Male rats weanling fed 20% gluten diet for 90 days of demonstrate to have about one-third lower level of carnitine per gram of skeletal muscle and heart muscle than the group which received the same diet supplemented with 1% of lysine. The carnitine level of the liver, however, was significantly higher in the unsupplemented versus the lysine supplemented groups.  相似文献

The influence of protein quality on the growth-depressing effect of excessive amount of 12 individual essential and semiessential amino acids was examined. Growing rats were fed for 3 weeks diets containing either 10.5% egg albumin or 11.6% wheat gluten (equivalent to the protein content of a 10% casein diet) supplemented with 5% of each of the l-amino acids. In general, the pattern of growth depression produced by the addition of excess amino acids to the egg albumin or the wheat gluten diet was similar to that of the case of casein diet obtained previously under the same experimental conditions. However, the extent of these effects was dependent not only upon the kind of amino acid supplemented with but also upon the source of protein used, and the depressing effect of each of excess amino acids added to the wheat gluten diet was usually severer than those added to casein and egg albumin diets. No evidence was noted of any striking changes in the liver protein and nucleic acid concentrations by either diets, but total liver protein, RNA and DNA contents were decreased in some amino acid groups of the egg albumin diet and in all amino acid groups of the wheat gluten diet except the lysine addition. The free amino acid level in plasma generally showed extreme elevation for the amino acid supplemented in excess in the diet, and in most cases the extent of the elevation was correlated with the growth depression.  相似文献

The metabolic fates of the carbon skeletons of leucine, lysine, and threonine were studied in growing rats on the diets containing graded levels of protein calorie percentages (10, 20, 30, and 40PC%) by use of either gluten or zein at 4100 kcal of metabolizable energy per kg of diets. In growth experiment for 21 days, body weight gain, food intake, and body fat increased at higher PC% in the gluten diets, but rats given zein did not maintain their initial weight even at 40PC%. The concentration of plasma free lysine remained low with the zein diets, but plasma threonine increased at 10 and 20PC% in the gluten and zein diets, respectively. Plasma leucine increased as the protein level increased either dietary protein. More than 70% of 14C was incorporated into body protein 12 h after an intraperitoneal injection of labeled lysine in all groups, but little 14CO2 was expired in rats on the gluten and zein diets. About 79% of 14C-threonine was incorporated into body protein in rats given the gluten and zein diets at 10PC%, but the values were gradually decreased with increasing the dietary protein levels. Some 40–50% of 14C-leucine was incorporated into the body protein in rats given the gluten diets, and the values for the zein diets were extensively decreased in the higher PC% groups where the expired 14CO2 was inversely increased to a great extent. These results showed that, when a specific amino acid was limiting or deficient in the diet, the major portion of the labeled amino acid was utilized for body protein synthesis and little was oxidized to carbon dioxide, whereas the oxidative degradation of essential amino acid other than limiting one was increased and the efficiency of the amino acid utilization was relatively decreased.  相似文献

Cadmium is an environmental pollutant of increasing worldwide concern. It has been reported to be high in the soil where food crops are grown in some parishes of Jamaica. Surprisingly, no adverse effect of cadmium has been reported among the Jamaican population. However, phytic acid has also been shown to be high in some food crops grown in Jamaica. In this study, we evaluated the effects of phytic acid (1 %) and exercise on the metabolism of cadmium (5 mg cadmium/kg body weight) in rats. Five groups of rats were fed as follows: rats fed control diet, control diet supplemented with cadmium and subjected to exercise, control diet supplemented with phytic acid plus cadmium and subjected to exercise, control diet supplemented with cadmium plus phytic acid, and control diet supplemented with cadmium only. The animals were fed for 4 weeks and then sacrificed. Blood samples were collected for some biochemical assays. Percentage weight loss (28.42 %) was greatest in the group that had cadmium supplement only. The group fed control diet supplemented with cadmium only displayed increased liver enzymes and electrolytes except for the significant decrease in bicarbonate compared to other test groups. Similarly, blood urea nitrogen and uric acid were increased in the group fed cadmium supplement only compared to other test groups. Total cholesterol trended downwards in the test groups compared to control. These observations suggest that consumption of diet high in phytic acid with relatively high physical activity may be protective against the adverse effects of cadmium.  相似文献

Two separate experiments were conducted to determine the dietary requirements of juvenile Asian sea bass Lates calcarifer Bloch for lysine and arginine. Fish (average initial weight: lysine experiment, 13.12 ± 0.12 g; arginine experiment, 2.56 ± 0.13 g) were given amino acid test diets for 12 weeks containing fish meal, zein, squid meal, and crystalline amino acids. Each set of isonitrogenous and isocaloric test diets contained graded levels of L ‐lysine or L ‐arginine. The feeding rate in the lysine experiment was at 4–2.5% of the body weight day?1, while in the arginine experiment it was at 10–4% of the body weight day?1. The fish (20 per tank, lysine experiment; 15 per tank, arginine experiment) were reared in 500‐L fibreglass tanks with continuous flowthrough sea water at 27 °C and salinity of 31 ppt in the lysine experiment and at 29 °C and salinity of 29 ppt in the arginine experiment. The experiments were in a completely randomized design with two replicates per treatment. Survival was high in fish given adequate lysine or arginine. Mean percentage weight gains were significantly different in fish fed varying levels of lysine or arginine. Fish fed high levels of L ‐arginine suffered high mortalities. No significant differences were obtained in the feed efficiency ratios (FER, g gain g?1 feed) of fish fed graded lysine, although the values tended to increase as the dietary lysine level was increased up to the requirement level. In contrast, in the arginine experiment, significant differences in FER of fish among treatments were obtained; the highest FER was observed in fish fed the diet containing an optimum arginine level. On the basis of the growth response, survival, and FER, the lysine and arginine requirements of juvenile Asian sea bass were estimated to be 20.6 g kg?1 dry diet (4.5% protein) and 18.2 g kg?1 dry diet (3.8% protein), respectively. These data will be useful in the further refinement of practical diet formulations for the Asian sea bass.  相似文献

In experiment 1, weanling rats were given, for 7 weeks, a commercial rat diet supplemented with hydrochloric acid at levels up to 560 mmol.kg-1 dry matter. The supplement increased water intake but did not significantly affect food intake, live-weight gain, blood haemoglobin and haematocrit values or acid-base balance. In experiment 2, adult rats were given, for 9 weeks, a commercial rat diet supplemented with hydrochloric acid at levels up to 1250 mmol.kg-1 dry matter. Food intake and liveweight gain were not affected by hydrochloric acid concentration up to 625 mmole but at 938 mmol.kg-1 they were considerably reduced and there was 100% mortality of the rats. In experiment 3, weanling rats were given, for 12 weeks, a commercial rat diet supplemented with hydrochloric or lactic acid each at 300, 600 and 900 mmol.kg-1 dry matter. Lactic acid at the three levels and hydrochloric acid at the two lower levels did not affect food intake or live weight gain and had only a slight effect on blood acid-base balance. At a dietary concentration of 900 mmol.kg-1 dry matter, hydrochloric acid decreased food intake, induced a mild degree of metabolic acidosis and resulted in 30% mortality of the rats. In the three experiments, the acid treatments dnot directly affect the length or composition of the femur of the rats.  相似文献

A 10-week feeding trial was conducted to study the effect of feeding level and dietary lysine concentration on growth, protein and lysine retention, and body composition in juvenile turbot. Maintenance requirement for lysine and the efficiency of lysine utilisation were determined as well. Two experimental diets were formulated based on fishmeal or wheat gluten as main protein sources, containing 6.4 g (Diet A, control) and 4.5 g lysine per 100 g CP (Diet B), respectively. Diets were fed once daily at six feeding levels (per day 0.3%, 0.6%, 0.9%, 1.2%, and 1.5% of body weight [BW] and ad libitum) to a total of 432 fish of 48 g initial BW. No differences in the growth parameters were observed between diets at the same feeding level, except a lower feed to gain ratio (p < 0.05) at the highest feeding level at Diet B. Whole-body composition was not affected by diet, whereas muscle protein concentration was significantly lower for fish fed Diet B. Amino acid concentration in whole-body protein was affected by dietary treatment and fish fed Diet B showed lower concentrations of all essential amino acids. In fish muscle protein, lysine, methionine, leucine, isoleucine, and valine concentrations were significantly lower in Diet B. Efficiency of lysine utilisation for growth (klys) was determined by linear regression analysis and amounted for 0.69 for Diet B. The maintenance lysine requirement defined at zero lysine retention was 6.5 mg · kg?0.8 · d?1. Lysine intakes at zero protein retention were 13.0 mg and 12.9 mg · kg?0.8 · d?1 for Diet A and B, respectively. Growth and nutrient retention were similar for both diets and, therefore, a lysine deficiency in Diet B did not occur. In conclusion, a proportion of 330 g wheat gluten per kg feed did not influence growth performance and maintenance requirement for lysine in juvenile turbot. However, the effect of diet composition on the amino acid profile of body protein might be relevant for the derivation of the amino acid requirement from protein retention.  相似文献

Faba bean, lentil or chick-pea raw seeds were used as the only protein food in the rat. The balanced diet was designed to suit growth requirements. Nitrogen apparent digestibility coefficient (CUDa) was 72% in the faba bean, 75% in the lentil, 76% in the chick-pea groups respectively, but the CUDa of some essential amino acids were much lower: 52-51-71% for methionine, 60-39-78% for cystine, 73-71-75% for valine, while arginine CUDa values (87-87-82) were higher than all other AA CUDa values. Growth rate (8.9-6.7-9.1 g/d) and food intake index (2.1-2.5-2.2) were satisfactory. Blood and muscle free amino acid concentrations related to digestible intake showed that methionine from the 3 legumes, threonine from chick-pea, arginine and lysine from faba bean were not fully available.  相似文献

The relationship between changes in blood plasma amino acids and the quantity of protein and energy self-selected by the weanling rat, simultaneously offered two diets varying only in gluten (15 and 55%) concentration, was examined. Gluten and energy intakes were manipulated by additions of lysine, arginine or ammonia to gluten. In two experiments groups of ten weanling rats were fed the diets for a two week or four week period and food intake selection recorded. Blood samples were obtained between 0900–1100 hr at the end of the two week or four week period. Correlation coefficients of protein intake with the plasma TRP/NAA (Tyr+Phe+Leu+Val+Ile) ratios were ?0.97 and ?0.98, and of energy intake with TYR/PHE ratios were 0.77 and 0.70 in experiments 1 and 2, respectively. It is suggested that the plasma TRP/NAA and TYR/PHE ratios reflect the mechanisms regulating protein and energy intakes, respectively.  相似文献

Compromisation of food intake when confronted with diets deficient in essential amino acids is a common response of fish and other animals, but the underlying physiological factors are poorly understood. We hypothesize that oxygen consumption of fish is a possible physiological factor constraining food intake. To verify, we assessed the food intake and oxygen consumption of rainbow trout fed to satiation with diets which differed in essential amino acid (methionine and lysine) compositions: a balanced vs. an imbalanced amino acid diet. Both diets were tested at two water oxygen levels: hypoxia vs. normoxia. Trout consumed 29% less food under hypoxia compared to normoxia (p<0.001). Under both hypoxia and normoxia trout significantly reduced food intake by 11% and 16% respectively when fed the imbalanced compared to the balanced amino acid diet. Oxygen consumption of the trout per unit body mass remained identical for both diet groups not only under hypoxia but also under normoxia (p>0.05). This difference in food intake between diets under normoxia together with the identical oxygen consumption supports the hypothesis that food intake in fish can be constrained by a set-point value of oxygen consumption, as seen here on a six-week time scale.  相似文献

The purpose of this study was to find whether the addition of dietary lysine affected the rate of brain protein synthesis in aged rats fed on a gluten diet. Experiments were done on two groups of aged rats (30 wk) given the diets containing 5% gluten or 5% gluten + 0.3% lysine for 10 d. The fractional rates of protein synthesis in brain, liver, and kidney increased with an addition of dietary lysine. In brain, liver, and kidney, the RNA activity [g protein synthesized/(g RNA x d)] was significantly correlated with the fractional rate of protein synthesis. The RNA concentration (mg RNA/g protein) was not related to the fractional rate of protein synthesis in any organ. The results suggest that the addition of the limiting amino acid for the low quality protein elevates the rate of protein synthesis in the brain of aged rats, and that RNA activity is at least partly related to the fractional rate of brain protein synthesis.  相似文献

Growth rates and efficiency of food conversion of young chicks fed on diets marginally limiting in total nitrogen and containing 150 g/kg diet of flash-dried microbial cells (MC) with and without amino acid supplements, were measured in three experiments. Performance of these animals was compared with that of groups fed on a methioninefortified soya bean meal (SBM) control diet. In all experiments, chicks fed on the SBM control diet grew faster and were more efficient than chicks fed on the basal unsupplemented MC diet. In Experiment 1, arginine supplementation markedly enhanced weight gain and efficiency of food utilisation of chicks offered the basal MC diet; methionine had no effect. The second experiment demonstrated that a supplement containing methionine, arginine and tryptophan was more effective in augmenting the nutritional value of MC than either methionine with arginine or tryptophan with arginine. In the final experiment, weight gain and food intake of chicks fed on MC with supplements of arginine, methionine and tryptophan were increased markedly by additions of lysine and glutamic acid but not by addition of lysine alone. In all experiments, performance of animals in MC supplemented groups was lower than that of animals fed on the SBM control diet.  相似文献

Diets were computed to contain equal concentrations of digestible crude protein either of wheat gluten (diet 1) or of grieves (diets 2–8). Per kg dry diet, 41 g crystalline amino acids were supplemented. All diets contained at least 1.2 g Lys per MJ digestible energy (DE). In diet 2, ratios of Met + Cys, Trp, Leu, Ile and Phe to Lys were about equal to those in diet 1. In each of diets 3–7, one of the respective amino acids, in diet 8 all five were replaced by Glu in the supplemented mixture of amino acids.

Each diet was fed to triplciate groups of 20 trout during a trial lasting 66 days. Trout fed the diet containing wheat gluten consumed more dry matter and showed higher growth rates as well as higher protein contents in their gained body mass than trout fed diets based on grieves. Supplementing Met plus Trp significantly improved dry matter intake, growth rate and protein content of gain, though not to the level of trout fed the wheat gluten diet, whereas Leu, Ile and Phe showed no such effect. When grieves were not supplemented with both Met and Trp, gain in body mass contained significantly more lipids. DE required per kg gain by trout fed wheat gluten, grieves + Met + Trp or grieves without supplementation of Met and Trp was 20.1, 21.2 and 29.9 MJ, respectively.  相似文献

Although many studies have shown that amino acid ingestion acutely stimulates protein anabolism, only few studies have investigated whether long-term supplementation promotes changes in body composition. We therefore tested the hypothesis that l-leucine (LEU) and l-phenylalanine (PHE) supplementation might have a positive impact on the body composition of rats submitted to intermittent periods of food restriction and refeeding (weight cycling or WC). The WC protocol comprised three cycles, each consisting of 1 week of 50% food restriction followed by 2 weeks of ad libitum ingestion. The groups submitted to WC ingested the control diet (WC-CON) or the diet supplemented with LEU+PHE (WC-AA). A pair-fed group receiving the control diet (PF-CON) was used as a reference for the effects of WC. Although food intake was the same in all groups, higher body weight and energy efficiency were observed in the WC-AA group compared to the PF-CON and WC-CON groups although not significantly in relation to the latter. These results were the consequence of a significant increase of lean body mass and body protein content in the WC-AA group compared to the PF-CON and WC-CON groups. The WC-CON and WC-AA groups presented 36.1% and 18.9% more body fat, respectively, than the PF-CON group but this difference was not significant. Neither fasting insulin nor glucose concentration nor postprandial insulin secretion was significantly affected by the supplemented diet. In conclusion, supplementation with LEU+PHE improved the body composition profile of rats submitted to WC, mainly by increasing lean body mass and body protein content.  相似文献

Using mounting casein and wheat gluten protein values (0-40%) in the animals' diet, the optimum and minimum physiological daily doses were determined in 49-day-old growing rats from changes in their body water, body nitrogen and protein intake. The optimum physiological doses were identical with the peak of linearity of the given parameters, which coincided with a 15% casein protein and a 20% gluten protein concentration in the diet. This was also confirmed by the maximum body amino acid values, which were found in animals given a 15% casein or 20% gluten protein diet. It was further confirmed by the finding of significantly elevated alanine aminotransferase and aspartate aminotransferase activity in the liver of animals with a higher intake of the above protein sources. The minimum physiological dose of the given protein was determined from the equations of the regression curves in the presence of zero changes in the body nitrogen or body water content. The optimum physiological daily doses of casein and wheat gluten protein were 3.25 g and 4.05 g respectively. The minimum physiological daily doses of casein protein were 268 mg (from body nitrogen changes) and 371 mg (from body water changes) and the minimum physiological daily doses of gluten protein were 892 mg (from body nitrogen changes) and 1,000 mg (from body water changes). The above indicators demonstrate, in the presence of higher and high dietary concentrations, that an intake of the given proteins over and above the optimum physiological daily dose is at the very least uneconomical (gluten), if not harmful (casein), making this a highly topical problem for further study.  相似文献

Objective: Food intake is known to be affected by macronutrient composition of the diet, and protein manipulation has been reported to alter food intake, but the effect of individual amino acids on eating behavior has not been fully studied. This study investigated the effect of diet supplementation with three individual amino acids on meal pattern in male rats. Research Methods and Procedures: Thirty‐two Sprague‐Dawley rats were randomly divided into four equal groups and fed control diet or histidine (5%)‐, leucine (5%)‐, or tyrosine (5%)‐supplemented diet for 2 weeks and were monitored for their meal pattern. Results: Total food intake and feeding rate of the different groups were not affected, although other components of meal pattern were altered. Histidine supplementation reduced diurnal meal size by 42% (p < 0.05), whereas that of leucine increased nocturnal meal size by ~35% (p < 0.05). Tyrosine supplementation increased food intake of the nocturnal period and decreased that of the diurnal period. Both histidine and tyrosine supplementation elevated fasting plasma insulin levels and suppressed fasting glucose significantly. Discussion: Individual amino acids were found to alter meal pattern differently. Further investigations are required to dissect the involvement of central and peripheral factors in these alterations.  相似文献

The objective of this study was to evaluate effects of dietary l-lysine on the intestinal mucosa and expression of cationic amino acid transporters (CAT) in weaned piglets. Twenty-eight piglets weaned at 21 days of age (Duroc × Landrace × Yorkshire; 6.51 ± 0.65 kg body weight) were assigned randomly into one of the four groups: Zein + LYS (zein-based diet + 1.35 % supplemental lysine), Zein ? LYS (zein-based diet), NF (nitrogen-free diet), and CON (basal diet). The experiment lasted for 3 weeks, during which food intake and body weight were recorded. At the end of the trial, blood was collected from the jugular vein of all pigs, followed by their euthanasia. Dietary supplementation with lysine enhanced villus height and crypt depth in the jejunum (P < 0.05). Jejunal mRNA levels for the b0,+-AT, y+LAT1 and CAT1 genes were greater (P < 0.05) in the Zein + LYS group than in the control, and the opposite was observed for CAT1. Dietary content of lysine differentially affected intestinal CAT expression to modulate absorption of lysine and other basic amino acids. Thus, transport of these nutrients is a key regulatory step in utilization of dietary protein by growing pigs and lysine in the diet influences the expression of amino acid transporters in the small intestine.  相似文献

When weanling male rats were fed amino acid diets which were gradedly varied tryptophan level only (0.18, 0.09 and 0.06%), allantoin and urea excretions were determined, and (allantoin/urea) × protein intake values were calculated. (Allantoin/urea) × protein intake values almost linearly increased with the increment of tryptophan level of the amino acid mixture. From these data, it was assumed that one of the main factors affecting these ratios is in amino acid balance in diet. It was also found that the equivalent weight gains observed between 0.18 and 0.09% Try. groups were mainly due to body fat accumulation of the latter.  相似文献

Eleven Poll Dorset times Merino crossbred female lambs 4 weeks of age were trained to suck liquid diets from bottles. In three separate experiments liquid diets providing 14-2% (expt 1) 10-6% (expt 2) or 8-0% (expt 3) of gross energy as protein and amino acids were fed. Responses in voluntary intake, growth rate and changes in plasma amino acid concentrations were studied when complete or incomplete mixtures of amino acids were added to the liquid diet. These mixtures supplied either: (1) all amino acids in quantities to bring the total of protein plus amino acids to provide more than 20% of dietary gross energy, the amino acids being provided in proportions estimated to meet adequately the lamb's requirements ('complete'); or (2) as the same total amount of amino acids but with the amino acid supplement devoid of threonine ('low-threonine', expts 1 and 2) or isoleucine ('low isoleucine', expt 3). In experiment 1, there was no food intake or growth depression after feeding the amino acid mixture lacking threonine. In both experiments 2 and 3, voluntary food intake was depressed to about 50% of that observed in lambs fed the low protein diet, when the amino acid mixture devoid of threonine or of isoleucine, respectively, was fed. Addition of the missing amino acid to the low threonine and low isoleucine diets resulted in recovery of voluntary intake in experiments 2 and 3 respectively, but no significant improvement above that found after feeding the low protein (basal) diet. In experiments 1 and 2, after feeding the low threonine diet the threonine concentration in the blood plasma decreased markedly, while concentrations of total amino acids were elevated. Although there was no improvement in growth or food intake, the feeding of the diet containing the complete amino acid mixture resulted in an elevation of all essential amino acids including threonin. Similarly in experiment 3, plasma isoleucine concentration decreased in the lambs fed the isoleucine imbalanced diet. Results indicate that the suckled, preruminant lamb exhibits sensitivity to dietary amino acid imbalance, in a manner analogous to that found in simple-stomached animals. These results also clearly illustrate a depression in food intake associated with the deletion of a specific essential nutrient from the diet of the lamb.  相似文献

Eight groups of 13-15 female rats were fed purified diets after littering. Four groups received a low protein (8% casein) diet (groups 8) and the others, a normal protein (20% casein) diet (groups 20). Carbohydrates were supplied either as starch (groups S) or as starch plus 40% fructose (groups F). Half the animals received a 0.4% methionine supplementation (groups M). Four or five dams per group were sacrificed on days 2, 7 and 14 after littering. The diet intake was increased by methionine supplementation, substitution of starch for fructose and increased protein content, mainly during the second week of lactation. This influenced weight variation of the dams and litter growth. On all days, the plasma levels of cholesterol esters, triglycerides and phospholipids were positively correlated with the dietary protein level. On days 7 and 14, the liver neutral lipid content was increased in rats fed the low protein diets supplemented with methionine (groups 8SM and 8FM) and the normal protein diets containing 40% fructose (groups 20F and 20FM). The plasma free threonine content was positively correlated with the protein level in the diet. On day 14, rats fed a low protein diet had a threonine deficiency, except those in groups 8S and 8F. The plasma free threonine content of these rats was not reduced, possibly due to an impaired utilization of this amino acid. The liver lipidosis observed during lactation, in contrast to that observed during growth with a low protein diet, was not due to a threonine deficiency.  相似文献
