Background

The pulmonary residence time of inhaled glucocorticoids as well as their rate and extend of absorption into systemic circulation are important facets of their efficacy-safety profile. We evaluated a novel approach to elucidate the pulmonary absorption of an inhaled glucocorticoid. Our objective was to monitor and compare the combined process of drug particle dissolution, pro-drug activation and time course of initial distribution from human lung tissue into plasma for two different glucocorticoid formulations.

Methods

We chose beclomethasone dipropionate (BDP) delivered by two different commercially available HFA-propelled metered dose inhalers (Sanasthmax®/Becloforte™ and Ventolair®/Qvar™). Initially we developed a simple dialysis model to assess the transfer of BDP and its active metabolite from human lung homogenate into human plasma. In a novel experimental setting we then administered the aerosols into the bronchus of an extracorporally ventilated and reperfused human lung lobe and monitored the concentrations of BDP and its metabolites in the reperfusion fluid.

Results

Unexpectedly, we observed differences between the two aerosol formulations Sanasthmax®/Becloforte™ and Ventolair®/Qvar™ in both the dialysis as well as in the human reperfusion model. The HFA-BDP formulated as Ventolair®/Qvar™ displayed a more rapid release from lung tissue compared to Sanasthmax®/Becloforte™. We succeeded to explain and illustrate the observed differences between the two aerosols with their unique particle topology and divergent dissolution behaviour in human bronchial fluid.

Conclusion

We conclude that though the ultrafine particles of Ventolair®/Qvar™ are beneficial for high lung deposition, they also yield a less desired more rapid systemic drug delivery. While the differences between Sanasthmax®/Becloforte™ and Ventolair®/Qvar™ were obvious in both the dialysis and lung perfusion experiments, the latter allowed to record time courses of pro-drug activation and distribution that were more consistent with results of comparable clinical trials. Thus, the extracorporally reperfused and ventilated human lung is a highly valuable physiological model to explore the lung pharmacokinetics of inhaled drugs.  相似文献

Concentric multilamellar microvesicles, named spherulites™, were evaluated as an oligonucleotide carrier. Up to 80% oligonucleotide was encapsulated in these vesicles. The study was carried out on two different spherulite™ formulations. The spherulite™ size and stability characteristics are presented. Delivery of encapsulated oligonucleotide was performed on a rat hepatocarcinoma and on a lymphoblastoid T cell line, both expressing the luciferase gene. We showed that spherulites™ were able to transfect both adherent and suspension cell lines and deliver the oligonucleotide to the nucleus. Moreover, 48–62% luciferase inhibition was obtained in the rat hepatocarcinoma cell line when the antisense oligonucleotide targeted to the luciferase coding region was encapsulated at 500 nM concentration in spherulites™ of different compositions.  相似文献

Retinoblastoma gene (Rb1) is required for proper cell cycle exit in the developing mouse inner ear and its deletion in the embryo leads to proliferation of sensory progenitor cells that differentiate into hair cells and supporting cells. In a conditional hair cell Rb1 knockout mouse, Pou4f3-Cre-pRb™/™, pRb™/™ utricular hair cells differentiate and survive into adulthood whereas differentiation and survival of pRb™/™ cochlear hair cells are impaired. To comprehensively survey the pRb pathway in the mammalian inner ear, we performed microarray analysis of pRb™/™ cochlea and utricle. The comparative analysis shows that the core pathway shared between pRb™/™ cochlea and utricle is centered on e2F, the key pathway that mediates pRb function. A majority of differentially expressed genes and enriched pathways are not shared but uniquely associated with pRb™/™ cochlea or utricle. In pRb™/™ cochlea, pathways involved in early inner ear development such as Wnt/β-catenin and Notch were enriched, whereas pathways involved in proliferation and survival are enriched in pRb™/™ utricle. Clustering analysis showed that the pRb™/™ inner ear has characteristics of a younger control inner ear, an indication of delayed differentiation. We created a transgenic mouse model (ER-Cre-pRbflox/flox) in which Rb1 can be acutely deleted postnatally. Acute Rb1 deletion in the adult mouse fails to induce proliferation or cell death in inner ear, strongly indicating that Rb1 loss in these postmitotic tissues can be effectively compensated for, or that pRb-mediated changes in the postmitotic compartment result in events that are functionally irreversible once enacted. This study thus supports the concept that pRb-regulated pathways relevant to hair cell development, encompassing proliferation, differentiation and survival, act predominantly during early development.Key words: hair cells, retinoblastoma, Rb1, proliferation, regeneration, apoptosis, inner ear  相似文献

Abstract Grazing behavior of both individual cells and populations of the marine hypotrich Euplotes mutabilis, a largely benthic ciliate that feeds on suspended particles, was studied using fluorescent latex microspheres. Microspheres of sizes 0.57-, 1.90-, 3.06-, 5.66-, and 10.0-μm diam were offered at concentrations from 102 to 106 ml−1. Their uptake in a ten-min period was determined. Food particles within such ranges of size and concentration are found under natural conditions. The ciliates ingested particles of all sizes offered. Uptake rates at all concentrations were dependent upon particle size, with 1.90- and 3.06-μm diam microspheres having the highest uptake rate in all cases. For all sizes, there was an increase in the percentage of feeding cells and in the uptake rate (the number of particles ingested cell−1 h−1), with increasing particle concentration. When uptake was expressed as the volume ingested, maximum values were obtained for 5.85-μm diam microspheres at a concentration of 106 ml−1. By taking a small number of large particles, present at a low concentration in the medium, a ciliate can ingest a greater biovolume than by taking a high number of small particles which are abundant in the medium. These results demonstrate that some ciliates can graze particles of a wide range of sizes. Also, its nutrition can be affected by changing ambient concentrations of different prey, both through effects on the proportion of feeding cells and through the relative food content of the particles. The data can also add to the understanding of food niche partitioning between species. At low particle concentrations, particularly, it is important to consider the behavior of individual ciliates as well as of the whole population. Several studies have proposed that the microbiological quality of the air in operating theatres be indirectly evaluated by means of particle counting, a technique derived from industrial clean-room technology standards, using airborne particle concentration as an index of microbial contamination. However, the relationship between particle counting and microbiological sampling has rarely been evaluated and demonstrated in operating theatres. The aim of the present study was to determine whether particle counting could predict microbiological contamination of the air in an operating theatre during 95 surgical arthroplasty procedures. This investigation was carried out over a period of three months in 2010 in an orthopedic operating theatre devoted exclusively to prosthetic surgery. During each procedure, the bacterial contamination of the air was determined by means of active sampling; at the same time, airborne particulate contamination was assessed throughout the entire procedure. On considering the total number of surgical operations, the mean value of the total bacterial load in the center of the operating theatre proved to be 35 CFU/m3; the mean particle count was 4,194,569 no./m3 for particles of diameter ≥0.5 µm and 13,519 no./m3 for particles of diameter ≥5 µm. No significant differences emerged between the median values of the airborne microbial load recorded during the two types of procedure monitored. Particulates with a diameter of ≥0.5 µm were detected in statistically higher concentrations (p<0.001) during knee-replacement procedures. By contrast, particulates with a diameter of ≥5 µm displayed a statistically higher concentration during hip-replacement procedures (p<0.05). The results did not reveal any statistically significant correlation between microbial loads and particle counts for either of the particle diameters considered (≥0.5 µm and ≥5 µm). Consequently, microbiological monitoring remains the most suitable method of evaluating the quality of air in operating theatres.  相似文献

Summary The photosynthetic pigments of the marine algaCryptomonas maculata are decreased under energy fluence rates of 4.4 Wm–2 (high light=HL). That this is a result of nitrogen deprivation following an increased cell growth rate triggered by high light in comparison to the control under 1.28 Wm–2 (low light=LL) is evident from combined pigment, growth rate and nutrient analyses. Fine structural studies by electron microscopy revealed that the electron opaque material in the thylakoid lumen of the plastid is lost under high light treatment parallel to the severe loss of almost 90% phycoerythrin-545. The reduction of chlorophylla andc is accompanied by a reversible disorganization of the thylakoid packing and the thylakoid membranes.In a combined freeze fracture study of HL and LL cells ofCryptomonas maculata it is demonstrated that the exoplasmic fracture face of the thylakoid membranes in HL cells possessed only 10 to 15% of the particles of the LL control; the 12.5 nm particle class was almost lacking, whereas particle populations with main sizes of 10 and 7.5 nm are preserved.The protoplasmic face, on the other hand, was less severely affected with only slight reduction in the particle frequency and a shift of the particle size from two populations with peaks at 10 and 7.5 nm to one class centred around 7.0 nm.  相似文献

The Purification Of B. megatherium G phage is described and it is shown that DEAE cellulose chromatography combined with conventional methods gave a phage preparation which was at least 95 per cent pure, and contained 2.16 µg nitrogen/1011 infective particles. The phage particle weight in molecular weight units was 91 x 106. The small amount of contaminating material appeared to represent phage "ghosts." An essentially 1:1 ratio of particles to infective units was found when data from electron microscopic counts or data from chemical analysis were related to phage infectivity. Comparison, by several methods, of the G phage and coliphage T2 shows that T2 is 2.6 times larger than G phage. The specific activity of the muralytic component obtained by disintegration of phage preparations with urea was unchanged by the purification indicating that the phage-"bound" muralytic activity is an integral part of the phage structure.  相似文献

CD4+ T cell enumeration is used to determine eligibility for antiretroviral therapy (ART) and to monitor the immune status of HIV-positive patients; however, many patients do not have access to this essential diagnostic test. Introducing point of care (POC) testing may improve access. We have evaluated Alere’s PIMA™, one such POC device, against conventional CD4+ testing platforms to determine its performance and validity for use in Kenya. In our hands, Alere PIMA™ had a coefficient of variability of 10.3% and of repeatability of 175.6 cells/µl. It differed from both the BD FACSCalibur™ (r2 = 0.762, mean bias −64.8 cells/µl), and the BD FACSCount™ (r2 = 0.874, mean bias 7.8 cells/µl). When compared to the FACSCalibur™ at a cutoff of 350 cells/µl, it had a sensitivity of 89.6% and a specificity of 86.7% in those aged 5 years and over (Kw = 0.7566). With the BD FACSCount™, it had a sensitivity of 79.4% and a specificity of 83.4% in those aged 5 years and over (Kw = 0.7790). The device also differed from PARTEC Cyflow™ (r2 = 0.781, mean bias −24.2 cells/µl) and GUAVA™ (r2 = 0.658, mean bias −0.3 cells/µl) platforms, which are used in some facilities in Kenya. We conclude that with refinement, Alere PIMA™ technology has potential benefits for HIV-positive patients. This study highlights the difficulty in selecting the most appropriate reference technology for technical evaluations.  相似文献

Pressurized metered dose inhalers (pMDIs) are frequently used for the treatment of asthma and chronic obstructive pulmonary disease. The aerodynamic particle size distribution (APSD) of the residual particles delivered from a pMDI plays a key role in determining the amount and region of drug deposition in the lung and thereby the efficacy of the inhaler. In this study, a simulation model that predicts the APSD of residual particles from suspension pMDIs was utilized to identify the primary determinants for APSD. These findings were then applied to better understand the effect of changing drug concentration and micronized drug size on experimentally observed APSDs determined through Andersen Cascade Impactor testing. The experimental formulations evaluated had micronized drug mass median aerodynamic diameters (MMAD) between 1.2 and 2.6 μm and drug concentrations ranging from 0.01 to 1% (w/w) with 8.5% (w/w) ethanol in 1,1,1,2-tetrafluoroethane (HFA-134a). It was determined that the drug concentration, micronized drug size, and initially atomized droplet distribution have a significant impact in modulating the proportion of atomized droplets that contain multiple suspended drug particles, which in turn increases the residual APSD. These factors were found to be predictive of the residual particle MMAD for experimental suspension HFA-134a formulations containing ethanol. Worldwide increases in fluvial fine sediment are a threat to aquatic animal health. Fluvial fine sediment is always a mixture of particles whose mineralogical composition differs depending on the sediment source and catchment area geology. Nonetheless, whether particle impact in aquatic organisms differs between mineral species remains to be investigated. This study applied an in vitro approach to evaluate cytotoxicity and uptake of four common fluvial mineral particles (quartz, feldspar, mica, and kaolin; concentrations: 10, 50, 250 mg L−1) in the rainbow trout epithelial gill cell line RTgill-W1. Cells were exposed for 24, 48, 72, and 96 h. Cytotoxicity assays for cell membrane integrity (propidium iodide assay), oxidative stress (H2DCF-DA assay), and metabolic activity (MTT assay) were applied. These assays were complemented with cell counts and transmission electron microscopy. Regardless of mineral species, particles ≤2 µm in diameter were taken up by the cells, suggesting that particles of all mineral species came into contact and interacted with the cells. Not all particles, however, caused strong cytotoxicity: Among all assays the tectosilicates quartz and feldspar caused sporadic maximum changes of 0.8–1.2-fold compared to controls. In contrast, cytotoxicity of the clay particles was distinctly stronger and even differed between the two particle types: mica induced concentration-dependent increases in free radicals, with consistent 1.6–1.8-fold-changes at the 250 mg L−1 concentration, and a dilated endoplasmic reticulum. Kaolin caused concentration-dependent increases in cell membrane damage, with consistent 1.3–1.6-fold increases at the 250 mg L−1 concentration. All effects occurred in the presence or absence of 10% fetal bovine serum. Cell numbers per se were marginally affected. Results indicate that (i.) natural mineral particles can be cytotoxic to gill epithelial cells, (ii.) their cytotoxic potential differs between mineral species, with clay particles being more cytotoxic, and (iii.) some clays might induce effects comparable to engineered nanoparticles.  相似文献

A novel cation-exchange resin, Eshmuno™ S, was compared to Fractogel® SO3 (M) and Toyopearl GigaCap S-650M. The stationary phases have different base matrices and carry specific types of polymeric surface modifications. Three monoclonal antibodies (mAbs) were used as model proteins to characterize these chromatographic resins. Results from gradient elutions, stirred batch adsorptions and confocal laser scanning microscopic investigations were used to elucidate binding behavior of mAbs onto Eshmuno™ S and Fractogel® SO3 and the corresponding transport mechanisms on these two resins. The number of charges involved in mAb binding for Eshmuno™ S is lower than for Fractogel® SO3, indicating a slightly weaker electrostatic interaction. Kinetics from batch uptake experiments are compared to kinetic data obtained from confocal laser scanning microscopy images. Both experimental approaches show an accelerated protein adsorption for the novel stationary phase. The influence of pH, salt concentrations and residence times on dynamic binding capacities was determined. A higher dynamic binding capacity for Eshmuno™ S over a wider range of pH values and residence times was found compared to Fractogel® SO3 and Toyopearl GigaCap S-650M. The capture of antibodies from cell culture supernatant, as well as post-protein A eluates, were analyzed with respect to their host cell protein (hcp) removal capabilities. Clostridium thermocellum JW20 and YM4 both synthesize cellulolytic enzyme complexes, cellulosomes, when grown on medium containing cellulose. Electron microscopic studies showed that, in the early stages of growth of strain JW20, clusters of tightly packed cellulosomes, i.e., polycellulosomes, were located on the cell surface and were bound to cellulose. The polycellulosome was estimated to have a particle mass of 50 × 106 to 80 × 106 daltons (Da), while that of the cellulosome was estimated to be 2 × 106 to 2.5 × 106 Da and to contain about 35 polypeptides ranging from 20 to 200 kDa. The cellulosome produced by strain YM4 was found to be somewhat larger, with the estimated particle mass being 3.5 × 106 Da, and the number of polypeptides was counted to be 45 to 50, ranging from 20 to 200 kDa. In the early stages of cultivation, the cellulosomes from both species exist as tightly packed complexes (tight cellulosomes). These subsequently decompose to loosely packed complexes (loose cellulosomes) and ultimately to free polypeptides. Examination of the loose cellulosomal particles showed that they contain rows of equidistantly spaced, similarly sized polypeptide subunits, with an apparently identical orientation arranged parallel to the major axis of the cellulosome. It is postulated that on binding of a cellulose chain alongside such a row of subunits a simultaneous multicutting event occurs that leads to the release of cellooligosaccharides of four cellobiose units in length (C4). Rows of smaller-sized subunits with lower center-to-center distances, which are also present in the cellulosome, subsequently cleave the C4 fragments (or cellulose) to C2 (cellotetraose) or C1 (cellobiose). In this way the cellulosome can catalyze the complete hydrolysis of cellulose.  相似文献

Gel filtration (GF) is an excellent tool to acquire information about sizing (identity), purity, and the multimeric state of a protein of interest. Superdex™ 200 and Superdex 75 are outstanding GF media for such analysis. To speed up analysis and keep sample and buffer consumption at minimum, two prepacked short GF columns have been developed, Superdex™ 200 5/150 GL and Superdex 75 5/150 GL. With lengths of only 15 cm and volume of 3 ml, these columns allow rapid analysis (6–12 min/run) with minimal sample (4–50μl) and buffer consumption.Purification of antibodies often generates dimers and higher aggregated forms, and during optimization of purification protocols, many samples may need to be analyzed for aggregate content. Gel filtration (GF) provides reliable information about size and the purity of a protein of interest, especially when it is in a multimeric state. GF, however, is often time-consuming, and the analyses become a bottleneck. A new column format—3 ml bed vol and 15 cm long—has been developed, enabling rapid and reliable GF with low sample and buffer consumption, using Superdex™ 75 and Superdex™ 200 media for determination of size and purity status.  相似文献

In recent years, several automated scale-down bioreactor systems have been developed to increase efficiency in cell culture process development. ambr™ is an automated workstation that provides individual monitoring and control of culture dissolved oxygen and pH in single-use, stirred-tank bioreactors at a working volume of 10–15 mL. To evaluate the ambr™ system, we compared the performance of four recombinant Chinese hamster ovary cell lines in a fed-batch process in parallel ambr™, 2-L bench-top bioreactors, and shake flasks. Cultures in ambr™ matched 2-L bioreactors in controlling the environment (temperature, dissolved oxygen, and pH) and in culture performance (growth, viability, glucose, lactate, Na+, osmolality, titer, and product quality). However, cultures in shake flasks did not show comparable performance to the ambr™ and 2-L bioreactors.  相似文献

1. A procedure is described for preparing 131I-labelled adrenocorticotrophin suitable for use in radioimmunoassay. 2. Adsorption of labelled and unlabelled adrenocorticotrophin at low concentrations occurs to various surfaces despite the presence of diluent protein. Adsorption and desorption errors are minimized by low pH and by the use of polystyrene vials. 3. Preparations with low initial damage are obtained if the radioiodination is performed rapidly and the separation of 131I-labelled adrenocorticotrophin from unchanged [131I]iodide is carried out on cellulose columns by using dilute acid. 4. The immunological activity of 131I-labelled α1–24-adrenocorticotrophin, but not of 131I-labelled porcine adrenocorticotrophin, decreases with increasing specific radioactivity. The involvement of tyrosine residues in the immunological specificity of the α1–24-adrenocorticotrophin only is suggested to explain this finding.  相似文献

Background

Ingestion of AndoSan, based on the mushroom Agaricus blazei Murill, has previously been shown to exhibit anti-inflammatory effects because of reduction of pro-inflammatory cytokines in healthy individuals and patients with ulcerative colitis. In this randomized single-blinded placebo controlled study we examined whether intake of AndoSan also resulted in clinical effects.

Methods and Findings

50 patients with symptomatic ulcerative colitis were block-randomized and blinded for oral daily intake of AndoSan or placebo for the 21 days’ experimental period. The patients reported scores for symptoms, fatigue and health related quality of life (HRQoL) at days 0, 14 and 21. Fecal calprotectin and general blood parameters were also analyzed. In the AndoSan group (n = 24) symptoms improved from baseline (day 0) to days 14 and 21, with respective mean scores (95% CI) of 5.88 (4.92–6.83), 4.71 (3.90–5.52) (p = 0.002) and 4.50 (3.70–5.30) (p = 0.001). Corresponding improved mean scores (±SD) for total fatigue were 16.6 (5.59), 14.1 (4.50) (p = 0.001) and 15.1 (4.09) (p = 0.023). These scores in the placebo group (n = 26) were not improved. When comparing the two study groups using mixed model statistics, we found significant better scores for the AndoSan-patients. HRQoL for dimensions bodily pain, vitality, social functioning and mental health improved in the AndoSan group. There were no alterations in general blood samples and fecal calprotectin.

Conclusions

Beneficiary effects on symptoms, fatigue and HRQoL from AndoSan consumption were demonstrated in this per-protocol study, supporting its use as a supplement to conventional medication for patients with mild to moderate symptoms from ulcerative colitis. The patients did not report any harms or unintended effects of AndoSan in this study.

Determination of lipoprotein particle size and number using advanced lipoprotein tests (ALTs) is of particular importance to improve cardiovascular risk prediction. Here we present the Liposcale test, a novel ALT based on 2D diffusion-ordered 1H NMR spectroscopy. Our method uses diffusion coefficients to provide a direct measure of the mean particle sizes and numbers. Using 177 plasma samples from healthy individuals and the concentration of ApoB and ApoA from isolated lipoprotein fractions, our test showed a stronger correlation between the NMR-derived lipoprotein particle numbers and apolipoprotein concentrations than the LipoProfile® test commercialized by Liposcience. We also converted LDL particle numbers to ApoB equivalents (milligrams per deciliter) and our test yielded similar values of LDL-ApoB to the LipoProfile® test (absolute mean bias of 8.5 and 7.4 mg/dl, respectively). In addition, our HDL particle number values were more concordant with the calibrated values determined recently using ion mobility. Finally, principal component analysis distinguished type 2 diabetic patients with and without atherogenic dyslipidemia (AD) on a second cohort of 307 subjects characterized using the Liposcale test (area under the curve = 0.88) and showed concordant relationships between variables explaining AD. Altogether, our method provides reproducible and reliable characterization of lipoprotein particles and it is applicable to pathological states such as AD.  相似文献

When logarithmically growing cultures of Clostridium botulinum types A and E are treated with penicillin in a liquid medium containing 8% polyethylene glycol, protoplast-like spherical bodies are formed. The penicillin effect shows a dose-response relationship; the largest yield of converted forms is obtained with 10,000 units/ml, but the treatment leaves many intact bacilli. Lower antibiotic concentrations produce smaller numbers of spherical bodies, but lysis of bacilli results in suspensions that are relatively free of rods. Cells grown under the same conditions and treated with 250 μg of lysozyme/ml do not form spherical bodies. However, a combination of 1,250 to 2,500 units of penicillin and 100 μg of lysozyme/ml yields suspensions which have sphere counts in excess of 1.0 × 108/ml and only a few intact rods. The state of the culture at the time of addition of the antibiotic and enzyme is critical. Suspensions of these protoplasts can be adapted to grow as stable L-form cultures producing the same toxin type as the parent cultures.  相似文献

Summary The densities and diameters of intramembranous particles in olfactory and nasal respiratory structures of frog, ox, rat and dog have been compared using the freeze-fracture technique. Dendritic endings and the various segments of the cilia of the olfactory receptor cells of a given species have identical particle densities (700–1,800 particles/m2 in P-and 100–600 in E-faces). Densities in P-faces of respiratory cilia are about 1/3 of those in the olfactory cilia. E-face particle densities of these respiratory cilia are often higher than P-face densities. Microvillus P-face densities range from 700–2,000 (respiratory cell microvilli) to 1,800–3,400 particles/m2 (olfactory supporting and Bowman's gland microvilli). Microvillus E-faces show no conspicuous mutual differences. Literature comparisons showed that odour concentrations at threshold are considerably lower (105–1010 times) than the concentrations of olfactory receptor ending intramembranous particles (5 M–30 M) expressed in the same units.Relative differences in particle distributions of the various cell structures studied are usually species-independent. Absolute values vary considerably with the species. Relative P-face particle densities of the supporting cell microvilli tend to correlate with those of dendritic ending structures. Particle diameters are usually similar for corresponding structures and fracture faces in the four species. Apical structures of supporting and Bowman's gland cells in rat and dog show rod-shaped particle aggregates in their P-and pits in their E-faces. Neither sex-dependency nor an influence related to physiological treatments on the particle distributions could be demonstrated.  相似文献
