Dietary cholesterol can affect both body lipid composition and steroid hormone concentration. We investigated whether a diet
rich in cholesterol influences torpor patterns of hibernating chipmunks (Tamias amoenus) and, if so, whether these changes are better explained by diet-induced changes in body lipid composition or the concentration
of testosterone, which at high levels inhibits torpor. Two groups of chipmunks were maintained either on a cholesterol diet
(rodent chow containing 10% cholesterol) or a control diet (rodent chow) during pre-hibernation fattening and throughout the
hibernation season. Torpid chipmunks on the cholesterol diet had significantly lower minimum body temperatures (−0.2 ± 0.2
vs +0.6 ± 0.2 °C), lower metabolic rates (0.029 ± 0.002 ml O2 g−1h−1 vs 0.035 ± 0.001 ml O2 g−1h−1), and longer torpor bouts at −1 °C (6.8 ± 0.5 vs 4.1 ± 1.0 days) than chipmunks on the control diet. Dietary cholesterol
resulted in a significant increase in blood plasma cholesterol (sevenfold), liver cholesterol content (6.9-fold) and liver
triglyceride content (3.5-fold) in comparison to controls. In contrast, dietary cholesterol had no detectable effect on the
concentration of plasma testosterone, which was very low in both groups. Since torpor was deeper and longer in animals on
the cholesterol diet our study suggests that torpor patterns of chipmunks were either directly affected by the dietary cholesterol
or via changes in body lipid composition.

Seasonal variations in the thermal response of liver mitochondrial membranes from Richardson's ground squirrels (Spermophilus richardsonii) were determined by measuring succinate-cytochrome reductase activity and spin label motion over a temperature range of 2 °C to 35 °C. For seven summer animals from the field the Arrhenius-type plots for enzyme activity and spin label motion were biphasic indicating a transition in structure and function at , respectively; typical of homeothermic mammals. For 12 winter animals maintained at 19°C, the transition in structure and function was lowered to and , respectively. The transition for 5 of 11 winter animals which were kept at 4°C and maintained normal activity and body temperature was similar to animals maintained at 19°C, while for the other six the transition was further lowered to less than 4°C. The transition for seven winter animals which were in deep hibernation was less than 4°C. The results for liver mitochondria show that lowering of the transition in membrane structure and function occurs as a two-stage process of about 10 deg. C for each stage and that the lowering is a requisite for hibernation rather than a response to the low-body temperatures experienced during hibernation. 相似文献

Physiological variables of torpor are strongly temperature dependent in placental hibernators. This study investigated how changes in air temperature affect the duration of torpor bouts, metabolic rate, body temperature and weight loss of the marsupial hibernator Burramys parvus (50 g) in comparison to a control group held at a constant air temperature of 2°C. The duration of torpor bouts was longest (14.0±1.0 days) and metabolic rate was lowest (0.033±0.001 ml O2·g-1·h-1) at2°C. At higher air temperatures torpor bouts were significantly shorter and the metabolic rate was higher. When air temperature was reduced to 0°C, torpor bouts also shortened to 6.4±2.9 days, metabolic rate increased to about eight-fold the values at 2°C, and body temperature was maintained at the regulated minimum of 2.1±0.2°C. Because air temperature had such a strong effect on hibernation, and in particular energy expenditure, a change in climate would most likely increase winter mortality of this endangered species.Abbreviationst STP

The physiological signal for torpor initiation appears to be related to fuel availability. Studies on metabolic fuel inhibition in placental heterotherms show that glucose deprivation via the inhibitor 2-deoxy-D-glucose (2DG) initiates a torpor-like state, whereas fatty acid deprivation via mercaptoacetate (MA) does not. As previous studies using inhibitors were limited to quantifying body temperature in placentals, we investigated whether inhibition of glucose or fatty acids for cellular oxidation induces torpor in the marsupial hibernator Cercartetus nanus, and how the response of metabolic rate is related to body temperature. Glucoprivation initiated a torpor-like state in C. nanus, but animals had much higher minimum body temperatures and metabolic rates than those of torpid food-deprived animals and arousal rates were slower. Moreover, 2DG-treated animals were thermoregulating at ambient temperatures of 20 and 12 °C, whereas food-deprived torpid animals were thermo-conforming. We suggest that glucoprivation reduces the hypothalamic body temperature set point, but only by about 8 °C rather than the approximately 28 °C during natural torpor. Reduced fatty acid availability via MA also induced a torpor-like state in some C. nanus, with physiological variables that did not differ from those of torpid food-deprived animals. We conclude that reduced glucose availability forms only part of the physiological trigger for torpor initiation in C. nanus. Diets rich in unsaturated and polyunsaturated fatty acids have a positive effect on mammalian torpor, whereas diets rich in saturated fatty acids have a negative effect. To determine whether the number of double bonds in dietary fatty acids are responsible for these alterations in torpor patterns, we investigated the effect of adding to the normal diet 5% pure fatty acids of identical chain length (C18) but a different number of double bonds (0, 1, or 2) on the pattern of hibernation of the yellow-pine chipmunk, Eutamias amoenus. The response of torpor bouts to a lowering of air temperature and the mean duration of torpor bouts at an air temperature of 0.5°C (stearic acid C18:0, 4.5±0.8 days, oleic acid C18:1, 8.6±0.5 days; linoleic acid C18:2, 8.5±0.7 days) differed among animals that were maintained on the three experimental diets. The mean minimum body temperatures (C18:0, +2.3±0.3°C; C18:1, +0.3±0.2°C; C18:2,-0.2±0.2°C), which torpid individuals defended by an increase in metabolic rate, and the metabolic rate of torpid animals also differed among diet groups. Moreover, diet-induced differences were observed in the composition of total lipid fatty acids from depot fat and the phospholipid fatty acids of cardiac mitochondria. For depot fat 7 of 13 and for heart mitochondria 7 of 14 of the identified fatty acids differed significantly among the three diet groups. Significant differences among diet groups were also observed for the sum of saturated, unsaturated and polyunsaturated fatty acids. These diet-induced alterations of body fatty acids were correlated with some of the diet-induced differences in variables of torpor. The results suggest that the degree of unsaturation of dietary fatty acids influences the composition of tissues and membranes which in turn may influence torpor patterns and thus survival of hibernation.Abbreviations bm

Wildfires have increased in frequency and intensity worldwide with climate change as a main driving factor. While a number of studies have focused on population changes in regard to fires, there are essentially no quantitative data on behavioural and physiological adjustments that are vital for the persistence of individuals during and after fires. Here we show that brown antechinus, a small insectivorous marsupial mammal, (i) endured a prescribed fire in situ, (ii) remained in their scorched home range despite unburned areas nearby, and (iii) substantially increased post-fire torpor use and thus reduced foraging requirements and exposure to predators. Hence, torpor is a physiological adaptation that, although not quantified in this context previously, appears to play a key role in post-fire survival for this and other heterothermic species. 相似文献

Circadian rhythms have been observed in most mammals, but their importance and function remain controversial with respect
to daily cycles during hibernation. We investigated the timing of arousals from and entries into hibernation for both free-living
and captive mountain pygmy-possums (Burramys parvus). Under both natural and laboratory conditions most arousals and entries were entrained with the light-dark cycle. Entries
occurred mainly during the night and arousals preferably around dusk, which coincides with the onset of the normal activity
phase for the nocturnal pygmy-possums. This entrainment prevailed throughout the hibernation season although only the laboratory
animals were constantly subjected to photoperiodic stimuli, whereas under natural conditions hibernacula are shielded from
photic cues and diurnal temperature fluctuations. Nevertheless, possums left their hibernacula frequently throughout winter
and were occasionally trapped close to the snow surface suggesting that during the periods of post-arousal normothermia they
can be exposed to environmental stimuli. It thus appears that the synchronisation with the photocycle was governed by a temperature-compensated
circadian clock which was reset periodically during short activity periods. For the mountain pygmy-possum, entrainment with
the photocycle probably has two functions:
1. Entrainment ensures that foraging bouts during the hibernation season remain synchronised with the dark phase.
2. Information about the prevailing climatic conditions sampled during short activity periods enables them to time final spring
emergence from hibernation when snow melt begins and ensures that the breeding season can commence as early as possible.

Chickpea (Cicer arietinum L.) is an important food legume crop, particularly for the arid regions including Indian subcontinent. Considering the detrimental effect of drought, temperature and salt stress on crop yield, efforts have been initiated in the direction of developing improved varieties and designing alternate strategies to sustain chickpea production in adverse environmental conditions. Identification of genes that confer abiotic stress tolerance in plants remains a challenge in contemporary plant breeding. The present study focused on the identification of abiotic stress responsive genes in chickpea based on sequence similarity approach exploiting known abiotic stress responsive genes from model crops or other plant species. Ten abiotic stress responsive genes identified in other plants were partially amplified from eight chickpea genotypes and their presence in chickpea was confirmed after sequencing the PCR products. These genes have been functionally validated and reported to play significant role in stress response in model plants like Arabidopsis, rice and other legume crops. Chickpea EST sequences available at NCBI EST database were used for the identification of abiotic stress responsive genes. A total of 8,536 unique coding long sequences were used for identification of chickpea homologues of these abiotic stress responsive genes by sequence similarity search (BLASTN and BLASTX). These genes can be further explored towards achieving the goal of developing superior chickpea varieties providing improved yields under stress conditions using modern molecular breeding approaches. 相似文献

Mitogen-activated protein kinase (MAPK) family members such as c-jun N-terminal kinase (JNK) may act as signal transducers early during pancreatitis development and evidence indicates that MAPK phosphatases (MKP) downregulate MAPK. We therefore investigated expression and regulation of pancreatic MKP in vivo. Pancreatic MKP mRNA levels were near or below the detection threshold in unstimulated animals. Cerulein hyperstimulation strongly induced MKP-1, MKP-3, and MKP-5 expression, peaking 30 to 60 min after treatment. Thus, MKP's clearly are early responsive genes during pancreatitis induction. Interestingly, inhibition of MKP-1 expression by Ro-31-8220 maximally induced activation of JNK but not of p38 and ERK in acutely isolated acini. These effects indicate that JNK may indeed be a preferred MKP-1 substrate in vivo. 相似文献

Peanut (Arachis hypogaea L.) ranks fifth among the world oil crops and is widely grown in India and neighbouring countries. Due to
its large and unknown genome size, studies on genomics and genetic modification of peanut are still scanty as compared to other
model crops like Arabidopsis, rice, cotton and soybean. Because of its favourable cultivation in semi-arid regions, study on abiotic
stress responsive genes and its regulation in peanut is very much important. Therefore, we aim to identify and annotate the abiotic
stress responsive candidate genes in peanut ESTs. Expression data of drought stress responsive corresponding genes and EST
sequences were screened from dot blot experiments shown as heat maps and supplementary tables, respectively as reported by
Govind et al. (2009). Some of the screened genes having no information about their ESTs in above mentioned supplementary tables
were retrieved from NCBI. A phylogenetic analysis was performed to find a group of utmost similar ESTs for each selected gene.
Individual EST of the said group were further searched in peanut ESTs (1,78,490 whole EST sequences) using stand alone BLAST.
For the prediction as well as annotation of abiotic stress responsive selected genes, various tools (like Vec-Screen, Repeat Masker,
EST-Trimmer, DNA Baser, WISE2 and I-TASSER) were used. Here we report the predicted result of Contigs, domain as well as 3D

The PI3K/Akt signaling pathway has been recently suggested to have controversial functions in models of acute and chronic inflammation. Our group and others have reported previously that the complement split product C5a alters neutrophil innate immunity and cell signaling during the onset of sepsis and is involved in PI3K activation. We report in this study that in vivo inhibition of the PI3K pathway resulted in increased mortality in septic mice accompanied by strongly elevated serum levels of TNF-alpha, IL-6, MCP-1, and IL-10 during sepsis as well as decreased oxidative burst activity in blood phagocytes. PI3K inhibition in vitro resulted in significant increases in TLR-4-mediated generation of various proinflammatory cytokines in neutrophils, whereas the opposite effect was observed in PBMC. Oxidative burst and phagocytosis activity was significantly attenuated in both neutrophils and monocytes when PI3K activation was blocked. In addition, PI3K inhibition resulted in strongly elevated TLR-4-mediated generation of IL-1beta and IL-8 in neutrophils when these cells were co-stimulated with C5a. C5a-induced priming effects on neutrophil and monocyte oxidative burst activity as well as C5a-induced phagocytosis in neutrophils were strongly reduced when PI3K activation was blocked. Our data suggest that the PI3K/Akt signaling pathway controls various C5a-mediated effects on neutrophil and monocyte innate immunity and exerts an overall protective effect during experimental sepsis. 相似文献
