A newly isolated strain, MU-2, which produces very high -fructofuranosidase activity, was identified asAspergillus japonicus. For enzyme production by the strain, sucrose at 20% (w/v) was the best carbon source and yeast extract at 1.5 to 3% (w/v) the best nitrogen source. Total enzymatic activity and cell growth were at maximum after 48 h, at 1.57×104 U/flask and 0.81 g dry cells/flask, respectively. The optimum pH value of the enzymatic reaction was between 5.0 and 5.5 and the optimum temperature 60 to 65°C. The enzyme produced 1-kestose (O--d-fructofuranosyl-(21)--d-fructofuranosyl -d-glucopyranoside) and nystose (O--d-fructofuranosyl-(21)--d-fructofuranosyl-(21)--d-fructofuranosyl -d-glucopyranoside) from sucrose by fructosyl-transferring activity. The strain was found to be very useful for industrial production of -fructofuranosidase.  相似文献

The major pentasaccharides Fuc(1-2)[GalNAc(1-3)]Gal(1-4)[Fuc(1-3)]Glc and Fuc(1-2) [Gal(1-3)]Gal(1-4)[Fuc(1-3)]Glc, which are normally present in the urine of bloodgroup A Leb and B Leb healthy subjects, were each found to be contaminated by a minor component when analysed by1H-NMR. Nicolaia elatior is used as an example to demonstrate that the mucronate tepals ofZingiberaceae correspond to hypsophylls (bracts) consisting of a leaf sheath and a rudimentary Oberblatt (= leaf petiole + lamina) represented by the mucro. Evidence for this interpretation is furnished by all available criteria: leaf sequence (exhibiting a complete continuum of forms from foliage leaves over cata- and hypsophylls to the tepals), nervature, and ontogeny.The present conception is compared with the well-founded thesis ofLeinfellner that the perigone ofLiliaceae is derived from the androecium. The different morphological status of the perigone in both families is not regarded as the result of different phylogenetic origin, but as a manifestation of morphogenetic transgressions from one phyllome category to an adjacent one: In theLiliaceae the perigone is under a strong morphogenetic influence of the androecium, and therefore displays staminal characters, in theZingiberaceae it is under the dominating influence of the extrafloral region, and thus appears as a hypsophyllous structure. If this assumption of a morphologically oscillating perigone is correct, it will be fundamentally impossible to demonstrate unequivocally the phylogenetic origin of the monocotyledonous perigone.

Some closely related members of the monocotyledonous familiesAlismataceae, Liliaceae, Juncaceae, Cyperaceae, Poaceae andAraceae with variable modes of pollination (insect- and wind-pollination) were studied in relation to the ultrastructure of pollenkitt and exine (amount, consistency and distribution of pollenkitt on the surface of pollen grains). The character syndromes of pollen cementing in entomophilous, anemophilous and intermediate (ambophilous or amphiphilous) monocotyledons are the same in principal as in dicotyledons. Comparing present with former results one can summarize: 1) The pollenkitt is always produced in the same manner by the anther tapetum in all angiosperm sub-classes. 2) The variable stickiness of entomophilous and anemophilous pollen always depends on the particular distribution and consistency of the pollenkitt, but not its amount on the pollen surface. 3) The mostly dry and powdery pollen of anemophilous plants always contains a variable amount of inactive pollenkitt in its exine cavities. 4) A step-by step change of the pollen cementing syndrome can be observed from entomophily towards anemophily. 5) From the omnipresence of pollenkitt in all wind-pollinated angiosperms studied one can conclude that the ancestors of anemophilous angiosperms probably have been zoophilous (i.e. entomophilous) throughout.

UDP-GlcNAc: Man1-6R (1-2)-N-acetylglucosaminyltransferase II (GlcNAc-T II; EC 2.4.1.143) is a key enzyme in the synthesis of complexN-glycans. We have tested a series of synthetic analogues of the substrate Man1-6(GlcNAc1-2Man1-3)Man-O-octyl as substrates and inhibitors for rat liver GlcNAc-T II. The enzyme attachesN-acetylglucosamine in 1-2 linkage to the 2-OH of the Man1-6 residue. The 2-deoxy analogue is a competitive inhibitor (K i=0.13mm). The 2-O-methyl compound does not bind to the enzyme presumably due to steric hindrance. The 3-, 4- and 6-OH groups are not essential for binding or catalysis since the 3-, 4- and 6-deoxy and -O-methyl derivatives are all good substrates. Increasing the size of the substituent at the 3-position to pentyl and substituted pentyl groups causes competitive inhibition (K i=1.0–2.5mm). We have taken advantage of this effect to synthesize two potentially irreversible GlcNAc-T II inhibitors containing a photolabile 3-O-(4,4-azo)pentyl group and a 3-O-(5-iodoacetamido)pentyl group respectively. The data indicate that none of the hydroxyls of the Man1-6 residue are essential for binding although the 2- and 3-OH face the catalytic site of the enzyme. The 4-OH group of the Man-O-octyl residue is not essential for binding or catalysis since the 4-deoxy derivative is a good substrate; the 4-O-methyl derivative does not bind. This contrasts with GlcNAc-T I which cannot bind to the 4-deoxy-Man- substrate analogue. The data are compatible with our previous observations that a bisectingN-acetylglucosamine at the 4-OH position prevents both GlcNAc-T I and GlcNAc-T II catalysis. However, in the case of GlcNAc-T II, the bisectingN-acetylglucosamine prevents binding due to steric hindrance rather than to removal of an essential OH group. The 3-OH of the Man1-3 is an essential group for GlcNAc-T II since the 3-deoxy derivative does not bind to the enzyme. The trisaccharide GlcNAc1-2Man1-3Man-O-octyl is a good inhibitor (K i=0.9mm). The above data together with previous studies indicate that binding of the GlcNAc1-2Man1-3Man- arm of the branched substrate to the enzyme is essential for catalysis. Two trisaccharide glycosides,p-trifluoroacetamidophenylethyl 3-O-(2-acetamido-2-deoxy--d-galactopyranosyl)-2-O-(-l-fucopyranosyl)--d-galactopyranoside andp-trifluoroa-cetamidophenylethyl 2-O-(-l-fucopyranosyl)-3-O-(-d-galactopyranosyl)--d-galactopyranoside, corresponding to the human blood group A and B determinants, were synthesized. A key fucosylgalactosyl disaccharide derivative was glycosylated with galactosaminyl or galactosyl donors, respectively. Two extracellular -glucosidases (cellobiase, EC 3.2.1.21), I and II, from Aspergillus nidulans USDB 1183 were purified to homogeneity with molecular weights of 240,000 and 78,000, respectively. Both hydrolysed laminaribiose, -gentiobiose, cellobiose, p-nitrophenyl--L-glucoside, phenyl--L-glucoside, o-nitrophenyl--L-glucoside, salicin and methyl--L-glucoside but not -linked disaccharides. Both were competitively inhibited by glucose and non-competitively (mixed) inhibited by glucono-1,5-lactone. -Glucosidase I was more susceptible to inhibition by Ag+ and less inhibited by Fe2+ and Fe3+ than -glucosidase II.  相似文献

A more precise taxonomic concept ofE. hirtella and its infraspecific synonymy is presented. Its diploid nature (2n = 22) is confirmed. Within the European area ofE. hirtella five different races may be recognised: typical, brandisii, capitulata, Rofan and Bretagne. Taxonomic rank is not yet attributed to these races. The heterogeneous taxonomic assembly E. drosocalyx is disentangled. The type refers to products of hybrid introgression ofE. rostkoviana-characters (long glandular hairs) intoE. minima.

An in vitro propagation technique based on axillary bud proliferation has been developed for matureSapium sebiferum trees. Nodal segments cultured on Murashige and Skoog (MS) medium supplemented with benzyl adenine (1–10 m and -naphthaleneacetic acid (0–0.5 m showed axillary bud proliferation. Shoots proliferated in vitro were multiplied on Murashige and Skoog medium containing 2.5 m benzyl adenine and 0.25 m -naphthaleneacetic acid. Seasonal changes affected the shoot proliferation potential of the initial explant. Shoots were rooted on a half-strength, growth-regulator-free, agar-gelled, MS medium after a 48-h treatment on half-strength MS liquid medium with 10 m indole-3-butyric acid. Rooted plantlets were potted and acclimatized in a growth chamber and then moved to the greenhouse. The induction by d,l-nicotine of the enantiozymes 6-hydroxy-L-nicotine oxidase and 6-hydroxy-D-nicotine oxidase in Archrobacter oxidans was differently affected by the inhibitors of Escherichia coli gyrase, novobiocin and nalidixic acid. These compounds inhibited 6-hydroxy-L-nicotine oxidase induction slightly, but led to an increase in the level of 6-hydroxy-D-nicotine oxidase activity. Summary Castellani's water culture method for microscopic fungi was re-examined and confirmed in its every detail. It is an ideal method for, at least, the smaller culture collection, in order to avoid continuous, short term subculturing. Benedek's suggestion for establishment of a mycotheca preserved in chlorallactophenol on the analogy of the herbarium of phanerogamists, may fill a long felt gap in mycological laboratories.
Zusammenfassung Castellani's Wasserkultur-methode für mikroskopische Pilze wurde einer Kontrolluntersuchung unterzogen. Sie konnte in all ihren Einzelheiten bestätigt werden. Sie ist eine ideale Methode, wenigstens für kleine Kultursammlungen, um eine fortgesetzte, kurz-fristige Überimpfung zu vermeiden. Benedek's Vorschlag für die Einrichtung einer Mykotheka in Chlorallactophenol in Analogie mit dem Herbarium für Blütenpflanzen, mag eine lang gefühlte Lücke im mykologischen Laboratorium beseitigen.

Résumé La méthode deCastellani, la culture en d'eau des champignons microscopiques, a été ré-examinée. Elle pouvait être confirmée en tous les détails. C'est une méthode idéale, au moins, pour la collection de cultures de petite dimension, pour éviter les transferts continus et d'intervalle courte.La suggestion deBenedek pour établir une mycothèque en chlorallactophénol, par analogie avec l' herbier pour les phanérogames peut remplir une lacune, existante pour long temps, aux laboratoires mycologiques.

We have analysed the mucins synthesized by the HT-29 MTX cell subpopulation, derived from the HT-29 human colon carcinoma cells through a selective pressure with methotrexate (Lesuffleuret al., 1990,Cancer Res 50: 6334–43), in the presence of benzyl-N-acetyl--galactosaminide (GalNAc-O-benzyl), which is a potential competitive inhibitor of the 1,3-galactosyltransferase that synthesizes the T-antigen. The main observation was a 13-fold decrease in the sialic acid content of mucins after 24 h of exposure to 5mm GalNAc-O-benzyl. This effect was accompanied by an increased reactivity of these mucins to peanut lectin, testifying to the higher amount of T-antigen. The second observation was a decrease in the secretion of the mucins by GalNAc-O-benzyl treated cells. -Glucuronidase from callus cultures of Scutellaria baicalensis Georgi was purified to apparent homogeneity by fractionated ammonium-sulfate precipitation and chromatography on diethylaminoethyl-cellulose, hydroxylapatite and baicalin-conjugated Sepharose 6B. A 650-fold purification was obtained by this purification system. When subjected to sodium dodecyl sulfate-polyacrylamide gel electrophoresis the purified protein migrated as a single band with a molecular mass of 55 kDa. We determined that the native enzyme has a molecular mass of 230 kDa using gel-filtration chromatography. These results suggested that the enzyme exists as a homotetramer composed of four identical 55-kDa subunits. The enzyme showed a broad pH optimum between 7.0 and 8.0. The K m values were 9 M, 10 M, 30 M and 40 M for luteolin 3 -O--d-glucuronide, baicalin, wogonin 7-O--d-glucoronide and oroxlin 7-O--d-glucuronide, respectively. Zusammenfassung Die Retinula im Ommatidium der Mehlmotte besteht aus einer wechselnden Anzahl (9–12, meist 11) langgestreckter, prismatischer Sinneszellen. Außerdem enthält jede Retinula nahe der Basalmembran im Zentrum zwischen diesen distalen Retinulazellen noch eine basale Retinulazelle. Die Längsachse der Retinula wird von der Achsenstruktur eingenommen, die aus Mikrovilli besteht. Ihr distaler Teil ist der Achsenfaden, der breitere, proximale Teil bildet das Rhabdom. Dieses erscheint im Querschnitt meist vierstrahlig gelappt, da seine Außenseite in Längsrichtung tief gekehlt ist. Der Rhabdomquerschnitt gliedert sich in mehrere Schöpfe parallel angeordneter Mikrovilli (Rhabdomsektoren); jeder Rhabdomsektor besteht aus 1 oder 2 Rhabdomeren. Die basale Retinulazelle entsendet einen kleinen Schopf von Mikrovilli in die proximale Spitze des Rhabdoms. Die distalen Retinulazellen setzen sich proximal in Neuriten fort, welche sich in Einkehlungen der basalen Retinulazelle bzw. der Tracheenendzelle einschmiegen. Jeweils eine Tracheole durchbricht zusammen mit dem Neuritenstrang einer Retinula die Basalmembran; sie verzweigt sich distal zu ca. 30 Tracheolen, die die Retinula umhüllen.Die Kristallkegelzellen grenzen distal an die Cornea; proximal laufen die Kristallkegelzellen eines Ommatidiums in einen gemeinsamen Fortsatz aus, der zwischen den Retinulazellen unmittelbar am Achsenfaden endet. — Nur das helladaptierte Auge wurde untersucht. Hierbei erscheint im distalen Teil der Retinula nur der Achsenfaden lichtdurchlässig, das Cytoplasma der Retinulazellen hingegen von Pigmentgrana durchsetzt und für Licht undurchlässig.
Fine structure of the eye of the meal moth, Ephestia kuehniella Zeller (Lepidoptera, Pyralididae)
Summary In each ommatidium of the meal moth a retinula is formed from a varying number (9–12, mostly 11) of elongated, prismatic sense cells. In addition, a basal retinular cell is situated near the basement membrane in the center of the other (distal) retinular cells. The axis of the retinula is occupied by many microvilli forming the axial structure, the distal section of which is the slender axial thread. Proximally, the axial structure widens (to 8.5 m instead of 1 m in diameter) and is now called rhabdom. Cross sections of the rhabdom mostly look like a petaloid with four petals; this figure is due to longitudinal infoldings along the length of the rhabdom surface. The rhabdom cross section is subdivided into several brushes of microvilli (rhabdom sectors), each one being characterized by an approximately parallel arrangement of its microvilli. One rhabdom sector may be composed of one or two rhabdomeres respectively.The basal retinular cell participates in rhabdom formation through a small brush of microvilli at the proximal end of the rhabdom. Proximally, the distal retinular cells taper into slender neurites which are embedded in grooves at the surface of the basal retinular cell and the tracheal end cell respectively. One tracheole piercing the basement membrane together with the neurites of one retinula branches into about 30 tracheoles surrounding the retinula.The crystalline cone cells touch the cornea; proximally, their cytoplasm forms a point which eventually terminates amongst the distal tips of the retinular cells, immediately at the axial thread.—Our work was restricted to light adapted eyes; in this condition, light transmission in the distal part of the retinula seems to be blocked by retinular cell pigment except inside the axial thread.

Callus growth and the production of anthocyanins were sustained on the salts and vitamins of Murashige and Skoog. Callus growth was stimulated at a concentration of 8–32 M -naphthaleneacetic acid (NAA) and 2,4-dichlorophenoxyacetic acid (2,4-d). Benzyladenine (BA) and zeatin at 8 M inhibited callus growth whereas isopentenyladenine (iP) stimulated callus growth. NAA repressed anthocyanin production with an increase in NAA from 8–32 M. Anthocyanin synthesis was promoted by an increase in 2,4-d from 0.5 to 2 M and decreased thereafter up to a concentration 32 M 2,4-d. A concentration of 8 M BA, thidiazuron and zeatin, respectively stimulated pigment production. N-acetylhexosaminidase fromNocardia orientalis catalysed the synthesis of lacto-N-triose II glycoside (-d-GlcNAc-(1-3)--d-Gal-(1-4)--d-Glc-OMe,3) with its isomers -d-GlcNAc-(1-6)--d-Gal-(1-4)--d-Glc-OMe (4) and -d-Gal-(1-4)-[-d-GlcNAc-(1-6)]--d-Glc-OMe (5) throughN-acetylglucosaminyl transfer fromN,N-diacetylchitobiose (GlcNAc2) to methyl -lactoside. The enzyme formed the mixture of trisac-charides3, 4 and5 in 17% overall yield based on GlcNAc2, in a ratio of 20:21:59. Withp-nitrophenyl -lactoside as an acceptor, the enzyme also producedp-nitrophenyl -lacto-N-trioside II (-d-GlcNAc-(1-3)--d-Gal-(1-4)--d-Glc-OC6H4NO2-p,6) with its isomers -d-GlcNAc-(1-6)--d-Gal-(1-4)--d-Glc-OC6H4NO2-p (7) and -d-Gal-(1-4)-[-d-GlcNAc-(1-6)]--d-Glc-OC6H4NO2-p (8). In this case, when an inclusion complex ofp-nitrophenyl lactoside acceptor with -cyclodextrin was used, the regioselectivity of glycosidase-catalysed formation of trisaccharide glycoside was substantially changed. The trisaccharide 2-(p-trifluoroacetamidophenyl)ethyl 2-acetamido-2-deoxy-4-O-[2-O-(-l-fucopyranosyl)--d-galactopyranosyl]--d-glucopyranoside 1 and the tetrasaccharide 2-(p-trifluoroacetamidophenyl)ethyl 2-acetamido-2-deoxy-3-O-(-l-fucopyranosyl)-4-O-[2-O-(-l-fucopyranosyl)--d-galactopyranosyl]--d-glucopyranoside 2 were synthesized. Root, hypocotyl and cotyledon explants of Sesbania bispinosa, Sesbania cannabina, Sesbania formosa, and Sesbania sesban were cultured on Murashige and Skoog medium with benzyladenine (BA; 2.22, 4.44, 8.88 M) in combination with 2,4-dichlorophenoxyacetic acid (2,4-d; 2.26, 4.52, 9.05 M), indolebutyric acid (IBA; 0.25, 0.49, 4.92 M) or naphthaleneacetic acid (NAA; 2.69, 5.37, 10.74 M). Although all explant types developed some callus, callus occurred earliest and continued to grow fastest with hypocotyls. Media including 2.4-d or NAA gave the fastest growing callus. Callus was subcultured up to 10 times at 20-day intervals and retained a rapid growth rate. Shoots regenerated readily from both hypocotyls or cotyledons but not from roots. Shoot organogenesis was most frequent with IBA (0.25–4.92 M) in combination with BA (4.44–8.88 M) and did not occur with 2,4-d. With each species at least one medium induced shoot differentiation from more than 50 percent of the callus pieces. With one exception, media containing IBA that induced shoot organogenesis on explants also did so in callus, but media containing NAA, even when effective with explants, did not cause differentiation of callus. Shoots that differentiated were excised and cultured on MS medium without growth regulators or with IBA (2.46, 4.92, 9.84 M). Roots developed after 3–8 days on an appropriate rooting medium, often without IBA. Rooted plantlets were transplanted to pots in a greenhouse and developed into normal plants. Suitable media and protocols for initiating and subculturing callus and regenerating whole plants in vitro from callus and explants have thus been established for four species of Sesbania.  相似文献

Homogenized tissues and their alkali-soluble and alkali-insoluble fractions of fruiting bodies of the basidiomycetes Laetiporus sulphureus and Piptoporus betulinus were investigated using X-ray diffraction, infrared spectrometry and chemical methods. The presence of (13)--d-glucan, (13)--d-glucan and chitin was established. The relative amounts of these polysaccharides were different in the two species and differences were also found between context and trama. The proportion of (13)--d-glucan was exceptionally high in the context of L. sulphureus (about 78%). In addition, the trama of both species contained a substance resembling a cyclic wax by its X-ray pattern and solubility properties. Summary According to the program Palynological Italian Flora, Aeropalynological section, the pollen morphological card ofPinus pinea L. is presented. The study is carried out on pollen coming from three Italian localities and regards fresh and acetolyzed pollen. For each sample, measurements are carried out on 30 fresh pollen grains in glicerol jelly with fuchsin and on 30 acetolyzed pollen grains in water/glicerol (1/1); general observations regard 1000 fresh and 1000 acetolyzed pollen grains/sample. Some observations on the main differences between fresh and acetolyzed pollen are mentioned.
