ABSTRACT

This investigation aims at assessing patterns of spatial genetic structure of the teleost fish Sardina pilchardus across the Siculo-Tunisian Strait (a well-known discontinuous biogeographic area) and delineating putative genetic stocks within the species. For this purpose, a total of 180 specimens, collected from 11 locations stretching across the western and eastern Mediterranean coasts of Tunisia, were analysed genetically by means of 18 nuclear allozyme loci. The outcome of this study revealed strong genetic differentiation among populations, with the marked genetic distinctiveness of the central Tunisian population at Mahdia. Despite the delineation of seven well-defined genetic groups, no significant correlation was found between genetic and geographic distances. Besides, the recorded population subdivision did not align with biogeographic boundaries, suggesting the presence of chaotic genetic patchiness. Recent genetic bottlenecks were evidenced in S. pilchardus populations. Patchy migration patterns were recorded among the examined pairs of sardine populations. Among the recorded 16 polymorphic loci, GPI-2 and SOD appeared to be subject to natural selection. Patterns of population genetic differentiation and structuring were not found to be driven by outlier loci that appeared to be under selection. Furthermore, the detected neutral GPI-1 locus was found to be responsible for most of the genetic variation among identified genetic clusters. Hence, natural selection cannot cause the detected genetic heterogeneity among sardine samples. Different explanations to the origin of chaotic genetic patterns, observed within S. pilchardus, were discussed.  相似文献

We investigate the influence of previously postulated biogeographic barriers in the Mediterranean Sea on the population genetic structure of a highly dispersive and continuously distributed coastal species. In particular, we examine nuclear and mitochondrial genetic variation in the marbled crab, Pachygrapsus marmoratus, across part of the African Mediterranean coast in order to assess the influence of the Siculo-Tunisian Strait on its population genetic structure. Four polymorphic microsatellite loci were genotyped for 110 individuals, collected from eight locations covering parts of the Algerian, Tunisian and Libyan coasts. In addition, mtDNA corresponding to the Cox1 gene was sequenced for 80 samples. The corresponding results show contrasting patterns of genetic differentiation. While mtDNA results revealed a homogeneous haplotype composition in our study area, microsatellite data depicted genetic differentiation among populations, but not associated with any geographic barrier. This pattern, already recorded for this species from different geographic regions, may hint at the involvement of a complex series of abiotic and biotic factors in determining genetic structure. Demographic history reconstruction, inferred from mtDNA data, supports demographic and spatial expansion for the North African metapopulation dating back to the Mid-Pleistocene and following an historical bottleneck. Comparison of these African mitochondrial sequences with new sequences from a Turkish population and previously published sequences revealed a weak but significant separation of Atlantic and Mediterranean populations across the Gibraltar Strait, which was not recorded in previous studies of this grapsid species.  相似文献

Nucleotide divergence in cytochrome c oxidase subunit I (COI) gene of mitochondrial DNA was analyzed to determine interpopulational variation of common octopus (Octopus vulgaris) species sampled along eastern Mediterranean. Nucleotide composition and nucleotide pair frequency analyses of 440 individuals representing 11 populations were investigated. Estimation of evolutionary divergence between sequences was conducted using Tamura-Nei model. Considering 440 individuals, 35 (5%) variable sites and an average variation of 1.9% were found between specimens. Highest divergence was found between Med11 and Med04-Med07 populations; with a genetic distance value of 0.039 (3.9%). Estimation of transition/transversion bias (R) was made under the Tamura-Nei model and resulted as 2.32. Phylogenetic tree was constructed using four methods: neighbor joining (NJ), minimum evolution (ME), maximum likelihood (ML) and unweighted pair-group method of arithmetic average (UPGMA). Results from clustering patterns of the consensus tree, with the sum of branch length = 0.05189, pointed out a correlation between genetic and geographic distance.  相似文献

Background  

The Strait of Gibraltar is a crucial area in the settlement history of modern humans because it represents a possible connection between Africa and Europe. So far, genetic data were inconclusive about the fact that this strait constitutes a barrier to gene flow, as previous results were highly variable depending on the genetic locus studied. The present study evaluates the impact of the Gibraltar region in reducing gene flow between populations from North-Western Africa and South-Western Europe, by comparing formally various genetic loci. First, we compute several statistics of population differentiation. Then, we use an original simulation approach in order to infer the most probable evolutionary scenario for the settlement of the area, taking into account the effects of both demography and natural selection at some loci.  相似文献

Vast amounts of effort have been devoted to investigate patterns of genetic diversity and structuring in plants and animals, but similar information is scarce for organisms of other kingdoms. The study of the genetic structure of natural populations of wild yeasts can provide insights into the ecological and genetic correlates of clonality, and into the generality of recent hypotheses postulating that microbial populations lack the potential for genetic divergence and allopatric speciation. Ninety‐one isolates of the flower‐living yeast Metschnikowia gruessii from southeastern Spain were DNA fingerprinted using amplified fragment length polymorphism (AFLP) markers. Genetic diversity and structuring was investigated with band‐based methods and model‐ and nonmodel‐based clustering. Linkage disequilibrium tests were used to assess reproduction mode. Microsite‐dependent, diversifying selection was tested by comparing genetic characteristics of isolates from bumble bee vectors and different floral microsites. AFLP polymorphism (91%) and genotypic diversity were very high. Genetic diversity was spatially structured, as shown by amova (Φst = 0.155) and clustering. The null hypothesis of random mating was rejected, clonality seeming the prevailing reproductive mode in the populations studied. Genetic diversity of isolates declined from bumble bee mouthparts to floral microsites, and frequency of five AFLP markers varied significantly across floral microsites, thus supporting the hypothesis of diversifying selection on clonal lineages. Wild populations of clonal fungal microbes can exhibit levels of genetic diversity and spatial structuring that are not singularly different from those shown by sexually reproducing plants or animals. Microsite‐dependent, divergent selection can maintain high local and regional genetic diversity in microbial populations despite extensive clonality.  相似文献

To determine, by means of quantitative analyses, the distribution patterns of the insectivores and rodents in Catalonia using physiographical regions as Operative Geographical Units (OGUs). Catalonia (north‐eastern Iberian Peninsula). Based on the presence/absence of twenty‐five small mammals in thirteen physiographical regions, which were used as OGUs, the following aspects were studied: (a) identification of biotic boundaries; (b) determination of chorological association of species; (c) climatic characterization of the biotic regions and the chorotypes. Groups of biotic regions and species were established, and their statistical significance was tested. The possible effect of several climatic factors on these groupings was studied using discriminant analyses. A significant biotic boundary was found to separate the central and eastern Pyrenees from the remaining physiographical regions. The climatic variables that defined this boundary were related to the severity and the availability of environmental energy. Four chorotypes were identified. One chorotype was constituted by Pyrenean or Pre‐Pyrenean species, an association determined by their mid‐European requirements; another chorotype was formed by Eurosiberian species, but showed a variable degree of tolerance to Mediterranean conditions; a third chorotype included species with a wide distribution that are Mediterranean, anthropic, generalist or have very specific habitats; and finally the fourth chorotype was constituted by a strictly Mediterranean species. The climatic factors that accounted for the distribution of these chorotypes were the mean July temperature and the mean annual precipitation. We conclude that the axial zone of the Pyrenees, except the coastal portion, determines two biotic regions in Catalonia. As for the classification of species, using quantitative techniques for the first time, we offer a new biogeographicalal configuration for the small mammals in this temperate‐Mediterranean transition.  相似文献

Posidonia oceanica is an endemic seagrass species in the Mediterranean Sea. In order to assess levels of genetic structure in this species, the microsatellite polymorphism was analysed from meadows collected in several localities, along the coasts of the Tyrrhenian Sea (Mediterranean Sea). The existence of single population units and the recruitment of seedlings collected in some localities were investigated. Moreover, genetic structure at different spatial scales and biogeographic relationships among populations were also assessed. Our analysis showed the existence of clear patterns of genetic structure in P. oceanica in the area considered in the analysis. P. oceanica, in fact, is present in separate meadows that represent discrete populations, characterized by low genetic diversity. Comparable levels of genetic variability between mature meadows and seedlings were found. Patterns of genetic relatedness among populations seem to be in accord with direction of dominant current flux in the whole area, separating South Tyrrhenian from North Tyrrhenian populations. Moderate levels of gene flow between populations and genetic substructure within populations, together with the finding of the limited role of sexual reproduction in increasing genetic variability, should be a cause for concern for the persistence of this essential resource in the Mediterranean basin.  相似文献

Differences in habitat use can bridge early and late stages of speciation by initiating assortative mating. Heliconius colour pattern races might select habitats over which each pattern confers a relative fitness advantage because signal efficacy of wing patterns can vary by environment. Thus habitat preferences could serve to promote the evolution of mimetic colour patterns for mate choice. Here I compare colour pattern genotype and phenotype frequencies to environmental variation across the H. erato hydara x H. erato erato hybrid zone in French Guiana to determine whether races exhibit habitat preferences. I found that genotype and phenotype frequencies correspond to differences in land cover moreso than to other environmental factors. Temporal shifts in colour pattern genotypes, phenotypes and land cover also were associated at individual sample sites, which further suggests that H. erato races differ in habitat use and that habitat preferences may promote speciation among Heliconius butterflies.  相似文献

应用地统计学的原理和方法,对降水量117.1mm、降水量和潜在蒸发量比值为0.05的荒漠绿洲过渡带斑块状植被优势植物泡泡刺、红砂种群的密度、盖度以及泡泡刺沙堆体积和高度的空间格局进行了研究.结果表明,泡泡剌和红砂种群平均盖度分别为1.56±2.34%和0.23±0.22%;密度分别为1.5±1.8丛·100m^-2和2.3±2.1株·100m^-2.泡泡刺灌丛沙堆是稳定戈壁下垫面受风沙流干扰形成的一种景观,灌丛高度和体积都比较小,平均值分别为0.18±0.15m和0.10±0.18m^3.泡泡刺种群在样地内靠近绿洲和浅山区有几条明显的分布带,而红砂分布相对均匀.用半方差函数分析表明,两种种群密度和盖度的块金值与基台值之比都比较小,介于0.08~0.12之间,说明在总空间异质性中随机因素所起的作用很小。占8%~12%.两种种群盖度和密度的变程相近,基本在34m以内,其中随机因素引起的异质性对应的尺度在10m以下,而结构性因素引起的异质性对应的尺度为10~34m.泡泡刺和红砂种群盖度和密度的空间异质性均具有各向同性和各向异性特征,其中红砂种群的各向同性的范围略大于泡泡刺种群.泡泡刺种群空间格局的塑造主要受降水量及其形成地表径流的控制.  相似文献

Fossil evidence shows that populations of species that currently inhabit arctic and boreal regions were not isolated in refugia during glacial periods, but instead maintained populations across large areas of central Europe. These species commonly display little reduction in genetic diversity in northern areas of their range, in contrast to many temperate species. The mountain hare currently inhabits both temperate and arctic-boreal regions. We used nuclear microsatellite and mtDNA sequence data to examine population structure and alternate phylogeographic hypotheses for the mountain hare, that is, temperate type (lower genetic diversity in northern areas) and arctic-boreal type (high northern genetic diversity). Both data sets revealed concordant patterns. Highest allelic richness, expected heterozygosity and mtDNA haplotype diversity were identified in the most northerly subspecies, indicating that this species more closely maps to phylogeographic patterns observed in arctic-boreal rather than temperate species. With regard to population structure, the Alpine and Fennoscandian subspecies were most genetically similar (F(ST) approximately 0.1). These subspecies also clustered together on the mtDNA tree and were assigned with highest likelihood to a common Bayesian cluster. This is consistent with fossil evidence for intermediate populations in the central European plain, persisting well into the postglacial period. In contrast, the geographically close Scottish and Irish populations occupied separate Bayesian clusters, distinct clades on the mtDNA maximum likelihood tree and were genetically divergent from each other (F(ST) > 0.4) indicating the influence of genetic drift, long isolation (possibly dating from the late glacial era) and/or separate postglacial colonisation routes.  相似文献

The common octopus, Octopus vulgaris Cuvier 1797, is a largely exploited cephalopod species in the Mediterranean Sea and the Atlantic Ocean, as well as along the coasts of Africa, Brazil and Japan, where its taxonomic identity is still debated. The assessment of its genetic structure is a pressing need to correctly manage the resource and to avoid overfishing and collapsing of local stocks. Here we analysed genetic variation and population structure of O. vulgaris using thirteen microsatellite loci in seven sampling localities from the Mediterranean Sea and one from the Atlantic Ocean. We also used a DNA barcoding approach by COI gene fragment to understand the phylogenetic relationships among the specimens here investigated and the ones whose sequences are available in literature. Our results reveal high levels of allelic richness and moderate heterozygosity in all samples investigated, and a pronounced differentiation of the Atlantic and Sicilian specimens. This latter aspect seems to support the isolation of the biota within the Strait of Messina. A certain degree of differentiation was detected among the other geographic samples within the Mediterranean Sea, which is more compatible with an island model than isolation by distance. The occurrence of null alleles affected more genetic diversity indices than population structure estimations. This study provides new insights about the genetic diversity and structure of O. vulgaris in the area of interest, which can be used as guidelines for a fisheries management perspective.  相似文献

Hybridization has presented a challenge for taxonomists and conservation biologists, since hybridizing forms could be stable evolutionary entities or ephemeral forms that are blending together. However, hybrid zones also provide a unique opportunity for evolutionary biologists who study the interaction between gene flow and reproductive isolation in speciation. Three forms of woodpeckers (sapsuckers; genus Sphyrapicus) in North America that are mostly geographically separated but hybridize with each other where they come into contact present a remarkable system for the study of hybridization. We provide the first comprehensive analysis of phenotypic and genetic variation across a hybrid zone between two of these forms, the red‐breasted Sphyrapicus ruber and yellow‐bellied S. varius sapsuckers. The objective was to infer whether selection maintains the differences between forms. Our analysis of eight morphometric and 20 plumage traits, and two molecular markers showed clear differences between the forms and roughly concordant clinal variation across a narrow hybrid zone. Thirty percent of sampled birds in the hybrid zone had mixed west/east genotypes at the genetic markers examined. The center of the genetic cline was located 20 km west of the crest of the Rocky Mountains. The width of the zone was 122 km, narrower than would be expected under neutral blending given reasonable estimates of the age of the zone and individual dispersal distances. Heterozygote deficit and cytonuclear disequilibrium at the centre of the hybrid zone suggested nonrandom mating or limited hybridization. Given these patterns and lack of evidence for habitat segregation we conclude that this hybrid zone is maintained by selection, most likely in the form of hybrid inferiority. This study provides an illustrative example of extensive hybridization between stable entities, providing additional evidence against the historical practice of treating hybridizing forms as members of the same species.  相似文献

A medium-sized Octopus species is described based on materialcollected in shallow equatorial waters around the oceanic islandsof Fernando de Noronha Archipelago, Rocas Atoll, St Peter andSt Paul Archipelago and the mainland of northeastern Brazil.The new species, Octopus insularis, is described morphologically,and also characterized by the large mitochondrial subunit ribosomalRNA gene (mt 16S rDNA). The new species has relatively shortand stout arms, rugose reddish brown skin in preserved specimens,8 to 11 gill lamellae on the outer demibranchs, small ligula,characteristic symmetrical radula, spermatophore and beak, smalleggs and high fecundity (213,000 oocytes under 1.5 mm diameter).The habitats and skin patterns of living animals are brieflydescribed. A comparison of the genetic structure of Chimaera monstrosa populations from the Atlantic Ocean and the Mediterranean Sea was carried out using mitochondrial DNA analysis. Results indicate high and significant pairwise ΦST values with no shared haplotypes between the two areas. Furthermore, migration rate estimates suggested absence of gene flow between the two basins. These findings, coupled with the species vertical distribution, suggest that the shallow depth of the Strait of Gibraltar may act as a barrier limiting the dispersal capabilities of these populations, which likely became separated at the end of the middle Pleistocene.  相似文献

The longstanding biotic interactions hypothesis predicts that herbivore pressure declines with latitude, but the evidence is mixed. To address gaps in previous studies, we measured herbivory and defence in the same system, quantified defence with bioassays, and considered effects of leaf age. We quantified herbivory and defence of young and mature leaves along a continental gradient in eastern North America in the native herb Phytolacca americana L. Herbivory in the field declined with latitude and was strongly correlated with lepidopteran abundance. Laboratory bioassays revealed that leaf palatability was positively correlated with latitude of origin. Young leaves were more damaged than mature leaves at lower latitudes in the field, but less palatable in bioassays. Both defence and palatability displayed non‐linear latitudinal patterns, suggesting potential mechanisms based on biological or climatic thresholds. In sum, observational and experimental studies find patterns consistent with high herbivore pressure and stronger plant defences at lower latitudes.  相似文献

Evidence is presented from fluorescence histochemistry studies that blood vessels in the viscera of octopus vulgaris are innervated by nerve fibres containing catecholamines. Summary Evidence is presented for acetylcholine as neurotransmitter in the sensory epithelia (macula and crista) of the statocyst of Octopus vulgaris, based on the following techniques: (i) histochemical assay of acetylcholinesterase at light- and electron-microscopical levels, in combination with the detailed knowledge of the ultrastructural and neuronal organization of the receptor epithelia; (ii) lesion/degeneration experiments of the efferent fibre system; (iii) radiochemical assay of acetylcholine; and (iv) bioassay of acetylcholine.
