A simplification of Brown’s fluorometry for monkey urinary estrogens has been attained. The method requires only a one-step extraction of estrogens instead of the complicated three-step extractions in the original Brown’s method. The procedure made it possible to shorten the time from the six to seven hours required in the original method to three hours. The present method is satisfactory in precision, sensitivity, accuracy, and specificity. Although the values of urinary estrogens obtained by the simplified method are lower than those obtained by the original method, the use of the present method is validated, since the correlation coefficient between the two assay procedures is excellent (γ=0.90) and also the application of the present method to the measurement of daily changes in urinary estrogens throughout the menstrual cycle has been successful. The simplified assay is expected to be useful in the routine assessment of urinary estrogens in Japanese monkeys. This work was supported by a grant from the Ford Foundation (No. 670-0558).  相似文献

Methods of artificial insemination (AI) for indoor breeding in the Japanese monkey and the Cynomolgus monkey were investigated. For the Japanese monkey AI was carried out in six females during the winter mating season and in six females during the summer non-mating season. During the mating season, semen was inseminated near ovulation time in natural menstrual cycles. In the mating season study, three females inseminated at the uterine cavity became pregnant. Three inseminated at the cervical canal failed to become pregnant. For the non-mating season study, ovulation was induced artificially by PMSG and hCG and AI was carried out near the induced ovulation time. In the non-mating season, no animals became pregnant. Of four Cynomolgus monkeys used, pregnancy occurred in two animals inseminated near ovulation time in natural menstrual cycles. AI occurred at the uterine cavity in one and cervical canal in the other. In both species ovulation was verified by laparoscopy. Semen was collected by penile electro-stimulation then diluted to 2.5 to 5.0×107/ml with Whitten's medium. Diluted semen of 0.2l was inseminated at the uterine cavity or cervical canal. Our results indicate the usefulness of vaginal AI as a method of artificial indoor breeding.  相似文献

Sexually mature bison (Bison bison) cows were tested for both pregnancy and ovulation by means of urinary steroid metabolites and fecal steroids. The accuracy of pregnancy diagnosis was determined among 18 bison cows, in approximately the third month of gestation, by means of urinary pregnanediol-3 delta-glucuronide (PdG), urinary estrone conjugates (E1C), and fecal total estrogens (TE). Urinary PdG was 100% accurate, urinary E1C was 89% accurate, and fecal TE was 100% accurate in predicting pregnancy. Fecal progesterone (P4) and TE, as well as urinary E1C and PdG concentrations all increased from conception (August) through January, but significant differences were not apparent until November. During the rutting season ovulation was detected by increases in either urinary PdG or fecal P4 concentrations. Both pregnancy and ovulation were detected in uncaptured bison with reasonable accuracy by means of urinary and fecal steroids or their metabolites.  相似文献

Practical aspects of urinary estrogen analysis were considered with regard to establishing simple and reliable methods for monitoring ovarian function in marmosets and tamarins. Changes in the hormone:creatinine ratio in small volumes of urine from the common marmoset were significantly correlated with changes in 24-h excretion. Comparison of the metabolism and excretion of estrogens during the ovarian cycle in the common marmoset and cottontop tamarin revealed interesting species differences. High concentrations of conjugated estrone were measured in marmoset plasma, but estradiol 17β was the predominant estrogen in urine. In contrast, estrone was the most abundant estrogen measured in tamarin urine. Both species excreted very little estriol. Sulfates and glucuronides were present in urine in similar proportions before ovulation in the marmoset, although after ovulation sulfates were the more abundant. Conversely, most of the estrogens in tamarin urine appeared to be conjugated as glucuronides. Direct assay for estrone sulfate was applied to the measurement of urinary estrogen excretion during the ovarian cycle in a marmoset. The results compared well with those for total estradiol 17β after hydrolysis and ether extraction. The use of direct assays for conjugated estrogens in small volumes of urine is suggested as a practical method for monitoring ovarian function in marmosets and tamarins.  相似文献

A direct immunoassay for urinary estrone conjugates (estrone sulfate and estrone glucuronide) was used to assess the preovulatory estrogen rise in normal gorilla menstrual cycles. Immunoreactive estrone conjugates in samples concomitantly assessed for total estrogen immunoreactivity reflected similar profiles throughout the cycle; however, the speed and resolution of the direct assay for conjugates indicate this method to be more accurate in monitoring ovulation than the measurement of total immunoreactive estrogens. In a single conceptive ovarian cycle, urinary estrone conjugate continued to rise in the luteal phase, indicating that this test may also be useful for detecting early pregnancy. The application of this technique provides clear profile of ovarian function in gorillas as well as in other primate species.  相似文献

Laparoscopic observations of ovaries before and after ovulation were made in 74 cycles in 39 Japanese monkeys (Macaca fuscata). Lengths of menstrual cycles, the follicular phase and the luteal phase in these animals were 25.3±2.8 days, 13.2±1.4 days, and 12.8±2.3 days, respectively. Ovarian morphology during the menstrual cycle in the Japanese monkey agreed in some features and disagreed in others, with findings inMacaca mulatta andMacaca fascicularis reported in the previous papers using laparotomy or laparoscopy. Follicular appearance before and after ovulation varied so considerably from ovary to ovary that it was hard to predict the accurate ovulation time or to estimate the precise age of corpus luteum by means of observations on the morphological appearance only. However, diagnosis of whether ovulation had already occurred or not, was possible when careful observations were made, although some ovaries without haemorrhagic and luteinized appearance after ovulation were very similar to pre-ovulatory follicle. Clear luteinization was detected without fail in many cases three days after ovulation. How ovariectomies and the administration of estrogen affected the vaginal smears, cervical mucus, and sexual skin of two Japanese monkeys (Macaca fuscata yakui) was examined. At the same time, these monkeys were determined for the level of urinary estrogens as an index of estrogen maintenance. The ovariectomy and the hormonal treatment had remarkable effects on these three characters, but the occurrence and duration of responses of these characters differed from one to another. The vaginal smears responded slowly to the treatments, and once the responses appeared, then continued for a long time, even after the estrogens were thoroughly excreted. Cervical mucus began to increase from one to two days after the increase of urinary estrogens, and it returned to its pre-operation level coinciding with disappearance of urinary estrogens. The appearance and disappearance of flushing of the sexual skin were nearly coincident with those of urinary estrogens. Urinary estrogens began to increase from the next day of the administration of estrogen (200 µg/head/day), and returned to the previous level in 12 days after the onset of the hormonal treatment in cases of 4 or 5-day-administration and in seven days in cases of a single injection.  相似文献

Time-series analysis was applied to the urinary total estrogen data from 142 ovulatory menstrual cycles to determine the first statistically significant increase as a marker for the beginning of the potentially fertile phase. Application of the Trigg's tracking signal to each cycle detected an increase in urinary total estrogens above the baseline in every case, with a cumulative probability of > or = 95%. The distribution of first increase days ranged from 10 days before 3.5 days before the presumed day of ovulation with a mean of 6.5 +/- 1.4 days. The significant parameters in the calculation of the tracking signal are the smoothing constant (alpha) which is related to the number of baseline observations, the baseline mean for the cycle, and the variation of the baseline mean. The method allows a calculation of the tracking signal as the cycle unfolds and a statistical assessment can be given each day. The procedure is easily adaptable for computer calculation, and because it is applied to individual cycles avoids the use of population means with the loss of information inherently associated with the combination of data from many cycles. The Trigg's tracking signal is an appropriate method of analysis of menstrual cycle data and represents a satisfactory alternative to the more usual cumulative sum procedure. The distribution of the first increases constitutes a reference standard for urinary estrogen assays.  相似文献

Lipomyces starkeyi is an oleaginous yeast, and has been classified in four distinct groups, i.e., sensu stricto and custers α, β, and γ. Recently, L. starkeyi clusters α, β, and γ were recognized independent species, Lipomyces mesembrius, Lipomyces doorenjongii, and Lipomyces kockii, respectively. In this study, we investigated phylogenetic relationships within L. starkeyi, including 18 Japanese wild strains, and its related species, based on internal transcribed spacer sequences and evaluated biochemical characters which reflected the phylogenetic tree. Phylogenetic analysis showed that most of Japanese wild strains formed one clade and this clade is more closely related to L. starkeyi s.s. clade including one Japanese wild strain than other clades. Only three Japanese wild strains were genetically distinct from L. starkeyi. Lipomyces mesembrius and L. doorenjongii shared one clade, while L. kockii was genetically distinct from the other three species. Strains in L. starkeyi s.s. clade converted six sugars, d-glucose, d-xylose, l-arabinose, d-galactose, d-mannose, and d-cellobiose to produce high total lipid yields. The Japanese wild strains in subclades B, C, and D converted d-glucose, d-galactose, and d-mannose to produce high total lipid yields. Lipomyces mesembrius was divided into two subclades. Lipomyces mesembrius CBS 7737 converted d-xylose, l-arabinose, d-galactose, and d-cellobiose, while the other L. mesembrius strains did not. Lipomyces doorenjongii converted all the sugars except d-cellobiose. In comparison to L. starkeyi, L. mesembrius, and L. doorenjongii, L. kockii produced higher total lipid yields from d-glucose, d-galactose, and d-mannose. The type of sugar converted depended on the subclade classification elucidated in this study.  相似文献

In captive Goeldi's monkeys, estrogen concentration was determined in fecal samples collected from 4 cycling/unmated females and 4 postpartum/mated females in order to ascertain the potential of fecal estrogen monitoring for providing basic information about reproductive status in this endangered Amazonian monkey. Subjects were fed an omnivorous diet and first-void feces were collected in the home cage at 1–3-day intervals for 30–50 days from the cycling females, and at 6–14-day intervals around the estimated time of the postpartum ovulation in each of the 4 mated females. Estimates for the duration of ovarian cycles (22–26 days) and the timing of ovulation were based on cyclic profiles of either blood progesterone or urinary pregnanediol-3α-glucuronide. Fecal estrogen values were normalized using these plasma or urinary profiles. HPLC analysis of estrogen from postpartum fecal samples demonstrated the presence of unconjugated estrone and estradiol-17β (“unconjugated estrogen”). Unconjugated estrogen was extracted and its fecal concentration estimated via EIA. The correlation (r) between plasma estrone conjugates and fecal unconjugated estrogen across nonconception ovarian cycles was 0.65 and measurement of the latter generated cyclic profiles. A range of 4–36-ng unconjugated estrogen g?1 feces was identified for follicular phases of nonconception cycles. Fecal unconjugated estrogen first exceeded the concentration range of the follicular phase 2–5 days after ovulation; the range was 49–402 ng g?1 feces in samples collected during the remainder of these nonconception luteal phases. Luteal phase concentrations were on average 10-fold higher than follicular phase concentrations. Each of the 4 mated females conceived at its postpartum ovulation; concentrations of fecal unconjugated estrogen excreted by 3 of these females demonstrated a marked postovulatory increase. This study demonstrates that fecal unconjugated estrogen can be applied to monitor ovarian cyclicity in Goeldi's monkey. © 1994 Wiley-Liss, Inc.  相似文献

Measurements of various parts of the head and body and weighing the body were carried out on about 170 adult Japanese monkeys (Macaca fuscata) and the results are noted with separate statistics for respective local groups. Intraspecific comparisons in the Japanese monkey and interspecific comparisons in macaques are discussed from the somatometrical point of view. Among macaques, the Japanese monkey has a comparatively large body, a very short tail, relatively wide biacromial and biiliac breadths, and markedly la ge intermembral and intercrural indices. The Japanese monkey itself shows various local variations. The most conspicuous difference is to be found between the so-called Yaku monkey living on Yaku islet (Yakushima), south of Kyushu, and the monkeys living in other parts of Japan, and, therefore, it is understandable that the Yaku monkey has been distinguished as a subspecies (M. f. yakui) of the Japanese monkey. The Yaku monkey has a somewhat small body, a relatively large head, wide hips, and slender hands and feet.  相似文献

Some dyads of Japanese monkey adult males and females show remarkable spatial proximity and frequent exchanges of social behaviors. It is suggested that some kind of “affinity” exists between them. Females obtain much unilateral benefit from “proximity effects”; even lowranking females can dominate high-ranking females as long as they stay nearby their “affinitive” males. Males acquire female followers in return. Mating relations and female mother-daughter relations play important roles in forming new “affinitive relations.” Once monkeys have formed “affinitive relations,” however, they seldom mate with each other, as if they were kin-related. Therefore, the acquisition of female followers appears inconsistent with a male's strategy for reproducing many genes in the next generation. This paper describes an original dot-enzyme-linked immunosorbent assay (ELISA) for predicting ovulation in women, based on the detection of the pre-ovulatory estrogen peak in urine. A monoclonal anti-estrogen antibody is used which recognizes not only free estrogens but also some of their urinary metabolites (17-glucuro- and sulfo-conjugates) allowing a direct assay on early morning urines. Antigen is immobilized as a spot on a nitrocellulose membrane which is immersed in urine in the presence of this antibody. A peroxidase-labeled second antibody allows the detection of the first antibody bound to the membrane. Antigen and anti-estrogen antibody concentrations are chosen to obtain a maximal enzymatic coloration of spots corresponding to basal urinary estrogen levels and no coloration corresponding to the pre-ovulatory surge. Six menstrual cycles were studied, comparing dot-ELISA results with patterns of: (1) urinary estrogens measured by RIA either directly or after hydrolysis and extraction, and (2) basal body temperatures. The validity of the pre-ovulatory signal obtained and the requirements for an adaptation of this methodology to a reliable home kit are discussed.  相似文献

Ateles generally lives in small temporary subgroups. The authors studied the grouping of the monkey for two months and obtained data concerning the subgroup size, composition, and inter-individual relationships. In general, the data agreed with that obtained byKlein (1972). However, they discovered that large subgroups of the monkey were observed only in relation to the utilization of a special place, the “salado” site. The authors discuss the reason for this. Criticism is given ofKlein's suggestions that the unique grouping ofAteles is a form of social adaptation to its palm-fruit eating behavior and that peripheral males exist in its social structure.  相似文献

The incidence of adenine phosphoribosyltransferase (APRT) deficiency is higher among Japanese nationals than among other ethnic groups, and the most common mutation (APRT*J, ATG to ACG mutation at codon 136) accounts for 68% of the disease-causing genes among Japanese. To investigate the origin of these mutations, we studied the geographical distribution of the mutant genes in Japan. The APRT*J mutation is distributed nearly uniformly in the four main islands of Japan and Okinawa, suggesting a very early origin. The products of PCR amplification between positions 2344 and 2750 of the genomic APRT sequence were examined by SSCP analysis in random blood samples from Japanese, Korean, and Taiwanese nationals. Among 955 random Japanese blood samples, 7 (0.73%) were heterozygous for the APRT*J mutation, giving a calculated heterozygote frequency of 1.1% among Japanese for the entire APRT deficiency. None of 231 Taiwanese samples contained heterozygotes for the APRT*J mutation, while 2 (0.53%) of 356 Korean samples were heterozygous. In addition to the APRT*J sequence, a total of five variant sequences was found. Sequencing one variant revealed a base substitution in intron 4, suggesting therefore that they are harmless mutations. Since the APRT*J mutation is present in Koreans and Okinawans who share ancestors only before the Yayoi era (third century bc to third century ad), the origin of the APRT*J mutation predates 300 bc. The troop fission of Japanese monkeys has been observed in 11 troops, and the total number of fissions observed numbered 22. Examined cases of fission relative to season, amount of food, troop size and socionomic sex ratio of the main troop and to individuals that acted as nucleus in forming a branch troop and females, exhibited variance, not being uniform. But inquiry into troop fission as seen from the standpoint of the mechanism maintained by Japanese monkeys' society indicated (1) troop fission checks the growth of troop size and increases the socionomic sex ratio of the main troop, (2) troop fission is an effective mechanism for the maintenance of class structure among adult males, and (3) the possibility that troop fission functions to check inbreeding.Thus, it must be said in general that troop fission, along with single desertions of males, plays a very important role as one mechanism for the maintenance of the society of Japanese monkey. However, the branch troop is often composed of a large number of males and a small number of females; in short, it is under very difficult conditions that a branch troop starts on its way to becoming a fully established Japanese monkey troop.The troop fission is an effective mechanism for the maintenance of the main troop, but not too effective for that of the branch troop.It cannot be said that there are abundant data on troop fission of primates other than Japanese monkey. But, from limited data, we can find various forms of troop fission according to each species, and it may be ultimately related to the maintenance mechanism of the primate society.  相似文献

 High resolution lectin-gold cytochemistry was used to quantitatively analyze the distribution of glycoconjugates in the hamster oviductal ampulla during the five stages of the estrous cycle. Lectins binding to N-acetyl-d-galactosamine-, d-galactose-, and sialic acid-associated glycoconjugates in the secretory granules of ampullary epithelial secretory cells showed staining of equal intensity throughout the five different stages of the estrous cycle. In contrast, the labeling intensity of glycoconjugates which contain N-acetylglucosamine as terminal sugar residues reached its maximum around the time of ovulation, i.e., at proestrus. Glycoconjugates which carry fucose and mannose as terminal sugar residues appeared to be totally absent from the secretory granules of the oviductal ampulla during the estrous cycle. Together, electron microscopic observations combined with quantitative results indicate that N-acetyl-d-galactosamine-, d-galactose-, and sialic acid-associated glycoconjugates may be secreted into the ampullary lumen irrespective of the stage of the estrous cycle, whereas the secretion of certain N-acetylglucosamine-associated glycoconjugates is stage specific and reaches its peak at the time of ovulation. These findings suggest that, at the time of ovulation, the ampullary epithelium changes its secretory activity and contributes its secretory products to the zona pellucida of oocytes freshly released from the ovary. Copulatory behavior unrelated to conception is sometimes observed in some non-human primates including the Japanese monkey. In the present study, the authors examined whether a mature follicle or a newly formed fresh corpus luteum was observed in the ovaries of female Japanese monkeys which displayed the copulatory behavior unrelated to conception. Post-conception copulatory behaviors were observed in three out of four females usually kept in individual cages in an air-conditioned room, and in two out of three females without infants kept in an outdoor group cage. However, neither a mature Graafian follicle nor a fresh corpus luteum formed newly after conception was observed in any of these females by laparoscopic examinations conducted immediately after termination of the copulatory behavior. In females with infants born in the preceding birth season, copulatory behaviors were observed in three out of four females kept in the outdoor group cage, and in two out of four females in a free-ranging troop. Ovulation was confirmed in one case out of the three kept in the outdoor group cage, but neither a mature follicle nor a newly formed corpus luteum was observed in the remaining four females. These findings suggest that copulatory behavior in the Japanese monkey is not always controlled by the development of a follicle or ovulation in the ovary.  相似文献
