对已分离到的产油酵母通过木糖摇瓶发酵,从中筛选到2株H2-1和J2-2利用蔗渣半纤维素水解液高产油脂酵母,其利用蔗渣半纤维素水解液油脂得率系数分别为13.49和10.28,均显示出对蔗渣半纤维素水解液的高转化率.根据常规生理生化特征和26S rDNA序列分析结果,确认H2-1为小红酵母(Rhodotorula minuta),J2-2为粘红酵母(Rhodotorula glutinis).  相似文献

作者从中国的北京、湖北等地的鸟粪,霉变柿子以及酒药中分离到了三株未描述的丝孢酵母菌。本文描述了这三个新种:板仓丝孢酵母(Trichosporon bancangense),北京丝孢酵母(Tr.beijingense)和中国丝孢酵母(Tr.sinense)并讨论了它们与属中近似种的差别。本文还发现了丝孢酵母属的4个新记录:皮刺丝孢酵母(Tr.aculeatum),埃利丝孢酵母(Tr.eriense),斐氏丝孢酵母(Tr.figueriae)和蜜二糖丝孢酵母(Tr.melibiosaceam)。  相似文献

生物质是一种可再生资源,生物质发酵可产生高端化工产品.本文主要探讨蒸汽爆破处理玉米秸秆及水解可发酵单糖,考察酵母发酵玉米秸秆糖化液产麦角甾醇的应用研究.实验结果表明:当固液比10%,盐酸浓度1.5%,90℃水解反应3 h,还原糖含量达到53.3%,纤维素转化率79%.发酵工艺参数为玉米秸秆糖化液6.0°Bx,玉米浆4%,pH 7.5,接种量10%,28℃摇床振荡培养32 h,细胞生物量达8.5 g/L,麦角甾醇含量可达2.35%.同时对玉米秸秆发酵产麦角甾醇晶体进行结构表征.  相似文献

采用新型常压室温等离子体射流诱变产油酵母,结合快速突变产油酵母操作条件及基于96孔板的高通量筛选手段,获得了一系列增殖速度和产油量发生变化的突变株。在等离子体对菌株致死率为99%的条件下获得的以突变株增殖速度为指标的正突变率达到27.2%。用含酵母粉 (10 g/L)、蛋白胨 (10 g/L) 及葡萄糖 (20 g/L) 的酵母膏胨葡萄糖培养基进行发酵实验表明,筛选得到的高产突变体产油量从对照株的1.87% (W/W) 增加到4.07% (W/W)。  相似文献

发酵性丝孢酵母HWZ004利用水稻秸秆水解液发酵产油脂   总被引:3,自引:0,他引:3  
为高效利用水稻秸秆中的纤维素和半纤维素产油脂,采用稀酸预处理和酶水解两步法对水稻秸秆进行水解,然后以水解液为碳源,培养发酵性丝孢酵母Trichosporon fermentans HWZ004产微生物油脂。结果表明,经简单overliming法脱毒后水稻秸秆水解液中乙酸、糠醛和5-羟甲基糠醛的浓度分别为0.4 g/L、0.1 g/L和0.05 g/L。只需添加少量氮源和微量CuSO4?5H2O,该水解液即可满足T. fermentans HWZ004发酵产油脂的要求。发酵最适接种量、初始pH和温度分别是5.0%、7.0和25 ℃。T. fermentans HWZ004在优化条件下培养7 d的生物量、油脂含量和油脂产量分别是26.4 g/L,52.2%和13.8 g/L;油脂得率系数为17.0,大大高于驯化前菌株T. fermentans CICC 1368在脱毒水稻秸秆半纤维素水解液中的对应值 (11.9)。所产油脂的脂肪酸组成与植物油相似,不饱和脂肪酸含量达70%以上,宜作为生物柴油的生产原料。  相似文献

本文研究了诱导热带假丝酵母子囊孢子产生和子囊破壁的条件,以及单倍体分离和鉴别方法.结果表明:NaAc、KCl这两种成分即可满足热带假丝酵母的产孢营养要求,在含有NaAc 8.2 g/L、KCl 1.8 g/L的琼脂培养基中,28℃培养7 d,热带假丝酵母细胞的产孢率可达到47.5%;单纯使用蜗牛酶对破除热带假丝酵母子囊壁的效率甚低,酶解液加入适量的石英砂同时振荡处理4 h左右,可以显著提高对热带假丝酵母子囊破壁速率.用这种方法处理,绝大多数子囊壁均可破除.子囊破壁处理后再以52℃处理10 min杀死残余的营养体细胞,分离物的单倍体孢子比例可由灭活处理前的48%,提高到76%以上.  相似文献

以发酵性丝孢酵母胞壁酯酶为研究对象,研究了其在不同理化条件下的稳定性。结果表明:0 ℃保存24 h,酯酶仍保持81.5%原酶活;在40 ℃完全失活;5 mmol/L K+、Mn2+和Fe2+提高酶活力6.9%以下,5 mmol/L Ca2+、Cu2+分别降低酶活力90.7%和73.3%;1%(质量分数,下同) Tween-40和TritonX-100保持77%以上原酶活,1% Tween-80提高酶活力23.3%,1%十二烷基磺酸钠(Sodium dodecyl sulfate,SDS)和十六烷基三甲基溴化铵 (Cetyl trimethyl ammonium bromide,CTAB)完全抑制酶活;5%甘油、阿拉伯胶和糊精分别提高酶活力57%、29.6% 和2%,5%可溶性淀粉和牛血清蛋白(Albumin from bovine serum,BSA)分别降低酶活力20.4%和71.5%。发酵性丝孢酵母酯酶热稳定性差,宜10 ℃以下贮存,甘油对其保护作用良好,BSA强烈抑制该酶活性。该酯酶对处理含油脂废水和增强洗涤剂洗涤效果有潜在的应用价值。  相似文献

用脱壁酶(纤维素酶、半纤维素酶和蜗牛酶)作用于皮状丝孢酵母ST851制备原生质体。本文就酶的种类、酶液浓度、酶作用时间与温度、酶混合时间、混酶作用效果、酵母细胞的不同生长期诸因素对ST851原生质体形成的影响,进行了较为详尽的探讨。结果表明采用混酶作用(即先用纤维素酶和半纤维素酶预处理后再用蜗牛酶作用)是较理想的破壁条件,在pH5.8、37℃条件下作用3.5-4hr,破壁率最高可达95-98%。  相似文献

最新的酵母菌分类系统,丝孢酵母属Trichosporon Behrend仅限于产生节孢子的担子菌酵母的无性型。因此,以前根据该属旧的定义在国内发表的三个新种板仓丝孢酵母T. bancangense、北京丝孢酵母T. beijingense和中国丝孢酵母T. sinense的分类地位,需要进行调整。根据26S rDNA D1/D2区的序列分析和最新标准方法重新测定的生理生化反应,发现上述三个种名分别是子囊菌酵母Saccharomycopsis fibuligera, Pichia burtonii和Issatchenkia orientalis的异名。  相似文献

丝孢酵母高甲硫氨酸突变株的选育及营养调控   总被引:4,自引:0,他引:4  
以丝孢酵母(Trichosporon Behr)ST851为原始菌株,经紫外线诱变,在含乙硫氨酸的双层平板上筛选到多株抗乙硫氨酸突变株。其中ST851-10株抗乙硫氨酸浓度达到350μg/ml,其菌体蛋白质含量由40.5%提高到44.3%,菌体甲硫氨酸含量由20.45mg/g-DCW增加到29.32mg/g-DCW。在以苹果渣为碳源、尿素为氮源、硫酸镁作硫源的最适培养条件下,固态发酵24h后,蛋白质和甲硫氨酸含量较原始菌株分别提高了15.8%和44.9%。培养基中C/N值低有利于甲硫氨酸的合成,C/N值高则适合于菌体生长。在苹果渣固态发酵过程中,适当补加氮源既有利于菌体生长和甲硫氨酸的合成,又可起到调节培养基pH值的作用。  相似文献

In this work, corncob acid hydrolysate and its simulated medium whose sugar composition was the same as the corncob acid hydrolysate were used as fermentation substrate for lipid production by oleaginous yeast Trichosporon dermatis. On the corncob acid hydrolysate, after 7 days of fermentation, the biomass, lipid content, lipid yield, and lipid coefficient of T. dermatis were 17.3 g/L, 40.2%, 7.0 g/L, and 16.5%, respectively. Interestingly, during the lipid fermentation on the corncob acid hydrolysate, glucose, xylose, arabinose, and even acetic acid could be well utilized as carbon sources by T. dermatis. Surprisingly, the lipid yield (7.0 g/L) of T. dermatis on the corncob acid hydrolysate was much higher than that (3.8 g/L) on the simulated medium, in spite of the fact that the lipid coefficient (17.4%) on the simulated medium was a little higher. This phenomenon further showed that lignocellulosic acid hydrolysate was a suitable substrate for lipid fermentation by T. dermatis. This work would help the comprehensive utilization of lignocellulosic biomass for lipid production.  相似文献

In this study, crude cellulase produced by Trichoderma reesei Rut-30 was used to hydrolyze pretreated straw. After the compositions of the hydrolysate of pretreated straw were optimized, the study showed that natural components of pretreated straw without addition of any other components such as (NH4)2SO4, KH2PO4, or Mg2+ were suitable for citric acid production by Yarrowia lipolytica SWJ-1b, and the optimal ventilatory capacity was 10.0 L/min/L medium. Batch and fed-batch production of citric acid from the hydrolysate of pretreated straw by Yarrowia lipolytica SWJ-1b has been investigated. In the batch cultivation, 25.4 g/L and 26.7 g/L citric acid were yields from glucose and hydrolysate of straw cellulose, respectively, while the cultivation time was 120 hr. In the three-cycle fed-batch cultivation, citric acid (CA) production was increased to 42.4 g/L and the cultivation time was extended to 240 hr. However, iso-citric acid (ICA) yield in fed-batch cultivation (4.0 g/L) was similar to that during the batch cultivation (3.9 g/L), and only 1.6 g/L of reducing sugar was left in the medium at the end of fed-batch cultivation, suggesting that most of the added carbon was used in the cultivation.  相似文献

毛壳霉CQ31的鉴定及固体发酵产木聚糖酶条件的优化   总被引:2,自引:0,他引:2  
从土壤中筛选出一株产木聚糖酶的真菌CQ31, 经鉴定后命名为毛壳霉CQ31。该菌能够利用几种农业废弃物固体发酵高产木聚糖酶, 玉米杆为最佳碳源。单因素优化试验表明: 以玉米杆为碳源, 胰蛋白胨为氮源, 初始水分含量80%, 初始pH值9.0为最佳产酶条件。在优化后的条件下培养7 d产木聚糖酶水平高达4897 U/g干基碳源, 此时甘露聚糖酶酶活达803 U/g干基碳源。因此, 毛壳霉CQ31固体发酵产木聚糖酶和甘露聚糖酶具有一定的工业化应用前景。  相似文献

以"凤丹"牡丹根际土壤中分离筛选到的产脂肪酶菌株Pseudomonas sp. RYXP作为出发菌株,对其进行了紫外线诱变选育,并采用单因素试验和正交试验方法对活性最强正突变株的产脂肪酶基本特性进行了测定。结果表明,出发菌株Pseudomonas sp. RYXP的紫外线诱变最佳条件为:15 W紫外灯30 cm距离照射1 min;将产脂肪酶活性最强的正突变菌株编号为RYXP-3,单因素试验表明RYXP-3产脂肪酶适宜的碳源为玉米淀粉,适宜氮源为豆饼粉,适宜的磷酸二氢钾含量是0.3%,适宜的初始pH值为7;正交试验表明RYXP-3的最佳的产酶培养基成分组成是:玉米淀粉7%,豆饼粉3%,磷酸二氢钾0.3%,初始pH值为8。在优化方案A1B2C2D3产酶条件下,突变株RYXP-3最高的产酶活性达到56.1 U/mL。突变菌株RYXP-3可作为产油牡丹"凤丹"专用促生菌肥开发的备选资源菌株。  相似文献

脂肪酶可以催化甘油三酯水解成脂肪酸和甘油,已广泛应用在工业领域,而获得产酶微生物是研究的基础。采用油脂平板法筛选出1株脂肪酶产生菌。经16S rRNA序列分析可知,该菌株属于柠檬酸杆菌(Citrobacter werkman and Gillen)。单因素试验对其进行产酶条件优化,优化后产酶条件(g/L):淀粉2.0,KH2PO4 1.0,K2HPO4·3H2O 2.2,(NH4)2SO4 1.0,MgSO4·7H2O 0.1,牛肉膏2.0,橄榄油10.0 mL,pH 7.5,接种量1.5%(v/v),37 ℃培养43 h。获得最大酶活为384 U/mL,是优化前的13倍。可以利用该菌制备脂肪酶。  相似文献

Cyathus stercoreus grown on wheat straw had a higher xylanase activity than when it was grown on rice husk or extracted hemicellulose. Inclusion of casein hydrolysate, Tween 80 and Mn2+ (at 0.02%, 0.2% and 0.075%, respectively) increased the production of extracellular xylanase. Optimal yield of xylanase (0.73 U/ml) was at pH 5.6 after 9 to 12 days at 30°C. 为了进一步提高极细链格孢菌产蛋白激发子的产量,通过单因子和多因子试验与分析,筛选优化了适于极细链格孢菌产生蛋白激发子的培养基和培养条件,并检测了发酵过程中pH、还原糖、氨基氮和菌丝量变化以及与蛋白激发子产量的关系。结果表明,土豆淀粉和黄豆粉对蛋白激发子产量影响最大,其次是蛋白胨和无机盐。优化的发酵培养基主要成分(g/L):碳源I 15、葡萄糖5、玉米淀粉5、土豆淀粉20、谷氨酸10、氮源I5、黄豆粉10、硫酸铵5。确定了优化的培养条件,调整培养基起始pH为7.0~7.5,将18h菌龄的种子培养液按10%接种量接种到装液量为75mL的500mL摇瓶中,在温度(28±1)℃、摇床转速180r/min下培养可获得理想的蛋白产量。在优化的培养基和培养条件下,发酵12~48h该菌进入对数生长期,48h进入稳定生长期,60h菌丝扣蛋白激发子产量达最高。蛋白产量与菌体生物量呈正相关,当还原糖、总糖量消耗到最低水平时,菌丝产量和蛋白激发子产量达最高。优化的培养基菌丝干重收率迭3.9g/100mL,蛋白激发子产量达到5.17g/L,比普通的土豆液体培养基提高近4倍。  相似文献

Bioethanol production from lignocellulosic materials has several limitations. One aspect is the high production cost of cellulases used for saccharification of substrate and inhibition of fermenting yeast due to inhibitors released in acid hydrolysis. In the present work we have made an attempt to achieve simultaneous cellulases production, saccharification and detoxification using dilute acid hydrolysate of Saccharum spontaneum with and without addition of nutrients, supplemented with acid hydrolyzed biomass prior to inoculation in one set and after 3 days of inoculation in another set. Organisms used were T. reesei NCIM 992, and Aspergillus niger isolated in our laboratory. Cellulase yield obtained was 0.8 IU/ml on fourth day with T. reesei. Sugars were found to increase from fourth to fifth day, when hydrolysate was supplemented with nutrients and acid hydrolyzed biomass followed by inoculation with T. reesei. Phenolics were also found to decrease by 67%.  相似文献

目的:米曲霉木聚糖酶生产菌的理性选育及发酵优化。方法:以米曲霉FS018为出发菌株,采用紫外和激光诱变先后处理3代,以抗高浓度葡萄糖阻遏效应为筛选模型获得一株高产木聚糖酶菌株FST036,并对该菌株的发酵培养基进行了初步优化。结果:突变株FST036产酶水平可达4200.57U/mL,产酶水平比出发菌株提高了24%。对该菌株的发酵培养基初步优化结果表明:培养基的最适氮源为牛肉膏;最适碳源为麸皮与玉米芯组合,总浓度为4%。二者最适比例为7:3;对该菌株的发酵条件初步优化结果表明:最适接种量1%,初始pH6.78,250mL三角瓶装液量为45mL,培养72h酶活可达5404.24U/mL。结论:反复多次诱变处理,并结合抗高浓度葡萄糖阻遏效应作为一种理性筛选模型,为通过诱变育种来获得曲霉木聚糖酶高产菌株提供了一条有效的途径。  相似文献

探讨了褐藻胶降解菌株S10的生长条件及其对产褐藻胶降解酶活力的影响。以分离自海参肠道的褐藻胶降解菌株S10为研究对象,采用形态学观察结合16S rDNA序列分析,对菌株S10进行菌种鉴定并对其生理生化特性进行测定。以降解酶活力为指标,利用单因素、Plackett-Burman(PB)和响应面法对培养基成分和培养条件进行优化;最后对优化前后的菌株生长量、产酶活力和粗酶液稳定性进行分析。结果表明,菌株S10属于溶藻孤菌(Vibrio algindyticus);当pH 7、接种量2%(体积分数)、装液量150 mL、温度26℃、转速150 r/min、NaCl 3%(质量分数,下同)、海藻酸钠含量1.12%、硫酸铵含量0.44%、培养时间35.95 h条件下,褐藻胶降解酶活力最大(188.18 U/min)。优化后产酶活力提高30%;4℃低温更有利于该酶保存。综上,优化后的菌株S10产褐藻胶降解酶活力较高,能更好地用于降解褐藻胶,可为提高褐藻胶的利用率和进一步发掘褐藻胶寡糖的利用价值提供参考。  相似文献
