近年来,越来越多的研究发现生物钟系统在许多生理活动中,包括心血管、内分泌、免疫、生殖等系统的生理,都起着重要作用。随着2006年卵巢生物钟的发现,生殖系统生物钟成为新的研究热点。研究发现卵巢生物钟不仅影响排卵,而且还控制类固醇激素的释放。卵巢生物钟属外围生物钟,受到中央生物钟(SCN)神经内分泌信号的调控。还发现下丘脑-垂体-卵巢(HPG)轴上各水平都存在生物钟,HPG轴上各生物钟失同步影响生殖能力,这可能导致一些疾病发生的病因。本文总结近十年的关于卵巢生物钟的研究,列举哺乳动物卵巢生物钟存在的证据,并阐述生物钟在雌鼠正常生殖生理过程,及在生殖系统疾病病理过程中的作用及其分子机制。  相似文献

Organisms must distribute sufficient energy among different and often competing physiological systems. This task can become challenging, however, as resources are often limiting, resulting in energetic trade-offs. For example, energetically based trade-offs between the reproductive and immune systems are common across taxa, yet the regulatory mechanisms underlying these trade-offs remain unclear. The adipose tissue hormone leptin is an ideal candidate for the modulation of energetic trade-offs between different physiological systems as this hormone serves as a gage of fat reserves and also modulates a range of physiological activities including the reproductive and immune processes. This article presents a review of the evidence for the role of leptin as a modulator of energetic trade-offs with the immune system and suggests its importance in disease ecology. In addition, we provide a case study of the ornate tree lizard (Urosaurus ornatus), testing whether leptin is involved in mediating a well-documented influence of energy state on the trade-off between reproductive activity and immune function. Overall, the combined results suggest that leptin serves as a proximate endocrine signal of available energy to the immune system, and therefore likely to affect susceptibility to diseases.  相似文献

The neuropeptide kisspeptin and its receptor are essential for activation of the hypothalamic-pituitary-gonadal (HPG) axis and regulating reproduction. While the role of kisspeptin in regulating the HPG axis in mammals has been well established, little is known about the functional ability of kisspeptins to activate the HPG axis and associated behavior in non-mammalian species. Here we experimentally examined the effects of kisspeptin on downstream release of testosterone and associated aggression and display behaviors in the side-blotched lizard (Uta stansburiana). We found that exogenous treatment with kisspeptin resulted in an increase in circulating testosterone levels, castration blocked the kisspeptin-induced increase in testosterone, and testosterone levels in kisspeptin-treated animals were positively related to frequency of aggressive behaviors. This evidence provides a clear link between kisspeptin, testosterone, and aggressive behavior in lizards. Thus, it is likely that kisspeptin plays an important role more broadly in non-mammalian systems in the regulation of reproductive physiology and related behaviors.  相似文献

哺乳动物的生殖功能受体内状态和外部环境综合作用的影响,这种综合作用通过作用于HPG轴的刺激因子和抑制因子之间的相对平衡来调控生殖。RFRP-3是目前下丘脑中唯一已知的HPG轴抑制因子。大量研究证实,RFRP-3能够抑制GnRH和LH的分泌,进而影响生殖功能。然而,RFRP-3对LH分泌的抑制作用是发生在垂体水平还是下丘脑水平尚不清楚。此外,RFRP-3还可能参与了MLT对哺乳动物季节性繁殖调控的信号通路,但是MLT对RFRP-3神经元的作用方式仍不清楚。此外,RFRP-3还可能在能量平衡和动物行为的调控中发挥着重要作用。文章就RFRP-3对HPG轴的调节机制以及其在能量平衡调节和行为调控中的作用进行了系统的阐述,并针对目前尚待解决的一些问题进行了探讨。  相似文献

The comparative approach in biological sciences has providedvaluable insights into the role of different organ systems inadaptation and evolution, and seeks to establish unifying themes.This approach also plays a key role in identifying model speciesand systems for the study of specific questions and problems.Further, by applying the concept of homology, information aboutnonmammalian species may be used either to directly understandmammalian/human regulatory processes, or to formulate hypothesesfor direct testing. Individual physiological systems functionin a milieu provided by the integrated activities of all ofthe systems to adapt, adjust and sustain the organism in itsenvironment. The overlapping interfaces between the differentphysiological systems provide fertile ground for new insightsand to enhance our knowledge. These interdisciplinary areasare of great importance if we are to understand the full complexityof organismal function. Of particular interest are the interactionsbetween the reproductive system and the immune system. The reproductivesystem is unique in that its primary role is to assure the continuityof the species, while the immune system provides internal protectionand thus facilitates continued health and survival. The modusoperandi of these 2 morphologically diffuse systems involveswidely distributed chemical signals in response to environmentalinput, and both systems must interact for the normal functioningof each. While the major focus of reproductive–immuneresearch has historically been with mammals, and has providedsubstantial insight into the interactions between these physiologicalsystems, comparative studies offer unique perspectives. Further,dysregulation of normal physiological interactions between thereproductive and immune systems can lead to disorders and diseaseseffecting one system or the other. Interactions between the immune system and reproductive system have important consequences for fertility and reproductive health in general. There is increasing evidence that many of the interactions between the immune and reproductive systems involve the Toll-like receptors (TLRs). While there is no doubt that TLRs are important in providing protection against infection in the reproductive tract, there is increasing evidence for the involvement of TLRs in more basic pathology and physiology of reproduction. In the female, TLRs have been implicated in critical aspects of ovarian, endometrial and placental function, as well as in ovarian cancer, pelvic inflammatory disease, intrauterine growth restriction, pre-eclampsia and preterm birth. In the male, TLRs appear to play a role in the control of testicular steroidogenesis and spermatogenesis in disease and, potentially, during normal function, as well. Recent studies also have begun to highlight the role of various TLRs in the aetiology of prostatitis and prostatic cancer. Given the nascent state of knowledge concerning this important area, it is clear that more studies are needed, which should provide valuable new insights into the biology of the TLRs and reproductive function in general.  相似文献

A major challenge in biology is understanding how organisms partition limited resources among physiological processes. For example, offspring production and self-maintenance are important for fitness and survival, yet these critical processes often compete for resources. While physiological trade-offs between reproduction and immune function have been documented, their regulation remains unclear. Most current evidence suggests that physiological changes during specific reproductive states directly suppress various components of the immune system; however, some studies have not found this clear relationship. We performed two experiments in female tree lizards (Urosaurus ornatus) that demonstrate the presence of trade-offs between the reproductive and immune systems under controlled laboratory conditions. These results also support the hypothesis that these trade-offs are a facultative response to resource availability and are not obligatory responses to physiological changes during reproduction. We found that (1) experimentally increasing reproductive investment under limited resources resulted in suppressed immune function and (2) experimentally limiting resources resulted in immunosuppression but only during resource costly reproductive activities. There seems to be a critical balance of resources that is maintained between multiple processes, and changes in the balance between energy intake and output can have major consequences for immune function.  相似文献

Myzostomids are minute, soft-bodied, marine worms associated with echinoderms since the Carboniferous. Due to their long history as host-specific symbionts, they have acquired a highly derived body plan that obscures their phylogenetic affinities to other metazoans. Because certain organs are serially arranged a closer relationship between polychaetes and myzostomids has repeatedly been discussion. We presented here a review on the ultrastructure of myzostomids with the most recent analyses that concern their phylogenetic position. The ultrastructure of the integument, digestive system, excretory system and nervous system are summarized. Unpublished information on the gametogenesis and reproductive systems of myzostomids are also exposed with a view on their reproductive process.  相似文献

Abstract The importance of genetic and environmental variation in condition in shaping evolutionary trade‐offs have recently been subject to much theoretical discussion, but is very difficult to investigate empirically in most field‐based systems. We present the results from mechanistic experimental manipulations of reproductive investment and condition in two female colour morphs (‘orange’ and ‘yellow’) of side‐blotched lizards (Uta stansburiana). We investigated the interactions between throat colour morphs, condition, local social environment and female survival using path‐analysis. Using follice‐ablation experiments, we show that large clutch size has a negative effect on field survival among yellow females, and that this effect is partly mediated by immunosuppressive effects of large clutches. In orange females these effects were less pronounced, and there was a negative survival effect of strong antibody responses. Hence, we experimentally confirmed our previous findings of correlational selection between female morphotype and immunocompetence, an important condition trait. Manipulation of corticosterone revealed multiple (‘pleiotropic’) direct and indirect effects of this hormone on both condition and reproductive traits. We argue that interaction effects (e.g. between local environments and genotypes) could explain a substantial fraction of variation in condition and reproduction in natural populations. Increased attention to such interaction effects and their fitness consequences will provide novel insights in field studies of selection and reproductive allocation.  相似文献

The role of a prominent surface-exposed loop (residues 88-97) in the alpha domain of streptokinase (SK), in human plasminogen (HPG) activation was explored through its selective mutagenesis and deletion studies. We first made a conformationally constrained derivative of the loop by the substitution of sequences known to possess a strong propensity for beta-turn formation. The mutant so formed (termed SK(88-97-Beta Turn)), when tested for co-factor activity against substrate HPG, after first forming a 1:1 molar complex with human plasmin (HPN), showed a nearly 6-fold decreased co-factor activity compared to the wild-type, native SK. The major catalytic change was observed to be at the k(cat) level, with relatively minor changes in K(m) values against HPG. Real-time binary interaction (i.e. the 1:1 complexation between SK, or its mutant/s, with HPG), and ternary complexation studies (i.e. the docking of a substrate HPG molecule into the preformed SK-HPG complex) using Surface Plasmon Resonance were done. These studies revealed minor alterations in binary complex formation but the ternary interactions of the substitution and/or deletion mutants were found to be decreased for full-length HPG compared to that for native SK.HPG. In contrast, their ternary interactions with the isolated five-kringle domain unit of plasminogen (K1-5) showed K(d) values comparable to that seen with the native SK.HPG complex. Taking into consideration the overall alterations observed in catalytic levels after site-specific mutagenesis and complete loop deletion of the 88-97 loop, on the one hand, and its known position at the SK-HPG interface in the binary complex, suggests the importance of this loop. The present results suggest that the 88-97 loop of the alpha domain of SK contributes towards catalytic turn-over, even though its individual contribution towards enzyme-substrate affinity per se is minimal.  相似文献

The oviduct is a key component of the reproductive system where essential states such as spermatozoa capacitation, fertilization and early embryo development take place. Recently, an additional role for the entire female reproductive tract has emerged with important implications for our understanding and management of reproductive health, namely, its role in mounting local immune responses against microbial pathogens. Most of the evidence about mucosal immune responses in the female reproductive tract is related to the vagina, with less information available for the uterus. The less known segment in this regard is the oviduct, which prompted us to review and summarize the current state of knowledge on the immune system at the level of the mammalian oviduct. A comprehensive search was conducted in Medline and--for the last two years--also in Current Contents. Result demonstrate that the mammalian oviduct is endowed with many of the elements that characterize a mucosal immune system. However, there are gaps in our knowledge that, in addition to important interspecies differences, make it essential to explore further some fundamental questions regarding this system.  相似文献

The free radical gas, nitric oxide is now known to be an important biological messenger in animals. Signal transmission by a gas that is produced by one cell, penetrates through membranes and regulates the function of another cell, represents new principles for signalling in biological systems. Nitric oxide is synthesised from L-arginine by enzyme nitric oxide synthase, which exists in multiple isoforms in a wide range of mammalian cells. Studies conducted in recent years point at a strong influence of NO in a wide range of reproductive functions. It is implicated in the control of gonadotrophin secretion at both hypothalamic and hypophyseal levels, LH surge mechanism, sexual behaviour, estradiol synthesis, follicle survival and ovulation. While considerable work lies ahead in unravelling the role of NO at the peripheral, cellular and molecular level in the domestic animal reproduction, findings presented in this review provide a general overview of growing appreciation of NO as a vital molecule controlling hypothalamic-pituitary-gonadal (HPG) axis.  相似文献

How does living in a social environment influence the brain? In particular, we ask the following questions: How do animals perceive and use social information? How does the perception of social information influence the reproductive system? Where is this represented in the brain? We present a model system in which these questions can be addressed, focusing on the brain's role in integrating information. In the social fish, Astatotilapia burtoni (Haplochromis), the relationship between social status and gonadotropin-releasing hormone (GnRH1) has been well established. Change in status results in numerous changes in the physiology of A. burtoni at every level of organization. Social status can regulate reproduction via the hypothalamic-pituitary-gonadal (HPG) axis. GnRH1 is used by the brain to signal the pituitary about reproductive state so reproductive control depends on regulation of this signaling peptide. In this fish, social dominance is tightly coupled to fertility. Here, we have exploited this link to understand the regulatory systems from circulating hormones, brain volume to gene expression.  相似文献

Trade-offs are widespread between life-history traits, such as reproduction and survival. However, their underlying physiological and behavioral mechanisms are less clear. One proposed physiological factor involves the trade-off between investment in male reproductive effort and immunity. Based on this hypothesis, we investigated differences in fitness between artificially selected immune response bank vole groups, Myodes glareolus . Significant heritability of immune response was found and a correlated response in testosterone levels to selection on immune function. Male reproductive effort, reproductive success, and survival of first generation offspring were assessed and we demonstrate a relationship between laboratory measured immune parameters and fitness parameters in field enclosures. We identify a trade-off between reproductive effort and survival with immune response and parasites as mediators. However, this trade-off results in equal male fitness in natural conditions, potentially demonstrating different male signaling strategies for either reproductive effort or survival. Females gain indirect genetic benefits for either genetic disease resistance or male reproductive effort, but not both. Immune response is genetically variable, genetically linked to testosterone and may indirectly maintain genetic variation for sexually selected traits. Evidence for both a genetic and a field trade-off between reproductive effort and survival indicates an evolutionary constraint on fitness traits.  相似文献

The specific mechanism regulating reproduction in invertebrates is a field of topical interest which needs to be explored in detail considering also the intriguing possible comparison with vertebrates. In this paper levels of Testosterone (T) and Estradiol (E2) and their reciprocal ratios were determined in ovaries and testis of the echinoid model species Paracentrotus lividus during the year 2004 by taking into account a putative relationship between steroid levels and reproductive cycle. T levels appeared to significantly vary during male reproductive cycle, thus suggesting a possible role of this hormone in regulation of spermatogenesis as demonstrated for other echinoderms. E2 levels were lower in males with respect to females; consequently E2 involvement in oogenesis is hypothesized. In parallel with steroid levels evaluation, variations in P450-aromatase activity and its possible role on regulation of gametogenesis were also considered. Clear correlations between steroid levels and gonad index (GI), as well as between GI and reproductive cycle were not detected, suggesting that GI alone is not a reliable parameter in describing the reproductive status of the gonads. Altogether the results obtained so far confirm the presence of a relationship between steroid levels and reproductive cycle as suggested by previous results on different echinoderm species.  相似文献

Hormonal control systems are complex in design and well integrated. Concern has been raised that these systems might act as evolutionary constraints when animals are subject to anthropogenic environmental change. Three systems are examined in vertebrates, especially birds, that are important for assessing this possibility: (i) the hypothalamic-pituitary-gonadal (HPG) axis, (ii) the activational effects of sex steroids on mating effort behaviour, and (iii) sexual differentiation. Consideration of how these systems actually work that takes adequate account of the brain's role and mechanisms suggests that the first two are unlikely to be impediments to evolution. The neural and molecular networks that regulate the HPG provide both phenotypic and evolutionary flexibility, and rapid evolutionary responses to selection have been documented in several species. The neuroendocrine and molecular cascades for behaviour provide many avenues for evolutionary change without requiring a change in peripheral hormone levels. Sexual differentiation has some potential to be a source of evolutionary inertia in birds and could contribute to the lack of diversity in certain reproductive (including life history) traits. It is unclear, however, whether that lack of diversity would impede adaptation to rapid environmental change given the role of behavioural flexibility in avian reproduction.  相似文献

The role of the maternal immune system during pregnancy has focused mainly on the aspect of immune tolerance to the invading trophoblast and, therefore, fetus. While this is a critical aspect of reproductive immunology, it is also important to consider the function of the maternal immune system in the promotion of implantation and maintenance of pregnancy. Apoptosis or cell death is not the final stage in tissue development. The quick and effective removal of apoptotic cells by tissue macrophages represents a vital process preventing "leak" of self-antigens and promoting the production of proliferative/survival factors. One of the key requirements of apoptotic cell clearance is the resolution of inflammatory conditions, which, as in the case of pregnancy, may have lethal consequences. This review will focus on decidual macrophages and their role on apoptosis and cell clearance during pregnancy.  相似文献

The hypothalamic–pituitary–gonadal axis (HPG) plays vital roles in reproduction and steroid hormone production in both sexes. The focus of this review is upon gene structures, receptor structures and the signaling pathways of gonadotropin-releasing hormone (GnRH), luteinizing hormone (LH) and follicle-stimulating hormone (FSH). The hormones' functions in reproduction as well as consequences resulting from mutations are also summarized. Specific characteristics of hormones such as the pulsatile secretions of GnRH are also covered. The different regulators of the HPG axis are introduced including kisspeptin, activin, inhibin, follistatin, androgens and estrogen. This review includes not only their basic information, but also their unique function in the HPG axis. Here we view the HPG axis as a whole, so relations between ligands and receptors are well described crossing different levels of the HPG axis. Hormone interactions and transformations are also considered. The major information of this article is depicted in three figures summarizing the current discoveries on the HPG axis. This article systematically introduces the basic knowledge of the HPG axis and provides information of the current advances relating to reproductive hormones.  相似文献
