Dogfish peripheral blood leucocytes were examined by electron microscopy after the injection of colloidal carbon. The cells were classified as lymphocytes, plasma cells, monocytes, thrombocytes and granulocytes. The granulocytes were further classified into four types according to the structure of their granules. Monocytes, thrombocytes and two types of the granulocytic cells were phagocytic.  相似文献

The leucocytes of the peripheral blood of the elasmobranch Scyliorhinus canicula L. were examined by light, transmission and scanning electron microscopy and histochemistry. Seven distinct leucocytes were identified including lymphocytes, thrombocytes, monocytes and granulocytes. The intracellular location of the binding site of antibody against purified myosin prepared from equine leucocytes was investigated in neutrophils and lymphocytes by electron microscopy using peroxidase-labelled antibody method. The myosin extracted from equine leucocytes could bind skeletal muscle F-actin and the formed complex showed the biophysical and biochemical properties and electron microscopic appearance of actomyosin. On immunodiffusion, the leucocyte myosin formed a single precipitin line with its antibody prepared in rabbits. The antibody also formed single precipitin lines with myosins from lymphocytes and thrombocytes, fusing with each other. The antibody against the leucocyte myosin did not react with myosins from skeletal or arterial smooth muscle. The specificity of the antibody was further established by determination of K+-EDTA-activated ATPase activity remained in the supernate of antigen-antibody mixture. Under electron microscope, the intracellular immunoreactive products of peroxidase labelled antibody were found in cytoplasm of neutrophils and lymphocytes incubated with antibody against leucocyte myosin, but not in neutrophils or lymphocytes treated with IgG from normal rabbits.  相似文献

The cell composition of the spleen of the dogfish, Scyliorhinus canicula L. was investigated by electron microscopy. It comprised areas of red and white blood cells. The red cells were observed in various stages of development and the spleen is probably the main erythropoietic organ, it is likely that thrombocytes are also produced in the spleen. The presence of plasma cells, and the appearance of rosette-like contacts between lymphocytes and phagocytic cells, probably macrophages, indicated that immunological processes were taking place. Destruction of effete blood cells, primarily erythrocytes, was indicated by the presence of macrophages containing residues of ingested blood cells.  相似文献

Optimum procedures for fish handling and sample processing for use when employing haematological parameters as health indicators in turbot, Psetta maxima (L.), have been established. We found thrombocytes to be the most abundant blood cell, representing approximately 52% of circulating leucocytes (lymphocytes, 40.8%; granulocytes, 5.6%; monocytes, 1.6%; total number of leucocytes=1.3 × 105 ml−1; packed cell volume=22.7%). The light- and electron-microscopical characteristics of these cell types are described, together with their cytochemical properties using Sudan Black B, Periodic Acid Schiff, Non-specific Esterase, and Acid and Alkaline Phosphatase. Turbot thrombocytes showed a high degree of shape alterations when observed in live preparations using phase contrast microscopy, while ultrastructural observations following the in vitro uptake of carbon particles supported an active process of phagocytosis by the thrombocytes, rather than passive entrapment. The lymphocytes of turbot are structurally similar to mammalian lymphocytes with the highest nuclear:cytoplasmic ratio of all the leucocytes observed. Small lymphocytes predominated, large lymphocytes forming less than 1% of the total white blood cell population. The most frequent granulocyte type was a neutrophil-like cell with an eccentric nucleus, only rarely seen in segmented form. In vitro uptake of carbon particles by granulocytes was not observed under the conditions of the experiment, although turbot granulocytes are capable of phagocytosis under different circumstances. These are discussed, along with other physiological and technical factors which can influence the blood parameter findings in fish.  相似文献

The ultrastructure of pike peripheral blood cells, lymphocytes, thrombocytes, granulocytes, and monocytes is described. At present there are no reliable criteria for differentiating between round thrombocytes and small lymphocytes of fish on a routine basis. At the ultrastructural level thrombocytes could be clearly differentiated from lymphocytes by cytoplasmic canals and vesicles, marginal microtubules, and large glycogen deposits. Electron microscopic identification of thrombocytes was confirmed by examining the ultrastructural features of a purified thrombocyte fraction. In addition, a preliminary investigation of the structure of the haemopoietic cells in the thymus, anterior kidney, and spleen was carried out.  相似文献

Summary Chicken thrombocytes were enriched for immunization by utilizing their strong capacity to adhere to plastic surfaces. The produced rabbit anti-thrombocyte serum ATS 3 reacted by means of the unlabeled antibody enzyme method (PAP) specifically with thrombocytes of fixed chicken-blood smears, but not with lymphocytes or other blood cells. When ATS 3 (substrate diaminobencidinetetrahydrochloride = DAB) and a 11 mixture of an anti-bursa serum and anti-thymus serum (ABS/ATS; substrate 4-chloro-1-naphthol = 4-Cl-1-N) were used simultaneously, thrombocytes revealed the brown color typical for DAB, whereas lymphocytes showed the blue stain of 4-Cl-1-N. The finding of a thrombocyte surface antigen not shared by lymphocytes is regarded as a further proof of the diversity of both cell systems, i.e., for the existence of a genuine thrombocyte system in chickens.  相似文献

Peripheral blood leucocytes were examined for their fine structure and their in vivo response to colloidal carbon particles and bacteria, Aeromonas salmonicida . Four main types of leucocyte were identified: lymphocytes, thrombocytes, monocytes and granulocytes. Previously, two types of granulocyte had been identified but at the electron microscope level a third type could be distinguished in the peripheral blood. This investigation sought to determine whether splenic lymphocytes obtained from Balb/C mice exposed to aflatoxin B1 (AFB1) showed any ultrastructural changes which could account for the immunodysfunction attributable to aflatoxins. Lymphocytes obtained from Balb/C mice administered aflatoxin B1 in olive oil daily for three weeks were studied using both transmission and scanning electron microscopy. The lymphocytes demonstrated ultrastructural changes primarily in the mitochondria where marked internal dissociation of the cristae was revealed by transmission electron microscopy. All other cellular organelles were unaffected. No significant alterations in external structure were observed under scanning electron microscopy. The findings of this study indicate that AFB1 administration does not affect the surface topography of lymphocytes, but AFB1, by causing extensive mitochondrial damage, may affect the way in which these cells function. The morphology of blood cells in the kidney and spleen of Xiphophorus helleri was characterized by electron microscopy. Erythrocytes, thrombocytes, lymphocytes, granulocytes and monocytes/macrophages were identified. Thrombocytes were elongate and contained bundles of microtubules and a canalicular system. Lymphocytes were heterogeneous in morphology. Granulocytes were divided into two types, based on the light microscopic results, which were extended by the present electron microscopic study. Neutrophils and eosinophils differed in abundance, cell shape, morphology of the nucleus and granula. Neutrophils displayed a spherical to three-lobulated nucleus and different types of granula. Several intermediate forms of granula were observed. The complement of granules displayed a different composition in the cells. These findings suggest a maturation process of a single type of granulum rather than several types. Eosinophils also contain different types of granula, described as G1–G4. The cell shape was variable and nuclei were small and eccentric. Monocytes and macrophages frequently showed autophagocytotic figures. A peroxidase reaction was observed only in the granula of neutrophils.  相似文献

Recent scanning electron microscopic studies on isolated follicular dendritic cells (FDC) showed that dendrites of certain FDC were "beaded," i.e., consisting of a series of interconnected immune complex coated bodies (termed "iccosomes," measuring 0.3 to 0.7 micron diameter). In vitro these iccosomes detach from one another with ease. The major objectives herein were to establish whether these structures can be detected in sections and whether iccosomes serve to disseminate antigen in vivo. Beginning at day 1, the time point used for isolating beaded FDC, the popliteal lymph nodes of immune C3H mice were studied with light and transmission electron microscopy for 2 wk (i.e., at days 1, 3, 5, 8, and 14) after hind footpad injection of the histochemically detectable antigen, horseradish peroxidase (HRP). Iccosomes (0.25 to 0.38 micron diameter), contoured by a peroxidase (PO)-positive coat of HRP-anti-HRP complexes, were first detected by transmission electron microscopy at day 1 adjacent to cell bodies of certain FDC. Within their limiting membrane they contained flocculent material that was PO positive. At day 3 by light microscopy, germinal centers were seen enlarged and the antigen-retaining reticulum, composed of antigen-bearing FDC, appeared diffuse. This coincided with the transmission electron microscopic visualization of a dispersed state of iccosomes among the follicular lymphocytes. At that time iccosomes were seen attached to the surface of lymphocytes via PO-positive immune complexes and were surrounded by microvillous processes of these cells. Germinal center lymphocytes and tingible body macrophages both responded to contact with iccosomes by endocytosis. Antigen-containing tingible body macrophage were most conspicuous by light microscopy at day 5, when transmission electron microscopy showed that the majority of germinal center lymphocytes contained endocytosed HRP in secondary lysosome-like granules associated with the Golgi apparatus. The number of dispersed iccosomes was markedly reduced by day 5. In controls injected with HSA, a PO-negative antigen, lymphocytes and tingible body macrophages were PO-negative. The presence of antigen in both cell types was confirmed through the use of a gold-conjugated antigen (goat IgG). Simultaneous immunoperoxidase labeling of the same tissues with anti-Ia showed the gold conjugate containing B cells to be Ia+. Antigen-positive B cells and tingible body macrophages were greatly reduced in numbers by day 14, suggesting the intracellular fragmentation of the antigen.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

The change of the venular permeability and endothelial structure was studied with the aid of fluorometry and electron microscopy after application of H2O2. Transmural transfer rate of FITC-albumin++ and permeable fraction of the vessel walls were increased. There were formed the "leaks" and transendothelial canals in the venular endothelium. Numerous local membrane injuries of the endotheliocytes and pericytes, degranulation of the mast cells and destruction of thrombocytes into the vessel lumen were detected.  相似文献

测定军曹鱼的血液指标,红细胞密度为2.69±0.86×106个/mm3,白细胞密度为1.50±0.09×104个/mm3;血 红蛋白含量为7.42±0.22g/L,红细胞渗透脆性为0.43±0.07g%,红细胞沉降速率为1.18±0.46mm/h。观察军曹鱼 外周血液涂片,可区分出红细胞、血栓细胞、淋巴细胞、嗜中性粒细胞和单核细胞,但没有发现嗜酸性粒细胞和嗜碱 性粒细胞。在外周血液涂片观察中还发现了较多未成熟的红细胞和嗜中性粒细胞以及少量正在分裂的红细胞。 血栓细胞、淋巴细胞、嗜中性粒细胞和单核细胞在白细胞中所占比例分别为61.20±6.30%,16.60±3.28%,16.00± 3.61%和6.20±3.90%。对肝脏、脾脏、头肾和中肾等四种造血组织进行了涂片观察,军曹鱼的血细胞主要在头肾 和肾脏产生;脾脏是军曹鱼粒细胞发生的另一个场所,而肝脏也具有产生粒细胞和淋巴细胞的作用。    相似文献

Summary Ultrastructures of human and rabbit thrombocytes reveal specific subcellular organelles within these elements. Serotonin granules are demonstrated containing extremely electron opaque material in vesicles with an average diameter of 1,700 Å and a considerable number of large dense bodies (average size 4,000 Å in diameter) is seen. The latter are less electron dense as compared to the serotonin granules. The appearance of serotonin granules in the human thrombocyte is rare, while rabbit platelets show a higher number of these granular vesicles.Acid phosphatase activity in the large dense bodies of human and rabbit platelets has been demonstrated by means of electron microscopy. Present results together with currently available biochemical information are briefly discussed in relation to the lysosomal activity within the thrombocytes.  相似文献

The morphological characteristics of the peripheral leucocytes of the cichlid O. mossambicus were studied using light microscopy. Cytochemical procedures and phagocytic studies were employed. Four main types of leucocytes could be recognized: lymphocytes, thrombocytes, monocytes and granulocytes. Two distinct types of granulocyte were identified. 用鳗弧菌(Vibrio anguillarum)和嗜水气单胞菌(Aeromonas hydrophila)分别感染海鳗(Muraenesoxcinereus),48 h后取其外周血液,瑞-吉(Wright-Giemsa)氏染色,光镜下观察,同时对其外周血液中的免疫细胞数量变化进行测定。鳗弧菌和嗜水气单胞菌感染组的单核细胞的伪足样突起与对照组比较明显增多,胞质内空泡增多、变大,数量明显增多,差异极显著;嗜中性粒细胞核多呈长杆状,数量与对照组比较明显增多,差异极显著;淋巴细胞伪足样突起明显,中、大型淋巴细胞较多,数量与对照组相比明显减少,差异极显著;血栓细胞的数量明显下降,差异极显著。  相似文献

Channel catfish were acclimated in vivo to 12, 17, 22 or 27 degrees C and their peripheral blood erythrocytes, thrombocytes, T lymphocytes and B lymphocytes assayed for cellular fatty acid composition. Excepting cells from 12 degrees C acclimated fish, all cells responded to acclimation to lower temperatures by exhibiting increased levels of phospholipid unsaturated fatty acids. Although temperature independent differences were observed between erythrocytes, thrombocytes and lymphocytes, no differences between T lymphocytes and B lymphocytes were seen.  相似文献

By the aid of histological special stainings, cytochemical proofs, phase contrast and electron microscopical methods the thrombocytes (spindle cells) of the European ell (Anguilla anguilla) and their stages of development were described as an independent series of cells. After an artificial blood loss thrombocytoblasts, prothrombocytes and mature thrombocytes could distinctly be demonstrates. Besides the kidney the spleen was found to be the main place of the thrombopoiesis. Phase contrast investigations provided evidence for the relatively strong locomotion of these cells. By means of cytochemical proofs a further differentiation of the thrombocytes from other white blood cells can be made. Electron microscopical investigations of the thrombocytes yielded a great similarity to the thrombocytes of the amphibians. The endoplasmatic ring and particular inclusions could be shown to be characteritstic features of the fine structure.  相似文献

Histological and ultrastructural studies of the thymus of the dogfish, Scyliorhinus canicula L., revealed paired lobular masses above the first two gill arches of embryonic and newly hatched dogfish, involuting at approximately 3 weeks. It did not have defined cortex and medullary regions and ultrastructural investigations showed the tissue to be composed mainly of a range of different sized lymphocytes within a connective tissue capsule and reticular epithelial cell framework. Lymphoblasts often in mitosis were also present being more electron lucent and larger than lymphocytes. Macrophages contained cellular debris and frequently whole small lymphocytes. A cell with cytoplasmic granules was occasionally observed.  相似文献

长薄鳅外周血细胞可分为红细胞、中性粒细胞、单核细胞、淋巴细胞和血栓细胞.在数量上,中性粒细胞、单核细胞、淋巴细胞和血栓细胞占白细胞总数的百分比分别是17.06%、5.83%、28.16%和48.94%.细胞化学染色显示所有白细胞均含有糖原物质,所有红细胞均不含酸性磷酸酶,中性粒细胞、单核细胞、淋巴细胞和血栓细胞均含有酸性磷酸酶.非特异件酯酶染色显示单核细胞呈阳性反应,中性粒细胞、淋巴细胞和血栓细胞均为部分呈阳性反应.所有细胞的碱性磷酸酶、过氧化物酶、苏丹黑显色反应均呈阴性.  相似文献
