Water stress is the major factor limiting plant productivity and quality in most regions of the world. In the present study, a two-year field experiment was conducted to determine the influence of putrescine (Put) on phytochemical, physiological, and growth parameters of Salvia officinalis L. under different irrigation regimes. The highest stem dry weight (56.05 and 65.21 g m−2) plus leaf dry weight (124.51 g m−2) were predicted in irrigation regimes of (20 and 40%) plus 20% available soil water was depleted (ASWD), respectively. Total phenolic content (TPC) was increased significantly under the irrigation regime of 80% with the application of distilled water in spring. TPC showed an increasing trend with increases in Put concentration under all irrigation regimes in both spring and summer. The highest total flavonoids content (TFC) in wavelengths of 415 and 367 nm were predicted in 2.25 mM Put. The highest ascorbate peroxidase (APX) activity (0.13 μmol mg−1 protein) was predicted in the irrigation regime of 20% with the application of distilled water in spring and summer. There was a significantly negative correlation coefficient between APX, TPC, and TFC. Indeed, there was a decreasing trend in APX and an increasing trend in TPC and TFC with increases in Put concentration under the irrigation regime of 20% ASWD. The highest hydroxyl radical scavenging activity (HRSA) values were obtained under irrigation regimes of 49.27% and 20% ASWD in spring and summer, respectively. There was an increasing trend in endogenous Put with increases in the Put concentration. The responses of compatible osmolytes to irrigation regime can be expressed by quadratic model, suggesting maximum proline (0.52 mg g−1), total reducing sugars (TRS) (0.37 mg g−1), xylose (0.68 mg g−1), and mannose (0.37 mg g−1) values would be obtained in irrigation regimes of 68.33%, 48.33%, 53.75%, and 56.25% ASWD, respectively.  相似文献

The study presents antioxidant, phytochemical, anti-proliferative, and gene repression activities against Hypoxia-inducible factor (HIF-1) alpha and Vascular endothelial growth factor (VEGF) of Elaeocarpus sphaericus extract. Elaeocarpus sphaericus dried and crushed plant leaves were extracted using water and methanol by ASE (Accelerated Solvent Extraction) method. Total phenolic content (TPC) and total flavonoid content (TFC) were used to measure the extracts′ phytochemical activity (TFC). Antioxidant potential of the extracts was measured through DPPH, ABTS, FRAP, and TRP. Methanolic extract of the leaves of E. sphaericus has shown a higher amount of TPC (94.666±4.040 mg/gm GAE) and TFC value (172.33±3.21 mg/gm RE). The antioxidant properties of extracts in the yeast model (Drug Rescue assay) showed promising results. Ascorbic acid, gallic acid, hesperidin, and quercetin were found in the aqueous and methanolic extracts of E. sphaericus at varying amounts, according to a densiometric chromatogram generated by HPTLC analysis. Methanolic extract of E. sphaericus (10 mg/ml) has shown good antimicrobial potential against all bacterial strains used in the study except E. coli. The anticancer activity of the extract in HeLa cell lines ranged from 77.94±1.03 % to 66.85±1.95 %, while it ranged from 52.83±2.57 % to 5.44 % in Vero cell lines at varying concentration (1000 μg/ml–31.2 μg/ml). A promising effect of extract was observed on the expression activity of HIF-1 and VEGF gene through RT-PCR assay.  相似文献

Propolis is known to exhibit various phytochemical compounds that aid in several biological activities. The current study investigates the phytochemical compounds of ethanolic extract of propolis of Tetrigona apicalis (EEP) using Q-TOF LC-MS, its antioxidant properties using DPPH and ABTS+ radical scavenging assays, total phenolic (TPC) and flavonoid content (TFC), using Folin-Ciocalteu and Aluminium Chloride method, respectively, as well as proapoptotic effects, based on the selected IC50 of the cytotoxic study conducted for EEP using annexin V-FITC assay. Terpene and polyphenol were among of 17 identified compounds. The EC50 of EEP for DPPH and ABTS+ was 1.78 mg/mL and 1.68 mg/mL, while the EEP exhibited TPC and TFC values of 31.99 mgGAE/g and 66.4 mgQCE/g, respectively in which the parameters were strongly correlated. The IC50 of EEP effectively induces apoptosis in MCF7 cells. In conclusion, EEP possessed important phytochemical compounds that work excellently as antioxidants and anticancer agents.  相似文献

The present experiment was conducted to study effects of Cu, Fe and Zn on activities of digestive enzymes of the hybrid tilapia Oreochromis niloticus×Oreochromis aureus. The acidic protease activities increased 65·5 and 55·1% by addition of homogenates of digesta‐containing stomach with copper (75 mg l−1) and zinc (50 mg l−1) respectively. Addition of Cu and Zn increased the activities of protease in the hepatopancreas homogenates by 132·7 and 38·1% respectively, and reduced the activity of protease in the digesta‐containing intestine homogenates by 11·0 and 13·8% respectively. Addition of Fe (50 mg l−1) increased the acidic protease activity by 96·7% but did not alter the activities of protease in the intestine and hepatopancreas. Addition of Cu markedly inhibited activities of amylase in intestine and hepatopancreas homogenates, while Zn addition showed no effects. Addition of Fe reduced activities of amylase in the intestine homogenates by 47·9% but had no effect on amylase activities in the hepatopancreas. When Cu (75 mg kg−1), Fe (50 mg kg−1) and Zn (50 mg kg−1) were supplemented to basal diet for 3 weeks, the activities of amylase in hepatopancreas homogenates increased 125·3, 215·6 and 70·0%, respectively, the activities of amylase in intestine increased 79·8, 74·6 and 48·5%, respectively, and the activities of lipase in intestine increased 90·5, 149·8 and 84·0%, respectively. Supplementation of Cu, Fe or Zn into diet had no effects on activity of protease in all digestive organs. Therefore, the results suggest that effects of Cu, Fe and Zn on activity of digestive enzymes in vitro were different from those seen in vivo, and that the positive effects of Cu, Fe and Zn supplemented to fish diet would be valuable information for formulating fish feed.  相似文献

The aim of this study was to determine the effects of Tribulus terrestris extract (TT) on growth performance, disease resistance and histopathological changes in intestine and liver tissues of Oreochromis mossambicus (Peters, 1852) first‐feeding fry before and after exposure to Streptococcus iniae. Five isonitrogenous and isocaloric diets were formulated to contain 0 (control), 200, 400, 600, and 800 mg kg?1 TT. After feeding for 45 days, fish were infected with S. iniae and mortalities recorded. Final weight, weight gain and SGR of tilapia fry fed the 400 mg kg?1 TT diet were significantly greater than that of control diet. In the challenge experiment, the best survival rate was obtained with 400 mg kg?1 TT supplementation. Infection by S. iniae appeared to have a negative effect on histopathological findings and outcome than did TT‐800 used alone. However, administration of TT (200 or 400 mg extract kg?1) resulted in overall improvement in the intestine and liver histopathology, emphasizing the protective potential of TT. The present study suggests the protective potential of TT in alleviating intestinal and hepatic damage that can occur after a S. iniae infection. It was concluded that 400 mg kg?1 TT can enhance growth and disease resistance during first–feeding of O. mossambicus fry. This suggests that TT may be an alternative to antibiotics in controlling streptococcal disease in tilapia culture.  相似文献

Plants have always been the prime focus in medicine industries due to their enormous ethnobotanical uses and multitude of biological and therapeutic properties. In the current study, preliminary phytochemical composition, Total phenolic content (TPC), and total flavonoid content (TFC) with the antioxidant and antibacterial activity of hydroalcoholic extract and n-hexane, chloroform and n-butanol fractions of five selected medicinal plants [Tephrosia purpurea (L.) Pers., Lavandula stoechas L., Aesculus indica (Wall. ex Cambess.) Hook, Iris ensata Thunb., and Kalanchoe pinnata (Lam.) Pers.] from Pakistan, have been evaluated. TPC and TFC were determined by Folin-Ciocalteu’s and AlCl3 methods respectively. The antioxidant activity was performed by DPPH, ABTS, FRAP, and CUPRAC while the antibacterial potential of these plants was determined by agar well diffusion assay. K. pinnata (Lam.) Pers. exhibited the highest TPC (695 ± 13.2 mg.GA.Eq.g-1DE ± SD) in n-butanol fraction and the highest TFC in its chloroform faction (615 ± 6.31 mg Q.Eq.g−1 DE ± SD). The n-butanol fraction and hydroalcoholic extract of I. ensata Thunb. exhibited strong antioxidant potential by DPPH and CUPRAC assays respectively, whereas K. pinnata (Lam.) Pers. n-butanol fraction exhibited the strongest reducing potential. The hydroalcoholic extract of all tested plants exhibited significant antibacterial activity against tested bacterial strains with ZI (12–18 mm). Conclusively, K. pinnata (Lam.) Pers. (Family: Crassulaceae) and I. ensataThunb. (Family: Iridaceae) exhibited the highest antioxidant and antibacterial potential. They can be explored for the isolation of phytoconstituents responsible for this potential and serve as a lead for the production of new natural antioxidants and antibacterial agents that can be used to cure various diseases.  相似文献

Total phenolic (TPC) and flavonoid (TFC) content, individual phenolic compounds and antioxidant activities of methanol extracts of wheat and corn straw were determined. Germination bioassay was conducted with Abutilon theophrasti Medik. , Asclepias syriaca L., and Chenopodium album L. seed. Samples were fermented by Lactobacillus plantarum and changes in TPC, TFC, antioxidant, and biological activity were investigated. TPC and TFC were significant in both samples and after fermentation their recovery was improved. All samples contain mainly quercetin, cinnamic acid, p‐coumaric acid, and ferulic acid. Fermentation changed the content of phenolic and flavonoid compounds, differently in each case. All tested extracts showed high DPPH activity with IC50 being significantly lower for fermented samples. FRAP activity was also high. Crude straw extracts were overall more effective than fermented ones concerning inhibition of germination and seedlings growth, mainly without statistically significant differences between wheat and corn. Compared with mesotrione, extracts were more effective in germination and seedling growth inhibition of C. album and in seedling growth inhibition of A. theophrasti.  相似文献

Salacia chinensis L. has various beneficial properties including antidiabetic and antioxidant activity. The S. chinensis fruit pulp (SCFP) was extracted with four different solvents (Methanol, ethanol, acetone and water) and was screened for total phenolic content (TPC), total flavonoid content (TFC) and antioxidant activity (AOA). The AOA was assessed by evaluating the 1, 1-diphenyl-2-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP) and metal chelating assay. Methanolic SCFP extract exhibited the highest phenolic content (3.20?±?0.12 mg GAE/g FW) whereas, ethanolic extract showed highest flavonoid content (0.31?±?0.68 mg RE/g FW). The methanolic extract possesses highest antioxidant activity towards DPPH (92.44 %), FRAP (1.939 O.D) and metal chelating activity (74.16 %). AOA (DPPH and FRAP) was significantly correlated with TPC. The results indicated that SCFP is a good natural source of antioxidant compounds for use in food and pharmaceutical industry.  相似文献

In our study, Central and Eastern European bee pollens of different botanical origins were compared, based on their antioxidant and color properties. Total phenolic content (TPC), total flavonoid content (TFC), and in vitro antioxidant capacity (by FRAP, CUPRAC, ABTS⋅+ and DPPH⋅ assays) were determined spectrophotometrically. Besides, Relative Antioxidant Capacity Indexes (RACI) were calculated. CIELAB color parameters (L*, a*, b*, chroma) were determined by using a tristimulus-based instrument. Potential correlations between the investigated parameters were also identified. Based on the results of the preliminary study, ethanol:distilled water (60 : 40) was chosen as an extraction solvent. The total phenolic content of our samples ranged between 9.41 and 27.49 mg GAE/g dw. Pollens showed TFC:TPC ratios between 9 and 44 %. RACI values indicate that rapeseed (Brassica napus), traveller's joy (Clematis vitalba) and phacelia (Phacelia tanacetifolia) pollens have relatively high, while pollens of certain plants of the Asteraceae family possess low antioxidant potential. Antioxidant properties correlated significantly in most cases. RACI values showed strong positive correlation with each of the other antioxidant capacity parameters, suggesting that this approach is well applicable for comparing the antioxidant potential of bee pollens. No clear correlation was found between the antioxidant and color parameters.  相似文献

The LD50 of roselle calyx extract and its effect on blood pressure were determined. The LD50 was found to be above 5000 mg kg−1. Roselle calyx infusion was found to lower significantly (p<0·05) both systolic and diastolic pressure in spontaneously hypertensive and normotensive Wistar–Kyoto rats at tested doses of 500 and 1000 mg kg−1 body weight. The reduction in blood pressure in both groups was positively correlated with weight. Continuous consumption of the infusion at 1000 mg kg−1 was discovered to lead to sudden death in spontaneously hypertensive rats but not in Wistar‐Kyoto rats. Water intake was not significantly different (p>0·05) in the control groups of the two strains of rats used, neither was there a significant difference in their urine output. The water intake in the treated spontaneously hypertensive and normotensive rats was not different from the corresponding control groups. However the urine output of the treated spontaneously hypertensive rats was significantly higher. A significant decrease in serum creatinine, cholesterol, and glucose in the treated rats compared with the control as well as a significant increase in serum uric acid was observed. The serum proteins (albumin and total protein) in the treated rats when compared with the control groups was not changed significantly. Copyright © 1999 John Wiley & Sons, Ltd.  相似文献

The aim of present work was to assess the total phenolic content (TPC), total flavonoid content (TFC), phenolic compounds and antioxidant properties of various extracts of three Ceropegia spp.: Ceropegia spiralis, Ceropegia panchganiensis and Ceropegia evansii from Western Ghats of India. TPC of the samples varied from 0.3 ± 0.2 to 28.5 ± 0.3 mg TAE/g FW, whereas, TFC of the samples ranged between 0.1 ± 0.1 and 15.3 ± 0.3 mg RE/g FW. The major phenolic compounds identified were gallic acid, vanillin, cathechol and ferulic acid. All the extracts possess 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, ferric reducing antioxidant power (FRAP) as well as metal chelating ability and this was also supported by significant correlation with TPC and TFC. To the best of our knowledge, this is the first paper presenting comprehensive data on TPC, TFC, phenolic profile and antioxidant properties of the Ceropegia spp.  相似文献

Diabetes is a chronic, lifelong condition due to inadequate production of insulin or the cells does not properly respond it. Recently, the significance and effectiveness of herbal drugs associated with diabetes has emerged. The aim of the present study was to determine the anti-diabetic effects of Terminalia catappa L. leaves on streptozotocin (STZ)-treated rats. Two different concentrations of ethanolic leaf extract (300 and 500 mg/kg) of T. catappa were used to treat diabetic rats, and biochemical parameters were analyzed in blood samples. The results of herbal treatments were compared with the standard drug, glibenclamide. The ethanol extract (500 mg/kg) had significant anti-diabetic activity by altering blood glucose, glycosylated hemoglobin, liver glycogen, glucose 6-phosphatase, fructose 1,6-bisphosphatase, glucokinase, aspartate transaminase, alanine transaminase, alkaline phosphatase, urea, uric acid and creatinine levels while increasing insulin levels. Thus, the present study suggests that the supplementation of the diabetic patients with T. catappa leaves can lead to recovery from diabetic effects.  相似文献

This article presents an optimized ultrasound-assisted ethanolic extraction (UAEE) and characterization of selected high-value components from Gemlik olive fruit (GOF) harvested from Potohar region of Pakistan. Response surface methodology (RSM), involving central composite design (CCD), was applied to optimize the extraction variables i. e., temperature (25–65 °C), extraction time (15–45 min) and aqueous ethanol concentration (60–90 %) for optimal recovery of bioactives extract, total phenolic contents (TPC) and DPPH free radical scavengers. Under the optimized set of conditions such as 43 °C temperature, 32 min extraction time and 80 % aqueous ethanol, the best extract yield (218.82 mg/g), TPC (19.87 mg GAE/g) and DPPH scavenging activity (63.04 %) were recorded. A quadratic polynomial model was found to be reasonably fitted to the observed results for extract yield (p<0.0001 and R2=0.9941), TPC (p<0.0001 and R2=0.9891), and DPPH radical scavenging activity (p<0.0001 and R2=0.9692). Potent phenolic compounds were identified by GC/MS in GOF extract and considerable amount of essential fatty acids were also detected. The current findings support the use of UAEE as an effective green route for optimized recovery of high-value components from GOF and hence its applications can be extended to functional food and nutra-pharmaceutical developments.  相似文献

Cylindrospermopsin (CYN) is a cyanotoxin that has raised serious concerns about public health in many parts of the world. It can bioaccumulate and affect the health of aquatic organisms, but despite this, few studies have been conducted on CYN uptake and clearance in fish. In this paper, the authors evaluate the uptake and clearance of CYN in the muscle tissue and viscera of juvenile tilapia (Oreochromis niloticus) after exposure to aqueous extracts and whole cells of Cylindrospermopsis raciborskii (CYN-producer). CYN blended with commercial fish food, and three experiments were conducted. In the first trial, fish food, and aqueous extracts containing 0.31 μg CYN g−1 of food per day, was administered to tilapia for 15 days. In the second trial, fish were provided food and intact cells (5.4 μg CYN g−1 of food per day) for 15 days. In the last trial, they were provided fish food and aqueous extracts (0.8 μg CYN g−1 of food per day) for 12 days, and for the next 10 days, the animals were fed food without toxic cell extracts (to simulate a clearance period). The concentration of CYN in muscle tissue and viscera was analysed using ELISA. In the case of juvenile tilapia, the presence of CYN was higher in viscera than in muscle tissue, and the toxin remained in the tissues even after 10 days without the addition of contaminated food. The results suggest that tilapia represents a potential source of CYN transfer through the food web, and this shows the need for a continuous monitoring of this compound in organisms that are used for human and animal consumption.  相似文献

In this study, chemical composition and in vitro biological activities of artichoke by‐products (leaves, floral stems and bracts) issued from two Tunisian varieties were evaluated. Analysis was performed by means of high‐performance liquid chromatography with diode array detection coupled to electrospray ionization mass spectrometric (LC/DAD/ESI‐MS). Total phenolic (TPC) and flavonoid (TFC) contents as well as the antioxidant activity conducted by three complementary methods, DPPH, ABTS and FRAP tests, were performed for each sample. Enzyme inhibitory effects against acetylcholinesterase, butyrylcholinesterase and α‐amylase were also studied. Results showed that TPC and TFC varied according to variety as well as the plant part. Bracts presented the highest TPC values (10–15 mg GAE/g DW), while leaves were distinguished by the highest TFC values (52–58 mg EQ/g DW). In vitro assays showed that Violet d'Hyères bracts and Blanc d'Oran leaves present the most antioxidant activities (30.040 and 20.428 mgET/gDW, respectively, by the DPPH method). Leaves demonstrated the highest acetylcholinesterase and butyrylcholinesterase inhibitory effects. Moreover, all organs displayed a noticeable inhibition towards α‐amylase. LC/DAD/MS analysis revealed that artichoke by‐products are a potential source of biopharmaceuticals such as luteolin derivatives from leaves and mono/dicaffeoylquinic acids in the other parts. This research demonstrates that globe artichoke by‐products, unexploited in our country, are a promising source of natural health promoting compounds with potential applications in the food and pharmaceutical industries.  相似文献

The effects of two common West African farm-made feeds on postprandial metabolism were assessed in adult Nile tilapia (Oreochromis niloticus) by quantifying the oxygen consumption rates (MO2) and total ammonia-nitrogen (TAN) excretion over a 24-h period. Measurements followed the ingestion of a single meal of a fishmeal-based control diet and two diets containing 30% copra (CM) or palm kernel meal (PKM) inclusions by groups of 15 adult O. niloticus per tank. The mean net MO2 (postprandial oxygen consumption corrected for routine metabolic rate) during digestion for the different tilapia groups varied narrowly between 112.2 ± 9.9 and 129.9 ± 20.4 mg O2 kg?1 h?1 with the fish fed the CM diet recording a significantly lower (p = 0.04) net MO2 response relative to the other two diets. Net TAN excretion rates of the different dietary groups varied between 3.4 ± 1.4 and 4.4 ± 1.6 mg TAN kg?1 h?1. Under the standardized experimental conditions, copra and PKMs appeared to be promising candidates as partial replacements to fishmeal in tilapia diets as far as rates of oxygen consumption and ammonia-nitrogen excretion are concerned.  相似文献

Litsea elliptica is traditionally believed to prevent and treat stomach ulcers, cancer, fever and headaches. This study investigates the phytochemical composition, antioxidant and cytotoxic effects of L. elliptica leaf extracts. The phytochemical content was determined via GCMS analysis and total phenolic content (TPC) and total flavonoid content (TFC) were analysed using the Folin-Ciocalteu and aluminium-chloride assays. Antioxidant activities were determined using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging, 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) scavenging and ferric-ion reducing antioxidant power (FRAP) assays, whereas cytotoxicity was determined using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and calcein/ethidium viability assays. The mechanism of cytotoxicity was investigated using Annexin V/propidium iodide. Modifications in the mitochondria were investigated using MitoTracker Red CMXRos. Ten and twenty-six compounds were characterized in the young-leaf and mixed-leaves extracts, respectively. The young-leaf methanolic extract demonstrated the highest antioxidant capacity of at least four-folds greater than the mixed-leaves and ethanolic extracts. The methanolic extract also had higher TPC and TFC values compared to the ethanolic extract. Although the mixed L. elliptica leaves had lower antioxidant capacities compared to the young leaves, the mixed leaves extract has demonstrated greater cytotoxicity against the A549 cancer cell line. Further investigation revealed that the L. elliptica leaves-induced cytotoxicity on A549 cells was possibly via the non-inflammatory mitochondria-mediated apoptotic pathway. Overall, our results showed the potential of the L. elliptica leaves possessing cytotoxic activities against carcinoma cells where the compounds present can be further investigated for its therapeutic application.  相似文献

Hardy kiwifruit (Actinidia arguta) is a highly appreciated exotic fruit endowed with outstanding bioactive compounds. The present work proposes to characterize the pulp from A. arguta organic fruits, emphasizing its radicals scavenging capacity and effects on intestinal cells (Caco-2 and HT29-MTX). The physicochemical properties and phenolic profile were also screened. The total phenolic and flavonoid contents (TPC and TFC, respectively) of pulp were 12.21 mg GAE/g on dry weight (DW) and 5.92 mg CE/g DW, respectively. A high antioxidant activity was observed (FRAP: 151.41 μmol FSE/g DW; DPPH: 12.17 mg TE/g DW). Furthermore, the pulp did not induce a toxic effect on Caco-2 and HT29-MTX cells viability up to 1000 μg/mL. Regarding in vitro scavenging capacity, the pulp revealed the highest scavenging power against NO. (IC50=3.45 μg/mL) and HOCl (IC50=12.77 μg/mL). These results emphasize the richness of A. arguta fruit pulp to be used in different food products.  相似文献

Ultrasonic-assisted extraction (UAE) was developed to extract phenolic and flavonoid antioxidants from Clerodendrum cyrtophyllum Turcz leaves. The optimal experimental parameters for antioxidant extraction from C. cyrtophyllum leaves were measured using single-factor experimentation combined with response surface methodology (RSM). Total phenolic content (TPC) and total flavonoid content (TFC) assays were used to quantify antioxidant compounds. Next, antioxidant radical scavenging capacity was measured using 2,2′-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′ -azino-bis(3-ethylbenzothiazoline-6-sulphonicacid) (ABTS) radicals. Optimized extraction conditions for UAE from C. cyrtophyllum leaves were as follows: 60.9% ethanol, 85.4 min, and 63.3°C for maximal TPC extraction (16.8±0.2 mg GAE/g DW); 67.7% ethanol, 82.9 min, and 63.0°C for maximal TFC extraction (49.3±0.4 mg RT/g DW); 48.8% ethanol, 85.1 min, and 63.9°C for maximal DPPH radical-scavenging capacity (86.8±0.2%); and 50.6% ethanol, 81.3 min, and 63.4°C for maximal ABTS radical-scavenging capacity (92.9±0.5%). Ethanol concentration was the most important factor in the extraction process. Our work offers optimal extraction conditions for C. cyrtophyllum as a potential source of natural antioxidants.  相似文献

Free radical scavenging activity (FRSA), total phenolic content (TPC), and total flavonoid content (TFC) of in vitro cultured and field grown Withania somnifera (Ashwagandha) roots were investigated. Withanolides analysis and comprehensive metabolic profiling between 100% methanol extracts of in vitro and field grown root tissues was performed using high performance thin layer chromatography (HPTLC) and gas chromatography-mass spectrometry (GC-MS), respectively. Significantly higher levels of FRSA, TPC, and TFC were observed in in-vitro cultured roots compared with field grown samples. In addition, 30 day-cultured in vitro root samples (1MIR) exhibited a significantly higher FRSA (IC50 81.01 μg/mL), TPC (118.91 mg GAE/g), and TFC (32.68 mg CE/g) compared with those in 45 day-cultured samples (1.5MIR). Total of 29 metabolites were identified in in vitro cultured and field grown roots by GC-MS analysis. The metabolites included alcohols, organic acids, purine, pyrimidine, sugars, and putrescine. Vanillic acid was only observed in the in vitro cultured root samples, and higher level of the vanillic acid was observed in 1MIR when compared to 1.5MIR. Therefore, it is suggested that 1MIR might serve as an alternative to field grown roots for the development of medicinal and functional food products.  相似文献
