Addition of ammonia to a suspension of photosynthesizing isolated mesophyll cells from P. somniferum quantitatively alters the pattern of carbon metabolism by increasing rates of certain key ratelimiting steps leading to amino-acid synthesis and by decreasing rates of rate-limiting steps in alternative biosynthetic pathways. Of particular importance is the stimulation of reactions mediated by pyruvate kinase and phosphoenolpyruvate carboxylase. The increased rates of these two reactions, which result in an increased flow of carbon into the tricarboxylic-acid cycle, correlate with a rapid rise in glutamine (via glutamine synthetase) which draws carbon off the tricarboxylic-acid cycle as -ketoglutarate. Increased flux of carbon in this direction appears to come mainly at the expense of sucrose synthesis. The net effect of addition of ammonia to mesophyll cells is thus a redistribution of newly fixed carbon away from carbohydrates and into amino acids.  相似文献

Rhodopseudomonas globiformis strain 7950 grew with a variety of amino acids, urea, or N2 as sole nitrogen sources. Cultures grown on N2 reduced acetylene to ethylene; this activity was absent from cells grown on nonlimiting NH 4 + . Glutamate dehydrogenase could not be detected in extracts of cells of strain 7950, although low levels of an alanine dehydrogenase were present. Growth ofR. globiformis on NH 4 + was severely inhibited by the glutamate analogue and glutamine synthetase inhibitor, methionine sulfoximine. High levels of glutamine synthetase (as measured in the -glutamyl transferase assay) were observed in cell extracts of strain 7950 regardless of the nitrogen source, although N2 and amino acid grown cells contained somewhat higher glutamine synthetase contents than cells grown on excess NH 4 + . Levels of glutamate synthase inR. globiformis were consistent with that reported from other phototrophic bacteria. Both glutamate synthase and alanine dehydrogenase were linked to NADH as coenzyme. Individual 14C-labelled amino acids are rapidly removed from dilute solution in artificial sea water (0.2 mol 1–1) by suspensions of Meliosira medocris. The rate of disappearance of radioactivity corresponds closely to removal of primary amines as determined by measurement of the rate of decrease of fluorescamine-positive material. Net removal of naturally occurring free amino acids from the sea water habitat from which the alga was isolated is demonstrated using high performance liquid chromatography. Removal of amino acids from natural sources makes a significant contribution to the carbon requirements of the alga as well as supplying significant amounts of amino nitrogen.  相似文献

Four strains of the green sulfur bacterium Chlorobium were studied in respect to nitrogen nutrition and nitrogen fixation. All strains grew on ammonia, N2, or glutamine as sole nitrogen sources; certain strains also grew on other amino acids. Acetylene-reducing activity was detectable in all strains grown on N2 or on amino acids (except for glutamine). In N2 grown Chlorobium thiosulfatophilum strain 8327 1 mM ammonia served to switch-off nitrogenase activity, but the effect of ammonia was much less dramatic in glutamate or limiting ammonia grown cells. The glutamine synthetase inhibitor methionine sulfoximine inhibited ammonia switch-off in all but one strain. Cell extracts of glutamate grown strain 8327 reduced acetylene and required Mg2+ and dithionite, but not Mn2+, for activity. Partially purified preparations of Rhodospirillum rubrum nitrogenase reductase (iron protein) activating enzyme slightly stimulated acetylene reduction in extracts of strain 8327, but no evidence for an indigenous Chlorobium activating enzyme was obtained. The results suggest that certain Chlorobium strains are fairly versatile in their nitrogen nutrition and that at least in vivo, nitrogenase activity in green bacteria is controlled by ammonia in a fashion similar to that described in nonsulfur purple bacteria and in Chromatium.Non-common abbreviations MSX Methionine sulfoximine - MOPS 3-(N-morpholino) propane sulfonic acid This paper is dedicated to Professor Norbert Pfennig on the occasion of his 60th birthday  相似文献

Summary The free amino acid pools in the nitrogen-fixing blue-green algae Anabaena cylindrica, A. flos-aquae and Westiellopsis prolifica contain a variety of amino acids with aspartic acid, glutamic acid and the amide glutamine being present in much higher concentrations than the others. This pattern is characteristic of that found in organisms having glutamine synthetage/glutamate synthetase [glutamine amide-2-oxoglutarate amino transferase (oxido-reductase)] as an important pathway of ammonia incorporation. Under nitrogen-starved conditions the level of acetylene reduction (nitrogen fixation) and the glutamine pool both increase but the free ammonia pool decreases, suggesting that ammonia rather than glutamine regulates nitrogen fixation.Glutamine synthetase has been demonstrated in Anabaena cylindrica using the -glutamyl transferase assay and also using a biosynthetic assay in which Pi release from ATP during glutamine synthesis was measured. The enzyme (-glutamyl transferase assay) is present in nitrogen-fixing cultures and activity is higher in aerobic than in microaerophilic cultures. Ammonium-grown cultures have lowest levels of all and activity in the presence of nitrate-nitrogen (150 mg nitrogen 1-1) is lower than in aerobic cultures growing on elemental nitrogen. Ammonium-nitrogen and nitrate-nitrogen have no effect on glutamine synthetase in vitro. Glutamate synthetase also operates in nitrogen-fixing cultures of Anabaena cylindrica.  相似文献

Suspension cultures of Chinese hamster cells (line CHO), which had stopped dividing and were arrested in G1 following growth to high cell concentrations in F-10 medium, could be induced to reinitiate DNA synthesis and to divide in synchrony upon addition of the appropriate amounts of isoleucine and glutamine. Both amino acids were required to initiate resumption of cell-cycle traverse. Deficiencies in other amino acids contained in F-10 medium did not result in accumulation of cells in G1, indicating a specific action produced by limiting quantities of isoleucine and glutamine. In the presence of sufficient glutamine, approximately 2 x 10-6 M isoleucine was required for all cells to initiate DNA synthesis in a population initially containing 1.5 x 105 cells/ml. Under similar conditions, about 4 x 10-6 M isoleucine was required for all G1-arrested cells to progress through cell division. In contrast, 1 x 10-4 M glutamine was necessary for maximum initiation of DNA synthesis in G1 cells, along with sufficient isoleucine. A technique for rapid production of G1-arrested cells is described in which cells from an exponentially growing population placed in F-10 medium deficient in both isoleucine and glutamine or isoleucine alone accumulated in G1 after 30 hr.  相似文献

Summary The seasonal changes in the pattern of 21 amino acids occurring in the xylem vessels of Populus twigs have been studied in connection to the mobilization of protein bodies in ray parenchyma cells at the electron microscopic level. Hydrolysis of protein bodies in spring and movement of amino acids into vessels are found to be closely linked. Comprising more than 75% of total amino acid content, glutamine (Gln) is by far the dominant N-constituent of the sap. Gln reaches up to 11 mol ml-1 in the spring sap while other amino acids only show 1/20 to 1/100 of this amount. From the measured Gln accumulation rates in the vessels in nature and in the vessels of isolated shoots, a minimum flux rate for Gln of 5.6 pmol cm-2 min-1 is calculated for the ray contact cell/vessel interface. Furthermore, because Gln constitutes 75% of the amino acid content of the sap but only 1.3% of the amino acids in the 32 kDa storage protein of the ray cells in the wood (Clausen and Apel 1991), it becomes evident that most amino acids originating from protein body mobilization do not enter the vessels but are used for Gln synthesis preceding Gln release into the vessels.  相似文献

Yellow lupin seeds (Lupinus luteus) cytoplasmic tRNAGlu was isolated and the primary structure was determined to be: pUCCGUUGUAGUCAGDDGGDCAGGAUAUUCGGCUCUCACCCGAAAGACm5CCGGGTCAm1 AGU CCCGGCGACGGAACCAOH. It is 76 nucleotides long and contains 8 modified nucleosides: 2 residues of pseudouridine, ribothymidine, 3 dihydrouridines, 5-methylcytosine and 1-methyladenosine. The primary nitrogen metabolism of the N2-fixing root nodule symbiosis Alnus incana (L.)–Frankia was investigated by 31P and 15N nuclear magnetic resonance (NMR) spectroscopy. Perfusion of root nodules in a pulse–chase approach with 15N- or 14N-labeled NH4+ revealed the presence of the amino acids alanine (Ala), -amino butyric acid, glutamine (Gln), glutamic acid (Glu), citrulline (Cit) and arginine (Arg). Labeling kinetics of the Gln amide-N and -amino acids suggested that the glutamine synthetase (GS; EC 6.3.1.2)–glutamate synthase (GOGAT; EC 1.4.1.13) pathway was active. Inhibition of the GS-catalyzed reaction by methionine sulphoximine abolished incorporation of 15N. Cit was labeled in all three N positions but most rapidly in the position, consistent with carbamoyl phosphate as the precursor to which Gln could be the amino donor catalyzed by carbamoyl phosphate synthase (CPS; EC 6.3.5.5). Ala biosynthesis occurred consistent with a flux of N in the sequence Gln–Glu–Ala. 31P NMR spectroscopy in vivo and of extracts revealed several metabolites and was used in connection with the 15N pulse–chase experiment to assess general metabolic status. Stable concentrations of ATP and UDP-glucose during extended perfusions showed that the overall root nodule metabolism appeared undisturbed throughout the experiments. The metabolic pathways suggested by the NMR results were confirmed by high activities of the enzymes GS, NADH-GOGAT and ornithine carbamoyltransferase (OCT; EC 2.1.3.3). We conclude that the primary pathway of NH4+ assimilation in A. incana root nodules occurs through the GS–GOGAT pathway. The effect of copper on the uptake of nitrogen and the tissue contents of inorganic nitrogen, amino acids and proteins were studied in cooper-sensitive Silene vulgaris (Moench) Garcke, grown at different nitrogen sources (NH4 + or NO3 -). All the toxic copper levels tested, i.e. 4, 8, 16 M Cu2+, strongly inhibited the uptake of nitrogen, especially of NO3 -, and decreased the content of NO3 -, amino acids and proteins. Especially at 4 and 8 M Cu2+, NH4 + accumulated in the plants, suggesting that the conversion of NH4 - into amino acids was inhibited.  相似文献

Abstract: CO2 fixation was measured in cultured astrocytes isolated from neonatal rat brain to test the hypothesis that the activity of pyruvate carboxylase influences the rate of de novo glutamate and glutamine synthesis in astrocytes. Astrocytes were incubated with 14CO2 and the incorporation of 14C into medium or cell extract products was determined. After chromatographic separation of 14C-labelled products, the fractions of 14C cycled back to pyruvate, incorporated into citric acid cycle intermediates, and converted to the amino acids glutamate and glutamine were determined as a function of increasing pyruvate carboxylase flux. The consequences of increasing pyruvate, bicarbonate, and ammonia were investigated. Increasing extracellular pyruvate from 0 to 5 mM increased pyruvate carboxylase flux as observed by increases in the 14C incorporated into pyruvate and citric acid cycle intermediates, but incorporation into glutamate and glutamine, although relatively high at low pyruvate levels, did not increase as pyruvate carboxylase flux increased. Increasing added bicarbonate from 15 to 25 mM almost doubled CO2 fixation. When 25 mM bicarbonate plus 0.5 mM pyruvate increased pyruvate carboxylase flux to approximately the same extent as 15 mM bicarbonate plus 5 mM pyruvate, the rate of appearance of [14C]glutamate and glutamine was higher with the lower level of pyruvate. The conclusion was drawn that, in addition to stimulating pyruvate carboxylase, added pyruvate (but not added bicarbonate) increases alanine aminotransferase flux in the direction of glutamate utilization, thereby decreasing glutamate as pyruvate + glutamate →α-ketoglutarate + alanine. In contrast to previous in vivo studies, the addition of ammonia (0.1 and 5 mM) had no effect on net 14CO2 fixation, but did alter the distribution of 14C-labelled products by decreasing glutamate and increasing glutamine. Rather unexpectedly, ammonia did not increase the sum of glutamate plus glutamine (mass amounts or 14C incorporation). Low rates of conversion of α-[14C]ketoglutarate to [14C]glutamate, even in the presence of excess added ammonia, suggested that reductive amination of α-ketoglutarate is inactive under conditions studied in these cultured astrocytes. We conclude that pyruvate carboxylase is required for de novo synthesis of glutamate plus glutamine, but that conversion of α-ketoglutarate to glutamate may frequently be the rate-limiting step in this process of glutamate synthesis.  相似文献

—The uptake of l [14C]glutamine by a crude isolated nerve ending fraction of rat brain was found to be linear with time for at least 5 min, profoundly temperature-dependent, apparently half-saturated at a substrate concentration of 0·26 mm , partially inhibited by dinitrophenol and ouabain and elevated [K+], weakly Na+-dependent, poorly inhibited by drugs which block uptake of biogenic amines and more strongly inhibited by glutamic acid (IC50= 0·5mm ) than by aspartic acid, GABA, glycine or methionine. The [14C]glutamine taken up appeared to be associated with nerve endings and was released by membrane-disruption; about 20 per cent was associated with free mitochondria. Glutamine, δ-aminolevulinic acid and several other amino acids were poor inhibitors of [3H]GABA-uptake; δ-aminolevulinic acid was a poor inhibitor of [3H]glutamine-uptake, whereas glutamine was a moderately effective competitive inhibitor (Ki= 1 mm ). [14C]glutamine and [3H]GABA were released from brain slices by electrical stimulation or 50 mm K+, while labeled δ-aminolevulinic acid, leucine, urea, amphetamine and tyramine were poorly released. [14C]glutamine was not released by unlabeled glutamate or several aromatic amines. We conclude that the neuropsychiatric features of porphyria are not likely due to a ‘false transmitter’ role for δ-aminolevulinic acid although such a role for glutamine in hepatic encephalopathy or other neuropsychiatric diseases should be considered.  相似文献

Summary A chloramphenicol concentration of 3 mg per ml inhibits uptake of 14C-labelled phenylalanine, lysine, and adenine by Chlorella cells. Incorporation into both the free pool and the TCA insoluble fraction is inhibited. The inhibition is not related to inhibition of protein synthesis since cycloheximide (a specific inhibitor of protein synthesis in Chlorella) does not inhibit uptake of the 14C-labelled amino acids. Uptake of 14C-uracil is not inhibited by chloramphenicol.Both chloramphenicol and 2.4-dinitrophenol stimulate endogenous respiration of Chlorella, but whereas the latter reduces the internal concentration of ATP, the former (in concentrations of 1–3 mg/ml) stimulates it about two-fold. Similar concentrations of chloramphenicol decreases slightly the concentration of ADP, and it is therefore suggested that in Chlorella, chloramphenicol concentrations of 1–3 mg per ml inhibit some energy-linked reactions by preventing ATP utilization.  相似文献

The synthesis of 5-aminolevulinic acid commences with the ligation of glutamate to a specific tRNAGlu by a glutamyl-tRNA synthetase (E.C. 6.1.1.17) (Huang et al., 1984, Science 225, 1482–1484). The synthetase from the yellow pigment mutant C-2A of the unicellular green alga Scenedesmus obliquus was purified by sequential column chromatography on Sephacryl S-300, Blue Sepharose, phosphocellulose P11 and by fast protein liquid chromatography (FPLC) on Mono Q. After denaturing sodium dodecylsulfate (SDS)-gel electrophoresis the purified enzyme preparation revealed a single protein band with a molecular mass of 55 kDa, proving the apparent homogeneity of the glutamyl-tRNA synthetase. A molecular mass of 105 ± 10 kDa was determined for the native protein by chromatography on Sephadex G-150. From these data it can be concluded that the glutamyl-tRNA synthetase from S. obliquus is a homodimer. The purified protein is active within a pH range from 7.0 to 9.0 with a maximum activity at pH 8.0. Changes in extracellular and intracellular free amino acids were followed during cyclic phases of N2-fixation (acetylene reduction) by cultures of the axenic, non-heterocystous cyanobacterium Gloeothece incubated under alternating light and darkness or continuous illumination. Changes in intracellular amino acids were minor, with only arginine (increasing during N2-fixation) and glutamate (decreasing during fixation) showing significant changes in cells incubated under 12 h light: 12 h dark. The intracellular concentration of glutamine in cultures was always very low and the value of the ratio glutamine: glutamate (GLN:GLU), used as an index of C–N status in eukaryote microbes, was consistently less than 0.05 suggesting that the cells were nitrogen-stressed. On addition of ammonium, there was a transient accumulation of intracellular glutamine, and the ratio GLN:GLU increased rapidly to a value greater than 0.5, typical of unstressed eukaryotes. In contrast to intracellular amino acids, there were significant changes in extracellular amino acids in cultures incubated under alternating light and darkness. Glycine, serine and alanine were released during the dark phase and were taken up again in the light, paralleling the diurnal pattern of nitrogenase activity (high in darkness). It is postulated that this release is usually retained in the mucilage surrounding the cells (but disturbed during even gentle filtration) and that this mucilage may constitute an extracellular vacuole.  相似文献

The mechanism of nitrate uptake for assimilation in procaryotes is not known. We used the radioactive isotope, 13N as NO3 -, to study this process in a prevalent soil bacterium, Pseudomonas fluorescens. Cultures grown on ammonium sulfate or ammonium nitrate failed to take up labeled nitrate, indicating ammonium repressed synthesis of the assimilatory enzymes. Cultures grown on nitrite or under ammonium limitation had measurable nitrate reductase activity, indicating that the assimilatory enzymes need not be induced by nitrate. In cultures with an active nitrate reductase, the form of 13N internally was ammonium and amino acids; the amino acid labeling pattern indicated that 13NO3 - was assimilated via glutamine synthetase and glutamate synthase. Cultures grown on tungstate to inactivate the reductase concentrated NO3 - at least sixfold. Chlorate had no effect on nitrate transport or assimilation, nor on reduction in cell-free extracts. Ammonium inhibited nitrate uptake in cells with and without active nitrate reductases, but had no effect on cell-free nitrate reduction, indicating the site of inhibition was nitrate transport into the cytoplasm. Nitrate assimilation in cells grown on nitrate and nitrate uptake into cells grown with tungstate on nitrite both followed Michaelis-Menten kinetics with similar K mvalues, 7 M. Both azide and cyanide inhibited nitrate assimilation. Our findings suggest that Pseudomonas fluorescens can take up nitrate via active transport and that nitrate assimilation is both inhibited and repressed by ammonium.  相似文献

Intact pea (Pisum sativum L.) cotyledons were incubated with [14C]glucosamine at several stages of seed development and the resultant radioactive proteins were analysed by gel electrophoresis combined with immunoaffinity chromatography and sucrose gradient fractionation. Glucosamine was incorporated into at least five vicilin polypeptides (approx. molecular weight 70,000; 50,000, two components; 14,000, two components). No incorporation was detected into the subunits of legumin. Tunicamycin at 50 g/ml largely inhibited glucosamine incorporation but had little effect on the incorporation of 14C-labelled amino acids into cotyledon proteins, including vicilin. The assembly of vicilin polypeptides into full-sized protein oligomers (7–9 S) was also unaffected by tunicamycin. Chromatography on concanavalin A confirmed that glycosylation of cotyledon proteins was inhibited by tunicamycin. Investigations of the uptake of ammonium (NH 4 + ) by Rhodopseudomonas capsulata B100 supported the presence of an NH 4 + transport system. Experimentally NH 4 + was determined by electrode or indophenol assay and saturation kinetics were observed with two apparent K m's of 1.7 M and 11.1 M (pH 6.8, 30°) and a V max at saturation of 50–60 nmol/min·mg protein. The optimum pH and temperature were 7.0 and 33° C, respectively. The Q10 quotient was calculated to be 1.9 at 100 M NH 4 + , indicating enzymatic involvement. In contrast to the wild type, B100, excretion of NH 4 + , not uptake, was observed in a glutamine auxotroph, R. capsulata G29, which is derepressed for nitrogenase and lacks glutamine synthetase activity. G29R1, a revertant of G29, also took up NH 4 + at the same rate as wild type and had fully restored glutamine synthetase activity. Partially restored derivatives, G29R5 and G29R6, grew more slowly than wild type on NH 4 + as the nitrogen source, remained derepressed for nitrogenase in the presence of NH 4 + , and displayed rates of NH 4 + uptake in proportion to their glutamine synthetase activity. Ammonium uptake and glutamine synthetase activity were also restored in R. capsulata G29 exconjugants which had received the plasmid pPS25, containing the R. capsulata glutamine synthetase structural gene. Summary Brush border membrane vesicles (BBMV) were prepared from the gills of the marine mussel,Mytilus edulis. These membranes contained two distinct pathways for cotransport of Na+ and -neutral amino acids. The major pathway in mussel gill BBMV was the alanine-lysine (AK) pathway, which had a high affinity for alanine and for the cationic amino acid, lysine. The AK pathway was inhibited by nonpolar -neutral amino acids and cationic amino acids, but was not affected by -neutral amino acids or imino acids. The kinetics of lysine transport were consistent with a single saturable process, with aJ max of 550 pmol/mg-min and aK t of 5 m. The AK pathway did not have a strict requirement for Na+, and concentrative transport of lysine was seen in the presence of inwardly directed gradients of Li+ and K+, as well as Na+. Harmaline inhibited the transport of lysine in solutions containing either Na+ or K+. The alanine-proline (AP) pathway transported both alanine and proline in mussel gill BBMV. The AP pathway was strongly inhibited by nonpolar -neutral amino acids, proline, and -(methylamino)isobutyric acid (Me-AIB). The kinetics of proline transport were described by a single saturable process, with aJ max of 180 pmol/mg-min andK t of 4 m. In contrast to the AK pathway, the AP pathway appeared to have a strict requirement for Na+. Na+-activation experiments with lysine and proline revealed sigmoid kinetics, indicating that multiple Na+ ions are involved in the transport of these substrates. The transport of both lysine and proline was affected by membrane potential in a manner consistent with electrogenic transport.  相似文献

  • 1.1. In eels captured in Roskilde Fjord in 1972 and 1975, a specifically enhanced synthesis was found from 14C-acetate of 14C-labelled mono-unsaturated fatty acids (C16:1 and C18:1) relative to saturated fatty acids (C16:0 and C18:0) in sea water 4 days after irradiation (10 Gy, 60Co).
  • 2.2. Corresponding experiments in 1976 and 1982 showed rather the opposite: irradiation resulted in more 14C-labelled saturated fatty acids relative to unsaturated fatty acids, both in fresh and sea water.
  • 3.3. The latter effect was less marked than that in 1972 and 1975, but still statistically clearly significant.
