Using RAPD marker analysis, two quantitative trait loci (QTLs) associated with earliness due to reduced fruit-ripening time
(days from anthesis to ripening = DTR) were identified and mapped in an F2 population derived from a cross between Lycopersicon esculentum’E6203’ (normal ripening) and Lycopersicon esculentum’Early Cherry’ (early ripening). One QTL, on chromosome 5, was associated with a reduction in both ripening time (5 days)
and fruit weight (29.3%) and explained 15.8 and 13% of the total phenotypic variation for DTR and fruit weight, respectively.
The other QTL, on chromosome 12, was primarily associated with a reduction only in ripening time (7 days) and explained 12.3%
of the total phenotypic variation for DTR. The gene action at this QTL was found to be partially dominant (d/a=0.41). Together,
these two QTLs explained 25.1% of the total phenotypic variation for DTR. Additionally, two QTLs associated with fruit weight
were identified in the same F2 population and mapped to chromosomes 4 and 6, respectively. Together, these two QTLs explained 30.9% of the total phenotypc
variation for fruit weight. For all QTLs, the ’Early Cherry’ alleles caused reductions in both ripening time and fruit weight.
The polymorphic band for the most significant RAPD marker (OPAB-06), linked to the reduced ripening time QTL on chromosome
12, was converted to a cleaved amplified polymorphism (CAP) assay for marker-aided selection and further introgression of
early ripening time (DTR) into cultivated tomato.

Black raspberry (Rubus occidentalis) is an important specialty fruit crop in the US Pacific Northwest that can hybridize with the globally commercialized red raspberry (R. idaeus). Here we report a 243 Mb draft genome of black raspberry that will serve as a useful reference for the Rosaceae and Rubus fruit crops (raspberry, blackberry, and their hybrids). The black raspberry genome is largely collinear to the diploid woodland strawberry (Fragaria vesca) with a conserved karyotype and few notable structural rearrangements. Centromeric satellite repeats are widely dispersed across the black raspberry genome, in contrast to the tight association with the centromere observed in most plants. Among the 28 005 predicted protein‐coding genes, we identified 290 very recent small‐scale gene duplicates enriched for sugar metabolism, fruit development, and anthocyanin related genes which may be related to key agronomic traits during black raspberry domestication. This contrasts patterns of recent duplications in the wild woodland strawberry F. vesca, which show no patterns of enrichment, suggesting gene duplications contributed to domestication traits. Expression profiles from a fruit ripening series and roots exposed to Verticillium dahliae shed insight into fruit development and disease response, respectively. The resources presented here will expedite the development of improved black and red raspberry, blackberry and other Rubus cultivars. 相似文献

Previous attempts to extract high-quality, total RNA from raspberry (Rubus idaeus) fruits using published protocols have proven to be unsuccessful. Even the use of protocols developed for the extraction
of RNA from other fruit tissue has resulted in low yields (1) or the isolation of degraded RNA (2). Here, we report on the development of a quick and simple method of extracting total RNA from raspberry fruit. Using this
method, high yields of good quality, undegraded RNA were obtained from fruit at all stages of ripening. The RNA is of sufficient

Protein content (PC) and protein index (PI) play important roles in determining nutritional quality in rice (Oryza sativa L.). We used 71 lines derived from “Asominori/IR24” to analyze the developmental behavior of PC and PI through unconditional
and conditional QTL mapping methods. In all, 10 unconditional QTLs and 6 conditional QTLs for PC, and 11 unconditional QTLs
and 9 conditional QTLs for PI, were identified at four stages of grain filling. More were identified in the first three stages
than at the final stage. Temporal patterns of gene expression for PC and PI differed over time, with several QTLs being expressed
across two or three stages but many being expressed at only one stage. Some of these QTLs were closely linked with maturity
QTLs reported previously. Many QTLs for PC and PI were co-localized, supporting the significant correlation found between
PC and PI. Our results suggest that dynamic QTL mapping might be a valid means for revealing more genetic information about

Female Osyris quadripartita plants exhibit uninterrupted reproductive activity throughout the year, due to the long duration of successive stages in the cycle and marked within-crop developmental asynchrony. Cycles corresponding to the flowering seasons of consecutive years overlap in each individual. Flowering takes place in spring, and fruits develop in the dry summer season and ripen at any time of the year. Variation in flowering time explains a negligible proportion of variation in ripening time. The greatest reproductive losses are incurred in the phase extending from closed flowers through unripe fruits, mostly due to ovary abortion. Only 30% of closed flowers eventually reach this latter stage. In contrast, 75% of unripe fruits complete their development, with subsequent dispersal of seeds. The probability of the setting of ripe fruit steadily decreases from early to late season flowers, due to increased ovary abortion rates. Resource limitation in the dry summer season seems responsible for this pattern of selective fruit maturation. 相似文献

Prolonged low temperature phases and short-term cold spells often occur in spring during the crucial stages of early maize
(Zea mays L.) development. The effect of low temperature-induced growth retardation at the seedling stage on final yield is poorly
studied. Therefore, the aim was to identify genomic regions associated with morpho-physiological traits at flowering and harvest
stage and their relationship to previously identified quantitative trait loci (QTLs) for photosynthesis and morpho-physiological
traits from the same plants at seedling stage. Flowering time, plant height and shoot biomass components at harvest were measured
in a dent mapping population for cold tolerance studies, which was sown in the Swiss Midlands in early and late spring in
two consecutive years. Early-sown plants exhibited chilling stress during seedling stage, whereas late-sown plants grew under
favorable conditions. Significant QTLs, which were stable across environments, were found for plant height and for the time
of flowering. The QTLs for flowering were frequently co-localized with QTLs for plant height or ear dry weight. The comparison
with QTLs detected at seedling stage revealed only few common QTLs. A pleiotropic effect was found on chromosome 3 which revealed
that a good photosynthetic performance of the seedling under warm conditions had a beneficial effect on plant height and partially
on biomass at harvest. However, a high chilling tolerance of the seedling seemingly had an insignificant or small negative

The production of ethylene by red raspberry (Rubus idaeus L.cv. Glen Clova) fruit increased climacterically during development.The concentration of ethylene within green fruit was low butincreased substantially as fruit abscission and ripening commenced.The receptacle contained higher concentrations than the drupeletsat all stages measured. In the mature ripening fruit the ethyleneconcentrations were found to be physiologically significant,and would accelerate the abscission of large green non-abscisingfruit if supplied as a fumigant. The addition of ethylene toripe fruit did not accelerate abscission, probably because saturatinglevels occurred naturally within these fruit. Reduction of ethylenesynthesis rates using the inhibitor of ethylene production aminoethoxyvinylglycine(AVG) reduced the rate of abscission zone weakening which occursin detached large green fruit. The rate of ethylene productionwas found to be dependent on the supply of the precursor l-aminocyclopropane-l-carboxylicacid (ACC). Seedless avocado fruit are produced alongside seeded fruit in the cultivar Arad, and both reach maturity at the same time. Using this system, it was possible to show that avocado seed inhibits the ripening process: seedless fruits exhibited higher response to exogenous ethylene already at the fruitlet stage, and also at the immature and mature fruit stages. They produced higher CO2 levels, and the ethylene peak was apparent at the fruitlet stage of seedless fruit, but not of seeded ones. The expression levels of PaETR, PaERS1 and PaCTR1 on the day of harvest at all developmental stages were very similar between seeded and seedless fruit, except that PaCTR1 was higher in seedless fruit only at very early stages. This expression pattern suggests that the seed does not have an effect on components of the ethylene response pathway when fruits are just picked. The expression of MADS-box genes, PaAG1 and PaAGL9, preceded the increase in ethylene production of mature seeded fruit, but not at earlier stages. However, only PaAGL9 was induced in seedless fruit at early stages of development. Taken together, these data suggest that these genes are perhaps involved in climacteric response in seeded fruit, and the seed is responsible for their induction at normal fruit ripening. 相似文献

The changes of the carotenoid pigments in the peel of ripening persimmon (Diospyros kaki) cv Triumph were followed for an entire season. During ripening, the total carotenoid decreased until colour break, then increased gradually and drastically at the last ripening stages. The chloroplast carotenoid pattern of the unripe fruit changed into a chromoplast pattern in which cryptoxanthin was the predominant pigment, reaching a level between 40 and 50% of the total carotenoids. It accumulated continuously at a rate of approximately 10% at each 2 week interval, its percentage being characteristic for each ripening stage. Other major pigments at levels of approximately 10% of the total carotenoids were zeaxanthin, antheraxanthin and violaxanthin. In the fully ripe fruit, ripened both on and off the tree, lycopene which was not present before was found as the second major pigment. This unusual pattern change is discussed. 相似文献

cDNA cloning and expression pattern of cinnamate-4-hydroxylase in the Korean black raspberry 总被引:1,自引:0,他引:1
Cinnamate-4-hydroxylase (C4H) is a key enzyme in the phenylpropanoid pathway, which is responsible for synthesizing a variety of secondary metabolites that participate in development and adaptation. In this study, we isolated a full-length cDNA of the C4H gene from the Korean black raspberry (Rubus sp.) and found that this gene existed as a single gene. By comparing the deduced amino acid sequence of Rubus sp. C4H with other sequences reported previously we determined that this sequence was highly conserved among widely divergent plant species. In addition, quantitative real time PCR studies indicated that the C4H gene had a differential expression pattern during fruit development, where gene expression was first detected in green fruit and was then remarkably reduced in yellow fruit, followed by an increase in red and black fruit. To investigate the two peaks in expression observed during fruit development and ripening, we measured the flavonoid content. The content of the major flavanol of Korean black raspberry fruits was determined to be highest at the beginning of fruit development, followed by a gradually decrease according to the developmental stages. In contrast, the content of anthocyanins during the progress of ripening was dramatically increased. Our results suggest that the C4H gene in Korean black raspberry plays a role during color development at the late stages of fruit ripening, whereas the expression of C4H gene during the early stages may be related to the accumulation of flavanols. 相似文献

This study focuses on four raspberry ( Rubus idaeus ) genotypes from two different genetic backgrounds: cvs Glen Prosen and Glen Clova, bred at the Scottish Crop Research Institute (SCRI) and genotypes bred at Horticulture Research International (HRI), East Malling (EM), EM 4997 and EM 5007. The ripe fruit of each genotype pair were characterised subjectively by raspberry breeders as relatively firm or soft, respectively. Different stages of fruit development from each genotype were used to quantify fruit firmness, rates of ethylene evolution and ripening rate. Penetrometry data confirmed suspected firmness differences. Firmness correlated with rates of ethylene evolution. Rates of ethylene production also correlated with receptacle size. Storage of green fruits in 20 μl l−1 ethylene reduced fruit firmness, enhanced respiration rate and colour (anthocyanin) development and stimulated the development of cell wall hydrolase activities. However, during natural ripening in the field, fruit respiration rate declined, which indicates a non-climacteric ripening pattern. In drupelets, the activities of polygalacturonase (PG), pectin methylesterase (PME), C x -cellulase (C x ) and β -galactosidase ( β -gal.) increased substantially as ripening progressed. More detailed studies with ripe fruit of cv. Glen Clova indicated major isoforms of PG at pIs 3.3, 8.6 and 10.1; of PME at pIs 7.2, 8.5, 8.7, 8.8; of C x at pI 2.4; and of β -gal. at pIs 6.3 and 6.7. 相似文献

For fruit flies, fully ripe fruit is preferred for adult oviposition and is superior for offspring performance over unripe
or ripening fruit. Because not all parts of a single fruit ripen simultaneously, the opportunity exists for adult fruit flies
to selectively choose riper parts of a fruit for oviposition and such selection, if it occurs, could positively influence
offspring performance. Such fine scale host variation is rarely considered in fruit fly ecology, however, especially for polyphagous
species which are, by definition, considered to be generalist host users. Here we study the adult oviposition preference/larval
performance relationship of the Oriental fruit fly, Bactrocera dorsalis (Hendel) (Diptera: Tephritidae), a highly polyphagous pest species, at the “within-fruit” level to see if such a host use
pattern occurs. We recorded the number of oviposition attempts that female flies made into three fruit portions (top, middle
and bottom), and larval behavior and development within different fruit portions for ripening (color change) and fully-ripe
mango, Mangifera indica L. (Anacardiaceae). Results indicate that female B. dorsalis do not oviposit uniformly across a mango fruit, but lay most often in the top (i.e., stalk end) of fruit and least in the
bottom portion, regardless of ripening stage. There was no evidence of larval feeding site preference or performance (development
time, pupal weight, percent pupation) being influenced by fruit portion, within or across the fruit ripening stages. There
was, however, a very significant effect on adult emergence rate from pupae, with adult emergence rate from pupae from the
bottom of ripening mango being approximately only 50% of the adult emergence rate from the top of ripening fruit, or from
both the top and bottom of fully-ripe fruit. Differences in mechanical (firmness) and chemical (total soluble solids, titratable
acidity, total non-structural carbohydrates) traits between different fruit portions were correlated with adult fruit utilisation.
Our results support a positive adult preference/offspring performance relationship at within-fruit level for B. dorsalis. The fine level of host discrimination exhibited by B. dorsalis is at odds with the general perception that, as a polyphagous herbivore, the fly should show very little discrimination in

Pericarp polypeptide profiles were analyzed at three ripening stages in the F1 hybrid and the F2 population from the cross between the accessions: LA1385 (Lycopersicon esculentum var. cerasiforme) and 804627 (L. esculentum, a homozygous genotype for the nor mutant). Six polymorphic polypeptides were observed in LA1385, while no polymorphic polypeptides among ripening stages was
observed in 804627. On the other hand, some polypeptides in the F1 hybrid were not observed in the parents whereas others were present in both parental genotypes and were unnoticeable in the
hybrid genotype. From a cluster analysis on the protein profiles of the F2 population, the differential expression of proteins allowed to distinguish mature green (MG) stage from the others two stages,
while for breaker stage (BR) and red ripe stage, the genetic background was more important in forming groups. The differential
expression of proteins could be associated with fruit morphology traits such as a 72 kDa polypeptide present in MG stage with

Quantitative trait loci (QTLs) identified so far in soybean were mainly derived in the final stage of plant development, which
did not apply to the exploitation of genetic effects that were expressed during a specific developmental stage. Thus, the
aim of this study was to identify conditional QTLs associated with yield traits at a specific developmental interval of soybean
plant. The 143 recombinant inbred lines developed from the cross of soybean cultivars ‘Charleston’ and ‘Dongnong 594’ were
used for the developmental QTLs analysis of pod number in the main stem and plant height by composite interval mapping method
combined with mixed genetic model. The results indicated that the number and type of QTLs and their genetic effects for the
two agronomic traits were different in a series of measuring stages. A total of 10 unconditional QTLs in 6 linkage groups
and 5 conditional QTLs in 3 linkage groups were identified for the pod number of the main stem, while 13 unconditional QTLs
in 7 linkage groups and 12 conditional QTLs in 6 linkage groups were identified for plant height. Many QTLs that were detected
in the early stages were different from those detected at the later stages. Some QTLs existed only at one stage and others
existed across two or three stages. Five marker intervals (satt509-satt251, sat_099-sat_113, sat_113-OPAW19_4, satt457-OPC10_85,
sat_095-OPBA08_5) were proven to be associated both with the development of pod number in the main stem and the development
of plant height. The present study suggested that the development of pods and plant height in soybean were governed by time-dependent

Fruit ripening process is associated with change in carotenoid profile and accumulation of lycopene in tomato (Solanum lycopersicum L.). In this study, we quantified the β-carotene and lycopene content at green, breaker and red-ripe stages of fruit ripening in eight tomato genotypes by using high-performance liquid chromatography. Among the genotypes, lycopene content was found highest in Pusa Rohini and lowest in VRT-32-1. To gain further insight into the regulation of lycopene biosynthesis and accumulation during fruit ripening, expression analysis of nine carotenoid pathway-related genes was carried out in the fruits of high lycopene genotype—Pusa Rohini. We found that expression of phytoene synthase and β-carotene hydroxylase-1 was four and thirty-fold higher, respectively, at breaker stage as compared to red-ripe stage of fruit ripening. Changes in the expression level of these genes were associated with a 40% increase in lycopene content at red-ripe stage as compared with breaker stage. Thus, the results from our study suggest the role of specific carotenoid pathway-related genes in accumulation of high lycopene during the fruit ripening processes. 相似文献

In studies of frugivorous tephritids, determining when offspring (i.e. egg and three larval instars) mortality occurs within the fruit can greatly improve the mechanistic understanding of the fly/host interaction. Previous research has demonstrated that the Queensland fruit fly, Bactrocera tryoni, has differential offspring performance in two tomato cultivars Cherry and Roma, but when juvenile mortality was occurring was not determined. We examined B. tryoni egg and larval survival in three different ripening stages (immature-green (IG), colour-break (CB) and fully-ripe (FR)) of Cherry and Roma tomato cultivars through destructive fruit sampling at 72 and 120 hr for eggs, and 48 (1st instar), 96 (2nd instar) and 120 hr (3rd instar) after fruit inoculation with neonates for larvae. Cultivar and ripening stage had no significant effect on egg survival, nor larval survival at 48 hr: the overall percentage of egg survival was at least 80% across all treatments, while 1st-instar larval was less than 52% across all treatments. In immature-green tomatoes of both varieties, nearly all mortality occurred during the first and second instars, but at 96 and 120 hr, there were significant interaction effects between cultivar and ripening stage on larval survival. In both colour-break Cherry and Roma tomatoes, there was significant larval mortality between 96 and 120 hr. However, in fully-ripe Cherry, no further significant larval mortality happened after 48 hr, while in fully-ripe Roma significant larval mortality occurred between the first and second larval instars but not thereafter. The difference in timing of larval mortality with ripening stage provides indirect evidence of active fruit defence which is strongest in immature-green fruit, less in colour-break fruit and absent in fully-ripe fruit. 相似文献

A cDNA library produced from mRNA isolated from the pericarp of wild-type tomato fruit (Lycopersicon esculentum Mill. cv Ailsa Craig) at the first visible sign of fruit ripening was differentially screened to identify clones whose homologous mRNAs were present at reduced levels in fruit of the tomato ripening mutant, ripening inhibitor,rin. Five clones were isolated (pERT 1, 10, 13, 14, 15). Accumulation of mRNA homologous to each of these clones increased during the ripening of wild-type fruit and showed reduced accumulation in ripening rin fruit. The levels of three of them (homologous to ERT 1, 13 and 14) were increased by ethylene treatment of the mutant fruit. A further clone, ERT 16 was identified for a mRNA present at a high level in both normal and mutant fruit at early stages of ripening. Database searches revealed no significant homology to the DNA sequence of ERT 14 and 15; however, DNA and derived amino acid sequence of ERT 1 both contain regions of homology with several reported UDP-glucosyl and glucuronosyl transferases (UDPGT) and with a conserved UDPGT motif. A derived amino acid sequence from the ERT 10 cDNA contains a perfect match to a consensus sequence present in a number of dehydrogenases. The ERT 13 DNA sequence has homology with an mRNA present during potato tuberisation. The presence of these mRNAs in tomato fruit is unreported and their role in ripening is unknown. The ERT 16 DNA sequence has homology with a ripening/stress-related cDNA isolated from tomato fruit pericarp. 相似文献
