Aims:  The hypothetical dithiolopyrrolone biosynthetic pathway includes a final step of pyrrothine nucleus acylation. The presence of an enzymatic activity catalysing this reaction was investigated in Saccharothrix algeriensis NRRL B-24137. To understand the effect exerted by organic acids on the level of dithiolopyrrolone production, their influence on enzymatic expression was studied.
Methods and Results:  The transfer of acetyl-CoA or benzoyl-CoA on pyrrothine was assayed in the cell-free extract of Sa. algeriensis NRRL B-24137. This study reports the presence of an enzymatic activity catalysing this reaction that was identified as either pyrrothine N -acetyltransferase or N -benzoyltransferase. The stimulation of benzoyl-pyrrothine (BEP) production by addition of benzoic acid at 1·25 mmol l−1 into the culture medium was demonstrated, and results showed that under the same conditions of growth, pyrrothine N -benzoyltransferase specific activity was doubled.
Conclusions:  This study shows that BEP production is enhanced in the presence of benzoic acid partly because of an induction of pyrrothine N -benzoyltransferase.
Significance and Impact of the Study:  The antitumor and antibiotic properties of dithiolopyrrolones are related to their variable acyl groups. New insights into regulation of biosynthetic pathway, especially the step of pyrrothine acylation, could lead after further studies to yield improvement and to selective production of dithiolopyrrolones with new biological activities.  相似文献

The amino acid and humic acid requirements of Saccharothrix algeriensis NRRL B-24137 for growth and production of the dithiolopyrrolone antibiotics were studied in a semi-synthetic medium (SSM). Nature and concentration of amino acids and humic acid strongly influenced the growth and dithiolopyrrolone specific production.

The highest value of thiolutin (acetyl-pyrrothine) specific production was obtained in the presence of 1 g/l humic acid (336 mg/g DCW), and in the presence of 5 mM l-cystine (309 mg/g DCW) as compared to 19 mg/g DCW obtained with the control. Furthermore, thiolutin production was increased about six-fold, four-fold and three-fold in the presence of l-proline, l-glutamic acid and dl-histidine, respectively. In contrast, the production of thiolutin was reduced by addition of other amino acids such as l-glutamine, dl-ethionine, l-methionine and l-arginine. The highest value of isobutyryl-pyrrothine production was obtained in the presence of 2,6-diaminopimelic acid and l-lysine (7.8 and 1.0 mg/g DCW, respectively). However, the highest value of butanoyl-pyrrothine production was obtained in the presence of humic acid (6.6 mg/g DCW), followed by l-cysteine and l-proline (3.6 and 3.2 mg/g DCW, respectively). In addition, the maximum specific production of senecioyl-pyrrothine (29 mg/g DCW) and tigloyl-pyrrothine (21 mg/g DCW) was obtained in the presence of humic acid. We found that, except for isobutyryl-pyrrothine, production of all dithiolopyrrolones was favoured by addition of l-proline. The maximum specific production was obtained with l-proline at concentrations of 2.50 mM for thiolutin (133 mg/g DCW), 1.25 mM for senecioyl-pyrrothine, tigloyl-pyrrothine and butanoyl-pyrrothine production (29, 23 and 3.9 mg/g DCW, respectively). Production of all dithiolopyrrolones strongly decreased as the l-methionine or dl-ethionine concentration was increased in the culture medium.  相似文献

Dithiolopyrrolone antibiotics, produced by several microorganisms, are known for their strong antimicrobial activities. This class of antibiotics generated new interest after the discovery of their anticancer and antitumor properties. In this study, four new antibiotics were purified from the fermentation broth of Saccharothrix algeriensis NRRL B-24137 and characterized as dithiolopyrrolone derivatives. These new dithiolopyrrolone antibiotics were induced by adding sorbic acid, as precursor, at a concentration of 5 mM to the semi-synthetic medium. The analysis of the induced antibiotics was carried out by HPLC. The maximal production of the antibiotics PR2, PR8, PR9 and PR10 was 0.08±0.04, 0.21±0.04, 0.13±0.03 and 0.09±0.00 mg L(-1) , respectively, obtained after 8 days of fermentation. The chemical structures of these antibiotics were determined by (1) H- and (13) C-nuclear magnetic resonance, mass and UV-visible data. The four new dithiolopyrrolone antibiotics - PR2, PR8, PR9 and PR10 - were characterized, respectively, as crotonyl-pyrrothine, sorbyl-pyrrothine, 2-hexonyl-pyrrothine and 2-methyl-3-pentenyl-pyrrothine. The minimum inhibitory concentrations of the new induced antibiotics were determined.  相似文献

A new bacterial species, Saccharothrix algeriensis NRRL B-24137, was isolated in 1992 in the Sahara desert. This filamentous bacterium is able to produce dithiolopyrrolones, molecules presenting antibacterial, antifungal, and anticancer properties. In this study, a “reaction engineering” approach was adopted to gain more knowledge on the growth of Sa. algeriensis and its dithiolopyrrolone production on a semi-synthetic liquid medium. The objective is to establish a reaction scheme of the bacterium metabolism from extracellular experimental information, relatively easy to obtain. The approach enabled us to show that Sa. algeriensis could grow using several substrates that were sequentially consumed and that substrate limitation may induce a secondary metabolism in antibiotic production. From these qualitative data, a general reaction scheme was extracted consisting of four reactions: growth via amino acids, glucose consumption for maintenance, growth using glucose, and thiolutin production. The stoichiometric coefficients and the reaction extends were identified using a factorial analysis based on the bilinear structure of the component mass balances in a batch reactor. The analysis of the reaction stoichiometry enabled us to draw some conclusions concerning the substrate consumption pathway.  相似文献

二硫吡咯酮类抗生素是一类具有独特的吡咯酮二硫杂环戊二烯(4H-[1,2]二硫[4,3-b]吡咯-5-酮)骨架的化合物的总称。基于N-7位酰基侧链的不同以及N-4位是否含有甲基,可分为N-methyl-Nacylpyrrothine、N-acylpyrrothine和thiomarinols等类别。迄今为止,已有27种该类化合物被报道,重要代表包括全霉素(holomycin)、硫藤黄菌素(thiolutin)、金霉素(aureothricin)以及最近发现的thiomarinols。就生物活性而言,二硫吡咯酮类抗生素具有广谱的抗细菌活性,对多种微生物,包括革兰氏阴性菌、革兰氏阳性菌以及寄生虫都有较好的杀灭活性。甚至一些二硫吡咯酮衍生物表现出较强的抗肿瘤活性。近几年来,多个二硫吡咯酮类抗生素的生物合成基因簇相继被报道,其生物合成机理也逐步被阐明。本文将针对目前国内外二硫吡咯酮类抗生素的生物合成研究进展,以及在组合生物合成与代谢工程领域所取得的成果进行综述,旨在为通过合成生物学的方法创造结构新颖、高效低毒的"非天然"二硫吡咯酮类化合物提供理论借鉴。  相似文献

The production of dithiolopyrrolones by Saccharothrix algeriensis was investigated after supplementing the culture medium with ethanol and/or 1-butanol. Optimal conditions for the addition of ethanol to the culture medium provided a maximal dithiolopyrrolone titer of about 200 mg⋅L−1 after 5 days of culture, roughly corresponding to a 600%-increase. Using NAD(P)H oxidase inhibitor (diphenyleneiodonium) or reactive oxygen species scavenger (para-aminobenzoic acid), we suppose that ethanol promotes the formation of reactive oxygen species in Saccharothrix algeriensis, which, in turn, could induce biomass decline and dithiolopyrrolone overproduction. However, the underlying mechanisms remain to be elucidated. These results may be helpful for the control of dithiolopyrrolone yields from Saccharothrix algeriensis cultures.  相似文献

The influence of organic acids on growth and dithiolopyrrolone antibiotic production by Saccharothrix algeriensis NRRL B-24137 was studied. The production of dithiolopyrrolones depends upon the nature and concentration of the organic acids in the culture medium. Study of the nature of organic acids showed that the most effective organic acids for thiolutin specific production were maleic, 4-hydroxybenzoic, benzentetracarboxylic, pantothenic, pivalic and pyruvic acids (which yielded almost five-fold over the starting medium) and pimelic acid (more than three-fold). 4-Bromobenzoic acid showed the best production of senecioyl-pyrrothine (59 mg g−1 DCW). Tiglic acid showed the best production of tigloyl-pyrrothine (22 mg g−1 DCW). The highest yield of isobutyryl-pyrrothine (7.6 mg g−1 DCW) was observed in the presence of crotonic acid. Sorbic acid yielded the best production of butanoyl-pyrrothine (26 mg g−1 DCW). Methacrylic, butyric, pyruvic and 4-bromobenzoic acids also exhibited the best production of butanoyl-pyrrothine (27–11-fold).Study of organic acid concentration showed that among the selected organic acids, pimelic acid yielded the highest specific production of thiolutin (91 mg g−1 DCW) at 7.5 mM; and senecioyl-pyrrothine (11 mg g−1 DCW), tigloyl-pyrrothine (9 mg g−1 DCW) and butanoyl-pyrrothine (3.5 mg g−1 DCW) at 5 mM. Pyruvic acid at 1.25 mM enhanced the production of senecioyl-pyrrothine (4.3 mg g−1 DCW). The maximum production of tigloyl-pyrrothine (18.6 mg g−1 DCW) was observed in the presence of tiglic acid at 2.5 mM. Maximum production of isobutyryl-pyrrothine was observed in the presence of 7.5 mM tiglic acid. In addition, methacrylic acid (at 5 mM) and butyric acid (at 2.5 mM) enhanced the production of butanoyl-pyrrothine (26 and 20 times, respectively).The above results can be employed in the optimisation of the culture medium for the production of dithiolopyrrolone in higher quantities.  相似文献

Addition of methyl oleate to a Streptomyces hygroscopicus NRRL B-1865 culture modified the metabolic properties of this strain. This addition decreased the pH of the medium, increased the valine uptake of the cells and reduced their consumption of glucose until the beginning of antibiotic biosynthesis, which was delayed. At the same time, an increase in growth (× 1.8) and a marked improvement in antibiotic production (× 20) could be observed. The use of labelled methyl oleate showed that methyl oleate was not a precursor of antibiotics produced by S. hygroscopicus NRRL B-1865. It is suggested that methyl oleate addition may cause some alteration in membrane permeability, inducing an increase in H+ extrusion and stimulating the accumulation of branched amino acids, known to be direct precursors of polyether antibiotics. Background and aim

Saccharothrix algeriensis NRRL B-24137, isolated from a Saharan soil, has been described as a potential biocontrol agent against Botrytis cinerea and other phytopathogens. However, the plant protection mechanisms involved still need to be described. The aim of this study was to determine this protection phenomenon as well as parts of the mechanisms involved, using Arabidopsis thaliana seedlings and B. cinerea.

Methods

The bacterial colonization process was evaluated on A. thaliana seedlings using fluorescence in situ hybridization. Protection of A. thaliana seedlings inoculated with NRRL B-24137 against B. cinerea was then evaluated. Parts of the mechanisms involved in the systemic protection against B. cinerea were evaluated using known mutants of genes involved in jasmonate (JA)/ethylene (ET)/salicylic acid (SA) signaling. Other Arabidopsis mutants, AtrhbohD-3, AtrhbohF-3, and ups1-1 were also screened to determine other parts of the mechanisms involved.

Results

The results showed that the strain NRRL B-24137 colonized, epi- and endophytically, the roots of Arabidopsis seedlings but the strain was not a systemic colonizer during the time of the experiment. The strain NRRL B-24137 also reduced B. cinerea symptoms and the protection was linked to known mechanisms of induced systemic resistance (ISR; JA/ET signaling), as well as to functionality of AtrbohF oxidase and of UPS1. Crosstalk between ET/JA and SA signaling could also be involved.

Conclusions

The isolate NRRL B-24137, after colonizing the root systems of A. thaliana, induces an ISR against B. cinerea, which is JA/ET dependent, but could also require SA crosstalk and protection could also require NAPDH oxidases and UPS1 functionalities.  相似文献

Protein obtained from several strains of Escherichia coli grown in the presence of [3,3′-14C]cystine contained the radiolabel in nearly all the other amino acids, suggesting catabolism of cysteine to pyruvic acid. Utilization in amino acid synthesis of the pyruvate thus generated can be blocked by growing the bacteria in a medium specifically enriched with most of the naturally occurring amino acids. Cysteine that is incorporated intact is diluted by de novo synthesis at low cystine concentrations; also, it was found that E. coli can use the sulfur of methionine for cysteine biosynthesis. Both of these latter two processes can be prevented by supplying an excess of exogenous cystine. This regiment leads to protein that is highly specifically labeled in the cysteine residues, with a minor amount (20–25%) of the label also appearing in alanine residues. Although this strategy was developed expressly for cysteine, it may be useful for incorporating other labeled amino acids that are also readily catabolized.  相似文献

Transport and metabolism of glutamine has been investigated in human diploid fibroblasts, IMR-90. Glutamine was taken up via System ASC (Na+-dependent amino acid transport system especially reactive with short or polar side chain amino acids). In the routine culture medium the cells contained a large quantity of glutamate; its major source was shown to be glutamine in the medium. Previously we described a transport system that mediates the entrance of cystine in exchange for the exit of glutamate (Bannai, 1986). Since the cystine taken up is reduced to cysteine and the cysteine readily exits to the medium where it is oxidized to cystine, a cystine-cysteine cycle across the plasma membrane has been postulated. When the cells were cultured in glutamate/glutamine-free medium, intracellular glutamate decreased, depending on the amount of cystine in the medium; in the absence of cystine, glutamate decreased very slowly. When the cells were cultured in ordinary medium, glutamine in the medium decreased, and glutamate in the medium increased. Both changes were well correlated with cystine concentration in the medium. These results are consistent with the view that the intracellular glutamate, of which the source is glutamine in the medium, is released from the cells into the medium in order to take up cystine and thereby to rotate the cystine-cysteine cycle. In the routine culture one-third to one-half of the total consumption of glutamine seems to be used for the uptake of cystine.  相似文献

AIMS: Optimization of bacteriocin production by L. lactis subsp. lactis CECT 539 and Ped. acidilactici NRRL B-5627 on mussel-processing wastes. METHODS AND RESULTS: The concentrations of glucose and five nitrogen sources that optimize nisin and pediocin production were determined using a second order orthogonal factorial design. NH4Cl, glycine and glutamic acid were poor nitrogen sources. Enhanced pediocin and nisin productions were achieved in a medium supplemented with yeast extract or Bacto casitone. CONCLUSIONS: Mussel-processing wastes could be successfully used as a culture medium for bacteriocin production at low production cost. SIGNIFICANCE AND IMPACT OF THE STUDY: Average optimization led to a threefold increase in pediocin production (from 322 to 934 BU ml(-1)) and in nisin production (from 32 to 100 BU ml(-1)) when compared with the unsupplemented medium. For this reason, mussel-processing waste warrants further investigation due to its potential use as a cheap culture medium for upscaling bacteriocin production.  相似文献

A synthetic medium SM-3 has been elaborated for growth of Streptococcus lactis strain 51, which contains the minimal number of organic components required for the growth of this strain and nisin production. This medium contains 9 amino acids, 4 vitamins from B group, glucose and mineral salts. Addition of biotin to the medium stimulated the growth of the strain, while the addition of purines and/or pyrimidines had no effect. Hitherto biotin has been considered to be necessary for the growth of S. lactis and purines and pyrimidines were believed to stimulate the growth of these bacteria. In strain 51 the minimal requirements for growth were also the minimal requirements for nisin biosynthesis. Strain 51 produced 3-4 times less nisin in medium SM-3 than in a complex medium. The addition of one of four amino acids (serine, proline, cysteine or cystine) to SM-3 medium increased the amount of antibiotic produced. The addition of all four amino acids simultaneously, caused formation of nisin amounts similar to those produced in complex medium.  相似文献

Abstract: The nature of cysteine and cystine uptake from the cerebral capillary lumen was studied in the rat using the carotid injection technique. [35S]-Cysteine uptake was readily inhibited by the synthetic amino acid 2-amino-bicyclo(2,2,1)heptane-2-carboxylic acid (BCH), the defining substrate for the leucine-preferring (L) system in the Ehrlich ascites cell. The addition of non-radioactive alanine or serine, representatives of the alanine, serine, and cysteine-preferring (ASC) system, produced no significant decrease in the uptake of cysteine after cysteine transport by the L system was blocked with BCH. This indicated that the major component of cysteine's transport from the brain capillary lumen was by the L system with no detectable uptake of cysteine by the ASC system. No carrier-mediated transport of cystine, the disulfide form of the amino acid, was detected, nor was there any inhibition by cystine of the transport of the neutral amino acid methionine or the basic amino acid arginine. These results suggest that the ASC system, if present, is not quantitatively important for the transport of neutral amino acids from the brain capillary lumen.  相似文献

Sixteen Pseudomonas aeruginosa strains, including patent strain NRRL B-18602, three recent isolates from composted materials amended with ricinoleic acid, and 12 randomly selected from the holdings of the ARS Culture Collection, were examined for their fatty acid converting abilities. The study examined the bioconversion of oleic acid to 7,10-dihydroxy-8(E)-octadecenoic acid (DOD) and ricinoleic acid to 7,10,12-trihydroxy-8(E)-octadecenoic acid (TOD). A new DOD-like compound from linoleic acid was observed. All strains except NRRL B-247 exhibited varying levels of DOD production. NRRL B-1000, NRRL B-18602 and NRRL B-23258 with yields up to 84% were among the best DOD producers. TOD production generally paralleled DOD production at a relatively lower yield of up to 15%. Strains NRRL B-1000 and NRRL B-23260 were the best TOD producers. A DOD-like product in low yields was obtained from linoleic acid. The fatty acid bioconversion capability was related neither to growth rate nor to variation in the greenish pigmentation of the strains. Production of significant quantities of DOD and TOD from oleic and ricinoleic acids, respectively, appeared to be a characteristic trait of P. aeruginosa strains. A number of highly effective strains for DOD production were identified.  相似文献

The effect of different amino acid supplements to the basal medium on poly(3-hydroxybutyrate) (PHB) accumulation by recombinant pha Sa + Escherichia coli (ATCC: PTA-1579) harbouring the poly(3-hydroxybutyrate)-synthesizing genes from Streptomyces aureofaciens NRRL 2209 was studied. With the exception of glycine and valine, all other amino acid supplements brought about enhancement of PHB accumulation. In particular, cysteine, isoleucine or methionine supplementation increased PHB accumulation by 60, 45 and 61% respectively by the recombinant E. coli as compared with PHB accumulation by this organism in the basal medium. The effect of co-ordinated addition of assorted combinations of these three amino acids on PHB accumulation was studied using a 23 factorial design. The three-factor interaction analyses revealed that the effect of the three amino acids on PHB accumulation by the recombinant E. coli was in the order of cysteine > methionine > isoleucine. The defined medium supplemented with cysteine, methionine and isoleucine at the concentration of 150 mgl–1 each and glycerol as the carbon source was the optimum medium that resulted in the accumulation of about 52% PHB of cell dry weight.  相似文献

A general procedure was devised for the determination of growth factor requirements of heterotrophic bacteria based upon identification of individual nutrients as they are successively depleted from a limited quantity of complex medium. By using this approach, it was possible to develop a defined medium for growth of Moraxella nonliquefaciens that contained nine amino acids and three vitamins. Three of the amino acids, proline, serine, and cysteine, were required in unusually high concentrations to obtain optimal growth. Methionine had a sparing action on the requirements for serine and cysteine. Glycine could substitute for serine. Although a required nutrient, cysteine was inhibitory for growth, but this inhibitory action was antagonized by valine or leucine. The requirement for cysteine was satisfied by cystine, glutathione, or sodium sulfide. M. nonliquefaciens could not use ammonia as a nitrogen source but could use glutamate or aspartate for this purpose. With the exception of 1 auxotrophic strain, the growth factor requirements of 23 independently isolated strains of M. nonliquefaciens were essentially the same.  相似文献

All cysteine and cystine in a protein are derivatized during hydrolysis in hydrochloric acid containing 3,3'-dithiodipropionic acid. The resulting derivative can be separated from other amino acids and used for quantitation of cysteine plus half-cystine. A procedure is presented for accurate determination by ion exchange chromatography and postcolumn derivatization of all amino acids from acid hydrolysis of a protein, including the Cys-derivative.  相似文献

Studies were carried out on the influence of various amino acids: cysteine, cystine, methionine, alanine, serine, tryptophan, and of thiourea and hydroquinone on the activity of cellulase synthesized by four strains of Fusarium. It follows from the results obtained that the addition of amino acids, thiourea or hydroquinone to the culture media stimulates or inhibits the activity of the enzyme studied.
