The slender mutant of barley resembles a normal barley plant treated with high doses of gibberellic acid (GA3). Expression of GA3-regulated and abscisic acid (ABA)-regulated mRNAs was studied in the endosperm and roots of mutant and wild-type (WT) plants.Production of -amylase (EC 3.2.1.1) by WT embryoless half-grains was dependent on the presence of GA3, and was prevented by ABA. In contrast, -amylase was produced by half-grains of the slender mutant in the absence of added GA3, although it was still reduced by ABA. The spectrum of -amylase mRNAs in slender embryoless half-grains incubated in the absence of added GA3 was the same as in WT endosperm half-grains incubated in the presence of GA3. These results indicate that the endosperm of the slender mutant exhibits similar properties to WT endosperm treated with GA3.In roots the expression of an ABA-inducible mRNA was similar in slender and WT seedlings either treated with exogenous ABA or exposed to dehydration. Seed maturation of Pisum sativum cv. Progress No. 9 proceeds more slowly in winter than in summer even when the parent plants are grown in greenhouse conditions with light-and heat-supplementation. For parent plants grown under summer and winter conditions the metabolism of [3H]GA9 in cultured seeds is qualitatively different in seeds of equivalent age and qualitatively the same in seeds of equivalent weight. 13-Hydroxylation of [3H]GA9[3H]GA20 is restricted to early stages of seed development. 2-Hydroxylation of [3H]GA92-OH-[3H]GA9 has only been observed at a stage of development after endogenous GA9 has accumulated. 2-OH-GA9 has been shown to be endogenous to pea and is named GA51. H2-GA31 and its conjugate have not been shown to be present in pea and may be induced metabolites of [3H]GA9. The metabolism of GA20GA29 is used to illustrate a technique of feeding [2H][3H]GAs in order to distinguish a metabolite from the same endogenous compound. The in vitro conversion of [3H]GA20[3H]GA29, and the virtual non-metabolism of [3H]GA29 have been confirmed for seeds in intact fruits. The metabolism of GA29 in maturing seeds of Pisum sativum cv. Progress No. 9 was further investigated, and the utility of 2H-labelled GAs in conjuction with GC-MS is illustrated. Using [2-2H1]GA29 as an internal standard, endogenous GA29 was shown to reach a maximal level (ca. 10 g/seed) 27 days from anthesis, and to decline to ca. 1.6 g/seed in mature seeds. In a time-course feed the metabolism of [2-2H1] [2-3H1]GA29 applied to 27 day old seeds, and of endogenous GA29, was compared from the 1H:2H ratios in the recovered GA29. Although both [2-2H1] [2-3H1]GA29 and endogenous GA29 were metabolised to the same limited extent to a putative conjugate, in the main metabolic process endogenous GA29 was preferentially converted to an untraceable (i.e. unlabelled) metabolite. In contrast, endogenous GA29 and [1,3-2H2] [1,3-3H2]GA29, derived from [1,3-2H2] [1,3-3H2]GA20 in a time-course feed, were metabolised in an identical manner. In the latter case isotope loss precluded identification of the metabolite. The structure (8) has been assigned to a GA catabolite present in maturing seeds and seedlings of pea. A time-course study is described relating the enzyme activities for GA20 metabolism with seed development in Phaseolus vulgaris L. Enzyme activity for the 3-hydroxylation of GA20 to GA1, and for the 2,3-desaturation of GA20 to GA5, was confined to the cotyledons and showed maximal specific activity at 21 d after anthesis. These enzyme activities co-occurred, together with a much lower level of activity for the 2-hydroxylation of GA20 to GA29. The observed rates of GA1, GA5 and GA29 formation from GA20 were constant under a range of incubation conditions. Enzyme activity for the conversion of GA1 to GA8 was detected only in embryos of seed from 40 d after anthesis. By deuterium-labelling and analysis of the products by gas chromatography-selected ion monitoring it was shown that 2-hydroxylation of GA1 to GA8 and 3-hydroxylation of GA20 to GA1 occur with retention of configuration and that the conversion of GA20 to GA5 occurs with loss of the 2- and 3-hydrogens. Isoelectrofocusing, product analysis, thermal denaturation studies and affinity chromatography on cycloheptaamylose-Sephadex were used to identify the amylolytic enzymes in internodes of deepwater rice (Oryza sativa L.). Amylolytic activity in internodes of deepwater rice consists of -amylase (sometimes separated into two isoforms) and of -amylase. During submergence of whole plants, -amylase activity increases in young, growing internodes, but -amylase activity declines. Although non-growing, mature internodes contain higher levels of -amylase than do the elongating younger internodes, the effect of submergence on amylase activities in both tissues follows the same trend. Submergence, gibberellic acid (GA3) and ethylene all promote -amylase activity in growing and non-growing internodes of excised deepwater-rice stem sections. Inhibitor studies showed that submergence and ethylene promote -amylase activity in the absence of endogenous gibberellin (GA), and GA3 enhances -amylase activity when ethylene action is inhibited. Therefore, ethylene and GA appear to increase -amylase activity independently of each other. Summary Solute osmotic potentials (x) in the vessels of hydroponically grown maize roots were measured to assess the osmotic-xylem-sap mechanism for generating root pressure (indicated by guttation). Solutes in vessels were measured in situ by X-ray microanalysis of plants frozen intact while guttating. Osmotic potentials outside the roots (o) were changed by adding polyethylene glycol to the nutrient solution. Guttation rate fell when o was decreased, but recovered towards the control value during 3–5 days when o was greater than or equal to –0.3 MPa, but not when o was equal to –0.4 MPa. In roots stressed to o = –0.3 MPa, x, was always more positive than o, and x changed only slightly (ca. 0.05 MPa). Thus the adjustment in the roots which increased root pressure cannot be ascribed to x, contradicting the osmotic-xylem-sap mechanism. An alternative driving force was sought in the osmotic potentials of the vacuoles of the living cells (v), which were analysed by microanalysis and estimated by plasmolysis. v showed larger responses to osmotic stress (0.1 MPa). Some plants were pretreated with abundant KNO3 in the nutrient solution. These plants showed very large adjustments in v (0.4 MPa) but little change in x (0.08 MPa). They guttated by 4 h after o was lowered to –0.4 MPa. It is argued that turgor pressure of the living cells is a likely alternative source of root pressure. Summary The ability of gibberellic acid (GA3) to prevent the light inhibition of stem elongation in peas was examined for several varieties under a wide range of irradiation conditions.A saturating dose of GA3 largely prevented the inhibitory effect of red light on total stem height in Duke of Albany (tall), Alaska (medium) and Meteor (dwarf) although a small, but statistically significant, effect persisted in all varieties after 3 days of light. The growth of the second internode was, however, markedly inhibited by red light even with a saturating dose of GA3. With gibberellin there was no difference between the effects of continuous red light and 15 minutes per day on height but the second internode was much shorter in the former treatment. The number of internodes present was the same in both cases and, therefore, the upper internodes in continuous light were as long or longer than in the 15-minute treatment. The number of internodes was only slightly fewer in darkness than in light so that, with GA3, the effect of red light was transient and only the growth of the lower internodes was inhibited. Without GA3 overall height was less in both red light treatments than in darkness for all three varieties.In blue light, on the other hand, there was no difference depending on whether height or internode length is considered, and even with a saturating dose of GA3 the growth rate remained depressed in continuous blue light. There was, however, some interaction between blue light and GA3.Red/far-red reversal experiments showed that in the varieties Alska and Duke of Albany the far-red stimulation of elongation persisted in the presence of a saturating dose of GA3 while for the dwarf variety Meteor there was a significant interaction between far-red and GA3.At least a quantitative difference was found between tall and dwarf peas in their response to light. Tall varieties showed a much greater effect of a prolonged exposure to blue and a smaller effect of a short exposure to red than dwarf varieties. Increasing the duration of exposure to red increasingly inhibited the growth of tall varieties. The medium variety Alaska grew to approximately the same height in continuous red and blue light.  相似文献

Plantlet formation through organogenesis in callus cultures of Himalayan yellow poppy,Meconopsis paniculata D.Don (Prain), a threatened taxon of ornamental value, is described. Hypocotyl segments from 3-month-old laboratory-raised seedlings produced callus on agar-solidified Murashige and Skoog medium (MS) containing 10 M -naphthaleneacetic acid and 1 M kinetin. Shoots differentiated best from callus on MS containing 10 M indolebutyric acid (IBA) and 1 M 6-benzyladenine. The regenerated shoots rooted best on MS medium containing 10 M IBA. Summary Exogenous gibberellic acid, A3 (GA3) inhibits phytochrome mediated betacyanin synthesis in seedlings of Amaranthus caudatus. The growth retardants, -chloroethyl-trimethylammonium chloride (CCC), 'isopropyl-4-(triethylammonium chloride)-5-methylphenyl piperidine carboxylate (AMO 1618) and tributyl-2,4,-dichlorobenzylphosphonium chloride (phosphon D) enhance pigment synthesis. Retardant stimulation of pigment synthesis is overcome by GA3 application. Besides lowering endogenous GA levels the retardants inhibit protein synthesis by as much as 25%. Retardant inhibition of protein synthesis is not overcome by GA3. The results suggest that amaranthin synthesis in Amaranthus caudatus can be directly controlled by endogenous GA. GA3 has no effect on kinin induced dark pigment synthesis. The effects of excess Cu as affected by the application of exogenous hormones (gibberellic acid - GA3 and indole-3-acetic acid - IAA) with respect to sunflower (Helianthus annuus L.) growth, physiology, and metabolism were studied. Application of 100 M IAA lessened the toxic effects of 80 M Cu in roots indicating greater root length and root hair formation, while addition of 100 M GA3 ameliorated the toxic effect mainly to the shoot. The content of photosynthetic pigments significantly declined under Cu stress, whereas application of hormones led to a substantial preservation of chlorophylls and carotenoids. Under Cu stress, the rate constant of energy trapping by photosystem 2 (PS2) reaction centres (RCs) was reduced as a result of physical dissociation of the light-harvesting complex (LHC) from PS2 core, while application of IAA and especially GA3 resulted in stability of the LHC of PS2 RCs. The drop in net photosynthetic (PN) and transpiration (E) rates with preserved or slightly reduced variable to maximum fluorescence ratio (Fv/Fm) in the presence of 80 M Cu could be explained by a possible inhibition of the enzymatic processes in the Calvin cycle. Application of 100 M IAA and 100 M GA3 lessened Cu effects mainly on P N. Water use efficiency was also improved under hormone exposure.  相似文献

The gibberellin (GA) 2-hydroxylases in mature and immature seeds of Pisum sativum have been partially purified and characterised. The enzymes are unstable when stored below pH 7.0 or in the absence of a thiol reagent. The optimum assay pH is between 7.4 and 7.8 and activity is dependent upon the presence of -ketoglutarate, Fe2+ and ascorbate. The 2-hydroxylase activities for GA1, GA4, GA9 and GA20 are chromatographically inseparable and correspond to a protein of Mr 44000. The rate of GA 2-hydroxylation varies according to substrate and some evidence indicates that the 2-hydroxylase activities for GA1 and GA4 and for GA9 and GA20 may reside in different proteins. During pea seed maturation, the specific activity of the enzyme(s) increases dramatically and reaches a maximum at a time when endogenous GA9, GA20, GA29 and GA51 are also at their greatest concentration. This correlation is not the result of substrate induction of enzyme activity. Since the GA 2-hydroxylases operate at maximal rate at low substrate concentrations they are incapable of rapidly 2-hydroxylating excessive quantities of (exogenously applied) GA1 or GA20. The influence of some macronutrients, especially NH4NO3 and KNO3, on root development of microcuttings from 3 apple scion cultivars is discussed. A reduction of the level of NH4NO3 in the medium from full strength to 1/4 strength significantly increased the percentage rooting of Gala and Royal Gala, but not Jonagold. Further reduction of NH4NO3 level from 1/4 strength to zero significantly reduced the percentage of rooting in Gala but not Royal Gala. Jonagold rooted best at zero concentration NH4NO3. Without NH4NO3, rooting percentages were as high as 100% for all 3 cultivars when KNO3 was provided at full strength. The results show that adventitious roots can be induced on apple scion cultivars by media manipulation.  相似文献

Zusammenfassung Von 1957–1990 blieb der Brutbestand der Blaumeise in 3 Kontrollgebieten (Flächengröße insgesamt 370 ha) gleich, doch ergaben sich beträchtliche jährliche Fluktuationen. Männliche Brutvögel hatten ein signifikant höheres Durchschnittsalter (2,01±0,04 Jahre) als weibliche (1,72±0,03 Jahre). Die danach ermittelte Mortalität beträgt 50 % () bzw. 58 % (). Im Durchschnitt siedelten gesichert näher am Geburtsort als ( =365 bzw. 700 m). Als maximale Ansiedlungsentfernung konnten 24 km () bzw. 470 km () nachgewiesen werden. Auch die Ortswechsel zwischen Bruthöhlen verschiedener Jahre waren bei gesichert geringer als bei ( =40 bzw. 75 m). Die weitesten Brutumsiedlungen fanden über 0,75 km () bzw. 37 km () statt. Emigration über >5 km vom Geburtsort wurde für maximal 4,2 der Nestlinge nachgewiesen. Abwanderungen vom Geburtsort über große Distanzen (>100 km) erfolgten bevorzugt nach WSW (mittlerer Vektor: 241,5°). Zwischen Brutpaardichte und dem -Wert der Zufallsfunde im 1. Lebensjahr (>5 km vom Geburstort) besteht von 1960–1989 ein gesichert positiver Zusammenhang. Seit 1976 konnte jedoch selbst in Jahren mit Bestandsspitzen keine überdurchschnittliche Emigration mehr festgestellt werden. Die Rückmelderate (alle Funde >5 km vom Geburtsort) nahm von 1960–1989 signifikant ab.
Population dynamics of the Blue Tit (Parus caeruleus): Longterm investigations in the Braunschweig region
Summary In more than 20 study areas, spread over approximately 1200 km2 near Braunschweig (52.16 N 10.32 E), the population dynamics of Blue Tits breeding in nestboxes has been studied. In three study areas (circa 370 ha overall) no significant long term trend was recognisable from 1957 to 1990. There were, however, considerable fluctuations in the particular years.Males had a significantly higher average age (2,01±0,04 years) than females (1,72±0,03 years). Accordingly the mortality rate of adult Blue Tits amounted to 50 % () and 58 % (). On average males settled significantly nearer from their birth place than females (median values: 365 and 700 m). Maximum distances of 24 km () and 470 km () were recorded. Change of locality between nestboxes of different years was significantly smaller for males than for females (median values: 40 and 75 m). The farthest breeding resettlement was in distances of 0,75 km () and 37 km ().Emigration over more than 5 km from birth place was established for a maximum of 4,2 of all Blue Tit nestlings of a particular years. Migrations over distances >100 km from birth place to recovery area were predominantly in WSW (average vector: 241,5°). There is a significantly positive relationship between the breeding-pair density of Blue Tits and the -rate of chance recoveries (more than 5 km from birth place during first year of life) of the years 1960 to 1989. Since 1976, however, above average emigration — even in years of peak populations — could not be established. The rate of recovery (all findings more than 5 km from birth place) declined significantly in the years 1960 to 1989.

A broad-host-range plasmid (pEA2-21) containing a Bradyrhizobium sp (F-4) nod DABC-lacZ translation fusion was constructed and used to monitor nod gene expression in response to pigeonpea root exudate. Two nod-inducing compounds were isolated and identified. Spectral analysis using ultraviolet absorption, infrared spectra, proton nuclear magnetic resonance, and mass spectrometry showed that the two inducers were 5,4-dihydroxy-6-(3-methyl-2-butenyl)-2, 2-dimethyl pyrano-[5, 6:7, 8]-flavanone (cajaflavanone) and 2,4,5-trihydroxy-5-isopentenyl-6, 7-dimethylchromene iso-flavanone (cajanone). When pEA2-21 was introduced into Rhizobium trifolii and R. meliloti cajanone and cajaflavanone did not induce nod gene indicating that specificity of induction appears to be influenced by the host-strain genome.  相似文献

The stem growth in darkness or in continuous red light of two pea cultivars, Alaska (Le Le, tall) and Progress No. 9 (le le, dwarf), was measured for 13 d. The lengths of the first three internodes in dark-grown seedlings of the two cultivars were similar, substantiating previous literature reports that Progress No. 9 has a tall phenotype in the dark. The biological activity of gibberellin A20 (GA20), which is normally inactive in le le geno-types, was compared in darkness and in red light. Alaska seedlings, regardless of growing conditions, responded to GA20. Dark-grown seedlings of Progress No. 9 also responded to GA20, although red-light-grown seedlings did not. Gibberellin A1 was active in both cultivars, in both darkness and red light. The metabolism of [13C3H]GA20 has also been studied. In dark-grown shoots of Alaska and Progress No. 9 [13C3H]GA20 is converted to [13C3H]GA1, [13C3H]GA8, [13C]GA29, its 2-epimer, and [13C3H]GA29-catabolite. [13C3H] Gibberellin A1 was a minor product which appeared to be rapidly turned over, so that in some feeds only its metabolite, [13C3H]GA8, was detected. However results do indicate that the tall growth habit of Progress No. 9 in the dark, and its ability to respond to GA20 in the dark may be related to its capacity to 3-hydroxylate GA20 to give GA1. In red light the overall metabolism of [13C3H]GA20 was reduced in both cultivars. There is some evidence that 3-hydroxylation of [13C3H]GA20 can occur in red light-grown Alaska seedlings, but no 3-hydroxylated metabolites of [13C3H]GA20 were observed in red light-grown Progress. Thus the dwarf habit of Progress No. 9 in red light and its inability to respond to GA20 may be related, as in other dwarf genotypes, to its inability to 3-hydroxylate GA20 to GA1. However identification and quantification of native GAs in both cultivars showed that red-light-grown Progress does contain native GA1. UDP-GlcNAc: Man1-6R (1-2)-N-acetylglucosaminyltransferase II (GlcNAc-T II; EC 2.4.1.143) is a key enzyme in the synthesis of complexN-glycans. We have tested a series of synthetic analogues of the substrate Man1-6(GlcNAc1-2Man1-3)Man-O-octyl as substrates and inhibitors for rat liver GlcNAc-T II. The enzyme attachesN-acetylglucosamine in 1-2 linkage to the 2-OH of the Man1-6 residue. The 2-deoxy analogue is a competitive inhibitor (K i=0.13mm). The 2-O-methyl compound does not bind to the enzyme presumably due to steric hindrance. The 3-, 4- and 6-OH groups are not essential for binding or catalysis since the 3-, 4- and 6-deoxy and -O-methyl derivatives are all good substrates. Increasing the size of the substituent at the 3-position to pentyl and substituted pentyl groups causes competitive inhibition (K i=1.0–2.5mm). We have taken advantage of this effect to synthesize two potentially irreversible GlcNAc-T II inhibitors containing a photolabile 3-O-(4,4-azo)pentyl group and a 3-O-(5-iodoacetamido)pentyl group respectively. The data indicate that none of the hydroxyls of the Man1-6 residue are essential for binding although the 2- and 3-OH face the catalytic site of the enzyme. The 4-OH group of the Man-O-octyl residue is not essential for binding or catalysis since the 4-deoxy derivative is a good substrate; the 4-O-methyl derivative does not bind. This contrasts with GlcNAc-T I which cannot bind to the 4-deoxy-Man- substrate analogue. The data are compatible with our previous observations that a bisectingN-acetylglucosamine at the 4-OH position prevents both GlcNAc-T I and GlcNAc-T II catalysis. However, in the case of GlcNAc-T II, the bisectingN-acetylglucosamine prevents binding due to steric hindrance rather than to removal of an essential OH group. The 3-OH of the Man1-3 is an essential group for GlcNAc-T II since the 3-deoxy derivative does not bind to the enzyme. The trisaccharide GlcNAc1-2Man1-3Man-O-octyl is a good inhibitor (K i=0.9mm). The above data together with previous studies indicate that binding of the GlcNAc1-2Man1-3Man- arm of the branched substrate to the enzyme is essential for catalysis. Container-grownEuphorbia lathyris plants were treated with foliar sprays of various combinations of BA and GA4+7 or 0–3600 mg L–1 Promalin (11 BA + GA4+7) in separate experiments. GA4+7 and Promalin stimulated plants to grow taller. BA and Promalin promoted axillary shoot growth. Multiple applications of Promalin stimulated branching more than single treatments. Dry weight accumulation was stimulated only if the growth regulators were applied to 28–33-cm and not to 56-cm tall plants. Chemical names used: (1, 2, 4a, 4b, 10)-2,4a,7-trihydroxy-1-methyl-8-methylenegibb-3-ene-1,10-dicarboxylic acid 1,4a-lactone (GA4+7),N-(phenylmethyl)-H-purin-6-amine (BA), and Promalin [11 (wt/wt) GA4+7 and BA].The use of the name Promalin or other trade names does not imply endorsement to the exclusion of other products or vendors that may also be suitable.  相似文献

Callus was initiated from immature leaf and stem segments of rose (Rosa hybrida cv. Landora) and subcultured every four weeks on a basal medium of half-strength Murashige & Skoog (1962) salts plus 30 g l-1 sucrose (1/2 MS) and supplemented with 2.2 M BA, 5.4 M NAA and 2.2–9.0 M 2,4-D. Embryogenic callus and subsequently somatic embryos were obtained from 8-week-old callus culture on 1/2 MS+2.2 M BA+0.05 M NAA+0.3 M GA3+200–800 mg l-1 L-proline. Long-term cultures were established and maintained for up to 16 months by repeated subculture of embryogenic callus on L-proline deficient medium. The proportion of spurs flowering on apple trees (Malus domestica Borkh. cv Golden Delicious) displaying a high degree of alternate-year flowering was increased in the off year by gibberellin A4 (GA4) and C-3 epi-GA4 applied in the previous year. When applied 4.5 weeks after anthesis amounts of GA4 ranging from 3 to 300 g per spur and 25 or 50 g of C-3 epi-GA4 per spur were effective. Treatments with GA4 made seven weeks after anthesis were less effective. A simple and rapid pea regeneration procedure was developed. An average of up to 20 shoots formed from hypocotyl explants of cvs. Sugar Ann and Patriot cultured on Murashige and Skoog basal medium supplemented with 0.5 or 1.0 M thidiazuron (N-phenyl-N-1,2,3-thiadiazol-5-ylurea). Hypocotyls of Puget and Sugar Daddy did not respond. Regenerated shoots rooted rapidly when cultured on Murashige and Skoog basal medium containing either 2.0 M -naphthaleneacetic acid or 1.0–2.0 M indole-3-butyric acid.
