(R)-β-acetylmercaptoisobutyric acid (RAM), a chiral compound, is an important intermediate for the chemical synthesis of various antihypertensive and congestive heart failure drugs. Microorganisms capable of converting (R,S)-β-acetylmercaptoisobutyric acid ((R,S)-ester) to RAM were screened from soil microorganisms. A strain ofPseudomonas sp. 1001 screened from a soil sample was selected to be the best. Cells showed an activity of 540 U/mL from culture broth and the enzyme was thermostable up to 70°C. This strain could produce RAM asymmetrically from (R,S)-ester.  相似文献

(22R,23R,24S)-22,23-Isopropylidenedioxy-5α-ergost-2-en-6-one 2b is an important intermediate of brassinolide. We found that the enone 2b can be prepared by transformation of (22R,23R,24S)-3α,5-cyclo-22,23-isopropylidenedioxy-5α-ergostan-6-one 5b with catalytic amount of both p-TsOH and NaBr in DMF under reflux. 5b was prepared from (22R,23R,24S)-3α,5-cyclo-22,23-dihydroxy-6β-methoxy-5α-ergostane 9b or a 6β-benzyloxy compound 9c, which was obtained in a manner similar to Mori’s brassinolide synthesis. The enone 2b was eventually prepared via a benzyl ether 9c from stigmasterol 3a in a 15.5% yield in 11 steps.  相似文献

(R)-(+)- and (S)-(?)-dihydrozeatins [(R)-(+)- and (S)-(?)-6-(4-hydroxy-3-methylbutylamino)purines, 1a and 1b] and their ribosides {(?)-6-[(R)-4-hydroxy-3-methylbutylamino]- and (?)-6-[(S)-4-hydroxy-3-methyl-butylamino]-9-β-D-ribofuranosylpurines, 3a and 3b} were synthesized and tested for their cytokinin activity by four bioassay systems, the growth of tobacco callus, the seed germination of lettuce, the fr. wt increase of excised radish cotyledons and the retardation of chlorophyll degradation in radish cotyledons. In tobacco callus bioassay, 1a was more active than 1b. The ribosides 3a and 3b were not less active than their corresponding aglycones 1a and 1b. In other bioassays used the activity followed the order: 1a >3a >1b >3b. In tobacco callus bioassay and lettuce seed germination, trans-zeatin [6-(4-hydroxy-3-methylbut-trans-2-enylamino)purine] showed stronger cytokinin activity than 1a.  相似文献

We investigated the effect of dietary phospholipid (PL) concentrate from bovine milk on the epidermis. Thirteen-week-old hairless male and female mice (Hos:HR-1) were separated into two experimental groups, each fed two experimental diets: the control group and the PL group. The mice were given the experimental diets for 6 weeks. Stratum corneum hydration and transepidermal water loss (TEWL) were measured using Corneometer CM825 and Tewameter TM300 (Courage and Khazaka Electronics, Cologne, Germany) at 3 weeks and 6 weeks. After the feeding period, ceramides in stratum corneum were analyzed. We found that stratum corneum hydration and ceramides in the PL group were significantly higher than those in the control group and that TEWL in the PL group tended to decrease.

These results indicate that dietary PL concentrate improves epidermal function by increasing the amount of ceramides, resulting in higher hydration.  相似文献

Abstract

(E)-3′,5′-Diamino-5-(2-bromovinyl)-2′,3′,5′-trideoxyuridine (5), the diamino analogue of BVDU (1), was synthesized from BVDU. The protonation behavior of 5 has been studied by means of pH-metric measurements and NMR spectroscopy. This study allows the determination of the basicity constants and the stepwise protonation sites. Thus, the main species at physiological pH is the monoprotonated form. The conformational analysis of this nucleoside analogue was also carried out through 1H NMR spectroscopy. In addition, a convenient synthesis of N-3′ and N-5′ acylated derivatives was developed by regioselective enzymatic acylation. Thus, Candida antarctica lipase B (CAL-B) selectively acylated the 5′-amino group, thus furnishing nucleosides 8. On the other hand, immobilized Pseudomonas cepacia lipase (PSL-C) exhibited the opposite selectivity, conferring acylation at the 3′-amino group, thus affording derivatives 9.  相似文献

Valinol is part of numerous pharmaceuticals and has various other important applications. Optically pure valinol (ee >99%) was prepared employing different ω-transaminases from the corresponding prochiral hydroxy ketone. By the choice of the enzyme the (R)- as well as the (S)-enantiomer were accessible. Reductive amination was performed in organic solvent (MTBE) using 2-propyl amine as amine donor whereas alanine was applied in or in aqueous medium. Transformations in phosphate buffer were successfully performed even at 200 mM substrate concentration (20.4 g/L) leading to 99% (R) and 94% (S) conversion with perfect optical purity (>99% ee).  相似文献

Musculature innervated by the N. facialis inTheropithecus gelada (Rüppell) is patterned on broad lines in agreement with related genera of catarrhine monkeys, but presents some specializations and divergences in detail. Noteworthy is the extension to the labial margins superficially of the combined levator labii superioris and zygomaticus in the upper and the pars mandibularis of trachelo-platysma in the lower lip. A specialization of the medial fibres of levator labii superioris forms a sling-like structure within the upper lip and serves to implement the lip-flip gesture characteristic of the genus. Its antagonist is the orbicularis oris. Large-scale preparation of (R)-(–)-1,3-butanediol (R-BDO), an important chiral synthon, from the racemate by Candida parapsilosis IFO 1396 was investigated. We found that ethanol accumulated during culture enhanced the secondary alcohol oxido-reduction activity of cells. Large-scale preparation of R-BDO was done using a fermentor. 3092 g of R-BDO was obtained from the racemate by the use of this strain with 94.0% enantiomeric excess.  相似文献

N2- (4-Hydroxyphenyl)-2 ′-deoxyguanosine-5 ′-O-DMT-3 ′-phosphoramidite has been synthesized and used to incorporate the N2-(4-hydroxyphenyl)-2 ′-dG (N2-4-HOPh-dG) into DNA, using solid-state synthesis technology. The key step to obtaining the xenonucleoside is a palladium (Xantphos-chelated) catalyzed N2-arylation (Buchwald-Hartwig reaction) of a fully protected 2 ′-deoxyguanosine derivative by 4-isobutyryloxybromobenzene. The reaction proceeded in good yield and the adduct was converted to the required 5 ′-O-DMT-3 ′-O-phosphoramidite by standard methods. The latter was used to synthesize oligodeoxynucleotides in which the N2-4-HOPh-dG adduct was incorporated site-specifically. The oligomers were purified by reverse-phase HPLC. Enzymatic hydrolysis and HPLC analysis confirmed the presence of this adduct in the oligomers.  相似文献

The photobromination of 1,5-anhydro-2,3-O-isopropylidene-β-d-ribofuranose gave the corresponding (5S)-5-bromo compound. The reduction of the bromide with triphenyltindeuteride gave (5S)-(5-2H1)-1,5-anhydro-2,3-O-isopropylidene-β-d-ribofuranose, with a chiral purity of 76% at C-5, which was converted to (5R)- and (5S)-(5-2H1)-d-riboses and other ribofuranose derivatives.  相似文献

The synthesis of (–)-muricatacin starting from 1-bromododecane and 2-pentyn-l-ol is described. 2-Pentadecyn-1-ol (4), which was prepared from 1-bromododecane (2) and 2-pentyn-1-ol (3), was converted to epoxy alcohol 6 through a two-step reaction sequence, 6 being successively submitted to tosylation, iodination, chain extension with tert-butyl lithioacetate, and acid-catalyzed cyclization to give (–)-muricatacin (1a). A fluorescent analog of angiotensin II was synthesized by reacting fluorescein 5′-isothiocyanate with (Asp1, Ile5)-angiotensin II. Nα-(N-Fluoresceinthiocarbamoyl)-(Asp1, Ile5)-angiotensin II was purified by chromatography on DEAE-cellulose and Sephadex G-25. Analysis of the analog by thin-layer chromatography, thin-layer electrophoresis, and reversed-phase high-performance liquid chromatography indicated that the analog was free of angiotensin II and fluorescein 5′-isothiocyanate. N-Terminal sequence analysis demonstrated that fluorescein 5′-isothiocyanate reacted with the N-terminal aspartic acid residue of angiotensin II. Nα-(N-Fluoresceinthiocarbamoyl)-(Asp1, Ile5)-angiotensin II has an absorption maximum at 492 nm, and the value of the molar extinction coefficient, ?, is 7.7 × 104m?1 cm?1. The fluorescence emission maximum occurs at 520 nm. Infusion of the analog (0.69 μg/min/kg body wt) directly into the renal artery of an anesthetized rat reduced the blood flow by 12 to 27% within 2 min. Infusion of angiotensin II (0.48 μg/min/kg body wt) reduced renal arterial blood flow by 35 to 53% within 2 min. Saralasin, a partial agonist and antagonist of angiotensin II, inhibited the biologic effect of the fluorescent analog and angiotensin II by 75 and 70%, respectively. The purity, spectral properties, and in vivo biologic activity of Nα-(N-fluoresceinthiocarbamoyl)-(Asp1, Ile5)-angiotensin II indicate that this analog should facilitate characterization of angiotensin II receptors.  相似文献

A series of (?)-β-d-(2R,4R)-dioxolane-thymine-5′-O-aliphatic acid esters as well as amino acid esters were synthesized as prodrugs of (?)-β-d-(2R,4R)-dioxolane-thymine (DOT). The compounds were evaluated for anti-HIV activity against HIV-1LAI in human peripheral blood mononuclear (PBM) cells as well as for their cytotoxicity in PBM, CEM and Vero cells. Improved anti-HIV potency in vitro was observed for the compound 24 (5′-O-aliphatic acid esters) without increase in cytotoxicity in comparison to the parent drug. Chemical and enzymatic hydrolysis of the prodrugs was also studied, in which the prodrugs exhibited good chemical stability with the half-lives from 3 h to 54 h at pH 2.0 and 7.4 phosphate buffer. However, the prodrugs were relatively labile to porcine esterase with the half-lives from 12.3 to 48.0 min.  相似文献

The enzymatic properties of P2-2 enzyme were determined by using cells of M. radiodurans. The enzyme was: most active at 60°C incubation temperature, stable at 40°C in neutral buffer, and inactivated by heating at 80°C for 15min. Maximal lytic activity occurred at pH 8.5 in Tris-HCl buffer. The range of enzyme stability was between pH 5.5 and 8. Bivalent metal ions, p-chloromercuribenzoate and monoiodo acetate inhibited lytic activity. The molecular weight was estimated to be 16,000 daltons by gel filtration on Sephadex G-75. The enzymatic digestion of peptidoglycans from the cell walls of M. radiodurans and M. lysodeikticus liberated free amino groups, but neither reducing groups nor N-acetylhexosamine, indicating that the enzyme was an endopeptidase. From analysis of the N-terminal amino acids of the digests, it is suggested that the P2-2 enzyme cleaves the peptide bond at the carboxyl group of D-alanine in peptidoglycan.  相似文献
