Genetic variation in plant growth under limited phosphorus (P) supply is necessary to obtain more productive cultivars on low P-available soils. Two pot experiments were conducted to evaluate the variability of some traits associated with efficiency of P absorption and utilization in wild and cultivated genotypes of common bean (Phaseolus vulgaris L.) under biological N2 fixation. At two P levels (20 and 80 mg P kg-1 soil, P1 and P2, respectively), 20 wild and 6 cultivated genotypes were grown in Experiment 1, and 4 wild and 27 cultivated genotypes were grown in Experiment 2. Plants were harvested at flowering, but in Experiment 1 wild accessions that did not flower were harvested at the beginning of leaf senescence. In Experiment 1, part of the genotypic variability of wild accessions was attributed to a less homogeneous ontogenetic stage at harvest, whereas in Experiment 2 some variation in biomass production was due to distinct phenologies of cultivated genotypes. Wild lines did not seem more tolerant to low P conditions, but the genotypic variation observed suggests these materials as a source of genetic diversity. Part of the variation in the root area and root efficiency ratio (total P content:root area) was compensatory, resulting in narrow genotypic differences in the total P content. The total P content and root efficiency ratio presented a wider amplitude of variation at P2 than at P1, and P uptake was more influenced by P supply than root production. Since the genotype × P level interaction was not significant for shoot biomass and shoot P concentration in Experiment 2, P utilization efficiency may be a useful selection criterion for cultivars between limited and adequate P supply. Within the sample of genetic diversity evaluated herein, there was large genotypic variability for traits related to P efficiency among wild and cultivated genotypes of common bean.  相似文献

Phytophagous insects generally feed on a restricted range of host plants, using a number of different sensory and behavioural mechanisms to locate and recognize their host plants. Phloem-feeding aphids have been shown to exhibit genetic variation for host preference of different plant species and genetic variation within a plant species can also have an effect on aphid preference and acceptance. It is known that genotypic interactions between barley genotypes and Sitobion avenae aphid genotypes influence aphid fitness, but it is unknown if these different aphid genotypes exhibit active host choice (preference) for the different barley genotypes. Active host choice by aphid genotypes for particular plant genotypes would lead to assortative association (non-random association) between the different aphid and plant genotypes. The performance of each aphid genotype on the plant genotypes also has the ability to enhance these interactions, especially if the aphid genotypes choose the plant genotype that also infers the greatest fitness. In this study, we demonstrate that different aphid genotypes exhibit differential preference and performance for different barley genotypes. Three out of four aphid genotypes exhibited preference for (or against) particular barley genotypes that were not concordant with differences in their reproductive rate on the specific barley genotype. This suggests active host choice of aphids is the primary mechanism for the observed pattern of non-random associations between aphid and barley genotypes. In a community context, such genetic associations between the aphids and barley can lead to population-level changes within the aphid species. These interactions may also have evolutionary effects on the surrounding interacting community, especially in ecosystems of limited species and genetic diversity.  相似文献

To assess the extent of variation in phosphorus acquisition efficiency of some winter wheat (Triticum aestivum L.), winter and spring barley (Hordeum vulgare L.) genotypes, depletion of inorganic phosphorus (P) extractable with 0.5 M NaHCO3 (NaHCO3-Pi) from the rhizosphere soil was studied. Nutrients supply, rhizosphere soil pH and soil water content was kept equal for all the genotypes with the aim to reduce the confounding variation due to these factors. The experimental set up implied that no difference in the relative growth rates, nitrogen, potassium and calcium content of shoot dry matter occurred among the genotypes.The winter wheat, winter barley and spring barley genotypes differed significantly (p>0.05) in their efficiency to acquire NaHCO3-Pi from the rhizosphere soil. The efficiency of the winter wheat genotypes to acquire NaHCO3-Pi from rhizosphere soil ranked Kraka > Gawain > Foreman > Sleipner = Obelisk > Kosack > Pepital > Arum. Winter wheat genotypes differed in extent of P depletion profiles in the rhizosphere, indicating variation in root hair length. The winter barley and spring barley genotypes also showed significant differences in their P depletion profiles near roots. The efficiency of the winter barley genotypes to acquire soil P in the rhizosphere ranked Hamu > Frost > Marinka > Astrid > Clarine = Angora. The efficiency of spring barley genotypes to acquire NaHCO3-Pi in the rhizosphere ranked Canut > Etna Riga > Digger > Peel > Semal > Alexis. The rhizosphere pH remained unchanged, suggesting that additional mechanisms such as root hair formation and root exudates play a significant role in causing variation in P acquisition among the genotypes.  相似文献

Knowledge of the virulence of a pathogen population and recognition of the risks of changes in the virulence spectrum are essential in breeding crops for disease resistance. Sexual recombination in a pathogen increases the level of genotypic diversity and can influence the virulence spectrum. This study aimed to determine how sexual recombination can change virulence of the barley pathogen Pyrenophora teres and whether the barley cultivation system, no‐tillage or normal tillage, influences P. teres virulence. The inheritance of avirulence/virulence in P. teres following sexual reproduction was studied in three artificially created pathogen populations. The first was a product of crossing two net forms of the pathogen, and the second and the third were products of crossing net and spot forms. None of the progeny generated resembled the parents exactly. The average similarity of the progeny isolates of the net by net cross with the parental type, based on avirulence/virulence tests, was 92%. That for net and spot form progenies was 58% in comparison with the net form parents and 73% with the spot form parents. The virulence reactions of the progeny isolates did not correlate with morphological traits of the isolates: growth rate on agar, spore production, spore width, spore length and numbers of septa per conidium. To study the effect of the barley cultivation method on P. teres virulence, 313 single‐spore cultures were obtained from barley fields. Two hundred and seventy‐six of the isolates represented the spot form and 37 represented the net form of P. teres. No association was established between the tillage method and virulence for either the net form or the spot form isolates.  相似文献

1 Sitobion avenae (F.) is a serious pest in Danish cereal crops. To understand the population genetic structure, aphids were sampled in seven different winter wheat (Triticum sativum Lamarck) fields throughout Denmark. The aphids were genotyped with seven microsatellite markers. In total, 2075 aphids were collected and 1203 of these were genotyped. 2 The Danish S. avenae populations displayed very high genotypic diversity, high percentages of unique genotypes and low linkage disequilibria; this is likely to be a result of genetic recombination encompassed by their holocyclic lifestyle. The populations showed very limited differentiation and no sign of isolation by distance. Almost all the genetic variation was ascribed within the populations rather than between populations, probably due to a high migration rate at approximate 10% per generation. 3 Seasonal changes in clonal diversity and distribution of asexual summer generations of S. avenae within the infestation period in a single winter wheat field were followed over two consecutive years by weekly sampling from 60 plots each of 20 × 20 m. Clonal diversity was high in all samples with no dominant clonal lineages and no significant difference in the genotypic diversity between weeks or between years. However, a temporal genetic differentiation effect, throughout the infestation, suggests that selective factors or high temporal migration play an important role in shaping the genetic structure S. avenae. 4 Analyses of fungal infected and uninfected aphids were performed to test whether some clonal linage were more often infected by fungi from the Entomophthorales under field conditions. In total, 54 progeny from aphids with Entomophthorales were genotyped and compared with 422 uninfected aphid genotypes. The Entomophthorales‐infected aphid genotypes did not cluster out together, suggesting that these fungal pathogens did not affect the population differentiation or clonal distribution of S. avenae in a Danish agroecosystem. 5 Our findings indicate that S. avenae populations can be controlled using conservation biological control  相似文献

Changes in the genetic structure and genotypic variation of the aphid Sitobion avenae collected from cereal crops in northern France were examined by analysing variation at five microsatellite loci across several years and seasons. Little regional and temporal differentiation was detected, as shown by very low FST among populations. Repeated genotypes, significant heterozygote deficits, positive FIS values and frequent linkage disequilibria were found in nearly all samples, suggesting an overall pattern of reproductive mode variation in S. avenae populations. In addition, samples from Brittany (Bretagne) showed greater signs of asexual reproduction than those from the north of France, indicating a trend toward increasing sexuality northward. These patterns of reproductive variation in S. avenae are consistent with theoretical models of selection of aphid reproductive modes by climate. Contrasting with little changes in allelic frequencies, genotypic composition varied substantially in time and, to a lesser extent, in space. An important part of changes in genotypic arrays was due to the variation in frequency distribution of common genotypes, i.e. those that were found at several instances in the samples. Genotypic composition was also shown to vary according to climate, as genotypic diversity in spring was significantly correlated with the severity of the previous winter and autumn. We propose that the genetic homogeneity among S. avenae populations shown here across large temporal and spatial scales is the result of two forces: (i) migration conferred by high dispersal capabilities, and (ii) selection over millions of hectares of cereals (mostly wheat) bred from a narrow genetic base.  相似文献

Ammonium-fed plants may acidify the rhizosphere and thus increaseavailability of Mn in calcareous alkaline soils. The importanceof N nutrition in the differential expression of tolerance toMn deficiency among cereal genotypes is not yet clear. Two factorialexperiments testing effects of the NH4-N/NO3-N ratio and Mnfertilization on growth of barley genotypes differing in toleranceto Mn deficiency were conducted in two calcareous alkaline soilsin pots in a controlled environment. In the soil containing80% CaCO3at pH 8.5, better root and shoot growth and highershoot Mn concentrations were achieved with nitrate supply, especiallyat lower rates of Mn fertilization. The Mn-efficient genotypeWeeah (tolerant of Mn deficiency) achieved better root and shootgrowth than Mn-inefficient Galleon barley (sensitive to Mn deficiency)regardless of experimental treatment. Fertilization with Mndid not influence total N concentration in barley roots andshoots. In the soil containing 5% CaCO3at pH 7.8, ammonium-fedplants had better root and shoot growth and, at shoot Mn concentrationsabove the critical level, Mn-inefficient Galleon performed betterthan Mn-efficient Weeah barley. It appears that differentialexpression of Mn efficiency among barley genotypes is not associatedwith differences in Mn availability expected to be producedby differential rhizosphere acidification as a response to differentforms of N supply. In this study we investigated the genotypic diversity and the migration patterns of Phytophthora infestans in the Nordic countries. Isolates of P. infestans from outbreaks in 43 fields sampled in 2008 were collected using stratified sampling with country, field, and disease foci as the different strata. Microsatellites were used as markers to determine the genotypic variation in the sampled material. The results show a high genotypic variation of P. infestans in the Nordic countries with most of the genotypes found only once among the collected isolates. The major part of the genotypic variation was observed within the fields, with low differentiation between the fields. The observed low association of alleles among loci is consistent with frequent sexual reproduction of P. infestans in the Nordic countries. Coalescence analyses did not support a single common population for the four countries, thus indicating some degree of geographic differentiation. The analyses of migration patterns showed differing levels of gene flow among the Nordic countries. No correlation between migration rates and geographical distance could be seen. This could be explained by different degrees of genetic similarity between the pathogen populations in the different countries.  相似文献

Barley is a major cereal grown widely and used in several food products, beverage production and animal fodder. Genetic diversity is a key component in breeding programs. We have analyzed the genetic diversity of barley accessions using microsatellite markers. The accessions were composed of wild and domesticated barley representing genotypes from six countries and three breeding programs in Brazil. A total of 280 alleles were detected, 36 unique to Brazilian barley. The marker Bmag120 showed the greatest polymorphism information content (PIC), with the highest mean value found on chromosome three, and the lowest on chromosomes four and six. The wild accessions presented the highest diversity followed by the foreign genotypes. Genetic analysis was performed using Principal Coordinates Analysis, UPGMA clustering, and Bayesian clustering analysis implemented in Structure. All results obtained by the different methods were similar. Loss of genetic diversity has occurred in Brazilian genotypes. The number of alleles detected in genotypes released in 1980s was higher, whereas most of the cultivars released thereafter showed lower PIC and clustered in separate subgroups from the older cultivars. The use of a more diverse panel of genotypes should be considered in order to exploit novel alleles in Brazilian barley breeding programs.  相似文献

Improvement of end-use quality in bread wheat (Triticum aestivum L.) depends on a thorough understanding of the genetic basis of important quality traits. The main goal of our study was to investigate the genetic basis of 1,000-kernel weight, protein content, sedimentation volume, test weight, and starch concentration using an association mapping approach. We fingerprinted 207 diverse European elite soft winter wheat lines with 115 SSR markers and evaluated the genotypes in multi-environment trials. The principal coordinate analysis revealed absence of a clear population but presence of a family structure. Therefore, we used linear mixed models and marker-based kinship matrices to correct for family structure. In genome-wide scans, we detected main effect QTL for all five traits. In contrast, epistatic QTL were only observed for sedimentation volume and test weight explaining a small proportion of the genotypic variation. Consequently, our findings suggested that integrating epistasis in marker-assisted breeding will not lead to substantially increased selection gain for quality traits in soft winter wheat.  相似文献

Low phosphorus (P) availability in soils and diminishing P reserves emphasize the need to create plants that are more efficient P users. Knowledge of P efficient germplasm among the existing cereal varieties may serve as the basis for improving soil P use by selection and breeding. We had identified some cereal cultivars (winter wheat: Kosack and Kraka; winter barley: Hamu and Angora; spring barley: Canut, Alexis, Salka, Zita;) which differed (p<0.05) in P depletion from thin slices (0.2 mm) of the rhizosphere soil under controlled conditions. In the present study, the same cultivars were studied under field conditions at three levels of P supply (no-P, 10 and 20 kg P ha-1) and the differences in P uptake as found in the previous work were confirmed. Under both conditions, the variation between the cultivars was greatest in soil without P fertilizers (no-P) for about 30 years. The variation in P uptake with most cultivars disappeared when 10 kg P ha-1 was applied. Root development did not differ between the cultivars much, but there was wide, consistent variation in their root hairs, regardless of growth media (solution, soil column and field). Increase in soil P level reduced the length of root hairs. The variation in root hairs between the cultivars was largest in no-P soil. When 10 kg P ha-1 was applied, the root hair lengths did not differ between the cultivars. Barley cultivars with longer root hairs depleted more P from the rhizosphere soil and also absorbed more P in the field. The relationship between root hairs and phosphorus uptake of the wheat cultivars was less clear. The wide variation in P uptake among the barley cultivars in the field and its relationship to the root hair development confirms that root hair length may be a suitable plant characteristic to use as criterion for selecting barley cultivars for P efficiency, especially in low-P soils. Developments in high-throughput genotyping provide an opportunity to explore the application of marker technology in distinctness, uniformity and stability (DUS) testing of new varieties. We have used a large set of molecular markers to assess the feasibility of a UPOV Model 2 approach: “Calibration of threshold levels for molecular characteristics against the minimum distance in traditional characteristics”. We have examined 431 winter and spring barley varieties, with data from UK DUS trials comprising 28 characteristics, together with genotype data from 3072 SNP markers. Inter varietal distances were calculated and we found higher correlations between molecular and morphological distances than have been previously reported. When varieties were grouped by kinship, phenotypic and genotypic distances of these groups correlated well. We estimated the minimum marker numbers required and showed there was a ceiling after which the correlations do not improve. To investigate the possibility of breaking through this ceiling, we attempted genomic prediction of phenotypes from genotypes and higher correlations were achieved. We tested distinctness decisions made using either morphological or genotypic distances and found poor correspondence between each method.  相似文献

Chickpea (Cicer arietinum L.) is one of the oldest and most important pulse crops grown and consumed all over the world, especially in developing countries. Magnesium (Mg) and manganese (Mn) are essential plant nutrients in terms of human health and many health problems arise in their deficiencies. The objectives of this study were to characterize genetic variability in the seed Mg and Mn concentrations and identify single nucleotide polymorphism (SNP) markers associated with these traits in 107 Cicer reticulatum and 73C. arietinum genotypes, using a genome wide association study. The genotypes were grown in four environments, characterized for Mg and Mn concentrations, and genotyped with 121,841 SNP markers. The population showed three-fold and two-fold variation for the Mg and Mn concentrations, respectively. The population structure was identified using STRUCTURE software, which divided 180 genotypes into two (K = 2) groups. Principal component analysis and neighbor joining tree analysis confirmed the results of STRUCTURE. A total of 4 and 16 consistent SNPs were detected for the Mg and Mn concentrations, respectively. The identified markers can be utilized in breeding of chickpea to increase Mg and Mn levels in order to improve human and livestock nutrition.  相似文献

Globally, over 7.4 million accessions of crop seeds are stored in gene banks, and conservation of genotypic variation is pivotal for breeding. We combined genetic and biochemical approaches to obtain a broad overview of factors that influence seed storability and ageing in barley (Hordeum vulgare). Seeds from a germplasm collection of 175 genotypes from four continents grown in field plots with different nutrient supply were subjected to two artificial ageing regimes. Genome‐wide association mapping revealed 107 marker trait associations, and hence, genotypic effects on seed ageing. Abiotic and biotic stresses were found to affect seed longevity. To address aspects of abiotic, including oxidative, stress, two major antioxidant groups were analysed. No correlation was found between seed deterioration and the lipid‐soluble tocochromanols, nor with oil, starch and protein contents. Conversely, the water‐soluble glutathione and related thiols were converted to disulphides, indicating a strong shift towards more oxidizing intracellular conditions, in seeds subjected to long‐term dry storage at two temperatures or to two artificial ageing treatments. The data suggest that intracellular pH and (bio)chemical processes leading to seed deterioration were influenced by the type of ageing or storage. Moreover, seed response to ageing or storage treatment appears to be significantly influenced by both maternal environment and genetic background.  相似文献

Patterns of polymorphism and linkage disequilibrium in cultivated barley   总被引:1,自引:0,他引:1  
We carried out a genome-wide analysis of polymorphism (4,596 SNP loci across 190 elite cultivated accessions) chosen to represent the available genetic variation in current elite North West European and North American barley germplasm. Population sub-structure, patterns of diversity and linkage disequilibrium varied considerably across the seven barley chromosomes. Gene-rich and rarely recombining haplotype blocks that may represent up to 60% of the physical length of barley chromosomes extended across the ‘genetic centromeres’. By positioning 2,132 bi-parentally mapped SNP markers with minimum allele frequencies higher than 0.10 by association mapping, 87.3% were located to within 5 cM of their original genetic map position. We show that at this current marker density genetically diverse populations of relatively small size are sufficient to fine map simple traits, providing they are not strongly stratified within the sample, fall outside the genetic centromeres and population sub-structure is effectively controlled in the analysis. Our results have important implications for association mapping, positional cloning, physical mapping and practical plant breeding in barley and other major world cereals including wheat and rye that exhibit comparable genome and genetic features.  相似文献

Drought tolerance is the essential trait that needs to be incorporated in cereal crops, particularly those grown under the rainfed cultivation. Drought tolerance being contributed by several regions of the genome requires identification of these regions, using suitable molecular markers. Therefore, present investigation was aimed at analyzing the genetic diversity present among the cultivars of rainfed and the irrigated areas with respect to the drought tolerant trait. In all, 14 RAPD and 90 ISSR markers were used to identify these genomic regions. Out of 14 RAPD markers, one RAPD primer exhibited polymorphic banding pattern with 18.6 % polymorphism, clearly separating drought tolerant and drought susceptible genotypes. Out of 90 ISSR primers, only 3 ISSR primers revealed polymorphism in relation to the drought tolerance trait exhibiting 21.38 % polymorphism.  相似文献

However, genotypic variation in root morphological characteristics of common bean (Phaseolus vulgaris L.) as affected by P availability has not been well clarified. In the present study, systematic studies were conducted in a P-buffered sand culture system using three pairs of common bean parental materials with contrasting root traits in response to P deficiency. The results indicate that P availability significantly affects bean root morphology. Common bean tends to have smaller root system, shorter and coarser roots at low P availability. Genotypic variation in root morphology was observed among different genotypes in response to P availability. The P efficient genotypes appear to have larger, finer and longer root systems than the P inefficient genotypes, and such a variation was particularly obvious in the basal roots. From allomeric analysis, we found that morphological characteristics of the basal roots contribute more to P efficiency than those of the tap roots. Further studies with the F9 recombinant inbred lines derived from one of the most contrasting parental pairs, DOR364 and G19833, confirmed the above findings, indicating that those morphological characteristics are inheritable hence provide potential for genetic improvement.
