Diseases affecting strawberry (Fragaria × ananassa Duch.) have been of major concern in recent years because of their widespread occurrence and potential for yield loss. Anthracnose, caused by the fungus Colletotrichum acutatum, is one of the most serious diseases of strawberry worldwide. Tissue-culture induced (somaclonal) variation provides one strategy for generating disease-resistant genotypes. As part of a program to generate strawberry germplasm resistant to anthracnose, an in vitro screening system was used to evaluate several commercial cultivars, Chandler, Delmarvel, Honeoye, Latestar, Pelican and Sweet Charlie propagated in vitro, and shoots regenerated from leaf explants of these cultivars for resistance to C.␣acutatum isolate Goff (highly virulent). Regenerants with increased levels of resistance were identified from all of the cultivars. The greatest increases in disease resistance were observed for regenerants from leaf explants of cultivars Pelican and Chandler that exhibited 17.5- and 6.2-fold increases in resistance, respectively. The highest levels of anthracnose resistance (2 to 6% leaf necrosis) were exhibited by regenerants from explants of cultivars Pelican and Sweet Charlie. These studies suggest that generating somaclonal variation may be a viable approach to obtaining strawberry plants with increased levels of anthracnose resistance.  相似文献

Actin depolymerizing factor (ADF) is an 18.5-kD protein with pH- dependent reciprocal F-actin binding and severing/depolymerizing activities. We previously showed developing muscle down-regulates ADF (J. R. Bamburg and D. Bray. 1987. J. Cell Biol. 105: 2817-2825). To further study this process, we examined ADF expression in chick myocytes cultured in vitro. Surprisingly, ADF immunoreactivity increases during the first 7-10 d in culture. This increase is due to the presence of a new ADF species with higher relative molecular weight which reacts identically to brain ADF with antisera raised against either brain ADF or recombinant ADF. We have purified both ADF isoforms from myocytes and have shown by peptide mapping and partial sequence analysis that the new isoform is structurally related to ADF. Immunoprecipitation of both isoforms from extracts of cells prelabeled with [32P]orthophosphate showed that the new isoform is radiolabeled, predominantly on a serine residue, and hence is called pADF. pADF can be converted into a form which comigrates with ADF on 1-D and 2-D gels by treatment with alkaline phosphatase. pADF has been quantified in a number of cells and tissues where it is present from approximately 18% to 150% of the amount of unphosphorylated ADF. pADF, unlike ADF, does not bind to G-actin, or affect the rate or extent of actin assembly. Four ubiquitous protein kinases failed to phosphorylate ADF in vitro suggesting that ADF phosphorylation in vivo is catalyzed by a more specific kinase. We conclude that the ability to regulate ADF activity is important to muscle development since myocytes have both pre- and posttranslational mechanisms for regulating ADF activity. The latter mechanism is apparently a general one for cell regulation of ADF activity.  相似文献

A strawberry genomic clone containing an osmotin-like protein (OLP) gene, designated FaOLP2, was isolated and sequenced. FaOLP2 is predicted to encode a precursor protein of 229 amino acid residues, and its sequence shares high degrees of homology with a number of other OLPs. Genomic DNA hybridization analysis indicated that FaOLP2 represents a multi-gene family. The expression of FaOP2 in different strawberry organs was analyzed using real-time PCR. The results showed that FaOLP2 expressed at different levels in leaves, crowns, roots, green fruits and ripe red fruits. In addition, the expression of FaOLP2 under different abiotic stresses was analyzed at different time points. All of the three tested abiotic stimuli, abscisic acid, salicylic acid and mechanical wounding, triggered a significant induction of FaOLP2 within 2-6h post-treatment. Moreover, FaOLP2 was more prominently induced by salicylic acid than by abscisic acid or mechanical wounding. The positive responses of FaOLP2 to the three abiotic stimuli suggested that strawberry FaOLP2 may help to protect against osmotic-related environmental stresses and that it may also be involved in plant defense system against pathogens.  相似文献

Chick brain actin depolymerizing factor (ADF) is a 19-kDa protein that severs actin filaments and binds actin monomers. We have obtained a cDNA encoding ADF by screening a chick embryo lambda gt11 cDNA library with both a rabbit anti-ADF antiserum and two oligonucleotide probes. Several non-full-length clones of 636 bases and one full-length clone of 1886 bases were isolated and sequenced. The full-length cDNA encodes a protein of 165 amino acids with a calculated molecular weight of 18,520. The deduced amino acid sequence shows 73% identity with the porcine brain actin binding protein cofilin. The coding region of the ADF cDNA has been placed in an expression vector, and the resulting protein shows immunoreactivity with an anti-ADF antiserum but not with an anti-cofilin antibody. The expressed ADF has been purified and has an actin depolymerizing activity identical with that of brain ADF. Like cofilin, ADF contains a sequence similar to the nuclear transport signal sequence of the SV40 large T antigen and a calcium/calmodulin-dependent protein kinase II phosphorylation consensus sequence. Northern blots of both embryonic chick brain and muscle RNA revealed two ADF mRNAs of length 2.1 and 0.9 kilobases. Southern blots suggest that the ADF gene is present in a single copy within the chicken genome. ADF contains regions of homology with other actin binding proteins including tropomyosin, gelsolin, and depactin.  相似文献

We have isolated cDNA and genomic clones encoding a mitochondrial HSP70 protein from Schizosaccharomyces pombe. Nucleotide sequence analysis indicates that the encoded protein is homologous to the HSP70s of other organisms. The highest degree of amino acid conservation is with the proteins encoded by the Escherichia coli dnaK gene, the SSC1 gene of Saccharomyces cerevisiae and the MTP70 gene of Trypanosoma cruzi, the latter two having recently been shown to be located in the mitochondria. Western-blot analysis with immunoglobulin G raised against a peptide corresponding to the C terminus of the SSP1 protein indicates a 70-kDa protein which is associated with the mitochondria.  相似文献

Actin is a ubiquitous and highly conserved microfilament protein that is hypothesized to play a mechanical force-generating role in the unusual gliding motility of sporozoan zoites and their active penetration of host cells. We have identified and isolated an actin gene from a Babesia gibsoni cDNA library by random sequencing. The complete nucleotide sequence of the actin gene is 1,243 bp; a single open reading frame encodes a polypeptide of 377 amino acid residues. The deduced amino acid sequence showed a high homology with actins from other species, especially with reported apicomplexan protozoans. The antiserum against recombinant actin expressed in Escherichia coli recognizes a 42-kDa native protein, which is consistent with its expected size. Immunofluorescence and confocal microscopic observation revealed that the protein is diffusely distributed throughout the B. gibsoni parasites.  相似文献

A cotton fiber is a single and highly elongated ovule epidermal cell. However, the mechanism that governs the development of fiber traits remains unclear. In this study, we cloned a calcium-dependent protein kinase (GhCPK1) and an actin depolymerizing factor (GhADF1) from Gossypium hirsutum. Real-time PCR analyses indicated that the expression of GhCPK1 and GhADF1 correlated with the elongation pattern of cotton fibers. Yeast two-hybrid assays using full-length GhCPK1 and truncated forms of GhCPK1 as baits identified GhADF1 as an interactor of GhCPK1. Furthermore, GhCPK1 is capable of phosphorylating GhADF1 in vitro in a calcium-dependent manner, and the phosphorylation of GhADF1 by GhCPK1 occurs on the Ser-6 of GhADF1. In addition, we observed that the heterologous expression of the GhCPK1 gene induced longitudinal growth of the host cells by 3.18-fold, with no apparent effect on other aspects of the host cells. The results strongly suggest that GhCPK1 may regulate the function of GhADF1 by phosphorylating this protein during cotton fiber elongation.  相似文献

A mouse genomic library in lambda Charon 4A was screened for putative ribosomal protein genes using a fragment of the gene encoding Drosophila ribosomal protein 49 as a hybridization probe under nonstringent hybridization conditions. A recombinant phage was selected and its restriction enzyme map determined. The major species of mouse poly(A)+ mRNA homologous to the putative gene is about 740 nucleotides long.  相似文献

We have isolated the gene encoding a methionine-rich 10-kDa zein protein from a lambda EMBL3 maize genomic 'mini' library of the inbred line BSSS-53 and determined its nucleotide sequence. The sequence matches perfectly with a cDNA clone from the inbred line W22 (which has the same restriction fragment length polymorphism as many inbred lines tested) indicating that we have isolated a functional storage protein gene that is very conserved in maize. This comparison also excludes any splicing of any precursor mRNA and therefore any presence of introns. A number of potential regulatory sequences have been located in the flanking regions. The 10-kDa-zein gene represents the last size class in the zein multigene family to be characterized. Its structure allows us now to re-examine the relationship of all the zein proteins and also to compare the structure of a new class of storage proteins that are rich in methionine, an essential amino acid in livestock fodder.  相似文献

We have cloned a putative yeast homolog of the gene encoding the human Tat-binding protein, TBP-1. The gene termed TBPY encodes a 45,243-dalton protein displaying a heptad repeat of hydrophobic amino acids reminiscent of a leucine zipper. Secondary structure predictions suggest the possibility of formation of an amphipathic helix that could further be organized into a coiled-coil. Additionally, the protein product of TBPY shows amino acid signatures characteristic of a large family of RNA and DNA helicases. We propose that the hydrophobic region of yTBP-1 participates in self-dimerization or heterodimerization.  相似文献

A gene encoding a putative membrane protein has been identified from Campylobacter jejuni NCTC 11168 following an immuno-screen of a lambda ZAP II genomic DNA library with antiserum raised against glycine-extractable proteins. The nucleotide sequence of the entire genomic insert revealed six open reading frames, all but one of which have sequence homologues in the complete genome sequence of Helicobacter pylori. The gene encoding the immuno-reactive protein was further identified by independent expression of these reading frames in Escherichia coli. The gene encodes an integral membrane protein, expression of which in E. coli results in a profound filamentous phenotype.  相似文献

We report the nucleotide sequence of a cDNA encoding an actin from amarine red alga, Porphyra yezoensis Ueda. A cDNA clone wasisolated from a leafy gametophyte cDNA library and analyzed for the sequence.The clone contained an open reading frame for a protein of 373 amino acidswhichexhibits sequence similarity to known actins. The GC content of the thirdposition (83.9%) was much higher than that at the first (56.3%) and second(42.4%) positions. The actin forms a gene family in the P.yezoensis genome. Comparison of the deduced amino acid sequenceshowed higher similarity to the Florideophycidae Chondruscrispus (85%) than to the ProtoflorideophycidaeCyanidioschyzon merolae (70%). The mRNA was detected inboth the leafy gametophytes and filamentous sporophytes.
