凝固剂对水稻花药培养愈伤组织诱导的影响   总被引:3,自引:1,他引:2  
本文初步研究了不同类型的凝固剂对水稻花药培养愈伤组织形成的影响。结果发现,用Gelrite、马铃薯淀粉、甘薯淀粉、可溶性淀粉代替琼脂可明显促进水稻花药培养愈伤组织的产生而尤以5.0%马铃薯淀粉为最佳。出愈率比琼脂增加5.2倍,达液体培养水平。以8个不同基因型,为材料研究发现,5.0%马铃薯淀粉作凝固剂,有7个材料出愈率高于对照,最高的BC 163比对照增加7.75倍,平均增加1.15倍。另外,以5.0%马铃薯淀粉作凝固剂代替0.8%琼脂可降低成本30%。因此,用马铃薯淀粉作凝固剂在水稻花药培养中可能具有潜在的应用价值。  相似文献

Summary A strain ofBacillus subtilis isolated from soil was found effective againstSclerotium rolfsii when grown on 20% potato extract for 48 hours and used along with the medium. Whole boiled potato was also a good medium but the bacteria needed 20 days to grow until insoluble starch was digested.In field trials the antagonist brought down the spread ofSclerotium and anthracnose diseases ofPiper betle. Against seed and air-borne diseases of jute caused byMacrophomina phaseoli,Diplodia corchori andColletotrichum corchori, the antagonist also showed promising results.Concluding part of a scheme supported by Food & Agriculture Council, Pakistan.  相似文献

Antibacterial and antifungal activity of crude extract, alcoholic extract and extracted phenol from various parts of tropical pteridophyta, Hemionitis arifolia were tested by agar diffusion and tube dilution assay. Both the crude and alcoholic extracts of vegetative and reproductive leaves of H. arifolia showed considerable antibacterial activity against Gram negative test strain of Escherichia coli (MTCC-739). Extract from reproductive leaves also showed moderate antibacterial activity against Bacillus subtilis (MTCC-441) (Gram positive test strain) but didn’t show any antifungal activity against Candida albicans (MTCC-7353). Mycorrhizal and other symbiotic association with the root system of H. arifolia was studied and it is revealed that a number of mycorrhizal strains were present in both vegetative and reproductive form. Presence of Dark Septate Endophytic Fungi (DSF) was also detected.  相似文献

A new species Trichophyton fischeri was isolated as a contaminant on blood agar plates. This fungus is believed to be a saprophyte. It may be confused with T. rubrum. On peptone dextrose agar plate, the growth is white and velvety to cottony. It occasionally forms furrows. The underside of the mature colony is brownish red. Clavate microaleuriospores are common. Trichophyton-type macroaleuriospores are produced occasionally on blood agar and potato dextrose agar. Erythritol does not stimulate T. fischeri to produce a red color on casamino erythritol albumen agar. Closterospore-like projections may be produced on the main filaments on peptone dextrose and potato dextrose agar.  相似文献

In the current study, twenty-eight soil samples were collected from coalmine sites of Telangana, India. The isolates were purified and identified based on their culture characterization on oatmeal agar, glycerol asparagine agar, yeast extract-malt extract agar, inorganic salt starch agar, and starch casein agar medium. Further, the supernatant of all the isolates were tested for antimicrobial and antifungal activities. The biochemical and microscopic studies of isolated strains results indicates the potential isolate strains belongs to Streptomyces genus. Among all the strains the biological activity of BHPL-KSKU5 showed higher anti-bacterial and anti-funagal activity. The molecular characterization of BHPL-KSKU5 16s rDNA gene sequence and phylogenetic tree showed that is mostly related to the Streptomysis felleus (S. felleus) strain. This isolate was submitted to gene bank NCBI with accession number MH553077. In addition, physiological studies such as utilization of carbon, nitrogen, amino acid sources of potential isolated were studied. Further, optimization, purification and characterization of the novel compound producing strain may be helpful for discovering the new therapeutic microbial agent.  相似文献

The phenolic extract of Acalypha leaves inhibited growth of Gloeophyllum sepiarium and Pleurotus sp. (test wood-rot fungi) in potato dextrose agar, starch agar, starch glucose agar, carboxyl methyl cellulose agar and carboxyl methyl cellulose glucose agar. Fungicidal or fungistatic concentration of the extract (10–14 mg/ml) depended on the medium. However a lower concentration of the extract (8–10 mg/ml) in combination with Trichoderma viride culture filtrate caused a similar inhibitory pattern. Degradation of obeche (Triplochiton scleroxylon), mahogany (Khaya ivorensis) and walnut (Lovoa trichilioides) by the test fungi was limited or prevented by extract treatment of 8–10 mg/g wood. A similar inhibitory effect again occurred when a combination of T. viride filtrate and lower extract concentration (6–8 mg extract per gram of wood) was used. On-going wood decay was limited or halted by a combined treatment involving 8–12 mg extract per gram of wood depending on the fungal residence period. Treated stakes exposed to 6 months of tropical wet season retained resistance to fungal attack including soft rot. The phenolic extract of A. hispida may prove useful in an integrated chemical and biological approach to wood treatment.  相似文献

The capacity of four bacterial strains isolated from productive soil potato fields to solubilize tricalcium phosphate on Pikovskaya agar or in a liquid medium was evaluated. A bacterial strain was selected to evaluate in vitro capacity of plant-growth promotion on Solanum tuberosum L. culture. Bacterial strain A3 showed the highest value of phosphate solubilization, reaching a 20 mm-diameter halo and a concentration of 350 mg/l on agar and in a liquid medium, respectively. Bacterial strain A3 was identified by 16S rDNA analysis as Bacillus pumilus with 98% identity; therefore, it is the first report for Bacillus pumilus as phosphate solubilizer. Plant-growth promotion assayed by in vitro culture of potato microplants showed that the addition of bacterial strain A3 increased root and stems length after 28 days. It significantly increased stem length by 79.3%, and duplicated the fresh weight of control microplants. In this paper, results reported regarding phosphorus solubilization and growth promotion under in vitro conditions represent a step forward in the use of innocuous bacterial strain biofertilizer on potato field cultures.Key words: Bacillus sp., phosphorus soluble, Pikovskaya agar, potato rhizosphere, plant growth promoting rhizobacteria  相似文献

采用琼脂打孔法和平板涂布法,观察五倍子提取物对常见致病菌的抑菌效果,并以金黄色葡萄球菌、大肠杆菌的抑菌环直径为参考指标,考察紫外光照时间、温度及介质pH值对五倍子提取物抑菌活性的影响。结果表明,五倍子提取物对革兰氏阳性菌的抑菌效果明显强于革兰氏阴性菌,在稳定性研究过程中发现,光照时间、温度及介质pH值对五倍子提取物的抑菌效果影响较大,因此在选用五倍子提取物作植物抑菌剂时应现配现用。  相似文献

Ten cardiovascular drugs, having diverse pharmacological action, were screened for possible antimicrobial property against known eight sensitive bacteria, belonging to Gram positive and Gram negative types. Although five drugs failed to show antimicrobial activity and three had moderate antimicrobial action, oxyfedrine HCl and dobutamine were seen to possess pronounced antimicrobial property. Oxyfedrine was further tested in vitro against 471 strains of bacteria from two Gram positive and fourteen Gram negative genera. The minimum inhibitory concentration (MIC) of oxyfedrine was determined by agar dilution method, which ranged from 50-200 microg/ml in most of the strains, while some strains were inhibited at even lower concentrations. In animal experiments, this compound was capable of offering significant protection to Swiss strain of white mice, challenged with 50 median lethal dose (MLD) of a virulent strain of Salmonella typhimurium at concentrations of 15, 30 and 60 microg/mouse. The sensitivity of two Gram positive (Staphylococcus aureus and Bacillus subtilis) and two Gram negative (Escherichia coli and Pseudomonas aeruginosa) pathogenic multi-drug resistant bacteria was tested against the crude extracts (cold aqueous, hot aqueous, and methanol extracts) of leaves and seeds of Argemone mexicana L. (Papaveraceae) by agar well diffusion method. Though all the extracts were found effective, yet the methanol extract showed maximum inhibition against the test microorganisms followed by hot aqueous extract and cold aqueous extract.  相似文献

为了克服水解马铃薯淀粉不易获得的困难,并使“水平切片淀粉凝胶电泳同工酶分析”更容易开展,普通的化学试剂马铃薯淀粉(或精制食用马铃薯淀粉)和可溶性淀粉混合物加入适当的添加剂被用来代替水解马铃薯淀粉制作凝胶。试验结果表明:用8~10%的上述混合淀粉(5∶3),添加1%的琼脂粉和2~4%的蔗糖,所制成的“复合淀粉凝胶”可以很好地被切片,并成功地对许多不同类群的植物材料的PGM、PGI、MDH、AAT、SKDH、6PGD、IDH和ME等酶进行染色。  相似文献

Microbacterium aurum strain B8.A was isolated from the sludge of a potato starch-processing factory on the basis of its ability to use granular starch as carbon- and energy source. Extracellular enzymes hydrolyzing granular starch were detected in the growth medium of M. aurum B8.A, while the type strain M. aurum DSMZ 8600 produced very little amylase activity, and hence was unable to degrade granular starch. The strain B8.A extracellular enzyme fraction degraded wheat, tapioca and potato starch at 37 °C, well below the gelatinization temperature of these starches. Starch granules of potato were hydrolyzed more slowly than of wheat and tapioca, probably due to structural differences and/or surface area effects. Partial hydrolysis of starch granules by extracellular enzymes of strain B8.A resulted in large holes of irregular sizes in case of wheat and tapioca and many smaller pores of relatively homogeneous size in case of potato. The strain B8.A extracellular amylolytic system produced mainly maltotriose and maltose from both granular and soluble starch substrates; also, larger maltooligosaccharides were formed after growth of strain B8.A in rich medium. Zymogram analysis confirmed that a different set of amylolytic enzymes was present depending on the growth conditions of M. aurum B8.A. Some of these enzymes could be partly purified by binding to starch granules.  相似文献

Biological and physiological characteristics of Neotyphodium gansuense were compared with Neotyphodium coenophialum and Epichlo? festucae at a range of temperatures and pH values, and on carbon and nitrogen amended media. N. gansuense was able to grow at 10-30 degrees C, but not at 5 degrees C, and slowly at 35 degrees C. The optimal temperature for both N. gansuense and N. coenophialum was 25 degrees C, but that of E. festucae was 20-25 degrees C. The optimal pH ranges for mycelial growth of N. gansuense, N. coenophialum and E. festucae were 5-9, 5-9 and 5-7, respectively. The Neotyphodium and Epichlo? endophytes varied in their ability to grow on media containing different carbon and nitrogen nutrients. The preference of N. gansuense for carbon source was sucrose>glucose, lactose, sorbitol, inulin, maltose, mannitol, starch, fructose>xylose. Growth of all three endophytes tested was significantly improved by peptone, tryptone, casein, yeast extract and l-proline. Yeast extract, peptone, casein, tryptone, l-proline, potassium nitrate, ammonium oxalic acid and l-leucine significantly improved growth of N. gansuense. However, ammonium nitrite was not utilized at all by any tested endophyte. N. gansuense grew significantly better on potato dextrose agar (PDA) and oat meal agar (OMA) than on corn meal agar (CMA) and drunken-horse-grass agar (DA), and most slowly on water agar (WA) and saltwater nutrient agar (SNA).  相似文献

The influence of nitrogen availability on the chemical structure of agar polymers isolated fromGracilaria verrucosa strain G-16 was ascertained by sequential solvent extraction and carbon 13 nuclear magnetic resonance (13C NMR) spectroscopy. Agar isolated fromG. verrucosa strain G-16 cultured under nitrogen limited conditions showed large non-polar components but produced spectra indicative of only minor amounts of methylation. These agars also produced spectra suggesting the presence of floridean starch. The nitrogen supplemented cultures ofG. verrucosa strain G-16 produced agars of a more polar nature (hot-water soluble) and contained little detectable starch. The data suggest that the higher gelling temperatures of agar from nitrogen limited plants is not due to markedly higher methylation. These data also suggest that nitrogen effects on agar content may not be as significant as previously thought.  相似文献

A raw starch utilizing microbe was isolated from mud in a milling factory. The 16S ribosomal DNA (rDNA) sequencing and morphological properties of the strain indicated that it belongs to the genus Streptomyces. A strongly raw starch digesting amylase was purified from the culture supernatant of the strain by chromatographic procedures. The specific activity of the enzyme was 11.7 U/mg, molecular mass 47 kDa, optimum pH 6.0, and optimum temperature 50 to 60 degrees C. The enzyme showed sufficient activity even at 70 degrees C. It was activated by calcium, cobaltous, and magnesium ions, and inhibited by copper, nickel, zinc, and ferrous ions. It formed maltose mainly from raw and gelatinized starch, and glycogen. No products were formed from glucose, maltose, maltotriose, pullulan, or cyclodextrins (CDs). The enzyme digested raw wheat, rice, and waxy rice starch rapidly, and raw corn, waxy corn, sweet potato, tapioca, and potato starch normally.  相似文献

A hyaluronic acid (HA)-producing strain, Streptococcus zooepidemicus NJUST01, can grow in a serum-free agar medium, with starch as exclusive carbon source, but not glucose, sucrose, dextrine, xylose, or lactose. In this starch medium, the strain NJUST01 reproduced successively at 37°C for 60 generations, with no obvious variation on morphology and physiology, but colonies of the strain after 60th generation could not produce a clear hemolytic zone on sheep blood agar plates. Hyaluronic acid production by the strain NJUST01 was analyzed relative to the starch medium. Employing a multifactor cross experiment, an optimum medium revealed for hyaluronic acid production was composed of 5% starch, 0.3% glucose, 0.5% peptone, 0.15% MgSO4, and 2.0% K2HPO4. The amount of HA 6.7 g/l was obtained in batch fermentation on a rotary shaker at 37°C, 220 rpm for 36 h.  相似文献

Pythium acanthophoron (two isolates) was as aggressive as P. oligandrum in hyphal interactions with Fusarium oxysporum on water agar films. It caused rapid post-contact lysis or cytoplasmic coagulation of the host, and often branched and penetrated the host at contact points. P. acanthophoron grew across a range of fungi on pre-colonized agar plates; the range was narrower than for P. oligandrum but broader than for P. mycoparasiticum. In chemically defined liquid media, P. acanthophoron was unique among the six known mycoparasitic Pythium spp. in requiring organic nitrogen but not thiamine. It also grew on mannitol as the sole carbon source in the presence of calcium or sterols or both. However, individual isolates of the three mycoparasitic species showed different responses to calcium and the sterols ergosterol, cholesterol and beta-sitosterol when grown on mannitol. All three mycoparasites required components of molasses, carrot extract or sunflower seed extract to produce oogonia in culture; they formed few oogonia on potato extract, with or without dextrose, even when supplied with sterols. The three mycoparasites were less tolerant of high NaCl levels in culture than were three phytopathogens (P. aphanidermatum, P. ultimum, P. graminicola), in contrast to a previous report for P. oligandrum and P. ultimum. These in vitro studies suggest that P. acanthophoron has potential for use as a biocontrol agent instead of, or in addition to, P. oligandrum.  相似文献

An endolithic bacterium, strain RSBr-1, was isolated from the inside of a piece of red sandstone from coastal areas of Scotland. RSBr-1 was Gram negative, oxidase and catalase positive, and cells were non-motile rods. Sodium was required for growth. The optimum sodium chloride concentration and pH for growth were 4% and pH 8.0, respectively. Eumelanin was produced in marine broth and in BY medium. RSBr-1 hydrolyzes chitin, esculin, gelatin, and starch, but not agar. Nitrate reduction is positive. Taxonomic characterization of this strain indicated that it belongs to the genus Microbulbifer. The difference between the aligned 16S rDNA sequences of RSBr-1 and the closest relative, M. elongata, is greater than the difference between the 16S rDNA sequences of M. hydrolyticus and M. elongata. On the basis of the phenotypic and genotypic comparison of this isolate with the other strains, RSBr-1 is proposed as a new species, Microbulbifer arenaceous, with type strain RSBr-1.  相似文献

Oxytetracycline–glucose–yeast extract agar (OGYA), gentamicin–glucose–yeast extract agar (GGYA) with the antibiotic added separately or sterilized with the medium, chlortetracycline–Rose Bengal agar (CRA), chloramphenicol-streptomycin agar (PYA) and to a lesser extent, oxytetracycline–gentamicin–glucose–yeast extract agar (OGGYA) with or without Rose Bengal added, have been compared for the selective enumeration of moulds in foods. The results obtained from dried cereal products show that the media are almost equally productive and selective when applied to such foods, but Rose Bengal limits the size of mould colonies. When examining fresh proteinaceous foods, such as minced meat and chicken, CR agars and to a certain extent gentamicin-containing agars show the distinct advantage of being much more inhibitory towards the psychrotrophic Gram negative rods that predominate in the associated flora of such foods. Oxytetracycline–glucose–yeast extract agar lost its bacteriostatic properties when heavily challenged with proteinaceous substrates and/or incubated for longer periods at 37° or even at 25°. For such applications chloramphenicol was found to be the antibiotic of choice.  相似文献
