD and its precursor fragments, preproIMD25–56 and preproIMD57–92, were measured by radioimmunoassay in 75 healthy subjects and levels of IMD were also compared to those of adrenomedullin (AM) and mid-region proadrenomedullin45–92 (MRproAM45–92) in 19 patients with systolic heart failure (LVEF < 45%). In healthy subjects, plasma levels (mean + SE) of IMD (6.3 + 0.6 pg ml−1) were lower than, but correlated with those of AM (25.8 + 1.8 pg ml−1; r = 0.49, p < 0.001). Plasma preproIMD25–56 (39.6 + 3.1 pg ml−1), preproIMD57–92 (25.9 + 3.8 pg ml−1) and MRproAM45–92 (200.2 + 6.7 pg ml−1) were greater than their respective bioactive peptides. IMD levels correlated positively with BMI but not age, and were elevated in heart failure (9.8 + 1.3 pg ml−1, p < 0.05), similarly to MRproAM45–92 (329.5 + 41.9 pg ml−1, p < 0.001) and AM (56.8 + 10.9 pg ml−1, p < 0.01). IMD levels were greater in heart failure patients with concomitant renal impairment (11.3 + 1.8 pg ml−1) than those without (6.5 + 1.0 pg ml−1; p < 0.05). IMD and AM were greater in patients receiving submaximal compared with maximal heart failure drug therapy and were decreased after 6 months of cardiac resynchronization therapy. In conclusion, IMD is present in the plasma of healthy subjects less abundantly than AM, but is similarly correlated weakly with BMI. IMD levels are elevated in heart failure, especially with concomitant renal impairment, and tend to be reduced by high intensity drug or pacing therapy.  相似文献

Concentrations of 28 polychlorinated biphenyls (PCBs) were assessed in soils from the Midway Atoll in the central North Pacific Ocean. The analytical procedure involved the application of accelerated solvent extraction (ASE) and gas chromatography coupled with ion trap mass spectrometric detection (GC/ITMS) for identification and quantification. Among the 28 PCB congeners studied, 26 of them, except CB195 and CB209, were detected in the analyzed samples at different frequencies. The total concentrations of 28 indicator PCBs (ΣPCBs) ranged from 2.6 to 148.8 ng g−1 with an average value of 50.7 ng g−1 and median of 39.5 ng g−1. Sources and congeners’ pattern of PCB were investigated in the soil of Midway Atoll. The principal component analysis indicated that the compositions of PCBs in most of the soil samples were similar. The total concentrations of PCBs were used to assess the cancer risk probabilities in humans via ingestion, dermal contact and inhalation of soil particles. Very low cancer risk was found in all soil samples caused by ΣPCBs.  相似文献

Experiments were conducted to determine the optimum requirements of non-phytin phosphorus (NPP) in commercial broilers and White Leghorn layers. Five levels of NPP (2.5, 3.0, 3.5, 4.0 and 4.5 g kg−1 diet) were tested to assess the NPP requirement of commercial broilers (3–30 days of age) fed maize–soya diets containing 10 g Ca kg−1. Each level of NPP was fed to quadruplicate groups of ten chicks each. Inclusion of graded levels of NPP significantly (P < 0.01) influenced body weight gain, feed intake, tibia ash content, phosphorus content in serum, tibia ash and phosphorus retention. The predicted NPP requirements for body weight gain, P content in serum and tibia ash were 4.4, 4.48 and 4.1 g kg−1 diet, respectively. The NPP requirement for tibia ash was the highest (7.4 g kg−1 diet). Similarly, four levels of NPP (2.0, 2.5, 3.0 and 3.5 g kg−1 diet) were tested with maize–soya diets containing 35 g Ca kg−1 for White Leghorn layers (266–350 days of age). Each diet was tested on four groups of 12 hens in each. Egg production was not influenced by the variation in dietary NPP levels. The predicted NPP requirements for better egg weight and shell thickness were 2.6 and 2.4 g kg−1 diet, respectively, while for the serum inorganic P level the value was 3.42 g kg−1 diet. Therefore, it can be concluded that commercial broilers need about 4.4 g NPP kg−1 diet for better performance, whereas, White Leghorn layers need not more than 2.0 g NPP kg−1 diet for better egg production. However, layers require 2.6 g NPP kg−1 diet to produce eggs with better egg size and shell quality.  相似文献

The dynamics of Dinophysis acuminata and its associated diarrhetic shellfish poisoning (DSP) toxins, okadaic acid (OA) and dinophysistoxin-1 (DTX1) as well as pectenotoxins (PTXs), were investigated within plankton and shellfish in Northport Bay, NY, USA, over a four year period (2008–2011). Over the course of the study, Dinophysis bloom densities ranged from ~104 to 106 cells L−1 and exceeded 106 L−1 in 2011 when levels of total OA, total DTX1, and PTX in the water column were 188, 86, and 2900 pg mL−1, respectively, with the majority of the DSP toxins present as esters. These cell densities exceed – by two orders of magnitude – those previously reported within thousands of samples collected from NY waters from 1971 to 1986. The bloom species was positively identified as D. acuminata via scanning electron microscopy and genetic sequencing (cox1 gene). The cox1 gene sequence from the D. acuminata populations in Northport Bay was 100% identical to D. acuminata from Narragansett Bay, RI, USA and formed a strongly supported phylogenetic cluster (posterior probability = 1) that included D. acuminata and Dinophysis ovum from systems along the North Atlantic Ocean. Shellfish collected from Northport Bay during the 2011 bloom had DSP toxin levels (1245 ng g−1 total OA congeners) far exceeding the USFDA action level (160 ng g−1 total OA of shellfish tissue) representing the first such occurrence on the East Coast of the U.S. D. acuminata blooms co-occurred with paralytic shellfish poisoning (PSP) causing blooms of Alexandrium fundyense during late spring each year of the study. D. acuminata cell abundances were significantly correlated with levels of total phytoplankton biomass and Mesodinium spp., suggesting food web interactions may influence the dynamics of these blooms. Given that little is known regarding the combined effects of DSP and PSP toxins on human health and the concurrent accumulation and depuration of these toxins in shellfish, these blooms represent a novel managerial challenge.  相似文献

Earthquake‐generated tsunamis threaten coastal areas and low‐lying islands with sudden flooding. Although human hazards and infrastructure damage have been well documented for tsunamis in recent decades, the effects on wildlife communities rarely have been quantified. We describe a tsunami that hit the world's largest remaining tropical seabird rookery and estimate the effects of sudden flooding on 23 bird species nesting on Pacific islands more than 3,800 km from the epicenter. We used global positioning systems, tide gauge data, and satellite imagery to quantify characteristics of the Tōhoku earthquake‐generated tsunami (11 March 2011) and its inundation extent across four Hawaiian Islands. We estimated short‐term effects of sudden flooding to bird communities using spatially explicit data from Midway Atoll and Laysan Island, Hawai'i. We describe variation in species vulnerability based on breeding phenology, nesting habitat, and life history traits. The tsunami inundated 21%–100% of each island's area at Midway Atoll and Laysan Island. Procellariformes (albatrosses and petrels) chick and egg losses exceeded 258,500 at Midway Atoll while albatross chick losses at Laysan Island exceeded 21,400. The tsunami struck at night and during the peak of nesting for 14 colonial seabird species. Strongly philopatric Procellariformes were vulnerable to the tsunami. Nonmigratory, endemic, endangered Laysan Teal (Anas laysanensis) were sensitive to ecosystem effects such as habitat changes and carcass‐initiated epizootics of avian botulism, and its populations declined approximately 40% on both atolls post‐tsunami. Catastrophic flooding of Pacific islands occurs periodically not only from tsunamis, but also from storm surge and rainfall; with sea‐level rise, the frequency of sudden flooding events will likely increase. As invasive predators occupy habitat on higher elevation Hawaiian Islands and globally important avian populations are concentrated on low‐lying islands, additional conservation strategies may be warranted to increase resilience of island biodiversity encountering tsunamis and rising sea levels.  相似文献

A new toxin-producing marine diatom, Nitzschia bizertensis sp. nov., isolated from the Bizerte Lagoon (Tunisia, Southwest Mediterranean Sea) is, based on studies on eight different strains, characterized morphologically by light microscopy, transmission and scanning electron microscopy, and phylogenetically using the nuclear rDNA regions: SSU, ITS1, 5.8S, ITS2 and D1–D3 of the LSU. The species belongs to the sections Lanceolatae or Lineares as defined by Cleve and Grunow (1880). These sections are characterized by species having linear-lanceolate valves with an eccentric raphe where the fibulae does not extend into the valve, and are otherwise famous for the lack of characters useful for delineation of species. Nitzschia bizertensis differs from most other species in these sections by having a high density of interstriae. The morphological and phylogenetic studies and comparisons with previously described Nitzschia species showed Nitzschia bizertensis sp. nov. to be a new species. Batch culture experiments were conducted for estimations of maximum growth rate and production of domoic acid (DA). Maximum cellular DA content of the examined strains ranged from 2 × 10−4 to 3.6 × 10−2 pg cells−1. The total DA concentration (pg mL−1) was high already in exponential growth phase maybe due to reinoculation of “old” stationary phase cells, and increased into stationary growth phase where it reached a stationary level varying among the strains from ca. 4500 to 9500 pg mL−1. Nitzschia bizertensis represents a new domoic acid-producing diatom and is the second toxin producing Nitzschia species. The resolution of Nitzschia bizertensis and Nitzschia navis-varingica in different parts of the LSU phylogenetic tree, and the recovery of the Pseudo-nitzschia species phylogenetically distant from those two species suggests that the ability to produce DA either evolved multiple times independently or was lost multiple times.  相似文献

A new bioprocess for the synthesis of lactosucrose was studied using a covalently immobilized β-galactosidase on macrospheres of chitosan. The effects of temperature and pH on the production of lactosucrose and other oligosaccharides were evaluated. At 30 °C and pH 7.0, the maximum concentration of lactosucrose reached to 79 g L−1. The change of the reaction conditions allowed to modify the qualitative profile of the final products without quantitative change in the total of oligosaccharides produced. At pH 7 and 30 °C, products profile was 79 g L−1 of lactosucrose, 37 g L−1 of galactooligosaccharides and 250 g L−1 of total oligosaccharides, while at pH 5 and 64 °C the concentrations for the same compounds were 40, 62 and 250 g L−1, respectively. The immobilization increased the thermal stability up to 260-fold. Using 300 g L−1 of sucrose and 300 g L−1 of lactose, and 8.5 mg of chitosan mL−1, 30 cycles of reuse were performed and the biocatalyst kept the maximal lactosucrose synthesis. These results fulfill some important aspects for the enzyme immobilization and oligosaccharides synthesis: the simplicity of the protocols, the high operational stability of the enzyme and the possibility of driving the final products.  相似文献

In order to evaluate the potential effects of rest grazing on organic carbon storage on the Stipa baicalensis steppe in Inner Mongolia, compared the S. baicalensis steppes after rest grazing for 3 years, 6 years, and 9 years, using potassium dichromate heating method, this study analyzed the organic carbon storage of plant and soil in the steppes among different periods of rest grazing. The results indicated that as the rest grazing years prolonged, the biomass included above-ground parts, litters and underground plant parts(roots) of the plant communities all increased, meanwhile the carbon content of the biomass increased with the rest grazing years prolonged. For the zero rest grazing (RG0) steppe and the steppes after a rest grazing of 3 years (RG3a), 6 years (RG6a), 9 years (RG9a), the carbon storage in above-ground parts of plant communities were 42.60 g C/m2, 66.33 g C/m2, 83.46 g C/m2, 100.29 g C/m2 respectively; the carbon storage of litters were 7.85 g C/m2, 9.12 g C/m2, 9.18 g C/m2, 11.54 g C/m2 separately; the carbon storage of underground plant parts (0–100 cm) were 281.40 g C/m2, 576.38 g C/m2, 745.33 g C/m2, 1279.61 g C/m2 respectively; and the carbon storage in 0–100 cm soil were 22991.14 g C/m2, 24687.75 g C/m2, 26564.86 g C/m2,33041.55 g C/m2. The results suggested that as the rest grazing years prolonged, the organic carbon storage in plant communities and soil increased. The carbon storage of underground plant parts and soil organic carbon mainly concentrated in 0–40 cm soil. After rest grazing for 3 years, 6 years, and 9 years, the increased soil organic carbon were as the 81.14%, 85.84%, and 89.46% of the total increased carbon; From the perspective of carbon sequestration cost, the total cost of RG3a, RG6a and RG9a were 2903.40 RMB/hm2, 5806.80 RMB/hm2, and 8710.20 RMB/hm2. The cost reduced with the extension of rest grazing years, 0.17 RMB/kg C, 0.16 RMB/kg C, 0.09 RMB/kg C for RG3a, RG6a and RG9a respectively. From the growth characteristics of grassland plants, the spring was one of the two avoid grazing periods, timely rest grazing could effectively restore and update grassland vegetation, and was beneficial to the sustainable use of grassland. From the available data, the organic carbon storage of RG9a was the highest, while the cost of carbon sequestration was the lowest. Therefore, spring rest grazing should be encouraged to continue for it was proved to be a very efficient grassland use measures.  相似文献

A high spatial resolution sampling of Alexandrium pacificum cysts, along with sediment characteristics (% H2O, % organic matter (OM), granulometry), vegetative cell abundance and environmental factors were investigated at 123 study stations in Bizerte Lagoon (Tunisia). Morphological examination and ribotyping of cells obtained from a culture called ABZ1 obtained from a cyst isolated in lagoon sediment confirmed that the species was A. pacificum. The toxin profile from the ABZ1 culture harvested during exponential growth phase was simple and composed of the N-sulfocarbamoyl toxins C1 (9.82 pg toxin cell−1), the GTX6 (3.26 pg toxin cell−1) and the carbamoyl toxin Neo-STX (0.38 pg toxin cell−1). The latter represented only 2.8% of the total toxins in this strain.High abundance of A. pacificum cysts correlated with enhanced percentages of water and organic matter in the sediment. In addition, sediment fractions of less than 63 μm were examined as a favorable potential seedbed for initiation of future blooms and outbreaks of A. pacificum in the lagoon. A significant difference in the cyst distribution pattern was recorded among the lagoon's different zones, with the higher cyst abundance occurring in the inner waters. Also, no correlation due to the specific hydrodynamics of the lagoon was observed in the spatial distribution of A. pacificum cysts and vegetative cells.  相似文献

Protocorm cultures of Dendrobium candidum were established in balloon type bubble bioreactors using Murashige and Skoog (MS) medium with 0.5 mg l−1 α-naphthaleneacetic acid (NAA), 2.5% (w/v) sucrose, 5:25 mM NH4:NO3 and 1% (v/v) banana homogenate for the production of biomass and bioactive compounds. In 3 l bioreactor containing 2 l medium, a maximum protocorm biomass (21.0 g l−1 dry biomass) and also optimum quantities of total polysaccharides (389.3 mg g−1 DW), coumarins (18.0 mg g−1 DW), polyphenolics (11.9 mg g−1 DW), and flavonoids (4.5 mg g−1 DW) were achieved after 7 weeks of culture. Based on these studies, 5 and 10 l bioreactor cultures were established to harvest 80 g and 160 g dry biomass. In 10 l bioreactors, the protocorms grown were accumulated with optimal levels of polysaccharides (424.1 mg g−1 DW), coumarins (15.8 mg g−1 DW), polyphenols (9.03 mg g−1 DW) and flavonoids (4.7 mg g−1 DW). The bioreactor technology developed here will be useful for the production of important bioactive compounds from D. candidum.  相似文献

We studied for two years the seasonal changes in plant available nitrate and ammonium nitrogen (N), nitrification, N-mineralization, microbial biomass carbon (MBC), nitrogen (MBN) and phosphorus (MBP) in two forest and three cropland sites, derived from a tropical forest ecosystem of India. Results indicated that seasonal values of nitrate N, ammonium N and phosphate P ranged from 7.33–12.99, 5.1–10.22 and 4.0–7.8 μg g?1 in forest and 4.13–9.26, 9.35–14.46 and 2.8–5.8 μg g?1 in cropland ecosystems, respectively, with maximum values in summer and minimum in rainy seasons. Nitrification and N-mineralization values varied from 6–28 and 4–26 μg g?1 mo?1 in forest and 3–14 μg g?1 mo?1 and 4–17 μg g?1 mo?1 in cropland, with maximum values in rainy season and minimum in summer season.MBC, MBN MBP ranged from 393–753, 34–80 and 16–36 μg g?1 in forests and 186–414, 21–41 and 11–22 μg g?1 in croplands, being maximum in summer and minimum in rainy seasons. There was gradual increase in the values of inorganic N, nitrification, N-mineralization and MBC, MBN and MBP along the age of cropland. Analysis of variance indicated significant difference in the concentration of inorganic N, nitrification and N-mineralization and MBC, MBN and MBP due to sites and seasons.Cultivation caused decline in the mean annual organic C, N and P by 42%, 29% and 13%. The values of nitrate N were decreased by 23–38%, while ammonium N was increased by 39–74%. Nitrification and N-mineralization values were reduced by 39–63% and 40–60%, respectively. Microbial C, N and P were reduced by 44–54%, 41–50% and 28–44%, respectively. Nonetheless, the contribution of soil microbial biomass reflected in total N was enhanced from 4.76% in forest to 5.03% in cropland ecosystem. Enhancement of plant available ammonium-N and microbial contribution in total N are an indicator of natural conserving mechanism to check the nitrogen loss from the nutrient poor agro-ecosystem.  相似文献

We investigated the abundance and biomass of planktonic ciliates in the sea area around Zhangzi Island, Northern Yellow Sea, from July 2009 to June 2010. Ciliates were sampled monthly from surface to bottom with a 10 m depth interval at 13 sample stations along three transects. A 1 L sample of water from each depth was collected with a 2.5 L Niskin water sampler and fixed in 1% acid Lugol’s iodine solution. Water samples were pre-concentrated using the Utermöhl method and observed using an Olympus IX51 inverted microscope at 100× or 200x. The dimensions of the ciliates were measured and the cell volume of each species was estimated using appropriate geometric shapes. The carbon:volume ratio used to calculate biomass was 0.19 pg C/μm3. Abundance and biomass of the ciliate in water column were calculated as the integral of the abundance and biomass from bottom to surface, respectively. The classification of tintinnids was based on taxonomic literature. The average abundance of non-loricate ciliates was 3066 ± 2805 ind/L, ranging from 165 ind/L (50 m depth of St. B6 in July) to 26,595 ind/L (surface of St. C1 in September). The average biomass of non-loricate ciliates was 2.88 ± 2.68 μg C/L, ranging from 0.05 μg C/L (10 m depth of St. A6 in July) to 20.51 μg C/L (surface of St. A5 in August). The average tintinnid abundance was 142 ± 273 ind/L, ranging from 0 ind/L (monthly) to 2756 ind/L (surface of St. A1 in July). The average tintinnid biomass was 0.84 ± 2.19 μg C/L, ranging from 0.00 μg C/L (every month) to 37.64 μg C/L (20 m depth of St. C5 in July). The results showed that the average abundance of total ciliates was 3208 ± 2828 ind/L, ranging from 166 ind/L (10 m depth of St. A6 in July) to 26,625 ind/L (surface of St. C1 in September); the average biomass of total ciliates was 3.73 ± 3.55 μg C/L, ranging from 0.05 μg C/L (10 m depth of St. A6 in July) to 38.29 μg C/L (20 m depth of St. C5 in July). Abundance and biomass were vertically homogeneous in February, November and December, but decreased dramatically from the surface down to the bottom in other months. 23 tintinnid species were identified, 12 of which were in genus Tintinnopsis. Tintinnid species were more abundant in February, July and August. Tintinnids occupied 6.6 ± 10.2% and 19.7 ± 23.3% of the total ciliate abundance and biomass, respectively, which increased during the warm season and at coastal stations, and decreased during the cold season and at offshore stations. Large non-loricate ciliate species were prevalent in spring, while smaller species dominated in summer and autumn. The average abundance of total ciliates in water column was 132 ± 72 × 106 ind/m2, with increases during spring and autumn. The average biomass of total ciliates in water column was 152.57 ± 93.10 mg C/m2, with increases during spring and summer. The average abundance and biomass of total ciliates in water column were greater at offshore stations than at coastal stations during spring and autumn, and were lower during summer and winter. Non-loricate ciliates, tintinnids and total ciliates showed significant positive correlation with temperature and significant negative correlation with salinity (p < 0.01). Non-loricate ciliates and total ciliates showed significant positive correlation with Chl a concentration (p < 0.01); however, relationship between Chl a concentration and tintinnids was not significant.  相似文献

Denitrification beds are a cost-effective technology for removing nitrate from point source discharge. To date, field trials and operational beds have primarily used wood media as the carbon source; however, the use of alternative more labile carbon media could provide for increased removal rate, lower installation costs and reduced bed size. While previous laboratory experiments have investigated the potential of alternative carbon sources, these studies were typically of short duration and small scale and did not necessarily provide reliable information for denitrification bed design purposes. To address this issue, we compared nitrate removal, hydraulic and nutrient leaching characteristics of nine different carbon substrates in 0.2 m3 barrels, at 14 and 23.5 °C over a 23-month period. Mean nitrate removal rates for the period 10–23 months were 19.8 and 15 g N m?3 d?1 (maize cobs), 7.8 and 10.5 g N m?3 d?1 (green waste), 5.8 and 7.8 g N m?3 d?1 (wheat straw), 3.0 and 4.9 g N m?3 d?1 (softwood), and 3.3 and 4.4 g N m?3 d?1 (hardwood) for the 14 and 23.5 °C treatments, respectively. Maize cobs provided a 3–6.5-fold increase in nitrate removal over wood media, without prohibitive decrease in hydraulic conductivity, but had higher rates of nutrient leaching at start-up. Significant difference in removal rate occurred between the 14 and 23.5 °C treatments, with the mean Q10 temperature coefficient = 1.6 for all media types in the period 10–23 months.  相似文献

A study was carried out in Coastal Kenya to evaluate the effect of supplementing Napier grass (Pennisetum purpureum) based diet with increasing level of Madras thorn (Pithecellobium dulce) on feed intake, digestibility and live weight changes of growing goats. Fifteen small East African goats 6 months old on average were randomly allocated to five treatments. Napier grass was either offered alone (control) or supplemented with 7.5, 15, 22.5 and 30 g DM/kg W0.75 of Madras thorn. Supplementing with Madras thorn up to 22.5 g DM/kg W0.75 had no (P > 0.05) significant effect on the intake of the basal diet, however there was a 29% depression in the intake of the basal diet at 30 g DM/kg W0.75 level of supplementation. A (P < 0.05) increase significant in the TDMI from 242 for the control to 258, 302, 357 and 458 g/kg DM, was recorded for 7.5, 15, 22.5 and 30 g DM/kg W0.75, respectively. Supplementation also resulted in increase (P < 0.05) in DM and OM digestibility. Feed N, N retained and loss increased linearly with increase in supplementation. Positive N balance was recorded for all goats 0.2, 1.4, 2.5, 3.2 and 3.6 g/day for control, 7.5, 15, 22.5 and 30 g DM/kg W0.75, respectively. Goats fed the control diet lost a mean of 8 g BW/day while those supplemented with 7.5, 15, 22.5 and 30 g DM/kg W0.75 gained 8, 23, 43 and 44 g/day, respectively. The use of Madras thorn forage as a protein supplement for goats could be an affordable source, especially to the resource-constrained farmers in the tropics.  相似文献

The uptake of paralytic shellfish poisoning (PSP) toxins and spirolides by the paddle crab (Ovalipes catharus) was investigated in two laboratory feeding trials using Greenshell? mussels (Perna canaliculus), which had been fed toxic strains of either Alexandrium catenella or A. ostenfeldii, as a vector. Toxin uptake by crabs occurred in both feeding trials and was limited to the visceral tissue; no toxins were detected in the body meat or the gills. The first trial utilized a strain of A. catenella that had high total PSP toxin content, 442.3 ± 91.6 fmol/cell, that was dominated by low toxicity N-sulfocarbamoyl toxins resulting in a low cellular toxicity, 5.5 ± 1.6 pg STXequiv./cell. In this trial, toxin accumulation in the crabs was highly variable and ranged from 3.8 to 221.5 μg STXequiv./100 g, with 3/4 of the crabs exceeding the regulatory limit of 80 μg STXequiv./100 g. Eight days after feeding on toxic mussels the crabs still retained high levels of toxin suggesting that depuration rates in this species may be slow. In the second feeding trial, the A. ostenfeldii strain fed to mussels produced low levels of both PSP toxins (52.0 ± 19.5 fmol/cell; 1.4 ± 0.3 pg STXequiv./cell) and spirolides (1.8 pg/cell) and, as a result, the concentration transferred to crabs via the mussels was very low-PSP toxins ranged from 2.5 to 6.8 μg STXequiv./100 g and spirolides from 6 to 7 μg/kg. The results of our study demonstrate that paddle crabs are capable of acquiring both PSP toxins and spirolides and suggest that this may occur in the wild during a toxic shellfish event. It also highlights the need to remove the viscera before consumption.  相似文献

The study present evaluated the levels of mercury (Hg) and methylmercury (MeHg) in hair samples of people from Barreiras community, riverside inhabitants of the Tapajós River (Pará, Brazil), an area impacted by clandestine gold mining, as well as we analyzed the levels of Hg and Se (selenium) in nine fish species (carnivores and non-carnivorous) from the Tapajós River, which stand out as the main species consumed by riverside inhabitants, to evaluate a relationship between frequency of fish consumption and Hg concentration, and also to evaluate possible mechanisms of fish protection (or non-protection) to Hg exposure by Se. Furthermore we analyze the water quality to evaluate the environmental trophic state, fact responsible by creating conditions that can potentiate the effects of toxic mercury. Concentrations of Hg and MeHg were analyzed in hair samples of 141 volunteers in different age band. Of those, 84.40% of samples present values above the threshold for biological tolerance, which is 6.00 μg g−1 of total Hg in hair. Total Hg, in men there was a variation of 2.07–24.93 μg g−1, while for women the variation was 4.84–27.02 μg g−1. Consequently, the level of MeHg in men presented a variation of 1.49–19.57 μg g−1, with an average of 11.68 μg g−1, while with women the variation was from 3.73 to 22.35 μg g−1, with an average of 10.38 μg g−1. In fish species, Hg concentrations in carnivorous species had an average of 0.66 μg g−1, higher than that permitted by current legislation, ranging from 0.30 to 0.98 μg g−1, while the non-carnivorous species have values below the recommended by the legislation averaging 0.09 μg g−1, ranging between 0.02 and 0.44 μg g−1. For Se in fish, show that among carnivores, the contents of Se ranged between 0.18 and 0.54 μg g−1 with a mean of 0.34 μg g−1, while for non-carnivores these values were of the order of 0.16–0.56 μg g−1, with an average of 0.32 μg g−1. In surface water quality variables at the sampling points all showed values in accordance with the range established by current legislation. In this regard, the results provided by this study, while not conclusive, are strong indicators that despite not having been shown the relationship between the concentration of mercury in hair and feeding habits along the Tapajós River basin communities showed that a plausible correlation exists between levels of mercury and selenium in fish. This fact may serve as a subsidy to research human health, because in the Amazon, there is still a lot to examine with regards to the full understanding of the Se cycle.  相似文献

Continuous anaerobic fermentations were performed in a novel external-recycle, biofilm reactor using d-glucose and CO2 as carbon substrates. Succinic acid (SA) yields were found to be an increasing function of glucose consumption with the succinic acid to acetic acid ratio increasing from 2.4 g g−1 at a glucose consumption of 10 g L−1, to 5.7 g g−1 at a glucose consumption of 50 g L−1. The formic acid to acetic acid ratio decreased from an equimolar value (0.77 g g−1) at a glucose consumption of 10 g L−1 to a value close to zero at 50 g L−1. The highest SA yield on glucose and highest SA titre obtained were 0.91 g g−1 and 48.5 g L−1 respectively. Metabolic flux analysis based on the established C3 and C4 metabolic pathways of Actinobacillus succinogenes revealed that the increase in the succinate to acetate ratio could not be attributed to the decrease in formic acid and that an additional source of NADH was present. The fraction of unaccounted NADH increased with glucose consumption, suggesting that additional reducing power is present in the medium or is provided by the activation of an alternative metabolic pathway.  相似文献

Rhamnolipids are surface-active molecules produced by Pseudomonas aeruginosa as congener mixtures. They are considered “green” alternatives to synthetic surfactants used in industrial, remediation and pharmaceutical applications. Optimizing yield as well as controlling congener distribution are necessary steps for successful commercialization of rhamnolipids. This study used a mixture of glucose and fatty acids of different chain length (C12–C22) and saturation (C18:1 and C18:2) to produce monorhamnolipids and determine the effect of fatty acid substrates on rhamnolipid yield, percent carbon conversion and congener distribution. Results show that 1% glucose + 0.25% stearic acid (C18) produced the greatest yield (2.1 g L−1) compared to other glucose–fatty acid combinations (0.8–1.8 g L−1). Various glucose + C18 ratios were then tested to optimize yield and percent substrate carbon conversion to monorhamnolipid. Results revealed a positive linear correlation between the mass percent of C18 used and the percent carbon conversion. A mass percent of 67% C18 was optimal resulting in a 44% carbon conversion and a yield of 13.7 g L−1 monorhamnolipid. For all fatty acid substrates tested, the RhaC10C10 was the most abundant and RhaC10C12:1 was the least abundant of the four major congeners produced. However, the relative amount of RhaC10C8 and RhaC10C12 congeners was dependent on several factors: in general, fatty acid substrates with relatively short chain length (C12 and C14), unsaturated fatty acid substrate (C18:2), and longer cultivation time resulted in a higher RhaC10C8/RhaC10C12 ratio. Studies were conducted with crossbred weanling pigs to determine the level of phosphorus needed to be fed when a maize–soyabean meal–whey diet was supplemented with exogenous phytase (Natuphos™). In Trial 1, phytase was added at 1200 phytase units (PTU) kg−1 as phosphorus decreased. The control diet in Phase I (0–14 days) contained 7.3 g kg−1 phosphorus and in Phase II (14–28 days) contained 6.5 g kg−1 phosphorus. Dietary phosphorus was calculated to decrease by 0.8, 1.6 or 2.4 g kg−1 when phytase was supplemented. Chromic oxide was added for estimation of apparent absorption of phosphorus. Performance was optimum when 5.7 and 4.8 g kg−1 phosphorus (analysed levels) were fed with 1200 PTU kg−1 phytase in Phases I and II, respectively. The lowest dietary phosphorus levels did not reduce performance for the overall 28-day period. Apparent phosphorus digestibility was increased by phytase in Phase I when 5.7 g kg−1 phosphorus was fed compared to the control diet and in Phase II when 6.0 g kg−1 phosphorus was fed with phytase. Faecal phosphorus excretion decreased in both phases as dietary phosphorus decreased. Faecal phosphorus excretion was minimized at the lowest phosphorus level with no decrease in performance. The estimated requirement for dietary phosphorus, as determined by the NLIN procedure, is 5.0 g kg−1 in Phase I and 4.3 g kg−1 in Phase II when 1200 PTU kg−1 is used. In Trial 2, phytase was supplemented at 500 PTU kg−1 when phosphorus was decreased in the diet. The control diet contained 6.6 and 6.0 g kg−1 phosphorus in Phases I and II, respectively, and phosphorus was calculated to decrease by 0.5, 1.0, 1.5, or 2.0 g kg−1 when phytase was added. Daily gain decreased when 5.0 g kg−1 phosphorus was fed in Phase I and when 4.6 or 4.2 g kg−1 (analysed levels) phosphorus was fed in Phase II with 500 PTU kg−1. Faecal phosphorus excretion decreased as dietary phosphorus decreased, but there were no treatment effects on apparent phosphorus digestibility. The dietary phosphorus requirement was estimated to be 5.7 and 5.0 g kg−1 in Phases I and II, respectively, when phytase is fed at 500 PTU kg−1. At the present recommendation of 500 PTU kg−1 in starter feed, phosphorus can be decreased by 0.10 g kg−1. However, higher levels of phytase are needed to actually increase apparent phosphorus digestibility.  相似文献
