The Waste Isolation Pilot Plant (WIPP) is a salt mine constructed 650 meters below the ground surface by the United States Department of Energy. The facility will be used for permanent disposal of transuranic wastes. This underground repository has been constructed in the geologically stable Permian age Salado salt formation. Of the wastes to be placed into the facility, 85% will be biodegradable cellulose. A 3-year survey of the bacterial populations existing within the facility was conducted. Bacterial populations were found to be heterogeneously distributed throughout the mine. Populations in some mine areas reached as high as 1.0 × 104 colony-forming units per gram of NaCl. The heterogeneous distribution of bacteria within the mine did not follow any recognizable pattern related to either age of the workings or to human activity. A biochemical comparison between ten known species of halophilic bacteria, and strains isolated from both the mine and nearby surface hypersaline lakes, showed the presence of extreme halophiles with wide biochemical diversity, some of which could prove to represent previously undescribed groups. The halophilic bacteria isolated from the mine were found to degrade cellulose and a wide variety of other carbon compounds. When exposed to two types of common laboratory paper, the cellulose-degrading halophiles attached to the substrate within 30 minutes of inoculation. Cultures enriched directly from a brine seep in the mine easily destroyed both papers and produced detectable amounts of oxalacetic and pyruvic acids. The combination of heterogeneity in the distribution of organisms, the presence of a physiologically diverse community, and the relatively slow metabolism of cellulose may explain several long-standing debates about the existence of microorganisms in ancient underground salt formations. Abstract A constitutive NAD+-dependent glycerol dehydrogenase activity was detected in Halobacterium salinarium and Halobacterium cutirubrum . Optimal activity was found at 3 M KCl and pH 8–10. No glycerol dehydrogenase activity could be demonstrated in representatives of the genera Haloferax and Haloarcula , even when grown in the presence of glycerol, or in Halobacterium saccharovorum and Halobacterium sodomense . Glycerol kinase activity was shown to be present constitutively in all halophilic archaea examined. The finding that glycerol dehydrogenase is found only in part of the halophilic arachaea makes dihydroxyacetone an improbable candidate as the precursor for the glycerol moiety of halobacterial lipids.  相似文献

A gene encoding NADP-dependent Ds-threo-isocitrate dehydrogenase was isolated from Haloferax volcanii genomic DNA by using a combination of polymerase chain reaction and screening of a lambda EMBL3 library. Analysis of the nucleotide sequence revealed an open reading frame of 1260 bp encoding a protein of 419 amino acids with 45837 Da molecular mass. This sequence is highly similar to previously sequenced isocitrate dehydrogenases. In the alignment of the amino acid sequences with those from several archaeal and mesophilic NADP-dependent isocitrate dehydrogenases, the residues involved in dinucleotide binding and isocitrate binding are well conserved. We have developed methods for the expression in Escherichia coli and purification of the enzyme from H. volcanii. This expression was carried out in E. coli as inclusion bodies using the cytoplasmic expression vector pET3a. The enzyme was refolded by solubilisation in 8 M urea followed by dilution into a buffer containing EDTA, MgCl(2) and 3 M NaCl. Maximal activity was obtained after several hours incubation at room temperature.  相似文献

Injection of a suspension of the Gram-positive bacterium Bacillus cereus into the haemocoel of Galleria mellonella larvae caused a rapid decrease in the number of circulating plasmatocytes. A similar phenomenon was observed following the injection of cell-free haemolymph collected from larvae previously challenged with bacteria, indicating that plasmatocyte depletion was mediated by a humoral factor released into the haemolymph. The release of plasmatocyte depletion factor could be effected in vitro by incubating adherent haemocytes with a suspension of B. cereus. The in vitro production of the factor did not require the presence of exogenous humoral components, was dose-dependent, was inhibited at low temperatures, and showed the same specificity with respect to bacterial species as observed in vivo. Activity of the plasmatocyte depletion factor destroyed by a 15-min incubation at 56°C. The release of the factor appears to be an early biochemical signal in the complex cellular response of this insect to bacterial infection.  相似文献

Excess brain manganese can produce toxicity with symptoms that resemble those of Parkinsonism and causes that remain elusive. Manganese accumulates in mitochondria, a major source of superoxide, which can oxidize Mn2+ to the powerful oxidizing agent Mn3+. Oxidation of important cell components by Mn3+ has been suggested as a cause of the toxic effects of manganese. Determining the oxidation states of intramitochondrial manganese could help to identify the dominant mechanism of manganese toxicity. Using X-ray absorbance near edge structure (XANES) spectroscopy, we have characterized the oxidation state of manganese in mitochondria isolated from brain, liver, and heart over concentrations ranging from physiological to pathological. Results showed that (i) spectra from different model manganese complexes of the same oxidation state were similar to each other and different from those of other oxidation states and that the position of the absorption edge increases with oxidation state; (ii) spectra from intramitochondrial manganese in isolated brain, heart and liver mitochondria were virtually identical; and (iii) under these conditions intramitochondrial manganese exists primarily as a combination of Mn2+ complexes. No evidence for Mn3+ was detected in samples containing more than endogenous manganese levels, even after incubation under conditions promoting reactive oxygen species (ROS) production. While the presence of Mn3+ complexes cannot be proven in the spectrum of endogenous mitochondrial manganese, the shape of this spectrum could suggest the presence of Mn3+ near the limit of detection, probably as MnSOD.  相似文献

Isolation, molecular identification and phylogenetic analysis were carried out to investigate the biodiversity of manganese bacteria in sediments which were collected from the Arctic Ocean during the 2nd Chinese Arctic Scientific Expedition. 21 and 19 species of cultivable strains were isolated from sediments at Stations P11 and S11, respectively, according to their distinct morphological character on the screening plate of manganese medium. Molecular identification and phylogenetic analysis showed that the cultivable manganese bacteria from Station P11 were basically composed of γ-Proteobacteria (γ subgroup of the Proteobacteria branch of the domain Bacteria) and Actinobacteria, which accounted for 86% and 14%, respectively. The isolates of γ-Proteobacteria mainly included Psychrobacter, Shewanella, Acinetobacter and Marinobacter, of which Psychrobacter was the major genus, which accounted for 67% of the γ-Proteobacteria. The cultivable manganese bacteria from Station S11 included α-Proteobacteria, γ-Proteobacteria and Flavobacteria of Bacteroides. The γ-Proteobacteria mainly included Shewanella, Marinomonas and Alteromonas. The majority of α-Proteobacteria was Sphingomonas. The phylogenetic analysis indicated that bacteria from sediments at Stations P11 and S11 had different cultivable manganese microbial communities. All tested strains had higher resistance to Mn2+, of which Marinomonas sp. S11-S-4 had the highest resistant ability.  相似文献

对中国第二次北极科学考察采集的北极海洋沉积物中的锰细菌进行了筛选、分离和系统发育分析。根据其在筛选平板上菌落的形态学特征,分别从站位P11和S11采集的沉积物中分离到了21株和19株锰细菌。系统发育分析表明,两个站位的锰细菌群落组成有着明显的差别。站位P11分离的可培养锰细菌主要由细菌域(Bacteria)中变形杆菌门的γ-变形杆菌纲(γ-Proteobacteria)和放线菌纲(Actinobacteria)组成,二者分别占86%和14%;γ-变形杆菌纲主要包括嗜冷杆菌属(Psychrobacter)、希瓦氏菌属(Shewanella)、假交替单胞菌属(Pseudoaheromonas)、不动杆菌属(Acinetobacter)、海杆菌属(Marinobacter),其中以嗜冷杆菌属为主,其比例可达67%。从站位S11分离到的可培养锰细菌主要包括细菌域中变形杆菌门的α-变形杆菌纲(α-Proteobacteria)和γ-变形杆菌纲以及拟杆菌门(Bacteroides)中的黄杆菌纲(Flavobaeteria);γ-变形杆菌纲主要包括希瓦氏菌属、海单胞菌属(Marinomonas)和交替单胞菌属(Aheromonas),α-变形杆菌纲主要为鞘氨醇单胞菌属(Sphingomonas)。实验菌株均对Mn^2+有着较强的抗性,其中以菌株Marinomonas sp.S11-S-4耐受性最高。  相似文献

Abstract Gene transfer among microorganisms has been well demonstrated in laboratory microcosms and in situ, under non-limiting nutrient conditions. The literature contains conflicting opinions, however, as to whether such processes could occur in the absence of nutrients. This review summarises the evidence for the occurrence of gene transfer by conjugation, transformation and transduction among non-growing bacteria in nutrient depleted environments. Conjugation by selftransmissible, or by non-selftransmissible but mobilisable, plasmids has been shown to occur among environmental isolates of Escherichia coli, Enterobacter cloacae, Pseudomonas aeruginosa and marine Vibrio strains. Transduction and transformation have been demonstrated in isolates of P. aeruginosa and marine Vibrio strains, respectively. It is possible that the mechanisms of these processes may be different in non-growing cells in nutrient depleted conditions, compared to those occurring in cells growing in rich media.  相似文献

Although health risk due to discoloured water is minimal, such water continues to be the source of one of the major complaints received by most water utilities in Australia. Elevated levels of iron (Fe) and/or manganese (Mn) in bulk water are associated with discoloured water incidents. The accumulation of these two elements in distribution systems is believed to be one of the main causes for such elevated levels. An investigation into the contribution of pipe wall biofilms towards Fe and Mn deposition, and discoloured water events is reported in this study. Eight laboratory-scale reactors were operated to test four different conditions in duplicate. Four reactors were exposed to low Fe (0.05?mg?l?1) and Mn (0.02?mg?l?1) concentrations and the remaining four were exposed to a higher (0.3 and 0.4?mg?l?1 for Fe and Mn, respectively) concentration. Two of the four reactors which received low and high Fe and Mn concentrations were chlorinated (3.0?mg?l?1 of chlorine). The biological activity (measured in terms of ATP) on the glass rings in these reactors was very low (~1.5 ng cm?2 ring). Higher concentrations of Fe and Mn in bulk water and active biofilms resulted in increased deposition of Fe and Mn on the glass rings. Moreover, with an increase in biological activity, an increase in Fe and Mn deposition was observed. The observations in the laboratory-scale experiments were in line with the results of field observations that were carried out using biofilm monitors. The field data additionally demonstrated the effect of seasons, where increased biofilm activities observed on pipe wall biofilms during late summer and early autumn were found to be associated with increased deposition of Fe and Mn. In contrast, during the cooler months, biofilm activities were a magnitude lower and the deposited metal concentrations were also significantly less (ie a drop of 68% for Fe and 86% for Mn). Based on the laboratory-scale investigations, detachment of pipe wall biofilms due to cell death or flow dynamics could release the entrapped Fe and Mn into the bulk water, which could lead to a discoloured water event. Hence, managing biofilm growth on drinking water pipelines should be considered by water utilities to minimize accumulation of Fe and Mn in distribution networks.  相似文献

Summary The effect of manganese on the development of vesicular-arbuscular (VA) mycorrhizae and on the population of Mn-oxidizing soil bacteria was studied using red clover as host plant and Glomus mosseae or G. aggregatum as VA-colonizing fungi. The addition of Mn to the substrate in which plants grew had a detrimental effect on root colonization, G. aggregatum being more susceptible than G. mosseae. Mn uptake was lower in mycorrhizal than in non-mycorrhizal plants, especially when colonized by G. mosseae. The development of mycorrhizae seems to favour the Mn-oxidizing bacteria population.  相似文献

外生菌根真菌对于酸性和锰污染土壤的植树造林和生态恢复有重要作用。采用液体培养方法,以大白菇Rd Fr(Russula delica Fr.)、彩色豆马勃Pt 715(Pisolithus tinctorius 715)、土生空团菌Cg Fr(Cenococcum geophilum Fr.)和厚环粘盖牛肝菌Sg Kl S(Suillus grevillei(Kl.)Sing)为供试对象,研究了Mn2+对外生菌根真菌生长、养分吸收、有机酸和氢离子分泌的影响,以及锰在菌丝细胞内外的分布情况。结果表明:在0—800 mg Mn2+/L的培养液中,Mn2+对Rd Fr生长无显著影响;低浓度的Mn2+刺激Sg Kl S生长,中、高浓度无抑制作用;但大幅度降低Pt 715和Cg Fr的生物量,说明Rd Fr和Sg Kl S抗(耐)锰的能力较强。在Mn2+胁迫下,供试菌株的氮、钾含量和吸收量显著降低;含磷量和吸收量,以及草酸和柠檬酸的分泌速率因菌株不同而表现出多样性,说明在减轻Mn2+毒的过程中,磷酸盐(或聚磷酸盐)对Mn2+固定作用和有机酸的络合作用因菌株不同而异。但是,Mn2+显著降低Rd Fr和Sg Kl S的氢离子分泌速率,菌丝和原生质中的含Mn量显著低于敏感性菌株,说明降低Mn2+的活性和减少吸收可能是外生菌根真菌抗(耐)Mn2+的重要机制。此外,菌丝吸收的Mn2+绝大部份存在于质外体,少量进入细胞,前者是后者的5.23—9.21倍,说明原生质膜是外生菌根真菌防御Mn2+进入细胞的重要屏障。  相似文献

Autopsied liver tissue samples collected from 42 males and 31 females were analyzed for copper, manganese and zinc using atomic absorption spectrometry (AAS). With the exception of two liver samples for which the copper levels were determined to be 74.8 and 104.0 g/g (dry weight), hepatic copper concentrations were found to range from 1.7 to 32.4 g/g with a mean concentration of 14.2 g/g and standard deviation of 7.0 g/g. Manganese concentrations (with the exception of one sample having 12.9 g/g) ranged from 0.22 to 4.6 g/g with a mean of 2.26 ± 1.00 g/g. Hepatic zinc levels averaged 118.3 ± 44.4 g/g and ranged from 38.5 to 231.3 g/g. There were no apparent trends for the levels of any metals versus age nor were there any differences in average hepatic metal concentrations for males and females. © Rapid Science 1998.  相似文献

【背景】目前对水库水体污染原因的研究往往专注于水体的富营养化、pH值、溶解氧、氨氮、菌落总数指标的变化,而重金属含量与环境因子的季节性变化相关性分析研究较少,同时对于典型季节原位微生物种群的多样性差异研究尚未见报道。【目的】研究浙江省台州市长潭水库底部水中正二价金属离子(二价锰离子Mn2+;二价铁离子Fe2+)浓度与不同环境因子的季节性变化规律,并对其相关性进行分析;富集平水期(2月)和丰水期(8月)水库底部水体中功能微生物菌群,分析其种类和丰度的差异。【方法】分别检测12个月的水库水Mn2+和Fe2+浓度及多种环境因子(水体溶解氧浓度、pH值、总磷浓度、浊度、水库环境温度及降水量),过滤并富集培养水库底部水体的功能微生物菌群,对其16S rRNA基因V3-V4区测序并分析其菌群结构。【结果】长潭水库水中Mn2+和Fe2+浓度呈季节性变化,每年的春夏交替季节水体中铁锰含量从零开始慢慢升高,至夏秋高温季节水体中Mn2+和Fe2+浓度达到最高值,然后慢慢降低,至秋末冬初检测不到含量。在检测的多种环境因子中,水体溶解氧浓度、水库环境温度及降水量呈明显的季节性规律变化。Mn2+和Fe2+浓度与温度、降水量和浊度有正相关性,与溶解氧浓度、pH值和总磷浓度有负相关性,其中在正负相关性分析中两种金属离子的浓度与溶解氧浓度的相关性最强,其次是环境温度及降水量。丰水期和平水期中富集获得的功能微生物菌群的种类和丰度差异很大,从属水平上分类,丰水期时菌群只包含不动杆菌(Acinetobacter)和鲑色沉积物杆状菌(Sediminibacterium)这2个属的菌株,含量各占约50%;平水期时菌群则主要由杆菌属(Bacillariophyta) (47.62%)和Limnohabitans(9.52%)等9个属的菌株构成。富集获得的平水期和丰水期的两个可培养的菌群均具有去除水库水中Mn2+的功能,去除率分别约为35.9%和11.4%。【结论】长潭水库底部水体中Mn2+和Fe2+浓度与不同环境因子均呈季节性规律变化,它们之间呈现不同的正负相关性,丰水期和平水期的功能微生物菌群结构差异很大。本研究为利用微生物进行重金属污染水体的治理储备了微生物资源,为实现国家“美丽乡村”的建设目标提供一定的参考价值。  相似文献

A number of essential trace elements play a major role in various metabolic pathways. Selenium (Se), manganese (Mn), copper (Cu), zinc (Zn), and iron (Fe) are essential trace elements that have been studied in many diseases, including autoimmune, neurological, and psychiatric disorders. However, the findings of previous research on the status of trace elements in patients with schizophrenia have been controversial. We studied these elements in patients with a DSM-IV diagnosis of schizophrenia and compared them with sex- and age-matched healthy controls. Plasma Cu concentrations were significantly higher (p<0.01) and Mn and Fe concentrations were lower (p<0.05 and p<0.05, respectively) in schizophrenic patients than in controls. Se and Zn concentrations and protein levels did not differ between patients and healthy controls. These observations suggest that alterations in essential trace elements Mn, Cu, and Fe may play a role in the pathogenesis of schizophrenia. However, findings from trace element levels in schizophrenia show a variety of results that are difficult to interpret.  相似文献

The goal of these experiments was to test the hypothesis that serotonin (5-HT) is involved in facilitation of the shortening reflex in the leech Hirudo medicinalis. For this reason, we have used the toxin 5-hydroxytryptamine (5,7-DHT) to deplete serotonin from the nervous systems of intact leeches and have assessed the effect on early facilitation, dishabituation, and sensitization of the touch-elicited shortening reflex using behavioral procedures previously developed in our lab (Boulis and Sahley, 1988). We find that 5,7-DHT lesions completely attenuate early facilitation and sensitization but only reduce dishabituation of the touch-elicited shortening reflex. Histological analyses of the ganglia from these leeches using glyoxilic acid staining procedures revealed an absence of staining in the Retzius cell of experimental leeches. All other serotonin-containing neurons showed glyoxilic acid staining comparable to that observed in the control leeches.  相似文献

The relationship between taurine concentrations of plasma, whole blood, serum and skeletal muscle during taurine depletion and repletion was investigated in cats, to identify the most useful indicators of taurine status. Sixteen cats were fed a purified diet containing either 0 or 0.15 g/kg taurine for 5 months. Treatments were then reversed and the taurine concentration was measured during repletion and depletion phases. Plasma taurine exhibited the fastest rate (slow component) of depletion (t 1/2 = 4.8 wk), followed by serum (5.3 wk), whole blood (6.2 wk), and skeletal muscle (11.2 wk). Whole blood taurine was the first to replete at a rate of 0.74 wk to 1/2 maximal repletion, followed by serum (2.1 wk), skeletal muscle (3.5 wk), and plasma (3.5 wk). Whole blood more closely reflected skeletal muscle taurine concentrations than plasma during depletion, while plasma taurine concentrations appear to be the most valuable predictor of skeletal muscle taurine concentrations during repletion. This study suggests that the best clinical method to evaluate the taurine status of the cat is the determination and interpretation of both plasma and whole blood taurine concentrations.  相似文献
