Volatiles stripped from rice bran are analyzed by gas chromatography and thin-layer chromatography either directly or after converted into 2, 4-DNPH’s or 3, 5-DNB’s. Most of prominent peaks revealed by a direct gas chromatography of the volatiles condensates behave as alcohols against classification reagents and are identified as methanol, ethanol, n-propanol, sec-butanol, isobutanol, n-butanol, isopentanol, n-pentanol and n-hexanol, respectively. Ethanol and methanol, being major components in the condensates are also detected by thin-layer chromatography of their 3, 5-DNB’s. Carbonyls identified by a flash exchange gas chromatography of 2, 4-DNPH’s of the volatiles are ethanal, propanal, isobutanal, acetone, n-butanal, isopentanal, n-pentanal and n-hexanal. Thin-layer chromatography of 2, 4-DNPH’s reveals presence of ethanal, acetone, isopentanal or/and isobutanal and n-hexanal in the volatiles. The experimental data have been integrated to furnish a basis for extensive comparison with other rice products and by-products.  相似文献

The enzyme showed the optimum pH between 7.5 and 8.0, and the optimum temperature at about 37°C. It was stable over the pH range from 4 to 9 and below 40°C, but completely lost the activity by heating 60°C for 15 min. The enzyme was activated by low concentration of calcium ion but inhibited partially by high concentration of calcium ion and by EDTA. With respect to substrate specificity, the enzyme exhibited a high specificity toward triglycerides and hydrolyzed ester bonds of short-carbon chain triglycerides faster than long-carbon chain triglycerides, whereas it catalyzed the hydrolysis of the oils from rice bran, olive and coconut. When 2-oleo-1,3-distearin was used as substrate, the enzyme was capable of preferentially hydrolyzing fatty acid ester bonds at the 1,3-position.  相似文献

Lipase extracted from defatted rice bran with calcium chloride solution was purified by ammonium sulfate precipitation, followed by successive column chromatographies on DEAE-cellulose, Sephadex G-75, CM-Sephadex C-50 in the presence of calcium ion. The specific activity of the purified enzyme was 4.7 units/mg protein and 480 times that of starting crude extract. The homogeneity of the enzyme protein was criticized by polyacrylamide gel disc electrophoresis and ultracentrifugation. The enzyme protein also behaved homogeneously in ampholine electrophoresis, indicating the isoelectric point of 8.56. The sedimentation coefficient of the enzyme was determined to be 2.97 S, and the molecular weight to be 40,000 by Archibald’s method. According to the measurement of optical rotatory dispersion of the enzyme, ORD constant, λc, Moffitt-Yang parameters, a0 and b0, were evaluated to be 239 mμ, ?164 and ?123, respectively.  相似文献

Some chemical properties of the rice bran lipase were studied. The enzyme protein contained 14.98% nitrogen and consisted of 312 amino acid residues. It also contained a certain amount of lipid. The amino-terminal amino acids of the enzyme protein were shown to be glutamic acid and the carboxyl-terminal amino acids to be glycine and serine. The treatment of the enzyme protein with 8 m urea containing 1×10?3m EDTA (ethyl-enediaminetetraacetic acid) seemed to cause dissociation of the subunits of the enzyme protein. From this observation and the results of the terminal amino acids analysis, it was presumed that the enzyme protein was composed of at least two types of subunits.  相似文献

Components of the unsaponifiable matter of crude rice bran oil were examined. A new compound having melting point of 157~158°C was obtained. Molecular weight was determined and ultraviolet and infrared analyses were carried out. Squalene and ferulic acid ester were confirmed to exist in the unsaponifiablc matter of crude rice bran oil. Quantitative relation of components was calculated and found to be 42% sterols, 24% higher alcohols. 20% ferulic acid esters, 10% hydrocarbons and 2% unreported compound.  相似文献

The anti-stress and anti-fatigue effects on rats and mice of a hot water extract of rice bran fermented with Saccharomyces cerevisae IFO 2346 were investigated. Oral administration (1 g/kg/day) of the hot water extract of fermented rice bran (FRB) inhibited major changes in the weight of the adrenal, thymus, spleen and thyroid, showing an anti-stress effect. The hot water extract of FRB also inhibited increases in the GPT and LDH activity, cholesterol and glucose in the serum. The administration (1 g/kg/day) for 2 weeks significantly prolonged the swimming time, resulting in an increase in the anti-fatigue effect. It is considered from these results that FRB had anti-stress and anti-fatigue effects.  相似文献

以水解度(DH%)和对DPPH自由基清除率为指标,筛选出制备米糠蛋白抗氧化活性肽的最适蛋白酶.研究最适蛋白酶的酶解条件,探讨底物浓度、蛋白酶的加入量、pH值、酶解时间等因素对水解度(DH%)和DPPH自由基清除率的影响;在单因素基础上采用Box-Behnken响应曲面中心组合设计法,对酶解米糠蛋白的工艺进行优化.试验结果表明,在加酶量13970.82 U/g,时间3.05h,底物浓度4.97%的水解条件下,米糠蛋白的水解度能够达到23.67%,活性肽对DPPH自由基清除率达到64.26%.  相似文献

A species of rice bran lipase (lipase II) was purified by ammonium sulfate precipitation, followed by successive chromatographies on DEAE-cellulose, Sephadex G–75 and CH-Sephadex C–50. Both polyacrylamide disc electrophoresis and ultracentrifugation demonstrated that the enzyme protein is homogeneous. The isoelectric point of the enzyme was 9.10 by ampholine electrophoresis. The sedimentation coefficient of the enzyme was evaluated to be 2.60 S, and the molecular weight to be 33,300 according to Archbald’s method. The enzyme showed the optimum pH between 7.5 and 8.0, and the optimum temperature at about 27°C. It was stable over the pH range from 5 to 9.5 and below 30°C. In substrate specificity, the enzyme exhibited a high specificity toward triglycerides having short-carbon chain fatty acids, although it was capable of hydrolyzing the ester bonds in the rice and olive oil.  相似文献

采用盐析法提取米糠蛋白,分别选用胃蛋白酶、胰蛋白酶、木瓜蛋白酶等单独或联合水解米糠蛋白,以从酶解米糠蛋白中分离获得具有血管紧张素转换酶(ACE)抑制活性的短肽组分。经Sephadex G-15凝胶层析和SP—Sephadex C-25离子交换层析分离各酶解组分,并检测各组分的ACE抑制活性。结果表明,米糠蛋白的酶解分离产物中含有较强ACE抑制活性的组分,其中经胃蛋白酶和胰蛋白酶共同水解时得到的小分子量寡肽组分的抑制活性最强,为后续对其进行结构分析奠定了基础。  相似文献

To examine the distribution of rice bran tocotrienol (T3), we gave rice bran T3 to rats after considering an acceptable daily intake of vitamin E for humans. Male SD rats (5 weeks of age) were fed for 3 weeks on a commercial diet containing 6.4 mg of vitamin E per 100 g wt and additively received vitamin E or the vehicle (vitamin E-free corn oil) by oral intubation. The animals were randomly divided into 4 groups depending on the type of test diet: control (vehicle), non-T3 (no T3 + 4.3 mg of tocopherol (TOC)/kg body weight (b.w.)/day), low-T3 (0.8 mg T3 + 3.5 mg TOC/kg b.w./day), and high-T3 (3.2 mg T3 + 1.1 mg TOC/kg b.w./day). The control rats and rats in the non-T3, low-T3, and high-T3 groups took 4.3 and 8.6 mg of vitamin E/kg b.w./day, respectively. Rice bran γ-T3 was significantly distributed to the adipose tissue and increased from 1.1 to 10.2 nmol/g of adipose tissue according to the rice bran T3 intake.  相似文献

A relatively guanine-specific endoribonuclease (RB-1) was isolated from rice bran. The pH optimum was 8.5 using yeast RNA as a substrate. The enzyme activity was inhibited by Cu2+, Zn2+, DTT and SDS, while EDTA, PCMB, IAA and heparin had no effect on the activity. The enzymic activity of RB-1 was inhibited by 3′-GMP as an end-product inhibitor. The enzyme protein was highly heat-stable. RB-1 did not hydrolyze native calf thymus DNA, heat-denatured DNA, poly A, poly U and poly C. Among synthetic substrates, only poly I was depolymerized. Only 2′,3′-cyclic GMP was identified in the hydrolysate of yeast RNA after 6hr hydrolysis.  相似文献

Extraction of oil from rice bran with supercritical carbon dioxide (SC-C02) at pressures of 150 to 350 kg/cm2 G at 40°C was demonstrated. The constituents of the selective fractions obtained at different pressures differed. Fractions obtained at higher pressures contained less free fatty acid and waxes or unsaponifiables. The phosphorus and iron contents were very low in the SC-C02 extracted oil, while the color of the oil was significantly lighter than that of hexane-extracted oil. The yield of low acid value oil was comparable to that for hexane extraction. One problem of the SC-C02 oil is its poor oxidation stability. This method of extraction may be effective in simplifying the processing of edible rice bran oil. Grinding the raw material was found to be effective in decreasing the C02 required and shortening the extraction period.  相似文献

Effect of protein to polysaccharide ratio (3:1, 6:1 & 9:1) and total biopolymer concentration (0.1, 0.2 & 0.4) on ζ-potential, particle size and particle distribution index (PDI) of complex coacervates were investigated. Furthermore, the physical, thermal and morphological characteristics of FG, RBP, RBP-FG coacervates and cross-linked RBP-FG coacervates by sodium tripolyphosphate were surveyed. Results showed that at low concentrations of FG (9:1 ratio) and a total concentration of 0.4, the ζ-potential of coacervate was close to zero and the coacervates had the largest size revealing the greatest interaction between biopolymers. SEM results showed a porous network structure which was varied from the RBP and FG. In contrast, the cross-linked coacervates showed a fine, uniform structure with less number of pores. FTIR findings revealed that the coacervate, due to the non-covalent interaction forces, was successfully developed. The fading of the pure peaks of protein and polysaccharide in XRD diffractogram indicated the interactions between the RBP and FG, as well as the structural changes of the complex. NaTPP cross-linked coacervate was indicated a reflection of slightly increased crystallinity. However, the dried powder of coacervates was generally amorphous. According to TGA and DSC results, cross-linked coacervates exhibited the highest thermal stability amongthe single biopolymers and non cross-linked coacervate.  相似文献

Cinnamic acid (CA) was added to rice bran oil (RBO) at concentrations range from 2 to 12% (w/w) to prepare functional fat-like organogel. The oil binding capacity and gel formation time of the obtained organogels were determined. The results showed that the oil binding capacity was increased and the gel formation time was significantly (p < 0.05) decreased with the increase in concentration of CA up to 10% (w/w). Based on these results, organogels prepared from RBO with 6, 8, and 10% CA (w/w) were chosen and subjected to further characterization analyses. The microscopy analysis showed different gel network structures and crystalline behavior for the organogels prepared with varying concentrations of CA. The rheological and thermal properties of the organogel were improved with the increase in CA concentration up to 10% (w/w). The X-ray diffractometer and Fourier Transform Infrared spectroscopy analyses indicated that the gel network formed based on crystalline of CA and physical entanglements among the organogel components. In addition, the organogel of RBO with 10% CA (w/w) showed a uniform and homogenized structure during storage at 5 °C for 28 days compared to the organogels prepared with 6 and 8%. Therefore, it can be suggested that cinnamic acid is a good organogelator for preparing functional fat-like organogel from rice bran oil.  相似文献

Aggregate sheath spot of rice caused by Rhizoctonia oryzae-sativae (Swa.) Mordue was found in several locations in Mazandaran Province. The polyacrylamide gel electrophoretic pattern of cell proteins of isolates from different regions were identical.
