The distribution of Peridinium willei and P. volzii was studied in Danish lakes. Both species were confined to lakes with concentrations of Total P < 0.15 mg 1-1, with the majority of occurrences at Total P concentration between 0.020–0.040 mg 1-1 and concentrations of PO4 P between detection limit and 0.040 mg 1-1. The occurrence of the species in relation to inorganic N compounds (NH4 N and NO2+ NO3 N) was significantly broader for P. willei than for P. volzii: P. willei had an almost even distribution within a wide range of NH4 N, whereas P. volzii mainly occurred between 0.001 and 0.10 NH4 N 1-1. P. willei had an almost even distribution at values beween 0.005 and 0.42 mg NO2+ NO3 N 1-1, whereas P. volzii mainly occurred below 0.050 mg NO2+ NO3 N 11. P. willei was found at pH values between 4.2 and 8.5, whereas P. volzii was confined to lakes with a slightly basic pH. The study confirmed the broad limits of P. willei and the much more narrow limits of P. volzii in relation to seasonal occurrence and pH, as well as an affinity of the former to ponds and lakes with a rich bottom vegetation. The study also showed, however, that the species were not as widespread and common in recent Danish lake phytoplankton as generally stated by previous authors. The use of different ecological factors to give weight to species separation is discussed. The inclusion of P. volzii in P. willei proposed by Popovsky & Phiester is not supported by the present study, as the two taxa appear to have different ecological tolerances.  相似文献

Abstract The effects of organic additions on nitrification and dentrification were examined in sediment microcosms. The organic material, heat killed yeast, had a C/N ratio of 7.5 and was added to sieved, homogenized sediments. Four treatments were compared: no addition (control), 30 g dry weight (dw) m−2 mixed throughout the 10 cm sediment column (30M), 100 g dw m−2 mixed throughout sediments (100M), and 100 g dw m−2 mixed into top 1 cm (100S). After the microcosms had been established for 7–11 days, depth of O2 penetration, sediment-water fluxes and nitrification rates were measured. Nitrification rates were measured using three different techniques: N-serve and acetylene inhibition in intact cores, and nitrification potentials in slurris. Increased organic additions decreased O2 penetration from 2.7 to 0.2 mm while increasing both O2 consumption, from 30 to 70 mmol O2 m−2 d−1, and NO3 flux into sediments. Nitrification rates in intact cores were similar for the two methods. Highest rates occurred in the 30M treatment, while the lowest rate was measured in the 100S treatment. Total denitrification rates (estimated from nitrification and nitrate fluxes) increased with increased organic addition, because of the high concentrations of NO3 (40 μM) in the overlaying water. The ratio of nitrification: denitrification was used as an indication of the importance of nitrification as the NO3 supply for denitrificaion. Abstract. Presence of 2.4-diaminobutyric acid (A2bu), a neurotoxin, in tissues of flatpea ( Lathyrus sylvestris L.) necessitates a thorough understanding of the regulation of this nonprotein amino acid before the species can be recommended to livestock producers for forage applications. To determine how different concentrations and ratios of NO3 and NH+4 in growth media influence the levels of A2bu and other free amino acids in the 'Lathco'flatpea cultivar, plants were grown hydroponically in controlled environments. The concentration of A2bu was highest in tissues when the NO3 to NH+4 ratio in the nutrient solution was low. Responses of amides and other nonprotein amino acids, especially in the roots, followed a similar trend. Free protein amino acids in leaves and stems were generally unaffected by changes in NO3 to NH+4 ratios. In roots, protein amino acids increased as the NO3 to NH+4 ratio in the growth medium increased. Ammonium inhibited shoot and root growth; NO3 alleviated the toxic effects of NH+4. Soluble protein concentrations were higher in the shoots of NO3-fed plants and in the roots of plants supplied with NH+4. These results suggest that accumulation of A2bu and other nonprotein amino acids, as well as asparagine and glutamine, plays a role in detoxification of NH+4 and storage of N.  相似文献

Abstract A reaction diffusion model was used to simulate the mineralization processes in an Arctic sediment. The simulation and the actual sediment were compared in relation to profiles of O2, NO3 and NH4+. The site of particulate organic matter (POM) degradation was the single most important factor in fitting the simulation profiles to those of the sediment. It was deduced that most POM degradation occurred close to the sediment surface. When a reasonably good simulation had been obtained, the sensitivity of the model to changes in other parameters was investigated. Increases in POM degradation in the upper sediment resulted in increases in concentration of NH4+ and NO3, but further increases in POM degradation created anoxic conditions below 3 mm, resulting in decreases in NO3 concentrations. The model was relatively intensive to changes in POM degradation in the lower sediment layers; increases led to more anoxic conditions and to less NO3. Increases in the C/N ratio of the POM in the lower sediment layers had little effect; increases in C/N in the upper layers led to a decrease in NH4+ and NO3. The model was sensitive to changes in the first order rate constant for nitrification, but not for denitrification. Decreases in the K m for O2 of the nitrifying bacteria had no effect on the profiles.  相似文献

The induction of activity of the enzyme nitrate reductase (NR, EC 1.6.6.1, 1.6.6.2) in needles of Norway spruce ( Picea abies [L.] Karst.) by nitrogen dioxide (NO2) was studied under laboratory and field conditions. In fumigation chambers an increase in nitrate reductase activity (NRA) was detected 4 h after the start of the NO2 treatment. During the first 2 days with 100 µg NO2 m−3, NRA reached a constant level and did not change during the following 4 days. At the same level of NO2, NRA was lower in needles from trees grown on NPK‐fertilized soil than on non‐fertilized soil. After the transfer of spruce trees from fertilized soil to NPK‐rich nutrient solution, NRA was transiently increased. This effect was assigned to root injuries causing nitrate transport to the shoot and subsequent induction of NRA. Neither trees on fertilized soil nor trees transferred to NPK‐poor nutrient solution had increased NRA unless NO2 was provided. The NO2 gradient in the vicinity of a highway was used to test the long‐term effect of elevated levels of NO2 on needle NRA of potted and field‐grown spruce trees. Compared with less polluted sites, permanently increased NRAs were detected when NO2 concentrations were above 20 µg m−3. Controls of field measurements some 10 years after the introduction of catalytic converters in cars showed no significant change neither in NO2 levels nor in the decreasing NRA of spruce needles with the distance from the highway.  相似文献

Dry deposition of NH3 and NOx (NO and NO2) can affect plant metabolism at the cellular and whole-plant level. Gaseous pollutants enter the plant mainly through the stomata, and once in the apoplast NH3 dissolves to form NH4+, whereas NO2 dissolves to form NO3 and NO2. The latter compound can also be formed after exposure to NO. There is evidence that NH3-N and NOx-N can be reversibly stored in the apoplast. Temporary storage might affect processes such as absorption rate, assimilation and re-emission. Once formed, NO3 and NO2 can be reduced, and NH4+ can be assimilated via the normal enzymatic pathways, nitrate reductase (NR), nitrite reductase and the glutamine synthetase/glutamate synthase (GS/GOGAT) cycle. Fumigation with low concentrations of atmospheric NH3 increases in vitro glutamine synthetase activity, but whether this involves both or only one of the GS isoforms is still an open question. There seems to be no correlation between fumigation with low concentrations of NH3 and in vitro GDH activity. The contribution of atmospheric NH3 and NO2 deposition to the N budget of the whole plant has been calculated for various atmospheric pollutant concentrations and relative growth rates ( RGRs ). It is concluded that at current ambient atmospheric N concentrations the direct impact of gaseous N uptake by foliage on plant growth is generally small.  相似文献

The reduction of nitrate by Pseudomonas denitrificans in a culture medium containing glycerol, yeast extract and 700 mg/1 NO3–N, was antagonized by Aeromonas hydrophila, Escherichia coli and Enterobacter aerogenes. Nitrate reduction by Ps. denitrificans in mixed culture with a fermentative heterotroph was inhibited when 100–150 mg/1 NO2–N had accumulated in the medium. The number of Ps. denitrificans declined concomitantly with the appearance of NO2 in the culture medium, but there was only a slight increase in the numbers of fermentative hetero-trophs. The fermentative heterotrophs did not antagonize nitrate reduction by Ps. denitrificans , when the culture medium contained 140 mg/1 NO3–N. When mixtures of equal parts of Ps. denitrificans and Esch. coli cultures were added to autoclaved river water relatively high concentrations of NO2 were produced from the nitrate present in the water.  相似文献

Pollutants like O3 and NO2 enter leaves through the stomata and cause damage during reactions with components of biological cell membranes. The steady-state flux rates of these gases into the leaf are determined by a series of physical and biochemical resistances including stomatal aperture, reactions occurring within the cell wall and the ability of the leaf to remove the products of apoplastic reactions. In the present study, multiple regression models incorporating stomatal conductance, apoplastic and symplastic ascorbate concentrations, and nitrate reductase (NR) activities were generated to explain the observed variations in leaf-level flux rates of O3 and NO2. These measurements were made on the plant Catharanthus roseus (Madagascar periwinkle). The best-fit model explaining NO2 flux included stomatal conductance, apoplastic ascorbate and NR activity. This model explained 89% of the variation in observed leaf fluxes and suggested physical resistances, reaction between NO2 and apoplastic ascorbate, and the removal rate of nitrate (generated by reactions of NO2 and water) from the apoplast all play controlling roles in NO2 flux to leaves. O3 flux was best explained by stomatal conductance and symplastic ascorbate explaining 66% of the total variation in leaf flux. Both models demonstrate the importance of measuring processes other than stomatal conductance to explain steady-state leaf-level fluxes of pollutant gases.  相似文献

The possibility to induce nitrate reductase (NR; EC 1.6.6.2) in needles of Scots pine ( Pinus sylvestris L.) seedlings was studied. The NR activity was measured by an in vivo assay. Although increased NR activities were found in the roots after application of NO3, no such increase could be detected in the needles. Detached seedlings placed in NO3 solution showed increasing NR activities with increasing NO3 concentrations. Exposure of seedlings to NOx (70–80 ppb NO2 and 8–12ppb NO) resulted in an increase of the NR activity from 10–20 nmol NO2 (g fresh weight)−1 h−1 to about 400 nmol NO2 (g fresh weight)−1 h−1. This level was reached after 2–4 days of exposure, thereafter the NR activity decreased to about 200 nmol NO2 (g fresh weight)−1 h−1. Analyses of free amino acids showed low concentrations of arginine and glutamine in NOx-fumigated seedlings compared to corresponding controls.  相似文献

The appearance of soil NO3 after forest disturbance is commonly ascribed to a higher availability of NH4+ to autotrophic nitrifiers, or to a reduction in available-C resulting in lower microbial assimilation of NO3. Alternatively, it has been proposed that increasing NH4+ pools following disturbance could increase net nitrification by reducing microbial assimilation of NO3. Forest floor material was collected from shelterwood harvest plots which displayed both low available-C and low NH4+ pools, and where previous experiments had suggested the prevalence of heterotrophic nitrification. Subsamples were amended with incremental rates of glucose-C or NH4+, and gross NO3 transformation rates were measured by isotope dilution. Glucose-C additions had little effect on the net difference between gross NO3 production and consumption rates. On the other hand, NH4+ additions caused gross NO3 consumption processes to decrease sharply, while gross NO3 production processes remained constant. The results suggest that NH4+ can have an immediate positive effect on net nitrification rates by suppressing NO3 assimilation and uptake systems.  相似文献

The dose- and time-response effects of 3 days of 6 h day-time sequential exposures to NO2, SO2 and SO2+NO2 of 0.45–1.81 μl l−1 (ppm) SO2 and 1.50–7.65 μl l−1 NO2 on photosynthesis, transpiration and dark respiration were examined for nine Carpatho-Ukrainian half-sib families and a population from the GFR ('Westerhof') of Norway spruce [ Piecea abies (L.) Karst.], all in their 5th growing season.
SO2+NO2 inhibited photosynthesis and transpiration and stimulated dark respiration more than SO2 alone. SO2 and SO2+NO2 at the lowest concentrations inhibited night transpiration, but increased it at the highest concentration, the strongest effects being obtained with combined exposures. Photosynthesis of the different half-sib families was affected significantly differently by SO2+NO2 exposures. NO2 alone had no effects.
Sensitivity to transpiration decline correlated negatively with branch density. Height of trees correlated postitively with decline sensitivity in the seed orchard. The distribution of photosynthesis and transpiration sensitivities over all tested half-sib families correlated negatively with the distribution of decline sensitivity of their parents in a rural Danish seed orchard. The relative photosynthesis and transpiration sensitivities may thus serve as diagnostic parameters for selecting against novel spruce decline.  相似文献

Fertilization of bean plants grown in perlite with 1 and 3 mM CaCl2 or Ca(NO3)2 reduced severity of grey mould as compared with control plants or plants fertilized with 5 mM of the compounds. Fertilization with Ca(NO3)2 reduced severity leaf grey mould and fruit ghost spots of tomato plants grown in perlite by 70 and 45%, respectively. The rate of decrease varied with the position of the fruits on the plants. Leaves from plants treated with calcium or otherwise [KNO3, (NH4)2SO4] produced less ethylene than leaves of nontreated plants. Rate of growth of B. cinerea was lower on growth medium prepared from washings from leaves of calcium fertilized plants than from leaves from other treatments. The fertilizer combination Ca(H2PO4)2+ CaSO4 (1 and 3 g/kg soil) applied once to tomato plants grown in soil reduced severity of leaf grey mould by 80 % (significant at P = 0.05) but 1–3 g CaSO4/kg soil only tended to reduce disease severity (30–40 %, not significant) as compared with the control. The compounds CaCl2 and Ca(NO3)2 increased significantly ( P = 0.05) the growth of B. cinerea on synthetic medium when applied at rates of 1 0–10.0 mM whereas reduction of growth was observed with 0.1 mM of the compounds and of CaSO4.  相似文献

Abstract A consortium was enriched from a humisol incubated with 3.6 kPa CH4 and NH4+. This consortium oxidized NH4+ to NO2 and NO3 (NO3/NO2 ratio about 20) with smaller amounts of N2O. This oxidation stopped in the stationary phase after depletion of CH4. CH3OH or CO2 did not support oxidation. Growth and resting cell experiments suggested that nitrification was associated with methanotrophic activity and that chemoautotrophic nitrifiers were absent.  相似文献

In order to investigate effects of limited NO3 availability in corn ( Zea mays L. cv. Brulouis) 17-day-old plants were grown for a further 25 days on sand in a growth chamber. The plants received frequent irrigation with a complete nutrient solution containing 0.2, 0.6, 1.5 or 3.0 mM NO3. With 0.2 mM NO; nitrate levels in both roots and leaves diminished rapidly and were almost zero after 10 days treatment. Concurrently, as signs of nitrogen deficiency appeared, shoot growth was restricted, whereas root growth was enhanced. In addition, the concentration of reduced nitrogen and malate in the leaves declined, and in vitro nitrate reductase activity (NRA. EC 1.6.6.1), soluble protein and chlorophyll levels of leaf tissue were depressed and starch concentration was enhanced. With 0.6 mM NO3 in the nutrient solution, the decrease in NO3 levels in the tissues and the increase in root development were similar to those observed with 0.2 mM NO3. However, shoot growth, reduced nitrogen concentration in leaves, and the above-mentioned biochemical characteristics were almost identical to those obtained at 1.5 and 3.0 mM NO3. This indicates that when supplied with 0.6 mM NO3, corn plants were able to absorb sufficient NO3 to support maximal biomass production without appreciable NO3 accumulation in roots or shoot. It is, thus, suggested that the plants responded to low NO3, availability in medium by enhancing root growth and by maximizing NO3 reduction relative to NO3 accumulation.  相似文献

Changes in water and nutrient movement are common disturbances resulting from human activities in arctic regions. To assess the influence of water and nutrient movement on different plant growth forms, we added water and NPK fertilizer along 10 to 20 m linear transects across small natural drainages on an Alaskan tundra slope. Water was added by continuous-flow emitters from a drip irrigation system at a rate of 450 L m−1 d−1 during the 1986 growing season and 110 L m−1 d−1 in the 1987 growing season. NPK in the form of Osmocote, a slow release fertilizer, was applied at 0.5 kg per linear meter in early and mid-season of the 1985 growing season. Tissue N and P contents, light-saturated photosynthetic rates, and aboveground biomass production were measured at peak season for key species 2 m above and 2 and 6 m below the water and nutrient applications in 1986 and 1987.
Mean leaf N and P of the species tested increased slightly 2 m below the water addition sites and dramatically below the fertilizer addition sites. Increases in tissue N and P were also found 6 m below the fertilizer addition points. Leaf photosynthesis tended to increase 2 m below the water and nutrient additions for the species tested. Six meters below the treatment application points, photosynthesis was minimally affected in 1986 but increased in 1987. Increases in leaf area on the irrigation treatments were found only for the evergreen species, Ledum palustre and Vaccinium vitis-idaea , 2 m below the site of addition. In contrast, fertilizer addition caused large increases in leaf area production for all species tested at 2 m below the treatment, and for some species at 6 m below treatment. The overall effects of the two treatments were similar, but were greater for the fertilizer addition.  相似文献

Peanuts ( Arachis hypogaea L. cv. Shulamit) grown with NO3 and saline water in hydroponics responded positively to addition of nitrogen (N) in their vegetative growth, but not in desert dune sand. In order to clarify these conflicting results, peanut plants were grown in a greenhouse pot experiment with fine calcareous sand. The nutrient solution contained 0 or 50 m M NaCl and 2 or 6 m M N in the form of Ca(NO3)2, NH4NO3 or (NH4)2SO4. Three replicates were harvested after 48 days (beginning of reproductive stage) and three after 109 days (pod filling). In addition, gynophores were treated with 0, 50, 100, 150 or 200 m M NaCl outside the growth pot to check their sensitivity to salt. Shoot dry weight became greater with increasing NH4+/NO3 ratio. Increasing the N concentration from 2 to 6 m M did not change shoot dry weight of the NH4NO3 or NH4+-fed plants, but caused a reduction in shoot dry weight of NO3-fed plants. Shoot dry weight was not affected by increasing the NaCl concentration to 50 m M . Salt caused an increase in the number of gynophores per plant and a reduction of the mean pod weight. A NaCl concentration of 100 m M and above reduced gynophore vitality. It is concluded that the salt sensitivity of peanut plants resides mainly in the sensitivity of the reproductive organs.  相似文献

To develop further the methods for estimation of NOx absorption by plants supplied with 15N-labelled fertilizer, we proposed a new calculation method, total N fixed method (TNF), and compared with the 15N dilution method and the classical mass balance method (MB).
Hydroponically grown soybean plants were supplied with 15N-labelled nitrate and exposed to 200–250 nl l−1 NO2 for 7 d. The proportions of the N derived from NO2 to total N in exposed plants were estimated by the three methods.
The reported rates of NO2 absorption by several plant species, estimated by the 15N dilution method, were recalculated using the TNF method. The results of the two methods were compared and showed that: (1) The 15N dilution method overestimated the content of NO2-N in exposed plants compared with the MB method whilst the TNF method produced estimations of NO2-N closer to those by the MB method when the plants were supplied with 5 m M nitrate. (2) The differences in estimations between the MB method and either the 15N dilution method or the TNF method increased with decreasing supply of 15N-labelled nitrate to roots.  相似文献

The immediate and posteffects of various concentrations of NaNO2 on ion uptake of wheat ( Triticum aestivum L. cv. GK Öthalom) seedlings were studied at different pH values. Without pretreatment, the higher the concentration of NaNO2 the greater was the decrease in uptake of K+ into the roots, both at pH 4 and pH 6. At pH 6 but not at pH 4 the reverse was true when the seedlings were pretreated with NaNO2. Due to the high Na+ content of the roots, an effect of Na+ in this process cannot be excluded. Nitrite was taken up by the roots more rapidly than nitrate. Nitrite at 0.1 m M in the medium induced the development of an uptake system for both NO2 and NO3 in wheat roots. At higher concentrations pretreatment with NO2 decreased NO3 uptake by the roots, but NO3 did not inhibit the uptake of NO2. The toxic effect of NO2 was strongly pH dependent. Lower pH of the external solution led to an increased inhibition by NO2 of both ion uptake and growth of seedlings. The inhibitory effect of NO2 differed considerably for roots and shoots. The roots and especially the root hairs were particularly sensitive to NO2 treatment.  相似文献

Ecosystem flux measurements using the eddy covariance (EC) technique were undertaken in 4 subsequent years during summer for a total of 562 days in an arctic wet tundra ecosystem, located near Cherskii, Far-Eastern Federal District, Russia. Methane (CH4) emissions were measured using permanent chambers. The experimental field is characterized by late thawing of permafrost soils in June and periodic spring floods. A stagnant water table below the grass canopy is fed by melting of the active layer of permafrost and by flood water. Following 3 years of EC measurements, the site was drained by building a 3 m wide drainage channel surrounding the EC tower to examine possible future effects of global change on the tundra tussock ecosystem. Cumulative summertime net carbon fluxes before experimental alteration were estimated to be about +15 g C m−2 (i.e. an ecosystem C loss) and +8 g C m−2 after draining the study site. When taking CH4 as another important greenhouse gas into account and considering the global warming potential (GWP) of CH4 vs. CO2, the ecosystem had a positive GWP during all summers. However CH4 emissions after drainage decreased significantly and therefore the carbon related greenhouse gas flux was much smaller than beforehand (475 ± 253 g C-CO2-e m−2 before drainage in 2003 vs. 23 ± 26 g C-CO2-e m−2 after drainage in 2005).  相似文献

The plant fraction of alfalfa ( Medicago sativa L. cv. Aragon) nodules contained both nitrate reductase (NR) and nitrite reductase (NiR). Specific activity of NADH-NR from the cytosol of nodules not treated with NO3- was about 30 nmol (mg protein)-1-h-1 and was not basically affected by NO3 addition. In contrast, typical specific activity for cytosolic NiR was 1.5 umol (mg protein)-1h-1 using methyl viologen as electron donor. This activity strongly increased with NO3 concentration, probably due to substrate induction. Maximal activity was 3.5 μmol (mg protein)-1h-1 at 50 to 200 mM NO3.
Estimates indicate that the contribution of cytosol to the overall NR and NiR activities of alfalfa nodules is distinctly different: less than 10% and about 70%, respectively. The increasing amounts of NO2 accumulating in the cytosol upon NO3, supply, and the different response to NO3 of bacteroid and cytosolic NRs support the concept that most of this NO2 comes from the bacteroids.  相似文献
