HPLC法测定新疆贯叶连翘中绿原酸和芦丁的含量   总被引:2,自引:0,他引:2  
采用液相色谱法测定贯叶连翘中绿原酸和芦丁含量。用Hyperil ODS色谱柱(12.5 cm×6.0 mm i.d.,5μm)进行分离。流动相为甲醇-1%冰醋酸梯度洗脱,流速为0.6 mL/min。检测波长为343 nm。绿原酸线性范围为0.0076~0.3040μg,相关系数R=0.9997。芦丁线性范围为0.312~3.120μg,相关系数R=0.9999。样品的平均回收率分别为101.2%,99.8%。此方法准确、快速,适用于绿原酸和芦丁的定量分析。  相似文献

为了探索绿原酸和芸香苷的新型植物资源和生物活性,同时为低等级烤烟叶的综合利用开辟有效途径,本文采用高效液相色谱技术,对中国和湖南省不同生态烟区以及不同部位烤烟叶中绿原酸和芸香苷的含量进行了研究。结果表明,东北烟区低等级烤烟叶中的绿原酸和芸香苷含量很高;湖南省湘南烟区烤烟叶中绿原酸和芸香苷含量显著高于湘北和湘中烟区;不同部位烤烟叶中绿原酸含量差异不显著,芸香苷含量差异极显著,并且上部叶>中部叶>下部叶。  相似文献

Chlorogenic acid (CQA) is one of the major polyphenols in apple and a good substrate for the polyphenol oxidase (PPO) in apple. Apple contains catechins as well as CQA, and the role of CQA quinone and its interaction with catechins in the enzymatic browning of apple were examined. Browning was repressed and 2-cysteinyl-CQA was formed when cysteine was added to apple juice. CQA quinone was essential for browning to occur. Although catechins and CQA were oxidized by PPO, some catechins seemed to be non-enzymatically oxidized by CQA quinone.  相似文献

The leaves of the white mulberry tree (Morus alba L.) are used worldwide in traditional medicine as anti-diabetics. Various constituents of mulberry leaves, such as iminosugars (i.e. 1-deoxynojirimicin), flavonoids and related compounds, polysaccharides, glycopeptides and ecdysteroids, have been reported to exert anti-diabetic activity, but knowledge about their contribution to the overall activity is limited. The objective of the present work was to determine the in vivo anti-diabetic activity of an extract of mulberry leaves (MA), and to examine to what extent three major constituents, chlorogenic acid, rutin and isoquercitrin, might contribute to the observed activity. Quantities of the three constituents of interest in the extract were determined by using HPLC-DAD. Activity was determined by using a type II diabetic rat model. After 11 days of per os administration of 250 or 750 mg/kg of MA or the corresponding amounts of each individual compound, a dose dependent decrease of non-fasting blood glucose levels were found for MA, chlorogenic acid and rutin, but not for isoquercitrin. Based on our results, chlorogenic acid and rutin might account for as much as half the observed anti-diabetic activity of MA, hence they can be considered as excellent markers for the quality control of mulberry products.  相似文献

建立了胎菊中绿原酸的提取方法及检测方法.采用超声波法对胎菊中的绿原酸进行提取,考查了提取溶剂、提取时间、提取料液比对提取量的影响,HPLC测定提取液中绿原酸的含量,确定最佳提取工艺.结果表明,以甲醇为溶剂、提取时间为80 min、提取料液比为1:20时,胎菊中绿原酸的提取量最高.该方法简单、快速、准确,可用于胎菊中绿原酸的含量检测.  相似文献

Developmental stages of tobacco (Nicotiana tabacum L. cv. Burley 21) flower and capsule were correlated with tissue contents of polyphenols and activities of phenylalanine ammonialyase, polyphenoloxidase, and peroxidase. Chlorogenic acid, scopolin, and scopoletin were present in most tissues, whereas rutin and two dihydroxyphenolic glycosides concentrated primarily in the corolla and placenta, respectively. Ovules contained only chlorogenic acid. As development progressed, polyphenols accounted for nearly 15% of the dry weight in the green capsule of field-grown plants. Fertilization triggered a rapid increase of chlorogenic acid in the ovary. When l-phenylalanine-U-14C was fed to the detached green capsules and capsule parts, an incorporation of radioactivity into chlorogenic acid and dihydroxyphenolic glycosides occurred which suggested in situ synthesis of these compounds. This was subtantiated by a positive correlation between phenylalanine ammonia-lyase activity and polyphenol accumulation. High polyphenoloxidase activity was associated mainly with the ovary, whereas peroxidase activity was maximal during senescence of all tissues. Polyacrylamide gel slab electrophoresis revealed five cathodic bands and one diffuse zone with poly-phenoloxidase activity in flower extracts. Two anodic poly-phenoloxidase isozymes appeared only in the fertilized ovary. Among 17 peroxidase isozymes, six cathodic forms were present throughout floral development, and the anodic ones increased in number and activity at the later stages of capsule growth.  相似文献

绿原酸对莴苣生长的化感作用及其机理研究   总被引:2,自引:0,他引:2  
酚酸类化合物是一类重要的化感物质,广泛存在于农作物等植物体内以及耕作土壤中,与植物的化感效应以及连作障碍密切相关。该研究以蔬菜类植物莴苣为受体,采用植物细胞和生理学方法,分析酚酸类化感物质绿原酸抑制莴苣生长的活性效应及其作用机理,以揭示绿原酸介导的化感作用及连作障碍机制。结果显示:(1)绿原酸对莴苣的根长、茎长以及鲜重等生长指标均表现出低浓度(0.1和1.0μmol/L)促进、高浓度(10、100和1 000μmol/L)抑制的活性作用模式,其在10μmol/L及以上浓度时对根长和鲜重具有显著抑制作用,但在各浓度下对莴苣茎长无显著影响。(2)绿原酸处理后,莴苣根尖细胞分裂指数明显下降,随处理浓度的升高,各分裂时期的细胞比例也显著降低,导致细胞分裂过程受阻。(3)较低浓度(0.1、1.0和10μmol/L)绿原酸对莴苣根尖细胞活力无明显影响,较高浓度绿原酸(100和1 000μmol/L)使莴苣根尖死细胞显著增多,根尖端绝大部分细胞失去活力。(4)DHE荧光染色显示,与对照相比,低浓度(0.1和1.0μmol/L)绿原酸对莴苣根部的活性氧积累无明显影响,当绿原酸处理浓度大于10μmol/L时,随处理浓度升高莴苣根部的活性氧积累明显大量增多。研究表明,绿原酸能够诱导莴苣体内活性氧产生并积累,从而导致莴苣细胞分裂受阻、细胞活力降低,最终影响莴苣幼苗的生长发育。  相似文献

In order to understand browning in tobacco plants during aging,age-dependent changes in the levels of ascorbic acid (AA) andchlorogenic acid (CGA) and its isomers were investigated inthe apoplast and the symplast of the leaves. Also activitiesof peroxidase (POX) and superoxide dismutase (SOD) were determined.AA decreased during aging until it was no longer detectablein the apoplast, while symplastic AA remained although the leveldecreased on aging. In contrast, levels of CGA and its isomersand activity of POX in the apoplast increased on aging, whilethose in the symplast remained nearly constant in mature andold leaves. The activity of SOD in the apoplast increased duringaging, while that in the symplast decreased. Oxidation of CGAby the apoplastic solution was observed in the absence of externallyadded H2O2 and the oxidation was inhibited by SOD and catalase.Brown components, which contained caffeic acid moieties, accumulatedin the apoplast on aging and the components produced O–2and H2O2 by autooxidation. From these results, we conclude (i)that brown components are formed in the apoplast by the CGA/POXsystem, (ii) that the H2O2 required for the reaction can beprovided by the CGA/POX system itself and by autooxidation ofthe brown components, and (iii) that apoplastic SOD functionsto generate H2O2 from apoplastically formed O–2. A novel acyltransferase from cotyledons of tomato (Lycopersicon esculentum Mill.), which catalyzes the transfer of caffeic acid from chlorogenic acid (5-O-caffeoylquinic acid) to glucaric and galactaric acids, was purified with a 2400-fold enrichment and a 4% recovery. The enzyme showed specific activities (theoretical Vmax per milligram of protein) of 625 nanokatals (caffeoylglucaric acid formation) and 310 nanokatals (caffeoylgalactaric acid formation). On sodium dodecyl sulfate-polyacrylamide gel electrophoresis it gave an apparent Mr of 40,000, identical to the value obtained by gel filtration column chromatography. Highest activity was found at pH 5.7, which was constant over a range of 20 to 120 millimolar K-phosphate. The isoelectric point of the enzyme was at pH 5.75. The reaction temperature optimum was at 38°C and the apparent energy of activation was calculated to be 57 kilojoules per mole. The apparent Km values were 0.4 millimolar for glucaric acid, 1.7 millimolar for galactaric acid, and with both acceptors as second substrates 20 millimolar for chlorogenic acid. The relative ratio of the Vmax/Km values for glucaric acid and galactaric acid was found to be 100:12. Substrate-competition experiments support the conclusion that one single enzyme is responsible for both the glucaric and galactaric acid ester formation with marked preference for glucaric acid. It is proposed that the enzyme be called chlorogenic acid:glucaric acid O-caffeoyltransferase (EC 2.3.1.-). The three caffeic acid-dependent enzyme activities involved in the formation of the glucaric and galactaric acid esters, the chlorogenic acid:glucaric acid caffeoyltransferase as the key activity as well as the caffeic acid:CoA ligase and the caffeoyl-CoA:quinic acid caffeoyltransferase as the preceding activities, were determined. The time course of changes in these activities were followed during development of the seedling in the cotyledons and growth of the young plant in the first and second leaf. The results from tomato seedlings suggest a sequential appearance of these enzymes.  相似文献

建立高效液相色谱—双波长同时检测咖啡豆提取物中绿原酸和咖啡因含量的方法。咖啡因和绿原酸在0.2~1.0 mg·mL-1范围内,峰面积与浓度呈现良好的线性关系。咖啡因:Y=44077X+227.78,R=0.9997,RSD=0.78%;绿原酸:Y=34896X+289.96,R=0.9995, RSD=0.93%。以绿原酸含量为指标,在单因素试验基础上建立正交试验,确定从绿咖啡豆中提取绿原酸的最佳条件为乙醇浓度20%,液料比6∶1,提取时间60 min,提取温度70 ℃。以咖啡因和绿原酸含量为指标,通过萃取试验,确定最佳脱咖啡因萃取剂为环己烷-三氯甲烷-丙酮混合溶剂,萃取工艺为1.5倍体积萃取剂萃取3次,产物中咖啡因含量由(3.52±0.08)%下降至(0.47±0.11)%。  相似文献

以绿原酸为对照品,利用紫外分光光度法和高效液相色谱法分别建立了测定海芦笋中多酚酸和绿原酸含量的方法.紫外分光光度法检测波长为338 nm;高效液相色谱法采用Zorbax Eclipse XDB-C187(4.6 mm×150mm,5um),以甲醇和0.5%冰醋酸水溶液梯度洗脱.结果发现海芦笋及其生物盐中含有大量的多酚酸和绿原酸,其中多酚酸含量分别为6.49 g/kg,3.37 g/kg,绿原酸含量分别为0.234 9/kg,0.180 g/kg.同时也表明高效液相色谱法可用于海芦笋中多酚酸含量的确定.这两种简单快捷的定量分析多酚酸和绿原酸的方法,不仅可用于海芦笋及其生物盐产品的质量控制,而且也为海芦笋的进一步研究和开发提供了一定的依据.  相似文献

The protective effects of chlorogenic acid on paraquat-induced oxidative stress were examined in rats. The activities of erythrocytes and liver glutathione peroxidase, and of both liver catalase and glutathione reductase, which were increased by feeding paraquat, declined to the levels in the control rats by supplementing chlorogenic acid to the paraquat diet. The activity of superoxide dismutase was not changed by dietary paraquat or by supplementing chlorogenic acid to the paraquat diet. Paraquat in the diet markedly decreased the liver triacylglycerol and phospholipid concentrations, as well as the food intake and body weight gain, while chlorogenic acid protected against these decreases. These in vivo results and the in vitro superoxide anion scavenging activity of chlorogenic acid suggest that chlorogenic acid acted preventively against paraquat-induced oxidative stress.  相似文献

Chlorogenic acid possessed a weak caffeine-like psychostimulant property when assessed for its effect on spontaneous locomotor activity in mice. In the evaluation of the effects for the major metabolites of chlorogenic acid which were detected upon incubation with rat feces and/or excreted in urine after oral administration to rats, caffeic and m-coumaric acids were found to be the principal active metabolites, while the others contributed little to this caffeine-like psychostimulant activity.  相似文献
