The aqueous extract of dried bonito (Katsuobushi) was distilled under reduced pressure. The resulting distillate with diethyl ether and the extract was separated into acidic, phenolic, basic and neutral fractions. The neutral fraction was further fractionated into ten sub-fractions by silica gel column chromatography. All these sub-fractions were analyzed by gas chromatography and gas chromatography-mass spectrometry.

One hundred and sixty-five compounds were identified and 12 compounds were tentatively identified from the neutral fraction. Among them, 111 compounds were newly identified as flavor components of Katsuobushi.  相似文献

The aqueous extract of dried bonito (Katsuobushi) was distilled by using a thin film evaporator. The resulting distillate was extracted with diethyl ether, and the extract was separated into basic, acidic, weak acidic, and neutral fractions.

The basic, acidic, and weak acidic fractions were analyzed by gas chromatography and gas chromatography-mass spectrometry.

Seventy-four compounds, including 24 acids, 24 phenols, 8 pyridines, 12 pyrazines, 3 thiazoles, and 3 other compounds were identified. Thirty-six of these compounds were newly identified as volatile flavor compounds of Katsuobushi.  相似文献

Adult rats with normal or X-irradiated testes were used in an experiment to test the possible existence of a chalone in the testis. On the 11th day following irradiation, i.e. as the type A spermatogonia proliferated actively to restore the partially destroyed spermatogonial population, the animals with irradiated testes were subdivided into three groups. Rats of the first group were injected intraperitoneally with a saline extract of normal adult rat testes. Animals of the second group were injected with an equal amount of physiological saline while the rats of the third group received equivalent injections of a saline liver extract. Two additional groups of rats with non-irradiated testes, injected with the testicular extract or saline solution, served as controls. Following the last injection all animals were injected with 3H-thymidine and sacrificed. From each animal one testis was used to determine the specific radioactivity of its DNA, the other testis was processed for radioautography. The testicular extract produced a significant decrease in uptake of radioactivity by the irradiated testes. There was no difference in the radioactivity uptake by the testes of non-irradiated rats. Correspondingly the labeling index of type A spermatogonia was significantly lower in animals of the first group than in the other two groups of animals with irradiated testes. However, there was no difference in the labeling indices of Intermediate and type B spermatogonia or of preleptotene spermatocytes in the animals receiving the extracts or the saline solution. In animals with non-irradiated testes there was no difference in the labeling indices of type A or other types of spermatogonia or of spermatocytes. These data were taken to indicate that a saline extract of normal adult testes contains a substance that can inhibit specifically the proliferation of type A spermatogonia during the repair phase of the spermatogonial population following irradiation. This substance was tentatively considered as a spermatogonial chalone.  相似文献

The biotransformation of the antidepressant drug amoxapine by Cunninghamella elegans formed three metabolites, 7-hydroxyamoxapine, N-formyl-7-hydroxyamoxapine, and N-formylamoxapine; two other compounds were only present when chloroform was used in the extraction process. All five of the compounds were separated by reversed-phase HPLC, then analyzed by 1H NMR and mass spectrometry, and by 13C NMR when sample quantities permitted. The artifacts were identified as N-carbomethoxy-7-hydroxyamoxapine and N-carbomethoxyamoxapine. Phosgene is a decomposition product of chloroform that can form carbomethoxy compounds at the secondary nitrogen of a piperazine ring in an alcoholic solution. Since N-carbomethoxy compounds were not observed when ethyl acetate was used for extraction of the culture medium, they were considered artifacts and not metabolites. These findings suggest that chloroform should be tested for the formation of phosgene before using it to extract any compound with a piperazine ring or any other amine-containing structure.  相似文献

A phosphate-solubilizing strain ofPenicillium bilaii was tested for the production of gliotoxin and other toxic compounds. The strain was fermented under five different conditions to allow the expression of various metabolites, including gliotoxin. These included Czapek-yeast extract medium under both shaken and still conditions as well as Czapek-yeast extract/malt extract/peptone medium and sucrose/glycerol medium in shake flasks. In addition, culture filtrate from an industrial fermentation of the fungus was examined. No gliotoxin was produced in any of the media. No other expectedP. bilaii metabolites were found. Three compounds were identified in all samples: dibutyl phthalate, 1-(4-hydroxy-phenyl)ethanone and 4-hydroxy-3,6-dimethyl-2H-pyran-2-one. The production of other metabolites was dependent on the culture conditions. Two hyalodendrin derivatives were found in some fermentations and two related compounds were tentatively identified. None of the compounds found have been reported as toxic. The identity of the culture was confirmed by comparison with the ex-type culture ofP. bilaii.  相似文献

Three bacterial strains identified as Paenibacillus sp., Aneurinibacillus aneurinilyticus and Bacillus sp. have been shown to decolourise kraft lignin in 6 days of incubation. The release of low molecular weight aromatic compounds by these bacterial strains during degradation of kraft lignin was analysed by GC–MS analysis. The total ion chromatograph (TIC) of ethyl acetate extract from kraft lignin sample inoculated by Paenibacillus sp. was similar to control except some minor changes in the chromatographic profile indicating incapability of this bacterium to modify kraft lignin. On the other hand the TIC of ethyl acetate extract from kraft lignin inoculated by A. aneurinilyticus and Bacillus sp. caused formation of several aromatic lignin-related compound that were not present in the extract of control. The compounds identified in extract of the sample degraded by A. aneurinilyticus were guaiacol, acetoguiacone, gallic acid and ferulic acid while t-cinnamic acid, 3,4,5 trimethoxy benzaldehyde, and ferulic acid by Bacillus sp. indicating oxidization of coniferylic (G units) and sinapylic (S units) alcohol of lignin polymer. Differences between the identified compounds from different bacterial treatment were strain-specific. Among the identified aromatic compounds, ferulic acid and 3,4,5-trimethoxy benzaldehyde could be useful to the industry of preservatives, aromas and perfumes.  相似文献

目的:考察蒲公英提取物对MCF-7增殖作用的影响,研究蒲公英抗肿瘤活性部位的化学成分。方法:采用MTT法比较蒲公英水提物、醇提物和不同极性提取物对MCF-7增殖的影响;以气相色谱.质谱(GC-MS)联用技术对具有抑制MCF-7人乳腺癌细胞增殖的部位进行分离和鉴定。结果:蒲公英水提物和醇提物对MCF-7细胞均有抑制作用,体外最大抑制率分别达到92.95%、93.59%,蒲公英不同极性溶剂提取物中,氯仿部位抑制MCF-7的增殖,且抑制作用高于醇提和水提物,乙酸乙酯和石油醚部位促进MCF-7的增殖。从蒲公英氯仿提取物中鉴定出80种化合物,占其总量的98.07%。结论:蒲公英氯仿部位具有抗肿瘤活性,为蒲公英抗肿瘤活性成分的研究提供了依据,拓展了筛选中药活性成分的方法。  相似文献

The rapid evaluation of antioxidant activity of crude borage (Borago officinalis L.) extract was determined by using DPPH free radical method. This borage extract resulted in a rapid decrease of the absorbance and showed very high hydrogen-donating capacity towards the 2,2'-diphenyl-1-picrylhydrazyl (DPPH) radical. A new HPLC-DPPH on-line method was applied for a screening of several radical scavenging components in this borage extract as well as for quantitative analysis. This on-line HPLC-DPPH method was developed using a methanolic solution of DPPH-stable radical. The HPLC-separated analytes reacted post-column with the DPPH solution in methanol. The induced bleaching was detected as a negative peak photometrically at 515 nm. The separation of antioxidative components was carried out by gradient HPLC with mobile-phase composition ranging from 2% to 80% acetonitrile with 2% acetic acid in water, UV detection was carried out at 280 nm. The HPLC analysis of borage extract revealed the presence of several radical scavenging components in the borage extract. The results obtained from the chromatograms suggest that some compounds present in the extract possess high radical quenching ability. The dominant antioxidative compound in the crude extract of borage leaves was identified as rosmarinic acid.  相似文献

为明确小桐子(Jatropha curcas L.)果壳的化感物质,对小桐子果壳水提物的水萃取物进行了分离和初步鉴定。结果表明,AO活性部位为10 g L-1时,对萝卜(Raphanus sativus Linn.)、苏丹草[Sorghum sudanense(Piper)Stapf]幼苗生长的抑制率均高于80%。经对AO部位进行HPLC-MS分析,选择鉴定了含量较高的6个化合物,其含量为总量的60.13%。其中,化合物6初步鉴定为楤木皂苷Ⅴ(AraliasaponinⅤ),化合物1、2、3、4、5相似,初步鉴定为肽和蛋白,其相对含量之和为56.13%。利用小桐子果壳可开发植物源除草剂。  相似文献

During our studies on the components in Japanese cedar, Cryptomeria japonica, we found that the crude methanol extract of C. japonica showed intense antifeeding activity against one snail species, Acusta despesta, which is well-known as a pest of many vegetables and crops. The active components in the extract were separated into the hexane and ethyl acetate soluble fractions. From the active ethyl acetate soluble fraction, two norlignans, sequirin-C and agatharesinol, were isolated and identified as the active compounds. Both compounds inhibited feeding behavior of A. despesta at 30 microg/cm2 and 40 microg/cm2 concentrations, respectively, when applied by an eggplant leaf or filter paper containing 20 microl of 5% sucrose solution.  相似文献

Seeds of anise (Pimpinella anisum) were exposed to doses of 0, 5, 10, 15 and 20kGy in a (60)Co package irradiator. Irradiated and unirradiated samples were stored at room temperature. Microbial populations on seeds, total and inorganic soluble solids in water extract and sensory properties of the latter were evaluated after 0, 6 and 12 months of storage. Results indicated that gamma irradiation reduced the aerobic plate counts of aniseed. Immediately after irradiation, the total soluble solids in an extract of irradiated seeds were greater than those of unirradiated ones. The total soluble solids in an extract of irradiated and un-irradiated seeds increased after 6 and 12 months of storage. There were no significant differences (p>0.05) in inorganic soluble solids between the water extract of irradiated and unirradiated aniseeds. Sensory evaluation indicated that gamma irradiation improved sensory characteristics of aniseed water extract tested immediately after irradiation; however, after 12 months of storage, no significant differences (p>0.05) were found in color, taste or flavor between extract of irradiated and unirradiated seeds.  相似文献

Opuntia ficus-indica L. is known for its beneficial effects on human health, but still little is known on cladodes as a potent source of antioxidants. Here, a direct, economic and safe method was set up to obtain water extracts from Opuntia ficus-indica cladodes rich in antioxidant compounds. When human keratinocytes were pre-treated with the extract before being exposed to UVA radiations, a clear protective effect against UVA-induced stress was evidenced, as indicated by the inhibition of stress-induced processes, such as free radicals production, lipid peroxidation and GSH depletion. Moreover, a clear protective effect against apoptosis in pre-treated irradiated cells was evidenced. We found that eucomic and piscidic acids were responsible for the anti-oxidative stress action of cladode extract. In conclusion, a bioactive, safe, low-cost and high value-added extract from Opuntia cladodes was obtained to be used for skin health/protection.  相似文献

Compounds from a soil isolate of Penicillium sp. MG-11 showed remarkable convulsive activity against silkworms. Activity-based fractionation of an acetone extract of the fermentation products resulted in the isolation of four compounds. One was identified as verruculogen, which was an active principle. Another compound, acetoxydehydroaustin, was new and was structurally characterized by spectral data and a single-crystal X-ray analysis. The two other compounds were identified as dehydroaustin and austin on the basis of the spectral data. Although the latter three compounds were not active themselves, they enhanced the convulsive activity of verruculogen in a silkworm bioassay. Three additional compounds with no identifiable activity were also isolated. Two of them were identified as austinol and dehydroaustinol, and the third was determined to be a new related compound named neoaustin by an X-ray analysis of crystals.  相似文献

Three new compounds, 1 – 3 , and 20 known compounds were isolated from the AcOEt and BuOH extract of edible Opuntia Milpa Alta. The petroleum ether extract was examined by GC and MS. A total of 26 compounds were identified, representing 95.6% of the total extract, phytosterol (36.03%) being the most abundant component, and polyunsaturated fatty acids (18.57%) represented the second largest group, followed by phytol (12.28%), palmitic acid, palmitate (13.54%), vitamin E (4.51%), and other compounds (7.47%). The effects of various extracts from edible Opuntia Milpa Alta (petroleum ether extract, AcOEt extract, BuOH extract, aqueous extract, H2O parts) and the positive control (received dimethylbiguanide) were tested on streptozotocin (STZ)‐induced diabetic mice. The results indicated that all the treatment groups could significantly decrease blood glucose levels in STZ‐induced diabetic mice compared to the model control group (P<0.01), except the aqueous extract group (P<0.05). Especially, the petroleum ether extract group and the positive control group showed remarkable decrease of blood glucose levels. Taken together, the results indicate that the petroleum ether extract is the major hypoglycemic part in edible Opuntia Milpa Alta, which may be developed to a potential natural hypoglycemic functional ingredient.  相似文献

The aqueous extract of yerba mate, a South American tea beverage made from Ilex paraguariensis leaves, has demonstrated bactericidal and inhibitory activity against bacterial pathogens, including methicillin-resistant Staphylococcus aureus (MRSA). The gas chromatography-mass spectrometry (GC-MS) analysis of two unique fractions of yerba mate aqueous extract revealed 8 identifiable small molecules in those fractions with antimicrobial activity. For a more comprehensive analysis, a data analysis pipeline was assembled to prioritize compounds for antimicrobial testing against both MRSA and methicillin-sensitive S. aureus using forty-two unique fractions of the tea extract that were generated in duplicate, assayed for activity, and analyzed with GC-MS. As validation of our automated analysis, we checked our predicted active compounds for activity in literature references and used authentic standards to test for antimicrobial activity. 3,4-dihydroxybenzaldehyde showed the most antibacterial activity against MRSA at low concentrations in our bioassays. In addition, quinic acid and quercetin were identified using random forests analysis and 5-hydroxy pipecolic acid was identified using linear discriminant analysis. We also generated a ranked list of unidentified compounds that may contribute to the antimicrobial activity of yerba mate against MRSA. Here we utilized GC-MS data to implement an automated analysis that resulted in a ranked list of compounds that likely contribute to the antimicrobial activity of aqueous yerba mate extract against MRSA.  相似文献

BackgroundChromolaena odorata, has been traditionally known for its insect repellent property. Aim of this study was to determine larvicidal tendency of C. odorata on Culex quinquefasciatus and isolate compounds responsible for this activity and to determine the mechanism of action of these compounds.MethodsC. odorata plant extract was screened for mosquito larvicidal activity. The extract was fractionated using chromatography and the bioactive fraction showing larvicidal activity was identified. The chemical nature of the compounds in the bioactive fraction was determined using NMR and Mass spectrometry.ResultsWe identified phytosterols and alkanols to be the compounds regulating larvicidal activity in the bioactive fraction of the plant extract. Stigmasterol and 1-hexacosanol were identified to be the chief orchestrators of larvicidal activity and their mode of action has been observed to be neurotoxicity. At a molecular level both stigmasterol and 1-hexacosanol were found to be inhibiting acetylcholinesterase activity in C. quinquefasciatus & A. aegypti. The acetylcholinesterase inhibitory effect was validated in vitro using recombinant acetylcholinesterase and ex vivo in larval homogenates of Culex and Aedes. Electrophysiological studies using electroantennography have shown enhanced neural response to these compounds.ConclusionsNeurotoxic effect of C. odorata derived stigmasterol and 1-hexacosanol, exerted through acetylcholinesterase inhibition was responsible for the mortality of C. quinquefasciatus, A. aegypti & Chironomus riparius. EAG studies pointed out hyper-excitability of the olfactory system by these compounds.General significanceThese compounds are natural agents for mosquito control that can be used in vector control as larvicidal compounds, pending further investigations.  相似文献

为了研究蟹爪叶盾蕨(Neolepisorus ovatus(Bedd.)Ching f. doryopteris(Christ)Ching)石油醚提取物的化学组成及抗菌活性,采用气相色谱-质谱法分析蟹爪叶盾蕨石油醚提取物的成分,采用琼脂平板打孔法检测提取物对大肠杆菌、金黄色葡萄球菌、藤黄八叠球菌的抑制活性。从蟹爪叶盾蕨石油醚提取物中鉴定出38种化学成分,占出峰总数的92. 76%,其中豆甾-4-烯-3-酮(19. 83%)、何帕烯(16. 40%)、β-香树脂酮(14. 93%)等甾体和萜类化合物含量丰富。抑菌活性结果显示,蟹爪叶盾蕨石油醚提取物对藤黄八叠球菌的抑菌圈为(7. 78±0. 55)cm,对大肠杆菌、金黄色葡萄球菌、白色念珠菌没有抑制作用。所鉴定的化合物均首次从蟹爪叶盾蕨检测到,蟹爪叶盾蕨石油醚提取物对藤黄八叠球菌有抑制作用。此研究结果为该植物的开发利用提供了科学依据。  相似文献

5-(Hydroxymethyl)uracil (HMUra) is a chemically stable derivative of thymine formed through the action of ionizing radiation which we previously identified in the DNA of gamma-irradiated HeLa cells [Teebor, G. W., Frenkel, K., & Goldstein, M. S. (1984) Proc. Natl. Acad. Sci. U.S.A. 81, 318-321]. In this report, we determine whether HMUra can be used as a marker of exposure of DNA to ionizing radiation. Dose-response curves for its formation in [3H]thymidine-labeled DNA were constructed by exposing the DNA to increasing amounts of gamma-radiation and measuring the HMUra content. DNA was irradiated both in solution and in intact cells. HMUra was identified as the 2'-deoxyribonucleoside 5-(hydroxymethyl)-2'-deoxyuridine (HMdU) by subjecting the irradiated DNA to enzymatic digestion and analyzing the mixture of 2'-deoxyribonucleosides by high-pressure liquid chromatography. The identity of the radiogenically formed HMdU was confirmed by acetylation and the structure of the acetyl derivative obtained by mass and nuclear magnetic resonance spectroscopies. At two different DNA concentrations in solution, the same number of thymidine moieties were converted to HMdU, indicating that within this range of concentration the formation of HMdU was mediated through the indirect action of ionizing radiation. Equal amounts of HMdU were formed in single- and double-stranded DNA at each radiation dose, indicating that DNA conformation did not affect HMdU formation. Surprisingly, the G value (number of HMdU molecules formed/100 eV) was higher in irradiated cellular DNA than in DNA irradiated in solution.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The composition of the volatile fraction from Cochlospermum tinctorium rhizome was investigated by GC and GC-MS; among 11 constituents detected, eight were identified as straight chain ketonic compounds.
