The contribution of Chrysonilia sitophila in cork stopper manufacture was studied and a simulation of the industrial processing of cork stoppers was performed. Stoppers cut from slabs where mold development was inhibited were compared with others cut from slabs colonized by C. sitophila alone or with several molds, in terms of physical properties and chemical taints. C. sitophila does not produce 2,4,6-trichloroanisole, guaiacol, or 1-octene-3-ol on cork slabs incubated for 66 days. Since some chlorophenol-related compounds contaminate cork slabs during the production processes, metabolic tests were performed to investigate the capability of molds to produce 2,4,6-trichloroanisole by methylation of 2,4,6-trichlorophenol. Degradation of 2,4,6-trichlorophenol by C. sitophila resulted in a very high level of degradation without production of 2,4,6-trichloroanisole. C. sitophila restricted growth of other molds on maturing slabs for at least 30 days. These results show that C. sitophila can be exploited by industrial producers of cork stoppers since it is able to inhibit the development of other molds and it does not produce the compounds responsible for ‘cork-taint’, even in the presence of chlorophenols. The industrialization of traditional processes relies on the scientific ability to understand the empirical evidence associated with traditional knowledge. Cork manufacturing includes one operation known as stabilization, where humid cork slabs are extensively colonized by fungi. The implications of fungal growth on the chemical quality of cork through the analysis of putative fungal metabolites have already been investigated. However, the effect of fungal growth on the mechanical properties of cork remains unexplored. This study investigated the effect of cork colonization on the integrity of the cork cell walls and their mechanical performance. Fungal colonization of cork by Chrysonilia sitophila, Mucor plumbeus Penicillium glabrum, P. olsonii, and Trichoderma longibrachiatum was investigated by microscopy. Growth occurred primarily on the surface of the cork pieces, but mycelium extended deeper into the cork layers, mostly via lenticular channels and by hyphal penetration of the cork cell wall.In this first report on cork decay in which specific correlation between fungal colonization and mechanical proprieties of the cork has been investigated, all colonizing fungi except C. sitophila, reduced cork strength, markedly altering its viscoelastic behaviour and reducing its Young's modulus.  相似文献

A particular fungal population is present in the main stages of the manufacturing process of cork discs. Its diversity was studied using both dependent (isolation) and independent culture methods (denaturing gel gradient electrophoresis and cloning of the ITS1-5.8S-ITS2 region). The mycobiota in the samples taken in the stages before and after the first boiling seems to be distinct from the population in the subsequent manufacturing stages. Most isolated fungi belong to the genera Penicillium, Eurotium and Cladosporium. The presence of uncultivable fungi, Ascomycota and endophytes in raw cork was confirmed by sequencing. The samples taken after the first boiling contained uncultivable fungi, but in a few samples some isolated fungi were also detected. The main taxa present in the following stages were Chrysonilia sitophila, Penicillium glabrum and Penicillium spp. All applied techniques had complementary outcomes. The main factors driving the shift in cork fungal colonization seem to be the high levels of humidity and temperature to which the slabs are subjected during the boiling process.  相似文献

The cork stopper manufacturing process includes an operation, known as stabilisation, by which humid cork slabs are extensively colonised by fungi. The effects of fungal growth on cork are not completely understood although they are considered to be involved in the so-called "cork taint" of wine. It is essential to (a) identify environmental constraints which define the appearance of the colonising fungal species and (b) trace their origin to the forest and/or the manufacturing space. The present article correlates two sets of data, from consecutive years and the same season, of systematic sampling of two manufacturing units, located in the North and South of Portugal. Chrysonilia sitophila dominance was confirmed, followed by a high diversity of Penicillium species. Penicillium glabrum, which was found in all samples, was the most frequently isolated species. P. glabrum intra-species variability was investigated using DNA fingerprinting techniques revealing highly discriminative polymorphic markers in the genome. Cluster analysis of P. glabrum data was discussed in relation to the geographical location of strains, and results suggest that P. glabrum arise from predominantly the manufacturing space, although cork specific fungi can contribute.  相似文献

A two-step protocol was used for the identification of 52 yeasts isolated from bark of cork oak at initial stages of the manufacturing process of cork stoppers. The first step in the identification was the separation of the isolates into groups by their physiological properties and RFLPs of the ITS-5.8S rRNA gene. The second step was the sequencing of the D1/D2 domains of the 26S rRNA gene of selected isolates representing the different groups. The results revealed a predominance of basidiomycetous yeasts (11 species), while only two species represented the ascomycetous yeasts. Among the basidiomycetous yeasts, members representing the species Rhodosporidium kratochvilovae and Rhodotorula nothofagi, that have been previously isolated from plant material, were the most abundant. Yeasts pertaining to the species Debaryomyces hansenii var. fabryii, Rhodotorula mucilaginosa and Trichosporon mucoides were isolated in small numbers.  相似文献

The compounds responsible for the so-called 'cork taint' include, among others, some microbial metabolites which can be produced by the microbial population colonizing the unprocessed cork and stoppers. This study was intended to obtain information on the mycobiota associated with Portuguese cork throughout the manufacturing process of stoppers. Samples of barks and stoppers of both 'normal' and 'green' cork were examined. Moulds were isolated from 'normal' and 'green' cork throughout the entire cork stopper manufacturing process. Yeasts were rarely detected in the corks. Fungal contamination was not detected in finished stoppers from the company under study.  相似文献

The production of enzymes by micro-organisms which are found on vegetal substrates is important due to their ability to decompose cellulose, lignin and other components, which guarantee the integrity of the vegetal cell. The objective of this study was to determine the enzymatic activity of filamentous fungi, yeasts and bacteria, isolated from natural cork stoppers for bottles of still and sparkling wines. Suspensions of fungal conidia, yeasts and bacterial cells of micro-organisms were established in concentrations of 10(6) CFU/ml. The enzymatic activity of these micro-organisms was evaluated by means of the API ZYM system, with which it was possible to determine and semi-quantify nineteen enzymatic activities simultaneously. The enzymes produced by all of the species were esterase (C1), esterase lipase and naphthol-AS-BI-phosphohydrolase. The micro-organisms with the greatest enzymatic activity were Monilia sitophila, Alternaria alternata, Aspergillus niger and Aeromonas sp.  相似文献

AIMS: The aim of this study was to determine the survival and growth characteristics of Salmonella enterica in sound and chill-injured tomatoes as influenced by co-infection with proteolytic moulds. METHODS AND RESULTS: Sound (not chill injured) raw, ripe tomatoes (Lycopersicon esculentum Mill. 'Roma') were inoculated with a five-serotype mixture of S. enterica and/or Alternata alternata (two strains), Cladosporium herbarum and C. cladosporioides. Simultaneous and delayed (3 days) inoculation of tomatoes with Salmonella and each mould was studied. Growth of moulds in sound tomatoes stored at 15 and 25 degrees C for up to 10 days was accompanied by increased pH of radial pericarp tissue (pulp), which enhanced the growth of Salmonella. Growth of moulds and Salmonella at 25 degrees C was enhanced in chill-injured tomatoes compared with sound tomatoes. CONCLUSIONS: Growth of proteolytic moulds in tomatoes stored at conditions simulating those commonly used in commercial postharvest storage and handling promotes the growth of Salmonella that may be an incidental contaminant. SIGNIFICANCE AND IMPACT OF THE STUDY: Discarding tomatoes that are infected by moulds is important in handling and minimal processing practices designed to minimize the risk of human salmonellosis.  相似文献

The presence of guaiacol in cork stoppers is responsible for some cases of cork taint causing unpleasant alterations to wine. We have performed a characterization of the cork-associated microbiota by isolating 55 different microorganisms: eight yeast, 14 filamentous fungi or molds, 13 actinomycetes and 20 non-filamentous bacteria. A screening for degradation of vanillic acid and guaiacol production showed that none of the filamentous fungi could achieve any of these processes. By contrast, five of the eight yeast strains isolated were able to degrade vanillic acid, although it was not converted to guaiacol. Guaiacol production was only detected in four bacterial strains: one isolate of Bacillus subtilis and three actinomycetes, Streptomyces sp. A3, Streptomyces sp. A5 and Streptomyces sp. A13, were able to accumulate this compound in both liquid media and cultures over cork. These results suggest that guaiacol-mediated cork taint should be attributed to the degradative action of vanillic acid by bacterial strains growing on cork.  相似文献

AIMS: The aims of the present study were to: (i) analyse a group of antimicrobial agents and to select the most active against test microbial strains; (ii) test the effect of the antimicrobial treatment on air filters in order to reduce microbial colonization. METHODS AND RESULTS: Different kinds of antimicrobial agents were analysed to assess their compatibility with the production process of air filter media. The minimal inhibitory concentration for each antimicrobial agent was determined against a defined list of microbial strains, and an antimicrobial activity assay of filter prototypes was developed to determine the most active agent among the compatible antimicrobials. Then, the most active was chosen and added directly to the filter during the production process. The microbial colonization of treated and untreated filter media was assessed at different working times for different incubation times by stereomicroscope and scanning electron microscope analysis. Some of the antimicrobial agents analysed were more active against microbial test strains and compatible with the production process of the filter media. Filter sections analysis of treated filter media showed a significantly lower microbial colonization than those untreated, a reduction of species both in density and varieties and of the presence of bacteria and fungal hyphae with reproductive structures. CONCLUSIONS: This study demonstrated the ability of antimicrobial treatments to inhibit the growth of micro-organisms in filter media and subsequently to increase indoor air quality (IAQ), highlighting the value of adding antimicrobials to filter media. SIGNIFICANCE AND IMPACT OF THE STUDY: To make a contribution to solving the problem of microbial contamination of air filters, by demonstrating the efficacy of incorporating antimicrobial agents in the filter media to improve IAQ and health.  相似文献

Drinking water and its delivery system are potential sources of variation in animal research. Concern arose that rubber stoppers used to cork water bottles might be a source of some nutritionally required minerals which could leach into drinking water. Six types of stoppers, each having different compositions, were cleaned with stainless-steel sipper tubes inserted into them and attached to polypropylene bottles filled with either deionized water (pH 4.5) or acidified-deionized water (pH 2.5). After six days of contact, water levels of copper, magnesium, iron, manganese, zinc, chromium, and selenium were determined by atomic absorption spectroscopy. Additionally, three of the stopper types were analyzed for mineral content. Minerals were present in both stoppers and drinking water. Acidified-deionized water generally leached minerals from the stoppers than did deionized water. The black stopper which is commonly used in animal facilities contained and leached measurable levels of some minerals, but it still can be recommended for typical animal husbandry uses, although other types of stoppers would be more suitable for specific nutritional and toxicologic studies.  相似文献

为满足三峡库区橙汁加工对甜橙品种熟期配套和品质调控的需要,通过对引进的特早熟品种早金(Early Gold)、早熟夏橙品种蜜奈(Midknight)和晚熟夏橙品种奥林达(Olinda),以及从本地主栽甜橙品种锦橙中选出的中晚熟品种晚熟锦橙的品质和熟期分析,初步证明这几个品种与本地主栽品种锦橙配套,可以实现从11月至次年6月的8个月成熟期配套,基本满足橙汁加工厂周年生产的需要。筛选出的长叶橙、宜圆3号等国内品种和从国外引进的埃及糖橙、早金等品种,其果汁具有较高的含糖量和固酸比,可用于调配降低三峡库区橙汁的酸度,改善果汁风味。  相似文献

No significant differences (P<0.05 occurred in the frequency of isolation from soil of four common moulds – Rhizopus stolonifer, Monilia sitophila, Aspergillus niger and Penicillium sp., on malt extract agar (MEA) and gelled gari (carbohydrate gel). Growth of the moulds on slide microcultures of potato dextrose agar (PDA) and gari showed typical diagnostic features of the organisms, which were often clearer on gari. It is concluded that gari, which is much cheaper and far more readily available in Nigeria than MEA or PDA, is an effective alternative to the two standard mycological media.  相似文献

BACKGROUND: Because different spectral sensitivities of human eye and image sensor lead to different perception of fluorescence signals, data generation is an important step in image analysis, because following work steps depend on it. METHODS: We developed a method to determine image parameters allowing an objective appraisal of quality of image data as well as a separation of object and background. RESULTS: Calculated parameters can be used for an automated adjustment of camera parameters in image analysis systems. DISCUSSION: Our approach for objective adjusted data generation achieves an improvement of analysis quality.  相似文献

AIM: To use ergosterol assay as a rapid tool to assess the extent of fungal contamination in grains and feeds. METHODS AND RESULTS: Ergosterol was extracted from moulds and quantified using a modified method based on Seitz et al. (1977). A good correlation coefficient of 0.9998 was obtained for ergosterol standards and a strong correlation (R(2) = 0 x 9645) was established between the ergosterol content and the growth of Aspergillus niger ATCC 24126. This shows that this assay can be used to estimate fungal growth. The ergosterol contents and amount of carbon dioxide produced in both Control and Myco CURB (brand) liquid-treated corns were measured over 36 days. Ergosterol contents of pelleted pig feeds were also determined. In both experiments, the levels of ergosterol in the control samples were significantly higher than those of the mould-inhibitor-treated samples. CONCLUSIONS: A strong positive correlation (Spearman, Rs = 0 x 7241) was observed between the ergosterol content and the degree of fungal growth. Hence, ergosterol assay can be used as a rapid tool to assess the microbiological status of grains and feeds. SIGNIFICANCE AND IMPACT OF THE STUDY: This study affirms that ergosterol assay is a rapid and accurate tool that can be used for the assessment of the feed quality.  相似文献

Nitrogen-doped carbon nanotubes consist of many cup-shaped graphitic compartments termed as nitrogen-doped carbon nanotube cups (NCNCs). These as-synthesized graphitic nanocups from chemical vapor deposition (CVD) method were stacked in a head-to-tail fashion held only through noncovalent interactions. Individual NCNCs can be isolated out of their stacking structure through a series of chemical and physical separation processes. First, as-synthesized NCNCs were oxidized in a mixture of strong acids to introduce oxygen-containing defects on the graphitic walls. The oxidized NCNCs were then processed using high-intensity probe-tip sonication which effectively separated the stacked NCNCs into individual graphitic nanocups. Owing to their abundant oxygen and nitrogen surface functionalities, the resulted individual NCNCs are highly hydrophilic and can be effectively functionalized with gold nanoparticles (GNPs), which preferentially fit in the opening of the cups as cork stoppers. These graphitic nanocups corked with GNPs may find promising applications as nanoscale containers and drug carriers.  相似文献

S. YASSIN AND A. WHEALS. 1992. Neurospora species may occur as spoilage organisms in bakeries and bakery products. A survey of two bakeries over a 4 month period produced 315 individually isolated strains which were characterized with respect to six features: conidial colour, protoperitheciality, mating type and percentage dark-coloured (fertile) ascospores in crosses with type specimens of Neurospora crassa, N. sitophila and N. intermedia. We concluded that (i) N: crassa of both mating types was by far the most abundant but N. sitophila and N. intermedia were also represented; (ii) conidial colour was not a good species criterion; (iii) the percentage of fertile ascospores produced in crosses with several type specimens provided a reliable and simple species indicator; (iv) identification of individuals based on these six characteristics suggested that a large number of genetically different organisms were present in the collection; and (v) bakery infection was not due to endemic contamination in the bakery or in raw materials but from repeated external infection, perhaps on contaminated returned goods.  相似文献

Background and Aims

Marginal populations of widely distributed species can be of high conservation interest when they hold a significant or unique portion of the genetic diversity of the species. However, such genetic information is frequently lacking. Here the relevance of genetic surveys to develop efficient conservation strategies for such populations is illustrated using cork oak (Quercus suber) from Minorca (Balearic Islands, Spain) as a case study. Cork oak is highly endangered on the island, where no more than 67 individuals live in small, isolated stands in siliceous sites. As a consequence, it was recently granted protected status.

Methods

Two Bayesian clustering approaches were used to analyse the genetic structure of the Minorcan population, on the basis of nuclear microsatellite data. The different groups within the island were also compared with additional island and continental populations surrounding Minorca.

Key Results

Very high genetic diversity was found, with values comparable with those observed in continental parts of the species'' range. Furthermore, the Minorcan oak stands were highly differentiated from one another and were genetically related to different continental populations of France and Spain.

Conclusions

The high levels of genetic diversity and inter-stands differentiation make Minorcan cork oak eligible for specific conservation efforts. The relationship of Minorcan stands to different continental populations in France and Spain probably reflects multiple colonization events. However, discrepancy between chloroplast DNA- and nuclear DNA-based groups does not support a simple scenario of recent introduction. Gene exchanges between neighbouring cork oak stands and with holm oak have created specific and exceptional genetic combinations. They also constitute a wide range of potential genetic resources for research on adaptation to new environmental conditions. Conservation guidelines that take into account these findings are provided.  相似文献

The development of sciarids was reduced in old casing and in casing to which a commercial composting additive was added in large amounts. In laboratory tests, adding water, soya bean, cotton seed or oatmeal to finished mushroom composts reduced the mean development time of the larvae of Lycoriella auripila. Soya bean meal significantly increased the number of adult sciarids which matured. While the presence of mushroom mycelium in large quantities inhibited the development of sciarids, smaller amounts increased either the number of larvae maturing or their rate of development in different tests. Larvae were seen to feed on mycelium. The addition of sucrose solution either reduced or prolonged development time in different tests. Nitrogenous additives encouraged the growth of ‘weed moulds’. Both these and the number of sciarids which developed were reduced by adding a solution of calcium nitrate. The results suggested that more selective composts, favouring rapid mycelial colonization, would provide ‘cultural’ control of mushroom sciarids.  相似文献

Aims:  To develop mathematical models for mycelium growth and ergosterol formation in conditions of periodic stationary culture; to verify possibilities of applying a model describing the relationship between ergosterol content and mycelium quantity.
Methods and Results:  The mould growth and ergosterol formation models covering all phases of mould growth are described using a modified logistic equation with the addition of an exponential function. The correlation between ergosterol and mycelium biomass depended on the growth phase of mould. Meaningful relation was obtained for initial two phases, when both parameters were growing equally. The quadratic function for estimation of the biomass based on ergosterol content was formulated. The error resulting from the application of this function in initial phases of moulds growth was small at 5–7%, in the following phases it was at 11–31%.
Conclusions:  Mycelium biomass can be precisely determined basing on the ergosterol content, when we know the moulds growth phase. In natural environments, without the information about growth phases, it will be possible, but with the higher error.
Significance and Impact of the Study:  Presented model based on the ergosterol content making possible to estimate the quantity of mycelium in moulds cultures and natural environments.  相似文献
