Soybean (Gtycine max (L.) Merr. cv. Fiskeby V), white clover(Trifolium repens L. cv Blanca) and lucerne (Medicago sativaL.cv. Europe) nodules grown in fluid culture of Perlite wereexamined by cryo-scanning electron microscopy. The surfacesof all three species consisted of loosely packed cortical cells,collapsed areas covering intact cells and in soybean, troughswhich lacked the layers of loosely packed cells and were coveredby an amorphous matrix. The superficial cortical cells werehydrophobic and their surfaces were covered by a solvent-extractablestippling. This stippling was absent from the surfaces of underlyingcells. Air-filled intercellular spaces in fractured noduleswere seen throughout the cross-section. Nitrogen fixation, intercellular pathways, hydrophobicity, oxygen diffusion, cryo-SEM, soybean, Glycine max (L.) Merr. cv. Fiskeby V, clover, Trifolium repens L. cv. Zinc-induced Vacuolation in Root Meristematic Cells of Cereals   总被引:1,自引:0,他引:1  
In the absence of Zn, vacuolar volume fractions of root meristematiccells of Secale cereale L. cv. K2, Triticum aestivum L. cv.Chinese Spring and Oryza sativa L. cv. IR34 were 5.64 x 10–2,2.17 x 10–2 and 1.63 x 10–2 µm3 vacuole µm–3tissue, respectively. A 4-d exposure to a subtoxic concentrationof zine (0.2 µg Zn cm–3) induced a 2.93-fold anda 6.78-fold increase in the total vacuolar volume fraction inOryza and Triticum, respectively, whereas no significant increasewas observed for Secale. It is proposed that this Zn-inducedvacuolation represents a compartmentalization mechanism. Theinitial total vacuolar volume fraction in Secale was greaterthan that for Oryza and Triticum and this may enable compartmentalizationof the metal soon after the onset of treatment so reducing itscytotoxic effects. These findings are similar to those observedin contrasting cultivars of Festuca rubra L. Triticum aestivum L, Secale cereale L, Oryza sativa L, zinc, root meristem, vacuolation  相似文献

A cultivar from each of four cereal species (Avena sativa L.,cv. Swan, Hordeum vulgare, L., cv. Clipper, Triticum aestivumL., cv. Gabo, and Secale cereale L., cv. South Australian Rye)was grown in controlled environment chambers in a 10-h photoperiod(short days) or 10-h photoperiod supplemented with a 6-h extensionby incandescent light. The developmental morphology of the inflorescenceswas followed to ascertain whether there were any common developmentalinterrelationships between the species. Inflorescence internodeelongation was initiated when the floret initial first appeared,irrespective of whether it occurred on the most advanced lateralspikelet or on the terminal spikelet of the rachis. The glumes(infertile bracts) of the terminal spikelet of the rachis wereinitiated when the first second-order inflorescence branch appeared,irrespective of whether the second-order inflorescence branchwas a floret initial or a lateral spikelet, as in Triticum sp.,or an inflorescence (panicle) branch, as in Avena sp. Cessationof the activity of the apical meristem, as measured by primordiumformation, was not correlated with any particular stage of floraldevelopment but appeared to be due to a lack of nutrients causedby an increasing competitiveness for the available nutrientsfrom the developing spikelets which are situated closer to thevascular system than the apical meristem.  相似文献

Second growth is an important physiological disorder of thepotato (Solanum tuberosum L.) plant. A model system to studysecond growth was developed using one-leaf cuttings. Photoperiod,temperature, decapitation and leaf removal treatments were carriedout on the plants from which the cuttings were taken and onthe cuttings themselves. Tuberized, one-leaf cuttings takenfrom moderately-induced plants and exposed to 35 °C afterleaf removal showed 95% second growth within 10 d after treatmentinitiation. Conditions that promoted second growth also reducedstarch and dry-matter content, even in tubers that did not developsecond growth. We have generated a telomere-specific probe by the polymerasechain reaction and used it to localize chromosome telomeresof ten unrelated angiosperm species in in situ. Concatenationof the simple monomers, 5'-(TTTAGGG)-3', derived from the sequenceof Arabidopsis thaliana telomeres, yielded a stable, versatileand reliable probe that gave a signal of high intensity followingfluorescence in situ hybridization. Most species, includingthose with known karyotype rearrangements, showed telomere labelonly at chromosome termini. Methionine sulfoximine caused ammonia accumulation and photosyntheticrate inhibition in leaf discs from two C4 species, Zea maysL. cv. F. M. Cross (Hybrid) and Sorghum bicolor L. Moench cv.NC-70X, as well as one C3 plant species, Datura stramonium L.cv. stramonium. Similar results were obtained earlier with theC3 species Spinacia oleracea L. The effect occurred in the absenceof inorganic nitrogen reduction and was light dependent. Germination responses to alternating daily cycles involvingtransfers between different temperatures over the range 0 toc. 40?C were investigated in four species: Lycopus europaeusL., Clarkia unguiculata Lindl., Apium graveolens L. cv. GiantRed, and Silene dioica (L.) Clairv. Comparisons of the germination of these species revealed distinctivedifferences which could be related to particular features ofthe geographical regions or ecological habitats in which theyoccur naturally. The effects of fluctuating as opposed to constant temperaturesvaried from one species to another. L. europaeus depended almostabsolutely on fluctuating temperatures for germination. Germinationof S. dioica was greatly increased by fluctuating temperatures.Moderate increases occurred over a restricted temperature rangein A. graveolens cv. Giant Red and C. unguiculata was affectedin only minor ways. These results are discussed in relationto previous attempts to explain the mechanisms responsible forthe frequently beneficial effects of fluctuating temperatureson germination.  相似文献

Oxygen Diffusion Pathways and Nitrogen Fixation in Legume Root Nodules   总被引:1,自引:0,他引:1  
The theoretical analysis presented in this paper suggests thatthe nature of the diffusion pathway from the surface of a noduleto the infected zone depends on the morphology of the nodule;in particular the cross-sectional area of the intercellularspaces in the inner cortex. If the diffusion barrier containscontinuous pathways there will be no pressure difference betweenthe atmosphere and the infected zone. The conditions under whichthe intercellular spaces of the inner cortex could be air-filled,water-filled, or a combination of both are explored. An experimentto resolve this issue is suggested. Information obtained usingcryo-scanning electron microscopy and oxygen electrode datahave been used to illustrate various points. Expressions arederived for the diffusion resistance of the nodule and its componentparts. To provoke further discussion a simple mechanism forthe control of diffusion is described in general terms. We studied the influence of internal ammonium and nitrate contenton the temperature response of ammonium and nitrate uptake inboth chilling sensitive and chilling resistant tomatoes. Threetaxa were examined: Lycopersicon esculentum Mill. cv. T-5, achilling sensitive cultivar, Lycopersicon hirsutum Humb. andBompl. LA 1264, a wild, chilling sensitive accession from thelowlands of Ecuador, and Lycopersicon hirsutum LA1778, a chillingresistant accession from the highlands of Peru. Short exposures(4 h) of L. esculentum cv. T-5 to chilling temperatures irreversiblyinhibited ammonium absorption for at least 6 h. Nitrate absorptionin this taxon and ammonium and nitrate absorption in the L.hirsutum accessions recovered fully and immediately from suchexposures. The chilling resistant accession, L. hirsutum LA1778,showed a lower Q10 for ammonium absorption (1?54?0?10, mean?s.e.)than its chilling sensitive relatives, L. hirsutum LA1264 (2?37?0?35)and L. esculentum cv. T-5 (1?92?0?11). The temperature responseof nitrate absorption depended on internal nitrate status; plantsgrown at high levels of ammonium and nitrate (200 mmol m–3)showed higher Q10's for nitrate uptake (2?29?0?10) than thosedepleted of internal (1?86?0?12). Young leaf segments of Zea mays L. seedlings were cultured onMurashige and Skoog's basal nutrient medium supplemented with2 mg l–1 2, 4-D and sub-cultured on medium containing8 mg l–1 2,4-D. Two types of callus tissues appeared—embryogenicand non-embryogenic. The embryogenic callus tissue producednumerous somatic embryos which on transfer to media containinglow amounts of 2,4-D or ABA produced plantlets. Profiles of shortwave radiation, net radiation and temperaturewere measured in swards of three grasses of contrasting structureLolium perenne cv. S24, L. perenne cv. Reveille and Festucaarundinacea cv. S170. Measurements were also made of the reflectionof shortwave radiation, leaf water potential and stomatal resistance.Differences in canopy structure influenced the absorption andreflection of radiation by the varieties. The absorption ofnet radiation and its influence on air temperature inside thecanopy was shown to vary with canopy structure. Calculationsshowed that diurnal changes in the reflection and transmissionof light (400–700 nm) would have little effect on canopyphotosynthesis. No clear relationship between leaf extensionrate, temperature and leaf water potential could be established,although decreases in water potential did appear to reduce thepotential response of leaf extension rate to temperature.  相似文献

A series of 13-hydroxygibberellins, gibberellin A1 (GA1), GA19,GA20, GA44 and GA53, were identified by GC/MS (full scan) fromvegetative shoots of tall (cv. Kentucky Wonder) and dwarf (cv.Masterpiece) Phaseolus vulgaris L. It is suggested that GA1is active per se in the control of shoot elongation of P. vulgarisL., and that dwarfism in Masterpiece is not due to shortageof the active GA, but to its low ability to respond to the bioactiveGA. The effect of high pH on the morphology and anatomy of the rootsof lupin (Lupinus angustifolius L. cv. Yandee) and pea (Pisumsativum L. cv. Dundale) was examined in buffered solution. Themorphology and anatomy of lupin roots were markedly altered,and root growth was reduced by increasing solution pH from 5·2to 7·5, whereas pea roots were unaffected. In lupin roots,pH 7·5 caused disintegration of the root surface andimpaired root hair formation. Lupin roots grown at pH 7·5also had decreased cell lengths but increased cell diameterin both the epidermis and the cortex in comparison to rootsgrown at pH 5·2. High pH reduced cell volume greatlyin the epidermis, to a lesser extent in the outer cortex andnot at all in the inner cortex. The rates of canopy and individual leaf photosynthesis and 14Cdistribution for three temperate forage grasses Lolium perennecv. S24, L. perenne cv. Reveille and Festuc'a arundinacea cv.SI70 were determined in the field during a summer growth period.Canopy photosynthesis declined as the growth period progressed,reflecting a decline in the photosynthetic capacity of successiveyoungest fully expanded leaves. The decline in the maximum photosyntheticcapacity of the canopies was correlated with a decline in theirquantum efficiencies at low irradiance. Changes in canopy structureresulted in changes in canopy net photosynthesis and dark respiration.No clear relationships between changes in the environment andchanges in canopy net photosynthesis and dark respiration wereestablished. The relative distributions of 14C in the shootsof the varieties gave a good indication of the amount of drymatter per ground area in the varieties.  相似文献

The effect of tris, choline, and ethanolamine chlorides on theactivity of Mg2–dependent ATPase in membrane fractions(cell walls, mitochondria, and microsomes) of Zea mays L. (cv.Neve Yaar 22), Avena saliva L. (cv. Mulga), and Hordeum vulgareL. (cv. Omer) was compared with the effect of KC1 and NaCl.Considerable salt effects on apparent Mg2+ATPase activity werefound only at relatively high pH values (8.2) at which Mg2+.ATPaseactivity was low in the absence of monovalent cation salts.The Mg2+-dependent ATP hydrolysis by ATPases from all the membranefractions increased in the presence of at least one of the organiccations to the same extent as in the presence of KCI or NaCl.The monovalent organic cations are only very slowly absorbedby corn roots in comparison with K+ and Na+. It is concluded that monovalent salt effects on ATPase fromthese plant roots are not cation specific and not related tothe capability of root cells to absorb cations. Present evidencefor the existence of a cation-transport ATPase in plant tissueis critically reviewed.  相似文献
