RNA/DNA ratio is a useful and reliable indicator of the nutritional status of fish larvae and juveniles. In order to assess the nutritional status of field-caught larval Pacific bluefin tuna Thunnus orientalis (Temminck et Schlegel), starvation experiments of hatchery-reared larvae were conducted and changes in the RNA/DNA ratio of fed and starved larvae were analyzed. Starvation experiments were conducted every 3 days after first feeding. The survival rate of Pacific bluefin tuna larvae ranged 10-50% after 1 day of starved conditions and growth retardation was observed immediately. These results suggest that Pacific bluefin tuna larvae have a very low tolerance to starvation. The RNA/DNA ratios of fed larvae were approximately 2.0-4.0. On the other hand, the value of starved larvae significantly decreased to 1.0-3.0. The nutritional status of 3 cohorts of field-caught tuna larvae collected in the northwestern Pacific Ocean was examined based on the value of the RNA/DNA ratio of the 1 day starved larvae. 4.35-25.77% of the cohorts were regarded as the “starving condition”, which was negatively correlated to the ambient prey densities. These findings suggest that the nutritional condition of larval Pacific bluefin tuna was influenced by the ambient prey density, and starvation itself and starvation-induced predation could greatly contribute to mortality in the larval period of Pacific bluefin tuna.  相似文献

Hatchery reared larval striped bass, Morone saxatilis , 8-days-post-hatching were subjected to various feeding/starvation regimes over a period of 14 days.
Batches of larvae from each treatment were sampled over the 14-day period and subdivided for determination of notochord length and RNA:DNA ratio. The best growth was found in fully fed F1000 larvae (exposed to 1000 Artermia nauplii l−1), which reached 8.2 mm after 11 days and 9.6 mm after 14 days. Starved animals after 11 days had notochord lengths of 4.9 mm. Growth curves from feeding-delayed larvae indicated that animals fed after up to 5 days starvation were capable of complete recovery. F100 larvae (exposed to 100 Anemia nauplii 1−l) had a slower growth rate than F1000 larvae, reaching a notochord length of 7.3 mm after 14 days. RNA:DNA ratios over time closely followed notochord growth curves, with clear differences between starved, F100 and F1000 larvae being established after only 2 days. Equilibrium RNA:DNA ratios of 3.0 and 2.25 were established in F1000 and F100 larvae, respectively, 6.8 days after the beginning of the experiment. The average lag time between a change from the starved to the fed condition and a change in RNA:DNA ratio as determined by the divergence of the nucleic acid curve from the starved condition was 0.66 days.
In treatments where starvation followed various periods of feeding, larvae regressed in notochord length such that the final length at 14 days reflected the degree of feeding. RNA:DNA ratios in these animals again closely followed growth curves with a lag time of 0.81 days.
It was concluded that RNA:DNA ratios provided very accurate indices of growth in striped bass larvae which were highly sensitive to feeding status.  相似文献

研究通过对岩原鲤仔鱼在饥饿和再投喂条件下其生存、生长率、RNA/DNA和RNA/蛋白质比率的测定,评估了仔鱼对饥饿的耐受能力和恢复能力。在(19.5±0.5)℃水温下,将岀膜后第16天的岩原鲤仔鱼随机分成6个组:1个持续投饲对照组,实验组分别禁食1、2、3、4、5d后再投喂,实验共进行10d。每天分别从各组取9尾鱼测定体重、体长、RNA、DNA、蛋白质含量。实验结果显示,饥饿处理组仔鱼存活率和以上各项生长指标均随饥饿时间的增加而下降,在恢复投喂后均表现不同程度的补偿生长,其中饥饿1、2、3d的仔鱼在恢复投喂后显示出完全补偿生长,几乎弥补了饥饿所产生的影响,平均终体重与对照组比较无显著差异。饥饿4、5d的仔鱼显示部分补偿生长,恢复投喂只少量减轻了饥饿的影响,平均终体重与对照组相比存在显著差异。饥饿1、2、3d的仔鱼和4、5d的仔鱼在恢复投喂后分别需要1—2d和4d时间才能达到与对照组无显著差异水平。仔鱼生长率变动范围从0.59%到8.00%WW/day,仔鱼RNA/DNA比率、RNA/蛋白质比率与生长率的回归方程为:GR=3.63RNA/DNA 1.74(R2=0.80)和GR=120.14RNA/Protein 2.33(R2=0.31),两种比率均与生长率呈显著线性相关,RNA/DNA比率对生长变化的拟合度更好。结果表明,仔鱼阶段食物缺乏很可能是影响岩原鲤仔鱼存活、生长的主要因素。RNA/DNA更适合作为评定岩原鲤仔鱼营养条件和生长的指标。  相似文献

A laboratory experiment was performed to study the responsiveness of selected condition measures to starvation in herring, Clupea harengus, larvae in relation to temperature and ontogeny. The larvae at two intervals of development, i.e. stage 1 larvae with initial exogenous feeding and stage 2 larvae with established feeding prior to notochord flexion, were reared at three temperatures (5, 8 and 11°C) and subjected to sub-lethal durations of starvation. Temporal changes in standard length, dry weight, DNA concentration (% of dry weight), RNA concentration (% of dry weight), Fulton's condition factor (CF) and RNA/DNA were assessed and compared with fed controls. Starvation led to decreases in dry weight, CF, RNA concentration and RNA/DNA, while it led to an increase in DNA concentration. Higher responsiveness to starvation was observed at higher temperatures, and the magnitude of the changes was higher in stage 2 larvae. The shortest latency in starvation response was found with respect to RNA/DNA which was length independent in the size range studied. RNA/DNA was also significantly related to average DNA growth rate, and the model for DNA growth rate was, SGRDNA=4.49 RNA/DNA + 7.14 T – 0.42 T2 – 37.5 ; n=32, r2=0.85, p < 0.001). While the model seemed to adequately represent the average temperature dependent DNA growth, a relatively low classification success made it unsuitable for depicting individually starving larvae. Critical levels in DNA concentration can be used (2.2% for stage 1 larvae, 2.9% for stage 2 larvae) to differentiate starving larvae (after 3 – 5 days) from feeding larvae. RNA/DNA was the most sensitive and suitable condition index studied in detecting early starvation of herring larvae.  相似文献

In order to clarify the nutritional conditions of larval milkfish in the surf zone, the following parameters were examined: 1) DNA and RNA content and RNA/DNA ratio of fed and unfed larvae collected from the surf zone and reared in the laboratory; 2) survival rate of the unfed larvae; and 3) total length, otolith increment counts and RNA/DNA ratio of wild larvae collected daily from the surf zone. The DNA and RNA content of the unfed larvae decreased, but increased in fed larvae. The RNA/DNA ratio decreased in unfed larvae, whereas in the fed larvae it decreased for the first three days after capture and increased thereafter. These results indicated that the values of DNA and RNA content and RNA/DNA ratio could be used as an indicator of nutritional condition of milkfish larvae after 6 days of starvation. Although total length of the wild-larvae did not show serial changes, their otolith increment counts showed continuous increases, indicating that the larvae sojourned in the surf zone for several days. In the same period, RNA/DNA ratios of the wild larvae decreased continuously, the ratios of larvae with fewer otolith increment counts being relatively higher than those of larvae with greater increment counts. Based on these results, the milkfish larvae remaining in the surf zone were concluded as being under insufficient nutritional conditions.  相似文献

The sea water temperature off the Norwegian coast was lower in 1994 than in 1992 and 1993. RNA/DNA ratios of Norwegian spring spawning herring increased with increasing larval dry weight all years, except for larvae sampled south of 62°N in 1994. The RNA/DNA ratios indicated that each year, only a small portion ( 0.7%) of the larvae were starving. RNA/DNA ratios and temperature were negatively correlated in 1992, but in other years no significant correlations were found. Residuals of In RNA v . In DNA and In W v. L s were poorly correlated in all years, but residuals of In RNA v. In DNA and In DNA v. In W were negatively correlated in all years. Principal component analysis showed that the RNA/DNA ratio and DNA (% of weight) were correlated with different axes. Abundance data for herring at the early larval and 0-group stages in 1992–1994 indicated higher mortality in 1994 compared with the other years. The data do not indicate that average larval condition was poorer in 1994 than in other years. However, the variability in larval condition was higher in 1994 than in other years, and the condition of later larval stages was relatively lower in 1994 than in other years.  相似文献

Growth in length and mass, improvements in condition, as well as final condition of c. 700 g Atlantic cod Gadus morhua were significantly less at 45% and 56% O2 saturation than at 65%, 75%, 84% and 93% O2 saturation. Hypoxia decreased food consumption. In turn, food consumption explained 97% of the variation in growth. Conversion efficiency varied slightly, but significantly, with level of dissolved O2, except that the group reared at 93% O2 had a lower than expected conversion efficiency. Slow growth in low O2 was not due to increased activity, because activity decreased in hypoxia. In the Gulf of St Lawrence, waters deeper than 200 m usually are <65% saturated in O2, and thus should impact negatively on cod growth.  相似文献

Wet mass and DNA, RNA and protein content increased significantly with standard length ( L S) of sardine Sardina pilchardus larvae, collected in January 1995, in the Bay of Málaga, North Alboran Sea. L S, wet mass and DNA, RNA and protein content were closely related allometrically to otolith radius ( R ). Larval daily length increments decreased but DNA, protein and wet mass daily increments increased with larval age. Daily length increments showed a negative and poor relationship with long-term otolith growth. In contrast, DNA, protein and wet mass daily increments were positively correlated. Differences between observed and back-calculated otolith radius-at-age indicated that larvae with slow otolith growth were under represented in older age groups, suggesting the existence of growth-selective mortality. Recent otolith growth, estimated from the mean widths of the last six increments, increased with age and R . Individual RNA: DNA and protein: DNA ratios were correlated significantly, although weakly, with L S and larval growth.  相似文献

The growth, nucleic acid and protein contents of walleye pollock Theragra chalcogramma larvae reared at prey densities of 10, 30, 50, and 500 prey 1-1 were measured for the first 9 days after the feeding initiation at 6° C. Incremental growth rates of larvae (mm day-1) were low and variable for the first 7 days after feeding initiation. Growth rates and rates of RNA, DNA, and protein accumulation by larvae reared at 500 prey 1-1 were positive while those of larvae reared at the lower prey levels did not differ significantly from zero. The RNA/DNA ratio was variable and exhibited no significant trend among food treatments. Estimates of instantaneous protein growth rates ranged from - 6·7 to 13·2% day-1 at food densities of 10 and 500 prey 1-1, respectively, and were moderately correlated with larval RNA/DNA ratios ( r = 0·628). The results suggest that in situ protein growth rates of first-feeding pollock larvae may be influenced by prey fields within the range of ambient food densities reported for sub-Arctic ecosystems.  相似文献

Unfertilised cod eggs showed a mean oxygen uptake rate at 5°C of 0.089 μl O2, dry wt.−1 h−1; this gradually rose to 0.768 μl O2 mg dry wt.−1 h−1 in eggs about to hatch. From hatching to complete yolk absorption larvae respired at 1.6 μl O2, mg dry wt.−1 h−1. During starvation following yolk absorption, uptake fell significantly to 1.1 μl O2, mg dry −1 h−1. Much of this decrease in oxygen consumption was shown to be caused by reduction in activity. Loss of weight during the embryo and larval phases could not easily be reconciled with total oxygen consumption; it is suggested that cod embryos and larvae may not rely solely upon endogenous energy reserves during development.  相似文献

Standard length, dry mass and RNA : DNA ratio measurements of 3876 Atlantic cod Gadus morhua larvae and juveniles from 26 families of recruit (fish during their first year of spawning) and repeat spawners (fish which were in their subsequent spawning season) reared in two mesocosms (2500 and 4400 m3) under semi-natural conditions were analysed over a period of 10 weeks using microsatellites. Larvae from recruit spawners were significantly longer and heavier at hatch and throughout the 10 weeks. RNA : DNA ratios from recruit spawner offspring were only significantly higher at week 1. The smaller (2500 m3) mesocosm was characterized by low plankton density during the transition from endogenous to exogenous feeding followed by a higher density during the metamorphosis period (weeks 4 and 5), with the reverse pattern evident in the 4400 m3 mesocosm. Patterns of larval growth followed patterns of zooplankton density. Significant differences in RNA : DNA ratios between the mesocosms at all comparable sampling dates were found and within each mesocosm individual fish exhibited a wide range of growth and condition responses under the same environmental conditions. RNA : DNA ratios as a function of size differed in the amount of variability between mesocosms, indicating that the higher food density led to a higher proportion of well-conditioned larvae in the first 3 weeks. Food availability probably has a major role in determining offspring growth and condition, with limited effects due to maternal effects in cases where the broodstock females are approximately of similar size and condition.  相似文献

DNA from archived otoliths was used to explore the temporal stability of the genetic composition of two cod populations, the Moray Firth (North Sea) sampled in 1965 and 2002, and the Bornholm Basin (Baltic Sea) sampled in 1928 and 1997. We found no significant changes in the allele frequencies for the Moray Firth population, while subtle but significant genetic changes over time were detected for the Bornholm Basin population. Estimates of the effective population size ( N e ) generally exceeded 500 for both populations when employing a number of varieties of the temporal genetic method. However, confidence intervals were very wide and N e 's most likely range in the thousands. There was no apparent loss of genetic variability and no evidence of a genetic bottleneck for either of the populations. Calculations of the expected levels of genetic variability under different scenarios of N e showed that the number of alleles commonly reported at microsatellite loci in Atlantic cod is best explained by N e 's exceeding thousand. Recent fishery-induced bottlenecks can, however, not be ruled out as an explanation for the apparent discrepancy between high levels of variability and recently reported estimates of N e  << 1000. From life history traits and estimates of survival rates in the wild, we evaluate the compatibility of the species' biology and extremely low N e / N ratios. Our data suggest that very small N e 's are not likely to be of general concern for cod populations and, accordingly, most populations do not face any severe threat of losing evolutionary potential due to genetic drift.  相似文献

The effect of the timing of first feeding (3, 4, 5, 6, 7 and 8 days post‐hatch, dph) on laboratory‐reared California halibut Paralichthys californicus larvae was evaluated by means of morphologic, morphometric and histological criteria. Larvae began to feed exogenously at 3 dph (2·7 ± 0·01 mm standard length, L S) at 18° C. Eye pigmentation, rather than mouth opening was the most distinctive trait of California halibut larvae at first feeding. Larval growth was significantly affected by the time of first exogenous feeding. At notochord flexion (21 dph), the L S of larvae fed for the first time at 3 dph was significantly larger (5·1 ± 0·1 mm) than that of those fed at 4 and 5 dph (4·9 ± 0·1 mm), although the latter fish had a more uniform size distribution. The point of no return was reached at 7 dph. Survival of larvae initially fed at 3, 4 and 5 dph was similar (58·4–60%), while no larvae were able to survive when food was offered for the first time between 6 and 8 dph. Food deprivation resulted in a progressive deterioration of the larval digestive system and atrophy of skeletal muscle fibres. Significant changes in the anterior and posterior enterocyte height were detected after 2 days of food deprivation. Similarly, tail height: L S and trunk length: L S ratios were the most sensitive morphometric indices to detect the effect of fasting on larval condition. Present results show that a combination of morphometric and histological variables can be used to evaluate the nutritional condition of California halibut larvae.  相似文献

A laboratory calibration study was undertaken with juvenile Sepia officinalis (80-85 g initial wet weight) to investigate the effects of different food rations and different starving intervals on RNA/dry weight (DW) ratios and RNA/DNA ratios in cephalopod mantle muscle at two different temperatures. The digestive gland index was also used as an additional indicator of recent growth. High food rations and low temperature went along with high RNA/DW ratios and high RNA/DNA ratios. Starving resulted in a linear decline in growth performance and a concomitant decrease in RNA/DW and RNA/DNA ratio, with RNA/DNA ratios representing the growth data better. RNA/DNA ratios decreased faster at higher temperatures. A fluorimetric assay for nucleic acid analysis was optimized for cephalopod mantle tissues and yielded reproducible RNA/DNA ratios with a relative variance below 10%. Thus, it may be possible to use this estimator of recently encountered feeding regime for the evaluation of mortality rates of early teuthid paralarvae to eventually support stock management. Also, log relative digestive gland weight showed a strong relationship with starving time, but, surprisingly, not with temperature. Data from the two temperatures analyzed could be combined to form a common regression line of relative digestive gland index with starving time. This indicator for recent growth might be especially suitable for large specimens with a well-developed digestive gland.  相似文献

Understanding the nutritional condition and survival of fish larvae is of primary importance in mass larva culture because intensive mortality is concentrated during the larval period. In order to estimate growth and nutritional status based on biochemical indices of slime flounder, Microstomus achne, larvae reared under starved and fed conditions in the hatchery for 58 days and the changes of RNA, DNA, and protein contents were described with the progress of growth and developmental. DNA contents increased gradually throughout the experimental period until 12 and 58 days post‐hatching (DPH) in starved and fed groups respectively. Although they fluctuated and decreased around 12 and 46 DPH, the RNA contents of the fed group increased gradually after hatching; however, in the starved group, they decreased after yolk absorption and 7 DPH. Subsequently, the RNA/DNA ratios in the starved group remained constant until 6 DPH and then decreased rapidly. In the fed group, this decreased slightly from hatching to 14 DPH, then increased gradually until the end of the experiment, except at the lower level of around 46 DPH. Namely, temperature shocks (around 14 DPH) and the dramatic changes in body shape (around 46 DPH) were accompanied by the decrease of the RNA/DNA ratios. Total protein/DNA in both groups decreased rapidly during yolk absorption 0–7 DPH, then decreased continuously until death in the starved group; in the fed group total protein/DNA increased again with feeding. It is suggested that the changes in these biochemical values reflect yolk absorption, feeding, morphogenetic changes, growth, and environmental conditions.  相似文献

Growth of 2659 Atlantic cod Gadus morhua aged 4 to 9 years examined in Placentia Bay, Newfoundland, peaked in most cases in June and was at a minimum in October or November. Water temperature, partial fullness index ( I P) and gonado‐somatic index ( I G) explained between 31 and 52% of the monthly variability in growth. Temperature and I P of capelin Mallotus villosus had significant effects on growth of all age groups and explained most of the variance for ages 6–8 and 4–5 years, respectively. The I P of large invertebrates (ages 4 to 7 years), sandlance ( Ammodytes sp. age 6 years) and demersal fishes (age 9 years) had age‐specific effects in the model. Overall, amphipods, decapods and echinoderms dominated the Atlantic cod diet in most seasons, but fish consumption by Atlantic cod was high in June and July, particularly on capelin. The rapid increase in somatic mass during June and July occurred despite cold water temperatures ( < 3° C at 50 m) and moderate to high gonado‐somatic index. The findings of this study suggest that when food was not a limiting factor, growth tended to increase even when Atlantic cod occupied colder waters, but when food was limiting, the opposite may have occured.  相似文献

The feeding ecology of the larvae of silver hake Merluccius bilinearis was examined during two time periods (October 1998 and December 1992) on the Western Bank, Scotian Shelf, north‐west Atlantic, and compared with the feeding ecology of Atlantic cod Gadus morhua larvae collected in the same samples in December 1992. During both time periods silver hake exhibited strong selection for late stage copepodids and adult copepods at a small size (>3·5 mm total length, L T). The niche width measured as the diet breadth index ( I DB) of silver hake declined rapidly as they increased in size and remained relatively constant from 3 to 11 mm L T, during each time period. Atlantic cod larvae exhibited a broader niche width that was curvilinear over the same L T. Atlantic cod were also less selective than silver hake, incorporating both naupliar and early stage copepodids in their diets throughout the length classes examined. Simple isometric relationships did not explain the differences in diet, as Atlantic cod larvae continued to feed on early stages of copepods at large size, while silver hake larvae quickly switched to large prey items. The strong selection and narrow I DB observed for silver hake probably reflects adaptation to spawning during the periods between major secondary production peaks in temperate waters.  相似文献

Summary The synthesis of protein and nucleic acids was studied by isotope incorporation and dilution in the plasmodia ofPhysarum polycephalum during periods of growth and differentiation (spherule formation). The total protein content decreased during starvation, but protein synthesis still occurred, probably at the expense of proteins previously synthesized during growth. Studies on leucine incorporation showed that protein synthesized during growth had a greater turnover than did protein formed by starving cultures, when both types of cultures were transferred to starvation conditions. Protein synthesis after prolonged starvation was rapidly and markedly decreased following the inhibition of RNA synthesis, whereas no such direct dependence on RNA synthesis was observed in growing cultures or during early starvation.The kinetics of RNA synthesis and the types of RNA formed were also shown to differ in growth and starvation. RNA turnover was low in growing cultures but substantial in starving cultures that were returned to growth medium. Qualitative differences in pulse-labeled RNA extracted from growing or starving cultures were revealed by methylated-albumin-kieselguhr column chromatography and sucrose gradient centrifugation. In starving cultures proportionately more labeled RNA was found in the lighter, non-ribosomal region of the gradient, and RNA from this region hybridized with denatured DNA to a greater extent than did other RNA fractions.This work was supported in part by Grant CA-07175 from the National Cancer Institute and by a grant from the Alexander and Margaret Stewart Trust Fund. Growth, survival and condition of redfish larvae Sebastes spp., reared in the laboratory (0, 500 1500 and 4500 prey l-1) were highest in the 1500 prey l-1 treatment. Significantly lower larval growth and survival in the 4500 prey l-1 treatment corresponded with lower prey bite: orient ratios in later weeks, suggesting that larvae were unable to forage efficiently at high prey densities. While these prey densities are higher than those reported in the field, naturally co-occurring Atlantic cod Gadus morhua larvae require higher prey densities when reared under similar conditions in the laboratory. These data suggest that prey availability may not be as limiting to redfish as for other commercially important marine species.  相似文献

Major temporal changes in the importance of euphausiids and Atlantic herring Clupea harengus in the diet of Atlantic cod Gadus morhua , 10–75 cm L F, occurred in shallow waters (<100 m depths) of the southern Gulf of St Lawrence between 1959 and 2000. Euphausiids represented 6–70% of prey mass for Atlantic cod 31–60 cm L F from 1959 to 1987 but only trace amounts were detected in stomachs collected from 1990 to 2000. Mysids and gammarid amphipods (for Atlantic cod ≤45 cm L F), and Atlantic herring (for Atlantic cod >45 cm L F) largely replaced euphausiids in the Atlantic cod diets from 1990 to 2000. This diet change suggested there has been a major perturbation of the food web of the southern Gulf of St Lawrence. The importance of fishes (mostly Atlantic herring) in the diet of Atlantic cod >45 cm L F increased significantly between the periods 1959–1980 and 1987–2000. Atlantic herring comprised 0–4% (mean 1.3%) of prey mass of Atlantic cod 46–60 cm L F from 1959 to 1980 and increased to 2–42% (mean 19.6%) of the diet from 1987 to 2000. Atlantic herring comprised 0–25% (mean 9.4%) of the prey mass of Atlantic cod 61–75 cm L F from 1959 to 1980 and increased to 42–81% (mean 54.2%) of prey mass from 1987 to 2000. This increased consumption of Atlantic herring was consistent with observed changes in abundance of Atlantic herring in the ecosystem between the late 1970s and 2000. The large changes in consumption of euphausiids and Atlantic herring represent highly significant changes and would need to be included in the development and interpretation of ecosystem-based management models for this ecosystem.  相似文献
