The objective of this study was to examine the role of antioxidant enzymes in waterlogging tolerance of pigeonpea (Cajanus cajan L. Halls) genotypes ICP 301 (tolerant) and Pusa 207 (susceptible). Waterlogging resulted in visible yellowing and senescence of leaves, decrease in leaf area, dry matter, relative water content and chlorophyll content in leaves, and membrane stability index in roots and leaves. The decline in all parameters was greater in Pusa 207 than ICP 301. Oxidative stress in the form of superoxide radical, hydrogen peroxide and thiobarbituric acid reactive substances (TBARS) contents initially decreased, however at 4 and 6 d of waterlogging it increased over control plants, probably due to activation of DPI-sensitive NADPH-oxidase. Antioxidant enzymes such as superoxide dismutase, ascorbate peroxidase, glutathione reductase and catalase also increased under waterlogging. The comparatively greater antioxidant enzyme activities resulting in less oxidative stress in ICP 301 could be one of the factor determining its higher tolerance to flooding as compared to Pusa 207. This study is the first to conclusively prove that waterlogging induced increase in ROS is via NADPH oxidase.  相似文献

Intact chloroplasts isolated from sulphur dioxide fumigatedHardwickia binata leaves showed inhibition of PS II electron transport activity without any significant effect on photosystem I. Sulphur dioxide exposed leaves accumulated more hydrogen peroxide than those from non-fumigated plants and this was caused by increase in superoxide radical production. Hydrogen peroxide formation was inhibited by addition of cytochrome C and superoxide disrnutase. In sulphur dioxide fumigated leaves, increase in superoxide dismutase activity showed resistance to sulphite toxicity. The localization of ascorbate peroxidase, glutathione reductase and dehydroascorbate reductase activities in chloroplasts provide evidence for the photogeneration of ascorbate. The scavenging of hydrogen peroxide in chloroplast due to ascorbate regenerated from DHA by the system: PS I → Fd → NADP → glutathione. We investigated the effect of water stress on yield and quality of tomato plants overexpressing Solanum lycopersicum thylakoid-bound ascorbate peroxidase gene (StAPX). APX activity, hydrogen peroxide content, net photosynthetic rate of tomato leaves, and yield and nutrition quality of tomato fruits were measured under soil moisture 70, 60, and 50 % of full field capacity. Results show that the capability of APX for scavenging hydrogen peroxide induced by water stress was higher in the transgenic than the wild type (WT) plants. The yield of fruits of the transgenic tomato plants was higher than that of WT plants under water stress and the fruit nutrition quality was not different. These results indicate that overexpression of StAPX might improve water stress tolerance in the transgenic tomato plants.  相似文献

Sesbania virgata (Leguminosae) is tolerant of long periods of soil inundation. However, its morphological adaptations to anoxia and its response to possible damage from oxidative stress are still unknown. Here, we provide new information that helps to explain the ability of S. virgata plants to grow in flooded environments. Plants containing six expanded leaves were placed in masonry tanks and were subjected to the following conditions: control (well watered), soil waterlogging (water to the setup level of 1 cm above the soil surface—roots and parts of the stems flooded), and complete submergence (whole plant flooded). Plants exposed to flooding (soil waterlogging and complete submergence) significantly increased their production of hydrogen peroxide (H2O2), indicating the extent of oxidative injury posed by stress conditions. We demonstrate that plants exposed to flooding develop an efficient scavenger of ROS (generated during stress) in the roots through the coordinated action of nonenzymatic ascorbic acid (Asc) and dehydroascorbate (DHA) as well as the enzymatic antioxidants superoxide dismutase (SOD), ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR), and glutathione reductase (GR) that are present in the tissues. Moreover, we observed the development of morpho-anatomical structures such as adventitious roots, lenticels, and cracks in the stem of plants under soil waterlogging. The secondary root of plants under soil waterlogging showed a thinner cortex and larger number of elements of small diameter vessels. Numerous aerenchymas were observed in the newly formed in the adventitious roots. We conclude that these antioxidative responses and morpho-anatomical adaptations in the roots are part of a suite of adaptations that allow S. virgata plants to survive long periods of flooding, notably under waterlogged conditions.  相似文献

In this study, an aqueous extract of leaves from Melothria maderaspatana was tested for in vitro antioxidant activity. Free radical scavenging assays, such as hydroxyl radical, hydrogen peroxide, superoxide anion radical and 2,2-diphenyl-1-picryl hydrazyl (DPPH), 2,2’-azinobis-(3-ethyl-enzothiazoline-6-sulfonic acid) (ABTS) radical scavenging, and reducing power assay, were studied. The extract effectively scavenged hydroxyl radical, hydrogen peroxide and superoxide anion radicals. It also scavenged DPPH and ABTS radicals. Furthermore, it was found to have reducing power. All concentrations of leaf extract exhibited free radical scavenging and antioxidant power, and the preventive effects were in a dose-dependent manner. The antioxidant activities of the above were compared to standard antioxidants such as butylated hydroxytoluene (BHT), ascorbic acid, and α-tocopherol. The results obtained in the present study indicate that the M. maderaspatana extract could be considered a potential source of natural antioxidant.  相似文献

从杨桃果中提取得到三种提取物为匀浆提取物、蛋白提取物和多糖提取物。采用化学发光法测定这三种提取物清除氧自由基的活性,实验结果:匀浆提取物清除羟自由基(·OH)和H2O2的活性大小相近,而清除超氧阴离子自由基(O2–·)的活性较小,其IC50约为前两者的4倍。蛋白提取物清除O2–·和·OH的活性大小相近,而清除H2O2的活性明显小于前两者,IC50约为前两者的9倍。多糖提取物清除.OH的活性明显大于清除O2–·和H2O2的活性,其IC50约为O2–·的1/22,约为H2O2的1/65。结果表明,杨桃果具有清除O2–·、·OH和H2O2的作用,不同提取物对这些活性氧自由基的清除能力有所不同。  相似文献

Hydrogen peroxide generation rates of uninfected and infected leaves of two tomato (Lycopersicon esculentum) cultivars showing differential susceptibility to Botrytis cinerea were determined. The superoxide anion, hydroxyl radical, ascorbate contents and changes in NADH peroxidase, superoxide dismutase (SOD), ascorbate peroxidase (APX) and catalase (CAT) activities in the apoplast fraction were analysed. Infected leaves had an increased hydrogen peroxide level. It was greater and generally occurred earlier in plants of the less susceptible cv. Perkoz than in those of the more susceptible cv. Corindo. Induction of nitrotetrazolium blue reducing activity and SOD levels in apoplast were higher in cv. Perkoz 24 h after inoculation. In the controls, NADH peroxidase activity in apoplast was higher in the more susceptible cv. Corindo, but after infection it increased faster and to a higher level in the less susceptible cv. Perkoz. NADH oxidation was inhibited by only 15% by a specific inhibitor DPI (diphenylene‐iodonium) but was completely inhibited by KCN and NaN3. Similar increases in APX activity after 48 h and a small increase in catalase activities were observed in both cultivars soon after infection. These results indicate that resistance of tomato plants to infection by the necrotrophic fungus B. cinerea may result from early stimulation of hydrogen peroxide and superoxide radical generations by NADH peroxidase and SOD in apoplastic space, and they confirm the important role of their enhanced production in apoplastic spaces of plants.  相似文献

To elucidate proline antioxidant properties in common sage (Salvia officinalis L.) plants, they were treated with paraquat (a producer of superoxide radical) and/or NaCl and also with paraquat and proline at the stage of 4–5 true leaves. The paraquat solution (1 ml containing 0.1 μmol of the agent) was applied to the leaf surface; NaCl (200 mM) and proline (the final concentration of 5 mM) were added to nutrient medium. Experimental plants were firstly kept in darkness for 12 h, then illuminated, and in 3, 6, and 12 h, leaves and roots were fixed for biochemical analyses. The results obtained are in agreement with the supposition of proline antioxidant properties. In particular, it was established that paraquat induced a slight increase in the proline level in the leaves during dark period of plant growth and also during subsequent 3 h after light switching on. This transient proline accumulation in the leaves was accompanied by its level decrease in the roots. Proline addition to the nutrient medium of paraquat-treated plants neutralized paraquat damaging action on the leaves. In the presence of paraquat, proline treatment reduced the accumulation in the roots of hydrogen peroxide and malondialdehyde, the product of membrane lipid peroxidation. It also affected indirectly the activities of superoxide dismutase (SOD) and free, covalently bound, and ionically bound peroxidases. Keeping in mind that, in the presence of paraquat, superoxide-induced changes in SOD activity in the roots were negatively correlated with the level of proline, which content was the highest during the last hours of experiments, we can conclude that proline antioxidant effects are manifested only after 12 h of stressor action, whereas antioxidant enzymes are involved in ROS scavenging during the earlier stage of damaging factor action.  相似文献

Activated oxygen species such as superoxide radicals, singlet oxygen, hydrogen peroxide and hydroxyl radicals can be produced in plants exposed to low, non-freezing, non-injurious temperatures. To prevent or alleviate oxidative injury, plants have evolved several mechanisms which include scavenging by natural antioxidants and enzymatic antioxidant systems such as superoxide dismutases, catalase and peroxidases. Although overproduction of hydrogen peroxide and increased tolerance to oxidative stress can be induced in wheat by low-temperature treatments, data concerning changes in the enzymatic antioxidant systems are almost absent. With the aim to provide this information, antioxidant enzyme (superoxide dismutases, catalase and peroxidases) activities were analysed in leaves and roots of Triticum aestivum cvs Brasilia (frost resistant in field) and Eridano (less frost resistant in field) seedlings grown at day/night temperatures of 24/22°C (control treatment) and 12/5°C (low-temperature treatment). Our data showed that superoxide dismutase activities were unaffected by low-temperature treatment both in leaves and roots. Catalase activity in leaves and roots was decreased in 12/5°C-grown seedlings, but Brasilia maintained higher catalase activity than Eridano. Differences were also observed in guaiacol peroxidase activities between control and acclimated seedlings: Higher guaiacol peroxidase activities were found in the leaves of 12/5°C-grown seedlings while in roots these activities were lower. Moreover, Brasilia guaiacol peroxidase activities were higher than Eridano. Superoxide dismutase and peroxidase zymogram analyses showed that synthesis of new isoforms was not induced by low-temperature treatment. Changes in the activities of antioxidant enzymes induced by cold acclimation support the hypothesis that a frost-resistant wheat cultivar, in comparison with a less frost-resistant one, maintains a better defence against activated oxygen species during low-temperature treatment.  相似文献

Pigeonpea [Cajanus cajan (L.) Millsp.] is a waterlogging-sensitive legume crop. We studied the effect of waterlogging stress on hydrogen peroxide (H2O2) content, lipid peroxidation and antioxidant enzyme activities in two pigeonpea genotypes viz., ICPL-84023 (waterlogging resistant) and MAL-18 (waterlogging susceptible). In a pot experiment, waterlogging stress was imposed for 6 days at early vegetative stage (20 days after sowing). Waterlogging treatment significantly increased hydrogen peroxide accumulation and lipid peroxidation, which indicated the extent of oxidative injury posed by stress conditions. Enzyme activities of peroxidase (POX), catalase (CAT), ascorbate peroxidase (APX), superoxide dismutase (SOD) and polyphenol oxidase (PPO) increased in pigeonpea roots as a consequence of waterlogged conditions, and all the enzyme activities were significantly higher in waterlogged ICPL-84023 than in MAL-18. POX activity was the maximum immediately after imposing stress, therefore, it was suggested to be involved in early scavenging of H2O2, while rest of the enzymes (CAT, APX, SOD and PPO) were more important in late responses to waterlogging. Present study revealed that H2O2 content is directly related to lipid peroxidation leading to oxidative damage during waterlogging in pigeonpea. Higher antioxidant potential in ICPL-84023 as evidenced by enhanced POX, CAT, APX, SOD and PPO activities increased capacity for reactive oxygen species (ROS) scavenging and indicated relationship between waterlogging resistance and antioxidant defense system in pigeonpea.  相似文献

【背景】微生物在荒漠生态系统中经常面临多重胁迫,包括干旱、高温、UV辐射,这些环境胁迫使得荒漠土壤微生物极易在体内外积累大量的超氧离子或过氧化物,抑制其生长或者直接造成死亡。【目的】荒漠土壤细菌为适应荒漠环境表现出抗氧化特性,作为荒漠生态系统重要组成部分,对其抗氧化特性的研究为荒漠地区抗氧化资源的开发提供科学依据和技术基础,也对荒漠微生物抗氧化机制的挖掘奠定了基础。【方法】利用过氧化氢氧化筛选出两株具有强抗氧化性的荒漠土壤细菌:海床动性微菌AX6(PlanomicrobiumokeanokoitesAX6)和海洋考克氏菌KD4(Kocuriamarina KD4),通过测定其在过氧化氢条件下的生长曲线、细胞受损程度、抗氧化酶活性以及自由基清除能力,探究荒漠土壤微生物的抗氧化生理生化特征。【结果】两株细菌在低浓度过氧化氢中细胞丙二醛含量显著低于阴性对照大肠杆菌,在1.5mmol/L过氧化氢中菌株AX6的谷胱甘肽过氧化物酶活性可达108.33 U/mL,同时DPPH、超氧阴离子自由基的清除能力显著升高;此外,在3 mmol/L过氧化氢中菌株KD4的过氧化氢酶活性升高至1.16 U/mL,显著高于阳性对照耐辐射球菌,羟自由基的清除能力也显著升高。【结论】不同荒漠土壤细菌的活性抗氧化酶种类、自由基清除能力存在较大差异,表明荒漠土壤微生物抗氧化过程的多样性。  相似文献

The effects of supplemental ultraviolet-B (s-UV-B; 3.6 kJ m?2 day?1 above ambient) radiation on plant metabolite profile and free radical scavenging activities of Withania somnifera (an indigenous medicinal plant) under field conditions were investigated. The metabolite profiles of both leaves and roots were analyzed via GC–MS. The methanolic extracts were examined for their DPPH radical-, superoxide radical-, hydrogen peroxide-, hydroxyl radical-, and nitric oxide radical scavenging activities, ferrous ion chelating activity, and reducing power. GC–MS profile of leaves revealed increment in compounds such as isophytol (138.1 %), β-stigmasterol (183.05 %), trans-squalene (233.3 %), and Withaferin A (155.0 %), while compounds such as eugenol, β-carotene, lycopene, and vitamin E were detected in s-UV-B-treated leaves only. In roots, compounds such as ledol, neophytadiene, palmitic acid, retinol, sitosteryl oleate, and campesterol registered their presence only under s-UV-B. Methanolic extracts of treated plant organs were found to be more potent as free radical scavengers (their EC50 values being lower than those of control extracts). Anomalies were observed for nitric oxide radical scavenging in both leaves and roots. The present study indicates that s-UV-B alters the composition and contents of plant metabolites leading to an increase in their free radical scavenging activities. Hence, s-UV-B-treated plant organs might be more effective in combating oxidative stress as well as from a nutritional and health perspective.  相似文献

L-carnitine plays an important regulatory role in the mitochondrial transport of long-chain free fatty acids. In this study, the antioxidant activity of L-carnitine was investigated as in vitro. The antioxidant properties of the L-carnitine were evaluated by using different antioxidant assays such as 1, 1-diphenyl-2-picryl-hydrazyl free radical (DPPH.) scavenging, total antioxidant activity, reducing power, superoxide anion radical scavenging, hydrogen peroxide scavenging and metal chelating activities. Total antioxidant activity was measured according to ferric thiocyanate method. alpha-tocopherol and trolox, a water-soluble analogue of tocopherol, were used as the reference antioxidant compounds. At the concentrations of 15, 30 and 45 microg/mL, l-carnitine showed 94.6%, 95.4% and 97.1% inhibition on lipid peroxidation of linoleic acid emulsion, respectively. On the other hand, 45 microg/mL of standard antioxidant such as alpha-tocopherol and trolox indicated an inhibition of 88.8% and 86.2% on peroxidation of linoleic acid emulsion, respectively. In addition, L-carnitine had an effective DPPH. scavenging, superoxide anion radical scavenging, hydrogen peroxide scavenging, total reducing power and metal chelating on ferrous ions activities. Also, those various antioxidant activities were compared to alpha-tocopherol and trolox as references antioxidants.  相似文献

Parsley (Petroselinum hortense L.) plants cultivated under controlled conditions were exposed to different doses of cadmium to investigate the antioxidant capacity and cadmium accumulation in the samples. Two-months-old parsley seedlings were treated with four different concentrations of CdCl2 (0, 75, 150, and 300 μM) for fifteen days. Cadmium level in leaves increased significantly by increasing the Cd contamination in the soil. Total chlorophyll and carotenoid content declined considerably with Cd concentration. Cd treatment caused a significant increase lipid peroxidation in tissue of leaf. Superoxide dismutase activity (SOD, EC 1.15.1.1) increased partially at 75 and 150 μM CdCl2 concentrations whereas the activity decreased at 300 μM CdCl2. Catalase (CAT, EC 1.11.1.6) and ascorbate peroxidase (APX, EC 1.11.1.11) activities were reduced by Cd application. Total phenolic compound amount increased significantly with increasing Cd concentration, as ferric reduction power, superoxide anion radical, and DPPH˙ free radical scavenging activities elevated slightly by the concentration. These results suggest that antioxidant enzymes activities were repressed depending on accumulation of cadmium in leaves of parsley while the non-enzymatic antioxidant activities slightly increased.  相似文献

Effects of flooding on the activities of some enzymes of activated oxygen metabolism, the levels of antioxidants, and lipid peroxidation in senescing leaves of tobacco were investigated. As judged by the decrease in chlorophyll and protein levels, flooding accelerated the senescence of tobacco leaves. Total peroxide and the lipid peroxidation product, malondialdehyde, increased in both control and flooding-treated leaves with increasing duration of the experiment. Throughout the duration of the experiment, flooded leaves had higher levels of total peroxide and malondialdehyde than did control leaves. Flooding resulted in an increase in peroxidase and ascorbate peroxidase activities and a reduction of superoxide dismutase activity in the senescing leaves. Glycolate oxidase, catalase, and glutathione reductase activities were not affected by flooding. Flooding increased the levels of total ascorbate and dehydroascorbate. Total glutathione, reduced form glutathione, or oxidized glutathione levels in flooded leaves were lower than in control leaves during the first two days of the experiment, but were higher than in control leaves at the later stage of the experiment. Our work suggests that senescence of tobacco induced by flooding may be a consequence of lipid peroxidation possibly controlled by superoxide dismutase activity. We investigated the impact of drought and arbuscular mycorrhizal (AM) fungi on the morphological structure and physiological function of shoots and roots of male and female seedlings of the dioecious plant Populus cathayana Rehder. Pot-grown seedlings were subjected to well watered or water-limiting conditions (drought) and were grown in soil that was either inoculated or not inoculated with the AM fungus Rhizophagus intraradices. No significant differences were found in the infection rates between the two sexes. Drought decreased root and shoot growth, biomass and root morphological characteristics, whereas superoxide radical (O2–) and hydrogen peroxide content, peroxidase (POD) activity, malondialdehyde (MDA) concentration and proline content were significantly enhanced in both sexes. Male plants that formed an AM fungal symbiosis showed a significant increase in shoot and root morphological growth, increased proline content of leaves and roots, and increased POD activity in roots under both watering regimes; however, MDA concentration in the roots decreased. By contrast, AM fungi either had no effect or a slight negative effect on the shoot and root growth of female plants, with lower root biomass, total biomass and root/shoot ration under drought. In females, MDA concentration increased in leaves and roots under both watering regimes, and the proline content and POD activity of roots increased under drought conditions; however, POD activity significantly decreased under well-watered conditions. These findings suggest that AM fungi enhanced the tolerance of male plants to drought by improving shoot and root growth, biomass and the antioxidant system. Further investigation is needed to unravel the complex effects of AM fungi on the growth and antioxidant system of female plants.  相似文献

In order to examine whether polyamines (PAs) modify the functioning of the scavenging system and oxidative stress levels in water-stressed plants, cucumber (Cucumis sativus L.) seedlings were treated with spermidine (Spd) prior to dehydration, and stress-evoked changes in superoxide dismutase (SOD) (EC 1.15.1.1), catalase (EC 1.11.1.6), guaiacol peroxidase (EC 1.11.1.7) activities, H(2)O(2) and superoxide radical levels were determined. Free PA content during Spd treatment and during the stress period were also determined. Exogenous application of Spd differentially influenced enzymes of the antioxidative system under stress conditions; we observed an increase of guaiacol peroxidase activity, and, to a lesser degree, a reduction of SOD and catalase activities in Spd-treated plants in comparison to untreated stressed plants. Hydrogen peroxide and superoxide radical contents were also reduced in stressed plants after Spd pretreatment. These positive effects were observed in the case of 1mM Spd concentration. A higher concentration (3mM) influenced negative, more significant stress-induced changes, but a lower concentration (0.1mM) had a very limited effect. In summary, PAs are able to moderate the activities of scavenging system enzymes and to influence oxidative stress intensity.  相似文献

The aim of the study was to implicate the generation of reactive oxygen species (ROS) and altered cellular redox environment with the effects of Cu-deficiency or Cu-excess in mulberry (Morus alba L.) cv. Kanva 2 plants. A study of antioxidative responses, indicators of oxidative damage and cellular redox environment in Cu-deficient or Cu-excess mulberry plants was undertaken. While the young leaves of plants supplied with nil Cu showed chlorosis and necrotic scorching of laminae, the older and middle leaves of plants supplied with nil or 0.1 μM Cu showed purplish-brown pigmented interveinal areas that later turned necrotic along the apices and margins of leaves. The Cu-excess plants showed accelerated senescence of the older leaves. The Cu-deficient plants showed accumulation of hydrogen peroxide and superoxide anion radical. The accumulation of hydrogen peroxide was strikingly intense in the middle portion of trichomes on Cu-deficient leaves. Though the concentration of total ascorbate increased with the increasing supply of Cu, the ratio of the redox couple (DHA/ascorbic acid) increased in Cu-deficient or Cu-excess plants. The activities of superoxide dismutase (EC 1.15.1.1), catalase (EC 1.11.1.6), peroxidase (EC 1.11.1.7), ascorbate peroxidase (EC 1.11.1.11) and glutathione reductase (EC 1.6.4.2) increased in both Cu-deficient and Cu-excess plants. The results suggest that deficiency of Cu aggravates oxidative stress through enhanced generation of ROS and disturbed redox couple. Excess of Cu damaged roots, accelerated the rate of senescence in the older leaves, induced antioxidant responses and disturbed the cellular redox environment in the young leaves of mulberry plants.  相似文献

The changes in superoxide (O2-) production, hydrogen peroxide (H2O2) content and active oxygen scavenging system in corn (Zea mays L. ) leaves under waterlogging stress were investigated to explore the relationship between O2- accumulation and waterlogging injury. Corn plants were grown in pots in a controlled environment. The results showed that prolonged waterlogging treatment conducted at 4-leaf stage caused a significant increase in the production of O2- and H2O2, while the extent of O2- change was more than that of H2O2. Malondialdehyde (MDA) accumulation, chlorophyll loss and electrolye leakage were positively correlated with O2- production in corn waterlogged leaves. Foliage spraying with 0. 1 mmol/L paraquat (02- producer) at the start of waterlogging treatment led to a significant increase in 02-, H202 and MDA levels. The addition of DDTC (SOD activity inhibitor) aggravated 02- formation in waterlogged leaves. Waterlogging apperantly reduced the activities of SOD. catalase (CAT), ascorbate peroxidase (AP) and the concentrations of ascorbic acid (ASA) and glutathione (GSH). It was noted that the decline in SOD activity proceeded the diminishment of H2O2 scavengers in chloroplasts (i. e. AP, AsA and GSH). Salinity influences the agricultural production all over the world. This constrain, similar to others biotic and abiotic stresses generate the reactive oxygen species such as superoxide, hydrogen peroxide and hydroxyl radicals. In the evolution process of halophyte plants the mechanisms to detoxify ROS, such as antioxidant enzymes, have been developed. Aeluropus littoralis is a special halophyte that selected to our research, so the plants treated with NaCl at different salt concentration (0, 250, 450 and 650 mM) for a period 45 days. Leaves and roots (separately) collected and their proteins extracted for superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) and peroxidase (POD) activity assay. Meanwhile the electrolyte leakage of leaves analyzed and increased at 450 and 650 mM of NaCl concentrations. Superoxide dismutase and catalase showed same pattern for changing in enzymatic activities (increasing activity by salt stress in roots and decreasing in shoot at 450 and 650 mM stress), also peroxidase and ascorbate peroxidase activity almost increased in all stress conditions.  相似文献
