Given the immense risk posed by widespread environmental pollution by inorganic and organic chemicals, novel methods of decontamination and clean-up are required. Owing to the relatively high cost and the non-specificity of conventional techniques, bioremediation is a promising alternative technology for pollutant clean-up. Advances in bioremediation harness molecular, genetic, microbiology, and protein engineering tools and rely on identification of novel metal-sequestering peptides, rational and irrational pathway engineering, and enzyme design. Recent advances have been made for enhanced inorganic chemical remediation and organic chemical degradation using various pathway-engineering approaches and these are discussed in this review. 相似文献

In the postgenome era, the analysis of entire subproteomes in correlation with their function has emerged due to high throughput technologies. Early approaches have been initiated to identify novel components of the circadian system. For example, in the marine dinoflagellate Lingulodinium polyedra, a chronobiological proteome assay was performed, which resulted in the identification of already known circadian expressed proteins as well as novel temporal controlled proteins involved in metabolic pathways. In the green alga Chlamydomonas reinhardtii, two circadian expressed proteins (a protein disulfide isomerase and a tetratricopeptide repeat protein) were identified by functional proteomics. Also, the first hints of temporal control within chloroplast proteins of Arabidopsis thaliana were identified by proteome analysis. 相似文献

This research aims towards developing an alternative antischistosomal drug using miltefosine, which is primarily used in the treatment of leishmaniasis. The treatment and control of schistosomiasis, a notable neglected tropical disease (NTD), rely on a single drug, praziquantel (PZQ). The dependency on PZQ exclusively is quite alarming, given the spread of the disease (over 200 million people infected and close to 800 million people at risk in three continents) and the threat of drug resistance. This study shows that the oral administration of miltefosine in a daily dose of 20 mg/kg for five successive days to mice infected with either invasive, juvenile or adult stages of S chistosoma mansoni resulted in significant reduction of worm burden, hepatic granulomata size and amelioration of hepatic pathology. Scanning Electron Microscopy revealed that miltefosine induced severe tegumental damage in adult schistosomes. In conclusion, we believe this is the first study highlighting miltefosine as a promising novel agent for schistosomiasis mansoni. 相似文献

BackgroundCirculating nucleic acids (CNAs) offer unique opportunities for early diagnosis of clinical conditions. Here we show that microRNAs, a family of small non-coding regulatory RNAs involved in human development and pathology, are present in bodily fluids and represent new effective biomarkers. Methods and ResultsAfter developing protocols for extracting and quantifying microRNAs in serum and other body fluids, the serum microRNA profiles of several healthy individuals were determined and found to be similar, validating the robustness of our methods. To address the possibility that the abundance of specific microRNAs might change during physiological or pathological conditions, serum microRNA levels in pregnant and non pregnant women were compared. In sera from pregnant women, microRNAs associated with human placenta were significantly elevated and their levels correlated with pregnancy stage. Conclusions and SignificanceConsidering the central role of microRNAs in development and disease, our results highlight the medically relevant potential of determining microRNA levels in serum and other body fluids. Thus, microRNAs are a new class of CNAs that promise to serve as useful clinical biomarkers. 相似文献

The proposed introduction of genetically modified herbicide tolerant (GMHT) crops, with claims of improved weed control, has prompted fears about possible environmental impacts of their widespread adoption, particularly on arable weeds, insects and associated farmland birds. In response to this, we have developed a novel weed-management system for GMHT sugar beet, based on band spraying, which exploits the flexibility offered by the broad-spectrum partner herbicides. Here, we show the results from two series of field experiments which, taken together, demonstrate that, by using this system, crops can be managed for enhanced weed and insect biomass without compromising yield, thus potentially offering food and shelter to farmland birds and other wildlife. These results could be applicable widely to other row crops, and indicate that creative use of GMHT technology could be a powerful tool for developing more sustainable farming systems in the future. 相似文献

Alzheimer's disease (AD) is a neuronal disorder with insidious onset and slow progression, leading to growing global concern with huge implications for individuals and society. The occurrence of AD has been increased and has become an important health issue throughout the world. In recent years, the care of more than 35 million patients with AD costs over $ 600 billion per year, it is approximately 1 percent of the global Gross Domestic Product. Currently, the therapeutic approach is not effective for neurological deficits especially after the development of these major neurological disorders. The discovery of the technique called cell-based therapy has shown promising results and made important conclusions beyond AD using the stem cells approach. LCA is a system-wide assessment, and the LCIA phase is confronted with the difficulties of local and regional effects in a
number of impact categories. We integrate three different environmental techniques to demonstrate how these effects can be
addressed in an environmental assessment. The techniques are life cycle inventory, environmental fate models, and an ecological
impact assessment using fuzzy expert systems.
Results of the LCI are mass and energy flows. In the environmental fate modelling step these mass flows are transformed into
concentration and immission values by dispersion-reaction models. A generalised fuzzy expert system for the environmental
mechanisms compares calculated exposure with site specific buffering capacities and formulates a generalised dose-response
relationship. This generalised fuzzy expert system is used as a template for the assessment of local and regional environmental
impacts. An application of this integrated approach is shown for a practical problem: production of magnesium car components.
The environmental fate of nitrogen oxides which are released due to the major combustion source within that production system
is simulated. Fuzzy expert models for crop damage, soil acidification and eutrophication determine the possible environmental
impact of the immited nitrogen oxides.
The important methodological extension of this integrated approach is a regionalised impact assessment depending on the spatial

We developed a computationally efficient algorithm AMBIENCE, for identifying the informative variables involved in gene-gene (GGI) and gene-environment interactions (GEI) that are associated with disease phenotypes. The AMBIENCE algorithm uses a novel information theoretic metric called phenotype-associated information (PAI) to search for combinations of genetic variants and environmental variables associated with the disease phenotype. The PAI-based AMBIENCE algorithm effectively and efficiently detected GEI in simulated data sets of varying size and complexity, including the 10K simulated rheumatoid arthritis data set from Genetic Analysis Workshop 15. The method was also successfully used to detect GGI in a Crohn's disease data set. The performance of the AMBIENCE algorithm was compared to the multifactor dimensionality reduction (MDR), generalized MDR (GMDR), and pedigree disequilibrium test (PDT) methods. Furthermore, we assessed the computational speed of AMBIENCE for detecting GGI and GEI for data sets varying in size from 100 to 10(5) variables. Our results demonstrate that the AMBIENCE information theoretic algorithm is useful for analyzing a diverse range of epidemiologic data sets containing evidence for GGI and GEI. 相似文献

Dendritic cells (DCs) are the most potent antigen presenting cells in the human organism. Ever since the discovery of their function in the self/nonself discrimination, DCs have been seen as potential candidates for therapy in malignant tumors. With the exception of differentiated thyroid cancer, endocrine malignancies are rare tumors and apart from surgical intervention there is no truly established method for their treatment. Therefore, the prognosis of many endocrine carcinomas is still poor and new therapeutic options are needed. In the last decade, different immunotherapeutic approaches have shown promising results in other solid tumors. In recent studies, immunotherapy using DCs has been proven to be safe and effective to induce antitumor immune responses leading to tumor regression and even rejection of cancer in some cases. This review will summarize the latest progress in DCs based immunotherapy with special focus on the limited experience in endocrine malignancies. With regard to these tumors, it is of special interest which antigens could serve as potential target antigens for future trials. We also discuss what steps have to be taken to develop a better immunotherapy in endocrine tumors. 相似文献

Never in Mitosis (NIMA) Related Kinase 2 (NEK2) plays a key role in regulating mitotic processes, including centrosome duplication and separation, microtubule stabilization, kinetochore attachment and spindle assembly checkpoint. NEK2 is aberrantly overexpressed in a wide variety of human cancers and has been implicated in various aspects of malignant transformation, including tumorigenesis, drug resistance and tumor progression. The close relationship between NEK2 and cancer has made it an attractive target for anticancer therapeutic development; however, the mechanisms of how NEK2 coordinates altered signaling to malignant transformation remains unclear. In this paper, we discuss the functional roles of NEK2 in cancer development; highlight some of the significant NEK2 signaling in cancer, and summarize recent advances in the development of NEK2 inhibitors. 相似文献

Cisplatin is a widely used antineoplastic agent; however, its major limitation is the development of dose-dependent nephrotoxicity whose precise mechanisms are poorly understood. Here we show not only that mitochondrial dysfunction is a feature of cisplatin nephrotoxicity, but also that targeted delivery of superoxide dismutase mimetics to mitochondria largely prevents the renal effects of cisplatin. Cisplatin induced renal oxidative stress, deterioration of mitochondrial structure and function, an intense inflammatory response, histopathological injury, and renal dysfunction. A single systemic dose of mitochondrially targeted antioxidants, MitoQ or Mito-CP, dose-dependently prevented cisplatin-induced renal dysfunction. Mito-CP also prevented mitochondrial injury and dysfunction, renal inflammation, and tubular injury and apoptosis. Despite being broadly renoprotective against cisplatin, Mito-CP did not diminish cisplatin's antineoplastic effect in a human bladder cancer cell line. Our results highlight the central role of mitochondrially generated oxidants in the pathogenesis of cisplatin nephrotoxicity. Because similar compounds seem to be safe in humans, mitochondrially targeted antioxidants may represent a novel therapeutic approach against cisplatin nephrotoxicity. 相似文献

PurposeThe population living in urban areas of the world continues to grow rapidly. It is, thus, a great priority for the planning practice to embed sustainability concept in their urban development endeavors. Currently, development and expansion of urban systems stress the need to control consumption of resources, especially non-renewable ones. There is also a need to reduce related environmental impacts, while stimulating a sustainable pathway for the population and urban growth. MethodsStrategic environmental assessment (SEA) is useful for policy design to build an integrated method for supporting the development of a sustainable society. It undertakes territorial assessments and describes urban flows and impacts related to them by using a variety of tools, including material flow accounting (MFA). This study employs MFA, as it fits well within the scope of SEA and supports the growing environmental attention in the urban metabolism approach. Although helpful, MFA has not been systematically applied in the urban development context; for this reason, this paper proposes the integration of SEA and MFA. Results and discussionIntegration of SEA and MFA generates a new framework for sustainable development planning. The framework is structured in phases oriented to the continual improvement based on the Deming cycle (i.e., plan, do, check, act), a key management approach mainly used in businesses for improving the effectiveness of an organization. It can also be implemented at the urban system level. In order to maintain normative compliance, each process (urban planning, strategic environmental assessment with urban metabolism approach, participatory processes) is standardized in line with a common and mandatory approach. While the processes are integrated among them, highlighting the reciprocal contact points, the results are combined in a holistic perspective. The framework, hence, transforms the voluntary MFA tool into a mandatory process. ConclusionsThe proposed SEA-MFA framework has the potential to unify and standardize the processes of categorizing and quantifying data in order to improve the understanding of urban metabolic principles and scale effects. It also supports management and policy development and meets the requirements of different stakeholders. The framework, thus, generated a novel approach for sustainable urban development planning by providing solutions for specific policy problems and ensuring urban ecological balance and sustainable urban futures. 相似文献

Phytoremediation is the use of plants for the removal of pollutants from contaminated soil or water. Phytoremediation is an environmentally friendly and cost-effective alternative to current remediation technologies. This review article outlines general aspects of phytoremediation, along with discussions about its advantages and limitations. It further reviews various phytoremediation processes in detail: phytoextraction, rhizofiltration, phytostabilization, phytodegradation, and phytovolatilization. Unlike previous review articles available in various journals, this paper presents a more comprehensive view of this issue, and deals with a much wider range of its applications to environmental pollution control. These include the treatment of wastewaters, removal of heavy metals and metalloids ( e.g. lead and arsenic), phytoremediation of organic pollutants, such as 2,4,6-trinitrotoluene (TNT) and polychlorinated biphenyls (PCBs), and cleanup of soil and water contaminated with radionuclides, such as cesium ( 137Cs) and strontium ( 90Sr). This paper also describes recent developments of transgenic plants for improving phytoremediation. Along the way, the present status of phytoremediation research in Korea is briefly introduced. Environmental problems, such as global warming, the limited supply of sustainable energy, the depletion of natural resources, hazardous emissions released into the atmosphere and waste, are increasing global concerns. Therefore, individuals, communities, and businesses need to address environmental protection and sustainability. Environmental impact assessments are needed to identify, mitigate, and control aspects that affect the environment or a company's products, services, or activities. In this study, a general environmental aspect and impact assessment approach, which can be applied to any company that is involved in the production or service sector, is created. An environmental impact pattern that consists of 10 main and 32 sub-categories was formed based on the ISO 14001, environmental studies and field applications. The developed approach was applied to the dyeing units of a manufacturing firm. Sixteen environmental aspects were identified and assessed using the environmental impact template via the environmental failure mode and effect analysis (E-FMEA) method. The developed-approach can be applied to each sector, which will enable us to perform a detailed analysis of the environmental aspects in the environmental impact category. This approach provides a checklist for the environmental impact studies of businesses and has been pioneered as an effective method for company resources to improve their environmental performance. 相似文献

The in vitro biological activity of N-benzylsalicylthioamides was evaluated against eight fungal strains by the broth microdilution method and the results were compared with those obtained with fluconazole. The compounds exhibited an in vitro antifungal activity against the fluconazole-susceptible as well as the fluconazole-resistant fungal strains.
