Nitrogen (N) is the key factor limiting photosynthetic processes and crop yield. Little is known about the response of leaf gas exchange of spring triticale (Triticosecale Wittm.) to N supply. The effect of N fertilizers on different gas exchange variables, i.e., photosynthetic rate (A), transpiration rate (E), stomatal conductance (g s), instantaneous water use efficiency (WUE) and maximum quantum yield of photosystem II (PSII) (F v/F m), chlorophyll index (SPAD, soil–plant analysis development), and the relationship of these variables with yield were studied in spring triticale grown under field conditions. Six treatments of N—0, 90, 180, 90 + 30, 90 + 30 + 30 kg ha?1 (applied as ammonium nitrate, AN) and one treatment of N 90 + 30 + 30 kg ha?1 (applied as urea ammonium nitrate solution, UAN) were compared. The analysis of variance showed that throughout the triticale growing season, N fertilization had significant effects on A, WUE, g s and SPAD. On average, N fertilizer application increased A values by 14–70%. E and F v/F m values were not influenced by N fertilization levels. The effect of growth stage and year on gas exchange variables and F v/F m and SPAD was found to be significant. At different growth stages, A values varied and maximum ones were reached at BBCH 31–33 (decimal code system of growth stages) and BBCH 59. With aging, values of A decreased independently of N fertilization level. The gas exchange variables were equally affected by both fertilizer forms. The interplay among grain yield, leaf gas exchange variables, F v/F m and SPAD of spring triticale was estimated. The statistical analysis showed that grain yield positively and significantly correlated with A and SPAD values throughout the growing season.  相似文献

The objectives of this study were to investigate stomatal regulation in maize seedlings during progressive soil drying and to determine the impact of stomatal movement on photosynthetic activity. In well-watered and drought-stressed plants, leaf water potential (Ψ leaf), relative water content (RWC), stomatal conductance (g s), photosynthesis, chlorophyll fluorescence, leaf instantaneous water use efficiency (iWUEleaf), and abscisic acid (ABA) and zeatin-riboside (ZR) accumulation were measured. Results showed that g s decreased significantly with progressive drought and stomatal limitations were responsible for inhibiting photosynthesis in the initial stages of short-term drought. However, after 5 days of withholding water, non-stomatal limitations, such as damage to the PSII reaction center, became the main limiting factor. Stomatal behavior was correlated with changes in both hydraulic and chemical signals; however, changes in ABA and ZR occurred prior to any change in leaf water status. ABA in leaf and root tissue increased progressively during soil drying, and further analysis found that leaf ABA was negatively correlated with g s (R 2 = 0.907, p < 0.05). In contrast, leaf and root ZR decreased gradually. ZR in leaf tissue was positively correlated with g s (R 2 = 0.859, p < 0.05). These results indicate that ABA could induce stomatal closure, and ZR works antagonistically against ABA in stomatal behavior. In addition, the ABA/ZR ratio also had a strong correlation with g s, suggesting that the combined chemical signal (the interaction between ABA and cytokinin) plays a role in coordinating stomatal behavior. In addition, Ψ leaf and RWC decreased significantly after only 3 days of drought stress, also affecting stomatal behavior.  相似文献

Pistachio (Pistacia vera L.) has a high tolerance to drought and soil salinity. Although adult pistachio trees are well known to be drought tolerant, the studies on physiological adaptation of pistachio cultivars to drought are limited. Therefore, three pistachio cultivars, i.e., Akbari, Kaleghochi, and Ohadi were subjected to three osmotic drought stress treatments: control (?0.1 MPa), moderate (?0.75 MPa) and severe drought (?1.5 MPa) stress using PEG 6000 for a 14-day period. All drought stress treatments decreased net photosynthesis (P n), stomatal conductance (g s), intercellular CO2 concentration (C i), and transpiration rate (E), but Ohadi maintained better its photosynthetic capacity compared to Akbari and Kaleghochi. Maximum quantum yield of PSII photochemistry (F v /F m), effective PSII quantum yield (ΦPSII) and photochemical quenching (qP) were also reduced. The chlorophyll fluorescence parameters indicated that Akbari was more susceptible to the applied drought stress. Drought stress levels decreased chlorophyll pigments, fresh weight, stem elongation, leaf nitrogen content (N), leaf water potential and increased water use efficiency (WUE). Proline increased strongly under drought stress for Akbari. After 2 weeks of stress a recovery of 2 weeks was applied. This period was insufficient to fully restore the negative effects of the applied stress on the studied cultivars. Based on the reduction of photosynthesis and the increase of the proline content Akbari seems more sensitive to the applied drought stress.  相似文献

The phytotoxic aluminum species (Al3+) is considered as the primary factor limiting crop productivity in over 40 % of world’s arable land that is acidic. We evaluated the responses of two wheat cultivars (Triticum aestivum L.) with differential Al resistance, cv. Yecora E (Al-resistant) and cv. Dio (Al-sensitive), exposed to 0, 37, 74 and 148 μM Al for 14 days in hydroponic culture at pH 4.5. With increasing Al concentration, leaf Ca2+ and Mg2+ content decreased, as well as the effective quantum yield of photosystem II (PSII) photochemistry (Φ PSII ), while a gradual increase in leaf membrane lipid peroxidation, Al accumulation, photoinhibition (estimated as F v /F m ), and PSII excitation pressure (1 ? q p ) occurred. However, the Al-resistant cultivar with lower Al accumulation, retained larger concentrations of Ca2+ and Mg2+ in the leaves and kept a larger fraction of the PSII reaction centres (RCs) in an open configuration, i.e. a higher ratio of oxidized to reduced quinone A (QA), than plants of the Al-sensitive cultivar. Four times higher Al concentration in the nutrient solution was required for Al-resistant plants (148 μM Al) than for Al-sensitive (37 μM Al), in order to establish the same closed RCs. Yet, the decline in photosynthetic efficiency in the cultivar Dio was not only due to closure of PSII RCs but also to a decrease in the quantum yield of the open RCs. We suggest that Al3+ toxicity may be mediated by nutrient deficiency and oxidative stress, and that Al-resistance of the wheat cultivar Yecora E, may be due at least partially, from the decreased Al accumulation that resulted to decreased reactive oxygen species (ROS) formation. However, under equal internal Al accumulation (exposure Al concentration: Dio 74 μM, Yecora E 148 μM) that resulted to the same oxidative stress, the reduced PSII excitation pressure and the better PSII functioning of the Al-resistant cultivar was probably due to the larger concentrations of Ca2+ and Mg2+ in the leaves. We propose that the different sensitivities of wheat cultivars to Al3+ toxicity can be correlated to differences in the redox state of QA. Thus, chlorophyll fluorescence measurements can be a promising tool for rapid screening of Al resistance in wheat cultivars.  相似文献

The Na+/H+ antiporters play an important role in salt tolerance in plants. However, the functions of OsNHXs in rice except OsNHX1 have not been well studied. Using the gain- and loss-of-function strategies, we studied the potential role of OsNHX2 in salt tolerance in rice. Overexpression of OsNHX2 (OsNHX2-OE) in rice showed the significant tolerance to salt stress than wild-type plants and OsNHX2 knockdown transgenic plants (OsNHX2-KD). Under salt treatments of 300-mM NaCl for 5 days, the plant fresh weights, relative water percentages, shoot heights, Na+ contents, K+ contents, and K+/Na+ ratios in leaves of OsNHX2-OE transgenic plants were higher than those in wild-type plants, while no differences were detected in roots. K+/Na+ ratios in rice leaf mesophyll cells and bundle sheath cells were higher in OsNHX2-OE transgenic plants than in wild-type plants and OsNHX2-KD transgenic plants. Our data indicate that OsNHX2 plays an important role in salt stress based on leaf mesophyll cells and bundle sheath cells and can be served in genetically engineering crop plants with enhanced salt tolerance.  相似文献

The plasticity response of Quercus variabilis and Quercus mongolica seedlings to combined nitrogen (N) deposition and drought stress was evaluated, and their performance in natural niche overlaps was predicted. Seedlings in a greenhouse were exposed to four N deposition levels (0, 4, 8, and 20 g N m?2 year?1) and two water levels (80 and 50 % field-water capacity). Plant traits associated with growth, biomass production, leaf physiology, and morphology were determined. Results showed that drought stress inhibited seedling performance, altered leaf morphology, and decreased fluorescence parameters in both species. By contrast increased N supply had beneficial effects on the nutritional status and activity of the PSII complex. The two species showed similar responses to drought stress. Contrary to the effects in Q. mongolica, N deposition promoted leaf N concentration, PSII activity, leaf chlorophyll contents, and final growth of Q. variabilis under well-watered conditions. Thus, Q. variabilis was more sensitive to N deposition than Q. mongolica. However, excessive N supply (20 g N m?2 year?1) did not exert any positive effects on the two species. Among the observed plasticity of the plant traits, plant growth was the most plastic, and leaf morphology was the least plastic. Therefore, drought stress played a primary role at the whole-plant level, but N supply significantly alleviated the adverse effects of drought stress on plant physiology. A critical N deposition load around 20 g N m?2 year?1 may exist for oak seedlings, which may more adversely affect Q. variabilis than Q. mongolica.  相似文献

Five isocaloric (430 kcal 100 g?1), isonitrogenous (40% CP) experimental diets were formulated with different concentrations of Bacillus licheniformis fb11 probionts (isolated from the gut of Chitala chitala) viz. Control (without probionts), 5 × 104 CFU g?1 (D1), 5 × 105 CFU g?1 (D2), 5 × 106 CFU g?1 (D3), 5 × 107 CFU g?1 (D4), 5 × 108 CFU g?1 (D5) to evaluate its efficiency in C. chitala juvenile. The best growth performance, feed utilisation, specific α-amylase, total protease and lipase activity were observed with the diet D3 (P < 0.05). The lowest Presumptive Pseudomonas Count, Motile Aeromonad Count, Total Coliform Count was observed for D3 (P < 0.05) on 90th day of trial. Two uppermost values were achieved in case of crude protein for D3 and D2 (P > 0.05). The highest lipid content (12.12 ± 0.4 g 100 g?1) was found for D5 (P < 0.05). The highest gross energy (18.75 ± 0.21 MJ 100 g?1) of carcass was recorded for D3. Thus B. licheniformis fb11 at the concentration 5 × 106 CFU g?1 as probiotic supplement promoted growth, digestion in C. chitala juvenile significantly by modulating intestinal microflora.  相似文献

Net photosynthetic rate (PN), transpiration rate (E), water use efficiency (WUE), stomatal conductance (gs), and stomatal limitation (Ls) were investigated in two Syringa species. The saturation irradiance (SI) was 400 µmol m-2s-1 for S. pinnatifolia and 1 700 µmol m-2s-1 for S. oblata. Compared with S. oblata, S. pinnatifolia had extremely low g s . Unlike S. oblata, the maximal photosynthetic rate (Pmax) in S. pinnatifoliaoccurred around 08:00 and then fell down, indicating this species was sensitive to higher temperature and high photosynthetic photon flux density. However, such phenomenon was interrupted by the leaf development rhythms before summer. A relatively lower PN together with a lower leaf area and shoot growth showed the capacity for carbon assimilation was poorer in S. pinnatifolia.  相似文献

Transgenic hairy roots of Datura spp., established using strain A4 of Agrobacterium rhizogenes, are genetically stable and produce high levels of tropane alkaloids. To increase biomass and tropane alkaloid content of this plant tissue, four Pseudomonas strains, Pseudomonas fluorescens P64, P66, C7R12, and Pseudomonas putida PP01 were assayed as biotic elicitors on transgenic hairy roots of Datura stramonium, Datura tatula, and Datura innoxia. Alkaloids were extracted from dried biomass, and hyoscyamine and scopolamine were quantified using liquid chromatography-tandem mass spectrometry analysis. D. stramonium and D. innoxia biomass production was stimulated by all Pseudomonas spp. strains after a 5-d treatment. All strains of P. fluorescens increased hyoscyamine yields compared to untreated cultures after both 5 and 10 d of treatment. Hyoscyamine yields were highest in D. tatula cultures exposed to a 5-d treatment with C7R12 (16.633 + 0.456 mg g?1 dry weight, a 431% increase) although the highest yield increases compared to the control were observed in D. stramonium cultures exposed to strains P64 (511% increase) and C7R12 (583% increase) for 10 d. D. innoxia showed the highest scopolamine yields after elicitation with P. fluorescens strains P64 for 5 d (0.653 + 0.021 mg g?1 dry weight, a 265% increase) and P66 for 5 and 10 d (5 d, 0.754 + 0.0.031 mg g?1 dry weight, a 321% increase; 10 d 0.634 + 0.046 mg g?1 dry weight, a 277% increase). These results show that the Pseudomonas strains studied here can positively and significantly affect biomass and the yields of hyoscyamine and scopolamine from transgenic roots of the three Datura species.  相似文献

Saltol, the major salinity tolerance quantitative trait loci (QTL) in rice, was introgressed from IR66946-3R-230-1-1 (FL530) into Khao Dawk Mali 105 (KDMl105) by two rounds of marker-assisted backcrossing (MAB). Twenty-eight BC2F2 introgression lines (BILs) with positive Saltol allele (BIL+Saltol) and 19 BILs with negative Saltol allele (BIL?Saltol) were validated for the effect of Saltol as key salinity tolerant trait at seedling stage. A hydrophonic system with salt stress of 12 dS m?1 (130 mM Na+) was conducted, and significant differences between BILs+Saltol and BILs?Saltol were observed for the period of plant survival (PPS), total K+ (T-K+) and Na+ (T-Na+) concentration, whole plant Na+-K+ ratio (T-Na+/K+), shoot Na+ (S-Na+) and K+ (S-K+) concentration, and shoot Na+-K+ ratio (S-Na+/K+). BILs+Saltol displayed higher PPS, uptake less Na+ (T-Na+; 43.4 ppm), and more K+ (T-K+; 30.9 ppm), while the BILs?Saltol uptake more Na+ (T-Na+; 45.7 ppm) and less K+ (T-K+; 28.2 ppm). Direct effects on PPS and salt injury score (SIS) were observed, indicating Na+/K+ homeostasis mechanism by the Saltol under hydrophonic salt stress. All BILs+Saltol recovered KDML105 cooking quality profile such as low apparent amylose content (AAC), high score of alkaline spreading value (ASV), intermediate gel consistency (GC), and strong fragrance. However, variation in agronomic traits was observed. The possibility of lowering S-Na+/K+ ratio under salt stress at seedling stage in KDML105 by introgression of the Saltol was demonstrated. Currently, BC2F7 of the BIL+Saltol selected lines are being tested for salinity tolerance in the salt-affected areas in the northeast of Thailand.  相似文献

Salt stress inhibited the growth of both tasg1 and wild-type (WT) wheat seedlings, but the inhibition in tasg1 plants was relatively weaker than that of WT. Compared to the WT, the chlorophyll content, thylakoid membrane polypeptides, Hill reaction activity, actual photochemical efficiency of PSII (ΦPSII), and Mg2+- and Ca2+-ATPase activities were higher in tasg1 under salt stress. At the same time, the photosynthetic activity of the tasg1 was significantly higher than that of WT. In addition, tasg1 plants displayed relatively less accumulation of reactive oxygen species and oxidative damage accompanied by higher activity of some antioxidant enzymes, and the up-regulation of antioxidant genes further demonstrated the improvement of antioxidant activity in tasg1 under salt stress. Furthermore, tasg1 plants also showed relatively weaker Na+ fluorescence and lower Na+ content, but relatively higher content of K+ in their roots and shoots, and then, the roots of tasg1 plants enhanced net outward Na+ flux and a correspondingly increased net inward K+ flux during salt stress. This might be associated with the relatively higher activity of H+-ATPase in tasg1 plants. These results suggest that the improved antioxidant competence and Na+/K+ ion homeostasis play an important role in the enhanced salinity tolerance of tasg1 plants.  相似文献

This study was performed to understand the mechanisms for Se-enhanced resistance of parsley (Petroselinum crispum L.) plants to salinity stress. Plant growth was negatively affected by salt stress; however, Se treatments at 1 mg/L significantly improved the growth rate and enhanced the salt tolerance of seedlings. This increased tolerance in Se-supplied plants was obtained by reduced damaging effect on maximal quantum yield of photosystem II (PSII) (F v/F m) coupled with higher levels of carotenoids and non-photochemical quenching (NPQ). The performance index (PIABS), as evidence for modulation of PSII function, was downregulated by salt stress; while Se mitigated this effect. Moreover, analysis of OJIP transients demon-strated that Se reduced salt damaging effect on PSII function through improvement of excitation energy trapping (TR0/CS) and electron transport (ET0/CS) per excited cross-section of leaf. The Na concentrations in shoots and roots of parsley seedlings considerably enhanced after NaCl treatment. Interestingly, treatment of salt-stressed plants with Se decreased the Na contents in shoots via the limitation of the root-to-shoot translocation of Na and exclusion of Na from cell sap, as well as the retention of K/Na and Ca/Na ratios. These data provide the first evidence that the Se application alleviates salinity stress by enhancing PSII function and by decreasing Na content in the shoot via binding of Na to the root cell wall.  相似文献

Objectives

Lycopene biosynthetic genes from Deinococcus radiodurans were co-expressed in Lactococcus lactis to produce lycopene and improve its tolerance to stress.

Results

Lycopene-related genes from D. radiodurans, DR1395 (crtE), DR0862 (crtB), and DR0861 (crtI), were fused in line with S hine-Dalgarno (SD) sequences and co-expressed in L. lactis. The recombinant strain produced 0.36 mg lycopene g-1 dry cell wt after 48 h fermentation. The survival rate to UV irradiation of the recombinant strain was higher than that of the non-transformed strain.

Conclusion

The L. lactis with co-expressed genes responsible for lycopene biosynthesis from D. radiodurans produced lycopene and exhibited increased resistance to UV stress, suggesting that the recombinant strain has important application potential in food industry.

Arthrospira (Spirulina) is widely used as human health food and animal feed. In cultures grown outdoors in open ponds, Arthrospira cells are subjected to various environmental stresses, such as high temperature. A better understanding of the effects of high temperature on photosynthesis may help optimize the productivity of Arthrospira cultures. In this study, the effects of heat stress on photosynthetic rate, chlorophyll a fluorescence transients, and photosystem (PS) II, PSI activities in a marine cyanobacterium Arthrospira sp. were examined. Arthrospira cells grown at 25 °C were treated for 30 min at 25 (control), 30, 34, 37, or 40 °C in the dark. Heat stress (30–37 °C) enhanced net photosynthetic O2 evolution rate. Heat stress caused over-reduction PSII acceptor side, damage of donor side of PSII, decrease in the energetic connectivity of PSII units, and decrease in the performance of PSII. When the temperature changed from 25 to 37 °C, PSII activity decreased, while PSI activity increased, the enhancement of photosynthetic O2 evolution was synchronized with the increase in PSI activity. When temperature was further increased to 40 °C, it induced a decrease in photosynthetic O2 evolution rate and a more severe decrease in PSII activity, but an increase in PSI activity. These results suggest that PSI activity was the decisive factor determining the change of photosynthetic O2 evolution when Arthrospira was exposed to a temperature from 25 to 37 °C, but then, PSII activity became the decisive factor adjusting the change of photosynthetic O2 evolution when the temperature was increased to 40 °C.  相似文献

The photosynthetic responses to salt stress were examined in a wheat (Triticum aestivum L. cv. Asakaze)–barley (Hordeum vulgare L. cv. Manas) 7H addition line having elevated salt tolerance and compared to the parental wheat genotype. For this purpose, increasing NaCl concentrations up to 300 mM were applied and followed by a 7-day recovery period. Up to moderate salt stress (200 mM NaCl), forcible stomatal closure, parallel with a reduction in the net assimilation rate (P N), was only observed in wheat, but not in the 7H addition line or barley. Since the photosynthetic electron transport processes of wheat were not affected by NaCl, the impairment in P N could largely be accounted for the salt-induced decline in stomatal conductance (g s), accompanied by depressed intercellular CO2 concentration and carboxylation efficiency. Both, P N and nonstomatal limitation factors (Lns) were practically unaffected by moderate salt stress in barley and in the 7H addition line due to the sustained g s, which might be an efficient strategy to maintain the efficient photosynthetic activity and biomass production. At 300 mM NaCl, both P N and g s decreased significantly in all the genotypes, but the changes in P N and Lns in the 7H addition line were more favourable similar to those in wheat. The downregulation of photosynthetic electron transport processes around PSII, accompanied by increases in the quantum yield of regulated energy dissipation and of the donor side limitation of PSI without damage to PSII, was observed in the addition line and barley during severe stress. Incomplete recovery of P N was observed in the 7H addition line as a result of declined PSII activity probably caused by enhanced cyclic electron flow around PSI. These results suggest that the better photosynthetic tolerance to moderate salt stress of barley can be manifested in the 7H addition line which may be a suitable candidate for improving salt tolerance of wheat.  相似文献

Photosynthetic parameters including net photosynthetic rate (PN), transpiration rate (E), water-use efficiency (WUE), and stomatal conductance (gs) were studied in indoor C3 plants Philodendron domesticum (Pd), Dracaena fragans (Df), Peperomia obtussifolia (Po), Chlorophytum comosum (Cc), and in a CAM plant, Sansevieria trifasciata (St), exposed to various low temperatures (0, 5, 10, 15, 20, and 25°C). All studied plants survived up to 0°C, but only St and Cc endured, while other plants wilted, when the temperature increased back to room temperature (25°C). The PN declined rapidly with the decrease of temperature in all studied plants. St showed the maximum PN of 11.9 μmol m?2 s?1 at 25°C followed by Cc, Po, Pd, and Df. E also followed a trend almost similar to that of PN. St showed minimum E (0.1 mmol m?2 s?1) as compared to other studied C3 plants at 25°C. The E decreased up to ≈4-fold at 5 and 0°C. Furthermore, a considerable decline in WUE was observed under cold stress in all C3 plants, while St showed maximum WUE. Similarly, the gs also declined gradually with the decrease in the temperature in all plants. Among C3 plants, Pd and Po showed the maximum gs of 0.07 mol m?2 s?1 at 25°C followed by Df and Cc. However, St showed the minimum gs that further decreased up to ~4-fold at 0°C. In addition, the content of photosynthetic pigments [chlorophyll a, b, (a+b), and carotenoids] was varying in all studied plants at 0°C. Our findings clearly indicated the best photosynthetic potential of St compared to other studied plants. This species might be recommended for improving air quality in high-altitude closed environments.  相似文献

Soil salinity reduces growth of Glycyrrhiza uralensis in arid and semi-arid areas of north-west in China. Silicon (Si) nutrition may alleviate salt stress in many crops including grain crop, fruit crop, and vegetable crop. In this study, the alleviating effects of Si on growth characteristics, antioxidant enzyme activity (SOD and POD) and MDA concentration, and K+ and Na+ concentrations in G. uralensis seedlings subjected to 50 mM NaCl stress were investigated. The results showed that NaCl stress imposed significant reduction in root length, secondary root number, leaf number, and stem and total dry weight of G. uralensis. NaCl stress also significantly reduced the activities of SOD and POD, and ration of K+/Na+, but significantly increased MDA concentration in leaves of G. uralensis seedling. The addition of Si increased SOD and POD activities, and reduced MDA concentration, which resulting in greater reactive oxygen species detoxification and lower lipid peroxidation. Si also significantly increased the ratio of K+/Na+ in stem and leaves of G. uralensis. In conclusion, Si could alleviate adverse effects of salt stress probably by decreasing Na+ concentration and improving antioxidant enzyme activity of G. uralensis, and these alleviating effects were dependent on Si concentration and on Si processing time.  相似文献
