A strain of Streptomyces produces a new substance capable of inactivating some amylases. This has not been reported by previous workers.

This amylase inhibitor was purified by means of acetone treatment, active carbon adsorption and column chromatography on DEAE-cellulose.

It was dialyzable through a cellophane membrane and soluble in water and methyl alcohol. The inhibitor had a small molecular weight and was a peptide-like substance. The inhibiting substance was resistant to the temperatures, and acted as inhibitor of glucoamylase, bacterial saccharogenic α-amylase, salivary and pancreatic α-amylases.  相似文献

The amylase inhibitor produced by Streptomyces sp. consisted of, at least, three kinds of inhibiting materials (inhibitor A, B and C). These inhibitors were separated by the Chromatographic techniques on Amberlite CG–120 and Dowex 1 × 2, and by paper chromatography etc.

Inhibitor A, B and C seemed to contain both carbohydrates and amino acids, but the specific activities (I. U./mg glucose) of the inhibitors A, B and C were different, that is, 18,800, 45,700 and 59,000, respectively. Also their ratios of amino acids to neutral sugars (expressed by moles of leucine per those of glucose) were approximately 0.09 (inhibitor A), 2.77 (inhibitor B) and 0.58 (inhibitor C).  相似文献

Talopeptin (N-(6-deoxy-α-l-talopyranosyloxyphospho)-l-leucyl-l-tryptophan), a specific inhibitor for thermolysin and some other endo-type metalloproteases, does not inhibit superoxide dismutase, alkaline phosphatase, or carbonic anhydrase, which are Zn(II)-enzymes, in the inhibitor concentration of the order of 10?4M. Although the enzyme activity of carboxypeptidase A, a Zn(II)-containing exo-type metalloprotease, is slightly reduced by the addition of talopeptin, the inhibition is not significant at the 10% level judged by Student's t test. On the other hand, talopeptin seemed to act as an inhibitor for all dehydrogenases examined, most of which do not contain Zn(II), at the inhibitor concentration studied (around 10?4M). It is possible that the phosphite group of talopeptin binds to the same site on the dehydrogenase as the one for the phosphate group of the coenzyme.  相似文献

The dialyzable proteinase inhibitor in the exocarp of the eggplant, Solanum melongena L. was partially purified by column chromatographies on DEAE-cellulose and Sephadex G–25. The inhibitor showed strong inhibitory activity on bovine trypsin [EC 3.4.4.4], Pronase and Nagarse. It also weakly inhibited α-chymotrypsin [EC 3.4.4.5], but, pepsin was not affected. The presence of three inhibitors in this preparation was demonstrated by isoelectrofocusing; their isoelectric points being pH 4.2, 4.7 and 6.3. The inhibitor with a pI value of 4.7 was present significantly more than the others. The molecular weight of the inhibitor was 5300 based on Sephadex G–75 gel filtration data. It was also very stable to heat treatment. This inhibitor was not the multi-headed type, and was gradually inactivated after a 4 hr incubation with Pronase.  相似文献

Acidocin 8912, a bacteriocin produced by Lactobacillus acidophilus TK8912, was purified by ammonium sulfate fractionation and successive chromatographies on CM-cellulose, Sephadex G-50, Sephadex G-25, and reversed-phase HPLC on Aquapore RP-300. The purified acidocin 8912 migrated as a single band on SDS–PAGE. The molecular weight was estimated to be 5200 by SDS–PAGE, and 5400 by HPLC gel filtration on TSKgel G3000PWXL. Both the amino acid composition and the N-terminal amino acid sequence analysis indicated that acidocin 8912 was a peptide composed of presumably 50 amino acids containing a Lys residue at the N-terminus. The purified acidocin 8912 showed a bactericidal effect on sensitive cells but not a bacteriolytic effect.  相似文献

水稻的糠皮和胚经生理盐水浸取、离心后的上清液加热至80℃处理10min,离心获得的上清液调pH至8.0,得到沉淀。沉淀溶解于0.01mol/LHCl,经透析冷冻干燥得水稻巯基蛋白酶抑制剂(CPI)粗品;粗品再经DEAE-Sepharose柱线性离子梯度洗脱和SephadexG-100柱分子筛层析,即可获得在PAGE、SDS-PAGE和HPLC上均为单一蛋白带的CPI样品。经上述步骤,CPI可被纯化58倍。经SephadexG-100和SDS-PAGE测定其分子量均为12000,N末端氨基酸为Pro,等电点5.6.水稻CPI经100℃处理10min后,其抑制活性无任何变化,在pH2.0~9.0之间,活性也不发生改变,但pH在9.0以上,其活性逐渐下降,水稻CPI对木瓜蛋白酶是一种高亲和性的抑制剂,它对木瓜蛋白酶和无花果蛋白酶有强抑制作用,对菠萝蛋白酶仅有弱抑制作用,但对胰蛋白酶则全无抑制作用;其抑制类型属竞争性抑制剂类型,K_i值约3.5×10 ̄(-8)mol/L对木瓜蛋白酶的抑制摩尔比约为1:1。  相似文献

A proteinaceous substance that inhibited the activity of papain (EC 3.4.22.2) was found in seeds of rice, Oryza sativa L. japonica. This cysteine proteinase inhibitor (CPI) was purified by a series of purification procedures including CM-Sephadex C-50, Sephadex G-75, and DEAE- Sephadex A-50 chromatography. The CPI was a single polypeptide with a molecular weight of about 12,000, with an isoelectric point at pH 5.3. The CPI was stable below 100°C and between pH 2.2 ~ 9.0. The inhibition of papain by the CPI was non-competitive, with a Ki value of 2.44 × 10-8 m. The complete inhibition of papain was reached by an equimolar concentration of the CPI.  相似文献

百合的鳞茎中含有一种对木瓜蛋白酶有强抑制作用的巯基蛋白酶抑制剂.百合的鳞茎经浸取加热处理,木瓜蛋白酶偶联的Sepharose4B柱亲和层析和SephadexG-100分子筛层析,可获得在PAGE和SDS-PAGE均为单一蛋白带的百合巯基蛋白酶抑制剂(CPI).此CPI为单链蛋白,含有0.307%的中性糖;N端氨基酸为Ile;SDS-PAGE测得亚基分子量为12000;SephadexG-100测得分子量为12500.百合CPI在100℃内和pH2~12范围内非常稳定;对木瓜蛋白酶的抑制属竞争性抑制类型,其Ki值为1.15×10~(-9)mol/L,对木瓜蛋白酶的抑制摩尔比为8.5:1.  相似文献

Amylase inhibitor (S-AI) was purified by about 25 times from culture filtrate of Streptomyces diastaticus subsp. amylostaticus No. 2476 through the methods of adsorption on active carbon, column chromatographies on Dowex 50 W × 2 (H-form) and Dowex 50 W × 2 (NH4-form), gel filtration on Sephadex G-25, El-complex formation with BLA, isolation of complex by gel filtration on Sephadex G-75, dissociation from complex by the method of acid denaturation, rechromatographies on Dowex 50 W × 2 (NH4-form) and Sephadex G-25. Homogeneity of this S-AI was examined by means of TLC, where S-AI gave a single spot in various solvent systems. S-AI specially inhibited α-amylases and glucoamylase, but not β-amylases and other glucoside hydrolases.

S-AI was a very stable substance, as it retained 100% of its original activity after being kept for 30 min at 100°C in a pH range between 3.0 and 10.0. The molecular weight of S-AI was estimated to be about 1500 by gel filtration on Sephadex G-15.

S-AI was regarded to be an oligosaccharide which was mainly composed of glucose in an amount of about 85 %. S-AI was hydrolyzed by β-amylase from non-reducing terminal and released two moles of maltose succesively.  相似文献

An isopropenyl ( = 3,3-dimethylallyl) 3-methoxyflavone (1) and its hydrate (5) were isolated from the roots of yellow lupin, Lupinus luteus L. cv. Topaz. Their structures were unambiguously determined to be 5,7,4′-trihydroxy-3-methoxy-6-(3,3-dimethylallyl)flavone (1) and 5,7,4′-trihydroxy-6-(3-hydroxy-3-methylbutyl)-3-methoxyflavone (5) by a combination of chemical and spectroscopic methods, and the new flavones were named topazolin and topazolin hydrate, respectively.

Antifungal tests against the growth of Cladosporium herbarum indicated that, in spite of its phenolic nature and the possession of an isopentenyl sidechain, topazolin (1) had only weak fungitoxic activity.  相似文献

In a previous report, the cDNA for human proteinase inhibitor 8 (PI8) was first identified, isolated, and subcloned into a mammalian expression vector and expressed in baby hamster kidney cells. Initial studies indicated that PI8 was able to inhibit the amidolytic activity of trypsin and form an SDS-stable approximately 67-kDa complex with human thrombin [Sprecher, C. A., et al. (1995) J. Biol Chem. 270, 29854-29861]. In the present study, we have expressed recombinant PI8 in the methylotropic yeast Pichia pastoris, purified the inhibitor to homogeneity, and investigated its ability to inhibit a variety of proteinases. PI8 inhibited the amidolytic activities of porcine trypsin, human thrombin, human coagulation factor Xa, and the Bacillus subtilis dibasic endoproteinase subtilisin A through different mechanisms but failed to inhibit the Staphylococcus aureus endoproteinase Glu-C. PI8 inhibited trypsin in a purely competitive manner, with an equilibrium inhibition constant (Ki) of less than 3.8 nM. The interaction between PI8 and thrombin occurred with a second-order association rate constant (kassoc) of 1.0 x 10(5) M-1 s-1 and a Ki of 350 pM. A slow-binding kinetics approach was used to determine the kinetic constants for the interactions of PI8 with factor Xa and subtilisin A. PI8 inhibited factor Xa via a two-step mechanism with a kassoc of 7.5 x 10(4) M-1 s-1 and an overall Ki of 272 pM. PI8 was a potent inhibitor of subtilisin A via a single-step mechanism with a kassoc of 1.16 x 10(6) M-1 s-1 and an overall Ki of 8.4 pM. The interaction between PI8 and subtilisin A may be of physiological significance, since subtilisin A is an evolutionary precursor to the intracellular mammalian dibasic processing endoproteinases.  相似文献

Streptomyces limosus was selected because it secreted a novel protease that catalyzed the synthetic reaction forming Pro-Pro-Pro from Pro-Pro. The protease was purified to an electrophoretically homogeneous state and an activity of more than about 20,000-fold that of the culture broth. The molecular mass of the enzyme was estimated to be 50 kDa by SDS-polyacrylamide gel electrophoresis. The enzyme was most active in alkaline pH for the synthetic reaction producing Pro-Pro-Pro from Pro-Pro, although for the hydrolytic reaction forming proline it was most active in neutral pH. The enzyme was inhibited by 1,2-epoxy-3-(p-nitrophenoxy)propane (EPNP) and diazoacetyl-DL-norleucine methyl ester (DAN). It can be considered that this enzyme belongs to the class of aspartic proteases. The substrate specificity indicates that this enzyme has a strong affinity for proline as a N-terminal amino acid of peptides.  相似文献

应用以胰蛋白酶为配体的亲和层析法,从生半夏蛋白粗提液的40%(NH。):SO4(M/V)沉淀中分离纯化出一种胰蛋白酶抑制剂(Rhizoma pinelliae trypsin inhibitor,RVn),经SDS.PAGE检测呈现单一条带,分子量为14kD左右,其N-端6个氨基酸残基顺序为DPVVDG。研究表明,其对胰蛋白酶的质量抑制比为l:4.72左右,对人低分化胃腺癌细胞系(BGC-823)的细胞增殖具有抑制作用,IC50值为121.53μg/mL。  相似文献

Streptomyces strain K1-02, which was identified as a strain of Streptomyces albidoflavus, secreted at least six extracellular proteases when it was cultured on feather meal-based medium. The major keratinolytic serine proteinase was purified to homogeneity by a two-step procedure. This enzyme had a molecular weight of 18,000 and was optimally active at pH values ranging from 6 to 9.5 and at temperatures ranging from 40 to 70°C. Its sensitivity to protease inhibitors, its specificity on synthetic substrates, and its remarkably high level of NH2-terminal sequence homology with Streptomyces griseus protease B (SGPB) showed that the new enzyme, designated SAKase, was homologous to SGPB. We tested the activity of SAKase with soluble and fibrous substrates (elastin, keratin, and type I collagen) and found that it was very specific for keratinous substrates compared to SGPB and proteinase K.  相似文献

The effect of Streptomyces-pepsin inhibitor (S-PI) on fruit-body formation of Lentinus edodes (Berk.) Sing, was studied. The addition of S-PI to the culture medium (5 ~ 10 µg/ml) shortened the time required for mature fruit-bodies, and increased the fruiting-percentage and the overall yield 3.4 times compared to the control.

The intracellular proteinase in the mycelium was investigated. Proteinases having an optimal pH of 2.7 and 7.0 were found in the vegetative mycelial extract. When S-PI was added to the culture medium, their activities were strikingly changed; the carboxyl proteinase activity was remarkably decreased, and, in the contrary, the metal proteinase activity was increased to 1.5 times that of the control.

The carboxyl proteinase was purified. This enzyme was strongly inhibited by S-PI and synthetic pepsin inhibitors such as DAN and EPNP. The molecular weight and isoelectric point were 43,000 and pH 3.4, respectively.  相似文献

To obtain actinomycetes capable of producing new enzyme affectors such as enzyme inhibitors or activators, a screening test was carried out. Streptomyces sp. strain No. BR-1381 isolated in our laboratory produced a proteinous lipase activator abbreviated as LAV. LAV was purified from the culture filtrate by salting-out with ammonium sulfate, DEAE-cellulose column chromatography and gel filtration on Sephadex G-100. LAV was stable in the pH range from 3 to 7 at 37°c for 20 hr and in a wider range of pH at 4°C for 5 days. LAV itself was very stable against heat treatment, but LAV did not have any effect on the thermal stability of Phycomyces nitens lipase. LAV activated several microbial lipases, but did not activate pancreatic or rice bran lipases. LAV particularly showed strong activation for Phycomyces nitens lipase.  相似文献
