(R)-β-acetylmercaptoisobutyric acid (RAM), a chiral compound, is an important intermediate for the chemical synthesis of various antihypertensive and congestive heart failure drugs. Microorganisms capable of converting (R,S)-β-acetylmercaptoisobutyric acid ((R,S)-ester) to RAM were screened from soil microorganisms. A strain ofPseudomonas sp. 1001 screened from a soil sample was selected to be the best. Cells showed an activity of 540 U/mL from culture broth and the enzyme was thermostable up to 70°C. This strain could produce RAM asymmetrically from (R,S)-ester.  相似文献

Crystalline aromatic l-amino acid decarboxylase from Micrococcus percitreus is inactive in the absence of pyridoxal phosphate (PLP). The inactive form of the enzyme shows absorption at 340 nm and contains one mol of PLP per mol of enzyme. Binding of PLP to the inactive form is accompanied by a pronounced increase in absorbance at 415 nm. The amount of PLP that binds to this holoenzyme is 2 mol per mol of enzyme. The inactive half-resolved form, i. e. semiapoenzyme, is obtained again by dialysis of the holoenzyme against phosphate buffer. When the semiapoenzyme is dialyzed against phosphate buffer containing 3,4-dihydroxyphenyl-l-alanine, it loses the absorption at 340 nm with the loss of PLP. This apoenzyme regains the activity and absorption at 340 nm and 415 nm on association with PLP.  相似文献

The effects of 1α,25-dihydroxyvitamin D3, 24R,25-dihydroxyvitamin D3 and 1α,24R,25-trihydroxyvitamin D3 on active calcium and phosphate transport by rat duodenum were studied in vitamin D-deficient rats that either underwent sham surgery or were bilaterally nephrectomized. Both 1α, 25-dihydroxy- and 1α,24R,25-trihydroxyvitamin D3 markedly stimulated calcium and phosphate absorption with similar effects in shamoperated and nephrectomized rats. A 10-fold higher dose of 24R,25-dihydroxyvitamin D3 was required for an equivalent stimulation of absorption in sham-operated rats, and this compound had no effect on duodena from nephrectomized rats. These data provide the first evidence that 24R,25-dihydroxy- and 1α,24R,25-trihydroxyvitamin D3 can stimulate the active intestinal absorption of phosphate. The lack of response to 24R,25-dihydroxyvitamin D3 in nephrectomized rats confirms prior results which indicated that renal metabolism of this secosteroid to 1α,24,25-trihydroxyvitamin D3 is required for biological activity. In addition, we describe a simple bioassay technique which apparently reflects, with reasonable accuracy, the changes in duodenal calcium and phosphate absorption which occur under more rigorous short-circuited conditions and, in particular, can be used for screening putative 1α-hydroxyl analogs of vitamin D in nephrectomized rats.  相似文献

(22R,23R,24S)-22,23-Isopropylidenedioxy-5α-ergost-2-en-6-one 2b is an important intermediate of brassinolide. We found that the enone 2b can be prepared by transformation of (22R,23R,24S)-3α,5-cyclo-22,23-isopropylidenedioxy-5α-ergostan-6-one 5b with catalytic amount of both p-TsOH and NaBr in DMF under reflux. 5b was prepared from (22R,23R,24S)-3α,5-cyclo-22,23-dihydroxy-6β-methoxy-5α-ergostane 9b or a 6β-benzyloxy compound 9c, which was obtained in a manner similar to Mori’s brassinolide synthesis. The enone 2b was eventually prepared via a benzyl ether 9c from stigmasterol 3a in a 15.5% yield in 11 steps.  相似文献

Terpene derivatives converted by microbial biotransformation constitute an important resource for natural pharmaceutical, fragrance, and aroma substances. In the present study, the monoterpene α-phellandrene was biotransformed by 16 different strains of microorganisms (bacteria, fungi, and yeasts). The transformation metabolites were initially screened by TLC and GC/MS, and then further characterized by NMR spectroscopic techniques. Among the six metabolites characterized, 6-hydroxypiperitone, α-phellandrene epoxide, cis-p-menth-2-en-1-ol, and carvotanacetone, which originated from (-)-(R)-α-phellandrene, are reported for the first time in this study. Additionally, the substrate and the metabolite 5-p-menthene-1,2-diol were subjected to in vitro antibacterial and anticandidal tests. The metabolite showed moderate-to-good inhibitory activities (MICs=0.125 to >4?mg/ml) against various bacteria and especially against Candida species in comparison with its substrate (-)-(R)-α-phellandrene and standard antimicrobial agents.  相似文献

[目的]构建能够专一性合成光学纯(R,R)-2,3-丁二醇的大肠杆菌工程菌,并进行发酵条件优化。[方法]将来源于多粘芽孢杆菌的(R,R)-2,3-丁二醇脱氢酶基因bdh,来源于阴沟肠杆菌的α-乙酰乳酸合成酶基因bud B和α-乙酰乳酸脱羧酶基因bud A与表达载体p Tr C99A连接,导入大肠杆菌中构建人工合成途径。筛选最适的培养基和发酵条件,提高(R,R)-2,3-丁二醇的产量、产率和得率。[结果]获得高效合成(R,R)-2,3-丁二醇的工程菌株GXASB,筛选到最适碳源及其浓度为120 g/L木薯淀粉,最适pH为6.5,最适接种量为10%,在发酵罐中进行同步糖化法发酵,(R,R)-2,3-丁二醇产量达到105.28 g/L,光学纯为99.1%,得率为0.47 g/g,生产强度为1.95 g/(L·h)。[结论]在大肠杆菌中表达基因簇bud B-bud A-bdh能够专一性合成光学纯(R,R)-2,3-丁二醇,经优化发酵条件后,能够显著提高(R,R)-2,3-丁二醇的合成效率。同时工程菌能够利用非粮原料木薯淀粉高效生产(R,R)-2,3-丁二醇,补料发酵产量达到105.28 g/L,为使用廉价原料工业化生产(R,R)-2,3-丁二醇提供参考。  相似文献

(R,R)-2,3-BD是一种重要的四碳平台化合物,在液晶材料、高附加值手性化合物,尤其是不对称合成光学纯药物等方面有天然优势.将来源于多粘芽孢杆菌(Paenibacillus polymyxa)DSM 365的α-乙酰乳酸合成酶(α-acetolactate synthase)基因 alsS、α-乙酰乳酸脱羧酶(α-acetolactate decarboxylase)基因alsD和(R,R)-2,3-丁二醇脱氢酶(2,3-butanediol dehydrogenase)基因R,R-bdh与表达载体pMA5连接,导入多粘芽孢杆菌P.polymyxa DSM 365中加强(R,R)-2,3-丁二醇的主代谢途径,构建可高效合成(R,R)-2,3-丁二醇的多粘芽孢杆菌工程菌株DM-5.利用工程菌株DM-5补料分批发酵60 h,(R,R)-2,3-丁二醇产量达54.91 g/L,得率为0.52 g/g,生产强度为0.92 g·L-1·h-1,与野生菌株相比(R,R)-2,3-丁二醇产量增加19.66%,且副产物甲醇浓度不变,乙醇、乙偶姻积累下降.本研究结果表明,在多粘芽孢杆菌中过量表达关键基因alsS、alsD和R,R-bdh 能够显著提高(R,R)-2,3-丁二醇的产量和生产强度,为多粘芽孢杆菌的代谢工程改造和工业化生产(R,R)-2,3-丁二醇提供参考.  相似文献

The cerebrosides were first isolated by Thudicum in 1874 and the structures were established by Carteret al. in 1950 (for review, see [2]). In 1961 Shapiro and Flowers [3] reported the first total synthesis of a cerebroside1 (Fig. 1) which was identified with the natural sample, only through comparison of their i.r. data. In order to confirm the absolute configuration at C-2 of natural cerebroside1, we describe here an unambiguous synthesis of two stereoisomeric cerebrosides1 and2, and found that the1H-NMR spectra of the synthetic1 (Fig. 2) was completely identical with that of the natural cerebroside reported recently by Dabrowskiet al. Total syntheses of both (2S, 3R, 4E)-1-O-β-d-galactopyranosyl-N-(2′R)-2′-hydroxytetracosanoylsphingenine 23 and the (2′S) stereoisomer were performed in an unambiguous way by employing either (2S, 3R, 4E)-N-(2′R)-2′-(tert-butyl-diphenylsilyloxy)tetracosanoylsphingenine or its (2′S) stereoisomer as the key glycosyl acceptors. The synthetic cerebroside 23 was shown to be identical with the natural product through comparison of their 400-MHz, 1H-n.m.r. spectra, thus providing synthetic evidence for the 2′R configuration of the natural cerebroside.  相似文献

We have performed an extensive analysis of Th1/Th2 cytokine receptors IL2Rα, IL4Rα, IL10Rα, and IFNγR1 gene polymorphisms to evaluate their impact on AIDS progression. The coding regions and promoters of these genes were sequenced in the genetics of resistance to immunodeficiency virus cohort, composed of 327 HIV-1-positive patients with extreme progression phenotypes, slow and rapid progressors, and of 446 healthy control subjects, all of them of Caucasian descent. Overall, 104 single nucleotide polymorphisms and four insertions/deletions with a minor allelic frequency higher than 1% were identified, 21 of them being newly characterized. We observed weak associations for 13 polymorphisms of IL2Rα, IL4Rα, IL10Rα, and IFNγR1, and 11 haplotypes of IL2Rα, IL4Rα, and IFNγR1. However, we could not relate these positive signals to any relevant biological information on the gene function. To affirm these putative associations in AIDS, further confirmation on other AIDS cohorts will be needed. This complete catalog of polymorphisms in IL2Rα, IL4Rα, IL10Rα, and IFNγR1 cytokine receptor genes should also be useful for investigating associations in other immune-related diseases.Electronic Supplementary Material Supplementary material is available for this article at  相似文献

Assessing population density is crucial for studying the ecology and evolutionary biology of species as well as for conservation purposes. Here we used point count methods to infer population density in a single-island endemic passerine bird, the Réunion Grey White-eye Zosterops borbonicus, that displays striking evidence of differentiation at a small spatial scale. Population density was estimated at 5.17 birds ha?1 (CL 4.85–5.50), a value somewhat higher than previously believed. This estimation provides the first detailed estimation of bird population density in the vulnerable summit ecosystems of Réunion and will possibly allow a better understanding of the evolutionary causes of this plumage colour variation.  相似文献

Twelve amino acids and amides at 0·1 to 0·75 or 1·0% in 35% sucrose solution were individually tested for their rôle in phagostimulation, growth, and survival in Acyrthosiphon pisum. Leucine and phenylalanine were phagostimulatory at all concentrations tested, tryptophan and valine at 0·1, 0·2, and 0·5%, and threonine at 0·1% only. Methionine was reported earlier by us to be phagostimulatory at 0·05 to 0·5%. Histidine and isoleucine had no effect, whereas arginine and lysine HCl reduced uptake when compared to sucrose alone. The non-essential amino acids, canavanine sulphate and glutamine, reduced uptake at all concentrations, whereas homoserine was phagostimulatory at 0·1 and 0·75%.Arginine, canavanine sulphate, glutamine, histidine, homoserine, isoleucine, leucine, and valine increased weight and prolonged survival, whereas lysine HCl, phenylalanine, threonine, and tryptophan neither promoted growth nor increased survival. Radioactive leucine (14C(U)) was incorporated into the protein fraction of the larval body and exuviae indicating that it took part in protein synthesis. This seems to be the first report in insects where peptide or protein synthesis occurred from single amino acids in sucrose.  相似文献

Studies of Río Negro subsistence farming and fishing activities are used to estimate the human carrying capacity for the region and the likely pattern of human land-use during prehistory. Ceramic evidence suggests human presence in the region more than 3000 years ago. Traditional farming is labor intensive and relatively unproductive. Nevertheless, farmers achieve an energy return of 15.21, and produce 2600 kcal per work hour. Fish are the major protein source, but fish catch per unit of effort and fish yield per hectare of floodplain are very low; fishermen are probably exploiting local fish resources very close to their limit. The low human population density would suggest that the Río Negro forest has been relatively undisturbed. Nevertheless, charcoal is widespread and abundant in forest soils. This charcoal is probably from anthropogenic or natural wildfires. These results suggest a much more complex history for Amazonia than previously thought.K. The rôle of the midgut, crop, and maxillae in the production and utilization of the cocoon-digesting enzyme was investigated in the silkworm, Bombyx mori.About a sixtyfold purified preparation of midgut protease was obtained by ammonium sulphate precipitation and column chromatography.Immunological studies by the agar diffusion method of Ouchterlony revealed that the crop and midgut proteases of the pharate adult are antigenically identical whereas that of the maxillary protease is different.From the results of extirpation experiments and previous studies it was shown that the midgut, crop, and maxillae play important rôles in the escape of moths from their cocoons.  相似文献

A phytochemical study on the alkaloids from Teclea nobilis yielded five furoquinolines, including the new acetylmontrifoline. The structures were elucidated from extensive 2D-NMR and MS experiments. The absolute configuration of the chiral center of montrifoline and of acetylmontrifoline was established as R by analysing the Mosher's derivatives and chemical modifications, respectively. All the five furoquinolines showed weak in vitro antiplasmodial activity against Plasmodium falciparum, montrifoline being moderately active (56 μg/ml) and not cytotoxic.  相似文献

Twelve taxa belonging to two subsections of section Idaeobatus in Rubus L. from southwestern China were characterized by karyotypic, palynological, and random amplified polymorphic DNA (RAPD) data as follows: (1) The 12 taxa were all diploid species (2n = 2x = 14), among which the chromosome counts for R. mesogaeus var. oxycomus, R. subtibetanus, R. ellipticus var. obcordatus, R. inopertus var. echinocalyx, and R. stans were reported for the first time; (2) All taxa except for R. ellipticus and R. pinfaensis could be distinguished from each other by karyotype, pollen morphology, and RAPD markers. Karyotypes were mainly characterized by the difference in numbers and positions of submetacentric chromosomes and chromosomes with satellited pair, the index of the karyotypic asymmetry, and the ratio of the longest to the shortest chromosome. Pollen morphology were mainly characterized by the discrepancy in specific pollen size, P/E ratio, colpi width, distance between the apices of two ectocolpi, and exine ornamentation characters. The cluster results based on RAPD markers were consistent with morphology classification except for R. pinfaensis; (3) Based on the general data of karyotypic, palynological, and RAPD, R. ellipticus var. obcordatus should be treated as a species R. obcordatus, R. ellipticus and R. pinfaensis should be combined as R. ellipticus, and it was more reasonable to place the combinants and R. obcordatus into subsection Stimulantes rather than into subsection Pungentes.  相似文献
