Genetic structure of juvenile fish from the populations of the Atlantic salmon Salmo salar inhabiting the rivers of Murmansk oblast, Arkhangelsk oblast, and Karelia, as well as of juveniles from hatcheries was examined at five allozymic loci: aspartate aminotransferase (AAT-4*), isocitrate dehydrogenase (IDHP-3*), iditol dehydrogenase (IDDH-2*), esterase D (ESTD*), and malic enzyme (MEP-2*). High genetic differentiation of both natural and “hatchery” juvenile fish was revealed. It was demonstrated that the gene pool of juveniles at three of the five loci tested was to a considerable degree formed by natural selection. In this case, the role of limiting factor was played by the duration of critical temperature regime in the rivers. The differentiation factors for juveniles from nature were not established, however, their clusterization pattern pointed to a possible role of natural selection in this process.  相似文献

Four natural Greek populations of Mediterranean fruit fly, Ceratitis capitata (Wiedemann), was studied for genetic variability at 25 enzyme loci. The comparison of polymorphism within and between populations shows two loci with high between-population heterozygosity (HT) and very high fixation index (F(ST)) values, suggesting the presence of balancing selection. The gradual decline of common allele frequency of the polymorphic loci tested indicated that latitudinal clines are present in Greece. Indirect estimates of gene flow based both on Wright's method (Nm*) and on the Slatkin's method (Nm*), which depends on the frequencies of rare alleles found in only one population, revealed a substantial amount of gene flow (Nm = 3.493 and Nm* = 3.197). These estimates of gene flow may well explain why the "introduced" Greek populations of C. capitata, in spite of their low genetic variability, display the same polymorphic loci. Gene flow in combination with natural selection and genetic drift may have played an important role to genetic differentiation in this species in Greece.  相似文献

Potential factors were analyzed that affected the formation of gene pools of two introduced beaver populations founded in the 1940s-1950s by releasing beavers from Voronezh oblast and Belarus into rivers of the Kirov and Novosibirsk oblasts. The populations from these two regions were shown to differ in four allozyme loci (Ahd-2, Gpi, Es-5, and Dia-2). Within Kirov oblast, the samples from three tributaries of the Vyatka River differed in three other loci (Ck-1, Gp-9, and Trf). It is suggested that these features of the allele distribution at these loci are explained by the history of the introduced populations, hybridization between beavers from Voronezh oblast and Belarus, and founder effect during the subsequent artificial and spontaneous migration. Generally, genetic differences between the populations in all studied loci were very small: D = 0.02 between populations from different regions and D = 0.01 between populations from the tributaries of third-order rivers of the Kirov oblast. At the same time, disequilibrium was detected for ten polymorphic loci in the combined sample from the populations of Kirov oblast, which indicates the existence of interpopulation heterogeneity at the level of local populations of fourth- and fifth-order rivers. The unexpected finding of a genetic similarity between one studied individual of Tuvinian beaver subspecies (Castor fiber tuvinicus) and C. f. orientoeuropeas was recorded.  相似文献

The 1985 introduction into the European North of Russia resulted in the formation of a large stock of pink salmon of the odd-year breeding line. To assess the divergence of the new population and the role of various microevolutionary factors, variation of four microsatellite loci and fifteen genes encoding proteins (allozymes) in samples of fish, running for spawning in rivers of the new area, and in samples from the donor population of the Ola River (Magadan oblast). In the generations 8 and 9 of the introduced pink salmon of the odd-year line, the genetic diversity (the number of alleles and the mean heterozygosity) both at allozyme and at microsatellite loci was significantly lower, than that in the donor population. The explanations of the decline in diversity are discussed. The first evidence for spatial genetic divergence in transplanted fish within the new area has been obtained; the divergence level may be comparable with that characteristic of native populations.  相似文献

Variations at three flight-related enzyme loci, -glycerophosphate dehydrogenase (-Gpdh), glucose-6-phosphate dehydrogenase (G6pd), and phosphoglucomutase (Pgm), ofEpiphyas postvittana (Walker) moths were investigated using starch gel electrophoresis. Among the three enzyme loci, -Gpdh andG6pd were found to be monomorphic, butPgm was polymorphic, with a total of seven different genotypes and five alleles identified in this study. Comparisons of allozyme variability at thePgm locus showed significant differentiation among five natural populations sampled from geographically distinct localities in New Zealand and Australia and between laboratory populations differentiated by artificial selection on flight capacity. ThePgm polymorphism was shown to be associated with the variation of flight capacity, but the role of the enzyme locus in the evolution of flight behavior is to be demonstrated in this species.This work was supported by the research scholarship of the University of New England.  相似文献

The variability of 21 allozyme and three microsatellite loci of chloroplast DNA (cpDNA) was studied in the populations of Siberian spruce (Picea obovata Ledeb.) from Irkutsk oblast, Magadan oblast, Buryatia, and Mongolia. It was demonstrated that the highest level of genetic diversity among the examined populations at both allozyme and microsatellite loci was observed in the Tulyushka population from Irkutsk oblast. The lowest level of genetic diversity was observed in marginal isolated populations of Bogd Uul and Magadan. In the relict spruce population from Olkhon Island, differing from the other populations in the lowest allelic diversity of both types of markers, no expected decline of expected heterozygosity and haplotype diversity was observed. In this population, the variability parameters mentioned were close to the population mean. The obtained intrapopulation and intraspecific variability parameters of allozyme and microsatellite loci of chloroplast DNA and the data on the population differentiation at these loci indicate that the given markers can be used for the analysis of the population structure of Siberian spruce.  相似文献

We determined the amount and temporal stability of genetic differentiation among brook cham sampled from five rivers on Cape Race, Newfoundland, with an electrophoretic analysis of 42 protein coding loci. Fish from four of these rivers were analysed for restriction fragment length polymorphisms in mitochondrial DNA (mtDNA). A single mtDNA clone was observed in all rivers sampled, except one, where 47% offish were from a different and relatively divergent clone (0.31 % sequence divergence). In contrast, Cape Race brook charr show large amounts of genetic differentiation at six enzyme coding loci; Nei's genetic distance ranged between 0,020 and 0.048. This differentiation is relatively stable as no significant differences in allele frequencies were detected between fish sampled from two rivers over two consecutive years. The most divergent population based on protein polymorphism is not that with two mtDNA clonal lineages. In contrast to the commonly held view, mtDNA analyses do not necessarily provide greater resolution of population structure than allozyme analyses.  相似文献

本文应用随机扩增多态性DNA (RAPD) 技术, 采用多态位点率、Shannon 信息多样性指数和Nei 遗传多样性指数作为种群遗传变异的指标, 研究了1998 和1999 年大仓鼠种群性别群体和年龄组群体的遗传多样性。主要的结论是: 性别群体间遗传分化较小, 年龄组群体间分化较大; 年龄组相距越大, 遗传分化也就越大; 幼年组的遗传多样性高于老龄组的遗传多样性; 1999 年秋季种群的年龄组遗传分化和差异要比1998 年四季种群更为明显。大仓鼠不同年龄组群体间的遗传分化支持遗传结构由于受时空上的选择压力而产生变异和适应性观点。大仓鼠幼体的遗传多样性比成体和老体的较高, 这一结果倾向于支持Ford 的假说, 说明种群繁殖出的幼体遗传多样性高, 其中有遗传质量好的, 也有遗传质量差的,但在生存过程中, 随着选择压力的作用, 种群趋于适应, 与环境不相适应的类型遭到淘汰,低质的幼体由于自然选择压力的增加而从种群中消失, 致使成年和老年群体多态性逐渐降低。  相似文献

Although kin associations between fish can develop in the laboratory, the importance of kin structure in natural social groups remains uncertain. Guppies ( Poecilia reticulata ) possess many behavioural and life-history traits that promote kin structure, and so constitute ideal candidates for testing its general significance amongst fish. We collected 25 adult only shoals from ∼150 m stretches along two Trinidadian rivers. The 272 sampled fish were genotyped from a pool of seven hypervariable microsatellite loci. Although mark-recapture experiments have previously detected male-biased dispersal, which is predicted from the guppy mating system and which may have important implications for population differentiation, we found no sex-biases in dispersal between shoals. Because sex-biased dispersal was not evident, males and females were pooled in analyses of kin structure. Relatedness levels within each shoal did not differ significantly from zero, indicating that kin selection does not contribute to gross structure. Instead, our data is more consistent with the hypothesis that individual-level selection underlies shoal composition dynamics. These dynamics have important implications for the evolution of cooperative behaviour in guppies.  相似文献

To manage populations of threatened fish species in modified habitats and regulated rivers requires an understanding of their reproductive biology and spawning cues. In particular, accurate information about early life stages in these species can be used to facilitate programmes that maximise recruitment and breeding success. This study aimed to develop methods to accurately age early juvenile freshwater native fish, Two‐spined Blackfish, (Gadopsis bispinosus), to allow the determination of spawning date. This was accomplished through the examination of otolith microstructure in early juveniles. The age at which the first ring was deposited and the relationship between days and number of rings were determined using both field and aquarium trials. Field trials of marked juvenile otoliths revealed daily deposition of rings (1.02 ± 0.02 rings per day for fish sacrificed six days postmarking). The strength of this relationship lessened slightly as juveniles aged (0.92 ± 0.02 rings per day for fish sacrificed 13 days postmarking). The first otolith ring was deposited 7.50 ± 1.09 days after spawning. The enumeration of daily rings combined with knowledge of the commencement of ring deposition enabled accurate estimation of spawning date. The current study is the first to examine otolith microstructure in juvenile Two‐spined Blackfish allowing accurate determination of spawning date. While more research is required to accurately age older juveniles, this technique has the potential to precisely correlate spawning with environmental cues, facilitating better management of temperature and flow during breeding periods, potentially increasing spawning and recruitment of this endangered species.  相似文献

Population differentiation can be driven in large part by natural selection, but selectively neutral evolution can play a prominent role in shaping patters of population divergence. The decomposition of the evolutionary history of populations into the relative effects of natural selection and selectively neutral evolution enables an understanding of the causes of population divergence and adaptation. In this study, we examined heterogeneous genomic divergence between alpine and lowland ecotypes of the allopolyploid plant, Anemone multifida. Using peak height and dominant AFLP data, we quantified population differentiation at non-outlier (neutral) and outlier loci to determine the potential contribution of natural selection and selectively neutral evolution to population divergence. We found 13 candidate loci, corresponding to 2.7% of loci, with signatures of divergent natural selection between alpine and lowland populations and between alpine populations (Fst  = 0.074–0.445 at outlier loci), but neutral population differentiation was also evident between alpine populations (FST  = 0.041–0.095 at neutral loci). By examining population structure at both neutral and outlier loci, we determined that the combined effects of selection and neutral evolution are associated with the divergence of alpine populations, which may be linked to extreme abiotic conditions and isolation between alpine sites. The presence of outlier levels of genetic variation in structured populations underscores the importance of separately analyzing neutral and outlier loci to infer the relative role of divergent natural selection and neutral evolution in population divergence.  相似文献

Habitat selection is an important phenomenon that may greatly affect individual fitness. Using an artificial stream, we examined the relationship between the percentage of prey captured, reactive distance, dominance, and water velocity for juvenile Chinook Salmon (Oncorhynchus tshawytscha) from the Chena River, Alaska, and tested the fitness-based microhabitat selection model of Grossman et al. (Ecol Freshw Fish 11:2–10, 2002). Recent declines in the abundance of Chinook accentuate our need for habitat selection studies on this species. We conducted three experiments: two with single fish (1st N?=?27, fish SL 58–84 mm, 2nd N?=?14, fish SL 49–56 mm) and one with pairs of dominant and subordinate fish (N?=?10 pairs, 64–96 mm, mean difference in SL?=?7 mm). We placed individual or pairs of fish in an artificial flume and recorded reactive distance and the percent prey capture with individual dead brine shrimp (Artemia spp.) as prey. Prey were presented at 10 cm/s velocity intervals ranging from 10 to 60 cm/s; velocities found in the natural habitat. Mean reactive distance in single fish experiments (henceforth SFE) averaged 33 and 29 cm respectively, and was not related to velocity. We detected a negative, curvilinear relationship between velocity and percent prey capture. Holding velocities for juvenile Chinook were significantly lower than prey capture velocities. The Grossman et al. (Ecol Freshw Fish 11:2–10, 2002) model yielded an optimal focal-point velocity prediction of 35 cm/s for juvenile Chinook, however focal-point velocities occupied by juveniles in the Chena River averaged 12 cm/s. Predicted optimal velocities were present in the Chena River; hence, this discrepancy suggests that other factors such as distraction from drifting debris or predation risk influenced habitat selection. There were no differences in reactive distances or holding velocity/capture velocity relationships for dominant and subordinate fish; however, dominants captured significantly more prey than subordinates. Being subordinate resulted in a decrease of 61% in mean percent prey capture (the difference between what was captured by the fish alone versus the difference with a dominant), whereas the mean cost to fish with dominant rank was a 21% decline between the percentage captured alone versus that with a subordinate.  相似文献

Compared to wild populations, cultured Atlantic salmon often sustain higher mortality rates and lower adult return rates when stocked as juveniles into natural streams. The ultimate causes for such differences in fitness, however, are not clear. Here we tested if relaxed levels of natural selection and improved survival in the hatchery environment could account for the observed degree of maladaptation among stocked fish. To do this, we assessed the degree of phenotypic mismatch between wild and cultured fish in three populations over five consecutive years. Significant differences were found in several phenotypic traits that are likely to have fitness implications. Thus, if the objective is to mimic wild individuals for restoration purposes, current hatchery practices aimed at maximising juvenile survival and enhancing growth may need to be revised.  相似文献

Sympatric populations of D. tryoni and D. neohumeralis are difficult to completely distinguish taxonomically. Using five pigmentation characters, each of some taxonomic value, a small proportion of individuals cannot be assigned to either species nor definitely classified as hybrids. To aid in species discrimination and hybrid identification gene frequencies in natural populations were estimated at three polymorphic protein loci, an alcohol dehydrogenase (Adh), an octanol dehydrogenase (Odh) and an esterase (E-2). Samples of flies were taken from four sites spread over 1200 miles along the Australian eastern coast. Within each species allelic frequencies at each locus were largely the same at all localities. Consistent differences in gene frequencies between species occurred at all three loci, strongly supporting the hypothesis of two distinct gene pools. The Adh locus best discriminated between species with a unique allele occurring in D. neohumeralis at a frequency of 0-85. None of the loci showed complete differentiation and hence it was not possible to find a quick and easy method to distinguish the species nor to detect field hybrids. Directional selection of laboratory populations for a change in callus colour (the best pigmentation character for separating the species) indicated that at the Adh and E-2 loci frequencies of major alleles were not genetically associated with major genes for callus colour. Thus genotype determination at these loci when considered together with pigmentation characters may be valuable taxonomically for further distinguishing between the species.  相似文献

The presence of bluestreak cleaner wrasse, Labroides dimidiatus, on coral reefs increases total abundance and biodiversity of reef fishes. The mechanism(s) that cause such shifts in population structure are unclear, but it is possible that young fish preferentially settle into microhabitats where cleaner wrasse are present. As a first step to investigate this possibility, we conducted aquarium experiments to examine whether settlement-stage and young juveniles of ambon damselfish, Pomacentrus amboinensis, selected a microhabitat near a cleaner wrasse (adult or juvenile). Both settlement-stage (0 d post-settlement) and juvenile (~5 weeks post-settlement) fish spent a greater proportion of time in a microhabitat adjacent to L. dimidiatus than in one next to a control fish (a non-cleaner wrasse, Halichoeres melanurus) or one where no fish was present. This suggests that cleaner wrasse may serve as a positive cue during microhabitat selection. We also conducted focal observations of cleaner wrasse and counts of nearby damselfishes (1 m radius) to examine whether newly settled fish obtained direct benefits, in the form of cleaning services, from being near a cleaner wrasse. Although abundant, newly settled recruits (<20 mm total length) were rarely (2 %) observed being cleaned in 20 min observations compared with larger damselfishes (58 %). Individual damselfish that were cleaned were significantly larger than the median size of the surrounding nearby non-cleaned conspecifics; this was consistent across four species. The selection by settlement-stage fish of a microhabitat adjacent to cleaner wrasse in the laboratory, despite only being rarely cleaned in the natural environment, suggests that even rare cleaning events and/or indirect benefits may drive their settlement choices. This behaviour may also explain the decreased abundance of young fishes on reefs from which cleaner wrasse had been experimentally removed. This study reinforces the potentially important role of mutualism during the processes of settlement and recruitment of young reef fishes.  相似文献

Horabagrus brachysoma (sun‐catfish, Bagridae, Siluriformes) is a valuable ornamental and food fish. The stock structure of H. brachysoma, necessary to conserve its declining natural populations, is not known. Twenty‐five primers developed for four fish species belonging to the orders Siluriform (3) and Osteoglossiform (1) were tested and eight primers amplified microsatellite loci in H. brachysoma. The results demonstrate that cross‐priming between fish species belonging to different families and even to different orders can yield microsatellite loci. Five of eight primers each amplified two loci. However, the loci that had repeat motifs after sequencing were considered only for genotyping. Finally, eight loci were polymorphic with hree to seven alleles. Individual fish genotype data (n = 42; 21 each in two rivers) at each locus was analysed. Significant genetic heterogeneity was detected at six loci. The identified loci exhibited potential for use in population genetics application in H. brachysoma.  相似文献

Anthropogenic alterations to river flow regimes threaten freshwater biodiversity globally, with potentially disproportionate impacts on species that rely on flow cues to trigger critical life history processes, such as migration for diadromous fishes. This study investigates the influence of river discharge on the abundance of juvenile fish moving into rivers by four temperate catadromous or amphidromous species (common galaxias Galaxias maculatus, spotted galaxias Galaxias truttaceus, climbing galaxias Galaxias brevipinnis and the threatened Australian grayling Prototroctes maraena). Fyke netting or fishway trapping was used to catch juvenile fish moving from estuaries into freshwater in five coastal waterways in south-eastern Australia during the spring migratory period. There was a positive relationship between the probability of high catch rates and mean discharge in September. We also found a positive relationship between discharge and the number of recruits captured 22–30 days later in a flow stressed system. In addition, day-of-year had a strong influence on catch rates, with the peak abundance of juveniles for three species most likely to occur midway through the sampling period (spotted galaxias in October, climbing galaxias in late October and Australian grayling in late October and early November). Our study shows that higher magnitudes of river discharge were associated with increased catches of juvenile catadromous and amphidromous fishes. With a limited supply of environmental water, environmental flows used to enhance immigration of these fishes may be best targeted to maintain small amounts of immigration into freshwater populations in waterways or years when discharges are low and stable. When there are natural, large discharge volumes, relatively large numbers of juvenile fish can be expected to enter coastal waterways and during these times environmental flows may not be required to promote immigration.  相似文献

A large-scale juvenile Japanese flounder (Paralichthys olivaceus) release-recapture experiment was undertaken to find the optimal release season by evaluating the nutritional status of released fish at different seasons during which food abundance was significantly different. Forty thousand fish were released at depths of 1.5 m for early-release (May 29, 1997) and 2 m for late-release (July 2, 1997) (42.1±3.5 and 40.9±4.2 mm body length, respectively) in an experimental field, Wakasa Bay, the Sea of Japan. Samples were taken, after the releases, at Wada beach at intervals of 1, 2, 3, 6, 10, 16 and 30 days after release (DAR), including pre-surveys before each release. Released fish recaptured from the two different release groups totaled 764; 467 from the early-release group (ER) and 297 from the late-release group (LR). A total of 1956 wild flounder juveniles were simultaneously collected (1041 ER, 915 LR). ER fish were subject to higher food availability and were exposed to less pressure from predation by smaller wild juvenile flounder. RNA/DNA ratios in ER juveniles were significantly higher than those of LR fish during all samples. Especially, RNA/DNA ratios in ER juveniles were higher than in wild juveniles from 3 to 50 DAR. In the LR group, the nutritional status of juveniles was relatively low in shallower water. These findings corresponded well with feeding incidence examined by coworkers. Mass release of hatchery-reared juveniles apparently reduced RNA/DNA ratio of the wild juveniles right after releasing. The present study showed that earlier release of hatchery-reared juvenile Japanese flounder with higher RNA/DNA ratio could increase the possibilities of survival right after release in the nursery ground, and that RNA/DNA ratio appeared to be a good tool in evaluating nutritional status of released juveniles as well as wild juveniles in Japanese flounder.  相似文献
