1. When Fucus eggs which have been fertilized for a sufficient length of time are irradiated unilaterally with monochromatic ultraviolet light (λ2804 Å) of adequate dosage, 97–100 per cent form rhizoids on the halves of the eggs away from the source of radiation (see Figs. 1 and 2). 2. The responsiveness of the eggs increases gradually after fertilization and does not reach a maximum until about 7 hours at 15°C. (see Fig. 3). The first rhizoids begin to form in a population at about 12 hours after fertilization. The responsiveness remains maximal until at least 11 hours after fertilization. 3. It is suggested that the low responsiveness of a population of eggs at an earlier period is due to recovery from the effects of irradiation before the rhizoids begin to form. 4. The response of eggs to λ2804 Å is proportional, over a wide range, to the logarithm of the dosage (see Fig. 1). Dosage was regulated by the duration of exposure during the period of maximum response. 5. High dosages of λ2804 Å, of the order of 10,000 ergs per mm.2, cause the rhizoids to form fairly precisely away from the source of radiation (see Fig. 2). Twice this dosage inhibits rhizoid formation altogether without causing cytolysis. 6. Other wave-lengths which have also been shown to be effective are: 3660, 3130, 2654, 2537, 2482, and 2345 Å. Only exploratory measurements have been made to test the effectiveness of these wave-lengths, but they show that much greater energy is necessary to obtain a strong response with λ3130 and 3660 Å, especially the latter. The wave-lengths shorter than 2804 Å, on the other hand, show the same order of effectiveness as λ2804 Å. Some may be more effective. 7. A beam of λ2804 Å which is incident on a single layer of Fucus eggs is completely extinguished at 2, 3, 6, or 6½ hours after fertilization. About 85 per cent of a beam of λ3660 Å is extinguished. The wave-length 3660 Å is thus not so completely absorbed as λ2804 Å, but the difference in proportion absorbed by the egg is not nearly so great as the difference in effectiveness.  相似文献

When uninjured cells of Valonia are placed in methylene blue dissolved in sea water it is found, after 1 to 3 hours, that at pH 5.5 practically no dye penetrates, while at pH 9.5 more enters the vacuole. As the cells become injured more dye enters at pH 5.5, as well as at pH 9.5. No dye in reduced form is found in the sap of uninjured cells exposed from 1 to 3 hours to methylene blue in sea water at both pH values. When uninjured cells are placed in azure B solution, the rate of penetration of dye into the vacuole is found to increase with the rise in the pH value of the external dye solution. The partition coefficient of the dye between chloroform and sea water is higher at pH 9.5 than at pH 5.5 with both methylene blue and azure B. The color of the dye in chloroform absorbed from methylene blue or from azure B in sea water at pH 5.5 is blue, while it is reddish purple when absorbed from methylene blue and azure B at pH 9.5. Dry salt of methylene blue and azure B dissolved in chloroform appears blue. It is shown that chiefly azure B in form of free base is absorbed by chloroform from methylene blue or azure B dissolved in sea water at pH 9.5, but possibly a mixture of methylene blue and azure B in form of salt is absorbed from methylene blue at pH 5.5, and azure B in form of salt is absorbed from azure B in sea water at pH 5.5. Spectrophotometric analysis of the dye shows the following facts. 1. The dye which is absorbed by the cell wall from methylene blue solution is found to be chiefly methylene blue. 2. The dye which has penetrated from methylene blue solution into the vacuole of uninjured cells is found to be azure B or trimethyl thionine, a small amount of which may be present in a solution of methylene blue especially at a high pH value. 3. The dye which has penetrated from methylene blue solution into the vacuole of injured cells is either methylene blue or a mixture of methylene blue and azure B. 4. The dye which is absorbed by chloroform from methylene blue dissolved in sea water is also found to be azure B, when the pH value of the sea water is at 9.5, but it consists of azure B and to a less extent of methylene blue when the pH value is at 5.5. 5. Methylene blue employed for these experiments, when dissolved in sea water, in sap of Valonia, or in artificial sap, gives absorption maxima characteristic of methylene blue. Azure B found in the sap collected from the vacuole cannot be due to the transformation of methylene blue into this dye after methylene blue has penetrated into the vacuole from the external solution because no such transformation detectable by this method is found to take place within 3 hours after dissolving methylene blue in the sap of Valonia. These experiments indicate that the penetration of dye into the vacuole from methylene blue solution represents a diffusion of azure B in the form of free base. This result agrees with the theory that a basic dye penetrates the vacuole of living cells chiefly in the form of free base and only very slightly in the form of salt. But as soon as the cells are injured the methylene blue (in form of salt) enters the vacuole. It is suggested that these experiments do not show that methylene blue does not enter the protoplasm, but they point out the danger of basing any theoretical conclusion as to permeability on oxidation-reduction potential of living cells from experiments made or the penetration of dye from methylene blue solution into the vacuole, without determining the nature of the dye inside and outside the cell.  相似文献

It can be demonstrated by an indicator method that living cells are as freely permeable to oxygen as dead cells, and that sudden admission of oxygen to the cell cannot account for increased oxidation as a result of stimulation. Oxygen penetrates as readily as carbon dioxide among the acids and ammonia among the alkalies. Exposure of living plant cells to high oxygen pressures does not initiate certain oxidations (except after some hours), which proceed readily in dead plant cells in the air. In the light of the preceding statement, about the permeability of cells for oxygen, this is interpreted to mean that more oxygen enters the cell at high pressure, but that the reacting substances (chromogen and oxidase) are kept apart by some phase boundary as long as the cell is alive. Increased oxygen concentration eventually produces injury to the cell.  相似文献

1. The nerve cord of the lobster (Homarus americanus Milne-Edwards) is very delicate and can be used as a living preparation for only a few hours after its removal from the animal. 2. During the first hour or so after removal it discharges CO2 at a steadily decreasing rate beginning at about 0.20 mg. CO2 per gram of cord per minute and ending at about 0.07 mg. 3. This discharge exhibits a steady decrease in rate and is not divisible into a period of gush and a period of uniform outflow as with the lateral-line nerve of the dogfish. It terminates in a very few hours with the complete death of the cord. 4. Both handling and cutting the cord temporarily increase the rate of CO2 output. 5. The stimulated cord discharges CO2 at a rate about 26 per cent higher than that of the quiescent cord, an increase of about 1.6 times that of the increase observed in the lateral-line nerve of the dogfish under similar circumstances.  相似文献

The respiration of the green alga Chlorella pyrenoidosa, suspended in Knop''s solution, has been studied in the dark as a function of time and of temperature. The rates of oxygen consumption and of carbon dioxide production (at constant temperature) decline for about 25 hours to a low, constant level. From an analysis of the curves it is suggested that two substances, A and B, are utilized, whose respiratory quotients are 1 and 0.65 respectively. The values of the temperature characteristics were found to be: for oxidation of A, 19,500 (0.6 to 11.5°C.) and 3,500 (11.5 to 28°C.); for oxidation of B, 5,600 (23.4 to 0.6°C.).  相似文献

In the larva of the butterfly Calpodes ethlius, the fat body begins to store protein in the form of granules at about 30 to 35 hours before pupation, at a time when the endocuticle is being resorbed. At least two sorts of granule can be distinguished. The first granules to arise are those within vesicles of the Golgi complex. These may increase in size by incorporating material from microvesicles at their surface and by coalescence with one another. Later, at about 10 hours before pupation, another sort of granule arises by the isolation of regions of the endoplasmic reticulum (ER) within paired membranes derived from Golgi vesicles. Several of these ER isolation bodies coalesce, with fusion of their outer isolating membranes. The ribosomes and membranes may then disappear and the granules become indistinguishable from the protein granules formed from Golgi vesicles, or the ribosomes may remain and be embedded in dense crystalline protein, forming a storage body for both protein and RNA. Mitochondria are isolated within paired membranes in the same way as regions of the ER. The isolated mitochondria also coalesce in a similar manner. When the inner membranes are lost, the structure of a group of isolation bodies is indistinguishable from that of a cytolysome. Isolation within paired membranes, as described here, may be of general importance in segregating regions of massive lysis or massive sequestration.  相似文献

1. With constant temperature and light intensity, the rate of elongation of a sporangiophore of Phycomyces is constant for many hours. 2. With constant light intensity, values of µ, the "critical thermal increment," have been computed for the elongation of different sporangiophores. These values group themselves closely about the modes 11,000, 16,000, 20,000, 26,000, and 33,000 calories. Sporangiophores from the same culture need not have similar increments. 3. In this paper it is shown that if the dry seeds of the cantaloupe (Cucumis melo) are soaked for 3 hours in solutions of ethyl alcohol of concentration ranging from 2 to 16 per cent by volume, and then germinated and grown in distilled water in the dark, the total growth attained is greater by amounts ranging from 9 to 35 per cent than is that made by seeds treated in every way identically except that they are initially soaked in distilled water instead of alcohol. It is shown that this result is not due simply to differences in osmotic pressure in the different alcohol solutions. It is probably due to a simple selective action of the alcohol which eliminates the constitutionally weak and defective seeds.  相似文献

Kidney cells, predominantly from Cercopithecus monkeys but also from baboons, were infected in vitro with the SV40 virus. The infectious cycle was studied with the electron microscope by means of thin sections of cells fixed from 3 hours up to 11 days after infection. The frequency of virus formation and various nuclear and cytoplasmic lesions in relation to the infection are described. The virus particles appear in the nucleus in close contact with the chromatin. In a small number of cells they have been observed as early as 10 to 12 hours after infection, but most often they appear 24 to 48 hours afterward. Their mean diameter is 33 mµ. They have no membrane and are frequently arranged as crystal-like structures. In addition to the appearance of virus, one observes various lesions in the nucleoplasm and particularly in the nucleolus, which shows an early hypertrophy and produces unusual, dense condensations in contact with the nucleolonema. The importance of these nucleolar lesions and the relationship between the SV40 virus and the polyoma, common wart, and Shope papilloma viruses are discussed.  相似文献

An electron microscopical study of the third eye of the Western Fence Lizard, Sceloporus occidentalis, fixed with 1 per cent osmium tetroxide, pH 7.4–7.6, for 16 to 20 hours at 0°C., revealed the following new facts. The fibrillar system of the retinal photoreceptor consists of nine double fibrils enclosed in a sheath. Pigment cells and lens cells possess similar systems. Two short cylindrical centrioles are associated with the fibrillar apparatus: one, from which striated rootlets extend inward, lies at the base of the fibrils, with the other at an oblique angle to the axis of the system. A Golgi complex, whorls of endoplasmic reticulum, lipid (?) droplets, and other organelles and inclusions in the photoreceptors are described. An axon leads from the base of the photoreceptor into the nervous layer of the retina which consists of many nerve fibers and large ganglion cells. Although the pattern of neural connections has not yet been determined, some synapses were found. The parietal nerve consists of about 250 non-medullated fibers. The capsule of the eye usually has a layer of iridocytes, which contain rows of guanine (?) rods. A few parietal eyes of the Granite Night Lizard, Xantusia henshawi, were also examined. 1. Mackerel egg development was followed to hatching at constant temperatures of 10°, 11°, 12°, 13°, 14°, 15°, 16°, 17°, 18°, 19°, 20°, 21°, 22°, and 24°C. Experiment showed that typical development could be realized only between 11° and 21°. 2. The length of the developmental period increases from 49.5 hours to 207 hours when the temperature is lowered from 21° to 10°C. 3. The calculated µ for the development of the mackerel egg is about 19,000 at temperatures above 15° and approximately 24,900 for temperatures below 15°C. 15° is, apparently, a critical temperature for this process. 4. The calculated values of µ for eight stages of development preceding hatching, i.e. 6 somites, 12 somites, 18 somites, 24 somites, three-quarters circles, four-fifths circles, five-sixths circles, and full circles, are essentially the same as the µ''s for hatching, indicating that the rate of differentiation up to hatching is governed by one process throughout. Critical temperatures for these stages approximate 15°. 5. The total mortality during the incubation period was least at 16°C. where it amounted to 43 per cent. At temperatures above and below this there was a steady increase in the percentage of mortality which reached 100 per cent at 10° and 21°.  相似文献

Cessation of mitosis was brought about in Vicia faba roots incubated for 24 hours in the thymine analogue, 5-aminouracil. Recovery of mitotic activity began 8 hours after removal from 5-aminouracil and reached a peak at 15 hours. If colchicine was added 4 hours before the peak of mitoses, up to 80 per cent of all cells accumulated in mitotic division stages. By use of single and double labeling techniques, it was shown that synchrony of cell divisions resulted from depression in the rate of DNA synthesis by 5-aminouracil, which brought about an accumulation of cells in the S phase of the cell cycle. Treatment with 5-aminouracil may have also caused a delay in the rate of exit of cells from the G2 period. It appeared to have no effect on the duration of the G1 period. When roots were removed from 5-aminouracil, DNA synthesis resumed in all cells in the S phase. Although thymidine antagonized the effects of 5-aminouracil, an exogenous supply of it was not necessary for the resumption of DNA synthesis, as shown by incorporation studies with tritiated deoxycytidine.  相似文献

In the rhabditid nematode Caenorhabditis briggsae the incorporation of thymidine-H3 has been studied by autoradiography after Feulgen staining, with animals maintained under axenic conditions in a medium of only partly defined composition. Labeling has been followed in adults left in the presence of thymidine-H3 for periods of from ½ to 24 hours, as well as in adults reared from larvae in the presence of the tritiated nucleoside. A massive incorporation is found in the nuclei of the gonads and intestine; also a less intense particulate cytoplasmic incorporation is clear in certain cells, especially those of the intestine. In general, all labeling has proved to be sensitive to DNase, but resistant to RNase. The label's stability has been tested by the transfer of adults into a medium containing "cold" thymidine. They remain there for up to 48 hours. A transfer for 24 hours results in a considerable decrease in the intensity of nuclear and cytoplasmic labeling; a stay of 48 hours leads to its complete disappearance from non-dividing (intestinal) as well as dividing (gonadal) nuclei. A phenomenon of DNA turnover is envisaged and discussed as a possible physiological attribute of C. briggsae.  相似文献

Large amounts of glycogen accumulate in rat skeletal muscle fibers during the late fetal stages and are mobilized in the first postnatal days. This glycogen depletion is relatively slow in the immature leg muscles, in which extensive deposits are still found 24 hr after birth and, to some extent, persist until the 3rd day. In the more differentiated psoas muscle and especially in the diaphragm, the glycogen stores are completely mobilized already during the early hours. Section of the sciatic nerve 3 days before birth or within the first 2 hr after delivery does not affect glycogen depletion in the leg muscles. Neonatal glycogenolysis in rat muscle fibers takes place largely by segregation and digestion of glycogen particles in autophagic vacuoles. These vacuoles: (a) are not seen in fetal muscle fibers or at later postnatal stages, but appear concomitantly with the process of glycogen depletion and disappear shortly afterwards; (b) are prematurely formed in skeletal muscles of fetuses at term treated with glucagon; (c) contain almost exclusively glycogen particles and no other recognizable cell constituents; (d) have a double or, more often, single limiting membrane and originate apparently from flattened sacs sequestering glycogen masses; (e) are generally found to contain reaction product in preparations incubated from demonstration of acid phosphatase activity. The findings emphasize the role of the lysosomal system in the physiological process of postnatal glycogen mobilization and appear relevant in the interpretation of type II glycogen storage disease.  相似文献

Sprague-Dawley rats that had been subjected 2 months previously to partial resection (10 per cent) of the small intestine and an equal number of control rats were injected with tritiated thymidine and sacrificed at intervals during the subsequent 16 hours. Segments of duodenum, jejunum and ileum were prestained by the Feulgen technique and radioautographed. The proportion of crypt cells bearing labeled nuclei, the percentage of labeled crypt cells in mitosis and the appearance of labeled crypt cells on the villi were determined. Comparison of control and resected rats showed that (a) the proportion of intestinal crypt cells incorporating thymidine was considerably greater and uniformly high throughout the shortened intestine, (b) the life cycle of crypt cells was slightly reduced, and was uniform throughout the shortened intestine, and (c) the time during which cells were retained in crypts was markedly reduced. On the basis of persistent, generalized increase in the production of crypt cells, and on prior evidence that the epithelial cells of shortened intestine continue to have a brief life span and evidence of metabolic immaturity, the existence of a humoral factor, tentatively called "intestinal epithelial growth hormone," is postulated.  相似文献

A study was made of the diffusion of the red pigment echinochrome from the eggs of the sea urchin, Arbacia punctulata, into sea water. Unfertilized eggs retained their pigment, over periods of hours. Outward diffusion of pigment from unfertilized eggs normally is entirely negligible, or does not occur at all. Enchancing the calcium or potassium content of the artificial sea water (while retaining isosmotic conditions) did not induce pigment release. Under anaerobic conditions, unfertilized eggs release pigment in small quantities. Fertilization alone brings about echinochrome release. Fertilized eggs invariably released pigment, whether in normal sea water, or sea water with increased calcium or potassium. This diffusion of the pigment began during the first cleavage, possibly soon after fertilization. The pigment release is not a consequence solely of the cell''s permeability to echinochrome (or chromoprotein, or other pigment combination) but is preceded by events leading to a release of echinochrome from the granules in which it is concentrated within the cell. These events may be initiated by activation or by anaerobiosis. The phenomenon was not due to cytolysis.  相似文献

Details are given of the elimination of mitochondria which occurs during the first 1 to 2 hours of the life of the egg of the fern Pteridium aquilinum (L.) Kuhn. During this phase the only mitochondria present are swollen and appear degenerate, but subsequently the cytoplasm of the egg becomes filled with large mitochondria containing numerous villi. Accompanying the appearance of these mitochondria, many, if not all of which have a peculiar umbo-like form, is the production by the nucleus of conspicuous and complex evaginations. The umbo-mitochondria are believed to be new, and a mechanism is suggested by which they may be generated from the complex evaginations of the nucleus.  相似文献

Three-week-old male rats of the Wistar strain were given tritiated thymidine, 1 µc/gm body weight, intraperitoneally and were killed at intervals from 0.25 to 72 hours later. Autoradiographs were made from 5 µ sections, stained by the Feulgen method. The replication time and its component intervals were determined from the scoring of the labeling of interphase nuclei as well as of prophase, metaphase, anaphase, and telophase nuclei. Absorption of the intraperitoneally injected label is rapid and is attended by "flash" labeling during interphase. The results show that at any one time about 4 per cent of the liver cells are synthesizing DNA preliminary to cell division. These cells alternate with waves of other cells and it is estimated that about 10 per cent of the liver cell population is engaged in cell duplication. The replication time is about 21.5 hours, and its component intervals occupy the following times: DNA synthesis, 9 hours; post-DNA synthesis gap, 0.50 hour; prophase, 1.3 hours; metaphase, 1.0 hour; anaphase, 0.4 hour; telophase, 0.3 hour; postmitosis gap, 9.0 hours. A group of liver cells has been recorded in at least 3 successive replication cycles.  相似文献

In the autotrophic bacterium, Thiobacillus thiooxidans, the oxidation of sulfur is coupled to transfers of phosphate from the medium to the cells. CO2 fixation is coupled to transfers of inorganic phosphate from the cells to the medium and is dependent, in the absence of concomitant sulfur oxidation, upon the amount of phosphate previously taken up during sulfur oxidation. The energy reservoir, which is formed by sulfur oxidation in the absence of CO2 and which can be released for the fixation of CO2 under conditions which do not permit sulfur oxidation, is a phosphorylated compound and the data suggest that the energy is stored in the cell as phosphate bond energy. It is possible to oxidize sulfur at a constant rate for hours in the absence of CO2. The phosphate energy formed during this process is probably released by cell phosphotases. It is possible to inhibit these phosphotases by means of inorganic phosphate and thus to inhibit sulfur oxidation in the absence of CO2. In the presence of CO2, where alternative uses for the phosphate energy are available, the inhibition is relieved. Sulfur oxidation (energy input) is coupled, not to CO2 fixation, but to phosphate esterification. CO2 fixation (energy utilization) is coupled with phosphate release.  相似文献

The integration of the taxonomy of lichenized and non-lichenized pyrenocarpous ascomycetes is still in progress. Within this group, at least 12 lichenization events, three re-lichenizations and at least 24 de-lichenization events, are supposed to have occurred during evolution. This is evident from the increasing amount of information regarding phylogeny and classification of pyrenocarpous fungi. The total number of lichenization events, including re- and de-lichenization, within all fungi is estimated to be at least 100. However, the vast majority of the pyrenocarpous lichens are concentrated in a few relatively large, presumably monophyletic, groups. In some cases, proposed integrations have proven to be untenable, for exampleMicrotheliopsisand with theHerpotrichiellaceae,Flavobatheliumwith theDimeriaceaeand most notably theNormandinathallus with the basidio-mycetes and its ascocarps withSphaerulinain theMycosphaerellaceae. The same is true for many cases where lichenized taxa were unwittingly included in otherwise non-lichenized groups, such asDidymosphaeriaandMassarina, or even described in new, presumably non-lichenized, genera likeHolstiella,StarbaeckiellaandTitanella.  相似文献
