An 8-week feeding trial was conducted to determine the dietary copper (Cu) requirement and its effect on the non-specific immune responses of juvenile grass shrimp, Penaeus monodon. Purified diets with seven levels (0, 10, 20, 30, 40, 80, 160 mg Cu kg diet(-1) of supplemental Cu were fed to P. monodon (mean initial weight 0.29 +/- 0.004 g). Each diet was fed to three replicate groups of shrimp. The rearing water contained 1.53 microg Cu 1(-1). Shrimp fed diets supplemented with 10 and 20 mg Cu kg diet(-1) had significantly (P < 0.01) greater weight gain, feed efficiency (FE) and protein efficiency ratio (PER) than those fed the unsupplemented control diet and diets supplemented with > or = 40 mg Cu kg diet(-1). Whole body Cu concentration in shrimp generally increased as dietary Cu supplementation increased. Total haemocyte count (THC) was higher in shrimp fed diets supplemented with 10-30 mg Cu kg diet(-1) than shrimp fed the unsupplemented control diet and diets supplemented with > or = 40 mg Cu kg diet(-1). Intracellular superoxide anion (O2-) production ratios were significantly higher in shrimp fed diets supplemented with 10-30 mg Cu kg diet(-1) than shrimp fed the diet supplemented with 160 mg Cu kg diet(-1). Analysis by polynomial regression of weight gain percent, FE and by linear regression of the whole-body Cu retention of shrimp indicated that the adequate dietary Cu concentration in growing P. monodon is about 15-21 mg Cu kg diet (-1). The immune indicators suggest that an adequate dietary Cu concentration for non-specific immune responses in P. monodon is about 10-30 mg Cu kg diet(-1).  相似文献

One hundred and sixty pigs were used to evaluate dietary copper (Cu) and zinc (Zn) supplementation on performance, fecal mineral levels, body mineral status and carcass and meat quality. Diets differed in mineral form (MF) (Cu and Zn in the form of proteinate amino acid chelate (organic) or sulfate (inorganic)) and inclusion level (IL) (27 mg/kg of total Cu and 65 mg/kg of total Zn ('low') or 156 mg/kg of total Cu and 170 mg/kg of total Zn ('high')) according to a 2 × 2 factorial arrangement of treatments. Pigs were used from 25 to 107 kg body weight (BW) and fed their respective diets ad libitum. Blood and fecal samples were collected on days 14 and 77 of the experiment. Blood was analyzed for concentration of Cu and Zn, hemoglobin (Hb), Cu content of red blood cells (RBC Cu) and alkaline phosphatase (ALP) and feces for Cu and Zn concentration. Hot carcass weight (HCW) and backfat depth were measured at slaughter and indices of meat quality were assessed on a section of longissimus thoracis. Liver, kidney and bone samples were collected immediately after slaughter and liver and kidney were tested for Cu and Zn content, while bone was only tested for Zn. Over the entire experimental period (25 to 107 kg BW) no significant treatment differences in average daily gain (ADG) or average daily feed intake (ADFI) occurred; however, feed conversion ratio (FCR) was improved by the inclusion of proteinate amino acid chelate (P = 0.012). Copper and Zn concentrations in feces were in direct proportion to the IL in the diet. Blood mineral levels were within normal physiological ranges in all treatments and tissue Cu and Zn concentrations increased with dietary IL (P < 0.05). Results indicate that Cu and Zn fecal concentrations were reduced by approximately 6-fold for Cu and by 2.5-fold for Zn by feeding 27 mg/kg Cu and 65 mg/kg Zn, in either the proteinate amino acid chelate or the sulfate form, compared with a diet containing 156 mg/kg Cu and 170 mg/kg Zn. This decrease in total dietary Cu and Zn did not reduce performance or mineral status of pigs.  相似文献

BackgroundComplementary feeding of breastfed infants with foods high in bioavailable zinc (Zn) can help meet physiological requirements for Zn. Some infant cereals contain high concentrations of phytic acid (PA) and calcium (Ca) that may reduce absorbable Zn.ObjectivesThis study measured PA, Zn and Ca concentrations in selected infant cereals sold in Canada and investigated the effects of dietary PA and Ca at concentrations present in infant cereals on Zn bioavailability in rats.Methods and resultsMale Sprague-Dawley rats (36-day old) were fed a control diet containing normal Zn (29.1 mg/kg) and Ca (4.95 g/kg) or six test diets (n = 12/diet group). Test diets were low in Zn (8.91–9.74 mg/kg) and contained low (2.16–2.17 g/kg), normal (5.00–5.11 g/kg) or high (14.6–14.9 g/kg) Ca without or with added PA (8 g/kg). After 2 weeks, rats were killed and Zn status of the rats was assessed. PA, Zn and Ca concentrations in infant cereals (n = 20) differed widely. PA concentrations ranged from undetectable to 16.0 g/kg. Zn and Ca concentrations ranged from 7.0–29.1 mg/kg and 0.8–13.4 g/kg, respectively. The [PA]/[Zn] and [PA × Ca]/[Zn] molar ratios in infants cereals with detectable PA (16 of 20 cereals) ranged from 22–75 and 0.9–14.9 mol/kg, respectively, predicting low Zn bioavailability. Body weight, body composition (lean and fat mass), right femur weight and length measurements and Zn concentrations in serum and femur indicated that diets higher in Ca had a more pronounced negative effect on Zn status of rats fed a PA-supplemented diet. Addition of PA to the diet had a greater negative effect on Zn status when Ca concentration in the diet was higher.ConclusionThese results show that, in rats, higher concentrations of dietary Ca and PA interact to potentiate a decrease in bioavailable Zn and may suggest lower Zn bioavailability in infant cereals with higher PA and Ca concentrations.  相似文献

The present study was designed to investigate the effects of Zn administration on metallothionein concentrations in the liver, kidney, and intestine of copper-loaded rats. Male CD rats were fed a diet containing 12 mg Cu and 67 mg Zn/kg body wt. They were divided into either acute or chronic experimental protocols. Rats undergoing acute experiments received daily ip injections of either Cu (3 mg/kg body wt) or Zn (10 mg/kg body wt) for 3 d. Chronic experiments were carried out on rats receiving Cu ip injections on d 1, 2, 3, 10, 17, and 24, Cu injections plus a Zn-supplemented diet containing 5 g Zn/kg solid diet, or a Zn-supplemented diet alone. Rats injected Zn or Cu had increased MT concentrations in liver and kidney. Zn produced the most important effects and the liver was the most responsive organ. Rats fed a Zn-supplemented diet had significantly higher MT concentrations in liver and intestine with respect to controls. Increased MT synthesis in the liver may contribute to copper detoxification; the hypothesis of copper entrapment in enterocytes cannot be confirmed.  相似文献

The high affinity copper transporter 1 (Ctr1), metallothionein (MT) and glutathione reductase (GR) are essential for copper uptake, sequestration and defense respectively. Following rearing on a normal commercial diet (12.6+/-0.2 mg kg(-1) Cu), sea bream were fed an experimental control diet lacking mineral mix (7.7+/-0.3 mg kg(-1) Cu), an experimental diet enhanced with Cu (135+/-4 mg kg(-1) Cu) or an experimental diet (7.7+/-0.3 mg kg(-1) Cu) whilst exposed to Cu in water (0.294+/-0.013 mg L(-1)). Fish were sampled at 0, 15 and 30 days after exposures. Fish fed the Cu-enhanced experimental diet showed lower levels of expression of Ctr1 in the intestine and liver compared to fish fed control experimental diets, whilst Ctr1 expression in the gill and kidney was unaffected by excess dietary Cu exposure. Waterborne-Cu exposure increased Ctr1 mRNA levels in the intestine and the kidney compared to experimental controls. Excess dietary Cu exposure had no effect on levels of metallothionein (MT) mRNA, and the only effect of dietary excess Cu on glutathione reductase (GR) mRNA was a decrease in the intestine. Both MT mRNA and GR were increased in the liver and gill after waterborne-Cu exposure, compared to levels in fish fed experimental control low Cu diets. Thus, Ctr1, MT and GR mRNA expression in response to excess Cu is dependent on the route of exposure. Furthermore, the tissue expression profile of sea bream Ctr1 is consistent with the known physiology of copper exposure in fish and indicates a role both in essential copper uptake and in avoidance of excess dietary and waterborne copper influx.  相似文献

The teratogenic effects of feeding a diet based on textured vegetable protein to Long-Evans rats were studied along with maternal and fetal mineral interactions and their relationship to diet composition. Pregnant rats were fed purified diets containing 18% protein as casein (CAS), textured vegetable protein (TVP, from defatted soy flour) with 18 mg Zn/kg, or TVP diet with 100 mg Zn/kg. A fourth group was fed diet NIH-31. The animals received their diets throughout pregnancy and were sacrificed on day 20 of gestation. Fetuses were examined for developmental effects, and mineral levels were determined in maternal and fetal tissues by inductively coupled argon plasma-atomic emission spectrometry. Females fed the casein diet or diet NIH-31 had normal weight gains throughout pregnancy and their progeny exhibited normal development. The animals on the TVP-containing diet with 18 mg Zn/kg had decreased food consumption and body weights, and their fetuses exhibited developmental anomalies as well as reductions in size and weight. These developmental alterations may be the result of decreased zinc levels in the fetal tissues, caused by reduced bioavailability of the trace element in the maternal diet. Significant increases in tissue iron accompanied the low zinc levels. No developmental effects were found in animals receiving the high Zn-TVP diet, and mineral data from these animals were not significantly different from the casein group.  相似文献

Zinc (Zn) is an essential nutrient that is required in humans and animals for many physiological functions, including immune and antioxidant function, growth, and reproduction. The present study was performed to investigate the effects of three Zn levels, including Zn adequate (35.94 mg/kg, as a control), Zn deficiency (3.15 mg/kg), and Zn overload (347.50 mg/kg) in growing male rats for 6 wk. This allowed for evaluation of the effects that these Zn levels might have on body weight, organ weight, enzymes activities, and tissues concentrations of Zn and Cu. The results showed that Zn deficiency has negative effects on growth, organ weight, and biological parameters such as alkaline phosphatase (ALP) and Cu−Zn superoxide dismutase (Cu−Zn SOD) activities, whereas Zn overload played an effective role in promoting growth, improving the developments of organs and enhancing immune system. Hepatic metallothionein (MT) concentration showed an identical increase tendency in rats fed both Zn-deficient and Zn-overload diets. The actual mechanism of reduction of Cu concentration of jejunum in rats fed a Zn-overload diet might involve the modulation or inhibition of a Cu transporter protein by Zn and not by the induction of MT.  相似文献

Equol, a metabolite of the phytoestrogen daidzein, is present at significant levels in some humans who consume soy and in rodents fed soy-based diets. Equol is estrogenic in vitro, but there have been limited studies of its activity in vivo. We evaluated equol effects on reproductive and non-reproductive endpoints in mice. Ovariectomized age-matched (30-day-old) female C57BL/6 mice were fed phytoestrogen-free diets and given a racemic mixture of equol by daily injections (0, 4, 8, 12, or 20 mg [kg body weight](-1) day(-1)) or in the diet (0, 500, or 1,000 ppm) for 12 days. Mice were killed, and serum concentrations of total and aglycone equol were measured. Total serum equol concentrations ranged from 1.4 to 7.5 microM with increasing doses of injected equol, but uterine weight increased significantly only at 12 and 20 mg (kg body weight)(-1) day(-1). Dietary equol at 500 or 1,000 ppm produced total serum equol concentrations of 5.9 and 8.1 microM, respectively, comparable with those in rodents consuming certain high-soy chows; the proportion of equol present as the free aglycone was much lower with dietary administration than injections, which may be a factor in the greater biological effects induced by injections. Dietary equol did not significantly increase uterine weight. Increasing dietary and injected equol doses caused a dose-dependent increase in vaginal epithelial thickness. Uterine epithelial proliferation was increased by equol injections at 8-20 mg (kg body weight)(-1) day(-1) and 1,000 ppm dietary equol. Neither dietary nor injected equol decreased thymic or adipose weights. In conclusion, equol is a weak estrogen with modest effects on endpoints regulated by estrogen receptor alpha when present at serum levels seen in rodents fed soy-based diets, but quantities present in humans may not be sufficient to induce estrogenic effects, although additive effects of equol with other phytoestrogens may occur.  相似文献

Different zinc (Zn) compounds have unique properties that may influence the amount of Zn absorbed particularly in the presence of phytic acid (PA), a common food component that binds Zn and decreases its bioavailability. In this study, 30-day-old male rats (n = 12/diet group) were fed diets supplemented with PA (0.8%) and low levels (8 mg Zn/kg diet) of inorganic (Zn oxide, Zn sulphate) or chelated (Zn gluconate, Zn acetate, Zn citrate, EDTA disodium Zn, Zn orotate) Zn compounds for 5 weeks. Two control groups were fed diets supplemented with low or normal (30 mg Zn/kg diet) Zn (as Zn oxide) without added PA. Control rats fed the low Zn oxide diet showed depressed Zn status. Addition of PA to this diet exacerbated the Zn deficiency in rats. Growth (body weight gain and femur length) and Zn concentrations in plasma and tissues were similar in rats fed Zn oxide, Zn sulphate, Zn gluconate, Zn acetate, Zn citrate or Zn orotate. Rats fed EDTA disodium Zn showed enhanced growth compared to rats fed Zn oxide or Zn gluconate and had higher Zn concentrations in plasma and femur compared to rats fed all other Zn compounds. Only the haematological profile of rats fed EDTA disodium Zn did not differ from control rats fed normal Zn. These data indicate that in rats fed a high PA diet, bioavailability of commonly used inorganic or chelated Zn compounds does not differ appreciably, but Zn supplied as an EDTA disodium salt has superior bioavailability.  相似文献

The iron bioavailability and acute oral toxicity in rats of a ferrous gluconate compound stabilized with glycine (SFG), designed for food fortification, was studied in this work by means of the prophylactic method and the Wilcoxon method, respectively. For the former studies, SFG was homogeneously added to a basal diet of low iron content, reaching a final iron concentration of 20.1 +/- 2.4 mg Fe/kg diet. A reference standard diet using ferrous sulfate as an iron-fortifying source (19.0 +/- 2.1 mg Fe/kg diet) and a control diet without iron additions (9.3 +/- 1.4 mg Fe/kg diet) were prepared in the laboratory in a similar way. These diets were administered to three different groups of weaning rats during 23 d as the only type of solid nourishment. The iron bioavailability of SFG was calculated as the relationship between the mass of iron incorporated into hemoglobin during the treatment and the total iron intake per animal. This parameter resulted in 36.6 +/- 6.2% for SFG, whereas a value of 35.4 +/- 8.0% was obtained for ferrous sulfate. The acute toxicological studies were performed in two groups of 70 female and 70 male Sprague-Dawley rats that were administered increasing doses of iron from SFG. The LD50 values of 1775 and 1831 mg SFG/kg body wt were obtained for female and male rats, respectively, evidencing that SFG can be considered as a safe compound from a toxicological point of view.  相似文献

Four groups of gilthead sea bream (Sparus aurata) were fed diets with additional metal contents: a basal diet (diet A) contained Zn at 60.9 ± 1.9 mg/kg diet, Cu at 3.9 ± 0.9 mg/kg diet, and Fe at 138.3 ± 6.8 mg/kg diet; the other diets were supplemented with copper (20 mg/kg, diet B), iron (100 mg/kg, diet C), or zinc (300 mg/kg, diet D). Two consecutive year-classes (0+ and 1+ age fish) from the same parent stock were examined. Several fish tissues were analyzed for metal contents in five different periods of each year in order to determine (1) the sensitivity of certain tissues as indicators of trace element metabolism and (2) possible seasonal variations. Growth data were similar for gilthead sea bream fed the basal diet and the metal-fortified diets. Mineral concentrations in tissues were found to be little affected by the dietary supplementation of trace elements, suggesting an efficient homeostatic control of these three metal concentrations. Tissues involved in metal metabolism (e.g., liver, kidney, gills) presented greater variations between minimum and maximum values with respect to other tissues (e.g., brain, muscle, eye). Seasonal variations were observed during the 2 yr of this study and were especially evident for zinc and copper concentrations in the liver. The overall pattern of metal variations showed a decreasing trend during the 2 yr. Results from this study indicate that (1) trace element concentrations in fish tissues vary with age and life cycle and (2) trace element requirements may vary in function of age and life cycle.  相似文献

Sensitivity of the assay for Cu,Zn superoxide dismutase 3 (SOD3), the predominant form of SOD in serum, can be increased, and interferences caused by low-molecular-weight substances in the serum can be reduced by conducting the assay at pH 10 with xanthine/xanthine oxidase and acetylated cytochrome c (cyt c) as superoxide generator and detector, respectively. Serum SOD3 activity was assayed under these conditions in an experiment where weanling, male rats were fed diets for 6 weeks containing 3, 5 and 15 mg Zn/kg with dietary Cu set at 0.3, 1.5 and 5 mg Cu/kg at each level of dietary Zn. Serum SOD3 responded to changes in dietary Cu but not to changes in dietary Zn. A second experiment compared serum SOD3 activity to traditional indices of Cu status in weanling, male and female rats after they were fed diets containing, nominally, 0, 1, 1.5, 2, 2.5, 3 and 6 mg Cu/kg for 6 weeks. Serum SOD3 activity was significantly lower (P < .05) in male rats fed diets containing 0 and 1 mg Cu/kg and female rats fed diet containing 0 mg Cu/kg compared with rats fed diet containing 6 mg Cu/kg. These changes were similar to changes in liver Cu concentrations, liver cyt c oxidase (CCO) activity and plasma ceruloplasmin in males and females. Serum SOD3 activity was also strongly, positively correlated with liver Cu concentrations over the entire range of dietary Cu concentrations (R(2) = .942 in males, R(2) = .884 in females, P < .0001). Plots of serum SOD3 activity, liver Cu concentration, liver CCO activity and ceruloplasmin as functions of kidney Cu concentration all had two linear segments that intersected at similar kidney Cu concentrations (18-22 microg/g dry kidney in males, 15-17 microg/g dry kidney in females). These findings indicate that serum SOD3 activity is a sensitive index of Cu status.  相似文献

为了更全面理解日本沼虾(Macrobrachium nipponense)的铜/锌营养生理作用,研究利用RACE技术从日本沼虾肝胰腺中克隆获得一金属硫蛋白基因cDNA全长(mn-MT),并对该基因分子特征、组织表达谱和饲料铜/锌水平对其表达的影响进行分析。结果显示:(1)mn-MT cDNA全长665 bp,含编码59个氨基酸的180 bp的开放阅读框,预测该多肽的理论分子量为6.085 kD,等电点为7.73。该蛋白中半胱氨酸含量最高(30.5%),其次是赖氨酸(16.95%)和丝氨酸(10.17%)。相似性分析显示mn-MT氨基酸序列与美洲海螯虾、斑节对虾和中华绒螯蟹MT的相似性分别达到78%、75%和75%。(2)qRT-PCR分析显示, mn-MT mRNA在肝胰腺、血细胞、鳃、胃、卵巢、肠和肌肉中都有表达,其中肝胰腺中表达量最高。(3)用4组铜添加量分别为0、20、40及160 mg/kg的饲料和3组锌添加水平分别为0、35和210 mg/kg的饲料饲喂初重为(0.1010.002) g日本沼虾56d后,分析各组虾肝胰腺的mn-MT mRNA表达。mn-MT mRNA表达随饲料铜水平的提高而升高,到40 mg/kg组达到最高(P0.05),而后开始下降;饲料中高锌(210 mg/kg)显著提高mn-MT表达(P0.05), 0和35 mg/kg组间差异不显著(P0.05)。结果表明饲料中铜/锌均可影响mn-MT表达,且呈现不同的剂量依赖效应。    相似文献

The removal of Cu, Ni and Zn from electroplating effluents by adsorption in cattle manure vermicompost has been discussed. A glass column 38 cm long and 7 cm i.d. was loaded with cattle manure vermicompost and effluents were passed through it. The metal concentrations were measured in the elutant. The experiments on adding effluent aliquots into the columns were continued until the metal concentrations in the elutant reached the maximum values established for effluent discharges in water courses by the Brazilian quality criteria, i.e., Cu=1.0 mg L(-1), Ni=2.0 mg L(-1), and Zn=5.0 mg L(-1). The amount of Cu retention by the vermicompost was determined at the natural effluent pH (2.0). The Zn and Ni retentions were evaluated at the natural effluent pH (6.9 and 7.4, respectively) as well pH 2.0. Vermicompost residues obtained from this process were used for lettuce cultivation. The vermicompost was found to be efficient in removing metals from the electroplating wastes, as well as in the increase of its pH values. Metal retention values were close to 100%. The Cu concentrations in lettuce leaves from the treatment with vermicompost enriched with this metal were below the range of critical toxicity level to plants, i.e., from 20 to 100 mg L(-1). However, the estimated Cu concentrations in the roots from the treatment with vermicompost enriched with Cu were much larger than that of the treatment with the natural vermicompost, reaching 246.3 mg L(-1). The Ni and Zn concentrations in lettuce leaves from the treatments, with vermicomposts enriched with the respective metals, were above the range of critical toxicity levels to plants, i.e., from 10 to 50 mg kg(-1) and from 15 to 30 mg kg(-1), respectively. However, no symptom of toxicity was found visually. Larger accumulations of Cu, Ni and Zn were found in the lettuce leaves than in the roots after the treatments with the uncontaminated vermicompost. A greater absorption of Cu and Ni by roots was found in treatments with vermicompost enriched with these elements, whereas Zn was found preferentially in the leaves. The statistical analysis was done by analyses of variance and regression.  相似文献

Studies of effects of diazepam on liver parenchyma are very scantly. In this study, adult albino rats were treated with diazepam in two different doses (0.25 mg and 0.30 mg/kg body wt) daily for 30 and 60 d. Through light microscopy and electron microscopy, prenecrotic and necrotic changes were noted in the high-dose group. Trace metal analysis indicated that zinc (Zn) was reduced by 30 and 60 day under both the doses, whereas iron (Fe) and copper (Cu) were reduced significantly in these groups only after 60 d of treatment. This reduction in metal contents may have some correlations with necrotic changes in liver parenchyma.  相似文献

A growth experiment with 108 lambs (breed: German Merino Landsheep) was carried out to examine the effect of gender, body weight (BW) and feeding intensity on the deposition of Fe, Zn, Cu and Mn in the empty body (whole animal minus contents of the gastrointestinal tract and bladder). The lambs (50% female and 50% male animals) were fed at three feeding levels ('low', 'medium' and 'high' by varying daily amounts of concentrate and hay) and slaughtered at different final BWs (30, 45 or 55 kg). Six male and six female animals were killed at a BW of 18 kg representing the animals' BW at the beginning of the comparative slaughter experiment. There were significant main effects for the treatments growth rate and final weight on the daily rate of accretion of the trace elements examined. Feeding intensity had a marked influence on the accretion rate for Fe (P < 0.001), Zn (P < 0.001), Cu (P < 0.001) and Mn (P = 0.003). With increasing feeding intensity (low, medium, high) the daily deposition of these trace elements increased (4.4, 5.2, 6.6 mg/day for Fe; 4.9, 5.5, 6.9 mg/day for Zn; 0.20, 0.36, 0.44 mg/day for Cu; 0.14, 0.16, 0.21 mg/day for Mn). Heavier final BW led to increased daily retention of Zn (P < 0.001) and Mn (P = 0.002). Gender had a marked influence only on the accretion rate for Zn (P < 0.001). Ram lambs had a higher daily deposition of this element than female lambs. Related to 1000 g empty body gain, the following concentrations were found for the trace elements examined: Fe 26.1 mg, Zn 30.0 mg, Cu 1.41 mg and Mn 1.04 mg. A feeding influence was given for Zn (P < 0.001) and Cu (P = 0.039). Feeding level low had higher Zn and lower Cu concentrations. Male animals showed less Fe (P < 0.001) and Zn (P = 0.034) per kg empty body gain than females.  相似文献

The effect of feeding Cu- and Zn-methionine to ewes was studied in a 240d feeding trial. The plasma and tissue Cu and Zn concentrations and Cu/Zn-superoxide dismutase (Cu/Zn-SOD) activity were employed to assess the relative bioavailability from Cu- and Zn-methionine. The macro and micronutrient intake, utilization, plasma mineral status, tissue accumulation of Cu and Zn as well as wool concentration of Cu and Zn were studied in ewes (n=12) fed a corn-soybean meal based basal diet with 50% more Cu and Zn supplementation over the basal diet either from Cu- and Zn-sulfate (Cu-Sulf+Zn-Sulf group) or Cu- and Zn-methionine (Cu-Meth+Zn-Meth group). The average daily feed intake and body weight gain of ewes did not differ due to dietary supplementation of Cu- and Zn-methionine. However, dry matter intake was comparatively lower and thus resulted in better feed: gain in Cu- and Zn-methionine group as compared to ewes fed Cu- and Zn-sulfate. Supplementation of Cu and Zn over the basal diet either from methionine-chelated or sulfate sources resulted in increased plasma Cu and Zn as well as Cu/Zn-SOD activity on d-30, which indicated a positive correlation between plasma Cu and Zn and Cu/Zn-SOD activity. The gut absorption, liver concentrations of Cu and Zn, and liver Cu/Zn-SOD activity were significantly (P<0.01) higher in ewes supplemented with Cu- and Zn-methionine compared to Cu- and Zn-sulfate. Periodical analysis of wool samples indicated no significant difference in Cu and Zn content between Cu-and Zn-methionine and Cu- and Zn-sulfate groups. Feeding of Cu and Zn from methionine-chelated source resulted in reduced (P<0.01) excretion of Cu and Zn in feces indicating their better utilization, and this will have positive implication on environment. The gut absorption values, plasma and liver tissue concentrations of Cu and Zn supported the hypothesis that Cu- and Zn-methionine supplements have better bioavailability compared to Cu- and Zn-sulfate and Cu- and Zn-dependent enzyme (Cu/Zn-SOD) could be used to determine the bioavailability of Cu and Zn.  相似文献

The hypothesis that copper (Cu) alters drug metabolizing enzymes and functions as an antioxidant nutrient in doxorubicin cardiotoxicity was tested. Male Sprague-Dawley rats were fed Cu adequate (+Cu; 5 mg Cu/kg of diet), marginally Cu deficient (MCu; 1.2 mg Cu/kg of diet), or severely Cu deficient (Cu; 0.5 mg Cu/kg of diet) diets for 6 wk. Doxorubicin (1, 2, or 4 mg/kg body wt) or saline were administered intraperitoneally 1 time/wk for 4 wk. Compared to control hearts, Cu, Zn superoxide dismutase activity was decreased by 9% in MCu rats and by 21–40% inCu rats. Glutathione peroxidase activity was elevated 5–15% inCu rats. Doxorubicin administration increased heart Cu, Zn superoxide dismutase activity in+Cu andCu rats 18 h after the last of 4 injections, but not 18 h after 1 injection. There was no synergism between doxorubicin and Cu deficiency on lipid peroxidation, plasma creatine phosphokinase, cardiac hypertrophy, electrocardiographic abnormalities, or morphological changes. Heart glutathione S-transferase activity was decreased by Cu deficiency, and like Cu, Zn superoxide dismutase activity, returned to normal inCu rats given doxorubicin. Thus, the Cu deficient rat heart may be able to compensate for doxorubicin-induced oxidant stress by increasing the activity of Cu,Zn superoxide dismutase and glutathione S-transferase.  相似文献

The present study focused on the effect of Zn containing diets on the activity of superoxide dismutase (Cu/ZnSOD), systolic blood pressure (SBP), lipid peroxides (ROOH) and lipids (LDL, HDL, triglycerides and cholesterol) in male spontaneously hypertensive rats (SHR). Three experimental groups of animals were studied: a control (G1-40 mg), and two with zinc-supplemented diets (G2-100 and G3-160 mg Zn/kg lab chow). The diets were introduced at the beginning of the development of hypertension (2 months after birth) and the animals were fed for 8 weeks. The activity of CuZnSOD in erythrocytes was determined by spectrophotometry with the use of RANSOD kit (RANDOX Laboratories Ltd., UK). Atomic-absorption spectrometry was used to determine Zn and Cu concentrations in the rat's sera. A significantly increased Cu/ZnSOD activity was found in G3 compared with rats fed with control diet G1 (p = 0.020). SBP was significantly decreased in G3 in relation to G1 (p = 0.0048). The lipid hydroperoxide concentration was significantly decreased in G3 compared with G1 (p = 0.016) and G2 (p = 0.005). Zinc supplement affected lipids profile by decreasing LDL and increasing HDL. The present data suggest that Zn concentration in the diet plays an important role in the regulation of SBP and can be a critical nutrient for maintenance of anti-oxidative events in SHR.  相似文献
