由于线粒体在生物氧化和能量转换过程中会产生活性氧,线粒体DNA又比核DNA更容易发生突变,因此线粒体是一种比较容易受到损伤的细胞器.及时清除细胞内受损的线粒体对细胞维持正常的状态具有重要的作用.细胞主要通过自噬来清除损伤线粒体,维持细胞稳态.越来越多的研究表明,线粒体自噬是一种特异性的过程,线粒体通透性孔道通透性的改变在这个过程中起着重要的作用.线粒体自噬在维持细胞内线粒体的正常功能和基因组稳定性上起着重要作用,但是线粒体发生自噬的信号通路及其调控机制还有待进一步深入研究.  相似文献

线粒体自噬是指细胞通过自噬的机制选择性地清除线粒体的过程。通过该途径,细胞可降解并清除受损或功能障碍的线粒体,以维持细胞内线粒体质量和数量的平衡,从而维持细胞稳态。在生理状态及应激状态下,多种因子可调控心肌细胞线粒体自噬,进而发挥保护心肌细胞的作用。本文就线粒体自噬及其调控机制以及其在心肌保护中的作用做一综述。  相似文献

线粒体是一种结构和功能复杂而敏感的细胞器,拥有独立于细胞核的基因组,在细胞的不同时相,生理过程和环境条件下,线粒体的形态,数量和质量,具有高度的可塑性。线粒体是细胞和生物体内最主要的能量供应场所,几乎存在于所有种类的细胞中,是一种动态变化的细胞器。正常情况下,线粒体的数量、形态以及功能维持相对稳定的状态,称之为线粒体稳态。当上述状态发生紊乱时,细胞乃至生物体形态、功能也将受到影响甚至死亡。线粒体质量控制是在细胞中维持正常状态的关键机制,决定着线粒体的命运。近年,随着线粒体研究的深入和具体,逐渐发现融合/分裂在其形态、数量、遗传物质等质量控制相关的方面挥了重要作用。本文通过探讨融合/分裂对线粒体质量控制的作用机制,总结和讨论相关前沿研究,为后期研究提供一定的理论依据。  相似文献

线粒体作为细胞的能量中心,在细胞内呈现高度的动态变化,其数量、质量及功能的稳定对维持细胞的正常活动至关重要。线粒体动力学与线粒体自噬之间可互相调控,共同构成线粒体质量控制的重要环节。泛素特异性蛋白酶30 (USP30)作为去泛素化酶,既可通过线粒体融合蛋白1/2 (Mfn1/2)、线粒体动力蛋白相关蛋白1 (Drp1)等融合与分裂蛋白参与调控线粒体动力学过程,还能通过E3泛素连接酶Parkin、泛素(Ub)及电压依赖性阴离子通道1 (VDAC1)等多种信号而调控PTEN诱导激酶1 (PINK1)/Parkin途径介导的线粒体自噬,但其详细机制尚未完全阐明。本文对USP30在调控线粒体动力学和线粒体自噬中的作用与其机制进行了综述。  相似文献

由于线粒体能敏感地感受机体内氧浓度的变化,缺氧时会影响线粒体氧化磷酸化过程中电子传递链的正常功能,抑制ATP生成,产生大量活性氧(ROS)。ROS蓄积导致氧化损伤细胞内脂质、DNA和蛋白质等大分子物质,线粒体肿胀,通透性转换孔开放,释放细胞色素C等促凋亡因子,最终严重影响细胞的存活。因此这些功能异常或受损线粒体是缺氧应激状态下细胞是否存活的危险因素,及时清除这些线粒体,对维持线粒体质量、数量及细胞稳态具有重要意义。线粒体自噬是近年来发现的细胞适应缺氧的一种防御性代谢过程,它通过自噬途径选择性清除损伤、衰老和过量产生ROS的线粒体,促进线粒体更新和循环利用,确保细胞内线粒体功能稳定,保护缺氧应激下细胞的正常生长发挥重要的调节作用。本文就线粒体自噬在缺氧条件下发生过程、参与相关蛋白及调节机制等方面研究进行了综述。  相似文献

线粒体自噬指细胞通过自噬机制选择性除去损伤或多余的线粒体。真核生物通过线粒体自噬调控线粒体质量,维持供能细胞器的功能。大量研究表明,帕金森病相关基因PINK1和parkin可通过线粒体自噬参与并维持线粒体功能。PINK1与parkin能协同特异性识别损伤的线粒体,PINK1作为线粒体质量调控的探测器被活化,此过程中泛素化酶和去泛素化酶对维持parkin活性及线粒体自噬的效率有重要作用。本文主要总结PINK1/parkin通路在线粒体自噬中的功能与作用。  相似文献

线粒体为细胞正常生命运动提供能量和物质;然而各种因素会导致线粒体损伤,衰老及功能紊乱,它们是细胞潜在的危险因素,必需及时清除,线粒体自噬可以起到这一作用,维持细胞稳态。当细胞处于恶劣环境时,线粒体自噬可通过降解线粒体补充生命必需物质,从而度过危机维持生存。另外线粒体自噬会在某些情况下通过降解正常线粒体来维持线粒体质量和数量的平衡。不同生物中具有不同的线粒体自噬途径和机制,酵母中主要通过Atg32磷酸化调控线粒体自噬;哺乳动物中则存在分别由Parkin-PINK1、Nix、FUNDC1等不同蛋白介导的线粒体自噬调控机制;植物线粒体自噬的研究主要集中在拟南芥,其途径及具体调控机制尚不明确。综述了近年来酵母、动物和植物中线粒体自噬的作用机制及调控因子等方面的研究进展。  相似文献

The accumulation of unhealthy mitochondria results in mitochondrial dysfunction, which has been implicated in aging, cancer, and a variety of degenerative diseases. However, the mechanism by which mitochondrial quality is regulated remains unclear. Here, we show that Mieap, a novel p53-inducible protein, induces intramitochondrial lysosome-like organella that plays a critical role in mitochondrial quality control. Mieap expression is directly regulated by p53 and is frequently lost in human cancer as result of DNA methylation. Mieap dramatically induces the accumulation of lysosomal proteins within mitochondria and mitochondrial acidic condition without destroying the mitochondrial structure (designated MALM, for Mieap-induced accumulation of lysosome-like organelles within mitochondria) in response to mitochondrial damage. MALM was not related to canonical autophagy. MALM is involved in the degradation of oxidized mitochondrial proteins, leading to increased ATP synthesis and decreased reactive oxygen species generation. These results suggest that Mieap induces intramitochondrial lysosome-like organella that plays a critical role in mitochondrial quality control by eliminating oxidized mitochondrial proteins. Cancer cells might accumulate unhealthy mitochondria due to p53 mutations and/or Mieap methylation, representing a potential cause of the Warburg effect.  相似文献

Maintenance of healthy mitochondria prevents aging, cancer, and a variety of degenerative diseases that are due to the result of defective mitochondrial quality control (MQC). Recently, we discovered a novel mechanism for MQC, in which Mieap induces intramitochondrial lysosome-like organella that plays a critical role in the elimination of oxidized mitochondrial proteins (designated MALM for Mieap-induced accumulation of lysosome-like organelles within mitochondria). However, a large part of the mechanisms for MQC remains unknown. Here, we report additional mechanisms for Mieap-regulated MQC. Reactive oxygen species (ROS) scavengers completely inhibited MALM. A mitochondrial outer membrane protein NIX interacted with Mieap in a ROS-dependent manner via the BH3 domain of NIX and the coiled-coil domain of Mieap. Deficiency of NIX also completely impaired MALM. When MALM was inhibited, Mieap induced vacuole-like structures (designated as MIV for Mieap-induced vacuole), which engulfed and degraded the unhealthy mitochondria by accumulating lysosomes. The inactivation of p53 severely impaired both MALM and MIV generation, leading to accumulation of unhealthy mitochondria. These results suggest that (1) mitochondrial ROS and NIX are essential factors for MALM, (2) MIV is a novel mechanism for lysosomal degradation of mitochondria, and (3) the p53-Mieap pathway plays a pivotal role in MQC by repairing or eliminating unhealthy mitochondria via MALM or MIV generation, respectively.  相似文献

Mieap, a p53-inducible protein, controls mitochondrial quality by repairing unhealthy mitochondria. During repair, Mieap induces the accumulation of intramitochondrial lysosomal proteins (designated MALM for Mieap-induced accumulation of lysosome-like organelles within mitochondria) by interacting with NIX, leading to the elimination of oxidized mitochondrial proteins. Here, we report that an additional mitochondrial outer membrane protein, BNIP3, is also involved in MALM. BNIP3 interacts with Mieap in a reactive oxygen species (ROS)-dependent manner via the BH3 domain of BNIP3 and the coiled-coil domains of Mieap. The knockdown of endogenous BNIP3 expression severely inhibited MALM. Although the overexpression of either BNIP3 or NIX did not cause a remarkable change in the mitochondrial membrane potential (MMP), the co-expression of all three exogenous proteins, Mieap, BNIP3 and NIX, caused a dramatic reduction in MMP, implying that the physical interaction of Mieap, BNIP3 and NIX at the mitochondrial outer membrane may regulate the opening of a pore in the mitochondrial double membrane. This effect was not related to cell death. These results suggest that two mitochondrial outer membrane proteins, BNIP3 and NIX, mediate MALM in order to maintain mitochondrial integrity. The physical interaction of Mieap, BNIP3 and NIX at the mitochondrial outer membrane may play a critical role in the translocation of lysosomal proteins from the cytoplasm to the mitochondrial matrix.  相似文献

Mitochondria play an important role in maintaining cardiac homeostasis by supplying the major energy required for cardiac excitation–contraction coupling as well as controlling the key intracellular survival and death pathways. Healthy mitochondria generate ATP molecules through an aerobic process known as oxidative phosphorylation (OXPHOS). Mitochondrial injury during myocardial infarction (MI) impairs OXPHOS and results in the excessive production of reactive oxygen species (ROS), bioenergetic insufficiency, and contributes to the development of cardiovascular diseases. Therefore, mitochondrial biogenesis along with proper mitochondrial quality control machinery, which removes unhealthy mitochondria is pivotal for mitochondrial homeostasis and cardiac health. Upon damage to the mitochondrial network, mitochondrial quality control components are recruited to segregate the unhealthy mitochondria and target aberrant mitochondrial proteins for degradation and elimination. Impairment of mitochondrial quality control and accumulation of abnormal mitochondria have been reported in the pathogenesis of various cardiac disorders and heart failure. Here, we provide an overview of the recent studies describing various mechanistic pathways underlying mitochondrial homeostasis with the main focus on cardiac cells. In addition, this review demonstrates the potential effects of mitochondrial quality control dysregulation in the development of cardiovascular disease.  相似文献

We and others previously reported that endogenous p53 can be located at mitochondria in the absence of stress, suggesting that p53 has a role in the normal physiology of this organelle. The aim of this study was to characterize in unstressed cells the intramitochondrial localization of p53 and identify new partners and functions of p53 in mitochondria. We find that the intramitochondrial pool of p53 is located in the intermembrane space and the matrix. Of note, unstressed HCT116 p53+/+ cells simultaneously show increased O₂ consumption and decreased mitochondrial superoxide production compared with their p53-null counterpart. This data was confirmed by stable H1299 cell lines expressing low levels of p53 specifically targeted to the matrix. Using immunoprecipitation and mass spectrometry, we identified the oligomycin sensitivity-conferring protein (OSCP), a subunit of the F₁F₀-ATP synthase complex, as a new partner of endogenous p53, specifically interacting with p53 localized in the matrix. Interestingly, this interaction seems implicated in mitochondrial p53 localization. Moreover, p53 localized in the matrix promotes the assembly of F₁F₀-ATP synthase. Taking into account that deregulations of mitochondrial respiration and reactive oxygen species production are tightly linked to cancer development, we suggest that mitochondrial p53 may be an important regulator of normal mitochondrial and cellular physiology, potentially exerting tumor suppression activity inside mitochondria.  相似文献

Capsaicin is widely used as a food additive and as an analgesic agent. Besides its well-known role in nociception, which is mediated by vanilloid receptor 1 specifically expressed in dorsal root ganglion neurons, capsaicin has also been considered as a potential anticancer agent, as it inhibits cell proliferation and induces apoptosis in various types of cancer cells. Here we identified a new molecular target of capsaicin from human myeloid leukemia cells. We show that capsaicin binds to prohibitin (PHB) 2, which is normally localized to the inner mitochondrial membrane, and induces its translocation to the nucleus. PHB2 is implicated in the maintenance of mitochondrial morphology and the control of apoptosis. We also provide evidence suggesting that capsaicin causes apoptosis directly through the mitochondria and that PHB2 contributes to capsaicin-induced apoptosis at multiple levels. This work will serve as an important foundation for further understanding of anticancer activity of capsaicin.  相似文献

Transient global brain ischemia induces dysfunctions of mitochondria including disturbance in mitochondrial protein synthesis and inhibition of respiratory chain complexes. Due to capacity of mitochondria to release apoptogenic proteins, ischemia-induced mitochondrial dysfunction is considered to be a key event coupling cerebral blood flow arrest to neuronal cell death. Ischemic preconditioning (IPC) represents an important phenomenon of adaptation of central nervous system (CNS) to sub-lethal short-term ischemia, which results in increased tolerance of CNS to the lethal ischemia. In this study we have determined the effect of ischemic preconditioning on ischemia/reperfusion-associated inhibition of mitochondrial protein synthesis and activity of mitochondrial respiratory chain complexes I and IV in the hippocampus of rats. Global brain ischemia was induced by 4-vessel occlusion in duration of 15 min. Rats were preconditioned by 5 min of sub-lethal ischemia and 2 days later, 15 min of lethal ischemia was induced. Our results showed that IPC affects ischemia-induced dysfunction of hippocampal mitochondria in two different ways. Repression of mitochondrial translation induced during reperfusion of the ischemic brain is significantly attenuated by IPC. Slight protective effect of IPC was documented for complex IV, but not for complex I. Despite this, protective effect of IPC on ischemia/reperfusion-associated changes in integrity of mitochondrial membrane and membrane proteins were observed. Since IPC exhibited also inhibitory effect on translocation of p53 to mitochondria, our results indicate that IPC affects downstream processes connecting mitochondrial dysfunction to neuronal cell death.  相似文献
