Nucellar explants from the immature fruit of Ataulfo mango, 3–4 cm long, were used. The process had a very low efficiency (<10%); however, Ataulfo was more embryogenic than other cultivars such as Tommy Atkins or Haden in our hands. Overall, the process from nucellus to plantlet stage took 5 months. Twelve regenerated plants were obtained and are currently growing in the greenhouse. They appear to be normal and free of abnormalities. Some biochemical and molecular tests are currently being carried out. 相似文献

白黄侧耳Pleurotus cornucopiae微卫星间区(ISSR)分析 总被引:3,自引:0,他引:3
本试验对我国1982年至2004年22年间栽培的白黄侧耳Pleurotus cornucopiae 30个菌株进行了锚定ISSR分析,试验表明,引物P4和P5都能对白黄侧耳P.cornucopiae进行多态性扩增,P4将供试菌株扩增出45个条带,大小在200~20000bp,P5将供试菌株扩增出39个条带,大小在500~15000bp,扩增出的条带100%具多态性。聚类分析在遗传相似性61%的水平下将30个供试菌株划分为15个类群,即15个具一定遗传差异的菌株;具有相同ISSR图谱、遗传相似性程度100%的可能为同一菌株,属于同物异名。试验表明我国的食用蕈菌野生环境面临人工栽培种质的污染,采自河北、山东、云南自然环境下的白黄侧耳P.cornucopiae与此前大量栽培的一些商业品种具完全相同的ISSR指纹图谱,聚类分析相似性系数100%。 相似文献

Endosperm at late stage of cell formation, excised from open-pollinated fruit of sweet orange (Citrus sinensis Osbeck. cv.‘Hongjiang’), was suitable for culture in vitro. The results indicated that 2,4-D was necessary for callus' induction, and supplemented with BA, CH in medium was more effective: the percentage of induction was as high as 33.3%. Endosperm tissues, excised from the fruits treated with low temperature (4–7℃) for 16 and 19 days, and from the young seeds precuhured on MT basal medium respectively with NAA, GA3, BA for 2–6 days, also stimulated to callus formation. When endosperm callus was transfered to the differentiation medium, embryoids and shoot-buds only developed in a sequence of culture conditions. Callus was first cultured on MT+BA/GA3, then transfered to different media with various nitrogen or hormone concentration, and finally transferred back to the first culture medium. Shoots were regenerated from shoot-buds in the medium in presence of hormones with only BA or with GA3. The whole plants were regenerated from embryoids in presence of GA3 or with BA. Analysis of endosperm plantlets showed that 79.4% of the observed celld have chromosome number of 2n=26–27, nearly the triploid number of 2n=3x=27. Through grafting on lemon seedlings as rootstocks in vitro, plantlets were growing successfully in soil. 相似文献

Immature embryos of apricot (Prunus armeniaca L.) cv. Royal with a PF index of 25–100 were used to regenerate plants in vitro using two methods. In the first case, callus was initiated on MS medium with 4.5 M 2, 4-D plus 0.44 M BA and regeneration of shoots from the callus occurred on MS medium with 4.4 M BA plus 1.0 M 2, 4-D. The objective of this paper is the structural and narrative reconstruction of representations of ‘nature’, ‘wildlife’ and
‘landscape’, held by rural residents of the Dadia Forest Reserve. Data collection involved in-depth interviews. Employing
a social representations’ approach, we recovered representational elements that are expected in the case of rural belief systems,
such as negative dispositions towards wolves and foxes, as well as elements of an urban adherence, such as nature’s independence.
Representational elements refer to visual aspects of the countryside, which seem compatible with the figurative nucleus of
the rural idyll. Concerning ‘wildlife’, residents focused on vultures, which comprise the main tourist attraction of the reserve.
Scientific knowledge adds to the complexity of the narrative schema, which corresponds to the representation of ‘wildlife’.
Interviewees perceived the rural landscape as an interface between the natural and the human-conditioned environment. Our
study shows that interviewees make no reference to environmental conservation or quality of life issues, as it could be expected
according to relatively wide definitions of the term ‘environmentalism’. Environmental messages reinforced by ecotourism development
seem to be recalled primarily in terms of their compatibility with the perceived economic benefit of local people. Despite
ecotourism development, representational elements that diverge from a tourist version of ‘nature’, ‘wildlife’ and ‘landscape’
were not pronounced within rural belief-systems. Further interventions within the study area are needed, in order to address
a variety of topics under the environmental conservation discourse and raise the environmental awareness of rural residents. 相似文献

‘Jin Zhui’ is a spontaneous self-compatible mutant of ‘Ya Li’ (Pyrus bretschneideri Rehd. S21S34), the latter displaying a typical S-RNase-based gametophytic self-incompatibility (GSI). The pollen-part mutation (PPM) of ‘Jin Zhui’ might be due to a natural mutation in the pollen-S gene (S34 haplotype). However, the molecular mechanisms behind these phenotypic changes are still unclear. In this study, we identified five SLF (S-Locus F-box) genes in ‘Ya Li’, while no nucleotide differences were found in the SLF genes of ‘Jin Zhui’. Further genetic analysis by S-RNase PCR-typing of selfed progeny of ‘Jin Zhui’ and ‘Ya Li’ × ‘Jin Zhui’ progeny showed three progeny classes (S21S21, S21S34 and S34S34) as opposed to the two classes reported previously (S21S34 and S34S34), indicating that the pollen gametes of ‘Jin Zhui’, bearing either the S21- or S34-haplotype, were able to overcome self-incompatibility (SI) barriers. Moreover, no evidence of pollen-S duplication was found. These findings support the hypothesis that loss of function of S-locus unlinked PPM expressed in pollen leads to SI breakdown in ‘Jin Zhui’, rather than natural mutation in the pollen-S gene (S34 haplotype). Furthermore, abnormal meiosis was observed in a number of pollen mother cells (PMCs) in ‘Jin Zhui’, but not in ‘Ya Li’. These and other interesting findings are discussed. 相似文献

Abstract Anopheles stephensi is the main vector of urban malaria in South Asia. Three ecological variants (‘type’, ‘mysorensis’and‘intermediate’) of An. stephensi have been reported on the basis of ecology and egg morphology. However, it is unclear if there is any genetic isolation between the three variants. We analyzed the three variants of An. stephensi using eight microsatellite loci and found that large and significant genetic differentiation exists between them (mean FST= 0.393 and mean RST= 0.422). Pairwise estimates of genetic differentiation between the variants were ‘type’ versus ‘mysorensis’ (mean FST= 0.411 and mean RST= 0.308), ‘type’ versus ‘intermediate’ (mean FST= 0.388 and mean RST= 0.518) and ‘intermediate’ versus ‘mysorensis’ (mean FST= 0.387 and mean RST= 0.398) and all were statistically significant (P < 0.05). The greater sensitivity of RST in differentiation indicated that mutations and not genetic drift had generated the differences between three variants of An. stephensi. The present study indicated large genetic differentiation and presence of non-significant low level of gene flow between the three variants (‘type’, ‘mysorensis’and‘intermediate’) of An. stephensi. 相似文献

Hotspots of biodiversity (taxonomic richness, endemism, taxonomic affinities between communities) at small (), medium () and larger () scales of diversity were examined for marine sponge populations throughout tropical and subtropical Australia, with the faunas of Vanuatu, Palau and Thailand used as outgroups for comparison. Spatial and numerical (ordination) models and hierarchic classifications delineated 37 and 13 scale faunas from 1343 investigated localities using a pool of 2324 species. The Australian taxonomic literature was ignored completely to avoid the many still unresolved taxonomic problems and to allow equal treatment of collecting localities. Richness and endemism varied considerably between marine areas, for species and genera at all spatial scales, with gradients strongly corroborated by hierarchic taxonomic relationships between faunas. Richness and endemism were equally effective indicators of biodiversity hotspots, whereas species-level vs. genus-level data produced differing patterns, with the latter substantially underestimating biodiversity and marine area relationships, and consequently a poor 'surrogate for species data. Patterns of taxa shared between adjacent areas were more informative than richness and endemism data alone, as they more accurately reflect the processes in these areas. Latitudinal gradients in sponge diversity were not evident, whereas various environmental factors were prominent at scales and biogeographic factors were prominent at and scales of diversity. An example of a small () scale diversity fauna revealed substantial spatial heterogeneity (mean of 41 spp/locality, 33% apparently endemic, and a total fauna of 226 spp) containing few ubiquitous species (40% or 78 spp), with adjacent reefs having relatively low faunal similarity (mean 33%). Faunas at the medium () scale of diversity were less heterogeneous (mean 127 spp/region, 27% apparently endemic to a particular region, with a total fauna of 2324 spp), containing a significantly larger dataset (829 spp) found in >1 region to assess taxonomic affinities. At the larger () scale of diversity faunas were far more heterogeneous (mean 263 spp/region, 47% apparently endemic to a particular region) containing a smaller dataset (only 588 spp or 26% of the fauna with >1 species/region) to assess taxonomic affinities. Consequently, sponge faunas at the and scales of diversity are ineffective and inappropriate as biodiversity models, respectively, with scale diversity also less relevant as a practical tool for marine resource management and marine area conservation. 相似文献

Freshwater species of the genus Euplotes (Protozoa, Ciliophora) change their morphology in the presence of some of their predators. The ciliates develop extended lateral wings as well as dorsal and ventral projections which make engulfment by predators more difficult. In a series of laboratory experiments ingestion rates of four protozoan predators, the ciliates Lembadion bullinum, Dileptus anser, Stylonychia mytilus and Urostyla grandis, and one metazoan predator, the turbellarian Stenostomum sphagnetorum, on three species of Euplotes (E. octocarinatus, E. patella and E. aediculatus) were determined. It was calculated that the probability of rejection by a predator changed from 1:1 for ovoid morphs of Euplotes to about 2:1–20:1 for winged morphs of Euplotes, dependent on the prey and predator species that were combined. The nutritional condition of the prey also had some influence. In mixed-species cultures of prey and predators, transformed cells of E. octocarinatus survived for several months. 相似文献

A simple and effective method of regenerating Syngonium podophyllum ‘Variegatum’ via direct somatic embryogenesis has been established. Leaf and petiole explants were cultured on Murashige
and Skoog (MS) medium supplemented with N-(2-chloro-4-pyridyl)-N′-phenylurea (CPPU) or N-phenyl-N′-1,2,3-thiadiazol-5-ylurea (TDZ) with either α-naphthalene acetic acid (NAA) or 2,4-dichlorophenoxyacetic acid (2,4-D). Somatic embryos directly formed at one or two sides of petiole explants on MS medium supplemented 2.5 mg l−1 TDZ with 0.5 mg l−1 NAA or 2.0 mg l−1 TDZ with 0.2 mg l−1 NAA or with 0.2 and 0.5 mg l−1 2,4-D, respectively. The frequency of petiole explants with somatic embryos produced was as high as 86% when cultured on medium
containing 2.5 mg l−1 TDZ with 0.5 mg l−1 NAA. Up to 85% of somatic embryos were able to germinate after transferring onto medium containing 2.0 mg l−1 6-benzylaminopurine (BA) and 0.2 mg l−1 NAA. Approximately 50–150 plantlets were regenerated from a single petiole explant. However, there was no somatic embryo
formation from leaf explants regardless of growth regulator combinations used. Regenerated plantlets from petiole explants

The mating system of a subsocial spider mite, Schizotetranychus miscanthi Saitō, which is closely related to Schizotetranychus longus Saitō (the long seta form of Schizotetranychus celarius (Banks) is a synonym of the latter) was studied in comparison with that of S. longus. Comparisons between nesting patterns of the two related species, S. miscanthi and S. longus revealed a difference in distribution of males among nests. Although more than one male sometimes occurred in the large nests of S. miscanthi, most nests were occupied only by a single male. On the other hand, many nests of S. longus included several males. Behavioral experiments revealed that the male and females of S. miscanthi which cohabited in a nest defended their offspring from phytoseiid predators. Observations and a census of the nesting pattern in a wild population indicate that this is the second example of biparental defense and of a subsocial life-pattern in spider mites. Differences in mating systems were experimentally demonstrated in the two species. Only a single male of S. miscanthi survived in a nest, as a result of highly aggressive male-male combat, while two males of S. longus cohabited in a nest. The mating system of the former species is thus considered as harem polygyny, while that of the latter as scramble type polygyny. Furthermore, observations by video recording and scanning electron microscopy showed that the winning male in the male-male combat in S. miscanthi often preyed on the loser, suggesting cannibalism among them. 相似文献

Chronic oral anticoagulant treatment is obligatory in patients (class I) with mechanical heart valves and in patients with atrial fibrillation with CHADS2 score >1. When these patients undergo percutaneous coronary intervention with placement of a stent, there is also an indication for treatment with aspirin and clopidogrel. Unfortunately, triple therapy is known to increase the bleeding risk. For this group of patients, the bottom line is to find the ideal therapy in patients with indications for both chronic anticoagulation therapy and percutaneous intervention to prevent thromboembolic complications such as stent thrombosis without increasing the risk of bleeding. Background
Epidemiological data have shown long-term health adversity in low birth weight subjects, especially concerning the metabolic syndrome and ‘diabesity’ risk. Alterations in adult food intake have been suggested to be causally involved. Responsible mechanisms remain unclear.Methods and Findings
By rearing in normal (NL) vs. small litters (SL), small-for-gestational-age (SGA) rats were neonatally exposed to either normal (SGA-in-NL) or over-feeding (SGA-in-SL), and followed up into late adult age as compared to normally reared appropriate-for-gestational-age control rats (AGA-in-NL). SGA-in-SL rats displayed rapid neonatal weight gain within one week after birth, while SGA-in-NL growth caught up only at juvenile age (day 60), as compared to AGA-in-NL controls. In adulthood, an increase in lipids, leptin, insulin, insulin/glucose-ratio (all p<0.05), and hyperphagia under normal chow as well as high-energy/high-fat diet, modelling modern ‘westernized’ lifestyle, were observed only in SGA-in-SL as compared to both SGA-in-NL and AGA-in-NL rats (p<0.05). Lasercapture microdissection (LMD)-based neuropeptide expression analyses in single neuron pools of the arcuate hypothalamic nucleus (ARC) revealed a significant shift towards down-regulation of the anorexigenic melanocortinergic system (proopiomelanocortin, Pomc) in SGA-in-SL rats (p<0.05). Neuropeptide expression within the orexigenic system (neuropeptide Y (Npy), agouti-related-peptide (Agrp) and galanin (Gal)) was not significantly altered. In essence, the ‘orexigenic index’, proposed here as a neuroendocrine ‘net-indicator’, was increased in SGA-in-SL regarding Npy/Pomc expression (p<0.01), correlated to food intake (p<0.05).Conclusion
Adult SGA rats developed increased ‘diabesity’ risk only if exposed to neonatal overfeeding. Hypothalamic malprogramming towards decreased anorexigenic activity was involved into the pathophysiology of this neonatally acquired adverse phenotype. Summary The life cycle of the Batrachospermales (freshwater florideophyte red such as Batrachospermum, Lemanea) is a shortened variant of the ‘normal’ marine florideophyte life cycle. The perennial Chantransia diploid phase is microscopic and encrusting. Each winter it produces one or more semi-erect haploid gametophytes by vegetative meiosis. Gamete production and fertilization is followed by production of diploid carposporophytes; these produce diploid carpospores which disperse, and regenerate the Chantransia phase. The question of the extent to which the Chantransia phase contributes resources to the gametophyte was approached by physiological-hydrodynamic modelling. These computations suggest that the photosynthetic rates in situ on an area basis are 20 times greater for the gametophyte than the Chantransia phase; this agrees with the observed ratios of peak biomass. The conclusion is that the Chantransia phase has a negligible role in provisioning the growing gametophyte, and that the role of the Chantransia phase is to occupy space with living biomass throughout the year, including exposure at summer drawdown, and (perhaps) by dispersal via production of monospores. A similar conclusion is arrived at on the basis of biomass data for the role of the perennial ‘mushroom’ phase of the semelparous marine Fucalean brown alga Himanthalia elongata in relation to the short-lived but much larger reproductive receptacles. 相似文献

A repeatable in vitro culture method was established and it could induce a series of abnormal embryos by which their scale leaves were substituted by petals. These petal-bearing embryos were derived from long-term root calli of an orchid cultivar, Oncidium ??Gower Ramsey??. The calli were induced and subcultured on a modified 1/2MS medium supplemented with five combinations of TDZ and dicamba. When 1-year-old callus, which induced and subcultured at 3?mg/l TDZ and 5?mg/l dicamba (line 13 callus), was transferred onto 1/2MS medium supplemented with 0.1?ml/l NAA and 3?mg/l TDZ, it gave the highest number of petal-bearing embryos. However, line 13 root explants gave one of the lowest percentage of callus formation (12.5%) and the number of somatic embryos per callus was also one of the lowest (along with lines 10, 11 and 12). The efficiency of embryogenesis decreased with age of callus and the significant decrease was started from the third year of culturing. Flowering characteristics of plantlets from normal embryos and petal-bearing embryos were both evaluated after 3?years of culture in the greenhouse. Parameters including length of the longest inflorescence, numbers of flowers per plant, length of flowers and width of flowers were all not significantly different between abnormal embryo-derived plants and normal embryo-derived plants. 相似文献

Oribatid mite and Carabid beetle communities were investigated at five sites in the ‘Pietraporciana’ and ‘Lucciolabella’ Nature Reserves (central Italy). In this part of southern Tuscany many attempts have been made to encourage the regeneration of native habitats and to preserve existing ones. Human-induced changes in the original forest landscape have had a direct impact on mite and carabid populations. Significant differences in species diversity and abundance among different sites were revealed throughout the sampling period. Species richness, abundance and diversity of oribatid mites decrease from woodland sites to open habitats where evenness was high. There is an inverse trend between the number of species and richness of carabid beetles and those of oribatid mites. Canonical correspondence analysis (CCA) of oribatid and carabid beetle compositions discriminated the sites, demonstrating how even small areas with different vegetation, composition, structure, environment and microclimate were characterised by distinct edaphic populations. 相似文献
