Female rats were exposed to odors from females that went through a birth cycle of pregnancy, birth and lactation. The estrous cycle was shortened by odors from pregnant rats and lengthened by odors from lactating rats and their pups. Changes in estrous cycle length were accompanied by changes in the length of the luteal phase (metestrus-early diestrus). A log survivor plot demonstrated that variation in cycle length was the result of different rates of ovulation within the population under each condition. Neither pregnancy nor lactation odors affected the intensity or timing of the lordosis reflex. These odors could modulate fertility and coordinate birth cycles within a group of rats. 相似文献

Summary. The aim of this study was to investigate the enhancing effect of polyamines on intestinal absorption of fluorescein isothiocyanate-labeled
dextran (MW 4400, FD-4) in the in situ loop study and in vivo oral absorption study. Absorption of FD-4 from the jejunum was
significantly enhanced by 5 mM spermine without serious membrane damage in the jejunum. An in vivo oral absorption study was
also performed, and plasma FD-4 levels increased significantly after co-administration of 30 mM spermine. In the in vitro
transport studies with Caco-2 cells, prolonged incubation with spermine resulted in a gradual decrease in transepithelial
electrical resistance. This finding suggests that the absorption-enhancing mechanism of spermine partly includes opening the
tight junctions of the epithelium via the paracellular route. These results indicate that excess oral ingestion of polyamines
may have widespread health effects via the modulation of the intestinal epithelial barrier function. 相似文献

In the present investigation, the deposition of aluminum in intestinal fragment and the appearance in blood were studied in a perfused rat intestine in situ for 1 h with several aluminum forms (16 mM). We observed that aluminum absorption was positively correlated with the theoretic affinity of aluminum and the functional groups of the chelating agent. The absorption of aluminum after ingestion of organic compounds is more important than after ingestion of mineral compounds, with the following order: Al citrate > Al tartrate, Al gluconate, Al lactate > Al glutamate, Al chloride, Al sulfate, Al nitrate. Absorption depends on the nature of the ligands associated with the Al3+ ion in the gastrointestinal fluid. The higher the aluminum retention in intestinal fragment, the lower the absorption and appearance in blood. However, the higher aluminum concentration is always in the jejunal fragment because of the influence of pH variation on this fragment. Another objective of the present study was to determine the influence of several parameters on aluminum citrate absorption: with or without 0.1 mmol dinitrophenol/L, with aluminum concentration from 3.2, 16, 32, and 48, to 64 mmol/L, media containing 0, 3, or 6 mmol Ca/L, with or without phosphorus or glucose. It is concluded that aluminum is absorbed from the gastrointestinal tract by (1) a paracellular energy independent and nonsaturable route, mainly used for high aluminum concentration, which is modified by extracellular calcium, and (2) a transcellular and saturable route, the aluminum level was not modified with enhancement of aluminum quantity in intestinal lumen. This pathway can be similar with calcium transfer through the intestine and is energy dependent because of a decrease of aluminum absorption that follows the removal of glucose and phosphorus. 相似文献

1. Rates of lipolysis were measured at different concentrations of glucagon in adipocytes prepared from parametrial adipose tissue of fed or starved rats in different reproductive states. All experiments were performed in the presence of a high concentration of adenosine deaminase (1 unit/ml). 2. Maximal rates of lipolysis (elicited by 25 nM-glucagon in each instance) were higher in adipocytes from peak-lactating rats than those from pregnant animals in both the fed and starved states. 3. Of adipocytes from fed animals, those from peak-lactating rats were the most sensitive to glucagon, whereas those from late-pregnant and early-lactating rats were 1-2 orders of magnitude less sensitive. 4. Adipocytes from 24 h-starved rats showed a much smaller stimulation of lipolysis by glucagon, making the assessment of sensitivity difficult. Therefore, rates of lipolysis were also measured in the presence of a maximally anti-lipolytic dose of insulin. The presence of insulin did not alter the relative sensitivities to glucagon of adipocytes from fed animals in different reproductive states, although all dose-response curves were shifted to the right. When lipolysis in adipocytes from starved animals was measured in the presence of insulin, it became evident that starvation for 24 h markedly increased the sensitivity of adipocytes from late-pregnant rats to glucagon, but did not affect that of cells from animals in the other reproductive states. 5. It is concluded that the large changes in sensitivity to glucagon that occurred during the reproductive cycle may enable the modulation of adipose-tissue lipolysis in vivo to satisfy the different metabolic requirements of the animal in the transition from pregnancy to peak lactation. 相似文献

脊髓损伤是一类常见的、高致残率的中枢神经系统疾病,由于多种复杂因素影响其损伤后的修复过程,损伤脊髓的再生能力非常有限。本研究采用cDNA微阵列技术筛选大鼠脊髓损伤后出现的差异表达基因。实验组动物在T8-T9进行脊髓全横断手术,对照组动物只打开椎板;4.5d后取脊髓进行RNA提取并在反转录过程中进行Cy3/Cy5标记,然后与预制的、带有4041条特异性探针的芯片进行杂交。Cy5/Cy3信号比值≥2.0视为脊髓损伤后出现差异表达的基因。通过筛选,我们得到了65个上调表达基因(21个已知基因,30个已知EST和14个未知基因)和79个下调基因(20个已知基因,42个已知EST和17个未知基因)。进一步通过半定量RT-PCR对其中的5个上调已知基因(Timpl,Tagln,Vim,Fc gamma receptor,Ctss)和三个下调已知基因(stearyl-CoA desaturase,F2,Ensa)的表达情况进行了验证,结果显示与芯片结果一致。这些基因可能在脊髓损伤后的修复过程中起一定的作用,对其深入研究将有助于揭示脊髓损伤修复的分子机制。 相似文献

We investigated the use of Gly in a high-energybeverage containing corn syrup (CS) by using a small intestineperfusion procedure in the rat, an approach shown earlier to providegood preclinical information. The effectiveness of several formulationswith Gly and CS was compared with commercial products and toexperimental formulas where Gly substituted for glucose (Glc). TheCS-Gly combination was more effective than preparations on the marketcontaining sucrose and Glc-fructose syrups (G-P and G-L, respectively)in maintaining a net water absorption balance in the test jejunal segment [CS-Gly = 0.021 ± 0.226, G-L = 1.516 ± 0.467, and G-P = 0.299 ± 0.106 (SE)µl · min1 · cm1(P = 0.0113)] and in reducingsodium release into the lumen [CS-Gly = 133.2 ± 16.2, G-L = 226.7 ± 25.2, and G-P = 245.6 ± 23.4 nmol · min1 · cm1(P = 0.0022)]. In otherpreparations, at equal CS concentrations (60 and 80 g/l, respectively),Gly clearly improved net water absorption over a comparableGlc-containing product [CS60-Gly = 0.422 ± 0.136 and CS80-Gly = 0.666 ± 0.378 vs. CS60-Glc = 0.282 ± 0.200 andCS80-Glc = 1.046 ± 0.480 µl · min1 · cm1(P = 0.0019)]. Four lactating cows fitted with permanent ruminal, duodenal and ileal cannulae were used to study the effect of extrusion of whole lupin seeds at 195 degrees C (Lupinus albus cv Lublanc) on organic matter (OM) and nitrogen (N) degradation in the rumen and their flow to and absorption from the small intestine. Raw whole lupin seeds (RWLS) and extruded whole lupin seeds (EWLS) were fed in diets containing 15.5% crude protein and composed of 22.6% whole lupin seeds, 56.5% corn silage, 10.2% corn grain and 10.7% Italian ray-grass on a DM basis, supplemented with vitamins and minerals. Chromium ethylenediaminotetraacetic (Cr-EDTA) and ytterbium chloride (YbCl3) were used as liquid and particulate markers respectively, while purines and 15N ammonium sulfate were utilized as bacterial markers. Cows fed EWLS had a similar ruminal ammonia N and volatile fatty acid concentrations and efficiency of bacterial protein synthesis compared to those fed the RWLS diet. Total tract OM and N digestion were not affected by inclusion of EWLS instead of RWLS; the corresponding mean values were 70 and 71%. Apparent degradation of OM and N in the rumen were 44 and 64% for diets containing RWLS, and 40 and 39% for EWLS diets. Feeding diets including EWLS both increased non ammonia N and dietary N flow to the duodenum compared with diets containing RWLS (472 vs 357 g/d) and (263 vs 153 g/d) respectively. Absorption from the small intestine (g/d and % entering) of dietary N was higher for EWLS diets (146 vs 62 g/d; 34 vs 15%). The PDIA, PDIE and PDIN contents (g/kg of DM) of RWLS were 18, 94 and 245 respectively; the corresponding values after extrusion were 145, 220 and 220. 相似文献

The present study investigated the role of apolipoprotein E (apoE) phenotype on intestinal cholesterol absorption and cholesterol synthesis. Studies were carried out in eight subjects homozygous for the apoE4 and 12 subjects homozygous for the E2 allele (six normocholesterolemic volunteers and six patients with type III hyperlipoproteinemia). Cholesterol absorption did not differ between the three groups of subjects and averaged 38 +/- 2% (mean +/- SEM) in normolipemic E2/2, 37 +/- 4% in type III hyperlipemic E2/2, and 41 +/- 3% in E4/4 subjects, respectively. Dietary intake of fat and cholesterol had no influence on cholesterol absorption efficiency. A positive correlation between efficiency of cholesterol absorption and the ratio of campesterol to cholesterol in plasma, an indirect marker for cholesterol absorption, was observed after combining the results of the three groups (r = 0.504; P < 0.02). Bile acid and total cholesterol synthesis were also not affected by the different apoE alleles, but the well-known relationship between body weight and cholesterol synthesis was noticed (r = 0.574; P < 0.01). Thus, the present study provides evidence that the efficiency of intestinal absorption and synthesis of cholesterol in humans are not related to the apoE phenotype. 相似文献

Summary Barley plants were grown, for periods of up to eleven weeks duration, in culture solutions in which the potassium concentrations were maintained at 0.01, 0.10, 1.0, and 50.0 ppm. Potassium added continuously during the day was augmented night and morning, as determined by solution analysis. The following observations were made:Plants grew equally well at pH levels of 5.5 and 7.0. Variations in root temperature between 15° and 25°C had no effect on K-content of tissue at either pH level. Yields were large as were the quantities of potassium absorbed from the solutions. No potassium-deficiency symptoms were observed at any of the concentrations used.Considerable variation in yield was observed between tanks and between plants growing in the same tank. The variation appeared to be due to a difference in the amount of tillering per plant.The results indicate that plants grow well in culture solutions at potassium concentrations approaching 0.01 ppm and this may have some implications on the mechanism of uptake of potassium by plant roots growing in a soil system. 相似文献

Tissue and organ deposition and blood parameters were evaluated as indices of mineral and trace element absorption in rats.
The absorption of elements was quantified in relation to nitrogen retention, i.e., considering the weight gain and new tissue
synthesis. A rapeseed meal diet was supplied with three levels of calcium, two levels of zinc, and two levels of copper in
a factorial design. In general, an increase in dietary mineral content increased the relative absorption, which in turn, increased
the tissue deposition progressively. Striated muscle, however, did not respond to either an increased calcium or zinc supply.
Furthermore, an increased calcium absorption caused a depression of the fractional phosphorus and magnesium content of femur
bones. The copper content of the kidneys and the heart muscle was directly proportional to the amount of absorbed zinc and
iron, respectively. The iron content of tissues was, in general, inversely proportional to zinc absorption and showed a tendency
to be directly proportional to copper absorption. The zinc level in tissues was, in a similar way, inversely correlated to
measured calcium absorption. In conclusion, interactions between elements do not only affect the intestinal element absorption,
but also the distribution of already absorbed elements in tissues and organs. 相似文献

The receptors and neurotransmitter pathways that may participate in the inhibitory action of 5-hydroxytryptamine (5HT) on prolactin release during late pregnancy and lactation in rats were studied. Administration of the 5HT synthesis inhibitor, p-chlorophenylalanine, to late pregnant rats induced a significant increase in serum prolactin concentrations at 17:00 h on day 19 of pregnancy that was partially blocked by injections of the 5HT precursor, 5-hydroxytryptophan, or the 5HT agonists, 8-hydroxy-2-(di-n-propylamino)-tetralin hydrobromide (S1a), 1-(2,5-dimethoxy-4-iodophenyl)-2-aminopropane (S2) and N-(3-chlorophenyl)imidodicarbonimide diamide HCl (S3), but not by RU 24969 (S1b) or 1-meta-(chlorophenyl)-piperazine-2-HCl (S1a-2c). The 5HT neurotoxins, fenfluramine and p-chloroamphetamine, which selectively destroy fine axon serotoninergic fibres but not coarse ones, prevented the increase in circulating prolactin observed at 18:00 h on pro-oestrus and on day 21 of pregnancy, but did not modify serum prolactin concentrations at 17:00 h on day 19 of pregnancy. Administration of the adrenergic antagonists, metoprolol or prazosin, also prevented the stimulatory effects of p-chlorophenylalanine or ketanserin in pregnant rats on day 19 (17:00 h) or on days 10-12 (16:30 h) in lactating rats separated from their litters. Administration of p-chlorophenylalanine to pregnant rats on day 19 reduced dopamine concentrations in the arcuate nucleus and in the anterior hypothalamus and noradrenaline concentrations in the anterior hypothalamus and the suprachiasmatic nucleus. These results indicate that the inhibitory actions of 5HT on prolactin release in pregnant and lactating rats are mediated by S1a, S2a and S3 receptors and by the coarse axon serotoninergic fibres. In addition, the inhibitory actions of 5HT may modulate the action of a stimulatory adrenergic pathway, as well as the concentrations of noradrenaline and dopamine in different hypothalamic areas, which, in turn, particularly arcuate nucleus dopamine, regulate prolactin release. 相似文献

The aim of present work was to study the effect of oral aluminium (Al) overload on intestinal calcium (Ca) absorption in the critical stages of pregnancy and lactation of rats and to find out possible relationships with prolactin (PRL) and 17beta-estradiol (E2) circulating levels. Adult female Wistar rats were orally treated from day 1 of pregnancy with 0 (control), or 50 mg elemental Al (as chloride)/kg body weight per day. Ca transport was determined by everted duodenal sacs technique using 2 microCi of (45)CaCl(2) as flux marker (JCa(ms)). Al treatment reduced JCa(ms) either in late pregnancy (day 19) or in middle lactation (day 9 postpartum). Oral administration of bromocriptine (BrC), an inhibitor of PRL secretion, at dose of 10 mg/kg body weight given 18 h before JCa(ms) measurements were done, produced a significant decrease in the inhibitory effect of Al on JCa(ms), expressed as percent of control, at day 9 of nursing (vehicle: 51+/-7%, BrC: 28+/-4%, P <0.05). A positive correlation between Al effects on JCa(ms) and the physiological variations of E2 serum levels along pregnancy and lactation in BrC-treated rats was also found (r(2)=0.277, P =0.001). We conclude Al could reduce transcellular Ca absorption in the duodenum by interfering with physiological mechanisms of Ca transport partially mediated by serum level increments of E2 and PRL, observed in late pregnancy and mainly during middle lactation of rats. 相似文献
