Bacteriosomes in axenic plants: endophytes as stable endosymbionts   总被引:1,自引:0,他引:1  
Observations of cells of axenic peach palm (Bactris gasipaes) microplants by light microscopy revealed movements of small particles within the cells. The phenomenon was characterized initially as Brownian movement, but electron microscopy revealed the presence of an intracellular bacterial community in these plants. Microscopy observations revealed the particular shapes of bacterial cells colonizing inner tissues of analyzed plants. Applying a molecular characterization by polymerase chain reaction and denaturing gradient gel electrophoresis, it was revealed the existence of bacterial rRNA within the plants. Sequencing of the rRNA identified three different phylogenetic groups; two bands had a high degree of similarity to sequences from Moraxella sp. and Brevibacillus sp., and a third sequence was similar to a non-cultivated cyanobacterium. The presence of those endosymbionts, called bacteriosomes, in axenic peach palm microplants raises the question of whether these stable endosymbionts were acquired in the process of evolution and how could they benefit the process of plants micropropagation.  相似文献

Summary Cell plate formation inChara zeylanica was compared with recent models of cytokinesis in higher plants in order to gain insight into the evolutionary origin of plant cytokinetic processes. Transmission electron microscopy (TEM) reveals that while cytokinesis inC. zeylanica bears many features in common with that in higher plants, there are significant differences. Unlike that in higher plants, cytokinesis inC. zeylanica begins with a congregation of smooth membrane tubules that are closely associated with endoplasmic reticulum (ER) and Golgi membranes. Mitochondria and other organelles excluded by the phragmoplast in higher plants are present as well. Unlike in higher plants, phragmoplast microtubules persist throughout cytokinesis inC. zeylanica, and the cell plate generally forms across the whole cell at once, though development is patchy, due to small regions developing at different rates; the ends of the plate form last. By identifying aspects of cytokinesis that are different inC. zeylanica and plants, our study indicates which cytokinetic features are more likely to be derived, and which are more likely to be ancestral. In addition, we demonstrated that all nodal cells ofC. zeylanica are interconnected via plasmodesmata, lending support to the idea that, whileChara spp. are generally considered to be filamentous organisms, nodal regions may be thought of as meristemlike tissues.Abbreviations HPF high-pressure freezing - KFe potassium ferricyanide - SCF stepwise chemical fixation - TEM transmission electron microscopy  相似文献

Particles of cassava latent virus (CLV) were purified by a method that yielded up to 3 mg per 100 g of systemically infected Nicotiana benthamiana leaf. Specific antiserum was prepared and used for enzyme-linked immunosorbent assay (ELISA), which detected purified virus at 5 ng/ml. As estimated by ELISA, CLV antigen reached a greater concentration in leaves of N. benthamiana plants kept at 20–25 °C than in those at 15 °C or 30 °C. CLV was also detected in leaf extracts of naturally infected cassava plants kept at 25 C but its concentration was only 1–7% of that in comparable extracts from N. benthamiana. Staining sections of N. benthamiana leaves with fluorescent antibody indicated that CLV particle antigen accumulates in the nuclei of many phloem cells and of some cells in other tissues. In tests on mosaic-affected cassava plants of Angolan origin, three plants were found in which CLV could not be detected by either ELISA or immunosorbent electron microscopy, or by transmission to indicator plants. This suggests that the mosaic symptoms were caused by a pathogen other than CLV, but no such agent was detected by electron microscopy of leaf extracts. Three kinds of serological test indicated that CLV is related to bean golden mosaic virus. Evidence was also obtained of a distant relationship to beet curly top virus but none was detected to four other geminiviruses.  相似文献

Summary Scanning electron microscopy, light microscopy, and gravimetric analysis was used to evaluate stomatal function, epicuticular wax, and the stem-root transition region of grape (Vitis sp. ‘Valiant’) plantlets grownin vitro, polyethylene glycoltreatedin vitro, and greenhouse-grown plants. Scanning electron microscopic studies of leaf surfaces ofin vitro-grown plants showed widely open stomata as compared to leaf stomata of polyethylene glycol-treatedin vitro-cultured and greenhouse-grown plants. Ultrastructurally, leaf epicuticular wax ofin vitro plants was less dense than in their polyethylene-treated and greenhouse counterparts. Quantitatively,in vitro-grown plants had reduced epicuticular was as compared to polyethylene glycol-treated and greenhouse-grown plants. Light microscopic studies showed no obvious differences in the vascular connections in the stem-root transition region ofin vitro-cultured, polyethylene glycol-treatedin vitro-cultured, and greenhouse-grown plants. It is therefore likely that the rapid wilting and desiccation observed after transplantingin vitro grape plantlets is due to their defective stomatal function and reduced epicuticular wax and may not be due to poor water transport associated with vascular connection.  相似文献

Transgenic Arabidopsis thaliana plants expressing cell antisense exhibit reduced levels of cell mRNA and protein compared with wild-type plants. The former display significant alterations in their phenotype. cell antisense plants have shorter stems and roots and are mechanically weaker than their wild-type counterparts. In cell antisense plants, the cell wall structure is markedly disrupted: both fluorescent confocal microscopy and scanning electron microscopy revealed `wrinkled' cell walls, thus indicating that CEL1 plays an important role in cell wall relaxation during cell growth and expansion. In cell antisense plants, the number of xylem elements per bundle is smaller than in the wild-type. In addition, both xylem elements and interfascicular fibers are significantly less lignified in the former. It is suggested that in A. thaliana, abnormal cell wall deposition affected by CEL1 depletion is associated not only with cell growth, but also with the differentiation process in the vascular and supporting tissues.Equal contributors  相似文献

In April 2022, Aristolochia plants with symptoms of mosaic were observed in a garden at Jardim Botânico Plantarum, Nova Odessa, São Paulo State, Brazil. Potyviridae-like particles were observed by transmission electron microscopy in leaf extracts. Total RNA extracted from symptomatic plants used in RT-PCR with universal and BCMV-specific primers detected the potyvirus bean common mosaic virus (BCMV). The cucumovirus cucumber mosaic virus (CMV) was identified only in Aristolochia littoralis plants that tested negative by RT-PCR for BCMV. Phylogenetic analysis grouped samples of Aristolochia in a different clade among samples of Phaseolus vulgaris. Phylogenetic analysis indicated that the CMV isolate from Aristolochia belongs to the CMV group IA. BCMV was mechanically transmitted to healthy plants of A. fimbriata, Chenopodium quinoa, P. vulgaris cv. Jalo and Macroptilium lathyroides. CMV was mechanically transmitted to plants of A. fimbriata and C. quinoa. The BCMV and CMV were aphid transmitted only by Aphis gossypii to Aristolochia plants. This is the first report of BCMV and CMV infecting Aristolochia plants in Brazil.  相似文献

As part of efforts to identify the causal agent of the rose rosette disease (RRD) of multiflora rose (Rosa multiflora Thunb.), root tip extracts from both symptomatic and nonsymptomatic roses were used to mechanically inoculate leaves of Nicotiana glutinosa. Pale green spots were observed along the margins of the major leaf veins only on leaves inoculated with extracts prepared from symptomatic rose plants. Light microscopy revealed abnormal development of the palisade and spongy mesophyll cells in the symptomatic tissue, although no virus‐like particles (VLPs) were observed by electron microscopy. However, VLPs were observed in cells from tissue adjacent to the leaf veins and bordered by the pale green spots. Inoculation of N. benthamiana with extracts from symptomatic N. glutinosa initially did not result in visible symptoms on N. benthamiana inoculated leaves. However, approximately 4 wk post inoculation, splitting of leaf tissue across and along major leaf veins in expanding leaves occurred. In later stages of leaf expansion some leaves split in regions not associated with veins. Light microscopy of thick sections revealed separation between palisade cells and groups of small dead cells in the mesophyll tissue of expanding systemically infected leaf blades. Electron microscopy revealed crystalline arrays in the cytoplasm of mesophyll cells. No abnormal cellular changes were observed in plants inoculated with extracts prepared from nonsymptomatic rose plants.  相似文献

Mycoplasma-like bodies were found by electron microscopy in the sieve tubes of blackberry, loganberry and raspberry plants showing symptoms of Rubus stunt disease, but not in those of normal plants. Watering diseased blackberry plants with aureomycin caused remission of the symptoms and greatly decreased the number of mycoplasma-like bodies.  相似文献

Plant pathogen effectors encoded by Avirulence (Avr) genes benefit the pathogen by promoting colonization and benefit plants that have a matching resistance (R) gene by constituting a signal that triggers resistance. The Hessian fly, Mayetiola destructor (Say) (Diptera: Cecidomyiidae), resembles a plant pathogen in showing R/Avr interactions. Because of these interactions, a wheat plant with the H13 resistance gene can be resistant or susceptible depending on the genotype of the larva that attacks the plant, being resistant if attack comes from a larva with a functional vH13 gene, but susceptible if attack comes from a larva with a non‐functional vH13 gene. In this study we asked: does this susceptible interaction involving plants with H13 look like susceptible interactions with plants lacking H13? Possibly, the H13 plant attacked by a larva with a non‐functional vH13 is induced to partial rather than complete resistance. Or the larva, lacking its vH13‐encoded effector, is compromised in its ability to induce susceptibility, which includes forcing the plant to create a gall nutritive tissue. Responses of epidermal cells to larval attack were explored using imaging techniques (light microscopy, scanning and transmission electron microscopy). Whole‐organism responses were investigated by measuring the growth of plants and larvae. No evidence was found for partial resistance responses by H13 plants or for a compromise in the ability of vH13 loss‐of‐function larvae to induce susceptibility. It appears that disrupting vH13 function is sufficient for overcoming the induced resistance mediated by the H13 gene.  相似文献

分别用考马斯亮蓝染色和间接免疫荧光标记,并运用荧光倒置显微镜和激光共聚焦显微镜,对慈姑根尖固定后酶解获得的去壁细胞和细胞团块以及根尖细胞分裂周期中微管骨架列阵进行详细观察,以探索高等植物微管周期的普遍性。结果表明:慈姑根尖固定后酶解可获得大量结构完整的去壁细胞与细胞团块;考马斯亮蓝染色观察可见,慈姑根尖细胞中丰富的蛋白物质以及处于不同分裂期的细胞核染色体;免疫荧光观察可见,慈姑根尖细胞周期中微管骨架保存较好,主要有周质微管、早前期带微管、纺缍体微管和成膜体微管4种循序变化的排列方式,构成了高等水生植物分裂细胞中典型的微管周期。实验结果证明,高等水生植物与陆生植物微管周期具有相似性,为植物微管周期概念提供了新的实例。  相似文献

Summary Black grama (Bouteloua eriopoda) and fourwing saltbush (Atriplex canescens) are important grass and shrub species in arid rangelands of the northern Chihuahuan Desert. They are naturally colonized by dark septate endophytic fungi that cannot be eliminated by seed disinfestation. Plants were regenerated from both species and appeared to be fungus-free in axenic cultures. Analysis of callus and regenerated plants of both species using dual staining with light and scanning electron microscopy revealed fungal endophytes intrinsically associated with cells, roots and leaves of regenerated plants that are also associated with native plants. Fungal layers and biofilms prevent direct exposure of callus, root or leaf tissues to the external environment. Micropropagation is a valuable tool for identifying key fungal endophytes that enhance drought tolerance in native desert plants.  相似文献

In greenhouse experiments, we investigated the potentials of Piriformospora indica (Pi) to penetrate and colonise roots of wheat and to induce beneficial effects on growth as well as to reduce seedlings damping-off disease of wheat caused by the soil-borne fungus Fusarium oxysporum (Fo). By microscopy we observed the intracellular hyphae within root cortex cells of 7–14 day-old plants and chlamydospores within root cortex cells of 14–28 day-old plants and within root hair cells of 21–28 day-old plants. Moreover, diagnostic PCR based on the β-tubulin gene marker confirmed the presence of Pi in roots of inoculated plants. Also, we found that plants inoculated with Pi exhibited a better growth of roots and shoots as well as early flowering as compared with non-treated plants. Moreover, Pi conferred tolerance of wheat to seedlings damping-off i.e. by reducing the harmful effects of Fo on infected plants.  相似文献

Infection of perennial ryegrass (Lolium perenne L.), Italian ryegrass (Loliummultiflorum L.), and timothy (Phleum pratense L.) by Xanthomonas campestris pv. graminis and Xanthomonas campestris pv. phlei occurred mainly via wounds rather than natural openings. Nevertheless, bacteria were detected by isolation and immunofluorescence in plants sprayed with the pathogen without prior wounding and in plants in which intact ears had been dipped in inocula. High concentrations of bacteria were observed around the stomata of perennial ryegrass and timothy by scanning electron microscopy 48 h after inoculation. Perennial ryegrass and Italian ryegrass and individual plants of ryegrass and timothy differedin susceptibility to the pathogens.  相似文献

Many aquatic plants act as biosorbents, removing and recovering metals from the environment. To assess the biosorbent activity of Egeria densa, a submerged freshwater macrophyte, plants were collected monthly from a circular drainage area in Lake Biwa basin and the Mn concentrations of the plants were analysed. Mn concentrations in these plants were generally above those of terrestrial hyperaccumulators, and were markedly higher in spring and summer than in autumn. Mn concentrations were much lower in plants incubated in hydroponic medium at various pH levels with and without Mn supplementation than in field‐collected plants. The precipitation of Mn oxides on the leaves was determined by variable pressure scanning electron microscopy‐energy dispersive X‐ray analysis and Leucoberbelin blue staining. Several strains of epiphytic bacteria were isolated from the field‐collected E. densa plants, with many of these strains, including those of the genera Acidovorax, Comamonas, Pseudomonas and Rhizobium, found to have Mn‐oxidizing activity. High Mn concentrations in E. densa were mediated by the production of biogenic Mn oxide in biofilms on leaf surfaces. These findings provide new insights into plant epidermal bacterial flora that affect metal accumulation in plants and suggest that these aquatic plants may have use in Mn phytomining.  相似文献

Barley yellow striate mosaic virus (BYSMV) was identified in Iran by electron microscopy and serology. The virus was widespread in the Fars province causing mosaic, stunting and head sterility in wheat and mosaic symptoms in Setaria spp. In 1989, about 1/3 of wheat plants in the Bajgah Experiment Station, 15 km north of Shiraz, were infected. The yield in individual plants was drastically affected. Rhabdovirus particles were consistently observed in leaf-dip preparations and thin sections from the infected plants. The virus was transmitted by Laodelphax stritellus to wheat. It reacted with BYSMV antisera from Italy and Morocco but not with antisera to several other rhabdoviruses of gramineous plants.  相似文献

A virus was isolated from squirting cucumber (Ecballium elaterium L.) collected in France (Alpes de Haute Provence). After mechanical inoculation, eight species belonging to the Cucurbitaceae were found susceptible to this virus with systemic symptoms of mosaic, chlorotic spots, and fruit deformations. The French ZYFV (ZYFV-Fr) isolate differed from the type strain in its ability to infect some differential host plants. Elongated flexuous particles with a modal length of 752 nm were observed by electron microscopy in leaf extracts. Cytoplasmic inclusions similar to those associated with infections by members of the potyvirus group were observed by light microscopy. The virus was found by SDS-immunodiffusion and DAS-ELISA to be serologically related but distinct from the type strain of zucchini yellow fleck virus from Italy (ZYFV-lt). ZYFV has not yet been found in cultivated cucurbit plants in France; this virus appears to be restricted to squirting cucumbers in a few localities. The anatomy of normal and hyperhydric shoots (leaves and stems) of in vitro Handroanthus impetiginosus was compared using light microscopy, scanning electron microscopy, and transmission electron microscopy. In contrast to normal shoots, hyperhydric shoots presented numerous anatomical abnormalities at the proliferation stage. Disorganized cortex, epidermal holes, epidermal discontinuity, collapsed cells, and other structural characteristics were observed in hyperhydric shoots. So, by using anatomical analysis of in vitro H. impetiginosus shoots at the proliferation stage, we can predict which plants will survive the rhizogenesis and acclimatization stages.  相似文献

Common bacterial blight, caused by Xanthomonas axonopodis pv. phaseoli, is an important disease affecting beans causing considerable yield losses in many producing areas. Our aim was to investigate Xap–beans interactions in the first 48 h after infection analyzing structurally relevant changes by electron microscopy. Scanning electron microscopy analysis demonstrated morphological changes on leaf surfaces of both species. Transmission electron microscopy showed different types of chloroplast structural disorganization in both parenchyma and bundle sheath cells of Vigna unguiculata which were only observed in parenchyma cells of Phaseolus vulgaris. Additionally, we report an unexpected physical association between the chloroplasts and mitochondria observed only to P. vulgaris-infected plants.  相似文献
